Developmental changes in hemolymph ecdysteroid level, ecdysteroid synthesis by prothoracic glands (PGs) in vitro, prothoracicotropic hormone (PTTH) activity in brain extracts, and PTTH activity in the hemolymph were measured during the fifth larval instar of the Eri silkworm, Samia cynthia ricini. The changing patterns of hemolymph ecdysteroid level and ecdysteroid synthesis by laGs in vitro are similar to each other, with maximums on day 9. However, on this day, hemolymph ecdysteroid level was substantially higher than ecdysteroid synthesis by PGs in vitro suggesting a high PTTH activity in the hemolymph on day 9. Moreover, the changing pattern of PTTH activity in brain extracts is also similar to that of PTTH activity in the hemolymph, both peaking on day 9. However, on this day, activity in brain extracts was much smaller than PTTH activity in the hemolymph implying that most PTTH synthesized by the brain is secreted to the hemolymph and the brain stores a very little amount of PTTH. This study provides unique insights onto the hormonal regulation of ecdysteroid synthesis in the Eri silkworm and is useful for our future studies on signal transduction of insect neurolaelatides.  相似文献

【目的】蓖麻蚕Samia cynthia ricini 属于鳞翅目昆虫,其体内存在的胰凝乳蛋白酶抑制剂(chymotrypsin inhibitor, CI)对蓖麻蚕的正常生长发育和自身防御能力有重要的作用。获得蓖麻蚕的CI种类分布信息,以及研究各种CI的功能,可以为鳞翅目害虫的生物防治提供理论依据。【方法】本实验通过非变性聚丙烯酰胺凝胶电泳(native-PAGE)和CI活性染色技术,对18个蓖麻蚕品系的主要组织器官中胰凝乳蛋白酶抑制剂的多态性分布进行了调查,并对各品系血液中的蛋白含量和CI活性进行了测定。【结果】在pH 8.6电泳下,各品系血液中共检测到7条CI活性带,在pH 4.0电泳下,各品系血液中可检测到3条CI活性带;血液中的CI含量从5龄第3天幼虫期到化蛹当天一直维持着很高的含量;头、体壁和脂肪体中的CI都不如血液中的CI含量高,而丝腺中未检测到任何CI。【结论】与家蚕 Bombyx mori 血液中的含量相比,蓖麻蚕血液中的CI含量更高,CI种类多态性分布不仅体现在不同品系的同一组织,还体现在同一品系的不同组织。  相似文献

Deforestation and exploitation has led to the fragmentation of habitats and scattering of populations of the economically important eri silkworm, Samia cynthia ricini, in north-east India. Genetic analysis of 15 eri populations, using ISSR markers, showed 98% inter-population, and 23% to 58% intra-population polymorphism. Nei's genetic distance between populations increased significantly with altitude (R(2) = 0.71) and geographic distance (R(2) = 0.78). On the dendrogram, the lower and upper Assam populations were clustered separately, with intermediate grouping of those from Barpathar and Chuchuyimlang, consistent with geographical distribution. The Nei's gene diversity index was 0.350 in total populations and 0.121 in subpopulations. The genetic differentiation estimate (Gst) was 0.276 among scattered populations. Neutrality tests showed deviation of 118 loci from Hardy-Weinberg equilibrium. The number of loci that deviated from neutrality increased with altitude (R(2) = 0.63). Test of linkage disequilibrium showed greater contribution of variance among eri subpopulations to total variance. D('2)IS exceeded D('2)ST, showed significant contribution of random genetic drift to the increase in variance of disequilibrium in subpopulations. In the Lakhimpur population, the peripheral part was separated from the core by a genetic distance of 0.260. Patchy habitats promoted low genetic variability, high linkage disequilibrium and colonization by new subpopulations. Increased gene flow and habitat-area expansion are required to maintain higher genetic variability and conservation of the original S. c. ricini gene pool.  相似文献

Insects produce silk for a range of purposes. In the Lepidoptera, silk is utilized as a material for cocoon production and serves to protect larvae from adverse environmental conditions or predators. Species in the Saturniidae family produce an especially wide variety of cocoons, for example, large, golden colored cocoons and those with many small holes. Although gene expression in the silk gland of the domestic silkworm (Bombyx mori L.) has been extensively studied, considerably fewer investigations have focused on members of the saturniid family. Here, we established expression sequence tags from the silk gland of the eri silkworm (Samia ricini), a saturniid species, and used these to analyze gene expression. Although we identified the fibroin heavy chain gene in the established library, genes for other major silk proteins, such as fibroin light chain and fibrohexamerin, were absent. This finding is consistent with previous reports that these latter proteins are lacking in saturniid silk. Recently, a series of fibrohexamerin‐like genes were identified in the Bombyx genome. We used this information to conduct a detailed analysis of the library established here. This analysis identified putative homologues of these genes. We also found several genes encoding small silk protein molecules that are also present in the silk of other Lepidoptera. Gene expression patterns were compared between eri and domestic silkworm, and both conserved and nonconserved expression patterns were identified for the tested genes. Such differential gene expression might be one of the major causes of the differences in silk properties between these species. We believe that our study can be of value as a basic catalogue for silk gland gene expression, which will yield to the further understanding of silk evolution.  相似文献

Both allatotropic and allatostatic activities were found in crude extracts of brain from adult and larval Eri silkworm, Samia cynthia ricini, but it seems that allatotropic activity dominates in each stage. There was a high level of allatotropic activity in the crude extract of brain from newly emerged female adults, but allatostatic activity appeared in the bioassay when excessive amounts of crude extracts of brain were added. Crude extracts of brain from premoulting fourth‐instar larvae and from newly ecdysed fifth‐instar larvae exhibited allatotropic activities, whereas extracts of brain from the second and third day of the fifth‐instar larvae inhibited juvenile hormone (JH) release slightly. Allatotropic activity from the brains of adults and larvae stimulated both adult and larval corpora allata (CA) to synthesize JH. Manduca sexta allatotropin (AT) (Mas‐AT) and M. sexta allatostatin (AST) (Mas‐AST) also stimulated and inhibited both adult and larval S. cynthia ricini CA to synthesize JH, respectively. Higher concentrations of Mas‐AT (10?4 or 10?3 M) showed an inhibitory effect on adult CA. CA from newly emerged female adults were the most sensitive to inhibition by Mas‐AST, whereas CA from female pharate adults at about 6 h before adult emergence were the most sensitive to stimulation by Mas‐AT and S. cynthia ricini brain allatotropic activity. An extract of brain and Mas‐AT induced some of the non‐active female pharate adult CA at 12 h before emergence to synthesize a small amount of JH.  相似文献

A cDNA clone encoding gallerimycin was isolated from larval fat body of immunized Samia cynthia ricini and named as Scr-gallerimycin. In naive larvae, no gene expression was detected, but strongly induced in fat body and hemocytes following immune challenge with bacteria or entomopathogenic fungus Beauveria bassiana. Strong expression of the gene was also induced by injection of peptidoglycan and zymosan, but very weakly by non-pathogenic fungus Aspergillus oryzae. Analysis of the sequence upstream from the cDNA shows the presence of motifs homologous to binding sites for NF-κB, C/EBP and CRE-BP1.  相似文献

During the period of adult emergence in the Eri silkworm, Samia cynthia ricini, the corpora allata (CA) are apparently reactivated in females, but not males. This creates a significant sexual dimorphism in juvenile hormone (JH) synthesis by CA. To determine the underlying molecular mechanisms in this process, we cloned cDNAs of two enzymes involved in the JH synthesis pathway: 3-hydroxy-3-methylglutaryl CoA reductase (HMGR) and juvenile hormone acid methyl transferase (JHAMT). Both Samcri-HMGR and -JHAMT mRNAs were detected in CA almost exclusively. However, their expression patterns were different from each other. During the period of adult emergence, Samcri-HMGR was expressed in CA at a constantly high level suggesting it plays little role for the regulation of JH synthesis. In contrast, the patterns of both Samcri-JHAMT mRNA level and enzyme activity were closely correlated with the patterns of JH synthesis, CA reactivation, and sexual dimorphism of JH synthesis. In addition, JHAMT mRNA levels were paralleled JH synthesis in the fifth-instar larvae of S. cynthia ricini and the pharate adults of the silkworm Bombyx mori. We infer from these results that JHAMT is a key regulatory enzyme for JH synthesis in the Eri silkworm.  相似文献

以家蚕Bombyx mori丝素重链基因、丝胶基因1和胰凝乳蛋白酶抑制因子13基因为探针,对野桑蚕B.mandarina、蓖麻蚕Philosamia cynthia ricini和家蚕B.mori基因组DNA进行限制性片段长度多态性分析。结果发现,在野桑蚕、蓖麻蚕基因组中存在着家蚕丝素重链基因、丝胶基因1的同源序列,而在中日野桑蚕以及蓖麻蚕品种间存在着限制性酶切位点差异;丝胶基因1在中国野桑蚕基因组的EcoRⅠ酶切图谱较日本野桑蚕与家蚕更为一致,表明家蚕与中国野桑蚕亲缘关系更近。此外,在野桑蚕基因组中发现了家蚕胰凝乳蛋白酶抑制因子13基因的同源序列,并且在家蚕品种间以及中日野桑蚕之间也存在着多态性。这些结果表明不同绢丝昆虫在适应生存环境的进化过程中,基因组发生了结构改变。  相似文献

There are many kinds of silks from silkworms and spiders with different structures and properties, and thus, silks are suitable to study the structure-property relationship of fibrous proteins. Silk fibroin from a wild silkworm, Samia cynthia ricini, mainly consists of the repeated similar sequences by about 100 times where there are alternative appearances of the polyalanine (Ala)(12-13) region and the Gly-rich region. In this paper, a sequential model peptide, GGAGGGYGGDGG(A)(12)GGAGDGYGAG, which is a typical sequence of the silk fibroin, was synthesized, and the atomic-level conformations of Gly residues at the N- and C-terminal ends of the polyalanine region were determined as well as that of the central Ala residue using (13)C 2D spin diffusion solid-state nuclear magnetic resonance (NMR) under off-magic angle spinning. In the model peptide with alpha-helical conformation, the torsion angle of the central Ala residue, the 19th Ala, was determined to be (phi, psi) = (-60 degrees, -50 degrees ), which was a typical alpha-helical structure, but the torsion angles of two Gly residues, the 12th and 25th Gly residues, which are located at the N- and C-terminal ends of the polyalanine region, were determined to be (phi, psi) = (-70 degrees, -30 degrees ) and (phi, psi) = (-70 degrees, -20 degrees ), respectively. Thus, it was observed that the turns at both ends of polyalanine with alpha-helix conformation in the model peptide are tightly wound.  相似文献

Background

All reported plant ferritins are heteropolymers comprising two different H-type subunits. Whether or not homopolymeric plant ferritin occurs in nature is an open question.

Methods

A homopolymeric phytoferritin from adzuki bean seeds (ASF) was obtained by various protein purification techniques for the first time, which shares the highest identity (89.6%) with soybean seed H-1 ferritin (rH-1). Therefore, we compared iron oxidation activity and protein stability of ASF with those of rH-1 by stopped-flow combined with light scattering or UV/Vis spectrophotography, SDS- and native- PAGE analyses. Additionally, a new rH-1 variant (S68E) was prepared by site-directed mutagenesis approach to elucidate their difference in protein stability.

Results

At high iron loading of protein, the extension peptide (EP) of plant ferritin was involved in iron oxidation, and the EP of ASF exhibited a much stronger iron oxidative activity than that of rH-1. Besides, ASF is more stable than rH-1 during storage, which is ascribed to one amino acid residue, Ser68.

Conclusions

ASF exhibits a different mechanism in iron oxidation from rH-1 at high iron loading of protein, and a higher stability than rH-1. These differences are mainly stemmed from their different EP sequences.

General significance

This work demonstrates that plant cells have evolved the EP of phytoferritin to control iron chemistry and protein stability by exerting a fine tuning of its amino acid sequence.  相似文献

This paper presents a new method for identifying retinoblastoma related genes by integrating gene expression profile and shortest path in a functional linkage graph. With the existing protein-protein interaction data from STRING, a weighted functional linkage graph is constructed. 119 consistently differentially expressed genes between retinoblastoma and normal retina were obtained from the overlap of two gene expression studies of retinoblastoma. Then the shortest paths between each pair of these 119 genes were determined with Dijkstra's algorithm. Finally, all the genes present on the shortest paths were extracted and ranked according to their betweenness and the 119 shortest genes with a betweenness greater than 100 and with a p-value less than 0.05 were selected for further analysis. We also identified 53 retinoblastoma related miRNAs from published miRNA array data and most of the 238 (119 consistently differentially expressed genes and 119 shortest path genes) retinoblastoma genes were shown to be target genes of these 53 miRNAs. Interestingly, the genes we identified from both the gene expression profiles and the functional protein association network included more cancer genes than did the genes identified from the gene expression profiles alone. In addition, these genes also had greater functional similarity to the reported cancer genes than did the genes identified from the gene expression profiles alone. This study shows promising results and proves the efficiency of the proposed methods.  相似文献

Exposure of the silkworm(Philosamia ricini) larvae to cold temperature (2‡C) and subsequent exposure to room temperature (29‡ C) resulted in the mortality of the larvae. Cold exposure brought about significant decrease in enzymic activity of proteases, aminotransferases, diacylglycerol lipase and in the amounts of some haemolymph sugars and polyols. However, glycerol increased sharply in response to severe cold exposure. There was also a marked increase in the levels of protein, pyruvate, total free amino acids, total lipid, phospholipid and triacylglycerols. In the colder environment, carbohydrates served as the energy source. Glycerol appears to have conferred cryoprotection to the cold-stressedPhilosamia ricini larvae.  相似文献

The outbreaks of fungal diseases in cultured fish have been severe in recent years, which is harmful to the healthy and sustainable development of fish farming. In this study, an investigation was conducted for significant fungal infections of 12 species of fish in four regions in Xinjiang, China, to understand the distribution of local fish fungal pathogens. Twenty-six fungal strains with pathogenicity were isolated, and the challenge experiment showed that eight strains from Changji area had high infection rate to fish eggs. Based on internal transcribed spacer sequence data and molecular analysis, the 26 strains were classified into nine different species of six fungal genera. Phylogenetic analysis showed that all strains were divided into two clades, namely Cluster 1 (contains only the genus Mucor) and Cluster 2 (consists of five small branches), and the distribution of strains from the same region was scattered in two clusters. There is no strict host selectivity for these fungi to infect fish. Mucor sp. are the main fungal pathogen of fish in these four regions, whereas Hypophthalmichthys molitrix and Carassius auratus are two types of fish that were susceptible to pathogen. In addition, the environmental adaptability experiments showed that eight highly pathogenic strains have different adaptability to the environment, and their optimum temperature and pH were 25°C and 7.0, respectively, whereas the concentration of NaCl was negatively correlated with the growth of strains. Therefore, these results indicated that the coinfection of multiple fungal pathogens in a culture region should be considered in the future study.  相似文献

Gastric cancer is one of the most common and lethal cancers worldwide. However, despite its clinical importance, the regulatory mechanisms involved in the aggressiveness of this cancer are still poorly understood. A better understanding of the biology, genetics and molecular mechanisms of gastric cancer would be useful in developing novel targeted approaches for treating this disease. In this study we used protein-protein interaction networks and cluster analysis to comprehensively investigate the cellular pathways involved in gastric cancer. A primary immunodeficiency pathway, focal adhesion, ECM-receptor interactions and the metabolism of xenobiotics by cytochrome P450 were identified as four important pathways associated with the progression of gastric cancer. The genes in these pathways, e.g., ZAP70, IGLL1, CD79A, COL6A3, COL3A1, COL1A1, CYP2C18 and CYP2C9, may be considered as potential therapeutic targets for gastric cancer.  相似文献

The economic consequences of bovine diarrhea are serious. Few long‐term epidemiological data are available concerning the causative pathogens of bovine diarrhea in Japan. From 2002 to 2011, surveillance of enteric pathogens was performed in cows of various breed and age from 302 farms in which diarrhea had occurred in Yamagata Prefecture, Japan. Differences between dairy and beef cows in the number of cases of diarrhea and rates of infection by Salmonella spp. and Eimeria spp. were found. Clinical symptoms (duration of epidemic, hematochezia and complications) caused by bovine rotavirus infection were milder than those caused by bovine coronavirus infection.  相似文献

Receptor-like cytoplasmic kinases (RLCKs) play important roles in various developmental processes and stress responses in plants. Whereas, the detailed information of this family in cassava has not clear yet. In this study, A total of 322 MeRLCK genes were identified in the cassava genome, and they could be divided into twelve clades (Clades I-XII) according to their phylogenetic relationships. Most RLCK members in the same clade have similar characteristics and motif compositions. Over half of the RLCKs possess cis-elements in their promoters that respond to ABA, MeJA, defense reactions, and stress. Under Xpm11 infection, the expression levels of four genes show significant changes, suggesting their involvement in Xpm11 resistance. Two RLCK (MeRLCK11 and MeRLCK84) genes potentially involved in resistance to cassava bacterial blight were identified through VIGS experiments. This work laid the foundation for studying the function of the cassava RLCK genes, especially the genes related to pathogen resistance.  相似文献
