The stembark of Afzelia bella yielded an acylated dihydroflavonol glycoside identified as 2R,3R-trans-aromadendrin-7-O-beta-D-glucopyranoside-6'-(4'-hydroxy-2'-methylene butanoate), along with five known flavonoids and the lignan glycoside (+)-isolariciresinol 9-O-xyloside. Their structures were determined by detailed 1D and 2D NMR analyses.  相似文献

In this paper we describe the solution properties of a new xyloglucan polysaccharide extracted from the African legume Afzelia africana Se. Pers. The polysaccharide is of high weight-average molecular weight (Mw), but application of the "pressure cell" method enabled a range of Mw fractions to be prepared. Results from the light scattering/intrinsic viscosity measurements on these fractions suggest that like other xyloglucans from tamarind and detarium it occurs in solution as a polymeric coil, with a small amount of excluded volume. Measurement of dilute and semidilute solution rheology suggests that, like these polymers, and the related galactomannan series, it forms viscous solutions at higher concentrations via entanglements.  相似文献

Ectomycorrhizal synthesis on seedlings of Afzelia quanzensis was initiated in the greenhouse and in the field using basidiospores or soil inoculum originating from fungi associated with adult trees of A. quanzensis, Brachystegia microphylla, B. spiciformis, and Julbernardia globiflora. Of the spore inocula used, only a Pisolithus sp. associated with adult A. quanzensis formed mycorrhizae. Seedlings raised in contact with all soil inocula formed mycorrhizae; however, the mycorrhizal types formed differed between soil inoculum used in the greenhouse and soil inoculum directly used in the field. A. quanzensis has a low specificity for mycorrhizal association. The concepts of ectomycorrhizal succession are also applicable to African savanna ecosystems.  相似文献

 The growth and mineral nutrition responses of seedlings of two provenances of Afzelia africana Sm. from Senegal and Burkina Faso, inoculated with four ectomycorrhizal (ECM) fungi (Scleroderma spp. and an unidentified isolate) from the same regions were assessed in a pot experiment in a savanna ECM-propagule-free soil deficient in NPK. There was little variation in the ability of the different fungal species to colonize roots of either provenance of A. africana or to produce external hyphal in soil. Root colonization by ECM fungi and their hyphal development were not related to mineral nutrition or ECM dependency. Differences in P, N, Mg and Ca concentrations in the leaves of inoculated and non-inoculated Afzelia seedlings were not always associated with production of biomass. Only leaf K concentration increased in both provenances after ECM inoculation. However, the Burkina Faso provenance responded better to inoculation with the two fungal isolates than the Senegal provenance in terms of biomass production. This was due to stimulation of root dry weight of the Burkina Faso provenance. Therefore, the hypothesis arises that non-nutritional rather than nutritional effects explain the contribution of ECM inoculation to the growth of A. africana seedlings. The logging of tree species of high commercial value is increasing throughout the African continent, yet the ecology of these species is generally poorly known. We studied the regeneration pattern and size class distribution of Afzelia quanzensis populations in northern South Africa over a 5‐year period. Recruitment was low as the annual seedling mortality was >65%. Seedlings were located under the canopy and were affected by drought and browsing. The adults were scattered or were in a clump‐dispersed pattern, which would result in higher recruitment of offspring near parents. Individuals of 0–10 cm diameter at breast height (DBH) were few, while there were 32 trees ha?1 at >10 cm DBH with an annual mortality of 0.8%. Annual diameter increments varied between 0.06 and 0.28 cm. It appears that the transition from the sapling into the juvenile stage could be a bottleneck in the regeneration of the species. A longer study, including more rainfall cycles, may reveal other patterns as dry and wet years have different impacts on dynamics.  相似文献

Two new free amino acids have been isolated from seeds of Afzelia bella: trans-4-hydroxy-L-proline and trans-4-carboxy-L-proline. They are accompanied by relatively large quantities of proline, pipecolic, acid, 4-methylene-DL-proline, cis-4-hydroxymethyl-L-proline and 4-methylene-L-glutamic acid. Small amounts of acetylornithine are also present. It is the first time that trans-4-hydroxy-L-proline has been found as a component of the free amino pool of plants.  相似文献

Stem length, number of secondary lateral roots, shoot dry weight and reducing sugar concentrations of root were significantly reduced when translocation of reserves from cotyledons to the roots of Afzelia africana seedlings was interrupted by complete or partial cotyledon excision. The sucrose but not the glucose concentration of lateral roots also decreased significantly after complete cotyledon excision. Hartig net development rather than fungal sheath formation was affected after inoculation with the early fungal isolate E1 and by both late-stage fungal isolates L1 and L2 after partial or complete cotyledon excision. However, mycorrhizal colonization by the early fungal isolate E2 was not affected by cotyledonary reserves, suggesting that this fungal isolate has a lower carbohydrate requirement than fungal isolates E1, L1 and L2. The late-stage fungal isolates L1 and L2 induced a hypersensitivity reaction by epidermal cell walls of the host plant after complete cotyledon excision, suggesting they are more dependent than the early fungal isolate E1 on available root carbohydrate substrates for ectomycorrhizal colonization. These results are discussed in the light of the hypothesis that early and late-stage fungi were different carbohydrate requirements, and that the time sequence of colonization was related to the root carbohydrate status, which increased with time.  相似文献

This paper is the first multi-scale characterization of the xyloglucan extracted from seeds of the African tree Afzelia africana Se. Pers. It describes the extraction and characterization of this polysaccharide in terms of both primary monosaccharide and oligosaccharide composition. It also includes a study of the seed morphology. Morphological characterization includes optical, transmission, and scanning electron microscopy. The polysaccharide exists in thickened cell walls of the cotyledonary cells, and the extracted xyloglucan is structurally quite similar to those from tamarind seed and detarium. Nevertheless there are some subtle differences in the fine structure, particularly in the oligomeric xyloglucan composition. The chain flexibility of the polysaccharide is also discussed in the light of our recent measurements reported elsewhere [Biomacromolecules2004, 5, 2384-2391].  相似文献

Afzelia xylocarpa is listed as an endangered species on the International Union for the Conservation of Nature and Natural Resources World list of Threatened Trees, due to overexploitation for its valuable timber and habitat loss. We isolated eight polymorphic microsatellite loci from A. xylocarpa using dual suppression polymerase chain reaction technique. These loci provide microsatellite markers with high polymorphism, as the number of alleles ranged from two to seven, and the estimate of gene diversity was between 0.285 and 0.795. The markers are now available for more detailed investigation of population genetic structure and gene flow among A. xylocarpa populations.  相似文献

Afzelia africana is a forest species used by local inhabitants for various purposes, especially as forage to feed cattle, as medicinal plant and its wood is used to make furniture or for cooking. Its utilisation in its current form constitutes a threat to this species. However, the lack of data on this species is a hindrance towards drawing up an efficient program for its sustainable management. In order to fill in some gaps in the knowledge of A. africana tree populations, dendrometric characteristics of this species were studied within different climatic zones where it occurs in Benin. Data collected on each of them included height and diameter, and with regard to the levels of pressure, five categories were defined namely: null, weak, moderate, severe and very severe. As far as diameter is concerned every size was taken into consideration in all the climatic zones. However, average diameter and height of the A. africana individuals varied significantly according to climatic zones. Anthropogenic pressure increased while moving from the humid zone towards the drier zone. Moreover, there was a noticeable significant change in the level and quality of pressure between trees found in the different climatic zones in the sense that the lower the height of the trees, the more severe the level of pressure exerted. Such a relationship was not significant when one considers tree diameter in accordance with the climatic zones in the country.  相似文献

Interactions between an isolate of the ectomycorrhizal fungus Pisolithus sp. and Afzelia africana Sm. seedlings were studied at the structural and ultrastructural levels. Several different conditions were tested with or without sugar and in a sterile or nonsterile medium. In the growth cabinet, the A. africana/Pisolithus sp. interactions did not produce ectomycorrhizas. A fungal sheath was formed but no Hartig net, and an unusual host epidermal cell wall was observed. Hyphae of Pisolithus sp. induced modifications of epidermal cells of 15-day-old A. africana seedlings indicative of non-mycorrhizal interactions, such as wall thickening, wall ingrowth, papillae formation, degraded host wall material and the presence of intracellular hyphae. Wall ingrowth consisted of depositions of host cell wall materials giving a positive reaction for polysaccharides; however, wall thickenings and papillae showed no homogeneous reactions for polysaccharides. In glasshouse conditions, inocula of Pisolithus sp. in the form of spores or mycelia entrapped in peat-vermiculite added to sterilized soil produced typical ectomycorrhizae only with 6-month-old A. africana seedlings. Under these conditions, no conspicuous cell wall reactions occurred on A. africana roots. The results demonstrate that the establishment of an association between an ectomycorrhizal fungus and a potential host plant is strongly influenced by seedling age and/or environmental conditions. Therefore, in vitro synthesis is not a conclusive demonstration of a symbiotic relationship.  相似文献

Basidiocarps of Pisolithus associated with indigenous ( Afzelia quanzensis Welw.) and introduced ( Pinus caribaea Mor. and Eucalyptus camaldulensis Dehnh.) hosts in the lowland forests of the Coast Province of Kenya are morphologically distinct. Genetic variability among 52 Pisolithus basidiocarps, collected beneath the various host plants, was examined based on sequence polymorphism within the internal transcribed spacer (ITS) and intergenic spacer (IGS1) of ribosomal DNA genes. Variability in ITS and IGS1 sequences indicated that the three host-associated morphotypes were genetically different. Consensus trees generated by bootstrap analysis of sequence data of Pisolithus isolates from Australia and Kenya are polyphyletic and strongly suggest that the three different morphotypes/genotypes present in Kenya represent separate biological species. In addition, our data indicate that little genetic exchange occurs in silva between these species.  相似文献

Tropical West African savannas are exposed to high climatic variability with potential impacts on tree growth, forest dynamics and ecosystem productivity. In such context, understanding the long-term ecological responses of savanna trees to changing environmental conditions is of great relevance for taking appropriate conservation actions. We conducted the first study on tree-ring analysis and quantitative wood anatomy on Afzelia africana Sm. in Burkina Faso, to investigate the life-span growth trajectories and wood anatomical adjustment to site and to climate variations. A total of 24 stem discs was collected in four protected forests along the Sudano-sahelian and the Sudanian climatic zones. Wood samples were analyzed using standard dendrochronological methods and quantitative wood anatomy. The mean annual growth rates varied from 1.002 (± 0.249) mm. year−1 in the Sudanian zone to 1.128 (± 0.436) mm. year−1 in the Sudano-sahelian zone. Analysis of growth trajectories showed high variations within sites and between climatic zones. Wood anatomical traits significantly varied between sites. Principal Component Analysis revealed strong relationships between ring width, wood density and vessel traits, with 82.81 % of the total variance explained. Vessel size significantly increased from the pith to the bark, highlighting the ontogenetic effects on xylem anatomical variations. Inverse relationships were found between vessel size and vessel density across the driest site and the wettest site, suggesting that the higher the rainfall, the taller the tree, the larger vessel size, but the lower vessel density. By contrast, more arid conditions and high evapotranspiration lead to smaller vessel sizes and higher vessel density. Such anatomical adjustments highlight the trade-offs between water conductance efficiency and hydraulic safety, and emphasize physiological responses to climate variability. These variations on the long-term dynamics and xylem anatomical patterns underline complex interactions between ontogenetic effects and contrasting environmental factors that affect the eco-physiological functioning of A. africana throughout the Sudanian region.  相似文献

I examined the fate of seeds from ten focal trees of Afzelia quanzensis (Leguminosae), a canopy tree in the Arabuko‐Sokoke, Kenya. The study was conducted for one fruiting season, between August 1990 and July 1991. Yellow baboons (Papio cynocephalus), Syke's monkeys (Cercopithecus albogularis), sun squirrels (Heliosciurus rufobrachium), and bush squirrels (Paraxerus palliatus) were all observed to interact with A. quanzensis seeds at various stages of pod development. Baboons and squirrels consumed high percentages of seeds when they were still immature, but the seeds were still unavailable to Syke's monkeys at this stage. Baboons bit open the hard green pods and squirrels gnawed through the pods to extract the immature seeds (hereafter referred to as seed predation), but monkeys were unable to open the pods. Upon maturity, the pods opened slightly, revealing red arils that were sought by baboons, monkeys, and squirrels. Monkeys removed the highest percentage of mature seeds from these pods. These mammal dispersers ate the arils from the mature seeds and discarded the viable part that germinates (hereafter referred to as seed dispersal). My data indicate that baboons and squirrels are seed predators while monkeys are seed dispersers of A. quanzensis.  相似文献

Understanding patterns of genetic diversity of plants is important in guiding conservation programs. The aim of our study was to characterize genetic diversity in Afzelia quanzensis, an economically important African tree species. We genotyped 192 individuals at 10 nuclear microsatellite loci. Samples were collected from nine sites in Zimbabwe, five in the north and four in the south, separated by a mountain range, the Kalahari-Zimbabwe axis. Overall, genetic diversity was relatively low across all sites (expected heterozygosity (H E)?=?0.452, mean number of alleles (A)?=?4.367, allelic richness (A R)?=?2.917, effective number of alleles (A E)?=?2.208, and private allelic richness (PAR)?=?0.197). Genetic diversity estimates, H E, A, A R, and PAR, were not significantly different between northern and southern sites. Allelic richness was significantly higher in southern sites. Significant population differentiation was observed among all sites (F ST ?=?0.0936, G′ ST ?=?0.1982, G ST ?=?0.1001, D JOST?=?0.0598). STRUCTURE analysis and principal components analysis identified two gene pools, one predominantly made up of southern individuals, and the other of northern individuals. A Monmonier’s function detected a genetic barrier that coincided with the Kalahari-Zimbabwe axis. The relatively low level of genetic diversity in A. quanzensis may reduce adaptability and limit future evolutionary responses. All sites should be monitored for deleterious effects of low genetic diversity, and genetic resource management should take into consideration the existence of the distinct gene pools to capture the entire extant genetic variation.  相似文献

This study was conducted to evaluate the competitiveness and effectiveness of Thelephoroid fungal sp. ORS.XM002 against native ectomycorrhizal fungal species colonizing potted Afzelia africana seedlings during 3 months of growth in different forest soils collected from under mature trees. Using morphotyping and restriction fragment length polymorphism (RFLP) analysis of the nuclear rDNA internal transcribed spacer (ITS), we were able to distinguish the introduced Thelephoroid fungal sp. ORS.XM002 among native ectomycorrhizal fungal species that form ectomycorrhizae in A. africana seedlings. The morphotype (MT) of the introduced fungus showed some color variation, with a shift from light- to dark-brown observed from younger to older mycorrhizal tips. We were able to differentiate the ITS type xm002 of the introduced fungus from the 14 ITS-RFLP types characterizing the 9 native MT that occurred in forest soils. The frequency of ITS type xm002 ranged from 40% to 49% depending on the forest soil used, and was always higher than those of ITS types from native dark-brown MT that occurred in inoculated seedlings 3 months after inoculation. We considered Thelephoroid fungal sp. ORS.XM002 to be responsible for stimulation of mycorrhizal colonization of inoculated A. africana seedlings when compared with control seedlings in forest soils. This fungus appeared to be more effective in increasing the root dry weight of A. africana seedlings. To identify the unknown introduced fungal species and native MT, we sequenced the ML5/ML6 region of the mitochondrial large subunit rRNA. Sequence analysis showed that these fungi belong to three ML5/ML6 groups closely related to the Cortinarioid, Thelephoroid, and Sclerodermataceous taxa. The molecular evidence for the persistence of Thelephoroid fungal sp. ORS.XM002 despite competition from native fungi argues in favor of using this fungus with A. africana in nursery soil conditions in Senegal.  相似文献
