The domesticated silkworm Bombyx mori is an economically important insect that produces large quantities of silk during its 5th instar larval stage. Polyamines are important regulators of growth and have been shown to affect silk production, however their role in larval development is not completely understood. L-ornithine decarboxylase (ODC), a key regulatory enzyme in the polyamine biosynthetic pathway catalyzes the conversion of ornithine to putrescine, which is further broken down to spermidine and spermine. In this study, we set out to understand the role of ODC on the growth and development of silkworm larvae. We fed 5th instar larvae with α-difluoromethylornithine (DFMO), an ODC inhibitor and studied its impact on larval silk glands. Feeding DFMO did not alter the expression of L-ODC but led to a significant reduction in putrescine and spermidine levels. Furthermore, reduced cellular levels of polyamine led to increased oxidative stress and decreased cell viability. Subsequently, this resulted in several developmental defects at the pupal and moth stages. These findings highlight the importance of ODC in the growth and development of B. mori larvae.  相似文献

The polyamine and amino acid composition of the mermithid nematode, Romanomermis culicivorax, and its host, Aedes aegypti, was determined. Putrescine, spermidine, spermine, cadaverine and two acetylated spermidine derivatives were present in parasitic juveniles, newly-emerged post-parasites, and eggs of R. culicivorax. Whole insect homogenates of fourth-instar A. aegypti contained the same array of polyamines, except that the putrescine:spermidine ratio was the inverse of that in parasitic R. culicivorax. Polyamines and amino acids were in greater concentrations in the nematode eggs than in other developmental stages investigated. Both the host and nematode possess the biosynthetic capacity for polyamine biosynthesis, as evidenced by measurable activities of ornithine decarboxylase in the host''s tissues and the nematode''s free-living stages.  相似文献

Polyamines are ubiquitous biologically active aliphatic cations that are at least transiently available in the soil from decaying organic matter. Our objectives in this study were to characterize polyamine uptake kinetics in Phytophthora sojae zoospores and to quantify endogenous polyamines in hyphae, zoospores, and soybean roots. Zoospores contained 10 times more free putrescine than spermidine, while hyphae contained only 4 times as much free putrescine as spermidine. Zoospores contained no conjugated putrescine, but conjugated spermidine was present. Hyphae contained both conjugated putrescine and spermidine at levels comparable to the hyphal free putrescine and spermidine levels. In soybean roots, cadaverine was the most abundant polyamine, but only putrescine efflux was detected. The selective efflux of putrescine suggests that the regulation of polyamine availability is part of the overall plant strategy to influence microbial growth in the rhizosphere. In zoospores, uptake experiments with [1,4-14C]putrescine and [1,4-14C]spermidine confirmed the existence of high-affinity polyamine transport for both polyamines. Putrescine uptake was reduced by high levels of exogenous spermidine, but spermidine uptake was not reduced by exogenous putrescine. These observations suggest that P. sojae zoospores express at least two high-affinity polyamine transporters, one that is spermidine specific and a second that is putrescine specific or putrescine preferential. Disruption of polyamine uptake or metabolism has major effects on a wide range of cellular activities in other organisms and has been proposed as a potential control strategy for Phytophthora. Inhibition of polyamine uptake may be a means of reducing the fitness of the zoospore along with subsequent developmental stages that precede infection.  相似文献

RNA interference (RNAi) is a useful tool for gene functional studies in non-model insects and pest insects. Establishment of experimental conditions for RNAi, which differ from insect to insect, is important for evaluating the effect of dsRNA injection of relevant genes. When injecting dsRNA into the green rice leafhopper Nephotettix cincticeps (Uhler), high mortality was observed. Therefore, the adverse effects of injection of dsRNA on leafhopper development were examined to assess the suitable conditions for RNAi in this species. Injection manipulation by using a glass capillary did not affect leafhopper survival but delayed the molt of the corresponding instar. High mortality was observed when large amounts of dsRNA were administered. This adverse effect of dsRNA was examined in 2 genes, exogenous EGFP gene and endogenous peptidoglycan recognition protein gene (NcPGRP12). Injection of a high dose (60 ng/insect) caused high mortality in all stages tested: 4th instar, 5th instar, and female adult. A relatively low dose (6 ng/insect) did not cause high mortality, retaining a high potential for gene silencing. Since RNAi is highly effective in this species, the deleterious effect of large amounts of dsRNA could be avoided by administering a low dsRNA dose.  相似文献

The three major polyamines—putrescine, spermidine, and spermine—were studied and changes of their levels were examined in extracts of cerebral ganglia and fat body from adult Acheta domesticus. In nervous tissue, only spermidine and spermine were present and spermine was two- to three-fold more abundant than spermidine. The polyamine levels were high up to day 3, decreased on day 4, and then remained relatively unchanged up to day 10. The spermidine/spermine ratios decreased during the imaginal life. Higher spermidine titres were observed in the neural tissue of egg-laying females compared to virgin females. In the fat body, putrescine was detected together with spermidine and spermine. Spermidine and spermine levels were two-fold higher than putrescine. Fat body of virgin females contained two times more polyamines than male fat body. Low at emergence, spermidine and spermine concentrations peaked on days 2–3 only in females, and egg-laying was characterized by an increase of putrescine and spermidine titres. Starvation did not change polyamine contents, implying homeostatic regulation of the intracellular polyamine metabolism. These data showing tissue specific changes in polyamine levels during the imaginal life of Acheta domesticus point to the physiological importance of polyamines as possible intracellular regulators during adult insect development. © 1993 Wiley-Liss, Inc.  相似文献

When the butterfly Eurema hecabe is infected with two different strains (wHecCI2 and wHecFem2) of the bacterial endosymbiont Wolbachia, genetic males are transformed into functional females, resulting in production of all-female broods. In an attempt to understand how and when the Wolbachia endosymbiont feminizes genetically male insects, larval insects were fed an antibiotic-containing diet beginning at different developmental stages until pupation. When the adult insects emerged, strikingly, many of them exhibited sexually intermediate traits in their wings, reproductive organs, and genitalia. The expression of intersexual phenotypes was strong in the insects treated from first instar, moderate in the insects treated from third instar, and weak in the insects treated from fourth instar. The insects treated from early larval instar grew and pupated normally but frequently failed to emerge and died in the pupal case. The dead insects in the pupal case contained lower densities of the feminizing Wolbachia endosymbiont than the successfully emerged insects, although none of them were completely cured of the symbiont infection. These results suggest the following: (i) the antibiotic treatment suppressed the population of feminizing Wolbachia endosymbionts; (ii) the suppression probably resulted in attenuated feminizing activity of the symbiont, leading to expression of intersexual host traits; (iii) many of the insects suffered pupal mortality, possibly due to either intersexual defects or Wolbachia-mediated addiction; and hence (iv) the feminizing Wolbachia endosymbiont continuously acts on the host insects during larval development for expression of female phenotypes under a male genotype. Our finding may prompt reconsideration of the notion that Wolbachia-induced reproductive manipulations are already complete before the early embryonic stage and provide insights into the mechanism underlying the symbiont-induced reversal of insect sex.  相似文献

Despite previous developmental studies on basally branching wingless insects and crustaceans, the evolutionary origin of insect wings remains controversial. Knowledge regarding genetic regulation of tissues hypothesized to have given rise to wings would help to elucidate how ancestral development changed to allow the evolution of true wings. However, genetic tools available for basally branching wingless species are limited. The firebrat Thermobia domestica is an apterygote species, phylogenetically related to winged insects. T. domestica presents a suitable morphology to investigate the origin of wings, as it forms the tergal paranotum, from which wings are hypothesized to have originated. Here we report the first successful CRISPR/Cas9-based germline genome editing in T. domestica. We provide a technological platform to understand the development of tissues hypothesized to have given rise to wings in an insect with a pre-wing evolution body plan.  相似文献

In an effort to understand the role of biogenic amines in insect development, changes in the levels of octopamine (OA), dopamine (DA), epinephrine (E), norepinephrine (NE), and serotonin (5-HT) in the brain, the optic lobes and the haemolymph of different developmental stages of Acherontia styx were analyzed using HPLC with electrochemical detector. In the brain, OA was the most abundant monoamine. DA, OA, and E levels in larvae peaked around the wandering stage (W). A dramatic increase in DA, 5-HT, and E levels was observed in the brain of the adult as compared to the pupal stage. NE, however, was not detected in the brain of most stages of the insect, except in the brain of 20-day-old pupae and adults. A 3-fold increase in OA levels was observed in the optic lobes of the adult as compared to late pupal stage. No changes were observed for E, DA, and 5-HT. NE was not detected in the optic lobes. In the haemolymph of 5th instar larvae, OA was also the most abundant amine. Both DA and OA peaked prior to the onset of the W stage. In contrast, E and NE concentrations decreased with development of the 5th instar larvae. 5-HT was not detected in the haemolymph. Finally, different profiles for amine levels were observed for the two forms of the 5th instar larvae (green vs brown). These results are interpreted in the light of the role of biogenic amines and their relation to development in the nervous system of lepidopteran insects.  相似文献

Polyamine biosynthesis and accumulation were studied during the early development of the nudibranch Phestilla sibogae. Ornithine decarboxylase activity increases over 40-fold in the first 4 days of embryogenesis, with the maximum (400 pmole of 14CO2/30 min/mg of protein) occurring between Days 3 and 4, the time of most rapid growth. Putrescine-stimulated S-adenosyl-l-methionine decarboxylase activity is not detectable until the second day of development and attains maximal activity (100 pmole of 14CO2/30 min/mg of protein) at Day 4. The pattern of spermidine-stimulated S-adenosyl-l-methionine decarboxylase activity is similar. Putrescine and spermidine double in concentration between Days 0 and 6. Adults contain fairly high levels of putrescine and spermidine, similar to prokaryotes. The increase in the polyamine biosynthetic enzymes and the accumulation patterns of the polyamines correlate well with data from other early developmental systems and support the hypothesis of key roles for the polyamine pathway in the control of growth processes.  相似文献

Naturally occurring levels of putrescine, spermidine, and spermine were analyzed in female gametophyte (nonfertilized) and reproductive tissues (cystocarps) at two different stages of development in the marine red algae Gracilaria cornea J. Agardh. Endogenous polyamine levels changed at differential stages of cystocarp maturation. Highest polyamine values were found on tissue from the early post‐fertilization stage, decreasing as the cystocarp matured. Incubation experiments revealed that exogenous polyamines induced cystocarp maturation and promoted carpospore liberation, developing cell masses within 4 to 7 days in treatments with spermine. This is the first report on the effect of polyamines on cystocarp maturation in marine algae.  相似文献

Polyamines were extracted from five different leishmanial strains (Leishmania sp., L. tropica major, L. mexicana, and two L. donovani isolates) and identified as pulrescine, spermidine, and spermine by thin-layer chromatography and mass spectrometry. These sensitive methods were also used to demonstrate the conversion of radioactive putrescine into spermidine and spermine. As in other types of cells, polyamine levels fluctuated during the growth cycle, maximal levels being attained during the logarithmic growth phase. In the five leishmanial strains, which were members of four different serotypes, the spermidine—putrescine ratios also varied, and in two strains of the same serotype, polyamine ratios were practically identical, suggesting that polyamine characteristics might serve as a further criterion for strain identification and classification.  相似文献

Effects of hexane extract of Lantana camara leaves were investigated on reproductive bioactivities of Dysdercus koenigii by assessing mating behavior, oviposition behavior and fertility of the adults emerged from treated fifth instar nymphs. Leaves of L. camara were extracted in hexane by ‘cold extraction method’. The fifth instar nymphs were treated with the extract by ‘dry film residual method’ for 24 h and the adults emerged from these nymphs were used for the experiments. The results indicated that treated males showed altered courtship behaviour, lesser number of mounting attempts and took relatively more time to mate than the normal males. The treated females, very often, exhibited mating refusal and non-receptive behaviour towards the courting males. This led to decrease in percent successful mating. Also, the mating in the treated insects frequently got disrupted and terminated prior to insemination. The treatment of the females with the extract resulted in alteration of their oviposition behaviour. Consequently, the treated females laid lesser number of egg batches and total number of eggs in their life span. Although the eggs laid by the treated females were fertile, the percent hatchability was lesser than normal. The results signify that hexane leaf extract of L. camara possesses phytochemicals, which adversely impaired the reproductive bioactivities of D. koenigii. Therefore, some of these compounds individually or synergistically can be employed in integrated pest management of D. koenigii by hampering its reproductive potential.  相似文献

Red cotton bug Dysdercus koenigii F. (Hemiptera: Pyrrhocoridae), is found destructive pest in various cotton growing areas. Under natural conditions insects are highly subjected to thermal stresses. In present work the developmental duration and survival rate of all immature stages, adult longevity and reproduction of D. koenigii by exposed to rapid changes in very low temperatures were studied. When 3 h short-stress of low temperatures (12–0 °C) was given to different stages of D. koenigii, the results revealed that survival rate of all stages were significantly reduced. Survival rate of female was significantly higher than male after exposed to cold temperature stress. Mating percentage, fecundity and hatching percentage were decreased significantly with the decrease of short-term cold temperature stress. Based on these results, we concluded that the developmental duration, survival rate and reproduction of D. koenigii significantly affected when they exposed to short term cold temperature stress.  相似文献

Modulation of differential gene expression and change of polyamine content by salt stress are analyzed for the first time in a well-known medicinal plant, Panax ginseng C.A. Meyer. Three ginseng genes (PgSPD, PgSAMDC, and PgADC) involved in polyamine biosynthesis showed differential up-regulation patterns after 1 and 7 days of salt treatments. The modulation of gene expression resulted in the elevation of total polyamine content with relatively high levels of spermidine and spermine, while putrescine level diminished depending on the salt concentration. Conversely, salt stress led to a significant increase in diamine oxidase and subsequent decline in polyamine oxidase. The proline content caused by salinity follows a similar pattern as the total polyamine content and exogenous spermidine also resulted in the alleviation of proline content under salinity. Further, polyamine biosynthesis inhibitors, such as cyclohexylamine and methylglyoxal bis-(guanylhydrazone) mediated down-regulation of PgSPD and PgSAMDC, and affected cellular polyamine levels. Thus, polyamines may enhance the ginseng plant tolerance in response to the salt stress by increasing the levels of endogenous polyamines.  相似文献

Glucose-regulated protein 78 (GRP78) is a member of the HSP70 family of proteins and is localized in the endoplasmic reticulum (ER) within cells. GRP78 and its gene has been identified in only a few species of insects, and its role is not clear. Here, we identified full-length grp78 cDNA from the Indian meal moth, Plodia interpunctella, and demonstrated the role of grp78 in developmental and physiological processes of the insect. The deduced amino acid sequence of GRP78 contained highly conserved functional motifs of the HSP70 family and the C-terminal motif of KDEL, which is characteristic of ER-localized HSP70. It also showed high identity (93–94%) with GRP78 and related HSP70 proteins of lepidopteran species. Gene expression analysis showed that grp78 mRNA levels were high in the egg, feeding larval, and adult stages, but low in the molting, wandering larval, and pupal stages of development. In a tissue comparison of fifth instar P. interpunctella, grp78 level was higher in the gut than in the integument or fat bodies. Grp78 level decreased greatly when fifth-instar larvae were starved for 48 h, but recovered within 3–6 h after re-feeding. Our data suggest that grp78 is highly associated with dietary energy conditions during development and may play an important role in the nutritional physiology of insects.  相似文献

The signal to induce diapause in H. virescens comes early in development (prior to the third instar in most insects), but the signal to break diapause can come shortly after entrance into diapause at pupation. Haemolymph ecdysteroid titres in both diapause-bound and non-diapause-bound Heliothis virescens larvae were similar in the first two thirds of the last-larval instar, when similar changes in morphology and behaviour occurred. However, the number of stepwise increases in titre and the timing of the steps was different in the two groups of larvae. Haemolymph ecdysteroid titres in the last third of the instar were approx, five times higher in non-diapause than in diapause-bound larvae. In diapausing pupae, haemolymph ecdysteroid titres dropped to levels found in larvae which had completed two thirds of the last instar. When diapausing pupae were warmed to break diapause, haemolymph ecdysteroid titres rose again. However, 2 of the 4 high ecdysteroid levels detected in pupae developing after diapause break were considerably lower than those detected for non-diapause pupae.  相似文献

Many biological processes result from the coupling of metabolic pathways. Considering this, proliferation depends on adequate iron and polyamines, and although iron-depletion impairs proliferation, the metabolic link between iron and polyamine metabolism has never been thoroughly investigated. This is important to decipher, as many disease states demonstrate co-dysregulation of iron and polyamine metabolism. Herein, for the first time, we demonstrate that cellular iron levels robustly regulate 13 polyamine pathway proteins. Seven of these were regulated in a conserved manner by iron-depletion across different cell-types, with four proteins being down-regulated (i.e., acireductone dioxygenase 1 [ADI1], methionine adenosyltransferase 2α [MAT2α], Antizyme and polyamine oxidase [PAOX]) and three proteins being up-regulated (i.e., S-adenosyl methionine decarboxylase [AMD1], Antizyme inhibitor 1 [AZIN1] and spermidine/spermine-N1-acetyltransferase 1 [SAT1]). Depletion of iron also markedly decreased polyamine pools (i.e., spermidine and/or spermine, but not putrescine). Accordingly, iron-depletion also decreased S-adenosylmethionine that is essential for spermidine/spermine biosynthesis. Iron-depletion additionally reduced 3H-spermidine uptake in direct agreement with the lowered levels of the polyamine importer, SLC22A16. Regarding mechanism, the “reprogramming” of polyamine metabolism by iron-depletion is consistent with the down-regulation of ADI1 and MAT2α, and the up-regulation of SAT1. Moreover, changes in ADI1 (biosynthetic) and SAT1 (catabolic) partially depended on the iron-regulated changes in c-Myc and/or p53. The ability of iron chelators to inhibit proliferation was rescuable by putrescine and spermidine, and under some conditions by spermine. Collectively, iron and polyamine metabolism are intimately coupled, which has significant ramifications for understanding the integrated role of iron and polyamine metabolism in proliferation.  相似文献

The main objective of this study was to identify potential forensic indicators in the insect fauna associated with pig carrion and the pattern of insect succession during the decomposition process in two environments of a rural area in Uberlândia, State of Minas Gerais, Brazil. The study was conducted at two locations: in a pasture and in a fragment of a semi-deciduous forest (vegetation profile of the Cerrado biome) in two different seasons (rainy and dry) of 2010. The decomposition process was more rapid in the rainy season. More than 32,000 insects belonging to 17 species of 6 families of Diptera and 2 species of Coleoptera bred in the carcasses. The majority of Diptera bred in the first three stages of decomposition. However, Phoridae and Coleoptera bred mainly in the last two stages. The insects bred more abundantly in the pasture and in the humid season. The exceptions were the Fanniidae (Diptera), which bred more abundantly in the forest and the Dermestidae and Cleridae (Coleoptera), which did not demonstrate any preference in terms of environments and were more abundant in the dry season, respectively. Species such as Chrysomya albiceps (Wiedemann), Peckia (Patonella) intermutans (Walker), Necrobia rufipes (De Geer), and Dermestes maculatus (De Geer) may be potential indicators of post-mortem interval. Hemilucilia segmentaria (Fabricius) and Ophyra aenescens (Wiedemann) may be indicators of localization of the natural environment, while Musca domestica Linnaeus may be an indicator of the anthropic environment. The study thus presented many species of potential forensic indicators in rural areas of this region.  相似文献
