The effect of the developing flowers and fruitlets on leaf expansionand composition has been determined in leafy inflorescencesof Citrus sinensis L. Osbeck. During leaf expansion the developingflowers do not compete with the leaves, and their early removaldoes not affect leaf size and composition. Competition for mineralelements is established after flower opening, once leaf expansionis complete. No effect of the fruit on metabolizable carbohydratesin the leaves was found up to day 22 after flower opening. The response to exogenously applied growth regulators suggeststhat the differences in weight and composition between inflorescenceleaves and leaves from vegetative sprouts may be due to hormonally-mediateddifferences in sink strength. Gibberellic acid enhances laminaexpansion and increases the sink strength of the leaves. Kinetinenhances lamina expansion without affecting sink strength. Leafgrowth is directly related to both soluble and wall-bound acidinvertase activities; however, the GA3 effects on leaf growthand sink strength are unrelated to invertase activity and tothe rate of starch accumulation. Carbohydrate accumulation in young, fully expanded leaves ofCitrus sinensis L. Osbeck is affected by the presence of thefruitlet on the shoot. Previous work gave evidence that gibberellinsmay be involved in this 'fruit effect'. In the present workwe have studied the effect of gibberellic acid (GA3) on 14C-sucroseuptake by leaf discs and whether its action could be due toa modulation of the plasma membrane ATPase, which maintainsthe H+ gradient that drives H+/sucrose co-transport. The effect of GA3 on 14C-sucrose uptake depended on the osmolarityof the assay medium. At 300 mOsm a reduction in the uptake ratewas observed. The inhibitory effect of the hormone disappearedafter preincubating the leaf discs with para-chloromercuri-phenylsulphonicacid (PCMPS), a sulphydril binding inhibitor. ATPase activityof isolated plasma membrane vesicles was inhibited by IAA treatments,while GA3 or ABA did not affect this enzyme, even after a 3h preincubation period. However, in the absence of a surfactantin the assay medium, GA3, together with turgor pressure, modulatedplasma membrane ATPase activity, possibly through modificationsof membrane permeability. Growth rates of seasonal leaf flushes of ‘Valencia’orange [Citrus sinensis (L.) Osbeck] were measured and waterrelations characteristics of young (new) and over-wintered (old)citrus leaves were compared. New flush leaves had lower specificleaf weights and lower midday leaf water potentials than comparablyexposed old leaves. Spring and summer flush new leaves had higherosmotic potentials than old leaves. These differences becamenon-significant as the new leaves matured. During summer conditions,water-stressed new leaves reached zero turgor and stomatal conductancealso began to decrease in them at higher leaf water potentialsthan in old leaves. Old leaves were capable of maintaining openstomata at lower leaf water potentials. Opened flowers and newflush leaves lost more water, on a dry weight basis, than flowerbuds, fruit or mature leaves. The results illustrate differencesin leaf water potential and stomatal conductance which can beattributed to the maintenance of leaf turgor by decreases inleaf osmotic potentials as leaves mature. These changes in citrusleaf water relations are especially important since water stressresulting from high water loss rates of new tissues could reduceflowering and fruit set. Citrus sinensis (L.) Flowering and vegetative shoots of ‘Shamouti’ orange[Citrus sinensis (L.) Osbeck] and ‘Marsh’ seedlessgrapefruit (Citrus paradisi Macf.) were examined for correlationof their morphology and anatomy with fruit set. Fruit set isfavoured on leafy inflorescences whereas abortion is nearlycomplete on leafless inflorescences. Leafless inflorescencesof ‘Shamouti’ with one flower were found to havea very thin stem which contained few vascular bundles, whereasthose with three flowers had better-developed vascular systems.The vascular system of leafy inflorescences is significantlydifferent from that of leafless ones and contains a distinctcentral xylem cylinder. The vascular area of leafless inflorescencesis only about one-quarter of that of the leafy ones. The vascularsystem of grapefruit resembles that of the ‘Shamouti’orange. This study emphasizes the importance of the dimensionof the vascular system for fruit set and provides a possibleexplanation for the better fruit set on both leafy and leaflessinflorescences with several flowers compared with single-floweredinflorescences. Yamashita, T. 1987. Modulated degradation of ribulose ftisphosphatecarboxylase in leaves on top-pruned shoots of the mulberry tree(Morus alba L.).—J. exp. Bot. 38: 1957–1964. The effects of pruning shoot tops on the synthesis and degradationof ribulose 1,5–Wsphosphate carboxylase (RuBPCase) inleaves on remaining shoots were investigated in mulberry trees.Leucine labelled with 14C was fed to leaf discs from field-grownmulberry trees and 14C incorporation into RuBPCase was examined.Proportion of 14C in RuBPCase to leucine–14C absorbedby leaf discs was remarkably lowered by top-pruning, thoughoccasionally a slight increase was observed soon after pruning.Yet RuBPCase content in leaves on top-pruned shoots became progressivelyhigher than that in leaves on intact shoots. Changes in 14Cin Ru1BPCase in leaves of mulberry saplings previously fed 14CO2were followed. Following 14CO2 feeding, the attainment of themaximal level of 14C in RuBPCase was retarded by top-pruning.The highest level of 14C in RuBPCase was maintained in leaveson top-pruned shoots but decreased in leaves on intact shoots.Specific radioactivity in RuBPCase continued to increase inleaves on top-pruned shoots even after attaining a maximum levelin the control leaves. These facts suggest that the increasein RuBPCase by top-pruning results from a cessation of its degradationfor the remobilization of nitrogen for newly developing leaveson shoot tops. CO2 uptake and diffusion conductance of Valencia orange fruits(Citrus sinensis L. Osbeck) were measured in the field duringthe growing season of 1977/78 to ascertain if, as in the leaf,stomata control photosynthesis and transpiration under changingenvironmental conditions. Measurements were made on 15 yearold trees grown in a sandy loam soil and receiving either adry or a wet treatment. Fruit diffusive conductance was measuredwith a modified water vapour diffusion conductance meter andgross photosynthesis was measured with a 14CO2 uptake meter.Photosynthetically active radiation (PAR) was measured witha quantum sensor. Fruits exposed to light assimilated CO2 ata rate which was 25–50% of that assimilated by leaves.The uptake was dependent on fruit size, PAR, chlorophyll content,and on diffusive conductance of the fruit epidermis. Epidermalconductance showed a diurnal trend which was similar in shapeto that of the leaf except in the late afternoon. Cuticularconductance of the fruit was calculated and ranged between 0.22and 0.30 mm s–1. It was speculated that the CO2 uptakeby the fruit could support the growth of flavedo cell layerswhen exposed to light. Dry soil caused an increase in the 14CO2uptake by fruit possibly caused by the increased potential areaof the stomatal opening per unit of fruit surface area.  相似文献

Carbohydrate Availability in Relation to Fruitlet Abscission in Citrus   总被引:2,自引:1,他引:1  
Abscission of flowers and fruitlets in the Washington navelorange (Citrus sinensis [L.] Osbeck) has been characterizedin relation to carbohydrate availability. A main wave of flowerabscission occurs shortly after anthesis while the carbohydratereserves in the tree are high. Fruitlet abscission starts approx.30 d after the commencement of flowering, while carbohydrates(mainly starch) are being accumulated in the leaves. Flowerand early fruitlet abscission are not caused by carbohydrateshortage. During late fruitlet abscission sucrose concentrationin the leaves falls to a low value demonstrating a limitationin supply and competition among the developing fruitlets forcarbohydrates. Concentrations of sucrose and reducing sugarsin the peel of the fruitlets also fall to low values, and arelationship could be demonstrated between these free sugarlevels and abscission. Ringing increases carbohydrate supplyto fruit and reduces late fruitlet abscission, but only hasa marginal effect on the growth of the fruitlets, which seemsless sensitive than abscission to carbohydrate shortage. Thelimitation of carbohydrate supply to the fruitlets occurs whilestarch levels in the leaves remain high. Slow mobilization ofstarch reserves may be one factor limiting set in Citrus. Activities of Carboxylation Enzymes in Freshwater Macrophytes   总被引:2,自引:0,他引:2  
Fifteen species of freshwater macrophytes, mainly from cool,temperate waters, were assayed for ribulose bisphosphate carboxylase-oxygenase(RuBPCase) and phosphoenolpyruvate carboxylase (PEPCase) activities.In extracts from all the species RuBPCase was the most activecarboxylation enzyme, and the RuBPCase/PEPCase ratio was atleast 2·0, even for the submersed species Isoetes lacustrisL. and Littorella unifiora (L.) Aschers. which have been reportedto show Crassulacean Acid Metabolism (CAM) activity. The PEPCaseactivity in I.lacustris was lower than that found in some non-CAM-likespecies. In this respect, I.lacustris and L unifiora differfrom most terrestrial CAM plants. However, these two species,along with Potamogeton praelongus Wulf. and Juncus bulbosusvar.fluitans L., had the lowest RuBPCASE/PEPCase ratios, lowerthan found in terrestrial C3 species; suggesting that the potentialfor substantial photosynthetic metabolism of C4 acids existsin some temperate, submersed plants. In the three amphibiousspecies (Potamogeton polygonifolius Pourr., Mentha aquaticaL., and Hippuris vulgaris L.) examined, the aerial leaves exhibitedhigher RuBPCase activities than the submersed leaves. Embryoids obtained from subcultured ovular callus of the Shamoutiorange (Citrus sinensis) were grown on Murashige and Tuckernutrient medium (BM) in the presence and absence of gibberellicacid (GA2), adenine sulphate (ADS) and combinations of these.Both GA2 and ADS stimulated significantly the rooting of smallembryoids with unorganized tissue, and larger embryoids withpartially developed root zones. Only large embryoids with fullydeveloped root zones rooted well on BM but better on BM+GA2+ADS.Rooting was best on media containing 1 to 5 mg per 1 ADS, andthese two combined. The optimum sucrose concentrations were5 to 6 per cent. Shoot formation on embryoids prior to rootingcompletely inhibited rooting which is otherwise enhanced byGA2+ADS. Filter-sterilized and autoclaved GA2, showed similaractivity in rooting.  相似文献

Changes in structure, size and number of oil glands locatedin the fruit rind were assessed in developing fruit of the WashingtonNavel orange (Citrus sinensis L. Osbeck) from pre-anthesis tofruit maturity. Initiation of oil glands was found to be restrictedto early fruit development. Glands continued to develop throughoutfruit growth, until all reached maturity by a fruit size of30 to 50 mm diameter. Mature glands continued to enlarge withfruit growth. Mature fruit had between 8 000 and 12 000 oilglands. Anatomical studies of the fruit rind were carried outusing light microscopy on samples prepared by different tissueprocessing methods. Glands were found to develop from a clusterof cells adjacent to the fruit epidermis, into a structure consistingof a central cavity surrounded by several layers of epithelialcells. All glands were joined to the fruit epidermis, irrespectiveof their stage of development. Neither lignin nor suberin waspresent in the gland. Gland cavity formation appeared to involveschizogeny. The activity of the enzyme ribulose bisphosphate carboxylase(RuBPCase) was estimated after rapidly extracting it from intactwheat leaves pretreated under different light and CO2 levels.No HCO3 was added to the extraction buffer since it isshown to inhibit RuBPCase. The activity increased as light intensityor CO2 concentration during pretreatment was increased. Enzymeactivity increased as temperature during pretreatment was decreased.Light activation did not affect the affinity of RuBPCase forCO2. A Km of 30 µM CO2 under air level O2 was determined.CO2, light and temperature are three main limiting factors ofphotosynthesis. It seems that the activity of RuBPCase is regulatedby these factors according to the requirements for CO2 fixation.  相似文献

The effect of tetraploidy on leaf characteristics and net gasexchange was studied in diploid (2x ) and autotetraploid (4x) ‘Valencia’ sweet orange (Citrus sinensis (L.)Osb.) and ‘Femminello’ lemon (Citrus limon (L.)Burm. f.) leaves. Comparisons between ploidy levels were madeunder high irradiance (I) in a growth chamber or low total Iin a glasshouse. Tetraploids of both species had thicker leaves,larger mesophyll cell volume and lower light transmittance thandiploids regardless of growth I. Mesophyll surface area perunit leaf area of 2x leaves was 5–15% greater than on4x leaves. Leaf thickness and mesophyll cell volume were greaterin high I leaves than low I leaves. In high I, average leafarea was similar for 2x and 4x leaves, whereas in low I it was30% greater in 4x than in 2x leaves. Nitrogen and chlorophyllconcentration per cell increased with ploidy level in both growthconditions. The ratio of chlorophyll a:b was 25% greater in2x than in 4x leaves. When net CO2assimilation rate (ACO2) wasbased on leaf area, 4x orange leaves had 24–35% lowerACO2than their diploids. There were no significant differencesin ACO2between 2x and 4x orange or lemon leaves when expressedon a per cell basis. Overall, lower ACO2per unit leaf area oftetraploids was related to increase in leaf thickness, largermesophyll cell volume, the decrease in mesophyll area exposedto internal air spaces, and the lower ratio between cell surfaceto cell volume. Such changes probably increased the resistanceto CO2diffusion to the site of carboyxlation in the chloroplasts. The solanifolia mutant (sf/sf) of tomato (Lycopersicon esculentum)produces leaves consisting of leaflets with entire margins,unlike the lobed leaflets of normal plants. Normal plants treatedwith gibberellic acid (GA3) produced leaves with entire marginswhereas mutant plants exposed to 2-chloroethyl-trimethyl ammoniumchloride (CCC)—an inhibitor of gibberellin biosynthesis—producedlobing of leaflets. The leaf area of the mutant was significantlygreater than that of the normal, but was not significantly differentfrom GA3-treated normal leaves. Similarly, in CCC-treated mutantleaves the leaf area was not different from that of normal untreatedleaves. These observations suggest that the sf/sf mutation affectsthe leaf shape through its effect on endogenous gibberellinsand/or inhibitory substances. The application of GA3in aqueous solution to leaves or flowersof hermaphrodite cultivars of strawberry, Redgauntlet and Rabunda,prevented growth of the receptacle despite hand pollination.This inhibitory effect occurred only when GA3 was applied priorto anthesis. Although viable pollen was produced and germinatedto grow down the styles of treated plants, no seeds were formed.Receptacle growth failed underneath the unfertilized carpels,but the basal region devoid of carpels enlarged and ripened.The effect of GA3 was the same in vivo and for flowers grownin vitro. ABA and BAP also inhibited growth of pollinated flowersin vitro, but neither substance stimulated growth of the baseof the receptacle. 2-NOA stimulated receptacle growth of pollinatedflowers but did not overcome the inhibitory effect of GA3. Removal of fertile carpels 9 days after pollination preventedfurther receptacle growth. GA3 treatment of the bare receptaclere-started growth but was less effective than 2-NOA. No growth substance treatment induced parthenocarpic developmentin these cultivars when unopened buds were emasculated and culturedwithout pollination, although GA3 induced some swelling of thereceptacle base. Photosynthetic CO2 assimilation, transpiration, ribulose-1,5-bisphosphate carboxylase (RuBPCase), and soluble protein were reduced in leaves of water-deficit (stress) `Valencia' orange (Citrus sinensis [L.] Osbeck). Maximum photosynthetic CO2 assimilation and transpiration, which occurred before midday for both control and stressed plants, was 58 and 40%, respectively, for the stress (−2.0 megapascals leaf water potential) as compared to the control (−0.6 megapascals leaf water potential). As water deficit became more severe in the afternoon, with water potential of −3.1 megapascals for the stressed leaves vs. −1.1 megapascals for control leaves, stressed-leaf transpiration declined and photosynthetic CO2 assimilation rapidly dropped to zero. Water deficit decreased both activation and total activity of RuBPCase. Activation of the enzyme was about 62% (of fully activated enzyme in vitro) for the stress, compared to 80% for the control. Water deficit reduced RuBPCase initial activity by 40% and HCO3/Mg2+-saturated activity by 22%. However, RuBPCase for both stressed and control leaves were similar in Kcat (25 moles CO2 per mole enzyme per second) and Km for CO2 (18.9 micromolar). Concentrations of RuBPCase and soluble protein of stressed leaves averaged 80 and 85%, respectively, of control leaves. Thus, reductions in activation and concentration of RuBPCase in Valencia orange leaves contributed to reductions in enzyme activities during water-deficit periods. Declines in leaf photosynthesis, soluble protein, and RuBPCase activation and concentration due to water deficit were, however, recoverable at 5 days after rewatering.  相似文献

Two Phaseolus vulgaris L. cultivars were grown at 20/15, 25/20,and 30/25 °C day/night temperatures in growth chambers witha 16 h thermoperiod corresponding to the photoperiod. When thefirst trifoliolate leaf was fully expanded rates of CO2 exchange(CER) were measured at 27 °C and saturating light usinginfrared gas analysis. Stomatal (rs) and mesophyll resistances,CO2 compensation points, activities of the enzymes ribulosebisphosphate carboxylase (RuBPCase), glycolate oxidase (GAO),malate dehydrogenase (MDH), and fructose-1, 6 diphosphate (FDP),chlorophyll content, Hill activities, and leaf anatomy at boththe light and electron microscope level were also investigatedin these leaves. Rates of CO2 exchange in the light, transpiration rate, andchlorophyll content increased with increasing growth temperaturewhile leaf thickness, specific leaf weight, RuBPCase activity,compensation point, and stomatal resistance decreased. Mesophyllresistance also decreased when calculated assuming zero chloroplastCO2 concentration (rm, o), but not when calculated assuminga chloroplast CO2 concentration equal to the CO2 compensationconcentration (rm, g). Average leaf size was maximal in 25/20°C plants while dark respiration, MDH activity, stomataldensity, and starch were minimal. Associations between photosynthesis, the activation state ofRuBP carboxylase and the contents of photosynthetic intermediateswere compared in soybean and wheat leaves before and after exposureto photoinhibitory treatments in the presence of atmosphericO2. Exposing attached leaves to a supra-saturating irradiance(3 800 µmol quanta m– 2 s–1) for 2 h in CO2-freeair decreased carboxylation efficiency and the light-saturatedphotosynthetic rate in air by approximately 50%. Exposure tothe photoinhibitory treatment for periods in excess of 2 h didnot cause a further decrease of photosynthesis in soybean. Althoughphotosynthesis was reduced, the initial and total (fully-activated)activities of ribulose 1,5-bisphosphate carboxylase (RuBPCase)in leaf extracts were unaltered in each species by the photoinhibitorytreatment. This was true for leaves sampled under both air andat a rate-limiting intercellular CO2 partial pressure (Ci) of75 µPa Pa–1. The contents of ribulose l,5-bisphosphate(RuBP) and 3-phosphoglyceric acid (3-PGA) were reduced by thephotoinhibitory treatment in soybean leaves sampled in air andat a rate-limiting Ci, although the RuBP/3-PGA ratio was unaffected.The relative reduction of RuBP content in soybean leaves atrate-limiting C1 was similar to the corresponding reductionof carboxylation efficiency. For wheat,the relative reductionof RuBP content at rate-limiting Ci (–19%) caused by thephotoinhibitory treatment was considerably less than the correspondingdecrease of carboxylation efficiency (–49%).The RuBP/3-PGAratio of wheat was also increased significantly by the photoinhibitorytreatment The significance of these observations to the regulationof CO2-limited photosynthesis in leaves experiencing photoinhibitionunder atmospheric oxygen is discussed. Consideration is alsogiven to the previous contention that contemporary measurementsof initial activity in crude extracts may provide a spuriousindication of the amount of the enzyme-CO2-Mg2 + form of RuBPcarboxylase present in the leaf. Antioxidative systems and the contents of pigments, malondialdehyde,soluble protein, and carbohydrate were investigated in sun-and shade-acclimated leaves of sour orange (Citrus aurantium)trees that had been grown for 7.5 years under ambient and elevated(+300 µmol mol–1) atmospheric CO2 concentrations.Sunacclimated leaves contained higher ascorbate, glutathioneand soluble carbohydrate contents and higher catalase activitiesthan shade-acclimated leaves. The activities of superoxide dismutases,which belonged to the family of Cu/Zn-isozymes, were similarin sunand shade-acclimated leaves and decreased in responseto enhanced CO2. In shade-acclimated leaves, none of the otherparameters studied was affected by elevated CO2. In sun-acclimatedleaves elevated CO2 caused increases in carbohydrate and ascorbatecontents. There was no evidence for enhanced lipid peroxidationas assessed from the determination of the malondialdehyde contentsunder either conditions. Effects of external nitrogen supply on quantity and activityof ribulose 1,5-bisphosphate carboxylase (RuBPCase) in mulberryleaves (Morus alba L. cv. Shin-ichinose) were examined. PhotosyntheticCO2 fixation and the contents of chlorophyll and phosphoruswere increased by the increased N supply. RuBPCase-protein concentrationand its proportion to total soluble-protein or to total-N rosecorrespondingly as the N supply increased. Normal, terminal inflorescences of Cordyline terminalis, a woodymonocotyledon, appeared 4–6 weeks after apical buds weretreated with gibberellin A3 (GA3) or GA4+7. There was no responseto GA13. Large plants, newly rooted cuttings, and seedlingsall responded, although there were clonal differences. Floweringwas induced under natural day lengths throughout the year. Untreatedcontrol plants in all experiments remained vegetative. Dracaenaspp. did not respond to the three gibberellins.  相似文献
