Allegheny woodrats (Neotoma magister) exist as groups of metapopulations due to their dependence on naturally disjunct rocky outcrops in the eastern United States. Severe demographic declines of Allegheny woodrats have occurred in many parts of the range due to a myriad of interacting processes, therefore identifying factors that help maintain the integrity of metapopulations is needed to guide conservation efforts. One factor considered critical to metapopulation persistence is maintaining functional connectivity. Therefore, our objective was to identify landscape factors that influence gene flow in Allegheny woodrats. We sampled 159 individuals from throughout the distribution in Virginia, genotyped them at 22 microsatellite loci, and modeled effects of land cover, roads, large rivers, and elevation on gene flow. The model that parameterized areas?≤?750 m in elevation as a barrier to gene flow performed best among the tested models, which indicates that functional connectivity is highest in high elevation habitats. We found little evidence for anthropogenic factors influencing gene flow, but continued study across extant metapopulations is needed in more peripheral areas to evaluate if anthropogenic barriers impact functional connectivity. Overall, our study emphasizes the importance of considering elevation for maintaining functional connectivity of Allegheny woodrats in the face of ongoing demographic declines in the eastern United States.  相似文献

Disruptions in metapopulation connectivity due to demographic pressure can leave genetically isolated subpopulations susceptible to genetic drift, accumulation of deleterious alleles, and inbreeding depression. Such a scenario may be playing out within Allegheny woodrat (Neotoma magister) metapopulations as a series of synergistic extrinsic pressures have contributed to the rangewide decline of the species over the last 40 years. Our goal was to elucidate the effects of demographic collapse on metapopulation function by using 11 microsatellites markers to quantify differences in patterns of connectivity and genetic diversity between a demographically stable metapopulation and one in severe demographic decline. The demographically diminished metapopulation had lower levels of genetic diversity than the stable metapopulation at all levels evaluated (metapopulation-, subpopulation-, and individual-scales). In contrast to patterns of connectivity observed within the stable metapopulation, peripheral subpopulations in the diminished metapopulation had become completely isolated and were drifting toward genetic fixation, likely as a result of the extirpation of stepping-stone subpopulations. The declining genetic parameters observed within these isolated peripheral subpopulations suggest that inbreeding depression may be contributing significantly to their demographic decline. Allegheny woodrats readily express the genetic consequences of metapopulation decline due to the low effective population sizes of subpopulations and the species’ limited dispersal capacity. Differences in genetic parameters observed between demographically stable and diminished Allegheny woodrat metapopulations emphasize the risks posed to metapopulation function and associated genetic processes introduced with demographic decline.  相似文献

The rising demand for bioethanol, the most common alternative to petroleum-derived fuel used worldwide, has encouraged a feedstock shift to non-food crops to reduce the competition for resources between food and energy production. Sweet sorghum has become one of the most promising non-food energy crops because of its high output and strong adaptive ability. However, the means by which sweet sorghum stalks can be cost-effectively utilized for ethanol fermentation in large-scale industrial production and commercialization remains unclear. In this study, we identified a novel Saccharomyces cerevisiae strain, TSH1, from the soil in which sweet sorghum stalks were stored. This strain exhibited excellent ethanol fermentative capacity and ability to withstand stressful solid-state fermentation conditions. Furthermore, we gradually scaled up from a 500-mL flask to a 127-m3 rotary-drum fermenter and eventually constructed a 550-m3 rotary-drum fermentation system to establish an efficient industrial fermentation platform based on TSH1. The batch fermentations were completed in less than 20 hours, with up to 96 tons of crushed sweet sorghum stalks in the 550-m3 fermenter reaching 88% of relative theoretical ethanol yield (RTEY). These results collectively demonstrate that ethanol solid-state fermentation technology can be a highly efficient and low-cost solution for utilizing sweet sorghum, providing a feasible and economical means of developing non-food bioethanol.  相似文献

Herbivores can act as dispersal vectors by purposely or accidentally ingesting ripe fruits, and thus endozoochory is one determining factor for plant distribution patterns. The objective of our study was to investigate to what extent plants of major taxonomic groups of the Knersvlakte (Succulent Karoo, South Africa) are endozoochorously dispersed. On three different farms in the central Knersvlakte, dung of domestic herbivores was collected and analysed by the seedling-emergence method. The resulting species composition was compared to the standing vegetation of thirty-four 1000-m2 plots each recorded on one hundred 400-cm2 subplots. Our results show that domestic livestock facilitated the dispersal of taxa characteristic of the Knersvlakte, in particular Aizoaceae. Among the taxa of this family, the local endemic dwarf shrub Drosanthemum schoenlandianum emerged with the highest frequency in dung (14.5% of all seedlings). For the Asteraceae, which are frequent in the standing vegetation of the Knersvlakte, however, endozoochorous dispersal by livestock was only of minor importance. Conservation planning should consider these dispersal patterns on behalf of future population dynamics. The complete exclusion of livestock might change current processes and thus alter vegetation patterns.  相似文献

The drivers of plant richness at fine spatial scales in steppe ecosystems are still not sufficiently understood. Our main research questions were: (i) How rich in plant species are the natural steppes of Southern Siberia compared to natural and semi-natural grasslands in other regions of the Palaearctic? (ii) What are the main environmental drivers of the diversity patterns in these steppes? (iii) What are the diversity–environment relationships and do they vary between spatial scales and among different taxonomic groups? We sampled the steppe vegetation (vascular plants, bryophytes and lichens) in Khakassia (Russia) with 39 nested-plot series (0.0001–100-m2 plot size) and 54 additional 10-m2 quadrats across the regional range of steppe types and measured various environmental variables. We measured β-diversity using z-values of power-law species–area relationships. GLM analyses were performed to assess the importance of environmental variables as predictors of species richness and z-value. Khakassian steppes showed both high α- and β-diversity. We found significant scale dependence for the z-values, which had their highest values at small spatial scales and then decreased exponentially. Total species richness was controlled predominantly by heat load index, mean annual precipitation, humus content and soil skeleton content. The positive role of soil pH was evident only for vascular plant species richness. Similar to other studies, we found that the importance of environmental factors strongly differed among taxonomic groups and across spatial scales, thus highlighting the need to study more than one taxon and more than one plot size to get a reliable picture.  相似文献

Phenanthrene-degrading bacteria were isolated from a 1-m2 intertidal sediment site in Boston Harbor. Samples were taken six times over 2 years. A total of 432 bacteria were isolated and characterized by biochemical testing. When clustered on the basis of phenotypic characteristics, the isolates could be separated into 68 groups at a similarity level of approximately 70%. Several groups (a total of 200 isolates) corresponded to well-characterized species belonging the genera Vibrio and Pseudomonas. Only 51 of the 437 isolates (<11.7% of the total) hybridized to a DNA probe that encodes the upper pathway of naphthalene and phenanthrene degradation in Pseudomonas putida NCIB 9816. A cluster analysis indicated that the species composition of the phenanthrene-degrading community changed significantly from sampling date to sampling date. At one sampling time, 12 6-mm-diameter core subsamples were taken within the 1-m2 site to determine the spatial variability of the degrading communities. An analysis of molecular variance, performed with the phenotypic characteristics, indicated that only 6% of the variation occurred among the 12 subsamples, suggesting that the subsamples were almost identical in composition. We concluded that the communities of phenanthrene-degrading bacteria in the sediments are very diverse, that the community structure undergoes significant change with time but does not vary significantly on a spatial scale of centimeters, and that the predominant genes that encode phenanthrene degradation in the communities are not well-characterized.  相似文献

Background and Aims

The dwarf eelgrass, Zostera noltii, is a predominant inhabitant of soft-bottom intertidal regions along the coasts of northern Europe. It is a monoecious, protogynous angiosperm in which the potential for self-fertilization and inbreeding are high, especially if clone sizes exceed pollen dispersal distances. The aim of the present study was to determine the relationship between mating system and clonal structure, examine the relative roles of geitonogamous selfing and biparental inbreeding, measure pollen availability (multiple paternities) and estimate pollen dispersal.

Methods

A 100-m2 plot was established in a large, intertidal Z. noltii meadow on the island of Sylt in the German Wadden Sea. A total of 256 adult shoots was sampled: one from the centre of 100 fixed 1-m2 quadrats (large scale resolution) and an additional 156 from within eight randomly selected 1-m2 sub-quadrats (small-scale resolution). DNA was extracted from seeds and leaf tissue of all samples and genotyped with nine microsatellite loci.

Key Results

Mating system analysis revealed high multilocus and single locus outcrossing rates. Average pollen dispersal distance was nearly the same as the average genet (clone) size. Multiple paternity was common and 20–30 % of mature seeds originated from matings within the plot. Among inbred seeds, most resulted from geitonogamy rather than biparental inbreeding.

Conclusions

Moderate disturbances intrinsic to the intertidal habitat appear to facilitate seed recruitment by gap formation. Pollen dispersal distances are sufficient to maintain outcrossing and high clonal diversity.  相似文献

Characteristics of social behavior and time budgets of common voles Microtus arvalis arvalis and M. a. obscurus were studied in 34-m2 enclosures. Ten family groups of each chromosomal form (a total of 60 animals) were used. The experiment consisted of four stages, the vole behavior being monitored (I) when they settled in a new territory; (II) when they were in a familiar territory; and upon placing (III) an unfamiliar male or (IV) an unfamiliar female of the same chromosomal race in the enclosure. Observations were performed for a total of 1120 h. The time budgets of the two vicarious forms of voles were compared with respect to the durations of sitting outside the nest boxes, grooming, active movement, and foraging. The social behavior of M. a. arvalis and M. a. obscurus was analyzed with respect to the numbers of intragroup interactions and the ratio between different types of contacts (exploratory, amicable, aggressive, contact evasion, and avoidance).  相似文献

Four plant functional types (PFTs) were used to compare the vegetation structure of an alien-invaded Acacia nilotica savanna with one of negligible invasions. Heights, canopy covers and species richness of three native PFTs (woody plants, grasses and herbs) and one alien PFT (woody plants) were measured in 14, 1-m2 quadrats sampled in a stratified-random pattern in a 400-m2 plot demarcated in each savanna. In the uninvaded plot, mean heights of native PFTs were stratified. In the invaded plot, the mean height of aliens extended into the native woody stratum with the lower range of native woody PFT heights reduced to the grass stratum. Discriminant analysis of canopy covers and species richness of the four PFTs revealed significant differences in composition between plots with the alien PFT being the most important variable correlated with these differences. Univariate analysis confirmed the dominance of alien woody plants in the invaded plot but also showed significant reductions in the canopy covers and species richness of native herbs and grasses compared to those in the uninvaded plot. These results suggest that PFTs can rapidly measure small-scale, spatial differences in the physiognomy, composition and species richness of A. nilotica savannas when invaded by alien woody plants.  相似文献

In Drosophila melanogaster, natural genetic variation in the foraging gene affects the foraging behaviour of larval and adult flies, larval reward learning, adult visual learning, and adult aversive training tasks. Sitters (for s) are more sedentary and aggregate within food patches whereas rovers (forR) have greater movement within and between food patches, suggesting that these natural variants are likely to experience different social environments. We hypothesized that social context would differentially influence rover and sitter behaviour in a cognitive task. We measured adult rover and sitter performance in a classical olfactory training test in groups and alone. All flies were reared in groups, but fly training and testing were done alone and in groups. Sitters trained and tested in a group had significantly higher learning performances compared to sitters trained and tested alone. Rovers performed similarly when trained and tested alone and in a group. In other words, rovers learning ability is independent of group training and testing. This suggests that sitters may be more sensitive to the social context than rovers. These differences in learning performance can be altered by pharmacological manipulations of PKG activity levels, the foraging (for) gene''s gene product. Learning and memory is also affected by the type of social interaction (being in a group of the same strain or in a group of a different strain) in rovers, but not in sitters. These results suggest that for mediates social learning and memory in D. melanogaster.  相似文献

Abstract. Several researchers have hypothesized that plant species richness has a unimodal relationship with biomass, while others have argued for a linear relationship. Data from various types of herbaceous communities show some support for the unimodal hypothesis, but this has not been tested extensively for forests and questions remain concerning its generality. We used linear and quadratic regression models to examine the relationship between overstory biomass and richness in a coastal Maine Quercus-Pinus forest across and within cover types using data from two plot sizes (2500-m2 quadrats and 625-m2 sub-quadrats). Understory data from 1-m2 plots were also analysed. Richness was quadratically related to biomass at both plot sizes for all cover types combined, but the amount of variation explained by the models was very low (R2s < 0.09). Richness and biomass were not significantly related at either plot size for the mixed mesic cover type, the most common type in the forest. The best fit (R2= 0.43) was obtained with a quadratic model for the conifer cover type at the sub-quadrat level, with the quadratic model for the 1-m2 data having the second highest R2 (0.24). Across all six data sets, the quadratic model was the only one with a significant fit in two cases, and had considerably higher R2s (1.3–1.9 ×) in two others. The remaining two data sets could not be fit with a significant model of either type. For this forest, these results suggest little support for a linear relationship between plant species richness and biomass and variable, often weak, support for a unimodal relationship. Density, a potentially confounding variable in this type of analysis, was only weakly correlated with richness and was not found to alter the relationship between biomass and richness.  相似文献

Trends in coral cover are widely used to indicate the health of coral reefs but are costly to obtain from field survey over large areas. In situ studies of reflected spectra at the coral surface show that living and recently dead colonies can be distinguished. Here, we investigate whether such spectral differences can be detected using an airborne remote sensing instrument. The Compact Airborne Spectrographic Imager (Itres Research Ltd, Canada) was flown in two configurations: 10 spectral bands with 1-m2 pixels and 6 spectral bands with 0.25-m2 pixels. First, we show that an instrument with 10 spectral bands possesses adequate spectral resolution to distinguish living Porites, living Pocillopora spp., partially dead Porites, recently dead Porites (total colony mortality within 6 months), old dead (>6 months) Porites, Halimeda spp., and coralline red algae when there is no water column to confuse spectra. All substrata were distinguished using fourth-order spectral derivatives around 538 nm and 562 nm. Then, at a shallow site (Tivaru) at Rangiroa Atoll, Tuamotu Archipelago (French Polynesia), we show that live and dead coral can be distinguished from the air to a depth of at least 4 m using first- and fourth-order spectral derivatives between 562–580 nm. However, partially dead and recently dead Porites colonies could not be distinguished from an airborne platform. Spectral differences among substrata are then exploited to predict the cover of reef substrata in ten 25-m2 plots at nearby Motu Nuhi (max depth 8 m). The actual cover in these plots was determined in situ using quadrats with a 0.01-m2 grid. Considerable disparity occurred between field and image-based measures of substrate cover within individual 25-m2 quadrats. At this small scale, disparity, measured as the absolute difference in cover between field and remote-sensing methods, reached 25% in some substrata but was always less than 10% for living coral (99% of which consisted of Porites spp.). At the scale of the reef (all ten 25-m2 quadrats), however, disparities in percent cover between imagery and field data were less than 10% for all substrata and extremely low for some classes (e.g. <3% for living Porites, recently dead Porites and Halimeda). The least accurately estimated substrata were sand and coralline red algae, which were overestimated by absolute values 7.9% and 6.6%, respectively. The precision of sampling was similar for field and remote-sensing methods: field methods required 19 plots to detect a 10% difference in coral cover among three reefs with a statistical power of 95%. Remote-sensing methods required 21 plots. However, it took 1 h to acquire imagery over 92,500 m2 of reef, which represents 3,700 plots of 25 m2 each, compared with 3 days to survey 10 such plots underwater. There were no significant differences in accuracy between 1-m2 and 0.25-m2 image resolutions, suggesting that the advantage of using smaller pixels is offset by reduced spectral information and an increase in noise (noise was observed to be 1.6–1.8 times greater in 0.25-m2 pixels). We show that airborne remote sensing can be used to monitor coral and algal cover over large areas, providing that water is shallow and clear, and that brown fleshy macroalgae are scarce, that depth is known independently (e.g. from sonar survey).  相似文献

We have previously described the safety and immunomodulatory effects of Lactobacillus paracasei CNCM I-4034, Bifidobacterium breve CNCM I-4035 and Lactobacillus rhamnosus CNCM I-4036 in healthy volunteers. The scope of this work was to evaluate the effects of these probiotic strains on the hepatic steatosis of obese rats. We used the Zucker rat as a genetic model of obesity. Zucker-Leprfa/fa rats received one of three probiotic strains, a mixture of L. paracasei CNCM I-4034 and B. breve CNCM I-4035, or a placebo for 30 days. An additional group of Zucker-lean+/fa rats received a placebo for 30 days. No alterations in intestinal histology, in the epithelial, lamina propria, muscular layers of the ileal or colonic mucosa, or the submucosae, were observed in any of the experimental groups. Triacylglycerol content decreased in the liver of Zucker-Leprfa/fa rats that were fed L. rhamnosus, B. breve, or the mixture of B. breve and L. paracasei. Likewise, the area corresponding to neutral lipids was significantly smaller in the liver of all four groups of Zucker-Leprfa/fa rats that received probiotics than in rats fed the placebo. Zucker-Leprfa/fa rats exhibited significantly greater serum LPS levels than Zucker-lean+/fa rats upon administration of placebo for 30 days. In contrast, all four groups of obese Zucker-Leprfa/fa rats that received LAB strains exhibited serum LPS concentrations similar to those of Zucker-lean+/fa rats. Serum TNF-α levels decreased in the Zucker-Leprfa/fa rats that received B. breve, L. rhamnosus, or the mixture, whereas L. paracasei feeding decreased IL-6 levels in the serum of Zucker-Leprfa/fa rats. In conclusion, the probiotic strains reduced hepatic steatosis in part by lowering serum LPS, and had an anti-inflammatory effect in obese Zucker rats.  相似文献

Abstract: We evaluated a chamber and nose cone method of isoflurane delivery for anesthetizing eastern gray squirrels (Sciurus carolinensis; summer n = 43, winter n = 48) and Allegheny woodrats (Neotoma magister; summer n = 24, winter n = 13) for use when pain or stress was possible from sampling procedures. Mean induction time for squirrels (from beginning of isoflurane administration to safe removal from trap), was 4.63 ± 0.58 minutes. Squirrels awoke more quickly in summer (1.40 ± 0.15 min) than in winter (3.62 ± 0.24 min) after removal of the nose cone. We manually restrained woodrats and administered the nose cone for 0.5 minutes to each animal. Woodrats awoke after 4.76 ± 0.58 minutes following the final dose of isoflurane for both seasons. These methods are useful for working with small mammals in the field and provide an appropriate anesthetic when there may be more than slight pain or distress.  相似文献

Abstract. Species composition patterns and vegetation-environment relationships were quantified for high-elevation rock outcrops of the Southern Appalachian Mountains, an infrequent and insular habitat in a forested landscape. Outcrops occur over a wide geographic range encompassing extensive variation in both geology and climate. Geographic-scale factors interact with site-scale factors to produce variation in vegetation among outcrops. Similarly, site-scale factors interact with micro-scale factors to produce variation in vegetation within outcrops. To provide a quantitatively-based classification of outcrop vegetation we used a TWINSPAN analysis of 154 100-m2 plots. We recognized nine communities that primarily correspond to different combinations of elevation, bedrock type, geography, and moisture. Within outcrops of a single bedrock type, vegetation composition of 100-m2 plots was consistently correlated with elevation and solar radiation, but relationships to soil nutrients varied with bedrock type. Both site-scale (100 m2) factors (e.g. elevation, slope, aspect, and bedrock type) and plot-scale (1-m2) microsite factors (e.g. soil depth, vegetation height, soil nutrients) were strongly correlated with species composition at the 1-m2 level. Environment can be used to predict composition more effectively for 100-m2 plots on a single bedrock type than either across bedrock types or at a 1-m2 scale. Composition-environment relationships resemble those described for outcrop systems from other regions with pronounced topographic relief more than they do those described for the nearby but flatter and lower-elevation outcrops of the Southeastern Piedmont. There is strong spatial autocorrelation in this community, perhaps owing to dispersal limitation. Consequently, a comprehensive conservation strategy must include reservation of both a range of geologic types and a range of geographic locations.  相似文献

Based on the hypothesis that, in Akodon azarae, polygyny operates through female defence, we studied inter-male aggression in order to test the following predictions: during the breeding period (1) resident males are more aggressive than intruder males in the presence of females (FP), and (2) aggressive behaviour is independent of male condition (resident or intruder) in the absence of females (FA). To test our predictions, we used the resident male behavioural response towards an intruder male in relation to FP or FA. We conducted 30 encounters in FP and 27 in FA in 0.79-m2 round enclosures placed in the Espinal Reservation. Our results support the prediction that, in FP, the intensity of aggressive behaviour exhibited by males varied in relation to resident or intruder condition. Resident males showed high levels of aggression towards intruders, and intruders exhibited the greatest values of submissive behaviours with residents. In FA, the intensity of aggressive behaviour did not vary in relation to resident or intruder condition. Both resident and intruder males exhibited low aggressive behaviour and inter-male encounters resulted mainly in non interactive behaviours. Our results support the hypothesis that, in A. azarae, the polygynous mating system operates through female defence.  相似文献

The in vitro deacetylation of N8-acetylspermidine by an enzyme activity in rat tissues is described. This deacetylase activity occurs as a soluble, cytoplasmic enzyme in rat liver and was detected in the 100,000g supernatant fraction of all tissues examined. The highest specific activity was found in liver. Spleen, kidney, and lung were found to contain 20–50% of the activity in liver, while heart, brain, and skeletal muscle exhibited from 2 to 10% of the activity in liver. Serum contained only barely detectable levels of activity, much lower than any of the tissues studied. The in vitro metabolism of N1-acetylspermidine differed from that observed for N8-acetylspermidine and does not appear to involve a simple deacetylation reaction.  相似文献

Large-scale (>100 m2/day) raids by tropical army ants have been linked to litter ant patchiness and diversity. In three Neotropical forests, densities of litter ants vary 10–20 fold at the 1-m2 scale. A survey of Barro Colorado Island. Panama, revealed that most army ant raids also occur on a 1-m2 scale with fronts 1 m wide. To explore the role that such small scale disturbance may play in creating litter ant patchiness, all litter ant nests were removed from 1-m2 plots. Control and disturbance plots were resampled 3 months later. In contrast to a previous study of large litter gaps, ant foundresses did not appear to prefer these smaller gaps. Nest densities, species richness, and species composition differed most from controls in a dry hilltop forest in Panama, second most in a wetter ravine forest nearby, and least in a Costa Rican wet forest. Disturbance may not leave a lasting signature in the wetter forests due to higher background levels of disturbance, faster recovery, or both.  相似文献
