Genetic diversity was studied among 21 accessions of lentil using SSR markers and morphological traits in order to assess the diversification of Indian gene-pool of lentil through introgression of exotic genes and introduction of germplasm. Among these , 16 genotypes either had ‘Precoz’ gene, an Argentine line in their pedigree or genes from introduced lines from ICARDA. Sixty five SSR markers and eight phenotypic traits were used to analyse the level of genetic diversity in these genotypes. Forty three SSR markers (66 %) were polymorphic and generated a total of 177 alleles with an average of 4.1 alleles per SSR marker. Alleles per marker ranged from 2 to 6. The polymorphic information content ranged 0.33 to 0.80 with an average of 0.57, suggesting that SSR markers are highly polymorphic among the studied genotypes. Genetic dissimilarity based a dendrogram grouped these accessions into two main clusters (cluster I and cluster II) and it ranged 33 % to 71 %, suggesting high level of genetic diversity among the genotypes. First three components of PCA based morphological traits explained higher variance (95.6 %) compared to PCA components based on SSR markers (42.7 %) of total genetic variance. Thus, more diversity was observed for morphological traits and genotypes in each cluster and sub-cluster showed a range of variability for seed size, earliness, pods/plant and plant height. Molecular and phenotypic diversity analysis thus suggested that use of germplasm of exotic lines have diversified the genetic base of lentil germplasm in India. This diversified gene-pool will be very useful in the development of improved varieties of lentil in order to address the effect of climate change, to adapt in new cropping systems niches such as mixed cropping, relay cropping, etc. and to meet consumers’ preference.  相似文献

Brassica rapa (2n = 20, AA genome) is an important oil yielding species of the family Brassicaceae and characterized by wide range of genetic and morphological subtypes suitable for cultivation under diverse agro-climatic regions of India. In this study, genetic diversity among three subspecies of B. rapa including yellow sarson, toria and outlier brown sarson was estimated using various agro-morphological traits and simple sequence repeat (SSR) markers. Maximum variability was recorded for siliqua angle (Coefficient of variation = 30.9%), followed by seeds/siliqua (CV = 18.8%), leaf length (CV = 10%) and plant height (CV = 16.8%). Principal component analysis explained more than 50% of the total observed morphological variability for first two components. Of the 107 SSR markers tested, 80 generated reproducible, clear and distinct amplicons of which, 65 (81.25%) were found polymorphic. The number of alleles at each locus ranged from 2 to 7, with an average of 3.03 alleles per marker. A total of 197 alleles were detected at 65 SSR loci with average PIC value of 0.457 and a mean resolving power of 3.04. Neighbor-Joining cluster analysis based on morphological traits and SSR markers separately classified all the 28 genotypes into five major groups. The population structure analysis resulted into three sub-populations with certain extent of admixture among the earlier established taxonomic sub-groups. Twenty-three unique alleles were detected in thirteen B. rapa varieties. The clustering analysis and principal coordinate analysis outlined the genetic relationships among different varieties belonging to the three subspecies of B. rapa. Genetically diverse genotypes as illustrated by score plots and from the clustering patterns brought out the wide range of diversity present among B. rapa genotypes and the underlying options available for selecting parental genotypes for hybridization and developing high yielding cultivars suitable for Indian conditions.  相似文献

The genetic distance analysis for selection of suitable parents has been established and effectively used in many crops; however, there is dearth of conclusive report of relationship of genetic distance analysis with heterosis in sesame. In the present study, an attempt was made to estimate the associations of genetic distances using SSR (GDSSR), seed-storage protein profiling (GDSDS) and agro-morphological traits (GDMOR) with hybrid performance. Seven parents were selected from 60 exotic and Indian genotypes based on genetic distance from clustering pattern based on SSR, seed-storage protein, morphological traits and per se performance. For combining ability analysis, 7 parents and 21 crosses generated from 7 × 7 half diallel evaluated at two environments in a replicated field trial during pre-kharif season of 2013. Compared with the average parents yield (12.57 g plant?1), eight hybrids had a significant (P < 0.01) yield advantage across environments, with averages of 26.94 and 29.99% for better-parent heterosis (BPH) and mid-parent heterosis (MPH), respectively, across environments. Highly significant positive correlation was observed between specific combining ability (SCA) and per se performance (0.97), while positive non-significant correlation of BPH with GDSSR (0.048), and non-significant negative correlations with GDMOR (? 0.01) and GDSDS (? 0.256) were observed. The linear regressions of SCA on MPH, BPH and per se performance of F1s were significant with R2 value of 0.88, 0.84 and 0.95 respectively. The present findings revealed a weak association of GDSSR with F1’s performance; however, SCA has appeared as an important factor in the determination of heterosis and per se performance of the hybrids. The present findings also indicated that parental divergence in the intermediate group would likely produce high heterotic crosses in sesame.  相似文献

Evaluation of breeding progress for spring barley varieties in Germany showed that both grain yield and malting quality were considerably improved during the last 33 years, and that genetic effects of protein concentration and malting traits were not associated.

Abstract

Based on historical data, this study aimed to investigate yield potential and malting quality of 187 varieties tested and released in German registration trials to evaluate the value for cultivation and use (VCU) during 1983–2015, and to quantify the environmental variability and the association among traits. We used mixed linear models with multiple linear regression terms to dissect genetic and non-genetic trend components. Grain yield increased by 43% (23.4 dt ha?1) in VCU trials and 35% (14.0 dt ha?1) on-farm relative to 1983. All yield components contributed significantly. Malting quality was also considerably improved by 2.3% for extract content up to 25.1% for friability, relative to 1983, nearly completely due to new varieties. Total variability of individual traits was very different between traits (2.4–24.4% relative to 1983). The relative influence of genotypes on total variation was low for grain yield and its components, whereas it was considerably larger for other traits. We found remarkable differences between phenotypic and genetic correlation coefficients for grain yield and protein concentration with malting traits. The observed positive phenotypic relation between grain yield and malting quality can be attributed to a shift of selection and environmental effects, but genetic correlations showed a negative association. Genetic effects of protein concentration and malting quality were not correlated indicating that both were not genetically linked. Considerable yield progress and improvement of malting quality were achieved despite of their weak to moderate negative genetic dependence.

Identification of stable quantitative trait loci (QTLs) across different environments and mapping populations is a prerequisite for marker-assisted selection (MAS) for cotton yield and fiber quality. To construct a genetic linkage map and to identify QTLs for fiber quality and yield traits, a backcross inbred line (BIL) population of 146 lines was developed from a cross between Upland cotton (Gossypium hirsutum) and Egyptian cotton (Gossypium barbadense) through two generations of backcrossing using Upland cotton as the recurrent parent followed by four generations of self pollination. The BIL population together with its two parents was tested in five environments representing three major cotton production regions in China. The genetic map spanned a total genetic distance of 2,895 cM and contained 392 polymorphic SSR loci with an average genetic distance of 7.4 cM per marker. A total of 67 QTLs including 28 for fiber quality and 39 for yield and its components were detected on 23 chromosomes, each of which explained 6.65–25.27 % of the phenotypic variation. Twenty-nine QTLs were located on the At subgenome originated from a cultivated diploid cotton, while 38 were on the Dt subgenome from an ancestor that does not produce spinnable fibers. Of the eight common QTLs (12 %) detected in more than two environments, two were for fiber quality traits including one for fiber strength and one for uniformity, and six for yield and its components including three for lint yield, one for seedcotton yield, one for lint percentage and one for boll weight. QTL clusters for the same traits or different traits were also identified. This research represents one of the first reports using a permanent advanced backcross inbred population of an interspecific hybrid population to identify QTLs for fiber quality and yield traits in cotton across diverse environments. It provides useful information for transferring desirable genes from G. barbadense to G. hirsutum using MAS.  相似文献

The DNA polymorphism among 92 selected tef genotypes belonging to eight origin groups was assessed using eight inter simple sequence repeat (ISSR) primers. The objectives were to examine the possibility of using ISSR markers for unravelling genetic diversity in tef, and to assess the extent and pattern of genetic diversity in the test germplasm with respect to origin groups. The eight primers were able to separate or distinguish all of the 92 tef genotypes based on a total of 110 polymorphic bands among the test lines. The Jaccard similarity coefficient among the test genotypes ranged from 0.26 to 0.86, and at about 60 % similarity level the clustering of this matrix using the unweighted pair-group method based on arithmetic average (UPGMA) resulted in the formation of six major clusters of 2 to 37 lines with further eight lines remaining ungrouped. The standardized Nei genetic distance among the eight groups of origin ranged between 0.03 and 0.32. The UPGMA clustering using the standardized genetic distance matrix resulted in the identification of three clusters of the eight groups of origin with bootstrap values ranging from 56 to 97. The overall mean Shannon Weaver diversity index of the test lines was 0.73, indicating better resolution of genetic diversity in tef with ISSR markers than with phenotypic (morphological) traits used in previous studies. This can be attributed mainly to the larger number of loci generated for evaluation with ISSR analysis as compared to the few number of phenotypic traits amenable for assessment and which are further greatly affected by environment and genotype x environment interaction. Analysis of variance of mean Shannon Weaver diversity indices revealed substantial (P < or = 0.05) variation in the level of diversity among the eight groups of origin. In conclusion, our results indicate that ISSR can be useful as DNA-based molecular markers for studying genetic diversity and phylogenetic relationships, DNA fingerprinting for the identification of varieties or cultivars, and also for genome mapping in tef.  相似文献

Twelve important pomological traits related to fruit quality were studied during 3 years in an F1 apricot progeny of 160 seedlings derived from a cross between the Spanish selection ‘Z701-1’ and the South African cultivar ‘Palsteyn’. Results indicated quantitative transmission of most of the fruit quality traits studied. In addition, a clear influence of the genetic background of parents was observed. In some seedlings, values outside the range of the parent were observed due to the influence of this genetic background. No correlations were found among most agronomic traits in apricot during the 3 years of the study. However, high correlations between years were described for most of the evaluated traits, and the environment had limited influence on the expression of the trait. A genetic map was developed using 41 apricot and peach SSR markers. The map obtained showed eight linkage groups (corresponding to the eight chromosomes) covering a total distance of 369.3 cM and an average distance between markers of 9 cM. Fifty-four QTLs associated with different traits were identified, including: blooming date (linkage groups G1, G4 and G7); ripening time (G4 and G6); fruit development (G4 and G6); fruit weight (G1 and G4); stone weight (G1 and G7); flesh color (G1 and G6); pH (G1, G2 and G4); malic acid (G1, G2 and G4); and soluble solids content (G4 and G5). We have highlighted several QTLs in G4 that explain the variability in various traits related to fruit quality such as blooming date, ripening time, and soluble solids content. In addition, we have also highlighted an important QTL on G2 that explains much of the variation in levels of acidity.  相似文献

Genetic distances between seven oil sunflower inbred lines expressed in the variability of five morphological traits were analysed in terms of their effect on the specific combining ability (SCA) effects in these lines for yield component traits. The multivariate analysis of variance (MANOVA) and other related multidimensional methods were used to recognize the differences between genotypes with respect to morphological traits. Canonical analysis was applied for a graphical configuration of parental lines and diallele hybrids in the two-dimensional space. As a measure of the difference between parental lines with respect to the analysed morphological traits jointly, i.e. as a measure of the morphological distance between these forms, Mahalanobis' distance was used. Phenotypic differences between lines were estimated using Mahalanobis' distance for each pair of crossed lines calculated for all the analysed morphological traits. The effect of morphological differences between inbred lines on the mean SCA effect for individual yield components, measured by the linear regression, was significant for oil yield per plant and for the number of seeds per plant.  相似文献

The clustering was ratified by using twenty three molecular markers (SSR, EST and STS) linked to stripe rust (Puccinia striiformis f. sp. tritici) resistant QTLs. The aim was to asses the extent of genetic variability among the genotypes in order to select the parents for crossing between the resistant and susceptible genotypes with respect to stripe rust. The Euclidian dissimilarity values resulted from phenotypic data regarding morphological traits and AUDPC were used to construct a dendrogram for clustering the accessions. Using un-weighted pair group method with arithmetic means, another dendrogram resulted from the similarity coefficient values was used to distinguish the genotypes with respect to stripe rust. Clustering based on phenotypic data produced two major groups and five clusters (with Euclidian dissimilarity ranging from 2.44 to 16.16) whereas genotypic data yielded two major groups and four clusters (with percent similarity coefficient values ranging from 0.1 to 46.0) to separate the gene pool into highly resistant, resistant, moderately resistant, moderately susceptible and susceptible genotypes. With few exceptions, the outcome of both type of clustering was almost similar and resistant as well as susceptible genotypes came in the same clusters of molecular genotyping as yielded by phenotypic clustering. As a result seven genotypes (Bakhtawar-92, Frontana, Saleem 2000, Tatara, Inqilab-91, Fakhre Sarhad and Karwan) of diverse genetic background were selected for pyramiding stripe rust lesistant genes as well as some other agronomic traits after hybridization.  相似文献

Abstract

Coppice yield and components of growth were quantified in eight clones of two widely distributed North American willows, Salix discolor (DIS) and S. eriocephala (ERI), in common-garden field tests on three sites of varying quality. Both species and clones within species varied significantly across different sites and interacted with site for plant yield and components of growth traits. At the species level, ERI was significantly more productive than DIS on the two most productive sites (85 and 57 % greater, respectively), but on the poorest site, a shale coal mine overburden, species ranking was reversed, with DIS showing a 60 % greater biomass yield than ERI. These sites had similar mean temperature, growing degree days, and seasonal precipitation. Site quality differences were thus most probably driven by soil nutrients, physical traits, and water-holding capacity quantified by 13 soil properties, 12 of which were significantly different and showed fairly consistent ranking among sites. At the clonal level, growth trends and differences were mostly consistent among clones across the three test sites, with the exception of one clone of DIS and to a lesser extent a clone of ERI, which showed abnormally strong clone by site interaction for specific growth traits. Productivity reached as high as 6.0 kg green mass in 2-year-old coppices for a clone from each of DIS and ERI on the most productive site. The strong expression of genetic by environment interactions at both the species and clonal levels suggests that biomass production can be optimized by taking advantage of such interactions and highlights the need for testing not only different species, but also a number of clones within a species before selecting clones for biomass production on different site types. Our results highlight variation in coppice form and the potential for genetic selection both among and within species.  相似文献

Mooseer or Persian shallot (Allium hirtifolium Boiss.) is a bulbous perennial herb belonging to Alliaceae family. Genetic diversity of 16 mooseer populations collected from different parts of Iran were evaluated using 14 morphological traits as well as antioxidant activities and RAPD markers. Statistical analysis showed significant differences among populations for all morphological traits. In a principal component analysis, four components showed 77.5 % of the total morphological variation. Based on the morphological traits, populations were divided into five groups by clustering. Antioxidant capacities differed among all populations, with the Sahneh and Nahavand populations having the highest and the lowest activities, respectively. Genetic diversity of populations was also investigated through RAPD technique using 14 random primers. Cluster analysis based on RAPD markers plus 47 % coefficient of similarity divided the genotypes into six distinct groups. The highest and lowest similarities between populations were detected to be 0.63 and 0.21, respectively. These results indicated that the assessment of genetic diversity in Iranian mooseer populations could be appropriately carried out using RAPD technique which correlates with morphological traits.  相似文献

Abstract

Study was carried out on 15 6-year-old candidate plus trees of Jatropha curcas adapted on semi-arid wasteland with an objective to assess variation in morphological, physiological and oil quality characters. Heritable and non-heritable components of the total variability of the characters were determined by genotypic (GCV) and phenotypic (PCV) co-efficient of variation, heritability and genetic advance (GA) and the best gain traits for Jatropha improvement through selection and breeding were assessed. Further, association among the traits were assessed and germplasm were separated into different clusters. Significant variation was found among the different genotypes for all the characters. The photosynthetic and transpiration rate correlated with oil content, seed and oil yield. The chlorophyll pigments correlated positively with the photosynthetic rate and oil content. The seed oil content varied considerably from 27.68 % (JCN01) to 37.49 % (JCN14) and had high heritability, but it had low PCV and GCV and moderate GA. The oil yield plant?1 had high genetic variability and varied significantly from 0.07 (JCN15) to 0.47 kg plant?1 (JCN09/IC 565733). Though the different fatty acids differed significantly with different germplasm and also had high heritability, they had low PCV, GCV and GA. Seed weight, fruit weight, seed weight fruit?1 and seed yield plant?1 strongly correlated with oil yield and had moderate to high GCV, PCV, coupled with high heritability and GA. Germplasm were separated into four distinct clusters with a maximum inter distance found between cluster II and IV, and minimum between cluster I and III. The study helped to identify the superior germplasm among diverse genotypes of J. curcas that can serve as parents with desirable characters like high oil yield, low stomatal conductance and high water use efficiency for further breeding purposes.

The study aims at identifying some submergence-tolerant rice genotypes through morphological and molecular characterization and their genetic variability analysis. Ten rice genotypes including two submergence-tolerant checks, two susceptible varieties and six advanced lines were evaluated for submergence tolerance in the laboratory and in the field during January–December 2015. The experiment was conducted in the field following randomized complete block design in a two-factor arrangement using five replications. Ten characters, viz. days to flowering, plant height, tiller number plant?1, effective tiller plant?1, and yield plant?1 etc. were studied for four treatments. A significant genotype × environment interaction was observed for all traits studied in this experiment. The yield was reduced for all genotypes at a different level of submergence stress compared to control. Binadhan-11, Binadhan-12, RC 249 and RC 251 showed tolerance, whereas RC 192, RC 193 and RC 225 showed moderate tolerance in submerged condition. The phenotypic coefficient of variance (PCV) was higher than the genotypic coefficient of variance (GCV) in all the studies traits. High heritability (75–97%) was found for all traits. High heritability along with high genetic advance was found for days to flowering (45.55) and plant height (40.05). Molecular characterization of the used genotypes was done with three SSR markers viz. RM 24, and submergence specific SC3 and SUB1. SC3 was found reliable for detection of submergence tolerant genotypes due to the highest gene diversity (0.840) compared to others. The banding pattern of the submergence specific markers SC3 and SUB1 identified in Binadhan-11, Binadhan-12, RC 192, RC 193, RC 225, RC 227, RC 249, and RC 251, which possess the SUB1 gene. Finally, clustering also separates the tolerant genotypes from the susceptible by dividing them into different clusters. The identified genotypes might be useful for the breeding programme for the development of submergence tolerant as well as resistant rice variety in Bangladesh.  相似文献

Sorghum, a cereal of economic importance ensures food and fodder security for millions of rural families in the semi-arid tropics. The objective of the present study was to identify and validate quantitative trait loci (QTL) for grain yield and other agronomic traits using replicated phenotypic data sets from three post-rainy dry sorghum crop seasons involving a mapping population with 245 F9 recombinant inbred lines derived from a cross of M35-1 × B35. A genetic linkage map was constructed with 237 markers consisting of 174 genomic, 60 genic and 3 morphological markers. The QTL analysis for 11 traits following composite interval mapping identified 91 QTL with 5–12 QTL for each trait. QTL detected in the population individually explained phenotypic variation between 2.5 and 30.3 % for a given trait and six major genomic regions with QTL effect on multiple traits were identified. Stable QTL across seasons were identified. Of the 60 genic markers mapped, 21 were found at QTL peak or tightly linked with QTL. A gene-based marker XnhsbSFCILP67 (Sb03g028240) on SBI-03, encoding indole-3-acetic acid-amido synthetase GH3.5, was found to be involved in QTL for seven traits. The QTL-linked markers identified for 11 agronomic traits may assist in fine mapping, map-based gene isolation and also for improving post-rainy sorghum through marker-assisted breeding.  相似文献

In this study, the genetic variability and relationships among wild and cultivated pomegranate genotypes from the north of Iran were investigated by morphological characters and RAPD molecular markers. Principal component analysis showed that the first three components explained 61.64 % of the total morphological variation for studied genotypes. Fruit neck diameter, anthocyanin index, TSS, aril juice, fruit flavor index, petiole length, fruit peel thickness and seed hardness were predominant in the first component and contributed most of the total variation. Fruit characteristics such as titratable acidity were negatively correlated (r = ?0.56) with TSS (r = ?0.56) and pH (r = ?0.86) and also, seed hardness showed negative correlation with aril length and aril diameter. Clustering from morphological data allocated individuals into two main clusters with high variation. Two hundred and twenty-nine fragments were scored of which 174 of them were polymorphic with 76.9 % polymorphism. Genetic similarity ranged from 0.15 to 0.78 with an average of 0.42, indicating high genetic variation among studied genotypes. High molecular and morphological variability indicated that this germplasm includes rich and valuable plant materials for pomegranate breeding.  相似文献

对资源圃保存的212份大蒜种质资源的表型性状进行了系统鉴定,分析表明我国大蒜种质资源的表型变异丰富。主成分分析表明,29个性状可以归并为反映植株生长发育、产品特征和产量构成的8个主成分;进一步的聚类分析将所有资源分为特点明显的2类5亚类。为了避免质量性状在种质评价中的主导作用,与产量相关的鳞茎数量性状的主成分分析显示,前3个主成分累积贡献率达74.83%,第一主成分中鳞茎重、鳞茎直径、鳞茎高和鳞芽数是影响产量的主要因子。主坐标排序将所有资源分为6类。通过综合评价,将大蒜鳞茎产量分为6个级别,筛选出单产大于15 t/hm2的资源3份。  相似文献

Phenotypic and genotypic variation and structure of 29 sour cherry (P. cerasus) and duke cherry (P. x gondouinii) genotypes from different regions of Iran were identified using random amplified polymorphic DNA (RAPD) markers and morphological characters. Furthermore, one Prunus mahaleb genotype was used as an outgroup for molecular analysis. For morphological analysis, 23 variables were recorded to detect similarities between and among studied sour and duke cherries. Most studied characteristics were showing a high degree of variability. Principal component analysis showed that the first three components explained a total of 73.87 % of the whole phenotypic variability. Based on the morphological cluster analysis, studied sour and duke cherry genotypes were placed into three main clusters. The first main cluster included 16 sour cherry genotypes. The second main cluster contained all duke cherry genotypes and eight sour cherry genotypes, while, only one sour cherry genotype was placed in third main cluster. For RAPD analysis, 17 primers generated a total of 233 discernible and reproducible bands across genotypes analyzed, out of which 214 (91.51 %) were polymorphic with varied band size from 300 to 3000 bp. According to the similarity matrix, the lowest similarity was obtained between P. mahaleb, as an outgroup, and sour cherry. Dendrogram based on molecular data separated genotypes according to their species and geographic origin. Low correlation was observed between the similarity matrices obtained based on morphological and RAPD data. The information obtained here could be valuable for devising strategies for conservation of Iranian sour and duke cherries.  相似文献

Data on genetic similarity among crop cultivars is of vital importance for the plant breeder. The objectives of this study were to group pepper (Capsicum annuum L.) genotypes into clusters according to their distances as estimated by morphological traits and amplified fragment length polymorphism (AFLP) markers and to assess the relationships between the two. Thirty-nine pepper genotypes obtained from different countries were grown in the greenhouse at University of the Free State, South Africa, during 2001 and 2002 in a randomized complete block design with three replications. A total of 20 different morphological traits were measured and six AFLP primer pairs were used to estimate pairwise genetic distances. Both datasets showed high genetic distances among the different genotypes, indicating high genetic diversity among them. The mean genetic distance among Ethiopian pungent elongated-fruit genotypes, was lower than that between them and the introduced ones. Morphological and AFLP distance estimations generally clustered together genotypes with similar fruit sizes. Significant, positive correlation was observed between morphological and AFLP diversity estimations. The narrow genetic basis among the Ethiopian pungent elongated-fruit cultivars suggests that the pepper breeding program of Ethiopia should focus on enriching its germplasm through local collection and introductions from other parts of the world.  相似文献
