Herein, the ecology of genus Propylea, with special reference to its three species — viz. P. dissecta (Mulsant), P. japonica (Thunberg), and P. quatuordecimpunctata Linnaeus is reviewed, with an eye toward its position in natural habitats and amongst other ladybirds and its biocontrol potential. Although Propylea is not as polyphagous as other successful species, it evidences a preference for certain prey, specifically aphids and whiteflies. This genus is a good model for studies of mating, reproduction, sexual selection, etc. Propylea is usually an intraguild prey in the guilds containing ladybirds, such as Harmonia axyridis (Pallas) and Coccinella septempunctata L. It also appears that it retains some intrinsic advantages that aid in the sustenance and survival of the species. Owing to its smaller size and level of victimization in the guild, its biocontrol potential is questionable, but its size disadvantages are counter-balanced by its high intrinsic rate of increase, high predation, reproductive potential, and bioconversion efficiency, as well as the ease with which it can be reared in the laboratory. Further research will be necessary to ascertain clearly the biocontrol potential of Propylea.  相似文献

Listeria monocytogenes is the etiologic agent of listeriosis, a food-borne disease affecting humans and a variety of animals. In order to combat this pathogen, it is crucial to have an understanding of its natural interplay with the environment. For this reason, the free soil nematode Caenorhabditis elegans was focused upon because of its shared natural habitat with Listeria and its potential as a model organism for Listeria pathogenesis. Previous studies have generated some contradictory results on Listeria’s ability to kill C. elegans, making additional interaction studies such as this more attractive. In our study, we carried out a series of killing assays in a systematic manner using different Listeria strains under different growth conditions. In addition to studying the effects of planktonic cells, we examined the interaction between C. elegans and sessile listerial cells. Our findings suggest that, rather than causing infection and death, L. monocytogenes may extend the life span of C. elegans. This indicates that Listeria is not pathogenic to C. elegans. We also found that C. elegans can feed and ingest sessile cells, as well as carry the pathogen in its gut, implying that C. elegans could be a vehicle for L. monocytogenes spread in the environment.  相似文献

The cell wall of M. tuberculosis is central to its success as a pathogen. Mycolic acids are key components of this cell wall. The genes involved in joining the α and mero mycolates are located in a cluster, beginning with Rv3799c and extending at least until Rv3804c. The role of each enzyme encoded by these five genes is fairly well understood, except for Rv3802c. Rv3802 is one of seven putative cutinases encoded by the genome of M. tuberculosis. In phytopathogens, cutinases hydrolyze the waxy layer of plants, cutin. In a strictly mammalian pathogen, such as M. tuberculosis, it is likely that these proteins perform a different function. Of the seven, we chose to focus on Rv3802c because of its location in a mycolic acid synthesis gene cluster, its putative essentiality, its ubiquitous presence in actinomycetes, and its conservation in the minimal genome of Mycobacterium leprae. We expressed Rv3802 in Escherichia coli and purified the enzymatically active form. We probed its activities and inhibitors characterizing those relevant to its possible role in mycolic acid biosynthesis. In addition to its reported phospholipase A activity, Rv3802 has significant thioesterase activity, and it is inhibited by tetrahydrolipstatin (THL). THL is a described anti-tuberculous compound with an unknown mechanism, but it reportedly targets cell wall synthesis. Taken together, these data circumstantially support a role for Rv3802 in mycolic acid synthesis and, as the cell wall is integral to M. tuberculosis pathogenesis, identification of a novel cell wall enzyme and its inhibition has therapeutic and diagnostic implications.  相似文献

Competitiveness for nodulation is a desirable trait in rhizobia strains used as inoculant. In Sinorhizobium meliloti 1021 mutation in either of the trehalose utilization genes thuA or thuB influences its competitiveness for root colonization and nodule occupancy depending on the interacting host. We have therefore investigated whether mutation in the thuA ortholog in Mesorhizobium loti MAFF303099 also leads to a similar competitive phenotype on its hosts. The results show that M. loti thuA mutant Ml7023 was symbiotically effective and was as competitive as the wild type in colonization and nodule occupancy on Lotus corniculatus and Lotus japonicus. The thuA gene in M. loti was not induced during root colonization or in the infection threads unlike in S. meliloti, despite its induction by trehalose and high osmolarity in in vitro assays.  相似文献

Tricholoma dulciolens is an ectomycorrhizal basidiomycete that produces “matsutake”-like mushrooms in association with Picea abies in Fennoscandia. The phylogenetic position of T. dulciolens relative to matsutake species is, however, unknown. In the present study, we demonstrated that T. dulciolens is phylogenetically independent from T. matsutake and its allied species. In addition, herbarium specimens identified as T. caligatum from Abies alba forests in Spain were phylogenetically related to but distinct from T. dulciolens. Based on ribosomal internal transcribed spacer DNA sequence data, T. dulciolens appears to exist in North America, as well. This is the first phylogeny of T. dulciolens in Fennoscandia and its related species in southern Europe.  相似文献

Selected yeasts classified as Candida sake van Uden et Buckley were examined for their physiological, morphological and immunological properties and their DNA relatedness. Candida maltosa Komagata, Nakase et Katsuya is herein recognized as a species separate from C. sake. Candida maltosa was distinguished from C. sake and from C. tropicalis by insignificant DNA reassociation. In addition, C. maltosa was distinguished from C. sake by its higher maximal growth temperature and lower guanine plus cytosine content of its DNA and from C. tropicalis by its failure to utilize soluble starch for growth and its resistance to cycloheximide. The species C. cloacae and C. subtropicalis are placed in synonymy with C. maltosa.  相似文献

Noctuidonema guyaneme is an interesting ectoparasite of adult Lepidoptera that feeds on hosts from at least five families with its long stylet. Noctuidonema guyanense spends its entire life on the adult moth and is sustained as it is passed from moth to moth during host mating. Overlapping host generations are essential for parasite survival. This nematode occurs throughout tropical and subtropical America and is transported by at least one of its hosts, Spodoptera frugiperda, during migration to northern sites in the United States each spring. Noctuidonema guyanense debilitates its hosts. Research conducted to help determine the biological control importance of this nematode is reviewed. Two additional species, N. daptria and N. dibolia, are now known for Noctuidonema.  相似文献

Oilseed rape (Brassica napus) is a valuable crop, attacked by several insect pests, of which the pollen beetle (Meligethes aeneus) is one of the most widespread and important in Europe. The relative attractiveness for the pollen beetle of Brassica nigra, Eruca sativa and Raphanus sativus was compared with that of spring oilseed rape, to assess the potential of these plant species as trap crops for the pest. At early growth stages, B. nigra and R. sativus were more attractive to over-wintered adult pollen beetles than B. napus. At the bud and flowering stages, B. nigra was the most attractive while E. sativa was the least attractive. At flowering, B. nigra was more attractive for oviposition than the other species. Thus, of the species tested, B. nigra has the most potential as a prospective trap crop to protect spring oilseed rape before flowering when it is at its most vulnerable developmental phase, due to its faster development and its acceptability both for feeding and oviposition to overwintered pollen beetle adults. Raphanus sativus was accepted both for feeding and oviposition, but pollen beetle larvae failed to develop in its flowers; it therefore has the potential for use as a dead-end crop. At the end of the growing season, both E. sativa and R. sativus, as late-flowering species, attracted the new generation of pollen beetles and thereby have potential to extend the effectiveness of a trap-cropping system at this time.  相似文献

A new spleenwort hybrid, Asplenium pinnatifidum×trichomanes, from southern Illinois is nicely intermediate in morphology between the parents, and shows no pairing of its genomes: two sets of chromosomes from A.×pinnatifidum and one from A. trichomanes. Taxonomically the cross is significant for its bearing on the interpretation of the taxon A. stotleri Wherry, previously believed to have originated from the same parents. New studies of A. stotleri based upon materials from the now extinct type colony in West Virginia, and from a newly discovered extant colony in Arkansas, show clearly that this taxon is not the same as A. pinnatifidum×trichomanes. Comparisons with A.×bradleyi D. C. Eaton (the tetraploid of A. montanum×platyneuron) indicate that the taxon A. stotleri is similar in all respects except for its more rounded segments. Asplenium stotleri is therefore reinterpreted as a synonym of A.×bradleyi, of which it is regarded as a slightly differentiated local form.  相似文献

Exocelina manokwariensis sp. n. from West Papua is placed into the Exocelina ekari-group based on the structure of its male genitalia. The new species is described, including its three subspecies, from the mainland of West Papua, Waigeo Island, Batanta and Salawati Islands, and Bomberai peninsula. Malaria remains as one of the most deadly diseases in developing countries. The Plasmodium causative agents of human malaria such as Plasmodium falciparum possess an organelle, the apicoplast, which is the result of secondary endosymbiosis and retains its own circular DNA. A type II topoisomerase, DNA gyrase, is present in the apicoplast. In prokaryotes this enzyme is a proven, effective target for antibacterial agents, and its discovery in P. falciparum opens up the prospect of exploiting it as a drug target. Basic characterisation of P. falciparum gyrase is important because there are significant sequence differences between it and the prokaryotic enzyme. However, it has proved difficult to obtain soluble protein. Here we have predicted a new domain boundary in P. falciparum GyrA that corresponds to the C-terminal domain of prokaryotic GyrA and successfully purified it in a soluble form. Biochemical analyses revealed many similarities between the C-terminal domains of GyrA from E. coli and P. falciparum, suggesting that despite its considerably larger size, the malarial protein carries out a similar DNA wrapping function. Removal of a unique Asn-rich region in the P. falciparum protein did not result in a significant change, suggesting it is dispensable for DNA wrapping.  相似文献

Neottia cordata is an orchid species that exclusively occurs in Picea abies subsp. abies and Pinus sylvestris stands and forms its southernmost distribution limits in Greece. Over the last decade, it has been recorded in numerous sites, in which it forms small colonies, both in terms of population and area. This research re-evaluates the threat status of Neottia cordata based on updated chorological and population data; it also checks the effectiveness of the Natura 2000 Network as far as its conservation is concerned by applying the MaxEnt model. The recently collected data justify the classification of N. cordata as "Vulnerable". The most significant variables that affect its distribution were found to be the vegetation type and the precipitation of the warmest quarter. The results of the MaxEnt model suggest that: (i) the Natura 2000 Network is characterised by areas of higher habitat suitability values when compared to the areas that fall outside this network; and, (ii) Picea abies subsp. abies forests are more suitable for its conservation than those of Pinus sylvestris. It is concluded that the most appropriate management measure for the species conservation is the maintenance of the tree layer canopy closed.  相似文献

The oligo-acyl-lysyl, C12(ω7)K-β12, is comprised of only three Lys residues. Despite its small size, it exhibits potent bacteriostatic activity against Gram-positive bacteria, but it is ∼10-fold less potent against Gram-negative bacteria. We followed the interactions of C12(ω7)K-β12 from its initial contact with the bacterial surface across the cell wall down to the cytoplasmic membrane. Binding to anionic lipids, as well as to negatively charged LPS and LTA, occurs with very high affinity. The C12(ω7)K-β12 does not cross the outer membrane of Gram-negative bacteria; rather, it achieves its action by depositing on the LPS layer, promoting surface adhesion and blocking passage of solutes. In Gram-positive bacteria, the thick peptidoglycan layer containing LTA allows passage of C12(ω7)K-β12 and promotes its accumulation in the small periplasm. From that location it is then driven to the membrane by strong electrostatic interactions. Despite its high potency against Gram-positive bacteria, this agent is not capable of efficiently breaking down the permeability barrier of the cytoplasmic membrane or of reaching an intracellular target, as suggested by the fact that it does not interact with DNA.  相似文献

Adhatoda vasica Nees, belonging to family Acanthaceae, is a well-known medicinal plant. It is endorsed for its pyrroloquinazoline alkaloids and its derivatives, such as vasicine and vasicinone. Germinating A. vasica seeds is a tedious task; on that account, vegetative propagation is the preferred method for its multiplication. For rapid and large-scale multiplication, germplasm conservation as well as secondary metabolites production, in vitro culture of A. vasica was preferred over conventional propagation by several researchers; however, some major applications of this tissue culture technique are still awaiting to undergo extensive research. The present review, for the first time, illustrates all the major achievements associated with in vitro regeneration of A. vasica, reported till date and highlights the future prospects.  相似文献

Dactylorhiza hatagirea (D.Don) Soó (Syn. Orchis hatagirea D.Don) is a perennial threatened medicinal plant species, which extends from subalpine to alpine zones (between 2800-4440 m a.s.l.) of the Himalayas. Commonly, it is known as Hatajari and Hatpanja in Uttarakhand, Salam panja in Kashmir, Wanglak or Angulagpa in Ladakh and Panchaunle in Nepal. Due to its high medicinal properties and overexploitation in nature, the species is threatened with extinction. The present study was carried out to assess the population status, distribution patterns and ethno-medicinal uses of D. hatagirea in the Kedarnath Wildlife Sanctuary (KWLS) of Western Himalaya. The vegetation analysis revealed that frequency, density, Importance value index (IVI) and area occupied by the species have declined considerably during recent times, possibly due to general habitat loss, and overexploitation. In order to conserve this species (D. hatagirea) from extinction, its categorization as ‘Critically Endangered’ in the KWLS is suggested. Due to some specific requirements, its efficient conservation becomes more important as well as a challenging task to accomplish. Hence, an integrated conservation approach is needed to conserve this Himalayan orchid.  相似文献
