Bragg and Cobb soybean [Glycine max (L.) Merr.] seeds were germinated in sand at temperatures ranging from 25 to 40°C. Emergence decreased with increasing temperature above 37°C, with virtually no emergence at 40°C. Emergence of 12 other cultivars at 38°C ranged from 25 to 95%. Foster and Coker 338 were more sensitive to high temperature than the other cultivars.  相似文献

Delicious apple (Malus domestica Borkh.) and several of its strains, which have been difficult to root in vitro, were successfully propagated with rooting percentages up to 100%. The combination of treatments used to achieve this result included placing the shoots on rooting medium in the dark at 30°C for the first week of the rooting stage, then moving them to a regime of 16 hr light-8 hr dark at 25°C. The rooting medium contained half strength Murashige and Skoog salts plus 1.2 M thiamine HCl, 0.56 mM myo-inositol, 1 mM phloroglucinol (PG), 1.4 M indolebutyric acid (IBA), 1.3 M gibberellic acid (GA3), 87.6 mM sucrose, and 7 g l–1 Difco Bacto agar. Dark treatment applied during the proliferation stage (etiolation) was less effective than one applied at the beginning of the rooting stage. The optimum length of dark treatment during rooting was 4 to 7 days. Increasing the temperature from 25°C to 30°C improved rooting of Delicious, Royal Red Delicious, and Vermont Spur Delicious in the absence of PG but generally had less effect in the presence of PG. Further increase in temperature to 35°C stimulated rooting of Royal Red Delicious but reduced rooting of Vermont Spur Delicious. Transfer of the cuttings to auxin-free medium after 1 week had no effect on percentage rooting and increased the number of roots per cutting for only 1 of 4 cultivars tested and then only in the presence of PG. In general PG stimulated rooting of Delicious and its strains, but had no effect on Golden Delicious.  相似文献

Summary Crude protein extracts from single seeds of nondomesticated Mexican bean accessions were analysed by SDS polyacrylamide gel electrophoresis for variability in phaseolin protein. Six new phaseolin types; M1, M2, M3, M4, M5, M6, which contained polypeptides within the same range of molecular weights (51,000 to 45,000 daltons) as occur in the S, T and C phaseolin types of cultivated beans were identified. No T and C types were found among the non-domesticated Mexican accessions, and the S type occurred in less than 7% of the seeds screened. Genetic analyses of F2 progenies from crosses between Sanilac (S), and five of the M types showed that each M phaseolin phenotype was allelic to the S type and expressed codominantly.  相似文献

Beans (Phaseolus vulgaris) are regarded as a susceptible crop to suboptimal temperatures. In temperate regions, low temperatures reduce establishment of beans when planted early in the growing season. Seeds of 14 cultivars/lines or beans were germinated in petri dishes at a constant 8, 10, 12, or 18°C or at 12 h alternating temperatures of 10/8, 12/8 or 18/8°C. Differences in germination percentages and rates between cultivars/lines were significant, especially at low temperatures. Cultivars/lines that germinated best and quickly at constant 8°C were Volare, Great Northern (G.N.) Tara, G.N. Belneb # 1, G.N. Spinel, and San Cristobal. Germination percent and rate of Pinto-UI-111 and Canadian Wonder increased significantly when temperatures were increased by 2 to 4°C for 12 h per 24 h, compared with a constant 8°C. Whereas, germination of G.N. Belneb # 1 was reduced. Polyacrylamide gel electrophoresis was used to study the effect of cold treatment on polypeptide patterns of seven cultivars/lines. Seeds were germinated at 18°C constant for 96 h or at 18°C for 48 h followed by 48 h at 2 or 8°C. During cold treatment the synthesis of some polypeptides increased. Volare, G.N. Tara Pinto-UI-111 and Canadian Wonder showed changes in polypeptide patterns, while Alubia-33-1, Michigan 84100 and BAT-1225 showed no changes in polypeptide patterns if compared to the control (96 h at 18°C in the dark). This suggests a likely essential role of these proteins in the development of chilling tolerance.  相似文献

Two seed lots of Calluna vulgaris were obtainedfrom English Nature (seed of Cornish provenance) (EN) and John ChambersWildflower Seeds (JCWS). In laboratory tests, under continuous light untreatedseeds of both seed lots were partially dormant at temperatures between14–35 °C, but JCWS seeds were more deeply dormant thanENseeds. The optimum temperature for germination for both lots was ca 18°C. Germination of EN seeds was much lower in the dark than inthe light at all temperatures; JCWS seeds did not germinate in the dark. In thelight at 22 °C, dormancy of both seed lots was broken whenseeds were incubated in GA4/7 solution(2×10–4 M). Dormancy ofJCWSseeds at 22 °C in the light was broken when seeds wereincubated in four different smoke solutions but more so when used incombinationwith GA4/7. Soaking seeds for 4h insmoke/GA4/7solutions before sowing improved both the speed andpercentage germination in pot experiments on a mist bench in the glasshouse byat least 10-fold. Soaking with GA4/7 alone produced a 5-fold increasein germination but seedlings were more etiolated than with thesmoke/GA4/7 mixtures. A seed advancement treatment modified from thatused commercially on sugar beet seeds also promoted germination in bothlaboratory and glasshouse tests. This entailed soaking seeds in 0.2% thiramsuspension for 4h followed by incubation in excess solution at 22°C for 4 days. This treatment was not as effective as thesmoke/GA4/7 seed soaks.  相似文献

CO2 compenstation point (), the concentration of CO2 at which photosynthesis and respiration are at equilibrium, is a commonly used diagnostic for the C4 photosynthetic pathway, since it reflects the reduced photorespiration that is a property of C4 photosynthesis. Geographic variation for was examined within Flaveria linearis, a C3–C4 intermediate species. Collections from four widely separated Floridian populations were propagated in a greenhouse and measured for . Little differentiation among populations was found, but significant within-population variation was present. Temperature is a hypothesized selective agent for the C4 photosynthetic pathway. To test this hypothesis, plants exhibiting a range of were cloned and placed in growth chambers at 25°C and 40°C. After 7 weeks, valves were remeasured and plants were harvested and weighed. There was a poor correlation between initial and final measures of for a given genotype (r=0.38, P>0.1). Broad sense heritability for was computed to be 0.10. At 25°C, there was no relationship between final size and . At 40°C, more C4-like plants, as indicated by their low , had grown larger. Differences in relative growth rate were attributable more to differences in net assimilation rate than in leaf area ratio. Taken together, these results demonstrate that although significant plasticity exists in the amount of photorespiration in this C3–C4 species, high temperature appears to be an effective selective agent for the reduction of photorespiration and the enhancement of C4-like traits.  相似文献

Seed maturation of Pisum sativum cv. Progress No. 9 proceeds more slowly in winter than in summer even when the parent plants are grown in greenhouse conditions with light-and heat-supplementation. For parent plants grown under summer and winter conditions the metabolism of [3H]GA9 in cultured seeds is qualitatively different in seeds of equivalent age and qualitatively the same in seeds of equivalent weight. 13-Hydroxylation of [3H]GA9[3H]GA20 is restricted to early stages of seed development. 2-Hydroxylation of [3H]GA92-OH-[3H]GA9 has only been observed at a stage of development after endogenous GA9 has accumulated. 2-OH-GA9 has been shown to be endogenous to pea and is named GA51. H2-GA31 and its conjugate have not been shown to be present in pea and may be induced metabolites of [3H]GA9. The metabolism of GA20GA29 is used to illustrate a technique of feeding [2H][3H]GAs in order to distinguish a metabolite from the same endogenous compound. The in vitro conversion of [3H]GA20[3H]GA29, and the virtual non-metabolism of [3H]GA29 have been confirmed for seeds in intact fruits. The production of erythritol and the erythritol yield from glucose by Torula sp. were improved, in increasing order, by supplementing with 10 mg MnSO44H2O l–1, 2 mg CuSO45H2O l–1, and both 10 mg MnSO44H2O l–1 and 2 mg CuSO45H2O l–1. Mn2+ decreased the intracellular concentration of erythritol, whereas Cu2+ increased the activity of erythrose reductase in cells. These results suggest that Mn2+ altered the permeability of cells, whereas Cu2+ increased the activity of erythrose reductase in cells.  相似文献

The sensitivity to gibberellic acid (GA3) of aleurone protoplasts isolated from a single harvest of an inbred line of Avena fatua seed that had been after-ripened over anhydrous CaCl2 at 25±2°C and 4±2°C for three years was assessed. Protoplasts isolated from aleurones of seed stored at 25°C produced substantially more -amylase in response to 10–7 M GA3 than those isolated from aleurones of seed stored at 4°C. The apparent difference in responsiveness does not appear to be due to a change in the duration of the lag phase between addition of GA3 and the production of -amylase. The dose response of aleurone protoplasts to GA3, measured as -amylase production, is complex and appears to have three phases. Protoplasts from seed stored at both temperatures respond appreciably to 10–14 M GA3. With increasing concentrations of GA3, up to 10–9 M, -amylase production increases similarly in protoplasts from both lots of seed, reaching a level approximately 2.7–3.8 times greater than when no GA3 is applied. GA3-induced -amylase production increases markedly as the concentration is raised from 10–9 M to 10–6 M, and the response then appears to be saturated. Over this part of the response curve protoplasts from the two seed lots differ markedly in their responsiveness to GA3. Those from seed stored at 25°C produce considerably more -amylase, >130-fold higher than the minus GA3 control, than those from seed stored at 4°C, <35-fold higher than the minus GA3 control. This apparent difference in the responsiveness of aleurone protoplasts to GA3 could be correlated with the loss of embryo dormancy in seed stored at 25°C. Seed stored at 4°C retained the dormancy characteristics present immediately after harvesting.  相似文献

The metabolism of GA29 in maturing seeds of Pisum sativum cv. Progress No. 9 was further investigated, and the utility of 2H-labelled GAs in conjuction with GC-MS is illustrated. Using [2-2H1]GA29 as an internal standard, endogenous GA29 was shown to reach a maximal level (ca. 10 g/seed) 27 days from anthesis, and to decline to ca. 1.6 g/seed in mature seeds. In a time-course feed the metabolism of [2-2H1] [2-3H1]GA29 applied to 27 day old seeds, and of endogenous GA29, was compared from the 1H:2H ratios in the recovered GA29. Although both [2-2H1] [2-3H1]GA29 and endogenous GA29 were metabolised to the same limited extent to a putative conjugate, in the main metabolic process endogenous GA29 was preferentially converted to an untraceable (i.e. unlabelled) metabolite. In contrast, endogenous GA29 and [1,3-2H2] [1,3-3H2]GA29, derived from [1,3-2H2] [1,3-3H2]GA20 in a time-course feed, were metabolised in an identical manner. In the latter case isotope loss precluded identification of the metabolite. The structure (8) has been assigned to a GA catabolite present in maturing seeds and seedlings of pea. The response of effective quantum yield of photosystem 2 (F/Fm) to temperature was investigated under field conditions (1 950 m a.s.l.) in three alpine plant species with contrasting leaf temperature climates. The in situ temperature response did not follow an optimum curve but under saturating irradiances [PPFD >800 µìmol(photon) m–2s–1] highest F/Fm occurred at leaf temperatures below 10°C. This was comparable to the temperature response of antarctic vascular plants. Leaf temperatures between 0 and 15°C were the most frequently (41 to 56%) experienced by the investigated species. At these temperatures, F/Fm was highest in all species (data from all irradiation classes included) but the species differed in the temperature at which F/Fm dropped below 50% (Soldanella pusilla >20°C, Loiseleuria procumbens >25°C, and Saxifraga paniculata >40°C). The in situ response of F/Fm showed significantly higher F/Fm values at saturating PPFD for the species growing in full sunlight (S. paniculata and L. procumbens) than for S. pusilla growing under more moderate PPFD. The effect of increasing PPFD on F/Fm, for a given leaf temperature, was most pronounced in S. pusilla. Despite the broad diurnal leaf temperature amplitude of alpine environments, only in S. paniculata did saturating PPFD occur over a broad range of leaf temperatures (43 K). In the other two species it was half of that (around 20 K). This indicates that the setting of environmental scenarios (leaf temperature×PPFD) in laboratory experiments often likely exceeds the actual environmental demand in the field.This revised version was published online in March 2005 with corrections to the page numbers.  相似文献

A rapid procedure is described for the separation of CMP-sialic acid:lactosylceramide sialyltransferase reaction components using Sep Pak C18 cartridges. The quantitative separation of the more polar nucleotide sugar, CMP-sialic acid, and its free acid from the less polar GM3-ganglioside is simple and rapid relative to previously described methods. Recovery of GM3 is optimized by the addition of phosphatidylcholine to the reaction mixture prior to the chromatographic step. Using rat liver Golgi membranes as a source of CMP-sialic acid: lactosylceramide sialyltransferase activity (GM3 synthase; ST-1), the transfer of [14C] sialic acid from CMP-[14C] sialic acid to lactosylceramide can be quantified by this assay. The 500-MHz1H-NMR characteristics of theN-linked carbohydrate chain Man1-6[Xyl1-2]Man1-4GlcNAc1-4[Fuc1-3]GlcNAc1-NAsn of the proteolytic enzyme bromelain (EC 3.4.22.4) from pineapple stem were determined for the oligosaccharide-alditol and the glycopeptide, obtained by hydrazinolysis and Pronase digestion, respectively. The seed storage globulins from sixHelianthus and four hybrids were studied using mono and bidimensional gel SDS electrophoresis (+ 2 mercaptoethanol). The polypeptide composition of each subunit was determined. Different pairs are specifically expressed according to the species studied. Three typical patterns were discriminated. All the studied species exhibit five subunits: two of them are expressed in all the species (11 and 22). The subunit corresponding to the 11 pair is present inH. petiolaris and in the three populations ofH. annuus studied. The 2b2 pair is common toH. annuus andH. argophyllus. H. petiolaris presents two specific 2a2 and 44 pairs andH. annuus a specific 33 pair. InH. argophyllus 11 33 or 44 are never observed but are replaced by 13 and 31 pairs. Some globulins, poorly represented, are of forms but present chains of higher molecular weights (in the range 54–56 kDa). Expressing variations in the banding patterns between these species by the use of a similarity index reveals complete identity between the three populations ofH. annuus. Identity between the twoH. petiolaris studied is also observed.H. annuus andH. argophyllus appear to be closer to each other thanH. petiolaris concerning the seed storage globulins.  相似文献

From June 1994 to October 1995, I conducted a study of the feeding ecology of one group of black-headed uacaris (Cacajao melanocephalus melanocephalus) in Pico da Neblina National Park, Brazil, near the Venezuelan border (01°10N to 00°26S, 65°03W to 66°52W). Of a total of 429 feeding records, 89% corresponded to fruits from which black-headed uacaris ate ripe and unripe seeds, mesocarps and arils. Seeds were the single most important food item year-around, corresponding to 81% of the feeding records for fruits. Black-headed uacaris used 120 different plant species in 32 families as food. The 3 most important plant species in their diet, Micrandra spruceana, Eperua leucantha, and Hevea braziliensis (all trees), accounted for 38% of the feeding records for fruits. These species produced fruits that were protected by very hard green husks, were barochorous, did not have fleshy mesocarps, and were slow to mature, thus being available as unripe fruits for long periods. The second and third most important species in the diet of black uacaris, Eperua leucantha and Hevea braziliensis, were dominant in the forest: approximately 30% of trees 10 cm dbh sampled in a 2-ha botanical transect at the study site. I discuss the formation of large social groups of black-headed uacaris in Pico da Neblina National Park in light of the peculiar floristic composition of the study site.  相似文献

Haustoria of two palm species, Phoenix dactylifera L. (date) and Washingtonia filifera (Lindl.) Wendl were carefully dissected from seeds, and the ultrastructural characteristics of the large, electron-opaque granules present in the cells of this tissue were compared using standard aldehyde and OsO4 fixations. In Washingtonia, the granules were smaller than those in date and were more variable in size and presence of non-opaque inclusions. All granules appeared to be membrane bounded although they often filled the bounded space. No protein, lipid, carbohydrate or tannins were found in the granules by standard staining procedures. The granules stained positively with two different metallic-phosphate stains which contained either bismuth or lead. Energy dispersive X-ray microprobe analysis, done on aldehyde-fixed granules and those stained with both phosphate stains, confirmed the fact that phosphorus and calcium were present in the granules. The granules also bound the metallic stains as expected. All procedures consistently confirmed the presence of phosphate in the granules. Summary A set of three 3D (1H, 13C, 15N) triple-resonance correlation experiments has been designed to provide H1-H8 intraresidue sugar-to-base correlations in purines in an unambiguous and efficient manner. Together, the HsCsNb, HsCs(N)bCb, and HbNbCb experiments correlate the H1 sugar proton to the H8 proton of the attached base by means of the {H1, C1, N9, C8, H8} heteronuclear scalar coupling network. The assignment strategy presented here allows for unambiguous H1-H8 intraresidue correlations, provided that no two purines have both the same H1 and C1 chemical shifts and the same C8 and N9 chemical shifts. These experiments have yielded H1-H8 intraresidue sugar-to-base correlations for all five guanosines in the [13C, 15N] isotopically labeled RNA duplex r(GGCGCUUGCGUC)2.  相似文献

The proportion of spurs flowering on apple trees (Malus domestica Borkh. cv Golden Delicious) displaying a high degree of alternate-year flowering was increased in the off year by gibberellin A4 (GA4) and C-3 epi-GA4 applied in the previous year. When applied 4.5 weeks after anthesis amounts of GA4 ranging from 3 to 300 g per spur and 25 or 50 g of C-3 epi-GA4 per spur were effective. Treatments with GA4 made seven weeks after anthesis were less effective. The growth and flowering response of a cold-requiring cauliflower (Brassica oleracea var. botrytis cv. 60 day) to a range of temperatures under 10 h photoperiod and to growth regulator application were investigated. Endogenous gibberellin A1(GA1) concentrations were also assessed under these treatments. Flowering and growth of the inflorescence stalk were correlated with plant developmental stage at the time of a vernalizing cold treatment. Temperature and its duration also affected flowering and inflorescence development. The most effective temperature for inflorescence induction was 10 °C. Flowering did not occur in non-vernalized plants (25 °C) even though they had been treated with GA3. Application of GA3 promoted inflorescence stalk elongation greatly in vernalized plants (10 °C), but less so in partially vernalized plants (15 °C or 20 °C). Paclobutrazol (PP333) sprayed at the 8–9 leaf stage significantly suppressed inflorescence stalk length and slightly delayed flower bud formation and anthesis. Vernalization at 10 °C increased endogenous GA1 content in both leaves and the inflorescence stalk irrespective of GA3 or PP333 treatment. Application of GA3 tended to increase GA1 levels, while PP333 significantly reduce GA1, both irrespective of vernalization. Vernalization is an important factor for flowering, but not curd formation in this cauliflower cv. 60 day and GA1 is likely a causal factor in inflorescence stalk elongation.  相似文献

Summary Seeds ofVerbascum thapsus were imbibed on filter papers in Petri dishes and were exposed to a series of temperatures of 4°C10°C20°C10°C4°C in darkness. After each temperature treatment lasting one month, samples were removed and tested in light and darkness over a range of alternating temperatures. The results showed us that the dormancy state changes gradually according to the imbibition temperatures simulating the seasonal cycle. The ecological implications of the dormancy phases for the regulation of germination in natural conditions are discussed.  相似文献
