Summary Six populations — P1,P2,F1,F2,B1 and B2 — each of five Upland Cotton (Gossypium hirsutum L.) crosses were used to evaluate gene effects in the inheritance of fibre properties by Gamble's six-parameter model for the analysis of generation means. Partial dominance of long fibres over short fibres and of mature fibres over immature fibres was found in the material studied. Overdominance in gene action governed fibre fineness while additive gene action governed the fibre strength. Besides additive and dominance effects, significant epistasis was present in all crosses. These results indicate a significant potential for improving fibre properties through reciprocal recurrent selection.  相似文献

Capillarity, fibre types, fibre area and enzyme activities of different skeletal muscles (pectoralis, extensor digitorum longus), tibialis anterior, plantaris and the myocardium were compared in Andean coot (Fulica americana peruviana) native to high altitude (Junín, Perú, 4200 m) and the same species nesting at sea level. Numbers of capillaries per square millimeter were higher in all high-altitude muscles when compared with sea-level muscles (P<0.0001). Moreover, values for capillaries per fibre and capillaries in contact with each fibre were higher in digitorum and tibialis high-altitude muscles. Muscle fibres were classified as Type I, Type IIA or Type IIB on the basis of their myofibrillar ATPase pH lability. Pectoralis muscle of high-altitude and sea-level coots presented only fibres of Type IIA. In contrast, all the leg muscles studied showed a mosaic pattern of the three fibre types. Fibre areas were determined using a Leitz Texture Analysis System. Significant differences in fibre area were observed (P<0.01) between high-altitude and sea-level muscles. Mean muscle fibre diameters were also lower in the high-altitude group than in the sea-level group. The enzyme activities studied were hexokinase, lactate dehydrogenase, citrate synthase and 3-hydroxyacyl-CoA-dehydrogenase. The oxidative capacity, as reflected by citrate synthetase and hydroxyacyl-CoA-dehydrogenase activities, was greater for myocardial and pectoralis than for leg muscles. However, analysis of maximal enzyme activities showed that there were no significant differences between the glycolytic and oxidative enzyme activities of high-altitude and sea-level coots. Summary Mammalian intrafusal fibre types (nuclear chain, nuclear bag1 and nuclear bag2 fibres) are known to differ in their ultrastructure, intensity of the myofibrillar histochemical ATP-ase reaction, type of innervation and time course of contraction. The present study concerns the myosin composition of these intrafusal fibre types in the soleus muscle (mouse) and the extensor digitorum longus muscle (rat). We used an immunohistochemical method with three myosin antisera raised in rabbits: anti chicken pectoral myosin, anti chicken heart myosin (1) and anti chicken heart myosin (2) (=anti chicken heart myosin (1) adsorbed with muscle powder from soleus muscle of guinea pig). The results showed that three intrafusal fibre types differed in their myosin composition. A comparison of intrafusal fibre types with extrafusal fibre types for the histochemical myofibrillar ATP-ase reactivity and the reactivity with myosin antisera showed a resemblance of nuclear chain fibres with extrafusal type II fibres and a difference between nuclear bag1 and nuclear bag2 fibres and all other fibre types.  相似文献

The changes in flax straw structure when treated with sulphur dioxide (SO2) and consequent effect on dew-retting of the treated straw were investigated. The active oxoanions released by sulfurous acid degraded phenolics bound to the polysaccharides and lipids leading to an increase in water-soluble carbohydrates and polyphenols. As a result, proportions of hemicellulose, nitrogen, sulphur, ash and caustic weight loss were higher in treated straw while lipid was reduced. The coarseness of treated fibre was possibly caused by in situ dryback of the hydrolysed polysaccharides. The deposition of salts containing high concentrations of sulphur, calcium and potassium in the vicinity of the fibre bundles and epidermal layer was observed and investigated. In a dew-retting trial comprising SO2-treated and untreated samples, the SO2 treatment delayed microbial colonisation and, consequently, retting by nearly 2 wk compared to the controls. The changes in pH, fungal population and polysaccharide degrading enzymes were monitored. The differences in fibre yield, fineness and strength are presented.  相似文献

Molecular markers linked to QTL contributing to agronomic and fibre quality traits would be useful for cotton improvement. We have attempted to tag yield and fibre quality traits with AFLP and SSR markers using F2 and F3 populations of a cross between two Gossypium hirsutum varieties, PS56-4 and RS2013. Out of 50 AFLP primer combinations and 177 SSR primer pairs tested, 32 AFLP and four SSR primers were chosen for genotyping F2 individuals. Marker-trait associations were studied for eight agronomic and five fibre quality traits through simple and multiple regression analysis (MRA) using a set of 92 AFLP polymorphic loci and four SSR markers. Simple linear regression analysis (SLRA) identified 23 markers for eight different traits whereas multiple regression analysis identified 30 markers for at least one of the 13 traits. SSR marker BNL 3502 was consistently identified to be associated with fibre strength. While all the markers identified in SLRA were also detected in MRA, as many as 16 of the 30 markers were identified to be associated with respective traits in both F2 and F3 generations. The markers explained up to 41 per cent of phenotypic variation for individual traits. A number of markers were found to be associated with multiple traits suggesting clustering of QTLs for fibre quality traits in cotton.  相似文献

The effects of feeding two fibre‐rich diets with contrasting solubility and a concentrated low dietary fibre on the rate of gastric emptying were examined in six gastric cannulated pregnant sows. Additionally, it was examined whether any effect could be related to the physico‐chemical properties of digesta, i.e. viscosity and/or water binding capacity.

The sows were fed each diet for one week in a 3 x 3 Latin Square design and the samples were taken in a randomised order 0.5, 1, 2, 3, 5 and 15.5 h after the morning meal. The stomach contents were evacuated through the gastric cannula once daily. The evacuated gastric digesta was quantified and a representative sample was taken to determine its viscosity, water binding capacity and its content of dry matter, dietary components and solid (Cr2O3) and liquid (polyethylene glycol) phase markers. The flow of liquid digesta was calculated as the difference between digesta and dry matter.

Increasing the content of dietary fibre in the diet led to higher recovery of liquid digesta but did not have any significant effect on the gastric emptying of dry matter and dietary components. The effect of dietary fibre could not be attributed to the viscosity of the liquid phase of digesta but might be related to the ability of the increased gastric dietary fibre content to hold water. The stomach selectively retained the insoluble dietary fibre components most noticeably seen with the bran‐supplemented diet where the concentration of insoluble NSP in digesta increased significantly from 2 hours and onward.  相似文献

Summary Optical sensors suitable for monitoring of O2 in water medium were prepared by immobilization of metallo-organic complexes in xerogels created by sol-gel technique and applied to optical fibre tip or glass slides by dipping. Tris (1,10-phenanthroline)ruthenium (II) chloride was immobilized on the tip of optical fibre. The fluorescence spectra of the complex were sensitive to O2 in water solution. The sensitivity of absorption spectra of bis(histidinato)cobalt (II) to O2 and pH was examinated both in the solution of precursors of silica sol-gel glasses and in the complex-containing coating of glass slides.  相似文献

Atlantic salmon Salmo salar eggs derived from a single family were incubated at two different water temperature regimes, with a mean temperature between fertilization and first feeding differing between 6 and 10° C (HT) and 2–6° C (LT). From first feed the fry were kept under the same rearing conditions and fed either high (50%) or low (45%) protein diet level of equivalent energy content until smoltification. All treatments were carried out in duplicate tanks. At first feeding the groups were similar in mass, but thereafter the HT‐fish were heavier and longer compared to the LT‐fish throughout the experiment. The groups fed the high protein diet were significantly heavier and longer compared with the corresponding low protein diet. A strong positive relationship was observed between LF and total white muscle cross‐sectional area (CSA), white muscle fibre diameter and fibre number. There were also equivalent relationships with body mass. There were no significant differences in CSA, the mean diameter or the number of white muscle fibres per CSA between groups at first feed. Muscle fibre number and CSA increased in all groups during the experiment, whereas fibre diameter reached a plateau when the fish reached > 9 cm LF. There were only minor effects of pre‐hatch and yolk sac stage temperature on CSA and fibre number per CSA during the juvenile stage. In short periods the LT‐group had larger CSA and higher fibre number than the HT‐groups, but this differences had disappeared by the end of the juvenile stage. No differences in mean fibre diameter were found between groups, except at the time of smoltification. When the fish approached smoltification a decrease in mean fibre diameter and an increase in muscle fibres <25 µm was seen and taken as an indication of recruitment of new fibres (hyperplasia). Only minor differences in CSA, fibre number or fibre diameter was observed between high and low protein diet groups.  相似文献

The aim of the present study was to determine equations that predict ME in total mixed rations (TMR) based on routine methods. The ME content of 30 TMR for dairy cows was determined based on digestible crude nutrients obtained with wether sheep. Concentrations in the TMR (in g/kg DM) varied between 118 and 234 for crude protein, 26 and 48 for crude lipid, 131 and 250 for crude fibre, 281 and 488 for NDF, and 173 and 304 for ADF. Gas production ranged from 40.7 to 54.1 ml/200 mg DM, and enzymatically degraded organic matter from 652 to 800 g/kg DM. Digestibility [%] ranged from 68.6 to 84.0 for organic matter, from 55.6 to 84.3 for crude lipid, from 55.0 to 77.8 for crude fibre, from 57.6 to 77.0 for NDF and from 53.1 to 79.6 for ADF. ME ranged from 9.6 to 11.9 MJ/kg DM, and NEL from 5.7 to 7.4 MJ/kg DM. ME content was highly correlated with the concentration of both crude fibre and enzymatically degradable organic matter as well as with organic matter digestibility. A multiple regression equation based on crude fibre and crude lipid predicted ME with a reasonable goodness of fit (r2 = 0.81; sy.x = 2.4%). The inclusion of other nutrients, of neutral and acid detergent fibre, neither of gas production did improve the goodness of fit. The best prediction was achieved with inclusion of enzymatically degraded organic matter (r2 = 0.90; sy.x = 1.7%).  相似文献

Summary The iliofibularis muscle ofXenopus laevis is reported to contain five types of fibres which have different force—velocity relationships. Ten fibres of each type were selected on the basis of succinate dehydrogenase activity, cross-sectional area and location in the muscle, in order to assess the validity of the fibre type classification.Maximum calcium-stimulated myofibrillar ATPase activity (V max) and apparent Michaelis constant (K m) for ATP were determined for these 50 fibres from serial sections. The values obtained varied according to the type of fibre. Type 1 had the highest and type 5 the lowest values forK m andV max.In a separate experiment, single freeze-dried fibres were used to determine the relationship between their ATP content and apparentK m for ATP. There was a tendency for high ATP concentrations in fibres with highK m values.When myofibrillar ATPase activity was related to the maximum velocity of shortening of the five fibre types, a significant correlation was found. It is concluded that calcium-stimulated myofibrillar ATPase histochemistry allows an estimate of the maximum shortening velocity of muscle fibres fromXenopus laevis.  相似文献

Background

It is hypothesized that decreased capillarization of limb skeletal muscle is implicated in the decreased exercise tolerance in COPD patients. We have recently demonstrated decreased number of capillaries per muscle fibre (CAF) but no changes in CAF in relation to fibre area (CAFA), which is based on the diffusion distance between the capillary and muscle fibre. The aim of the current study is to investigate the muscle-to-capillary interface which is an important factor involved in oxygen supply to the muscle that has previously been suggested to be a more sensitive marker for changes in the capillary bed compared to CAF and CAFA.

Methods

23 COPD patients and 12 age-matched healthy subjects participated in the study. Muscle-to-capillary interface was assessed in muscle biopsies from the tibialis anterior muscle using the following parameters:1) The capillary-to-fibre ratio (C:Fi) which is defined as the sum of the fractional contributions of all capillary contacts around the fibre2) The ratio between C:Fi and the fibre perimeter (CFPE-index)3) The ratio between length of capillary and fibre perimeter (LC/PF) which is also referred to as the index of tortuosity.Exercise capacity was determined using the 6-min walking test.

Results

A positive correlation was found between CFPE-index and ascending disease severity with CFPE-index for type I fibres being significantly lower in patients with moderate and severe COPD. Furthermore, a positive correlation was observed between exercise capacity and CFPE-index for both type I and type IIa fibres.

Conclusion

It can be concluded that the muscle-to-capillary interface is disturbed in the tibialis anterior muscle in patients with COPD and that interface is strongly correlated to increased disease severity and to decreased exercise capacity in this patient group.  相似文献

Background

Airway remodelling is a feature of asthma including fragmentation of elastic fibres observed in the superficial elastin network of the airway wall. Fibered confocal fluorescence microscopy (FCFM) is a new and non-invasive imaging technique performed during bronchoscopy that may visualize elastic fibres, as shown by in vitro spectral analysis of elastin powder. We hypothesized that FCFM images capture in vivo elastic fibre patterns within the airway wall and that such patterns correspond with airway histology. We aimed to establish the concordance between the bronchial elastic fibre pattern in histology and FCFM. Second, we examined whether elastic fibre patterns in histology and FCFM were different between asthmatic subjects and healthy controls. Finally, the association between these patterns and lung function parameters was investigated.

Methods

In a cross-sectional study comprising 16 subjects (8 atopic asthmatic patients with controlled disease and 8 healthy controls) spirometry and bronchoscopy were performed, with recording of FCFM images followed by endobronchial biopsy at the airway main carina. Elastic fibre patterns in histological sections and FCFM images were scored semi-quantitatively. Agreement between histology and FCFM was analysed using linearly weighted kappa κw.

Results

The patterns observed in histological sections and FCFM images could be divided into 3 distinct groups. There was good agreement between elastic fibre patterns in histology and FCFM patterns (κw 0.744). The semi-quantitative pattern scores were not different between asthmatic patients and controls. Notably, there was a significant difference in post-bronchodilator FEV1 %predicted between the different patterns by histology (p = 0.001) and FCFM (p = 0.048), regardless of asthma or atopy.

Conclusion

FCFM captures the elastic fibre pattern within the airway wall in humans in vivo. The association between post-bronchodilator FEV1 %predicted and both histological and FCFM elastic fibre patterns points towards a structure-function relationship between extracellular matrix in the airway wall and lung function.

Cotton provides us the most important natural fibre. High fibre quality is the major goal of cotton breeding, and introducing genes conferring longer, finer and stronger fibre from Gossypium barbadense to Gossypium hirsutum is an important breeding strategy. We previously analysed the G. barbadense fibre development mechanism by gene expression profiling and found two homoeologous fibre‐specific α‐expansins from G. barbadense, GbEXPA2 and GbEXPATR. GbEXPA2 (from the DT genome) is a classical α‐expansin, while its homoeolog, GbEXPATR (AT genome), encodes a truncated protein lacking the normal C‐terminal polysaccharide‐binding domain of other α‐expansins and is specifically expressed in G. barbadense. Silencing EXPA in G. hirsutum induced shorter fibres with thicker cell walls. GbEXPA2 overexpression in G. hirsutum had no effect on mature fibre length, but produced fibres with a slightly thicker wall and increased crystalline cellulose content. Interestingly, GbEXPATR overexpression resulted in longer, finer and stronger fibres coupled with significantly thinner cell walls. The longer and thinner fibre was associated with lower expression of a number of secondary wall‐associated genes, especially chitinase‐like genes, and walls with lower cellulose levels but higher noncellulosic polysaccharides which advocated that a delay in the transition to secondary wall synthesis might be responsible for better fibre. In conclusion, we propose that α‐expansins play a critical role in fibre development by loosening the cell wall; furthermore, a truncated form, GbEXPATR, has a more dramatic effect through reorganizing secondary wall synthesis and metabolism and should be a candidate gene for developing G. hirsutum cultivars with superior fibre quality.  相似文献

The crystal and molecular structure of a tri-O-ethylamylose polymorph, TEA 3, has been solved by stereochemical conformation and packing analysis, combined with X-ray fibre diffraction analysis. The unit cell is orthorhombic, space group P212121, with a  15.36 (±0.03) Å, b  12.18 (±0.05) Å, and c (fibre repeat)  15.48 (±0.01) Å. The actual chain conformation is a 43 helix with the EtO-6 group in the tg position, as was found in the polymorph TEA 1.  相似文献

Electron-microscopic examination in conjunction with extraction procedures and chemical analysis have confirmed that a suberin-like lipid biopolymer is located within the concentric polylamellate layers found in the secondary cell walls of green cotton fibres (Gossypium hirsutum cv. green lint). A polymer of similar ultrastructure and chemical constitution also occurs mainly in the secondary seed-coat walls of the outer epidermis of both green and white varieties of G. hirsutum. The suberins composed of predominantly C22 compounds are, however, markedly different from those present in the periderms of the same plants; these comprise mainly C16 and C18 compounds. Long-chain 1-alkanols (C26–C36) and alkanoic acids (C16–C36) are the principal components of the wax from white fibres but these lipid classes comprise a much smaller proportion of that from green fibres. unidentified highmolecular-weight compounds were the major constituents of the green-fibre was extract which also contains a number of yellow-green pigments, probably flavonoid in nature. These pigments are thought to be associated with the ultrahistochemical reaction with silver proteinate that was observed only in the green-fibre cell walls. A total of 16 wild and cultivated cotton species were examined with the electron microscope for the presence of suberin. The outer seed-coat epidermis of all the examined species but only the fibres of the wild ones were found to be suberized. Functionalization of rayon fibre has been carried out by grafting acrylic acid (AAC) both by a chemical method using a Ce4+-HNO3 redox initiator and by a mutual irradiation (γ-rays) method. The reaction conditions affecting the grafting percentage have been optimized for both methods, and the results are compared. The maximum percentage of grafting (50%) by the chemical method was obtained utilizing 18.24 × 10−3 moles/L of ceric ammonium nitrate (CAN), 39.68 × 10−2 moles/L of HNO3, and 104.08 × 10−2 moles/L of AAc in 20 mL of water at 45 °C for120 min. For the radiation method, the maximum grafting percentage (60%) was higher, and the product was obtained under milder reaction conditions using a lower concentration of AAc (69.38 × 10−2 moles/L) in 10 mL of water at an optimum total dose of 0.932 kGy. Swelling studies showed higher swelling for the grafted rayon fibre in water (854.54%) as compared to the pristine fibre (407%), while dye uptake studies revealed poor uptake of the dye (crystal violet) by the grafted fibre in comparison with the pristine fibre. The graft copolymers were characterized by IR, TGA, and scanning electron micrographic methods. Grafted fibre, prepared by the radiation-induced method, showed better thermal behaviour. Comparison of the two methods revealed that the radiation method of grafting of acrylic acid onto rayon fibre is a better method of grafting in comparison with the chemical method.  相似文献

This paper presents methods for the stereological analysis of spatial fibre systems on the base of planar or thin sections. Under the assumption that the cross-section figures of the tubular fibres can be measured, the orientation distribution of the fibre system and its line density Lv can be determined from one section only and without distributional assumptions. A simple way to study the degree of randomness of fibre systems consists in the statistical analysis of the point pattern of centres of intersection figures. More sophisticated methods are of stereological nature and yield the spatial reduced second moment measure. Similarly also correlations between two fibre systems can be quantified. The methods are demonstrated by two examples concerning samples of human brain.  相似文献

Summary Tench (Tinca tinca) were acclimated to either aerated (P O 2 17.6 KPa) or hypoxic water (P O 2 1.5 KPa) at 15° C. Fish acclimated to P O 2 17.6 KPa had a routine oxygen consumption (mls O2/Kg bodyweight/h) of 32.7 in aerated water. Upon acute exposure to P O 2 1.5 KPa oxygen consumption decreased to 10.8 and 15.6 in fish acclimated to aerated and hypoxic water, respectively.On the basis of staining for glycogen and for the activities of myofibrillar ATPase and succinic dehydrogenase, three main fibre types can be differentiated in the myotomal muscle.Fibres have been classified as slow, fast aerobic and fast glycolytic. Fast aerobic fibres can be distinguished histochemically by their alkaline-stable Ca2+-activated myofibrillar ATPase activity and their intermediate levels of staining for glycogen and succinic dehydrogenase activity.The patterns of innervation of the fibre types have been investigated by staining neuromuscular endplates and peripheral axons for acetylcholinesterase activity. Motor axons to slow fibres branch extensively giving rise to a number of diffuse endplate formations on the same and adjacent fibres. Fast glycolytic fibres also have a complex pattern of innervation with 8–20 endplates per fibre. A large proportion of the endplates belonged to separate axons.Cross-sectional areas and perimeters of fibres, the number of capillaries/fibre and the lengths of contacts between capillaries and fibres were determined from low-magnification electron micrographs.Acclimation to hypoxia resulted in a decrease in the number of capillaries per fibre for both slow (1.8 to 1.0) and fast (0.8 to 0.2) muscles. The capillary perimeter supplying 1 m2 of fibre cross-sectional area decreased by 43 % and 76 % for slow and fast fibres, respectively.  相似文献

Summary The mitochondria in type-I and -II muscle fibres in the pectoralis major muscle of the pigeon (Columba livia) have been analysed using stereological techniques not previously applied in muscle biology.Mitochondrial volume fractions (VV) were estimated in different regions of each type of muscle fibre using randomly orientated sampling sectors within fibre profiles. These sectors were sub-divided into smaller sampling regions to provide accurate data on the intracellular distribution of mitochondria. Estimates of the external surface densities of mitochondria per unit volume of fibre, SV total surface , and also the densities of mitochondrial cristae, SV total cristae , were obtained using a specific technique derived for analysing anisotropic structures (Saltykov, 1958). The relative amounts of the random and orientated mitochondrial membranes were also estimated.Significiant differences were found to exist between the different types of muscle fibres and considerable though constant variations in the intracellular arrangement of mitochondria were also found.  相似文献
