THE RELATIONSHIP OF GIBBERELLIN AND AUXIN IN PLANT GROWTH   总被引:5,自引:0,他引:5  
No synergism was found between IAA and gibberellin in the Avenucurvature test and this bioassay thus measures changes in diffusibleauxin resulting from gibberellin treatment and not a synergisticaction of the gibberellin on the curvature response to auxin.Gibberellin treatment causes an increase in diffusible auxinfrom the stem apex of dwarf pea (Pisum sativum L. var. LittleMarvel) 24 to 48 hours before the elongation response in thestem. The increase in diffusible auxin in the stem apex of Centaureacyanus L. var. Blue Boy occurs four to six days before the boltingresponse to gibberellin treatment under short days. The stemtissues of both the dwarf pea and Centaurea show an elongationresponse to IAA when the IAA is applied in a manner simulatingthe stem apex. Gibberellin-like substances and an auxin similar to IAA weredetected by bioassays in extracts of flowers of Chrysanthemummorifolium. The activity of these substances was shown to reacha maximum early in the development of the flower when its relativegrowth-rate was at a maximum, and then to decline with the relativegrowth-rate. The leaves of lateral flowering shoots were found to containgibberellins similar to those detected in the flowers whilea different gibberellin, which appeared to decrease in activitywith the age of the shoot, was detected in the stem. An auxinsimilar to indol-3yl-acetic acid (IAA) was also detected inthese stems. Growth-promoting substances were not detected inthe old stems and leaves from the main shoot. Gas-liquid chromatographyrevealed the presence of a number of additional gibberellinsin the flowers. The chemical nature of the growth substances is discussed inrelation to their biological and chromatographic behaviour.  相似文献

A synthetic brassinosteroid, 22,23(S,S)-homobrassinolide (hBR),was examined for its interaction with IAA and GA3 in the elongationof hypocotyl sections of light-grown cucumber (Cucumis salivusL. cv. Aonagajibai) seedlings. hBR alone was less active thanIAA. Its optimal concentration was around 10 µM and thelowest effective concentration between 10 and 100 µM,which is more than 100 times higher than that of brassinolide.hBR was more active in sections from younger seedlings. Itsgrowth-promoting effect was negated or greatly reduced by inhibitorsof auxin-induced elongation such as p-chlorophenoxyisobutyricacid and kinetin. hBR acted synergistically with IAA and 2,4-Dbut not with GA3 showing only an additive effect. Sequentialtreatment of sections with hBR and then with IAA also resultedin synergistic enhancement of auxininduced elongation, but whenthe order of treatment was reversed, hBR was inactive. The synergisticeffect was obtained with 1 h pretreatment with hBR and couldbe reduced by subsequent washing with water. There was no sequentialinteraction between hBR and GA3. The synergistic pretreatmenteffects of hBR and GA3 were simply additive to each other. Amembrane-bound ATPase inhibitor, dicyclohexylcarbodiimide, inhibitedthe hBR-induced elongation, but did not affect GA3-induced elongation.The findings led to the conclusion that brassinosteroids enhanceauxin action and possess growth-promoting activity which isindependent of that of gibberellin. Effects of uniconazole-P, a triazole-type growth retardant,on endogenous levels of IAA, cytokinins, ABA and ethylene inrice seedlings were investigated. Endogenous levels of IAA andABA were similar between control and uniconazole-P-treated riceshoots. Evolution of ethylene was promoted slightly, being 1.8times greater under 0.3 ppm uniconazole-P treatment than thatof control. The most obvious effect was the increase of trans-Zand trans-RZ in shoots. Shoots treated with uniconazole-P (10mg/m2 nursery box) contained 3.4 times and 3 times more trans-Zand trans-RZ than control, respectively. No significant differencesof cytokinin levels were recognized in roots except for cis-RZ.The increase of ethylene and active forms of cytokinins, andthe decrease of gibberellin in the shoots may be the basis forphysiological phenomena caused by uniconazole-P, namely thepromotion of flowering in woody plants and the enhancement offemaleness in cucumber. The distribution of exogenously-supplied radioactive labelledindol-3-yl-acetic acid (IAA) and gibberellin A1 (GA1) in geotropicallystimulated roots of Norway spruce (Picea abies (L.) Karst.)has been demonstrated. Seedlings were positioned with theirroot tips in 2.1 x 10–6 M [14C]IAA or 1.3 x 10–8m 3H-GA1 for 4 and 20 h, respectively. After geotropic stimulationfor 90 min in the horizontal position the root tips were cutlongitudinally in 50 µm thick sections, using a freeze-microtome.The radioactivity in the 14C-IAA treated roots occurred in higherconcentration in the lower than in the upper halves (ratio 1.25:1). A similar trend was observed in the [3H]GA1-treated rootswhere the ratio lower: upper halves was 2.04: 1. The ratio ofradioactivity in right and left halves of vertical roots wasapproximately the same in roots supplied with [14C]IAA and [3H]GA1(1.09: 1). The supplied radioactive compounds were analysed chromatographicallyafter extraction in methanol of 6 mm apical root segments. Onlya small fraction (7–8 per cent) of the supplied [14C]IAAwas revealed unchanged in the segments. The major part of thechromatographed, labelled compound has not been identified,but on basis of its RF value it is suggested that it may beindol-3-acetyl-aspartic acid (IAAasp). The chromatographic analysis of the [3H]GA,-treated segmentsshowed that only small fractions of this gibberellin has beenconverted to other compounds. These results have been discussed and correlated with knowledgeof plant growth regulators and their participation in root geotropism. Auxin activates pumping of protons from the symplast to theapoplast and causes hyperpolarization of the symplast membranein the elongation zone of Vigna stems prior to the accelerationof growth. This auxin-induced hyperpolarization has been studiedin most cases in hypocotyl segments excised from the elongationzone. In the present study, mature-zone segments were perfusedwith IAA by the xylem perfusion technique in an effort to determinewhether or not IAA has any effects in the mature zone. Althoughno hyperpolarization of the symplast membrane was observed uponthe perfusion with auxin alone, auxin-induced hyperpolarizationwas observed when mature-zone segments had been pretreated withGA3, in the absence of an increase in the growth rate. Theseresults suggest that cells in the mature zone have lost theability to activate the proton-pumping machinery in responseto auxin but that this ability can be restored by treatmentwith GA3. This effect of GA3 suggests the possibility that theconcentration of gibberellin in a tissue controls one of thecell's responses to auxin, namely, activation of the protonpump. The morphological effects of gibberellin A3 (GA3) on the dgtmutant of tomato were investigated. The mutant effectively showedthe normal range of responses, including a promotion of stemlength due to an increased number of longer internodes, a dramaticincrease in apical dominance, and effects on leaf shape andcolour. In the case of stem elongation, the quantitative responseof the mutant was greater than normal. The morphological abnormalitiescharacteristic of the dgt mutant, such as horizontal growth,a thin stem and hyponastic leaves, were not normalized by GA3. The natural chlorinated auxins 4-chloroindole-3-acetic acid(4-Cl-IAA) and its methyl ester (4-Cl-IAA Me ester) were found,in addition to IAA and its Me ester, by gas chromatography-massspectrometry in immature seeds of Vicia amurensis, a Vicieaespecies. In contrast, only non-chlorinated, IAA and IAA Me esterwere present in immature seeds of three Phaseoleae species.These results are further evidence of the wide distributionof 4-Cl-IAA and its Me ester in various Vicieae. The recessive gene na results in peas with extremely short internodes(phenotype nana). In intact plants, na prevents expression ofthe Le/le gene pair which determine the tall/dwarf difference.na blocks a step early in the gibberellin biosynthetic pathwaywhile le prevents conversion of gibberellin A20 to GA1. Whengrafted to leafy Na stocks, le na and Le na scions become phenotypicallydwarf and tall, respectively. In azuki bean (Azukia angularis = Vigna angularis) epicotylsections, gibberellin A3 (GA3) enhanced the growth promotingeffect of indoleacetic acid (IAA), but showed no growth effectwhen applied alone. Sections showed practically no cell division.The promoting effect of GA3 on section growth seems to be dueto its promoting effect on cell elongation. The diameters of sections treated with IAA increased, but thediameters of sections treated with GA3 together with IAA remainedconstant. GA3 seems to suppress cell expansion in a directiontransverse to the cell axis. Colchicine at a concentration with no inhibiting effect on IAA-inducedelongation almost completely reversed the effect of GA3 On the basis of these results, the participation of wall microtubulesin GA3-induced elongation is discussed. Indole-3-acetaldehyde (IAAId) was detected in the culture supernatantof Bradyrhizobium elkanii. Deuteriumlabelled L-tryptophan (Trp)was incorporated into IAAId and indole-3-acetic acid (IAA),suggesting that B. elkanii produces IAA via IAAId from Trp.In B. elkanii cell suspension, indole-3-pyruvic acid (IPyA)was converted to IAAId, and exogenously added IAAId was rapidlyconverted to IAA. Furthermore, the activity of indolepyruvatedecarboxylase (IPDC), which catalyzes the decarboxylation ofIPyA to produce IAAId and is a key enzyme for IPyA pathway,was detected in B. elkanii cell-free extract. The IPDC activitydepended on Mg2+ and thiamine pyrophosphate, cofactors of decarboxylation.This mounting evidence strongly suggests that IAA synthesisoccurs via IPyA pathway (Trp IPyA p IAAId IAA) in B. elkanii. Indol-3-ylacetic acid (IAA), gibberellin A3 (GA) and 6 benzylaminopurine(BAP) were applied factorially each at 3x10–2 M in lanolinto the roots and stems of Sitka spruce seedlings and the activityof the two secondary meristems, the vascular cambium and phellogen,and of the parenchymatous tissues between them, was examined.All the treatments, with the exception of GA produced a localizedstimulation of radial growth at the point of application andthere was a similarity in the response of the various tissuesin both the root and stem. Radial growth of the xylem was notsignificantly affected in the roots whereas in the stems BAPand IAA stimulated growth. In the phloem BAP produced significantstimulation in both roots and stems and IAA stimulated growthin the roots. Growth of the parenchyma and periderm externalto the phloem was also strongly stimulated by both BAP, andIAA in roots and stems. In roots and stems the application of BAP altered the derivativesproduced by the vascular cambium, resulting in the productionof large multiseriate rays in the xylem, and giving rise toan overall increase in the proportion of ray tissue. In a study of various phytopathogenic fungi, we found that fungithat belong to the genus Rhizoctonia produce IAA efficientlyfrom tryptophan. R. solani Kühn MAFF-305219, in particular,produced large amounts of tryptophol (Tol), which was assumedto be a specific by-product of the indole-3-pyruvate (IPy) pathway,in addition to IAA. Therefore, this fungus seemed suitable foranalysis of the function and the regulation of the biosynthesisof auxin by a fungal pathogen. Under normal aerobic conditions,the ratio of IAA to Tol synthesized by this strain was higherthan that under less aerobic conditions. In metabolic studieswith various indole derivatives, R. solani converted L-tryptophanand indole-3-acetaldehyde to IAA and Tol, but other indole derivativeswere scarcely metabolized. Of many varieties of rice plant tested, a dwarf mutant "Tamanishiki"was the most suitable material for gibberellin assay. It respondsto neither kinetin nor indoleacetic acid in the concentrationrange from 0.01 to 10 µg/l. Seeds were sterilized withsodium hypochiorite solution (chlorine content, one per cent)for one hour and soaked in water for 2 days. Germinating seedsselected for uniformity were then placed in test tubes containing1–3 ml of test solution, and the length of the 2nd leafsheath of the seedlings was measured after 7 days of incubation.The best conditions for the incubation were: pH of the testsolution, 4; temperature, 30°; continuous illumination ofintensity stronger than 50 lux. Under the best conditions statistically significant growth promotionwas found with 7 seedlings grown in one ml of the test solutionwhich contained 0.001 µg gibberellin As In double logarithmicplot, the growth promotion over control increased linearly withincreasing gibberellin concentration up to 1 µg in oneml test solution. Segments excised from the upper and the lower parts of cowpea(Vigna unguiculata L.) hypocotyls were compared in terms oftheir responses to exogenous indole-3-acetic acid (IAA) in relationto their endogenous levels of gibberellin. Growth of the segmentswas measured continuously during xylem perfusion with a lineardifferential transformer. IAA induced a burst of elongationin the upper segments but only slight promotion of growth inthe lower segments. Treatment with uniconazole, a potent inhibitorof the biosynthesis of gibberellins, reduced the responsivenessof the upper segments to exogenous IAA to about one half ofthe control value. Pre-perfusion with GA3 of such segments fortwo hours prior to application of IAA, partially restored theresponsiveness to IAA. Analysis by GC/MS identified GA1, GA4,GA9 GA20 and GA51 as native gibberellins in the hypocotyls ofcowpea seedlings. Analysis by GC/SIM also showed that the physiologicallyactive gibberellins (GA1 and GA4) were located mainly in theupper part of the hypocotyl and the treatment with uniconazolemarketly reduced the endogenous level of gibberellins thereto less than 11% of the control level. The relative activities of seven gibberellins have been testedin their ability to induce antheridia in prothallia of Anemiaphyllitidis. The spectrum of activities differs from the spectradescribed for other gibberellin reactions. Allogibberic acidproved to be highly active in the Anemia test. Additionallyit causes a change in the polarity of the germinating spore.Under the influence of this compound the primary rhizoid istransformed into an additional chloronema. In high concentrationsof allogibberic acid more than 80% of the prothallia developinto those "twins". Out of the polypodiaceous ferns only fewspecies respond to gibberellin application. In roots of sweet potato (Ipomoea batatas Lam. cv. Kokei 14),the metabolic response to wounding was remarkable in the proximalside and developed in the acropetal direction. We assumed thatthe polarity resulted from the increase in polar movement ofindoleacetic acid (IAA) (1977, Plant Physiol. 60: 563–566).Transport of IAA and change of the IAA level in the woundedtissue of sweet potato roots were investigated. Transport ofthe label from 14C-IAA was obviously polarized in the acropetaldirection. 14C-IAA administered to the wounded tissue was mainlymetabolized into two conjugates of IAA. The amount of IAA inthe wounded tissue, determined by the spectrofluorometric method,increased about 3-fold after 18 hr of incubation prior to thedevelopment of activities of some enzymes. The increase in IAAcontent was not affected with aseptic incubation, therefore,the possibility of IAA production by microorganisms on the woundedtissue was excluded. The results obtained strongly support ourhypothesis that IAA plays an important role in the metabolicresponse to wounding. (Received May 2, 1979; )  相似文献

Further studies have been made on the interactions of plant-growthregulators during uptake by Avena sativa coleoptile and Phaseolusradiatus hypocotyl segments. 2, 4-Dichlorophenoxyacetic acid(2, 4-D) had no effect on the uptake of either indol-3yl-aceticacid (IAA) or -naphthylacetic acid (NAA) by Avena. On the otherhand, a-(i-naphthylmethylthio)-propionic acid (NMSP) stronglyinhibited IAA uptake non-competitively but was much less effectiveon NAA uptake by Avena. The ‘metabolic’ uptake ofIAA by hypocotyl segments of Phaseolus radiatus was very stronglyinhibited by 2, 3, 5-tri-iodobenzoic acid (TIBA).  相似文献
