The extra small virus (XSV) is a satellite virus associated with Macrobrachium rosenbergii nodavirus (MrNV) and its genome consists of two overlapping ORFs, CP17 and CP16. Here we demonstrate that CP16 is expressed from the second AUG of the CP17 gene and is not a proteinase cleavage result of CP17. We further expressed CP17 and several truncated CP17s (in which the N-or C-terminus or both was deleted), respectively, in Escherichia coli. Except for the recombinant plasmid CP17^AC10, all recombinant plasmids expressed soluble protein which assembled into virus-like particles (VLPs), suggesting that the C-terminus is important for VLP formation.  相似文献

利用PCR方法从烟草曲茎病毒 (TbCSV)Y35分离物的病株中获得复制蛋白 (Rep)基因 ,将其克隆到原核表达载体pGEX 4T 1上获得重组质粒pGEX Y35Rep。重组质粒导入大肠杆菌BL2 1(DE3)pLysS中 ,IPTG诱导表达后发现部分Rep融合蛋白以可溶性形式表达。利用GST Sepharose 4B亲和层析柱纯化了Rep的融合蛋白 ,免疫家兔获得Rep蛋白的抗体。对TbCSV侵染烟草中Rep蛋白的亚细胞分布研究发现 ,Rep蛋白主要分布于含有细胞核的组份中。利用免疫胶体金技术对感病烟草中Rep蛋白进行了定位 ,发现Rep蛋白存在于细胞核内  相似文献

Virus-derived small interfering RNAs (vsiRNAs) play important roles in regulating host endogenous gene expression to promote virus infection and induce RNA silencing to suppress virus infection. However, to date, how vsiRNAs affect geminivirus infection in host plants has been less studied. In this study, we found that tobacco curly shoot virus (TbCSV)-derived vsiRNA18 (TvsiRNA18) can regulate TbCSV infection in Nicotiana benthamiana plants. The virus-mediated small RNA expression system and stable transformation technique were used to clarify the molecular role of TvsiRNA18 in TbCSV infection. The results indicate that TvsiRNA18 can aggravate disease symptoms in these plants and enhance viral DNA accumulation. ATP-dependent RNA helicase (ATP-dRH) was proven to be a target of TvsiRNA18, and down-regulation of ATP-dRH in plants was shown to induce virus-like leaf curling symptoms and increase TbCSV infection. These results suggest that TvsiRNA18 is an important regulator of TbCSV infection by suppressing ATP-dRH expression. This is the first report to demonstrate that TbCSV-derived vsiRNA can target host endogenous genes to affect symptom development, which helps to reveal the molecular mechanism of symptom occurrence after the virus infects the host.  相似文献

近年来对一些单组分菜豆金色花叶病毒属病毒(begomoviruses)的研究发现,这类单组分begomoviruses伴随着一种新型卫星分子-DNAβ。对我国云南省发生的双生病毒的研究表明,云南begomoviruses普遍伴随DNAβ分子,DNAβ分子与致病性紧密相关,且begomoviruses与其伴随的卫星DNA的是共进化的。为了弄清卫星DNAβ是否存在类似于卫星RNA的高度变异,将烟草曲茎病毒(Tobacco curly shoot virus,TbCSV)及其伴随卫星DNAβ的侵染性克隆共同接种本氏烟(Nicotiana benthamiana)和心叶烟(N.glutinosa),对TbCSV卫星DNAβ在不同寄主中的分子变异进行了比较。  相似文献

Cacao swollen shoot virus (CSSV) is a major pathogen of cacao (Theobroma cacao) in Africa, and long‐standing efforts to limit its spread by the culling of infected trees have had very limited success. CSSV is a particularly difficult virus to study, as it has a very narrow host range, limited to several tropical tree species. Furthermore, the virus is not mechanically transmissible, and its insect vector can only be used with difficulty. Thus, the only efficient means to infect cacao plants that have been experimentally described so far are by particle bombardment or the agroinoculation of cacao plants with an infectious clone. We have genetically transformed three non‐host species with an infectious form of the CSSV genome: two experimental hosts widely used in plant virology (Nicotiana tabacum and N. benthamiana) and the model species Arabidopsis thaliana. In transformed plants of all three species, the CSSV genome was able to replicate, and, in tobacco, CSSV particles could be observed by immunosorbent electron microscopy, demonstrating that the complete virus cycle could be completed in a non‐host plant. These results will greatly facilitate the preliminary testing of CSSV control strategies using plants that are easy to raise and to transform genetically.  相似文献

Rice stripe virus (RSV) causes severe diseases in Oryza sativa (rice) in many Eastern Asian countries. Diseasespecific protein (SP) of RSV is a non-structural protein and its accumulation level in rice plant was shown to determine the severity of RSV symptoms. Here, we present evidence that expression of RSV SP alone in rice or Nicotiana benthamiana did not produce visible symptoms. Expression of SP in these two plants, however, enhanced RSV- or Potato virus X (PVX)- induced symptoms. Through yeast two-hybrid screening, GST pull-down, and bimolecular fluorescence complementation assays, we demonstrated that RSV SP interacted with PsbP, a 23-kDa oxygen-evolving complex protein, in both rice and N. benthamiana. Furthermore, our investigation showed that silencing of PsbP expression in both plants increased disease symptom severity and virus accumulation. Confocal microscopy using N, benthamiana protoplast showed that PsbP accu- mulated predominantly in chloroplast in wild-type N. benthamiana cells. In the presence of RSV SP, most PsbP was recruited into cytoplasm of the assayed cells. In addition, accumulation of SP during RSV infection resulted in alterations of chloroplast structure and function. Our findings shed light on the molecular mechanism underlying RSV disease symptom development.  相似文献

Eukaryotic cells coordinate chromosome duplication by the assembly of protein complexes at origins of DNA replication by sequential binding of member proteins of the origin recognition complex (ORC), CDC6, and minichromosome maintenance (MCM) proteins. These pre-replicative complexes (pre-RCs) are activated by cyclin-dependent kinases and DBF4/CDC7 kinase. Here, we carried out a comprehensive yeast two-hybrid screen to establish sequential interactions between two individual proteins of the mouse pre-RC that are probably required for the initiation of DNA replication. The studies revealed multiple interactions among ORC subunits and MCM proteins as well as interactions between individual ORC and MCM proteins. In particular CDC6 was found to bind strongly to ORC1 and ORC2, and to MCM7 proteins. DBF4 interacts with the subunits of ORC as well as with MCM proteins. It was also demonstrated that CDC7 binds to different ORC and MCM proteins. CDC45 interacts with ORC1 and ORC6, and weakly with MCM3, -6, and -7. The three subunits of the single-stranded DNA binding protein RPA show interactions with various ORC subunits as well as with several MCM proteins. The data obtained by yeast two-hybrid analysis were paradigmatically confirmed in synchronized murine FM3A cells by immunoprecipitation of the interacting partners. Some of the interactions were found to be cell-cycle-dependent; however, most of them were cell-cycle-independent. Altogether, 90 protein-protein interactions were detected in this study, 52 of them were found for the first time in any eukaryotic pre-RC. These data may help to understand the complex interplay of the components of the mouse pre-RC and should allow us to refine its structural architecture as well as its assembly in real time.  相似文献

Mixed infections of Nicotiana benthamiana plants by Cucumber mosaic virus (CMV) and Tobacco necrosis virus (TNV) exhibit a synergistic interaction and result in symptom enhancement. Accumulation of CMV(+) RNA as well as capsid protein (CP) in mixed infection was considerably higher than that of singly‐infected plants. There was also a slight increase in TNV(+) RNA and CP levels in doubly infected plants. Synergistic infection by CMV‐ and TNV‐induced higher increase in the levels of malonyldialdehyde, hydrogen peroxide (H2O2) and more decline in the activities of catalase than singly infected ones. Both peroxidase and superoxide dismutase activities increased rapidly for the first 10 days post inoculation (dpi) in doubly‐infected plants and then declined, whereas the enzyme activities continued to increase after 10 dpi in singly infected plants and had higher enzyme activities in the late stages than that of co‐infected plants. These results suggest that synergistic infection by CMV and TNV produced severes oxidative stress in N. benthamiana plants and the synergy between the two viruses was mutual.  相似文献

Origin recognition complex (ORC), a six-protein complex, is the most likely initiator of chromosomal DNA replication in eukaryotes. Throughout the cell cycle, ORC binds to chromatin at origins of DNA replication and functions as a 'landing pad' for the binding of other proteins, including Cdt1p, to form a prereplicative complex. In this study, we used yeast two-hybrid analysis to examine the interaction between Cdt1p and every ORC subunit. We observed potent interaction with Orc6p, and weaker interaction with Orc2p and Orc5p. Coimmunoprecipitation assay confirmed that Cdt1p interacted with Orc6p, as well as with Orc1p and Orc2p. We mapped the C-terminal region, and a middle region of Orc6p (amino acids residues 394-435, and 121-175, respectively), as important for interaction with Cdt1p. Cdt1p was purified to examine its direct interaction with ORC, and its effect on the activity of ORC. Glutathione-S-transferase pull-down analysis revealed that Cdt1p binds directly to ORC. Cdt1p neither bound to origin DNA and ATP nor affected ORC-binding to origin DNA and ATP. These results suggest that interaction of Cdt1p with ORC is involved in the formation of the prereplicative complex, rather than in regulation of the activity of ORC.  相似文献

Acquisition and transmission was studied of Tomato yellow leaf curl China virus (TYLCCNV) and Tobacco curly shoot virus (TbCSV) by the B and a non‐B biotype (China‐ZHJ‐1) of Bemisia tabaci from Zhejiang, China. The frequency of TYLCCNV and TbCSV detection by PCR in whitefly adults increased with increasing length of feeding on virus‐infected plants. The virus DNA was detected by PCR in 40% of the B biotype adults tested after a period of 30 min access to infected plants and in all adults after a 12‐h period of access. All ZHJ‐1 adults acquired TYLCCNV and TbCSV after a 48‐h period of access to the virus‐infected plants. Viruliferous B and ZHJ‐1 adults retained TYLCCNV DNA for their entire life when placed on healthy cotton plants. Viruliferous ZHJ‐1 adults retained TbCSV DNA for their entire life when placed on healthy cotton plants but the B biotype adults did not. Transmission of TYLCCNV was achieved with one B or ZHJ‐1 adult per plant, and the probability of transmission reached 100% when the number of adults was increased to 10 per plant. The efficiency for TYLCCNV transmission to healthy plants by adults of both B and ZHJ‐1 was much higher than that for TbCSV.  相似文献

榆黄蘑中一种抗病毒蛋白的纯化及其抗TMV和HBV的活性   总被引:16,自引:1,他引:16  
采用阴离子交换层析和凝胶层析方法从新鲜食用菌榆黄蘑(Plearotus citrinopileatus)中进行了抗病毒蛋白的纯化,结果获得了一个纯化蛋白YP46-46,经SDS-PAGE可确定其分子量为27.4kD.以半叶法在枯斑寄主心叶烟上检测该蛋白对烟草花叶病毒(TMV)的抑制率,发现有较好的抗TMV活性,其抑制TMV的中浓度为0.24μg/mL.同时以HepG2.2.2.15细胞株为模型,对所获得的蛋白进行体外抗乙型肝炎病毒效果的评价.结果表明该蛋白对HBsAg的50%抑制时的浓度为0.08μg/mL,但对HBeAg效果不大.  相似文献
