The stickleback family (Gasterosteidae) of fish is less than 40 million years old, yet stickleback species have diverged in both diploid chromosome number (2n) and morphology. We used comparative fluorescence in situ hybridization (FISH) on 2 stickleback species, Gasterosteus aculeatus (2n = 42) and Apeltes quadracus (2n = 46), to ascertain the types of chromosome rearrangements that differentiate these species. The A. quadracus karyotype contains more acrocentric and telocentric chromosomes than the G. aculeatus karyotype. By using bacterial artificial chromosome probes from G. aculeatus in our FISH screen, we found that 6 pericentric inversions and 2 chromosome fusions/fissions are responsible for the greater number of acrocentric and telocentric chromosomes in A. quadracus. While most populations of G. aculeatus have an XX/XY sex chromosome system, A. quadracus has a ZZ/ZW sex chromosome system, as previously reported. However, we discovered that a population of A. quadracus from Connecticut lacks heteromorphic sex chromosomes, providing evidence for unexpected sex chromosome diversity in this species.  相似文献

For the first time, we studied the karyotype of the flathead sculpin Megalocottus platycephalus taeniopterus (Kner, 1868) from Vostok Bay of the Sea of Japan. The karyotype is stable: 2n = 42 (2 metacentric, 2 submeta-subtelocentric, 30 subtelocentric, and 8 acrocentric chromosomes), NF = 44 + 2. The nucleolar organizers (NOs) were identified using Ag banding in two pairs of chromosomes: in the telomeric parts of the short arm of the medium-size subtelocentric chromosome and the long arm of the large acrocentric chromosome. Variations in the number of nucleolar organizer chromosomes and in the number of NO staining blocks were found. Comparison of the karyotypes of M. p. taeniopterus and previously studied M. p. platycephalus (Pallas, 1814) from the northern Sea of Okhotsk revealed their similarity in the number and morphology of chromosomes and the number of chromosome arms and difference between the subspecies in the number and location of NO, which allows their discrimination.  相似文献

Przewalski's wild horse (E. przewalskii, EPR) has a diploid chromosome number of 2n = 66 while the domestic horse (E. caballus, ECA) has a diploid chromosome number of 2n = 64. Discussions about their phylogenetic relationship and taxonomic classification have hinged on comparisons of their skeletal morphology, protein and mitochondrial DNA similarities, their ability to produce fertile hybrid offspring, and on comparison of their chromosome morphology and banding patterns. Previous studies of GTG-banded karyotypes suggested that the chromosomes of both equids were homologous and the difference in chromosome number was due to a Robertsonian event involving two pairs of acrocentric chromosomes in EPR and one pair of metacentric chromosomes in ECA (ECA5). To determine which EPR chromosomes were homologous to ECA5 and to confirm the predicted chromosome homologies based on GTG banding, we constructed a comparative gene map between ECA and EPR by FISH mapping 46 domestic horse-derived BAC clones containing genes previously mapped to ECA chromosomes. The results indicated that all ECA and EPR chromosomes were homologous as predicted by GTG banding, but provide new information in that the EPR acrocentric chromosomes EPR23 and EPR24 were shown to be homologues of the ECA metacentric chromosome ECA5.  相似文献

The karyotype of Saguinus labiatus labiatus was determined by the Giemsa-banding technique on leukocytes cultured from 10 marmosets. The diploid chromosome number (2n = 46) was the same and the chromosome complement similar to other marmosets of genus Saguinus. Small karyotypic differences were found between S. l. labiatus and white-lipped marmosets (Saguinus fuscicollis) in the size of the X chromosome and in the banding pattern of one pair of metacentric chromosomes. A karyotypic variant was detected in 1 S. l. labiatus, characterized by a diploid chromosome number of 45 with balanced autosomal translocation involving two pairs of acrocentric chromosomes (T 16/19).  相似文献

The karyotype of the snow sculpin Myoxocephalus brandti, 2n = 44, NF = 46, from Peter the Great Bay was studied. Two-armed chromosomes were presented by one pair of metacentric chromosomes of medium size; one-armed chromosomes included two pairs of large subtelocentric chromosomes and a pair of large acrocentric chromosomes. Ag-NOR-staining in the telomere vicinity revealed nucleolus-organizing regions in one metacentric chromosome and in one medium size acrocentric chromosome in one of the fishes, in two homological small acrocentric chromosomes in three fishes, and in one acrocentric chromosome of average size in six fishes. No difference between the male and female karyotypes and any type of variability was revealed. The karyotypes of the snow sculpin M. brandti and the frog sculpin M. stelleri were compared. Summary The material consists of 84 metaphase plates from 17 individuals with clearly distinguishable Y chromosomes. The plates were obtained from leucocyte cultures. In making the preparations, exactly the same procedure was employed in all cases, including among other things, air-drying and light flaming.It was found that the size of the Y chromosome is subject to interindividual variation. The size of the Y chromosome has been expressed in relation to the mean length of the other small acrocentric chromosomes. The chromosomes have been tentatively classified into the following main groups:1. Y/G = 1.8; 2. Y/G =1.5; 3. Y/G is somewhat larger than G or 1.1, and 4. Y/G equals the mean of the small acrocentric chromosomes, or Y/G = 1. In the long Y chromosome two secondary constrictions have been observed.The location of the Y chromosome has been determined as peripheral or non-peripheral. The proband material has been divided into three main groups. The first comprises the individuals with a large Y chromosome (Y/G = 1.8). The second group includes individuals showing Down's syndrome and having 47 chromosomes, and the third comprises individuals with 46 chromosomes and possessing a Y/G sized 1 to 1.5. Preferential peripheral location of the Y chromosome has not been statistically verified in any one of these groups.  相似文献

The volumes and morphology of human chromosomes in mitotic reconstructions   总被引:1,自引:1,他引:0  
Summary Ten unpretreated normal human male fibroblast cells in mitosis were completely reconstructed from micrographs of between 82 and 119 consecutive serial sections. All 46 chromosomes and their centromeres could be reconstructed in every cell. Measurements of chromosome volumes and centromere indices are presented. The data enabled allocation of all chromosomes to their groups (A to G), and chromosomes 1, 2, 3, 6, 9, 16, 17, 18 and Y were individually identified. Comparisons with published karyotypes showed that volume measurements correlated well with measurements of DNA content and chromosome length. Centromere indices also showed good correlation, but the acrocentric chromosomes were more unequally armed than found by length measurement. Secondary constrictions at the nucleolar organising region were visible on about a third of the acrocentric chromosomes. One chromosome of the C group, number 9, had a diffuse subcentromeric region (DSR) on the long arm, at the position of the constitutive heterochromatin and (in meiotic cells) the paramere.  相似文献

A new karyotype for blind mole rats was recorded in Tunceli province in Eastern Turkey. The karyotype contained 44 chromosomes, including 13 biarmed pairs, 7 acrocentric pairs, and one heteromorphic pair with a submetacentric and an acrocentric homologue in the autosomal complement (FNa=69). The X chromosome was submetacentric and the Y chromosome medium-sized subtelocentric (FN=73). Distinct dark centromeric C-bands were observed on most of the biarmed and three pairs of the acrocentric autosomes. The NORs were detected on short arms of three subtelocentric pairs and one acrocentric pair of autosomes. The diploid number of chromosomes and the karyotype characteristics observed are obviously unique among hitherto studied populations of blind mole rats and the complement can be evaluated as a new chromosome race of Nannospalax xanthodon. The distribution ranges of individual chromosome races of the species recorded in Eastern Anatolia are revised and possible interracial hybridization is discussed in respect of the finding of a new race.  相似文献

Karyotype of Omisus caledonicus with 2n = 14 is described, all chromosomes are acrocentric, heterochromatin centromere is vacuolized. The following sequence discovered: A1, A2, B1, B2, C1, C2, D1, D2, E1, F1, G1 and G2. Simple inversions are found in chromosomes A-C and G, a complex reconstruction with 3 break points are observed in chromosome D. All reconstructions are large (from 66 to 99% of bands). According to phylogenetic reconstructions for morphological characters (Saether, 1979) and data on karyotypes it may be considered that O. caledonicus is one of much more primitive species in the tribe Chironomini.  相似文献

The karyotype and other chromosomal markers as revealed by C-banding and Ag-staining were studied in Lutjanus quinquelineatus and L. kasmira (Lutjanidae, Perciformes). While in latter species, the karyotype was invariably composed of 48 acrocentric chromosomes in both sexes, in L. quinquelineatus the female karyotype had exclusively 48 acrocentric chromosomes (2n = 48) but that of the male consisted of one large metacentric and 46 acrocentric chromosomes (2n = 47). The chromosomes in the first meiotic division in males showed 22 bivalents and one trivalent, which was formed by an end-to-end association and a chiasmatic association. Multiple sex chromosome system of X1X1X2X2/X1X2Y type resulting from single Robertsonian fusion between the original Y chromosome and an autosome was hypothesized to produce neo-Y sex chromosome. The multiple sex chromosome system of L. quinquelineatus appears to be at the early stage of the differentiation. The positive C-banded heterochromatin was situated exclusively in centromeric regions of all chromosomes in both species. Similarly, nucleolus organizer region sites were identified in the pericentromeric region of one middle-sized pair of chromosomes in both species. The cellular DNA contents were the same (3.3 pg) between the sexes and among this species and related species.  相似文献

Concordant/discordant associations at chromatid level were compared and found significant (P less than 0.05) in females with primary amenorrhea. This probably suggested that the acrocentric association pattern in this group of ASD and infertility did not follow a random segregation in subsequent cell divisions and that the concordant acrocentric chromosomes have regularly established physical connections with one another, held together for several cell cycles. It could only be speculated that the association of acrocentric chromosome anomalies in some females with abnormal sex chromosomes are due to this reason. In the event that chromosome association has a bearing on chromosome aberrations, the non-random pattern of acrocentric association probably would increase the choice for translocation and non disjunction in the somatic cells in females with primary amenorrhea during ontogenesis.  相似文献

The chromosomes of Australotettix montanus Richards and A. carraiensis Richards have been studied. Both species share the following characteristics: the chromosome number is 2n=45 ♂-46 ♀ the sex mechanism is of the XO ♂-XX ♀ type with the X submetacentric; the X chromosome as well as two bivalents (H1 and H2) are heterochromatic during first prophase.
The species differ in: (a) the pattern of distribution of heterochromatic blocks in H1; (b) the H1 pair consists of two metacentric autosomes in A. carraiensis while these chromosomes are acrocentric in A. montanus ; (c) a medium-sized pair of autosomes is formed by subacrocentric elements in A. carraiensis while they are acrocentric in A. montanus.
The karyotypes of both species do not differ basically from that which is typical for the Macropathinae.  相似文献

Karyotypic and cytogenetic characteristics of catfish Harttia carvalhoi (Paraíba do Sul River basin, S?o Paulo State, Brazil) were investigated using differential staining techniques (C-banding, Ag-staining) and fluorescent in situ hybridization (FISH) with 18S and 5S rDNA probes. The diploid chromosome number of females was 2n = 52 and their karyotype was composed of nine pairs of metacentric, nine pairs of submetacentric, four pairs of subtelocentric and four pairs of acrocentric chromosomes. The diploid chromosome number of males was invariably 2n = 53 and their karyotype consisted of one large unpaired metacentric, eight pairs of metacentric, nine pairs of submetacentric, four pairs of subtelocentric, four pairs of acrocentric plus two middle-sized acrocentric chromosomes. The differences between female and male karyotypes indicated the presence of a sex chromosome system of XX/XY1Y2 type, where the X is the largest metacentric and Y1 and Y2 are the two additional middle-sized acrocentric chromosomes of the male karyotype. The major rDNA sites as revealed by FISH with an 18S rDNA probe were located in the pericentromeric region of the largest pair of acrocentric chromosomes. FISH with a 5S rDNA probe revealed two sites: an interstitial site located in the largest pair of acrocentric chromosomes, and a pericentromeric site in a smaller metacentric pair of chromosomes. Translocations or centric fusions in the ancestral 2n = 54 karyotype is hypothesized for the origin of such multiple sex chromosome systems where females are fixed translocation homozygotes whereas males are fixed translocation heterozygotes. The available cytogenetic data for representatives of the genus Harttia examined so far indicate large kayotype diversity.  相似文献

Cytogenetic and molecular analyses enabled identification of two cytotypes among individuals of the spotted scorpion fish Scorpaena plumieri from Margarita Island, Venezuela. Cytotype 1 was characterized by 48 subtelo‐acrocentric chromosomes and fundamental number (number of chromosome arms; FN) equalled 48, while cytotype 2 was characterized by two metacentric and 46 subtelo‐acrocentric chromosomes and FN was 50. These cytotypes also differed in the location of the ribosomal gene clusters and in the distribution of the constitutive heterochromatin. Moreover, fish from the cytotypes 1 and 2 were found to belong to distinct mitochondrial lineages. The presence of two S. plumieri cytotypes from two lineages separated by high genetic distance suggests that they correspond to sympatric cryptic species.  相似文献

A procedure for obtaining G-bands on chromosomes of mammals is outlined. The procedure was utilized in an investigation of the idiogram and banding pattern of the mitotic chromosomes of the cotton rat, Sigmodon arizonae. The diploid number of this species is 22, and each pair of homologues is easily separated on the basis of size, centromeric position, and banding pattern. The autosomes are represented by four pairs of large submetacentric chromosomes, three pairs of medium to small submetacentric chromosomes, two pairs of large subtelocentric chromosomes and one pair of small acrocentric chromosomes. The X chromosome is acrocentric and averages from 5.42% to 5.46% of the haploid female complement. The Y chromosome is a minute acrocentric and easily separated from the smallest acrocentric autosome. The usefulnes of Sigmodon arizonae as a laboratory animal for cytogenetic studies is substantiated.  相似文献

江豚(Neophocaena phocaenoides)是鲸目(Cetacea)鼠海豚科(Phocaenidae)的一种小型齿鲸,在淡水和海洋中均有分布。关于江豚染色体的研究,国外文献中尚未见记载,国内亦无报道。Pilleri和Gihr(1972,1975)根据江豚的形态解剖学的研究,认为我国产的江豚和印度洋的及日本海的江豚不属同一个种,但国际上对此尚有不同意见。因此,搞清江豚染色体的核型,将可有助于澄清江豚属的的分类问题。本文就我国长江产江豚的染色体核型作初步探讨。  相似文献

Three different karyotypes have been found so far among Saimiri originating from five different South American localities. All animals examined have the same diploid number (44) of chromosomes but the number of acrocentric and submetacentric chromosomes varies, presumably as a result of pericentric inversions. Saimiri originating from Iquitos, Peru, consistently have ten acrocentric chromosomes; animals originating from Leticia, Colombia, have 12 acrocentric chromosomes. Hybrids produced in our laboratory have the expected 11 acrocentrics and one unpaired submetacentric chromosome. Animals originating from Guyana have fourteen acrocentric chromosomes and the expected two fewer submetacentric chromosomes. Squirrel monkeys from Costa Rica, Panama, and Pucallpa, Peru, studied to this date conform to the Iquitos type with ten acrocentric chromosomes. These findings point to genetic differences which may result in variable responses to laboratory situations. The evolutionary factors involved in this rearrangement of chromosomes and possible influences on phenotypes are subjects of interest for future study. The importance of identifying the source of squirrel monkeys used in biomedical research is apparent if results from different laboratories are to be repeated or compared.  相似文献

The polytene chromosomes of the salivary glands of Nilodorum biroi (Kieffer) comprise three long metacentric or submetacentric chromosomes. It is considered that this is a derived condition from a diploid number of 8, by the tandem fusion of a small acrocentric element (C) to a metacentric chromosome (AB). A standard chromosome map for the genus is provided and the chromosomal polymorphism of Indian and Australian populations is described.  相似文献
