Summary Experimental evidence is advanced which contradicts the view that the intercepts on the ordinate of plots of amount of cation absorbedvs time may be interpreted as representing the amount of ion initially bound to the sites involved in its active absorption and may be used to calculate the concentration of these sites.  相似文献

In Petunia pollen tubes growing in the style there appear to be two ways of callose deposition. The first one is callose deposition outside the plasma membrane as a distinct layer closely appressed to the cell wall. The second one is callose deposition within the cytoplasm as distinct callose grains, leading to the formation of callose plugs. This second way is accompanied by a characteristic ultrastructure of the cytoplasm, namely strong electron-density of the plasma matrix, partial absence of the plasma membrane and the absence of plastids and dictyosomes. Data of compartmental analysis of sulphate were compared with the sterol content of roots of differently yielding maize genotypes. In conditions of steady state nutrient supply, sterol content was significantly correlated only with sulphate efflux (co). This increased at increasing concentration of sterols in the roots. Influx to cytoplasm (oc) was evaluated after sulphate deprivation leading to an induced rate of sulphate uptake. This was negatively correlated with sterol content, which was lower in the high than in the low yielding genotypes. When the highest yield genotype was grown at different sulphate concentrations, influx, efflux, root content of sulphate and sterols were positively correlated with the concentration of sulphate in the nutrient medium. Sterol content in roots appears to be controlled by both the genetic settlement and the nutritional status in maize. Low sterol content is connected with a high efficiency of sulphate utilization.  相似文献

Summary Barley leaves extracted 35 min after a 15-min pulse of 10-6 M 14C-labelled cis,trans-(±)-abscisic acid (ABA) showed very little breakdown of the ABA. Leaves extracted 2 h after a 30-min pulse of 10-5 M labelled ABA showed at least 34% degradation of the hormone. The pattern of degradation was very similar for leaves kept in either light or darkness following the labelling pulse, and the products formed resembled those described in tomato fruit and bean axes. No detectable isomerization to the physiologically inactive trans,trans-(±)-ABA isomer occurred in these leaves during the 2-h period. ABA-induced stomatal closure is partially reduced 30 min after the pulse. The reversal cannot be attributed to catabolism of the hormone and must be explained by removal of the hormone into storage sites where it cannot act on the stomata.  相似文献

Ophiobolin A, a sesterterpene metabolite of Helminthosporium maydis, Nisikado and Miyake, stimulates net leakage of electrolytes and glucose from maize (Zea mays L.) seedling roots. Treatment of the roots with ophiobolin A at a concentration of 10 mug/ml (25 mum) inhibits uptake of 10 mm 2-deoxyglucose by 50% and of 0.5 mm 2-deoxyglucose by 85%. Compartmental analysis of the efflux of 3-O-methylglucose failed to show a similar effect of ophiobolin A on the rate of efflux of hexose. The inhibition of uptake is not reversible by washing. There is no difference in the effects on roots from cytoplasmic male sterile or normal cytoplasm plants, and exposure of carrot (Daucus carota L.) root discs to ophiobolin A also causes inhibition of 2-deoxyglucose uptake by this tissue.  相似文献

Summary The uptake of C14O2 by the roots of intact tomato plants from solution containing Na2C14O3 was studied at different light intensities as well as in darkness.Where plants had previously been starved for CO2 for 12 hours, a higher rate of C14 uptake was observed than with plants which had been transferred directly from the soil to the radioactive solution.In general, the C14 content of the roots was slightly higher than that of the shoots. At light intensities under the compensation point and in darkness the C14 content of the shoots relative to the roots decreased. This was accompanied by release of C14O2 during respiration, indicating that the absorbed C14 was readily translocated upwards and released as C14O2 under these conditions. At light intensities above the compensation point no C14O2 was released.  相似文献

The addition of 1 micromolar 2,3,5-triiodobenzoic acid (TIBA) to solutions containing KCl resulted in the inhibition of K and Cl uptake in excised barley roots. The effectiveness of TIBA as an inhibitor increased as the pH of the treatment solution decreased and approached the pKa of TIBA. A lag period of approximately 20 minutes existed prior to the onset of TIBA induced inhibition of ion uptake. Respiratory activity was also inhibited by TIBA. The data suggest that in this material, TIBA functions by entering the cytoplasm and inhibiting metabolism. Comparisons made on the effect of added Ca, showed that at pH 5.7 and higher, Ca had no effect on ion uptake whereas at lower pH values the presence of Ca enhanced uptake by offsetting the deleterious effects of H+.  相似文献

Abstract The patterns of naturally growing ion currents associated with horizontally growing roots of Trifolium repens L. seedlings were measured in a simple low-salt bathing medium using a highly sensitive vibrating electrode. Current consistently enters the main elongation zone of the root and leaves from mature, elongated tissue. This current enters and leaves with densities of ca. 4.0–11.0 mA m?2 and 4.0 mA m?2, respectively. Current was also delected entering the zone of emerging root hairs and also the root-hair tips themselves. These results are a further example of the involvement of self-generated electrical fields in plant developmental processes. Possible, secondary rhizosphere-associated effects of the extracellular loop of the developmental current are also suggested.  相似文献

Soil water movement with root water uptake is a key process for plant growth and transport of water and chemicals in the soil-plant system. In this study, a root water extraction model was developed to incorporate the effect of soil water deficit and plant root distributions on plant transpiration of annual crops. For several annual crops, normalized root density distribution functions were established to characterize the relative distributions of root density at different growth stages. The ratio of actual to potential cumulative transpiration was used to determine plant leaf area index under water stress from measurements of plant leaf area index at optimal soil water condition. The root water uptake model was implemented in a numerical model. The numerical model was applied to simulate soil water movement with root water uptake and simulation results were compared with field experimental data. The simulated soil matric potential, soil water content and cumulative evapotranspiration had reasonable agreement with the measured data. Potentially the numerical model implemented with the root water extraction model is a useful tool to study various problems related to flow transport with plant water uptake in variably saturated soils. The uptake of beryllium by excised barley roots from a solutioncontaining 111 µM beryllium and 500 µM calcium wasstudied. The exchange-adsorbed portion was 15% of the totalBe uptake after 30 min of exposure. The total Be content couldbe reduced to 55% of the control in 1 hr with demineralizedwater. At pH levels above 6, Be uptake was increased dramatically.A Q10 of 1.2 was found to exist between 1 and 23°C. Mg (111µM) did not influence Be absorption while Zn and Al atthe same concentration reduced it, possibly competitively, 27and 62% of the control, respectively. Dinitro-phenol (500 µM),cyanide (500µM), azide (500 µM) and m-chlorocarbonylcyanidephenylhydrazone (100 µM) at least doubled it indicatingthe existence of a possible metabolic barrier to beryllium entrywhich was disrupted by these compounds. Absorption kineticsover an external beryllium concentration range of 11 to 444µM suggested a number of binding sites. Absorption isotherms for chloride and rubidium ions have been determined through a wide concentration range for nonvacuolate root tips, and for vacuolate subapical sections of corn root. In the range 0 to 0.5 mm, chloride absorption is hyperbolic with concentration in both tips and proximal sections. At high concentrations, 1 to 50 mm, a second multiple-hyperbolic isotherm for chloride is noted in vacuolate tissue, while the isotherm for nonvacuolate tips rises exponentially. A linear to exponentially rising isotherm is taken to signify diffusive permeation.  相似文献

Effects of several organic acids on ion uptake and retention and on respiration in barley roots having low and high KCl contents were assayed by measurements of K+, Na+, Ca2+, Cl, and oxygen uptake. Organic acids with high pKa values increase the permeability of roots to ions and decrease respiration when present in sufficient concentrations at pH 5 but have no inhibitory effects at pH 7. Absence of respiratory inhibition in short times and at lower organic acid concentrations, under conditions that immediately produce a permeability increase, indicate that the permeability change is not a result of respiratory inhibition. Effects of formate, acetate, propionate, and glutarate are attributed to entry of undissociated acid molecules into the effective membranes. Lack of a permeability increase with succinate, which has lower distribution coefficients to lipid solvents than do the aliphatic acids, can be explained by failure of sufficient amounts of the hydrophilic succinic acid molecules to penetrate the membranes involved. These experiments suggest that undissociated acid in root membranes can increase permeability of the roots.  相似文献

Abstract The concentration dependence of phosphate uptake was studied using root sections of corn (Zea mays L. cv. Ganga 5). Detailed and wide-range (57 concentrations in the range 1 μmol m?3-75 mol m?3), precise (average SEM < 2.5%, n= 6) and reproducible (similar patterns in three independent experiments and for 5, 10, 15, 20, 25 and 30°C) data revealed six (or seven) concentration-dependent phases separated by ‘jumps’ or sharp breaks. These transitions were independent of temperature and occurred over relatively narrow concentration ranges (0.0001–0.0004, 0.08–0.31, 1.0–3.5, (7.5–10), 18–20 and 57–59 mol m?3). The intermediate phases obeyed Michaelis-Menten kinetics, whereas sigmoidal kinetics were observed at lower concentrations. Uptake within each of the two highest phases increased more rapidly with increasing external phosphate concentration than predicted from Michaelis-Menten kinetics but also saturated more rapidly. The latter finding is not consistent with free diffusion across the plasmalemma at high external phosphate concentrations. Kinetic models yielding continuous isotherms, e.g. the sum of one or two Michaelis-Menten terms and a diffusion term, cannot account for the data.  相似文献

Mechanistic models of nutrient uptake consider diffusion and mass flow acting simultaneously to supply nutrients to the sorbing root surface. Plant parameters that determine nutrient uptake include those describing changes in root geometry and size due to root growth and others describing kinetics of the nutrient uptake process. Mechanistic models generally assume that nutrient uptake occurs evenly along the roots that are uniformly distributed in homogeneous and isotropic soil having no temporal and spatial gradients in volumetric moisture content. Uptake of immobile nutrients (like P and K) is mainly determined by the soil-supply parameters and is well predicted by the simulation models. In contrast, uptake of mobile nutrients (e.g. Ca and Mg) that usually accumulate at the root surface is determined mainly by the plant-uptake parameters; prediction of uptake of those nutrients is subject to a much wider error due to uncertainties of applying kinetic parameters measured on hydroponically-grown plants to soil-grown plants. Comparison of model-predicted and experimentally-observed uptake values should be done by calculating the mean squares of deviates instead of performing regression analysis, especially if data that encompass a relatively wide range in root length are considered. Complementary-ion effects occurring at the soil-root interface raise the need for developing a multi-nutrient uptake model that will simultaneously calculate uptake of several essential nutrients taking into account interactions among them.  相似文献
