Aims:  To evaluate the antioxidant effect of carotenoids from Deinococcus radiodurans on protein.
Methods and Results:  Deinococcus radiodurans strain R1 (ATCC 13939) and its mutant strain R1ΔcrtB were used for this study. The total carotenoids (R1ex) from D. radiodurans were obtained by extraction with acetone/methanol (7 : 2, by vol), and their antioxidant activity was measured using the DPPH˙ (2,2-diphenyl-1-picrylhydrazyl) system. The protein oxidation level, in vitro and in the cell, was measured using the DNPH (2,4-dinitrophenyl hydrazine) method. The carotenoid extract R1ex scavenged 40·2% DPPH˙ radicals compared to β-carotene (31·7%) at a concentration of 0·5 mg ml−1. The intracellular level of protein oxidation in mutant R1ΔcrtB, which does not contain carotenoid, was 0·0212 mmol mg−1 protein which is significantly greater than that in the wild type (0·0169 mmol mg−1 protein) following the treatment with H2O2. The purified major carotenoid product (deinoxanthin) from the wild type showed a greater inhibition of oxidative damage in bovine serum albumin than lycopene or lutein.
Conclusions:  Carotenoids prevent protein oxidation and contribute to the resistance to cell damage in D. radiodurans .
Significance and Impact of the Study:  Our results provide the evidence that carotenoids can protect proteins in D. radiodurans against oxidative stress.  相似文献

Rod and cone cells of the mammalian retina harbor two types of a membrane bound guanylate cyclase (GC), rod outer segment guanylate cyclase type 1 (ROS-GC1) and ROS-GC2. Both enzymes are regulated by small Ca2+-binding proteins named GC-activating proteins that operate as Ca2+ sensors and enable cyclases to respond to changes of intracellular Ca2+after illumination. We determined the expression level of ROS-GC2 in bovine ROS preparations and compared it with the level of ROS-GC1 in ROSs. The molar ratio of a ROS-GC2 dimer to rhodopsin was 1 : 13 200. The amount of ROS-GC1 was 25-fold higher than the amount of ROS-GC2. Heterologously expressed ROS-GC2 was differentially activated by GC-activating protein 1 and 2 at low free Ca2+ concentrations. Mutants of GC-activating protein 2 modulated ROS-GC2 in a manner different from their action on ROS-GC1 indicating that the Ca2+ sensitivity of the Ca2+ sensor is controlled by the mode of target–sensor interaction.  相似文献

Aims:  To expose bacteria to anolyte and subsequently investigate the effect of anolyte on the protein profiles of treated bacteria.
Methods and Results:  Proteins were extracted from bacteria treated with different concentrations of anolyte and analysed using SDS-PAGE. Fewer and more faint protein bands were observed for concentrated halide anolyte treated bacteria when compared to untreated bacteria while extra protein bands were observed for bacteria exposed to dilute concentrations.
Conclusions:  The undiluted and the 10−1 dilution of halide derived anolyte was effective in killing the test bacteria. Anolyte caused bacterial death by complete destruction of proteins or by causing oxidative stress which resulted in protein fragmentation.
Significance and Impact of the Study:  The results of this study provide information on the antimicrobial mechanism of anolyte on other bacteria for which the information is currently unavailable.  相似文献

Aims:  To investigate the effects of salicylates in Saccharomyces cerevisiae exposed to oxidative stress induced by hydrogen peroxide (H2O2).
Methods and Results:  Saccharomyces cerevisiae was cultured through to the postlogarithmic phase of growth. Stress was induced by the addition of 1·5 mmol l−1 H2O2 for 1 h, while N-acetyl-l-cysteine (NAC) and glutathione (GSSG) were used as control agents that affect the redox balance. Sodium salicylate, at 0·01–10 mmol l−1or acetylsalicylic acid, at 0·02–2·5 mmol l−1 was administered at various times before hydrogen peroxide stress. Both agents conferred resistance to a subsequent hydrogen peroxide stress, similarly to the induction of the adaptive response observed upon pretreatment with NAC and GSSG. Sodium salicylate was more potent as a short-term, but not as a long-term pretreatment agent, compared to acetylsalicylic acid.
Conclusions:  Pharmacological pretreatment with salicylates resulted in dose related increases in cell survival, indicating the induction of the protective response in yeast.
Significance and Impact of the study:  The possible role of salicylates in the modulation of the hydrogen peroxide stress response in eukaryotic cells address questions on the effects of these commonly used therapeutic agents in a number of disorders exhibiting an oxidative stress component.  相似文献

SUMMARY 1. Two experiments were performed with periphytic diatoms originating from the River Lot (France) and allowed to grow on clean substrata within indoor artificial streams. Three cadmium (Cd) levels (control, low Cd=10 μg L−1 and high Cd=100 μg L−1) were used to test the effects of Cd on (i) the settlement and development of diatom communities (Experiment 1, 4 weeks Cd exposure) and (ii) predeveloped communities grown over a 2-week period without contamination (Experiment 2, 2 weeks Cd exposure).
2. Experiment 1 revealed that growth and taxonomic composition of diatom communities clearly differed with Cd exposure. Biofilms were more adpressed to substrata under low Cd concentration, and were thinner and patchy under high Cd concentration.
3. Exposure of developed diatom communities (Experiment 2) to Cd revealed only minor variations in taxonomic composition, possibly linked to the protective role of the developed organic matrix against metal stress.
4. These results support the validity of periphytic diatom communities as indicators of metal pollution, although significant structural changes would take longer within developed communities.  相似文献

Aims:  The ability to transform Vibrio spp. is limited by the extracellular nuclease that their cells secrete. The reported transformation efficiency of this organism is 102–105 transformants per microgram DNA. We tried different buffers and conditions, aiming to elevate its transformation efficiency.
Methods and Results:  MgCl2 and sucrose are often included in the washing and/or electroporation buffers to stabilize the cell membrane. However, Mg2+ is required for production and activity of the extracellular nuclease. A simple electroporation buffer lacking Mg2+ was found to increase transformation efficiency dramatically, to levels 50-fold more than the buffers containing Mg2+. To maintain the stability of the cell membranes, Mg2+ was replaced with high concentrations of sucrose, from 272 to 408 mmol l−1. With the new buffers, the transformation efficiency of Vibrio parahaemolyticus was increased to 2·2 × 106 transformants per microgram DNA.
Conclusions:  Mg2+ in the buffer adversely affected transformation of V. parahaemolyticus by electroporation. The cell membranes of vibrio can be stabilized by high concentration of sucrose when Mg2+ is absent.
Significance and Impact of the Study:  A greater transformation efficiency can facilitate the genetic analysis of an organism and its pathogenicity. Buffers lacking Mg2+ can be used for other nuclease-producing organisms.  相似文献

Aims:  To investigate the antibacterial mechanism of carvacrol and thymol against Escherichia coli.
Methods and Results:  The time-kill curve results showed that carvacrol and thymol at 200 mg l−1 could inhibit the growth of E. coli . Flow cytometry and fluorescent dyes were used to explore the effect of two components on membrane permeability and membrane potential. In membrane permeability experiment, the mean fluorescence intensity of cells treated with 200 mg l−1 carvacrol or thymol were lower than nonexposed cells. The ratio of red to green fluorescence intensity of DiOC2(3) reflected the change of membrane potential. Carvacrol and thymol at 200 mg l−1 caused the ratio of red/green decreasing from 0·42 of control to 0·08 and 0·07, respectively.
Conclusions:  Carvacrol and thymol had desired antimicrobial effect on E. coli . The antibacterial effects were attributed to their ability to permeabilize and depolarize the cytoplasmic membrane.
Significance and Impact of the Study:  This study showed the potential use of flow cytometry as a suitable method to investigate the mode of antibacterial action of essential oil components.  相似文献

Aims:  To investigate the effect of liquid smoke on growth, survival, proteomic pattern and haemolytic potential of Listeria monocytogenes.
Methods and Results:  Growth and survival curves were recorded in brain–heart infusion broth supplemented with three concentrations of liquid smoke. L. monocytogenes growth was inhibited in the presence of 15 μg ml−1 phenol while a rapid decrease in cell viability occurred in the presence of 30 μg ml−1 phenol. The proteome of L. monocytogenes cytosoluble proteins was slightly modified after 2-h incubation with 30 μg ml−1 phenol but no protein already characterized in response to other known stresses was induced, except the protease ClpP. Liquid smoke inhibited the haemolytic potential without affecting hly gene expression, showing a potential inhibition of protein activity or stability.
Conclusions:  The presence of liquid smoke in a rich medium strongly affected growth and survival of L. monocytogenes . Brief smoke stress affected the metabolic pathways and inhibited the haemolytic activity of L. monocytogenes .
Significance and Impact of Study:  This study is a first step in the investigation of the influence of a smoked product on L. monocytogenes strains.  相似文献

Aims:  The aim was to explore the mechanisms by which a blend of orange ( Citrus sinensis ) :  bergamot ( Citrus bergamia ) (1 : 1 v/v) EO (essential oil) (2% v/v) and its vapour (15 mg l−1 air) brings about its antimicrobial effect against Enterococcus faecium and Enterococcus faecalis .
Methods and Results:  Cells were exposed to the blend in oil or vapour form in a sealed unit. Membrane permeability was measured using an NPN assay and intra and extracellular ATP concentrations were assessed using luminescence. Assays using 3,3-dipropylthiacarbocyanine and carboxyfluorescein diacetate succinimidyl ester measured membrane potential and intracellular pH changes. TEM images of treated cells indicate morphological differences and show the possible uptake of the EO into the cell. After cells were exposed to EO or vapour, cell permeability increased by ×2 and ×40 respectively. A decrease of 1·5 in intracellular pH, 20 a.u. in membrane potential and 18 pmol mg−1 protein of intracellular ATP occurred.
Conclusions:  The EO blend affects the cell membrane and cell homeostasis resulting in inhibition of growth or cell death.
Significance and Impact of the Study:  Understanding the mechanisms by which EOs bring about their antibacterial effect could lead to an alternative to chemical-based bactericides for use against Enterococcus sp.  相似文献

Aims:  The source and routes of diarrhoeagenic Escherichia coli (DEC) have not been clarified because it is difficult to detect these organisms in samples with numerous coliform bacteria. We have developed multiplex real-time PCR assays for exhaustive detection of DEC.
Methods and Results:  Primers and TaqMan probes were designed to amplify and quantify one gene ( eae , stx1 , stx2 , elt , est , virB , aggR , astA, and afaB ) from each of seven pathotypes of DEC, in duplex or triplex reactions under the same PCR cycling conditions. Specificity was confirmed using 860 strains including 88 DEC strains. The fluorescence threshold cycle and DNA concentrations correlated with decision coefficients of more than 0·99. Subsequently, meat samples and enrichment broths were spiked with DEC and the assays used to detect the genes. The detection limits varied from 7·1 × 102 to 1·1 × 104 CFU ml−1, depending on the target genes. All meat samples spiked with a variety of DEC (more than 10 CFU 10 g−1) were found to be positive by the method.
Conclusions:  The present system allows for the efficient and simultaneous determination of various DEC pathotypes.
Significance and Impact of the Study:  This system makes epidemiological investigations for DEC sensitive and quick, and is a useful tool to clarify the source and routes of DEC.  相似文献

Aim:  To investigate the effects of feeding and induction strategies on the production of Bm R1 recombinant antigen.
Methods and Results:  Fed-batch fermentation was studied with respect to the specific growth rate and mode of induction to assess the growth potential of the bacteria in a bioreactor and to produce high yield of Bm R1 recombinant antigen. Cells were grown at a controlled specific growth rate (μset) during pre-induction, followed by constant feeding postinduction. The highest biomass (24·3 g l−1) was obtained during fed-batch process operated at μset of 0·15 h−1, whereby lower μset (0·075 h−1) gave the highest protein production (9·82 mg l−1). The yield of Bm R1 was increased by 1·2-fold upon induction with 1 mmol l−1 IPTG (isopropyl-β- d -thiogalactoside) compared to using 5 mmol l−1 and showed a further 3·5-fold increase when the culture was induced twice at the late log phase.
Conclusions:  Combination of feeding at a lower μset and twice induction with 1 mmol l−1 IPTG yielded the best result of all variables tested, promising an improved method for Bm R1 production .
Significance and Impact of the Study:  This method can be used to increase the production scale of the Bm R1 recombinant antigen to meet the increasing demand for Brugia Rapid, a commercial diagnostic test for detection of brugian filariasis.  相似文献

Abstract— Effects of an increased concentration of K+ (55 m m ) in the medium on fluxes of glutamate and other amino acids in the presence and absence of 10 m m -glutamate were studied. The following observations were made:
(1) The efflux of glutamate is slightly increased by excess K+. The glutamate efflux is smaller than the potassium fluxes.
(2) The K+-induced increase of glutamate efflux is enhanced under anoxia or in glutamate-containing media.
(3) The influx of glutamate is unaffected or slightly increased by excess K+.
(4) The efflux of GABA is increased by excess K+, both in the absence and in the presence of glutamate.
(5) Efflux of glutamine, leucine and lysine is increased by excess K+, but only provided that glutamate is also present in the medium.
(6) Efflux of glutamate and of GABA is increased by addition of 10 m m -glutamate.  相似文献

Expression of mel gene improves the UV resistance of Bacillus thuringiensis   总被引:1,自引:0,他引:1  
Aims:  To improve ultraviolet (UV) resistance of Bacillus thuringiensis for increasing the duration of the Bt product applied in the field, a genetically engineered strain Bt TD841 that produced both melanin and Cry1A protein was constructed, and its UV resistance was evaluated in the laboratory.
Methods and Results:  Melanin quantitative analysis revealed that the recombinant strain Bt TD841 could synthesize 0.15 mg melanin ml−1 sporulated culture. Atomic force microscopy confirmed the production of diamond crystal and SDS-PAGE results showed the expression of the 130 kDa Cry1A protein. Bioassay results demonstrated that the LC50 value of Bt TD841 was 3.69 μl ml−1 against Helicoverpa armigera and the UV resistance of this recombinant was enhanced 9.7-fold compared to its parental strain Bt HC42 after 4-h UV irradiation.
Conclusion:  Expression of the mel gene can significantly increase UV resistance of B. thuringiensis.
Significance and Impact of the Study:  This is the first report on genetically engineered Bt strain with co-expression of melanin and the insecticidal crystal proteins gene, and the results may offer a practical solution for improving the photoprotection of Bt products in field application.  相似文献

1. The effects of emergent macrophytes on water turbidity and sediment resuspension in the shallow Kirkkojärvi basin of Lake Hiidenvesi were studied with sediment traps, and concomitant sediment and water samples. The study was conducted during May–August in three different zones of a stand of the emergent Typha angustifolia .
2. Within the stand (5 m from the edge), both the concentration of suspended solids and the rate of sediment resuspension were significantly lower than at the edge and outside the stand (5 m from the edge). The differences between the zones increased towards the end of summer together with the growing stem density. During the study period (82 days), 2210 g dw m−2 of sediment was resuspended in the outer zone. At the edge and in the inner zone, the corresponding numbers were 1414 and 858 g dw m−2, respectively.
3. With the resuspended sediment, 39.4 mg P  m−2 day−1 was brought to the water column outside the stand, 22.4 mg P  m−2 day−1 at the edge and 13.4 mg P  m−2 day−1 within the stand.
4. In early summer, the concentration of suspended solids had a highly significant positive effect on soluble reactive phosphorus (SRP) concentration in the water, whereas in late summer no effect was found. During the study period, phosphorus retention by emergent macrophyte stands corresponded to 3–5% of the present annual external phosphorus loading of the Kirkkojärvi basin.  相似文献

Aims:  To evaluate the anti-oxidant properties of extracts from 20 medicinal herbs growing in western Siberia using microbial test systems and different in vitro methods.
Methods and results:  In vivo anti-oxidant activity of extracts was evaluated for their capacity to protect bacteria, Escherichia coli , against bacteriostatic and bactericidal effects of H2O2 and menadione, and action on anti-oxidant gene expression. In vitro anti-oxidant activity has been examined by a number of methods including: the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH)-scavenging assay, chelating activity and capacity to protect plasmid DNA against oxidative damage. In addition, total polyphenol content was determined. The extracts of Fragaria vesca , Rosa majalis , Pentaphylloides fruticosa , Alchemilla vulgaris and Pulmonaria mollis possessed the highest levels of anti-oxidant activity in vivo and in vitro . The protective properties were more closely related to the DPPH radical-scavenging activity, tannin content and action on anti-oxidant gene expression than to other parameters.
Conclusion:  The extracts of medicinal plants may have anti-oxidant effects on bacteria simultaneously through several different pathways, including direct inhibition of reactive oxygen species, iron chelation and anti-oxidant genes induction.
Significance and Impact of the Study:  Using microbial test systems, we revealed herbs that may be used as potential sources of natural anti-oxidants.  相似文献

Aim:  The capacities of live and heat-killed cells of Saccharomyces cerevisiae at 45°C for the removal of copper, nickel and zinc from the solution were compared.
Methods and Results:  Kinetic studies have shown a maximum accumulation of Ni2+ and Zn2+ after 10 min for both types of cells, while for Cu2+ this was attained after 30 and 60 min for dead and live cells, respectively. Equilibrium studies have shown that inactivated biomass displayed a greater Zn2+ and Ni2+ accumulation than live yeasts. For Cu2+, live and dead cells showed similar accumulation. Fluorescence, scanning electron microscopy and infrared spectroscopy studies have shown that no appreciable structural or molecular changes occurred in the cells during the killing process. The increased metal uptake observed in dead cells can be most likely explained by the loss of membrane integrity, which allows the exposition of further metal-binding sites present inside the cells.
Conclusions:  Heat-killed cells showed a higher degree of heavy metal removal than live cells, being more suitable for further bioremediation works.
Significance and Impact of the Study:  Dead flocculent cells can be used in a low cost technology for detoxifying metal-bearing effluents as this approach combines an efficient metal removal with the ease of cell separation.  相似文献

Aims:  To determine the role of oxidative stress and chronological ageing on the propensity of brewing yeast strains to form respiratory deficient 'petites'.
Methods and Results:  Four industrial yeast strains (two ale and two lager strains) were exposed to oxidative stress in the form of H2O2 (5 mmol l−1) for two hours. Cell viability and occurrence of petites were determined by the slide culture and TTC-overlay techniques, respectively. Increases in petite frequency were observed but only in those strains sensitive to oxidative stress. Chronological ageing under aerobic conditions led to an increase in petites in strains sensitive to oxidative stress. No such increase was observed under anaerobic conditions.
Conclusion:  Ageing may contribute to mitochondrial DNA damage and increase the propensity of brewing yeast cells to become respiratory deficient. Tolerant strains may be less likely to generate petites as a result of serial re-pitching.
Significance and Impact of the Study:  Continuous re-use of brewing yeast is associated with an increase in the frequency of petites within brewery yeast slurries, a phenomenon resulting in reduced fermentative capacity. The cause of petite generation during brewery handling is unknown. We show that endogenous oxidative stress has the potential to generate petites within brewing yeast populations.  相似文献

Abstract: The growth cone is responsible for axonal elongation and pathfinding by responding to various modulators for neurite growth, including neurotransmitters, although the sensor mechanisms are not fully understood. Among neurotransmitters, GABA is most likely to demonstrate activity in vivo because GABA and the GABAA receptor appear even in early stages of CNS development. We investigated the GABAA receptor-mediated signaling pathway in the growth cone using isolated growth cones (IGCs). Both the GABAA binding site and the benzodiazepine modulatory site were enriched in the growth cone membrane. In the intact IGC, GABA induced picrotoxin-sensitive Cl flux (not influx but efflux) and increased the intracellular Ca2+ concentration in a picrotoxin- and verapamil-sensitive manner. Protein kinase C (PKC)-dependent phosphorylation of two proteins identified as GAP-43 and MARCKS protein was enhanced in the intact IGC stimulated by GABA, resulting in the release of MARCKS protein and GAP-43 from the membrane. Collectively, our results suggest the following scheme: activation of the functional GABAA receptor localized in the growth cone membrane → Cl efflux induction through the GABAA-associated Cl channel → Ca2+ influx through an L-type voltage-sensitive Ca2+ channel → Ca2+-dependent phosphorylation of GAP-43 and MARCKS protein by PKC.  相似文献
