The two-dimensional polyacrylamide gel electrophoresis technique has been adapted for the analysis of human cerebrospinal fluid proteins. Proteins were detected by Coomassie brilliant blue stain and/or by silver stain. Highly reproducible protein patterns were obtained. We analyzed ten normal CSF specimens, thirty pathological CSF specimens and the corresponding sera. We mapped the protein patterns observed by examination of serum/CSF differences and by immunofixation. Preliminary observations on the changes in protein patterns in CSF specimens from patients with neurological disorders are reported.  相似文献

Improved methods for the identification and grouping of bacteria by polyacrylamide gel electrophoresis of soluble proteins are described. Electrophoretic protein patterns were obtained in rigorously standardized comditions. The results were much more reproducible than any described previously. Some of the factors affecting reproducibility were; growth conditions, time and speed of centrifugation of extracts, and conditions of gel electrophoresis. Protein patterns were compared by computing correlation coefficients from normalized densitometric tracings and clustering the strains by the unweighted average pair group method. As model systems, both Agrobacterium and Zymomonas were used because of differences in the sharpness of the peaks. The methodwas applied to 42 Agrobacterium strains. The agreement with the results of clustering by either phenotypic tests or DNA:DNA hybridization was excellent. Computerized comparisons of electrophoretic protein patterns can be a fast, easy and powerful tool for classification and identification of bacteria.  相似文献

Summary Variation in the native conformation of bean lectins was examined using electrophoresis of non-denatured total protein extracts and purified albumin and globulin lectin. The observed variation was related to the genetic variation reported previously for lectin polypeptide composition as revealed by two-dimensional isoelectricfocusing-sodium dodecyl sulfate polyacrylamide gel electrophoresis (IEF-SDS/PAGE). When eleven cultivars with different IEF-SDS/PAGE lectin polypeptide compositions were compared, eight had unique non-denatured lectin patterns and three had identical patterns. For some cultivars differences in non-denatured lectin patterns were observed between the purified albumin and globulin lectin preparations.  相似文献

本文对槐生疫霉(Phytophthora robinicola)、掘氏疫霉(P.drechsleri)、樟疫霉(P.cinnamomi)、烟草疫霉(P.nicotianae)、棕榈疫霉(P.palmivora)及辣椒疫霉(P.capsici)等6种疫霉81株菌菌体可溶性蛋白质和酯酶进行了聚丙烯酰胺凝胶平板电泳研究。结果指出,种间蛋白质图谱差异明显,种内菌株间基本一致。就酯酶图谱来说,种内菌株间存在一定差异,但种间差异更为显著。因此,本试验进一步支持槐生疫霉新种的建立。同时表明,菌体可溶性蛋白质电泳图谱在疫霉种的鉴定和分类上具有重要意义,而酯酶电泳图谱在一定程度上也有助于疫霉种的研究。  相似文献

Purified cauliflower (Brassica oleracea var. botrytis) mitochondrial proteins fractionated into soluble, membrane, integral membrane and peripheral membrane samples were analyzed by 2D- PAGE (isoelectric focusing/ SDS polyacrylamide gel electrophoresis). 2D gels patterns were compared using the Imager Master 2D Elite software. 561 silver stained protein spots were resolved after electrophoresis under standard conditions of a whole protein extract. In the soluble fraction a prevalent number of more intense protein spots was observed. The cauliflower protein 2D patterns resembled Arabidopsis thaliana 2D patterns. The two protein spots selected which occupied a similar isoelectric point positions on both gels represented the same proteins as revealed by ESI-MS analysis of cauliflower proteins. The third selected spot belongs to unidentified proteins. The comparative analysis of mitochondrial suborganellar fractions proved the usefulness of this approach.  相似文献

Capillary zone electrophoresis and high-resolution agarose gel electrophoresis were compared to detect protein components in serum and cerebrospinal fluid of patients. Both electrophoretic methods proved to be useful for detection of protein abnormalities (e.g., mono- and oligoclonal bands) in biological fluids, but capillary electrophoresis offered several important advantages, such as sample application without preliminary concentration, lack of staining procedures, and on-line evaluation of patterns. Furthermore, capillary electrophoresis exhibits shorter analysis time and high resolution with low baseline noise. The results were proven to be powerful in diagnosis and monitoring of dyscrasias in routine laboratory practice.  相似文献

对北京、河北、青海、山东、湖北、广东等地不同寄主上收集的,属于Phytophthora capsici, P. colocasiae, P. heveae, P. infestans, P. melonis P. nicotianae var. nicotianae和P.nicotianae var.parasitica 7个种和变种的17株菌的蛋白质,进行了聚丙烯酰胺凝胶盘状电泳的分析和比较,经多批次试验,7个种和变种各自恒定地呈现典型的蛋白质图谱,显示出种间的差异。其中P.infestans占11株,它们虽采自不同地区和寄主,又分属5个生理小种,经多次电泳,电泳图谱基本相同,但不同菌株存在一些差异。同一株菌,培养条件对电泳图谱影响不大,不论是培养在含苹果酸盼培养基上,还是培养在延胡索酸培养基上,电泳图谱基本一致,只是电泳材料必须新鲜。  相似文献

为了建立一套适合小麦根蛋白质组分析的双向电泳系统(2-DE),得到更加清晰的电泳图谱,本研究以小麦幼苗根系为材料,对根蛋白的提取方法、上样量等进行了优化。研究发现,上样量为1200μg时,Trizol抽提法提取的蛋白能够获得图像清晰、分辨率高、重复性好的双向电泳图谱,基本满足小麦根系蛋白质组学的分析和研究。  相似文献

Protein fractions of salivary glands were analyzed from 30 wildtype strains of eight species belonging to the Drosophila nasuta subgroup by SDS-polyacrylamide gel electrophoresis. The electrophoretic patterns indicated several prominent bands which could be shown to represent the major glue protein fractions. The glue protein fractions are species-specific as well as wildtype strain-specific. Wildtype strain specificities are characterized by variations of the species-specific patterns. The patterns of the different wildtypes, species, and hybrids were used for taxonomic identification within the nasuta subgroup, in which the females are morphologically indistinguishable and the males differ only by the markings of their frons. The hybrids provide evidence for gonosomal as well as autosomal linkage of individual genes coding for the major glue protein fractions.  相似文献

为建立适用于杉木(Cunninghaimia lanceolata)叶片蛋白质组研究的双向电泳技术,对杉木叶片蛋白质的溶解方法、上样量、IEF及SDS-PAGE电泳等关键步骤进行了优化。结果表明,杉木叶片蛋白质主要分布在pH4-7范围;裂解液中含有硫脲(2mmol/L)才能较充分地溶解蛋白,DTT浓度为60mmol/L、上样量1.5mg时得到的图谱分辨率较好且蛋白斑点分布均匀、清晰,拖尾现象明显减少,平衡液Ⅱ中碘代乙酰胺浓度为450mg(15ml)-1时能提高图谱分辨率;采用与质谱兼容的考马斯亮兰进行染色,得到近700个蛋白点。  相似文献

The proteins of lumbar CSF have been investigated by two-dimensional gel electrophoresis, and their patterns have been compared with the corresponding serum protein patterns. Serum proteins in CSF have been identified by electroblotting and immunoreaction with antiserum against total human serum proteins. Proteins derived from brain have been identified with antiserum against human brain proteins. The most prominent CSF protein group has been identified as a multiple form of apolipoprotein E. The correct position of the glial fibrillary acidic protein has also been determined. The prefractionation of CSF proteins by size exclusion chromatography or by affinity chromatography followed by two-dimensional electrophoresis has facilitated the detection of trace components in CSF and the corresponding serum.  相似文献

Interfacing a minicomputer with a high resolution densitometer permits the online acquisition and statistical analysis of population parameters for experimental and control sample patterns of various macromolecules obtained by polyacrylamide gel electrophoresis. This technique, illustrated in this paper using protein from the brains of Japanese quail, enables the experimenter to make comparisons which were previously not readily available for electrophoresis.  相似文献

Numerical solution of the relevant continuity equations has been used to examine the possible effects of intramolecular sulfhydryl oxidation on the electrophoresis of proteins. Simulations of moving boundary electrophoresis, based on variants of a previous model [J. R. Cann, N. H. Fink, and D. J. Winzor (1983) Arch. Biochem. Biophys.221, 57–63], show that the Schlieren patterns for the ascending and descending limbs are likely to exhibit pronounced nonenantiography. Whereas the pattern for one limb may comprise essentially a single peak, that for the conjugate side can exhibit bimodality, the nature of which is time dependent. Bimodality of the Schlieren pattern can develop in either the ascending or descending limb of the electrophoresis cell, depending basically upon the number of sulfhydryl groups available for oxidation, and on the relative magnitudes of the rate constants describing the oxidation and the isomerization of the oxidized protein species. Whether the faster-moving or slower-moving peak grows with time is shown to depend upon the magnitude of the electrophoretic mobility of the resultant isomer in relation to that of the oxidized protein species. Schlieren patterns for fish muscle creatine kinase and rabbit muscle aldolase are then used to support the relevance of these predictions to moving boundary electrophoresis of proteins undergoing intramolecular sulfhydryl oxidation. Finally, numerical simulation of the zonal electrophoretic behavior of such systems serves to illustrate that bimodal patterns may also obtain, thereby giving a false impression of inherent protein heterogeneity. Emphasis is therefore placed on the importance of maintaining an adequate concentration of reducing agent throughout the medium in which the protein migrates, a potential problem in polyacrylamide gel electrophoresis at neutral pH.  相似文献

In this study, a total of fifteen staphylococcal strains belonging to different species were characterized by whole-cell and extracellular protein profiles using sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The results are presented as dendrograms after quantitative analysis of the band patterns with a computer program. Visual inspection of protein bands and cluster analysis of protein patterns of 15 strains representing 10 Staphylococcus species showed that whole-cell and extracellular protein profiles differed in several protein bands in Staphylococcus aureus, S. epidermidis, S. simulans and other species of Staphylococcus; however, the differences were insufficient for reliable differentiation of Staphylococcus species by the SDS-PAGE method.  相似文献

A multiple mini two-dimensional electrophoretic method which results in three two-dimensional protein spot patterns being positioned side by side in an individual gel has been developed. Preparation time has been minimized by employing disposable capillary tubes for the isoelectric focusing gels and reducing the number of second-dimensional gels required. Commercially available vertical slab units were used for the second-dimensional electrophoresis. The protein spot patterns were visualized either by staining the second-dimensional gel with silver or fluorescently labeling the focused proteins while present in the isoelectric focusing gel and subsequently electrophoresing them into the second-dimensional gel. The fluorescently labeled second-dimensional gel was imaged while still present in the glass mold immediately following electrophoresis. Two fluorophores were compared: 2-methoxy-2,4-diphenyl-3(2H)-furanone and 5-(4,6-dichlorotriazin-2-yl)aminofluorescein hydrochloride. A rapid imaging system based on a cooled charge-coupled device was used to view both the silver-stained and fluorescently labeled two-dimensional spot patterns. The sensitivity of detection of protein spots in the mini two-dimensional gels was similar for the two types of fluorescently labeled gels and the silver-stained gels.  相似文献

In an attempt to explain the unusual electrophoretic behavior of fish muscle creatine kinase, a phenomenological theory of transport of reacting systems has been formulated for the electrophoresis of a sulfhydryl protein undergoing oxidation in the presence of a gradient of molecular oxygen. The model assumes slow O2-oxidation of sulfhydryl groups followed by rather rapidly reversible sulfhydryl-disulfide interchange with concomitant change in the electrophoretic mobility of the protein. The computed electrophoretic patterns for this model exhibit a sharp, unimodal ascending boundary but a bimodal descending boundary in which the proportions of the two peaks are time dependent. There is a striking similarity between the theoretical patterns and their experimental counterparts (M.D. Doherty, D.A. Bergman, V.M. Re-Miller, and D.J. Winzor, 1980, Arch. Biochem. Biophys. 202, 558-564); and hence support for consideration of the electrophoresis of fish muscle creatine kinase in these terms.  相似文献

Varietal differences in the leaf protein profiles of 17 diploid (2n = 28) varieties of mulberry (Morus spp.) were demonstrated by 2D-gel electrophoresis and silver staining; similarity was statistically evaluated by means of principal component analysis. A close correlation was found between inter-varietal similarity, as estimated by the electrophoretic banding patterns of certain enzymes, and the leaf protein profiles obtained by 2D-gel electrophoresis. Little apparent varietal difference in the amount of ribulose-1,5-bisphosphate carboxylase was detected immunoelectrophoretically in the leaves. Varietal differences in crude protein content were obtained with leaves and this was discussed in relation to the observed 2D-gel electrophoretic patterns.  相似文献
