Treatment failure (TF) jeopardizes the management of parasitic diseases, including leishmaniasis. From the parasite’s point of view, drug resistance (DR) is generally considered as central to TF. However, the link between TF and DR, as measured by in vitro drug susceptibility assays, is unclear, some studies revealing an association between treatment outcome and drug susceptibility, others not. Here we address three fundamental questions aiming to shed light on these ambiguities. First, are the right assays being used to measure DR? Second, are the parasites studied, which are generally those that adapt to in vitro culture, actually appropriate? Finally, are other parasite factors – such as the development of quiescent forms that are recalcitrant to drugs – responsible for TF without DR?  相似文献

We present experimental data on the survivorship of damselflies infested by parasitic water mites from a population in field cages. In addition, we show correlative laboratory data under simulated severe weather conditions. In the manipulative experiment, parasitized females' individual condition, which was measured as weight at emergence, was an important determinant of survival under field conditions. In contrast, such a relationship did not occur in males and unparasitized females. It was found in the laboratory experiment that water mites as well as weight at emergence both contributed significantly to the reduced survivorship of male and female damselflies. It was concluded that the impact of parasitism depends on environmental conditions and that host sexes differ in their responses to parasitism. This is discussed in the light of immunocompetence in invertebrates.  相似文献

Summary Waage's (1982) hypothesis that host-size-dependent sex ratios will occur in parasitoids of nongrowing hosts and not in parasitoids of growing hosts is examined using published data on parasitoid wasps. Waage's hypothesis is supported as a general, but not absolute, rule: among solitary parasitoid wasps, a significantly greater proportion of parasitoids of nongrowing than of growing hosts show some evidence of host-size-dependent sex ratios (85% versus 42%, G=6.54, P< 0.05). The premise of Waage's hypothesis-that for parasitoids which develop in a growing stage, host size at oviposition is not a good predictor of the amount of resources available to the developing parasitoid-is also examined. It is suggested that across host species Waage's premise will hold for some, but not all, parasitoids of growing hosts. Likely exceptions to Waage's premise, and thus his prediction, are discussed. Parasitoids of growing hosts which are expected to have evolved hostsize-dependent sex ratios include parasitoids which utilize a narrow size range of host species, parasitoids which can distinguish among host species by some criterion other than size, and parasitoids which utilize host species whose susceptible instars do not overlap in size.  相似文献

Asymmetry patterns across the distribution range: does the species matter?   总被引:1,自引:0,他引:1  
An important question in evolutionary ecology is whether different populations across a species range, from core to periphery, experience different levels of stress. The estimation of developmental instability has been proposed as a useful tool for quantifying the degree of environmental and genetic stress that individuals experience during their development. Fluctuating asymmetry, the unsigned difference between the two sides of a bilaterally symmetrical trait, has been suggested to reflect the levels of developmental instability in a population. As such, it has been proposed as a useful tool for estimating changes in developmental instability and in stress response in populations across a range of environmental conditions. Recent studies focusing mostly on birds have detected increasing fluctuating asymmetry from core to periphery across the distribution range, suggesting that peripheral populations may experience higher levels of environmental and/or genetic stress. Most of these comparisons were done for single taxa across a single gradient. However, different species are predicted to respond differently to environmental shifts across the range. We compared asymmetry patterns in wing morphology in populations of two Euchloe butterfly species across their opposing ranges in Israel. Contrary to the patterns observed in birds across the same gradient, bilateral asymmetry did not increase or shift towards the periphery in either of the butterfly species. If fluctuating asymmetry in these traits reflects levels of stress, these results may partly reflect the fact that the range of these two butterfly species is limited by the distribution of their host plant, rather than by abiotic environmental variables. In addition, developing pierids can diapause during harsh seasons and can persist in resource‐rich patches, thus minimizing the environmental stress perceived by developing individuals. The lack of therapies fostering remyelination and regeneration of the neural network deranged by the autoimmune attack occurring in multiple sclerosis (MS), is raising great expectations about stem cells therapies for tissue repair. Mesenchymal stem cells (MSCs) have been proposed as a possible treatment for MS due to the reported capacity of transdifferentiation into neural cells and their ability at modulating immune responses. However, recent studies have demonstrated that many other functional properties are likely to play a role in the therapeutic plasticity of MSCs, including anti-apoptotic, trophic and anti-oxidant effects. These features are mostly based on the paracrine release of soluble molecules, often dictated by local environmental cues. Based on the modest evidence of long-term engraftment and the striking clinical effects that are observed immediately after MSCs administration in the experimental model of MS, we do not favor a major role for transdifferentiation as an important mechanism involved in the therapeutic effect of MSCs.  相似文献

Background and Aims

The underlying evolutionary processes of pollinator-driven floral diversification are still poorly understood. According to the Grant–Stebbins model speciation begins with adaptive local differentiation in the response to spatial heterogeneity in pollinators. Although this crucial process links the micro- and macroevolution of floral adaptation, it has received little attention. In this study geographical phenotypic variation was investigated in Patagonian Calceolaria polyrhiza and its pollinators, two oil-collecting bee species that differ in body size and geographical distribution.

Methods

Patterns of phenotypic variation were examined together with their relationships with pollinators and abiotic factors. Six floral and seven vegetative traits were measured in 45 populations distributed across the entire species range. Climatic and edaphic parameters were determined for 25 selected sites, 2–16 bees per site of the most frequent pollinator species were captured, and a critical flower–bee mechanical fitting trait involved in effective pollination was measured. Geographical patterns of phenotypic and environmental variation were examined using uni- and multivariate analyses. Decoupled geographical variation between corolla area and floral traits related to the mechanical fit of pollinators was explored using a Mantel test.

Key Results

The body length of pollinators and the floral traits related to mechanical fit were strongly correlated with each other. Geographical variation of the mechanical-fit-related traits was decoupled from variation in corolla size; the latter had a geographical pattern consistent with that of the vegetative traits and was mainly affected by climatic gradients.

Conclusions

The results are consistent with pollinators playing a key role in shaping floral phenotype at a geographical scale and promoting the differentiation of two floral ecotypes. The relationship between the critical floral-fit-related trait and bee length remained significant even in models that included various environmental variables and an allometric predictor (corolla area). The abiotic environment also has an important role, mainly affecting floral size. Decoupled geographical variation between floral mechanical-fit-related traits and floral size would represent a strategy to maintain plant–pollinator phenotypic matching in this environmentally heterogeneous area.  相似文献

Extruded pellets containing activated carbon (AC) can be used to sow native seeds while simultaneously applying herbicide to control invasive species. Incorporating AC in pellets has been demonstrated to protect native seeds; however, there may be unintended detrimental impacts to seedling emergence. We aimed to optimize seed position within pellets to maximize emergence and survival of the perennial shrub Jacksonia furcellata. Seeds were positioned at 2 mm (top), 6 mm (middle), and 12 mm (bottom) within pellets (with or without AC), sown on or below the soil surface, and compared to non-pelleted seeds sown under the soil surface in the equivalent positions (2, 6, and 12 mm depth). Trays were treated with a pre-emergent herbicide (Simazine) or left unsprayed. Emergence (without herbicide) was significantly higher from seeds positioned at the bottom of pellets without AC sown on the soil surface (70%), compared to non-pelleted seeds sown at the bottom (12 mm below the soil surface; 57%). However, emergence was inhibited when seeds were positioned in the middle (6 mm) of pellets with AC (32%). When treated with Simazine, survival was highest from seeds positioned at the bottom of pellets with AC (60%), compared to pellets without AC (15%) and non-pelleted seeds sown at the bottom (12 mm below the soil surface; 15%). Jacksonia furcellata seeds positioned at the bottom of pellets, sown on the soil surface, shows promise to minimize negative impacts to emergence, and maximize herbicide protection. Further testing with additional species is required to refine pellet production (e.g. recipe, extrusion, and shape) for optimal emergence.  相似文献

Background

Interest in translational studies aimed at investigating biologic markers in predicting response to primary chemotherapy (PCT) in breast cancer has progressively increased. We conducted a pilot study to evaluate feasibility of evaluating biomarkers of response to PCT at one & 21 days after first cycle.

Methods

Adult, non-pregnant, non-lactating women with histologically confirmed infiltrating duct carcinoma underwent serial core biopsies after first cycle of PCT and these were scored for Ki-67, Bcl-2 and Caspase-3 using immunohistochemistry.

Results

We recruited 30 patients with a mean age of 51 years. We were successful 95.6% times in performing a core biopsy and of these 84.6% had adequate tissue in the cores harvested. After a mean of 4 cycles of PCT, 26 patients underwent surgery and good response was noted in 9 patients (30%) using Miller-Payne criteria. There was a trend noted in all markers, which appeared different in those with good response and poor response. Good responders had significantly higher Ki-67 and significantly lower Bcl-2 at baseline and a significant decrease in Ki-67 and Caspase-3 at 21 days after the first chemotherapy.

Conclusion

We report a detectable change in biomarkers as early as 24–48 hours after the first chemotherapy along with a definite trend in change that can possibly be used to predict response to chemotherapy in an individual patient. The statistical significance and clinical utility of such changes needs to be evaluated and confirmed in larger trials.  相似文献

Immunostimulation in crustaceans: does it really protect against infection?   总被引:10,自引:0,他引:10  
There is a growing need to control, prevent or minimise the devastating effects of disease in crustacean culture without recourse to toxic chemicals or antibiotics. In keeping with approaches to disease control in fish and higher mammals, interest is developing in compounds that confer protection and/or enhance immune reactivity to likely pathogens in shellfish (sometimes, erroneously, referred to as as "shellfish vaccines"). The agents currently under scrutiny for crustaceans include glucans, lipopolysaccharides and killed bacterial cells. They are thought to act as "immuno-stimulants" because of their known effects on the crustacean immune system in vitro. A number of papers are now appearing in the literature claiming to demonstrate their positive impact on immunity and immunity and disease resistance. This review article considers the problem of disease and its control in crustacean farming, describing the types of capability in cultured species. Analysis of the validity of the results of many of the published studies raises questions about the value of these compounds for cost-effective control of infection in aquaculture, especially for long lasting protection in both adults and juveniles. This review further discusses the potential risks to the wellbeing of the stock animals from repeated use of these agents and makes the case for rigorous testing of putative stimulants, at the gene, protein and functional levels, as well as for the need to consider alternative strategies and approaches to disease control.  相似文献

We studied male locomotory response to trails and patches of sex pheromone (left respectively by free-ranging females and females constrained to stay on a small area) in the two parasitoids Aphelinus asychis (Hymenoptera: Aphelinidae) and Trichogramma brassicae (Hymenoptera: Trichogrammatidae). Under the hypothesis that the spatial distribution of virgin females differs between these species (scattered among host plants in A. asychis, gregarious at emergence sites in T. brassicae), we predicted that male locomotory response to their sex pheromones should also differ: A. asychis males should follow pheromone trails on plants in order to encounter the females along these trails, whereas T. brassicae males should stay on pheromone patches, at emergence sites, and mate the females on these patches. Using an improved video-tracking system, we found that males of both species respond to conspecific sex pheromone trails and patches, but that the response does not differ much between species. Males released on marked substrates walked in a more convoluted pattern (i.e. higher path fractal dimension and higher number of crossings within tracks) than males released on unmarked substrates. On pheromone patches, males turned persistently in the same direction when leaving the patch, which explains a higher number of visits on marked patches than on unmarked patches, and possibly, higher track convolution on pheromone trails. Contrary to our hypothesis, male A. asychis did not follow female trails more accurately than male T. brassicae, and male T. brassicae did not stay longer on pheromone patches than male A. asychis. We argue that these discrepancies between our predictions and the observed responses originates from discrepancies between the assumed spatial distribution of virgin females and their actual distribution in the wild.  相似文献

If the cestodes are excluded, then the parasitic platyhelminths of fishes divide neatly into the external and monoxenous Monogenea and the internal and heteroxenous Digenea. Both groups have apparently had long associations of coevolution, host switching and adaptation with fishes and have become highly successful in their respective habitats. Current estimates of species richness for the two groups suggest that they may be remarkably similar. Here we consider the nature of the diversity of the Monogenea and Digenea of fishes in terms of richness of species and higher taxa to determine what processes may be responsible for observed differences. The Monogenea includes at least two super-genera (Dactylogyrus and Gyrodactylus) each of which has hundreds of species; no comparable genera are found in the Digenea. Possible reasons for this difference include the higher host specificity of monogeneans and their shorter generation time. If allowance is made for the vagaries of taxonomic 'lumping' and 'splitting', then there are probably comparable numbers of families of monogeneans and digeneans in fishes. However, the nature of the families differ profoundly. Richness in higher taxa (families) in the Digenea is explicable in terms of processes that appear to have been unimportant in the Monogenea. Readily identifiable sources of diversity in the Digenea are: recolonisation of fishes by taxa that arose in association with tetrapods; adoption of new sites within hosts; adoption of new diets and feeding mechanisms; adaptations relating to the exploitation of ecologically similar groups of fishes and second intermediate hosts; and adaptations relating to the exploitation of phylogenetic lineages of molluscs. In contrast, most higher- level monogenean diversity (other than that associated with the subclasses) relates principally to morphological specialisation for attachment by the haptor.  相似文献

Although intrasexual contests generally favor bigger or stronger individuals, the relevance of body size to war of attrition-typedisputes between weaponless animals such as butterflies isunclear. In this study I aimed to investigate the significanceof size in this context by studying territorial contests inHypolimnas bolina (L.), a species that exhibits consistentseasonal plasticity in body size. In this species adult ageis positively correlated with large size in spring but withsmall size in autumn. This shift allowed independent evaluationof the relevance of each variable (size and age) to intrasexualcontest success. Observation of a population of marked individualsindicated that only age appeared important, with the winnersof pair-wise contests significantly older than losers in both seasons, and with contests lasting longer when the age differencebetween the combatants was small. Age was also linked to residency;residents won 99% of all contests. This study suggests thatsize does not matter in these aerial disputes, but age andresidency do. It is not yet possible to determine whether olderbutterflies are intrinsically better competitors, or whether they simply have greater opportunity to find a vacant territory.  相似文献

Aim Species geographic ranges are the ‘fundamental units’ of macroecology. Range size is a major correlate of extinction risk in many groups, and is also critical in studies of biotic responses to climate change. Despite this, there is a lack of studies exploring the role of environmental, historical and anthropogenic processes in determining large‐scale patterns in range size. We perform the first global analysis of putative drivers of range size variation in any group, choosing amphibians as our study taxon. Our aims are to disentangle the many hypothesized causes of range size variation and evaluate support for ‘Rapoport's rule’, the observation that range size correlates with latitude. Location Global. Methods We develop a global map of gridded median range size using the International Union for Conservation of Nature (IUCN) distribution maps. From this we perform spatial and non‐spatial regressions to explore relationships between range size and nine hypothesized variables in six biogeographic realms. We use information‐theoretic model selection to compare multiple competing variables, simultaneously evaluating the relative support for each one. Results Current climate – environmental water and energy, and temperature seasonality – is consistently highly ranked in spatial and non‐spatial analyses. Human impacts and other environmental measures (topographic and landscape complexity, effective area, climate extremes) show mixed support, and glacial history is consistently unimportant. Our findings add further evidence to the view that Rapoport's rule is a regional, not global, phenomenon. Main conclusions The primary importance of temperature seasonality may explain why Rapoport's rule is largely restricted to northern latitudes, as this is where seasonality is most pronounced. More generally, the dominance of contemporary climate in our analyses (even when accounting for space) has stark implications for the future status of amphibians. Changes in climate will almost certainly interact with the anthropogenic processes already threatening a third of amphibians globally, with the effects being most keenly felt by species with a restricted range.  相似文献

This study examined the relationship between otolith size and growth in juvenile cod (Gadus morhua L.). Two groups of juvenile cod were reared under different food ration and temperature regimes to obtain fish of similar somatic size but with different sized otoliths. The two groups were subjected to alternating temperature regimes and intermediate ration levels. Large otoliths grew significantly faster than the small ones and variation between individuals was extensive. The ratio of otolith growth during cold and warm temperature exposure did not differ between groups, and the observed growth pattern is therefore not attributable to differential growth within individual temperature periods. The ratio decreased with otolith size, presumably as a result of ontogenetic decrease in otolith protein composition. These results suggest that processes coupled to the metabolic rate of the endolymphatic epithelium are the key driver behind otolith growth.  相似文献
