 Restriction fragment length polymorphism (RFLP) analysis of mitochondrial DNAs (mtDNAs) from nine Japanese wild isolates of Phellinus linteus was carried out to examine their genetic variability. BamHI and EcoRI digests of mtDNAs from these isolates produced four and five distinct RFLP patterns, respectively. By combining the RFLP patterns obtained with the two endonucleases, mtDNAs from the nine isolates could be assigned to five different genotypes, but no mtDNA variation was detected among the isolates collected from a small area. Distance values calculated among all pairs of mtDNA genotypes, based on the presence or absence of comigrating restriction fragments, were clearly smaller than those among the mtDNA genotypes of Lentinula edodes and Pleurotus ostreatus samples collected worldwide, suggesting the necessity of collecting P. linteus wild isolates for genetic resources from geographically wider areas. Summary It is well documented that chloroplast DNA (cpDNA) recombination occurs at a relatively high frequency during sexual reproduction of unicellular green algae from the Chlamydomonas genus. Like the cpDNAs of most land plants, those of Chlamydomonas species are divided into two single-copy regions by a large inverted repeat sequence, part of which encodes the chloroplast rRNA genes. In the present study, we scored the inheritance of polymorphic loci spanning the entire chloroplast genome in hybrids recovered from reciprocal interspecific and F1 crosses between Chlamydomonas eugametes and C. moewusii, and from these data, estimated the density of recombination junctions within each region of recombinant cpDNAs. Our results indicate that recombination junctions occur at highly variable frequencies across the three main domains of the chloroplast genome. The large inverted repeat sequence was found to exhibit at least a five-fold higher density of recombination junctions compared to one of the singlecopy regions, whereas junctions in the latter region were five-fold more abundant relative to those in the other single-copy region. This marked difference in the densities of recombination junctions implies that the extent of genetic linkage between two given chloroplast loci will depend not only on their physical distance, but also on their locations within the genome.  相似文献

We evaluated the use of amplified fragment length polymorphism (AFLP) markers to distinguish genotypes, populations and species of Lolium. Accessions of two species Lolium perenne and Lolium multiflorum and their hybrid Lolium x hybridum, collected by the Institute of Grass and Environmental Research in 1995 from locations across Portugal, were used. The genetic variation within and between populations from the extremes of latitude and altitude was determined and assessed. Three primer pair combinations generated 765 polymorphic bands. Principal coordinate analysis of similarities between 127 plants showed high dimensionality in the data. Axes 1-3 were associated primarily with species differences, axes 4-14 with population differences within species and axis 15 onwards with within population differences. UPGMA analysis confirmed the groupings. The three populations of L. perenne formed a discrete cluster widely separated from all other populations. There were two distinct groups of L. x hybridum, of which one was similar to and overlapped with L. multiflorum and the second formed a distinct cluster. Analyses of individual bands showed that every inter- and intraspecific contrast involved a different sets of bands, again confirming the high dimensionality of the data. No single band was strictly diagnostic of any population or species. Nevertheless, the UPGMA analysis showed little or no overlap between populations. Thus, despite the high ratio of within-to-between population genetic variance, the full AFLP banding pattern of each genotype is a relatively reliable fingerprint diagnostic of its parent population. The high dimensionality implies that many different factors contribute to the differences observed. This adds to the potential value of the methodology, since it implies that there is a reasonably high likelihood of finding bands relevant to a given environmental gradient or other factor influencing the distribution of genetic diversity.  相似文献

在人或动物的基因组中,存在着类似于小卫星DNA的简单衔接重复单位,如(GACA)_4、(GATA)_4、(TCC)_5、(CA)_8和(CAC)_5等,由于这些重复单位在人或动物基因组中出现的数目和频率不同而表现出多态性。本文用人工合成的寡核苷酸探针(CAC)_5/(GTG)_5,调查了北京地区的50名无关个体,经过统计学处理,计算出无关个体的相关机率是3.8×10~(-10);此外,还对2个家系中的11名成员和1对双胞胎进行了检测,其结果显示子代中的杂交带分别来自父亲和母亲;双胞胎的DNA指纹图完全一致。研究结果表明,(CAC)_5/(GTG)_5探针检出的谱带具有高度的个体特异性(同卵双生子除外),并且谱带在亲代与子代间的传递符合孟德尔遗传规律。  相似文献

The wild Bolivian potato, Solanum berthaultii Hawkes, has been used as a source of resistance to the Colorado potato beetle (CPB), Leptinotarsa decemlineata Say, one of the most significant pests of potato. In this study, two reciprocal backcross S. tuberosum x S. berthaultii potato progenies, BCB and BCT, were mapped with RFLP markers and screened for resistance to CPB consumption, oviposition and defoliation. The genotypic and phenotypic data were combined and analysed to locate quantitative trait loci (QTLs) for resistance to CPB. Three QTLs on three chromosomes in BCB, and two QTLs on two chromosomes in BCT influenced resistance. The QTLs were generally additive but one instance of epistasis was noted. Each QTL accounted for 4–12% of the phenotypic variation observed in resistance. In the more resistant BCB population, a three QTL model explained ca. 20% of the variation in CPB oviposition. When alleles at the three QTLs were homozygous S. berthaultii, oviposition was reduced ca. 60% compared to the heterozygotes. The QTLs for resistance to CPB were compared to those previously identified for the type A and B glandular trichomes, which have been implicated in resistance in the same progenies. Generally, the QTLs for resistance to CPB coincided with loci associated with the glandular trichomes confirming the importance of the glandular trichomes in mediating resistance. However, a relatively strong and consistent QTL for insect resistance in both BCB and BCT on chromosome 1 was observed that was not associated with any trichome traits, suggesting the trichomes may not account for all of the resistance observed in these progenies.  相似文献

Restriction fragment analysis was conducted to determine interspecific chloroplast DNA (cpDNA) variation and genetic relationships among Populus deltoides, P. nigra, P. x canadensis (P. deltoides x P. nigra), and P. maximowiczii. Total cellular DNAs of these poplars were digested with 16 restriction endonucleases, and Southern blots of the restriction digests were probed with six different cloned cpDNA fragments from Petunia. P. deltoides, P. nigra, and P. maximowiczii each had a distinct chloroplast genome, separated by many restriction-site and restriction-fragment-length mutations, predominantly in the large single-copy region of the genome. P. x canadensis shared the same cpDNA restriction fragment patterns as P. deltoides var. deltoides. P. nigra was most diverged from P. deltoides, and P. deltoides showed close cpDNA relationships to P. maximowiczii. Nucleotide substitutions per site in cpDNA were 0.0036 between P. deltoides and P. maximowiczii, 0.0071 between P. nigra and P. maximowiczii, and 0.0077 between P. deltoides and P. nigra. We suggest that P. nigra should be classified in a new separate section, the Nigrae.  相似文献

DNA genetic markers, such as restriction fragment length polymorphisms (RFLPs) and random amplified polymorphic DNA markers (RAPDs), are powerful tools for studying the genetics of plant growth and development. DNA markers are defined sequences of DNA that can be used in traditional linkage mapping. Using DNA marker technology, scientists can uncover relationships between cloned cDNA sequences and classically characterized genes. DNA markers make it possible to dissect the contributions of multiple genetic loci underlying complex developmental processes. Moreover, changes in genome organization that occur during development or in response to environmental signals can be monitored using RFLP technology. In the future, it may be possible to clone any gene based solely on its map position. This will involve the use of tightly linked DNA markers as entry points for chromosome walking, in which a series of overlapping genomic clones reaching from the tightly linked DNA marker to the gene of interest are identified.  相似文献

利用花粉管通道技术对国槐DNA导入花生栽培品种79266,D2代变异株98D010-7经连续自交,D3代获得9个株系,61个株行,641个单株,观察D5代群体内每个单株的开花型、株型、主茎高、第一对侧枝长、分枝数及单株结果数等主要农艺性状的表现型。结果表明:(1)79266和98D010-7均为连续开花型,D5代出现了交替开花型变异株,占总株数4.37%,开花型分离株行占16.39%;(2)79266株型直立,98D010-7株型葡匐,D5代表现型有直立,葡匐和半葡匐3种类型,分别占34.95%、14.04%和51.09%;表现分离的株行占73.77%;(3)株高和第一对侧枝长与79266相比显著降低;(4)单档分枝数与受体平均值相近,只有1个株系分枝数显著少于受体79266,其它8个株系差异不显著;(5)D5代群体有3个株系的单株结果数显著大于受体79266的单株结果数。  相似文献

Summary The Avena sterilis collection in the National Small Grains Collection (NSGC) is an invaluable source of genetic variation to be exploited by oat breeding programs. Prior knowledge of the structure and distribution of genetic variation within the A. sterilis collection would be useful to efficiently screen the collection for valuable traits. To determine genetic structure within a subset of the collection, restriction fragment length polymorphisms were analyzed in a stratified sample of 173 accessions originating in eight countries of Africa and Southwest Asia. Of the 48 probes used for this study 43 detected polymorphism among accessions. The average number of RFLP patterns per probe ranged from 2.9 among Ethiopian accessions to 3.7 among those from Iran. Genetic variation, as measured by genetic distances and polymorphic indexes, was highest in Iran and lowest in Ethiopia. The probability of drawing a genotype from Iran or Iraq that is not present in the more western regions was high, indicating large genetic divergence of the Iran-Iraq accessions from the other regional collections surveyed. Cluster analysis of genetic distances and probabilities of unique genotypes clearly differentiated the eastern region (Iran and Iraq) from the western region (Algeria, Ethiopia, Israel, Lebanon, Morocco, and Syria). The western region could be further subdivided into two clusters, an African cluster (Algeria, Ethiopia, and Morocco) and a southwestern Asia cluster (Israel, Lebanon, and Syria). The molecular methods that are presently being used for studying phylogenetics, phylogeography and population genetics can also be applied to study bird migration. They are powerful and can supplement the information obtained from ringing, telemetry, morphometrics, ringing, radar tracking and isotope analysis. This short review describes the principles, scopes and limitations DNA methods and DNA markers that are relevant for migration research, such as DNA sequences, short tandem repeats (microsatellites), single nucleotide polymorphisms, amplified fragment length polymorphism, inter simple sequence repeats and molecular sexing.  相似文献

Summary Cellular membrane stability (CMS) is a physiological index widely used to evaluate thermostability in plants. The genetic basis of the character has been studied following two different approaches: restriction fragment length polymorphism (RFLP) analysis, and the effects of segregating heat shock protein (HSP) loci. RFLP analysis was based on a set of recombinant inbreds derived from the T32 × CM37 F1 hybrid and characterized for about 200 RFLP loci. Heritability of CMS estimated by standard quantitative analysis was 0.73. Regression analysis of CMS on RFLPs detected a minimum number of six quantitative trait loci (QTL) accounting for 53% of the genetic variability. The analysis of the matrices of correlation between RFLP loci, either within or between chromosomes, indicates that no false assignment was produced by this analysis. The effect of HSPs on the variability of the CMS was tested for a low-molecular-weight peptide (HSP-17) showing presence-absence of segregation in the B73 × Pa33 F2 population. Although the genetic variability of the character was very high (h 2=0.58) the effect of HSP-17 was not significant, indicating either that the polypeptide is not involved in the determination of the character or that its effect is not statistically detectable.  相似文献

Three doubled dihaploid potatoes were hybridised with six tetraploid cultivars and the offspring examined for commercially important characters. Doubled dihaploids used as parents did not reduce the within progeny phenotypic variation for most characters below that found in material produced by intercrossing heterozygous tetraploids. They could however prove useful for improving some characters in a breeding programme. The products of the hybridisations were used to examine the genetic control of discrete variables such as tuber skin pigmentation, tuber flesh colour, and skin russetting. Doubled dihaploids were found to be useful for such studies because they have only three out of the five possible tetraploid genotypes. Presence or absence of red tuber skin in the progenies of cv. Désirée was concluded to be due to the segregation of a dominant major gene which was simplex in the parental cultivar. The yellow flesh of Desiree was found to be recessive. On the hypothesis that russet skin was due to three complementary dominant genes, possible genotypes of the doubled dihaploids and cultivars could also be deduced.  相似文献

We analysed polymorphism of 13 microsatellites in two Indian domesticated pig types (North Indian and Northeast Indian). Heterozygosity, polymorphism information content, and probability of identity of two random individuals were calculated for all microsatellites in both types. The number of alleles observed at a locus varied between five and 12. The evaluated microsatellites exhibited a very high heterozygosity and polymorphism information content. The probability of identity of two random individuals from different populations taking into account all the 13 microsatellites was as low as 3.51 × 10-19. On the basis of these results, we propose that these microsatellite markers may be used with reliability for studying the genetic diversity and for identification of individuals in Indian pig types.  相似文献

Abstract Genomic DNA was extracted from seven species of Verticillium and digested with the restriction endonucleases Eco RI or Hae III. Hybridization with an homologous V. albo-atrum ribosomal RNA gene probe revealed restriction fragment length polymorphisms (RFLPs) which could differentiate V. lateritium, V. lecanii, V. nigrescens, V. nubilum and V. tricorpus . Digestion with Eco RI did not provide RFLPs which could distinguish between V. albo-atrum and V. dahliae . Digestion of genomic and mitochondrial DNA with Hae III showed distinctive patterns on ethidium bromide gels which allowed each species to be distinguished. Some intra-species variation in patterns occurred and a combination of mitochondrial and ribosomal RNA gene complex RFLPs has potential as an aid for the characterization of species and sub-species populations in the genes Verticillium .  相似文献

本文系统地研究了广东地区汉族人群中FⅧ:C基因内BclⅠ,XbaⅠ和BgⅡ位点RFLP的基因频率。多态性位点BclⅠ,XbaⅠ及BglⅠ的切点阳性率分别为63.5%、43.5%和100%。对Bcll和Xbal多态性切点连锁情况研究显示,19.5%的Bcll切点阳性纯合子为Xbal切点杂合子,证明联合应用此两位点RFLP可以把甲型血友病基因连锁分析的有效率提高到65.9%。用RFLP连锁分析对两例甲型血友病家系中的女性进行了致病基因携带者检测,对另一例家系进行了基因产前诊断。  相似文献

A comparison of photosynthetic characteristics of 20 cultivars of grapevine ( Vitis vinifera L. ) from Mallorca (Balearic Islands, Spain) and two widespread cultivars, Cabernet Sauvignon and Chardonnay, was made under irrigation as well as in response to drought. Although these cultivars share a common origin, a high variability was found for several photosynthetic characters under irrigation. Interestingly, these variations were significant for gas-exchange parameters (net CO2 assimilation, stomatal conductance and intrinsic water use efficiency) but not for chlorophyll fluorescence parameters (maximum photochemical efficiency, electron transport rate and non-photochemical quenching). Since water stress is the most limiting factor for plant production under the Mediterranean climate, it is presumable that these findings reflect specific selection pressures over physiological characteristics related to a balance between net carbon gain and water use. Some cultivars presented high carbon assimilation at the expense of a high water loss, whereas others were water savers, accompanied by low CO2 assimilation even under irrigation. Escursach was found to be an interesting cultivar, presenting low water consumption at the same time as reasonably high carbon assimilation. These cultivars also showed different responses to drought, which allowed their classification in two main groups: alarmist cultivars, which showed strong reductions of stomatal conductance in response to relatively low decreases of leaf water potential, and luxurious cultivars, showing low reductions of stomatal conductance under water stress.  相似文献

Nuclear DNA clones and sequence information derived from human genetic analyses were used to detect and characterize intra- and inter-species DNA variation at several nuclear loci in hominoids and cercopithecoids. Restriction fragment length polymorphisms were found at five loci among captive rhesus monkeys. Cross-species polymerase chain reaction (PCR) amplification detected an insertion within the beta-globin gene cluster in hylobatids. The combined use of cross-species PCR and denaturing gradient gel electrophoresis detected both species differences and intra-species polymorphism in the homeobox cluster 2 of hominoids. These results a) demonstrate that DNA clones and nucleotide sequence information from human molecular genetics can be used to facilitate studies of the molecular genetics of nonhuman primates, and b) document specific examples of intra- and inter-species molecular variability at several loci. © 1992 Wiley-Liss, Inc.  相似文献

The polymorphism of 23 microsatellites in the four main cattle breeds in Belgium (Holstein Friesian, Belgian Blue, Belgian Red Pied and East Flemish) was analysed. Heterozygosity, polymorphism information content, the effective number of alleles, exclusion probability and the probability of genotypic identity for two random individuals were calculated for all microsatellites and all breeds. The Belgian Blue breed is generally a little less polymorphic in comparison with the other three breeds. Estimates of the genetic distances between these breeds confirmed the widely accepted proposition that the Belgian Blue is the most genetically distinct of these breeds. The three other breeds are likely to become one population, given current breeding strategies. Exclusion probabilities in parentage control cases are >0·9999 in all four breeds when all 23 microsatellites are used and >0·98 with only the two most polymorphic multiplexes.  相似文献

The Swedish sand lizard (Lacerta agilis) is a relict species from the period of warmth following the last glacial episode and has a fragmented distribution in central Sweden and a more continuous distribution in the southern part of the country. We used this model system of colonization–extinction for a study of genetic variability within and among Swedish populations from different parts of the distribution range using multilocus DNA fingerprinting. The results from the Swedish populations are then contrasted with those from a large Hungarian population in the centre of the species geographical distribution range, which is likely to closely resemble the ancestral founding population of Sweden. Swedish populations have a low level of genetic variability compared with the Hungarian reference population, which showed a genetic variability within the range described for outbred populations. Within the Swedish populations, the average bandsharing was 0.61, the mean heterozygosity 0.45 and the estimated number of alleles 2.7. The figures for the Hungarian population were a bandsharing of 0.19, a heterozygosity of 0.89 and an estimated number of alleles of 9.8. A population bottleneck, common to all Swedish sand lizards, is indicated by less than 20% of the alleles in the Hungarian population being retained in the Swedish populations, and higher bandsharing similarity between different Swedish populations (0.33) as opposed to the Hungarian population (0.19). The limited variability found in Swedish sand lizards is strongly subdivided between populations, with an average FST of 0.32, indicating a very limited gene flow between the isolated populations, as well as between populations in the region where the sand lizard has a more or less continuous distribution (FST = 0.41).  相似文献
