We studied the distribution of the phosphophoryn present in rat incisors by immunolocalization and histochemical techniques. The polyclonal antibody used reacts with both phosphorylated and de-phosphorylated phosphophoryn. Technical problems encountered in immunostaining and in preparing sections from mineralized dentin were resolved by use of peroxidase-conjugated protein A as the "second antibody" in indirect immunostaining reactions and by surface etching of partially demineralized sections. Staining with anti-rat incisor alpha-phosphophoryn antibody showed light staining over the odontoblasts and proximal odontoblastic processes, no stain over the predentin, dense staining over the intertubular dentin, and no stain over the mantle dentin. In the intertubular dentin the stain intensity was directly related to the distribution of mineral. These findings were directly corroborated by staining with Stains All. The mineralization of dentin and the distribution of phosphophoryn within the dentin may be much less uniform than previously supposed.  相似文献

目的:探讨预酸蚀乳牙牙本质对自酸蚀粘接系统粘接强度的影响。方法:随机选取28颗健康乳磨牙,磨除颊舌面釉质,暴露牙本质粘接面,沿近远中向劈开形成56个样本,随机分为7组(n=8)。直接涂布组(A1组,A2组和A3组)分别涂布AdperTM Easy One(AEO),Xeno-V(XV)和OptiBond All In One(AIO)三种自酸蚀粘接剂,预酸蚀组(B1组,B2组和B3组)在涂布三种自酸蚀粘接剂前先使用35%磷酸酸蚀乳牙牙本质15 s,对照组(C组)使用Prime&Bond NTTM(NT)全酸蚀粘接剂,每个样本用Z350复合树脂堆砌成直径为3 mm的树脂小柱,通过剪切试验测试剪切粘接强度,并通过扫描电子显微镜观察断裂表面形态。结果:B1组,B2组的剪切粘接强度值明显高于A1组,A2组(P<0.001);B3组与A3组的剪切粘接强度值比较无明显差别(P=0.94)。A2组的剪切粘接强度值低于C组(P<0.05);B1组的剪切粘接强度值明显高于C组(P<0.001)。扫描电镜观察结果显示各组试件断裂面形态多为牙本质和复合树脂界面破坏。直接涂布组(A1组,A2组和A3组)断裂多发生在混合层的底部,树脂突较少且低于小管口。B1组和B2组试件断裂面可见多数牙本质小管被树脂突填满,断裂多发生在混合层的中上部。B3组试件断裂面可见牙本质小管空虚,树脂突较少。结论:预酸蚀乳牙牙本质可以提高AEO,XV两种自酸蚀粘接剂的剪切粘接强度。自酸蚀粘接剂处理乳牙牙本质可以达到全酸蚀粘接剂处理的粘接强度,但应用自酸蚀粘接剂前预酸蚀乳牙牙本质可以获得更高的粘接强度。  相似文献

Dentin constitutes the major part of human tooth. It is composed of a large number of tubules with both variational radii and radially parallel pattern. In addition, peritubular dentin surrounds each tubule lumen and has a higher elastic modulus than the matrix of dentin, i.e. intertubular dentin. Considering the above microstructural characteristics, a micromechanics model is used in this paper to evaluate the overall elastic properties of dentin. Five independent effective elastic parameters in transverse isotropic elasticity matrix can be expressed analytically by the material parameters of peri- and intertubular dentin and the volume fraction of tubules. To determine the effectivity of this theoretical model, a finite element (FE) model simulating a longitudinal tooth slice in moire fringe testing of Wang and Weiner (J. Biomech. 31 (1998) 135) was performed. Furthermore, the FE model was developed incorporate modeling of variation of tubule's diameter and softer characteristic of intertubular dentin near the dentin-enamel junction and around the pulp chamber. It turned out that the isoline figure of longitudinal displacement by FE calculation has very similar patterns to the moire fringe results. However, the FE results of displacement by traditional stress-strain models which regard dentin as a homogeneous and isotropic material show an obviously different strain distributions as compared to published moire fringes results. Thus the inhomogeneous and anisotropic model developed in this paper more accurately reflects the true physical nature of human dentin.  相似文献

Non‐carious cervical lesions (NCCLs) involve various forms of tooth loss with different etiologies. This study aimed to utilize swept‐source optical coherence tomography (SS‐OCT) at 1300 nm wavelength range in vitro and in vivo to evaluate and clarify the mechanism of NCCLs. In the in vitro phase, a dentin attenuation coefficient (μt) derived from the SS‐OCT signal at NCCL was compared with mineral loss obtained from transverse microradiography (TMR) to determine a μt threshold to discriminate demineralization of cervical dentin in vivo. In the clinical study, 242 buccal surfaces were investigated in 35 subjects. Presence and dimensions of NCCLs, cervical cracking and the degree of demineralization at the exposed cervical dentin were determined using SS‐OCT. Dentin demineralization was observed in 69% of NCCLs. SS‐OCT results confirm that dentin mineral loss and occlusal attrition were associated with larger NCCLs, and can be considered as an etiological factor in formation and progress of these lesions.

The occlusion of dentinal tubules is an effective method to alleviate the symptoms caused by dentin hypersensitivity, a significant health problem in dentistry and daily life. The in situ mineralization within dentinal tubules is a promising treatment for dentin hypersensitivity as it induces the formation of mineral on the sensitive regions and occludes the dentinal tubules. This study was carried out to evaluate the in vitro effect of a whole generation poly(amidoamine) (PAMAM) dendrimer (G3.0) on dentinal tubule occlusion by inducing mineralization within dentinal tubules. Dentin discs were treated with PAMAM dendrimers using two methods, followed by the in vitro characterization using Attenuated total reflection Fourier-transform infrared spectroscopy (ATR-FTIR), X-ray diffraction (XRD), Field emission scanning electron microscopy (FE-SEM) and Energy-Dispersive X-ray Spectroscopy (EDS). These results showed that G3.0 PAMAM dendrimers coated on dentin surface and infiltrated in dentinal tubules could induce hydroxyapatite formation and resulted in effective dentinal tubule occlusion. Moreover, crosslinked PAMAM dendrimers could induce the remineralization of demineralized dentin and thus had the potential in dentinal tubule occlusion. In this in vitro study, dentinal tubules occlusion could be achieved by using PAMAM dendrimers. This could lead to the development of a new therapeutic technique for the treatment of dentin hypersensitivity.  相似文献

Teeth were incompletely demineralized by immersion in unchanged 10% formic acid for 7 days. Reprecipitation deposits of secondary calcium phosphate were present in the dentin and soft tissues of the dental pulp and, if the final pH was 3 or greater, in the remnants of the periodontal ligament. The deposits in the dentin appeared to be intratubular. Deminieralized sections of teeth suspended in supersaturated solutions of brushite contained similar deposits in the soft tissues. It is suggested that reprecipitation of secondary calcium phosphates is a frequent intermediate stage during demineralization with formic acid.  相似文献

To examine whether Cl-coupled HCO3 transport mechanisms were present on the basolateral membrane of the mammalian proximal tubule, cell pH was measured in the microperfused rat proximal convoluted tubule using the pH-sensitive, intracellularly trapped fluorescent dye (2',7')- bis(carboxyethyl)-(5,6)-carboxyfluorescein. Increasing the peritubular Cl concentration from 0 to 128.6 meq/liter caused cell pH to decrease from 7.34 +/- 0.04 to 7.21 +/- 0.04 (p less than 0.001). With more acid extracellular fluid (pH 6.62), a similar increase in the peritubular Cl concentration caused cell pH to decrease by a similar amount from 6.97 +/- 0.04 to 6.84 +/- 0.05 (p less than 0.001). This effect was blocked by 1 mM SITS. To examine the Na dependence of Cl/HCO3 exchange, the above studies were repeated in the absence of luminal and peritubular Na. In alkaline Na-free solutions, peritubular Cl addition caused cell pH to decrease from 7.57 +/- 0.06 to 7.53 +/- 0.06 (p less than 0.025); in acid Na-free solutions, peritubular Cl addition caused cell pH to decrease from 7.21 +/- 0.04 to 7.19 +/- 0.04 (p less than 0.05). The effect of Cl on cell pH was smaller in the absence of luminal and peritubular Na than in its presence. To examine whether the previously described Na/(HCO3)n greater than 1 cotransporter was coupled to or dependent on Cl, the effect of lowering the peritubular Na concentration from 147 to 25 meq/liter was examined in the absence of ambient Cl. Cell pH decreased from 7.28 +/- 0.03 to 7.08 +/- 0.03, a response similar to that observed previously in the presence of Cl. The results demonstrate that Cl/HCO3 (or Cl/OH) exchange is present on the basolateral membrane. Most of Cl/HCO3 exchange is dependent on the presence of Na and may be coupled to it. The previously described Na/(HCO3)n greater than 1 cotransporter is the major basolateral membrane pathway for the coupling of Na and HCO3 and is not coupled to Cl.  相似文献

Thin semi-serial ground sections of coronal dentin were examined radiographically. The bulk of the coronal dentin was characterized by the majority of the tubules having a distinct peritubular zone. With the exception of the tubules running from the tip of the cusp to the pulp cornu, the bulk of peritubular matrix forming the walls of the tubules was disposed eccentrically. The matrix was thicker on the cervical sides of the tubule than it was on the incisal sides. In a relatively narrow layer of the coronal dentin between the bulk of the dentin and the predentindentin border area the thickness of the peritubular matrix varied considerably. It was extremely narrow or absent in some tubules and reached its greatest thickness in others. The tubules in the predentin border area showed little or no evidence of peritubular matrix. The area of dentin beneath the central developmental groove differed somewhat from the bulk of the dentin. Many of the tubules at all levels of this area showed little radiographic evidence of peritubular matrix. Obliterated tubules were seen in some of the sections taken immediately above the predentin-dentin border area in the region of the pulp cornu and were always seen at the junction of the mantle dentin and the circumpulpal dentin beneath the central developmental groove.  相似文献

High‐resolution deformation measurements in a functionally graded hard tissue such as human dentin are essential to understand the unbound water‐loss mediated changes and their role in its mechanical integrity. Yet a whole‐field, 3‐dimensional (3D) measurement and characterization of fully hydrated dentin in both macro‐ and micro‐scales remain to be a challenge. This study was conducted in 2 stages. In stage‐1, a stereo‐digital image correlation approach was utilized to determine the water‐loss and load‐induced 3D deformations of teeth in a sagittal section over consecutively acquired frames, from a fully hydrated state to nonhydrated conditions for a period up to 2 hours. The macroscale analysis revealed concentrated residual deformations at the dentin‐enamel‐junction and the apical regions of root in the direction perpendicular to the dentinal tubules. Significant difference in the localized deformation characteristics was observed between the inner and outer aspects of the root dentin. During quasi‐static loadings, further increase in the residual deformation was observed in the dentin. In stage‐2, dentin microstructural variations induced by dynamic water‐loss were assessed with environmental scanning electron microscopy and atomic force microscopy (AFM), showing that the dynamic water‐loss induced distention of dentinal tubules with concave tubular edges, and concurrent contraction of intertubular dentin with convex profile. The findings from the current macro‐ and micro‐scale analysis provided insight on the free‐water‐loss induced regional deformations and ultrastructural changes in human dentin.   相似文献

High-resolution studies of dental tissues are of considerable interest for biomedical engineering and clinical applications. In this paper, we demonstrate the application of piezoresponse force microscopy (PFM) to nanoscale imaging of internal structure of human teeth by monitoring the local mechanical response to an electrical bias applied via a conductive tip. It is shown that PFM is capable of detecting dissimilar components of dental tissues, namely, proteins and calcified matrix, which have resembling morphology but different piezoelectric properties. It is demonstrated that collagen fibrils revealed in chemically treated intertubular dentin exhibit high piezoelectric activity and can be visualized in PFM with spatial resolution of 10 nm. Evidence of the presence of protein inclusions of 100-200 nm wide and several micrometers long in tooth enamel has been obtained. Furthermore, it is found that the peritubular dentin and intertubular dentin exhibit different piezoelectric behavior suggesting different concentration of collagen fibrils. The obtained results demonstrate a high potential of PFM in providing an additional insight into the structure of dental tissues. It is suggested that the PFM approach can be used to study the structure of a wide range of biological materials by monitoring their electromechanical behavior at the nanoscale.  相似文献

EDTA-insoluble organic structures of the hypercalcified peritubular matrix (PM) in horse dentine were observed by scanning electron microscopy. The PM was enveloped in double cylindrical structures composed of fibrillar sheaths in the inner and outer peripheries. Between the outer fibrillar sheath and intrinsic fibrils of the intertubular matrix, a calcified cementing membrane existed. Within the PM, warped cone-shaped structures of fibrillar sheaths, overlapping at intervals of 4-6 microns and semiconcentrically surrounding the dentinal tubule, extended from the inner fibrillar towards the outer fibrillar sheath. The cone-shaped fibrillar sheaths following the inner and outer fibrillar sheaths were identified as the incremental lines of the PM. Most of these fibrils may be collagen although it could not be confirmed, whereas non-collagenous organic materials in the lateral branches of the dentinal tubule are radially arranged in the PM. These EDTA-insoluble structures were three-dimensionally illustrated using an image-analysing system.  相似文献

Teeth contain several calcified tissues with junctions that provide interfaces between dissimilar tissues. These junctions have been difficult to characterize because of their small size. In this work a new technique using a combination of atomic force microscopy (AFM) and a force-displacement transducer was used to simultaneously study the surface topography and map mechanical properties of the junctions and adjacent hard tissues. Prepared specimens from human third molars were scanned by an AFM piezo-tube in contact mode. To measure the dynamic viscoelastic properties of the material a small sinusoidal force was superimposed on the contact force and the resulting displacement amplitude and the phase shift between the force and amplitude were measured. This force modulation technique was used to map the local variation of nanomechanical properties of intertubular dentin, peritubular dentin, enamel, dentin-enamel junction (DEJ) and peritubular-intertubular dentin junction (PIJ). This new technique allowed us to measure the widths of these junctions in addition to local variation in dentin and enamel without causing plastic deformation to the material and with 2 orders of magnitude increase in spatial resolution compared with previous studies that used discrete nanoindentation techniques. Due to the ability to analyze the sample line-by-line, the distribution functions associated with the width of the DEJ and PIJ were conveniently obtained for specific intratooth locations. The data suggested, for three third molar specimens, a DEJ width of 2-3 microm with full-width half-maximum (FWHM) of 0.7 microm and PIJ width of 0.5-1.0 microm with 0.3 microm FWHM. The intertubular dentin storage modulus variation was between 17 and 23 GPa with a mean value of 21 GPa. The range of storage modulus for enamel near the DEJ was between 51 and 74 GPa with a mean value of 63 GPa.  相似文献

Experimental periodontal regeneration studies have revealed the weak binding of repair cementum to the root surface, whereas attachment of cementum to dentin preconditioned by odontoclasts appears to be superior. The aim of this study has been, therefore, to analyze the structural and partial biochemical nature of the interface that develops between resorbed dentin and repair cementum by using human deciduous teeth as a model. Aldehyde-fixed and decalcified tooth samples were embedded in acrylic or epoxy resins and sectioned for light and transmission electron microscopy. Antibodies against bone sialoprotein (BSP) and osteopontin (OPN), two noncollagenous proteins accumulating at hard tissue interfaces in bone and teeth, were used for protein A-gold immunocytochemistry. Light microscopy revealed a gradually increasing staining intensity of the external dentin matrix starting after the withdrawal of the odontoclast. Labeling for both BSP and OPN was first detected among the exposed collagen fibrils and in the intratubular dentin matrix when odontoclasts had withdrawn but mesenchymal cells were present. Subsequently, collagen fibrils of the repair cementum were deposited concomitantly with the appearance of labeling for BSP and OPN over the intratubular, intertubular, and peritubular dentin matrix. Labeled mineralization foci indicated the advancing mineralization front, and the collagenous repair matrix became integrated in an electron-dense organic material that showed labeling for BSP and OPN. Thus, no distinct planar interfacial matrix layer lies between the resorbed dentin and the repair cementum. In this study, simultaneous diffraction and fluorescence mapping with a (250 nm)2, 10.1 keV synchrotron X-ray beam investigated the spatial distribution of carbonated apatite (cAp) mineral and elemental Ca (and other cations including Zn) around dentin tubules. In 1 μm thick sections of near-pulp root dentin, where peritubular dentin (PTD) is newly forming, high concentrations of Zn, relative to those in intertubular dentin (ITD), were observed adjacent to and surrounding the tubule lumens. Some but not all tubules exhibited hypercalcified collars (high Ca signal relative to the surrounding ITD), and, when present, the zone of high Ca did not extend around the tubule. Diffraction rings from cAp 00.2 and 11.2 + 21.1 + 30.0 reflections were observed, and cAp was the only crystal phase detected. Profiles of Ca, Zn and cAp diffracted intensities showed the same transitions from solid to tubule lumen, indicating the same cAp content and organization in ITD far from the tubules and adjacent to them. Further, the matching Ca and diffraction profiles demonstrated that all of the Ca is in cAp or that any noncrystalline Ca was uniformly distributed throughout the dentin. Variation of 00.2 and 11.2 + 21.1 + 30.0 diffracted intensity was consistent with the expected biaxial crystallographic texture. Extension of X-ray mapping from near 1 μm resolution to the 250 nm level, performed here for dentin and its tubules, will provide new understanding of other mineralized tissues.  相似文献

To investigate age and site-related changes to human dentin collagen, sound human teeth collected from donors aged 13–29 (young) and 50–74 (aged) years (n = 9/group) were cut to shallow and deep sites. Dentin collagen orientation and fibril bundling was investigated using the Picrosirius Red (PSR) stain observed under cross-polarized light microscopy (Pol), and collagen distribution was investigated using Confocal Laser Scanning Microscopy (CLSM). Collagen types III to I distribution in peritubular dentin (PTD) was revealed using Herovici stain and brightfield microscopy. Image analysis software and linear mixed modelling quantified outcomes. In situ dentin collagen was observed using Xenon Plasma Focussed Ion Beam Scanning Electron Microscopy (Xe PFIB-SEM). The PSR-Pol analysis revealed less coherently aligned and more bundled collagen fibrils in aged dentin (P = 0.005). Deep inner dentin collagen in both groups were less coherently aligned with reduced bundling. Regardless of age, CLSM showed collagen distribution remained stable; and more collagen type III was detectable in PTD located in inner dentin (Young: P = 0.006; Aged: P = 0.008). Observations following Xe PFIB-SEM cross-sectioning showed apatite-like deposits surrounding large intratubular collagen fibers, and evidence of smaller intertubular dentin collagen fibrils in situ. In conclusion, aging changes collagen network architecture, but not distribution or content.  相似文献

Differences in dentin microstructure have been used as a tool for dietary reconstruction; however, the extent that diet is associated with this aspect of dental morphology has yet to be empirically tested. We conducted microhardness tests of mammalian dentin sections, hypothesizing that species with adaptations to particularly hard diets would have softer dentin, owing to a higher proportion of soft intertubular dentin. Species adapted to abrasive diets, in contrast, should have harder dentin, resulting from a higher proportion of hypermineralized peritubular dentin. We examined molar dentin hardness in ten mammalian taxa with durophagous diets, abrasive diets, and a comparative “control” group of mechanical generalists. Samples included six primate taxa and four non-primate species representing various dietary regimes. Our results reveal significant variation among taxa in overall hardness, but the data do not distinguish between hard and abrasive diets. Several taxa with generalized (i.e., mechanically diverse) diets resemble each other in exhibiting large variance in hardness measurements and comparably soft dentin. The high variation in these species appears to be either a functional signal supporting the niche variation hypothesis or indicate the absence of sustained unidirectional selective pressure. A possible phylogenetic signal of dentin hardness in the data also holds promise for future systematic investigations.  相似文献

The dentin layer of the tooth is a complex mineralized tissue traversed by a closely packed system of tubules. Each tubule is surrounded by highly mineralized tissue referred to as peritubular dentin (PTD). The remaining mineralized collagen network between the tubules is the intertubular dentin (ITD). A TOF-SIMS analysis of the PTD constituents has been used to compare the PTD to the ITD. The PTD differs from the ITD not only in the degree of mineralization but also in the amount and nature of the mineral elements and amino acids. The organic matrix of the PTD consists of a unique collagen free assembly of proteins rich in glutamic acid, where the ITD organic matrix is collagen-rich and Asp-rich. The apparent concentration of organic fragment ions observed in the PTD in the TOF-SIMS negative ion mode was much higher than expected. The PTD was found to be rich in Ca2+, Na+, Mg2+, and K+. The amount of Mg2+ and K+ in the PTD was significantly reduced after deproteination, while Ca2+ and Na+ were still accumulated in the PTD. This implies that Mg2+ and K+ are mainly associated with the organic matrix rather than with the mineral of the PTD.  相似文献

Chitin is an organic polymer and it is the most frequent marine natural polysaccharide after cellulose. The main natural sources of chitin are exoskeletons of insects, mollusks, the cell walls of certain fungi and crustaceans such as crabs, shrimps and lobsters. The waste of these marine exoskeletons are pollutant for the environment, but these waste raw materials could be useful for production of commercial products like chitin. Chitin is an important raw material used for water treatment, agricultural, biomedical, biotechnological purposes, food and paper industry and cosmetics. Based on the variety of importance, the present targets of this study are to optimize the demineralization process for the removal of calcium and phosphate contents from the waste of Portunidae segnis (P. segnis) by using acid at ambient temperature and to characterize the isolated demineralized sample as well as the percentage of remaining calcium and phosphorus contents by using Inductively Coupled Plasma-Optical Emission Spectrometry (ICP-OES). The prepared waste carbs coarse powder samples of P. segnis were demineralized with seven different concentrations of hydrochloric acid at ambient temperature for 1 h. All the demineralization samples by the different concentrations were analyzed by using sensitive ICP-OES. The results based on ICP-OES showed that among the seven different concentrations used in the demineralization process for the isolation of chitin, the best was 2 M of HCl concentration for the production of chitin. The results also showed that the optimized concentration 2 M HCl gave the minimum concentration of calcium and phosphorus compared to other concentrations applied in this experiment. In conclusion, the optimized concentration for demineralization process could be used commercially for the isolation or commercial production of chitin for agricultural, biomedical and biotechnological purposes.  相似文献

Dentin is a biocomposite possessing elegant hierarchical structure, which allows it to resist fracture effectively. Despite the considerable efforts to unravel the peculiar fracture behavior of dentin, the effect of microstructural features on the fracture process is largely unknown. In this study, we explore the interaction between the primary crack with crack tip located in intertubular dentin (ITD) and microcracking of peritubular dentin (PTD) ahead of the primary crack. A micromechanical model accounting for the unique composite structure of dentin is developed, and computational simulations are performed. It is found that the microcracking of PTD located in the crack plane in front of the primary crack tip can promote the propagation of the primary crack, increasing the propensity of coalescence of primary crack and microcracks nucleating in PTD. We show that the two-layer microstructure of dentin enables reduction in driving force of primary crack, potentially enhancing fracture toughness. The high stiffness of PTD plays a critical role in reducing the driving force of primary crack and activating microcracking of PTD. It is further identified that the microcracking of PTD arranged parallel to the crack plane with an offset could contribute to the shielding of primary crack.  相似文献

We examined immunocytochemically the type and distribution of glycosaminoglycans and proteoglycans (PG) in predentin and dentin demineralized with EDTA after aldehyde fixation of rat incisors using (a) four monoclonal antibodies (1-B-5,9-A-2,3-B-3, and 5-D-4) which recognize epitopes in unsulfated chondroitin (C0-S), chondroitin 4-sulfate (C4-S), chondroitin 6-sulfate (C6-S), and keratan sulfate (KS) associated with the PG, and (b) monoclonal (5-D-5) and polyclonal antibodies specific for the core protein of large and small dermatan sulfate (DS) PG. Light microscope immunoperoxidase staining after pre-treatment of tissue sections with chondroitinase ABC localized the majority of stainable PG (C4-S, KS, DSPG, C0-S, and C6-S) in predentin and, to a lesser extent (C4-S and small DSPG), in the dentin matrix. The former site demonstrated relatively homogeneous PG distribution, whereas the latter site revealed that strong staining of C4-S and small DSPG was confined mostly to dentinal tubules surrounding odontoblastic processes, with only weak staining in the rest of the dentin matrix. These results indicate that there is not only a definite difference between PG of predentin and dentin but also a selective decrease in the concentration or alteration of these macromolecules during dentinogenesis and mineralization.  相似文献
