Soluble microbial products (SMP) are organics produced by microorganisms as they degrade substrates. The available literature does not reveal how SMP affect and regulate microbial activities. In this study, we monitored variations in pH, dissolved oxygen concentration, soluble biological and chemical oxygen demands (sBOD5 and sCOD) as a measure of microbial activity in synthetic wastewater. Aerobic degradation tests were carried out under the following conditions: aeration, 1,500 cm3 /min; initial sBOD5, 515±5 mg/l; initial sCOD, 859±6 mg/l; initial biomass concentration (defined as mixed liquor suspended solids), 1,200±25 mg/l; sludge retention time, 24 h; and temperature, 20±1°C. The study involved non-acclimated biomass (R0 flora), biomass developed in the presence of SMP (R1 flora), and biomass developed in reduced level of SMP (R2 flora). We also determined which of these flora produced more refractory SMP. The results showed that R2 flora utilized the synthetic feed more quickly, and produced less refractory organic matter than R0 and R1 flora. The production of more refractory organics by R0 and R1 flora shows that not all the biomass was active. R1 flora degraded the substrates irregularly, suggesting that some microbes were dependent on the metabolic products of those that could utilize the feed components. These results show that production of SMP also depends on the prior substrates and on the ability of the flora to respond to changes in substrate composition.  相似文献

The kinetics of soluble microbial product (SMP) formation under substrate-sufficient conditions appear to exhibit different patterns from substrate-limited cultures. However, energy spilling-associated SMP formation is not taken into account in the existing kinetic models and classification of SMP. Based on the concepts of growth yield and energy uncoupling, a kinetic model describing energy spilling-associated SMP formation in relation to the ratio of initial substrate concentration to initial biomass concentration (S 0/X 0) was developed for substrate-sufficient batch culture of activated sludge, and was verified by experimental data. The specific rate of energy spilling-associated SMP formation showed an increasing trend with the S 0/X 0 ratio up to its maximum value. The SMP productivity coefficient (α p/e) was defined from the model on the basis of energy spilling-associated substrate consumption. As a part of the investigations on the microbial lipid production using the yeast Rhodotorula gracilis, CFR-1, kinetics of the biomass synthesis has been studied using shake flask experiments. Using a medium containing a carbon to nitrogen ratio of 701, the rates of biomass production were followed at different initial substrate concentrations in the range of 20–100 kg/m3. A carbon mass balance methodology employing minimal measurements was applied to heterotrophic and photoheterotrophic marine bacteria grown under constant dilution and exposed to 12-h intervals of light or darkness. Carbon mass balance calculations using measurements taken every 3 h closed to within 93–103% using dissolved organic carbon, biomass carbon and CO2 production data only, indicating that background interference from dissolved inorganic carbon variations in the amended seawater medium was not significant. Neither strain was observed to sustain a net CO2 fixation using paramagnetic measurement of oxygen uptake rates (OUR), indicating a need for more sensitive on-line measurement techniques for OUR. Photoheterotrophic growth demonstrated lower carbon-mole biomass yields (0.41±0.026 vs. 0.64±0.013 mol mol–1) despite higher specific glucose uptake rates (0.025 vs. 0.02 mol mol–1 h–1), suggesting that bioreactor-based study of marine bacteria can present growth modes that are different from those encountered in the marine environment.  相似文献

There are a number of controversies surrounding both biomass estimation and carbon balance in tropical forests. Here we use long-term (from 1978 through 2000) data from five 0.5-ha permanent sample plots (PSPs) within a large tract of relatively undisturbed Atlantic moist forest in southeastern Brazil to quantify the biomass increment (MI), and change in total stand biomass (Mstand), from mortality, recruitment, and growth data for trees 10 cm diameter at breast height (DBH). Despite receiving an average of only 1,200 mm annual precipitation, total forests biomass (334.5±11.3 Mg ha–1) was comparable to moist tropical forests with much greater precipitation. Over this relatively long-term study, forest biomass experienced rapid declines associated with El Niño events, followed by gradual biomass accumulation. Over short time intervals that overlook extreme events, these dynamics can be misinterpreted as net biomass accumulation. However for the 22 years of this study, there was a small reduction in forest biomass, averaging –1.2 Mg ha–1 year–1 (±3.1). Strong climatic disturbances can severely reduce forest biomass, and if the frequency and intensity of these events increases beyond historical averages, these changing disturbance regimes have the capacity to significantly reduce forest biomass, resulting in a net source of carbon to the atmosphere.  相似文献

Sugar cane bagasse hemicellulosic fraction was hydrolysed by treatment with 70 mg of sulphuric acid per gram of dry mass at 125 °C for 2 h. The hydrolysate was used as the substrate to grow Candida langeronii RLJ Y-019 at 42 °C; initial pH 6.0; stirring at 700 rev/min and aeration at 1.0 and 2.0 v/v/min. The utilization of D-xylose, L-arabinose, and acetic acid were delayed due to the presence of D-glucose, but after D-glucose depletion the other carbon sources were utilized. The kinetic parameters calculated for both cultivations at 1.0 and 2.0 v/v/min included: maximum specific growth rate (max) of 0.29 ± 0.01 h–1 and 0.43 ± 0.016 h–1, yields (Y x/s) of 0.36 ± 0.012 and 0.40 ± 0.012 gx/gs and productivity (Q x) of 0.81 ± 0.016 and 0.97 ± 0.012 gx/l/h, respectively, and compared favourably with published results obtained with Candida utilis and Geotrichum candidum. Candida langeronii appeared superior to C. utilis for biomass production from hemicellulose hydrolysate, in that it utilized L-arabinose and was capable of growth at higher temperatures. The biomass contained 48.2, 1.4, 5.8 and 23.4% of total protein, DNA, RNA and carbohydrate, respectively and contained essential amino acids for animal feed.  相似文献

Soluble microbial products (SMP) are ubiquitously present in the effluents of biological wastewater treatment systems. In sequencing batch reactor (SBR) systems, effects of influent concentration and temperature on the amount and the molecular weight (MW) distribution of SMP were investigated for the two substrates, glucose and phenol. The values of effluent SMP/S0 of phenol were higher than those of glucose at different influent concentrations and temperatures. It was found that the effluent SMP (Se) was linearly correlated to the influent total organic carbon (TOC) (S0) for both substrates. The slope and intercept of the equation were affected by the temperature. According to the analysis of the MW distribution, it was shown that there exists a bimodal pattern with the majority of SMP having a MW<1 kDa or >10 kDa. The low MW fraction (<1 kDa) amounts to 47.3–70.4% of the effluent SMP. The high MW fraction (>10 kDa) slightly fluctuates in the range of 21.2–32.8% of the effluent SMP.  相似文献

The aim of this project has been to study and compare the ciliate populations present in roptating biological reactors treating three different wastewaters. Wastewaters chosen were a maize mill (nejayote), a sugarcane/ethyl alcohol plant (vinasses) and a recycled paper mill (whitewaters). The initial dissolved organic contents, measured as soluble chemical oxygen demand (COD) and biochemical oxygen demand in five days (BOD5), were 2040±150 mg COD L–1 and 585±5 mg BOD5L–1 for nejayote; 2000±20 mg COD L–1 and 640±5 mg BOD5 L–1 for vinasses and 960±200 mg COD L–1 and 120±10 mg BOD5 L–1 for whitewaters. Results obtained indicate that ciliated protozoa proliferated in the different chambers of each rotating biological reactor (RBR). Saprobity indices, as a quantitative evaluation parameter, indicate that there are no universal species of ciliates associated with specific BOD5 concentrations. Therefore, the number of species of ciliates present in the effluent indicate qualitatively the efficiency of removal of pollution from the wastewaters during treatment in the rotating biological reactors.  相似文献

Two capsulated, exopolysaccharide-producing cyanobacteria, Cyanospira capsulata and Nostoc PCC7936, were tested with regard to their metal removal capability by using copper as model metal. The experiments, carried out with the sole cyanobacterial biomass suspended in distilled water and confined into small dialysis tubings, showed that C. capsulata biomass is characterized by the best efficiency in metal removal, with a qmax (maximum amount of copper removed per biomass unit) of 96 ± 2 mg Cu(II) removed per g of protein in comparison with the value of 79 ± 3 of Nostoc PCC7936 biomass. The experimental data obtained with both cyanobacterial biomass best fit the Langmuir sorption isotherm. The sorption of copper started from the first minutes of contact with the metal and attained the equilibrium state, when no more copper removal was evident, after 5 and 6 hours, for C. capsulata and Nostoc PCC7936, respectively. The best efficiency in Cu(II) removal was obtained at pH 6.1–6.2, while the presence of Mg2+ or Ca2+ reduced copper removal capability of both species to 60–70% of their qmax. The results showed that the biomass of C. capsulata and Nostoc PCC7936 possesses a high affinity and a high specific uptake for copper, comparable with the best performances shown by other microbial biomass, and suggest the possibility to use the capsulated trichomes of the two cyanobacteria for the bioremoval of heavy metals from polluted water bodies.  相似文献

In order to investigate the influence of hydraulic retention time (HRT) on organic pollutant removal in a submerged membrane bioreactor (SMBR), a laboratory-scale experiment was conducted using domestic sewage as influent. The dissolved oxygen (DO) concentration was controlled at 2.0– during the experimental period. The experiments demonstrated that when HRT was 3, 2 and 1 h, the reduction of chemical oxygen demand (COD) was 89.3–97.2, 88.5–97.3 and 80–91.1%, and the effluent COD was 38.9–11.2, 41.6–10.8 and 63.4–, respectively. It is suggested that an HRT of 1 h could meet the normal standard of discharged domestic sewage, and an HRT of 2 h could meet that of water reclamation. In addition, we use mathematical software MATLAB to analyse the relation of mixed liquor suspended solids (MLSS) and COD removal. The results showed that the optimum MLSS concentration should be maintained at around in the SMBR. The results also showed that the COD removal was related to HRT (τ), influent concentration (S0) and sludge loading rate for COD removal (NS). Moreover, the high COD removal could be achieved through adjusting τ, S0 and NS.  相似文献

Biomass behaviour and COD removal in a benchscale activated sludge reactor have been studied alternating anaerobic and aerobic conditions. Particular attention has been paid to the influence of the ratio of the initial substrate concentration (S 0) to the initial biomass concentration (X 0) on the reactor performance. Tests at very low ratios (S 0/X 0<2) demonstrate the existence of a threshold below which the reactor performance is seriously affected (S 0/X 0=0.5). Under conditions of total suppression of cell duplication, substrate maintenance requirements have also been calculated for the microbial consortium present in the activated sludges. A pilot-scale test was conducted in a submerged membrane bioreactor (SMBR) for 452 days to treat high-strength traditional Chinese medicine wastewater from two-phase anaerobic digest effluent. This study focuses on the effects of operational parameters on effluent quality of a SMBR. The parameters include shorter hydraulic retention time (HRT), higher influent COD concentration, higher COD loading rate and mixed liquor suspended solids (MLSS). The experimental results demonstrated that when HRT was 5 h and the influent COD was less than 3000 mg L−1, the effluent quality of the SMBR evaluated from its COD content (CODfilt) could meet the accepted Chinese standards for water reclamation; when HRT was 3.2 h and the influent COD was less than 3000 mg L −1, or HRT was 5 h and the influent COD fluctuated between 3000 and 6000 mg L−1, the effluent quality of the SMBR could meet the normal Chinese discharged standard. Statistical analyses showed that CODfilt correlated positively with the COD loading rate. It correlated negatively with the MLSS for MLSS values between 7543 and 13 694 mg L−1. When MLSS was >13 694 mg L−1 it correlated positively with CODfilt. Based on experimental values from SMBR and on values predicted by a simulation model generated using the back propagation neural network (BPNN) theory, the optimum operational parameters for the treatment of a high-strength TCM wastewater were as follows: HRT was 5 h, SRT was 100 day, COD loading rate was<20.5 kg m−3 d−1, the range of MLSS was 7543–13 694 mg L−1.  相似文献

The applicability of a recently published modification of the chemostat, named titrostat, for microbial continuous-flow purification of toluene-contaminated air is discussed. This article describes the operative range and the toluene elimination efficiency of a 2-l titrostat running with a mixed bacterial culture dominated by two Acinetobacter species: A. calcoaceticus and A. radioresistens. The study focuses on the kinetics and stoichiometry of the process. Special attention is paid to the peculiarities of toluene as an unconventional growth substrate having high carbon and energy content. Removal productivity as high as 2.24 g l–1 h–1 with 99.9% elimination efficiency was observed at air flow rate 60 l h–1, temperature 32°C, pH 6.2, toluene concentration in the inlet air 37.4 mg l–1 and titrant solution containing NH3 at 1.87 g l–1. The maximum biomass yield from assimilated toluene, Y s m=0.880±0.011, and a rate of substrate expenditures for cell maintenance, m s=0.022±0.002 h–1, were estimated.  相似文献

Microorganisms catalyse the reaction and in this study, mainly the effect of different concentration of biomass on COD removal was investigated. Three sets of two-compartment reactors were established. The cation exchange membrane (CEM) was employed in each reactor and 0.5 V of electricity was supplied. Graphite rod employed in cathodic part and a combination of graphite rod and graphite granules were used in anodic chamber. The highest rate of COD removal (40 ± 2.0 ppm/h) was achieved in the reactor which had initial VSS at 6130 mg/l, whereas the slowest rate of 23 ± 1.2 ppm/h in the reactor started with 3365 mgVSS/l. Some ammonia removal was also noticed during the operation. Further understanding and improvement is needed to be competitive against traditional wastewater treatment processes.  相似文献

Removal of toluene in waste gases using a biological trickling filter   总被引:12,自引:0,他引:12  
The removal of toluene from waste gas was studied in a trickling biofilter. A high level of water recirculation (4.7 m h–1) was maintained in order to keep the liquid phase concentration constant and to achieve a high degree of wetting. For loads in the range from 6 to 150 g m–3 h–1 the maximum volumetric removal rate (elimination capacity) was 35±10 g m–3 h–1, corresponding to a zero order removal rate of 0.11±0.03 g m–2 h–1 per unit of nominal surface area. The surface removal was zero order above the liquid phase concentrations of approximately 1.0 g m–3, corresponding to inlet gas concentrations above 0.7–0.8 g m–3. Below this concentration the surface removal was roughly of first order. The magnitude of the first order surface removal rate constant, k1A , was estimated to be 0.08–0.27 m h–1 (k1A a=24–86 h–1). Near-equilibrium conditions existed in the gas effluent, so mass transfer from gas to liquid was obviously relatively fast compared to the biological degradation. An analytical model based on a constant liquid phase concentration through the trickling filter column predicts the effluent gas concentration and the liquid phase concentration for a first and a zero order surface removal. The experimental results were in reasonable agreement with a very simple model valid for conditions with an overall removal governed by the biological degradation and independent of the gas/liquid mass transfer. The overall liquid mass transfer coefficient, KLa, was found to be a factor 6 higher in the system with biofilm compared to the system without. The difference may be explained by: 1. Difference in the wetting of the packing material, 2. Mass transfer occurring directly from the gas phase to the biofilm, and 3. Enlarged contact area between the gas phase and the biofilm due to a rough biofilm surface.  相似文献

This study investigated characteristics of a sequencing batch reactor (SBR) system which was varied with respect to sludge retention time (SRT) (5.9, 8.2, 10.5, 12.2, and 16.2 days). The removal efficiencies of chemical oxygen demand (COD) were more than 90% under all SRT conditions, and the greatest efficiency (92.2%) occurred with a SRT of 16.2 days. As the SRT increased, the denitrification rate per mixed liquor suspended solids (MLSS) during the anoxic(I) period decreased significantly from 166.3 mg NO(X)(-)-N/g MLSS d to 68.8 mg NO(X)(-)-N/g MLSS d. As the SRT increased, the phosphorus removal efficiency decreased from 47.1% (SRT of 5.9 days) to 31.0% for a SRT of 16.2 days, because active phosphate release and uptake occurred under shorter SRT conditions. The mass balance of nitrogen (with respect to nitrogen in the influent) at a SRT of 16.2 days (the highest nitrogen removal efficiency) showed 14.9% of nitrogen was removed in clarified water effluent, 49.7% was removed by the sludge waste process and 33.3% was removed by denitrification. Nitrogen processing was well accounted for in the SBR system as the nitrogen mass balance was close to 100% (97.9%).  相似文献

Removal of Cr(VI) from ground water by Saccharomyces cerevisiae   总被引:1,自引:0,他引:1  
Chromium can be removed from ground water by the unicellular yeast, Saccharomyces cerevisiae. Local ground water maintains chromium as CrO4 2- because of bicarbonate buffering and pH and E h conditions (8.2 and +343 mV, respectively). In laboratory studies, we used commercially available, nonpathogenic S. cerevisiae to remove hexavalent chromium [Cr(VI)] from ground water. The influence of parameters such as temperature, pH, and glucose concentration on Cr(VI) removal by yeast were also examined. S. cerevisiae removed Cr(VI) under aerobic and anaerobic conditions, with a slightly greater rate occurring under anaerobic conditions. Our kinetic studies reveal a reaction rate (Vmax) of 0.227 mg h-1 (g dry wt biomass)-1 and a Michaelis constant (Km) of 145 mg/l in natural ground water using mature S. cerevisiae cultures. We found a rapid (within 2 minutes) initial removal of Cr(VI) with freshly hydrated cells [55–67 mg h-1 (g dry wt biomass)-1] followed by a much slower uptake [0.6–1.1 mg h-1 (g dry wt biomass)-1] that diminished with time. A materials-balance for a batch reactor over 24 hours resulted in an overall shift in redox potential from +321 to +90 mV, an increase in the bicarbonate concentration (150–3400 mg/l) and a decrease in the Cr(VI) concentration in the effluent (1.9-0 mg/l).  相似文献

Summary Fermentation yields of Lactobacillus plantarum were measured at controlled pH between 4.0 and 8.0 and initial lactate concentrations of 0–90 g/l. Optimal growth conditions at pH 6.0 without addition of lactate gave a growth rate of 0.57 h–1 and 20 g dry biomass/mol ATP formed (Y ATP). The pH variations resulted in a decrease in growth rate but the effect on Y ATPwas insignificant. The addition of lactate to the medium at 0 h resulted in linear decrease in the growth rate of the culture, and all the metabolic activities were completely inhibited at 110 g/l. Summary Cell K activity,a k, was measured in the short-circuited frog skin by simultaneous cell punctures from the apical surface with open-tip and K-selective microelectrodes. Strict criteria for acceptance of impalements included constancy of the open-tip microelectrode resistance, agreement within 3% of the fractional apical voltage measured with open-tip and K-selective microelectrodes, and constancy of the differential voltage recorded between the open-tip and the K microelectrodes 30–60 sec after application of amiloride or substitution of apical Na. Skins were bathed on the serosal surface with NaCl Ringer and, to reduce paracellular Cl conductance and effects of amiloride on paracellular conductance, with NaNO3 Ringer on the apical surface.Under control conditionsa k r was nearly constant among skins (mean±SD=92±8mM, 14 skins) in spite of a wide range of cellular currents (5 to 70 A/cm2). Cell current (and transcellular Na transport) was inhibited by either apical addition of amiloride or substitution of Na by other cations. Although in some experiments the expected small increase ina k r after inhibition of cell current was observed, on the average the change was not significant (98±11mM after amiloride, 101±12mM after Na substitution), even 30 min after the inhibition of cell current. The membrane potential, which in the control state ranged from –42 to –77 mV, hyperpolarized after inhibition of cell current, initially to –109±5mV, then depolarizing to a stable value (–88±5mV) after 15–25 min. At this time K was above equilibrium (E k=98±2mV), indicating that the active pump mechanism is still operating after inhibition of transcellular Na transport.The measurement ofa k r permitted the calculation of the passive K current and pump current under control conditions. assuming a constant current source with almost all of the basolateral conductance attributable to K. We found a significant correlation between pump current and cell current with a slope of 0.31, indicating that about one-third of the cell current is carried by the pump, i.e., a pump stoichiometry of 3Na/2K.  相似文献
