Abstract

This paper provides a general review of the nature of tolerance to alcohol and describes specific studies designed to evaluate genetic influences on the acquisition of tolerance. Both environmental and genetic factors are involved in acquisition of either acute or chronic tolerance. Studies showing inbred strain differences in rates of acquisition of tolerance in mice are prima facie evidence for genetic influences, but the possible interactions between genes and environmental factors have not been investigated. Potential relationship between acutely and chronically acquired tolerance to alcohol is discussed.  相似文献

Opioids are extensively used for the management of both chronic malignant and non malignant pains. One major serious limitation associated with chronic use of opioids is the development of tolerance to its analgesic effect. The effect of Bacopa monnieri, a renowned ayurvedic medicine for acquisition and expression of morphine tolerance in mice, was investigated. Bacopa monnieri, n-Butanol fraction was analyzed on High performance liquid chromatography (HPLC), for Bacopaside A major components i.e. Bacoside A3, Bacopaside ll and Bacosaponin C. Antinociceptive effect of n-Butanol extract of Bacopa monnieri (n Bt-ext BM) (5, 10 and 15 mg/kg) was assessed on hot plate. Effect of different doses of n Bt-ext BM on morphine antinociception was also assessed. n Bt-ext BM was also screened for development of tolerance to antinociceptive effect of Bacopa monnieri by administering 15 mg/kg n Bt-ext BM for seven days. Tolerance to morphine analgesia was induced in mice by administering intraperitoneally (I.P.) 20 mg/kg morphine twice daily for five days. Acute and Chronic administration of 5, 10 and 15 mg/kg n Bt-ext BM significantly reduced both expression and development of tolerance to morphine analgesia in mice. Additionally, Bacopa monnieri was found to enhance antinociceptive effect of morphine in intolerant animals. However, no tolerance to Bacopa monnieri antinociceptive effect was observed in seven days treatment schedule. These findings indicate effectiveness of Bacopa monnieri for management of morphine tolerance.  相似文献

Based on the ability of phytosiderophores to chelate other heavy metals besides iron (Fe), phytosiderophores were suggested to prevent graminaceous plants from cadmium (Cd) toxicity. To assess interactions between Cd and phytosiderophore-mediated Fe acquisition, maize (Zea mays) plants were grown hydroponically under limiting Fe supply. Exposure to Cd decreased uptake rates of 59Fe(III)-phytosiderophores and enhanced the expression of the Fe-phytosiderophore transporter gene ZmYS1 in roots as well as the release of the phytosiderophore 2'-deoxymugineic acid (DMA) from roots under Fe deficiency. However, DMA hardly mobilized Cd from soil or from a Cd-loaded resin in comparison to the synthetic chelators diaminetriaminepentaacetic acid and HEDTA. While nano-electrospray-high resolution mass spectrometry revealed the formation of an intact Cd(II)-DMA complex in aqueous solutions, competition studies with Fe(III) and zinc(II) showed that the formed Cd(II)-DMA complex was weak. Unlike HEDTA, DMA did not protect yeast (Saccharomyces cerevisiae) cells from Cd toxicity but improved yeast growth in the presence of Cd when yeast cells expressed ZmYS1. When supplied with Fe-DMA as a Fe source, transgenic Arabidopsis (Arabidopsis thaliana) plants expressing a cauliflower mosaic virus 35S-ZmYS1 gene construct showed less growth depression than wild-type plants in response to Cd. These results indicate that inhibition of ZmYS1-mediated Fe-DMA transport by Cd is not related to Cd-DMA complex formation and that Cd-induced phytosiderophore release cannot protect maize plants from Cd toxicity. Instead, phytosiderophore-mediated Fe acquisition can improve Fe uptake in the presence of Cd and thereby provides an advantage under Cd stress relative to Fe acquisition via ferrous Fe.  相似文献

The effect of p-chlorophenylalanie (p-CPA) --300 mg/kg-- on reproduction has been studied in the female rat. Groups of animals were injected with a dose of 300 mg/kg of p-CPA 48 hours before proper copulation conditions at different moments along the ovarian cycle. Presence of spermatozoa in the vaginal frotis was negative in treated rats; in the control groups however, positivity was found in variable proportions according to the phase of the ovarian cycle: 30 ad 90% in diestrus and proestrus respectively. Treated animals showed continuous diestrus phases and diffuse luteinitation of the ovary. The results may indicate that a decrease of cerebral 5-HT, caused by p-CPA, lessens the reproductive behaviour of the female rat through mechanisms depending probably on the liberation of gonadrotrophins.  相似文献

An earlier study had shown a decreased tolerance to hexobarbital after electrically induced convulsions in the abstinence after chronic barbital treatments in the rat. The effect of spontaneous convulsions was investigated in the present study.In two experiments tolerance was induced by treatment with barbital in the drinking water. Two treatment periods with a total duration of about 55 weeks were given in each experiment. The average consumption of barbital was approximately 200 mg/kg/day during the last part of the treatments. In the abstinence after the second treatment period tolerance was measured with a hexobarbital threshold method. Convulsions were recorded with a jiggle cage. Records were obtained from all barbital treated animals during the first three days of abstinence.In both experiments the hexobarbital thresholds were reduced in barbital treated animals if a convulsion had occurred within the 25 hour period prior to a threshold determination performed on the third abstinent day. If no convulsion had occurred there was a more marked tolerance to hexobarbital.The convulsion which is regarded as a sign of physical dependence can thus reduce the tolerance to hexobarbital in the abstinence after chronic barbital treatments. The convulsion could not only be the sign but also the means to at least temporarily decrease physical dependence.  相似文献

The effect of p-chlorophenylalanine (PCPA: 300 mg/kg) on the rate of ovulation and plasma LH, FSH and prolactin secretion has been studied in rats at preovulatory periods (18th hour of diestrus) and post-ovulatory periods (9th hour of metaestrus). In both experimental groups, results showed that administration of PCPA caused an increase in both prolactin concentration and number of mature ovarian follicles (p less than 0.001). No changes were observed in FSH levels. LH concentration, however, decreased (p less than 0.001) and ovulation became totally inhibited. Rats treated at the 9th hour of metaestrus exhibited a marked luteinization as well as an increased number of corpus luteum in the ovaric tissue (p less than 0.001), whereas those treated at the 18th hour of diestrus underwent no luteinization and merely showed a greater number of mature ovarian follicles (p less than 0.001). PCPA, therefore, seems not to have a double effect on ovulation, LH, FSH, and prolactin secretion regardless of the pre or post-ovulatory periods. Changes observed in the ovaric tissue might be due to an increase in plasma prolactin concentration which appears earlier in the preovulatory than in the post-ovulatory treated animals. This difference may explain the double effect that has been attributed to the ovaric cycle and reproductive behavior.  相似文献

Rats developed cross-tolerance to the motor-impairing effects of ethanol after daily oral administration of pentobarbital. Chronic administration of p-chlorophenylalanine (p-CPA), in a dosage regimen previously demonstrated to maintain extensive brain serotonin (5-HT) depletion, slowed down cross-tolerance development. p-CPA did not appear to exert this effect by altering the disposition of ethanol, since blood ethanol levels measured 20 min after ethanol administration were not affected by p-CPA treatment. This study extends our previous findings with respect to the inhibitory effects of p-CPA on tolerance development to ethanol and pentobarbital, and suggests that 5-HT may play a role in cross-tolerance development between ethanol and pentobarbital.  相似文献

High concentrations of sucrose are often used to cryopreserve regenerable plant cell cultures in liquid nitrogen. A 21-h pretreatment of carrot somatic embryos in medium containing 0.4 M sucrose allows 80 % of them to germinate after freezing. Substitution of sucrose by polyethylene glycol 6000 led to lower germination rates. However, a high level of freezing tolerance was restored by addition of 1 μM abscisic acid in the pretreatment medium. Using these different media, both total water soluble protein, using SDS-PAGE, and boiling-stable protein, using 2-D electrophoresis, were studied in relation to acquisition of cryopreservation tolerance. Only boiling-stable protein patterns showed some changes: five polypeptides accumulated in 0.4 M sucrose-pretreated embryos or in embryos pretreated by media containing abscisic acid. This accumulation was not detected with polyethylene glycol 6000 used as sole cryoprotectant. Although over-accumulation of polypeptides was highest with media containing ABA, the best germination rates were linked to pretreatment with 0.4 M sucrose. The addition of okadaic acid in 0.4 M sucrose medium led to embryo death after freezing, confirming the existence of a message leading to metabolic changes and acquisition of cryotolerance. Water-soluble proteins obtained from 0.4 M sucrose-pretreated embryos appeared more active than those extracted from control embryos in protecting in vitro a freeze-labile enzyme. Boiling-stable proteins, corresponding to a part of total proteins, were more active than total proteins. These results suggest that these polypeptides may be involved in a mechanism of protection needed for cell survival during freezing stress.  相似文献

The horizontally-transmitted acanthocephalan parasite Polymorphus minutus and the vertically-transmitted microsporidian parasite Dictyocoela roeselum have both been shown to influence on the antitoxic responses of mono-infected Gammarus roeseli exposed to cadmium. The present study investigates the effect of this co-infection on the antitoxic defence responses of naturally infected females exposed to cadmium stress. Our results revealed that, depending on the cadmium dose, bi-infection induced only slight, significant increased cell damage in G. roeseli as compared to non-infection. In addition, the antitoxic defence pattern of cadmium-exposed bi-infected hosts was similar to the pattern of cadmium-exposed D. roeselum-infected hosts. Reduced glutathione concentrations, carotenoid levels and γ-glutamylcystein ligase activity decreased, while metallothionein concentrations increased. This similar pattern indicates that host physiology can be controlled to some extent by microsporidia under stress conditions. It supports the hypothesis of a disruption of acanthocephalan effects in the presence of microsporidia. However, the global negative effects of bi-infection on host condition should be tested on more biological models, since competition between parasites depends on life history trade-off.  相似文献

Reproduction in female rats kept on artificial cycles of light/darkness, continuous lighting and continuous darkness, has been studied. Three weekly doses of p-chlorophenylalanine (p-CPA), 100 mg/kg, were given to half of the rats in each group during the two preconditioning months before copulation. An evaluation has been made of the insemination, fecundation, number of embryos, ovarian cycle and level of 5-HT in the hypothalamus. The administration of p-CPA had an inhibitory effect on the reproductive behavior, especially pronounced in rats kept under continuous lighting and under continuous darkness. The difference in 5-TH levels were greater among the groups kept on cycles of light/darkness without treatment and with p-CPA (0.05 greater than P greater than 0.01). The importance of 5-HT in the mechanisms of reproductive behavior is emphasized.  相似文献

Changes in ultrastructural, biochemical and biophysical characteristics of embryonic axes of Aesculus hippocastanum during development are related to changing levels of desiccation tolerance. Histodifferentiation was complete 30 days after flowering (DAF) and fruits were shed about 120 DAF. During this period, the dry mass of embryonic axes increased from about 0.5 to 4 mg and the water content decreased from 10.2 to 2.0 g H2O g?1 dry mass (g g?1). In spite of the large changes in water content, water potentials of freshly harvested material declined slightly during development from ?0.65 to ?2.0 MPa. Tolerance of desiccation increased as embryos matured. If evaluated on the basis of critical water contents for survival, tolerance appeared to increase continuously, maximum tolerance being achieved at 120 DAF when embryos survived water contents as low as 0.30 g g?1. When evaluated from critical water potentials, three distinct levels of desiccation tolerance could be recognized at ?1.8 MPa (30-40 DAF), ?4 M Pa (48-90 DAF) and ?12 MPs (100-120 DAF). During development, total dry matter increased while sugar content (g g?1' dry mass) and osmotically active material (mmol g?1 dry mass) decreased. The subcellular organisation of axes was always typical of metabolically active tissues. Maximum tolerance (?12 MPa) was associated with a reduced amount of monosaccharides and the appearance of water with unusual calorimetric behaviour. Our data are consistent with several of the current hypotheses regarding the mechanisms of desiccation tolerance. Accumulation of dry matter reserves, reduced levels of monosaccharides, presence of dehydrin-like proteins and ability to form glasses appear to be associated with the changes in desiccation tolerance. However, none of these factors allow A. hippocastanum embryos to achieve the extreme level of desiccation tolerance typical of orthodox seeds. This may be because A. hippocastanum embryos do not reach physiological maturity and remain metabolically active even after they are shed from the parent plant. Also, embryos may acquire incompetent protectants or lack as yet unidentified protective mechanisms.  相似文献
