Role of mitochondria in ethanol tolerance of Saccharomyces cerevisiae   总被引:7,自引:0,他引:7  
The presence of active mitochondria and oxidative metabolism is shown to be essential to maintain low inhibition levels by ethanol of the growth rate (), fermentation rate (v) or respiration rate () of Saccharomyces cerevisiae wild type strain S288C. Cells which have respiratory metabolism show K i (ethanol inhibition constant) values for , v and , higher (K i>1 M) than those of petite mutants or grande strains grown in anaerobiosis (K i=0.7 M). In addition, the relationship between or v and ethanol concentration is linear in cells with respiratory metabolism and exponential in cells lacking respiration. When functional mitochondria are transferred to petite mutants, the resulting strain shows K i values similar to those of the grande strain and the inhibition of and v by increasing ethanol concentrations becomes linear.  相似文献

Psychological parameters of mental activity (30 in total) and their genetic determination were studied in 67 families of schizophrenia patients (67 patients, 107 parent, and 30 sibs). Abnormalities of most of the examined characteristics were found in both the patients and their healthy relatives. Parameters of attention shifting and emotionality revealed the largest genetic component (25–75 and 17–98%, respectively) in all analyzed groups of relatives (probands–affected sibs, probands–healthy sibs, healthy parents–healthy children, affected parents–affected children). Significant impact of genetic factors was also found in parameters stability of attention under conditions of continuous concentration, mediated retention span productivity of voluntary retention by reproduction data, personal anxiety level, reflection of unusual social groups, and self-assessment. The relationships among the characteristics examined in the system of mental activity were established by means of cluster analysis. The results of this study can be used in medical genetic counseling for identifying subjects at high risk for schizophrenia.  相似文献

Synopsis We examined the prey capture process in walleye larvae (9.8–18.0 mm mean length) feeding on zooplankton (density 1501–1) in laboratory aquaria at 15,18.5 and 22°C. Larvae were starved for 8 h prior to the experiment and only allowed to feed for 10 min during the experiment in order to minimize the influence of gut processing or satiation on feeding behaviour. Prey consumption (g min–1) increased exponentially and prey capture success (%) increased logarithmically with mean walleye length. Prey consumption and attack rate (strikes min–1) increased significantly with increasing temperature. The effect of temperature on capture success was slight (2% increase from 15 to 22°C) and not significant. Walleye showed positive electivity for medium-sized prey (0.3–0.6 mm body width, mostlyCeriodaphnia quadrangula), negative electivity for small prey (0.3 mm, mostly cyclopoid copepods) and neutral electivity for large prey (0.6 mm, mostlyDaphnia sp.). Neither prey species electivity nor prey size electivity were significantly affected by temperature. However, prey size electivity did show a fairly large effect size with respect to temperature and we suggest that this relationship should be examined further. These results indicate that temperature affects prey capture in walleye larvae primarily by influencing attack rate.  相似文献

In southern African savannas, bush encroachment is a major problem for range managers. However, little is understood of the actual regeneration processes leading to it, and in particular the role of soil seed banks. The horizontal (between microsites) and vertical (with depth in litter and soil) distribution of soil seed banks of the microphyllous woody species, Acacia tortilis, A. nilotica and Dichrostachys cinerea (all legumes of the Mimosoideae), were quantified in an area with low intensity grazing (reserve), and a bordering cattle farm with high intensity grazing (farm). Species differed in seed bank densities between microsites and sites. Seed densities for all species were highest below parent tree canopies and decreased with distance beyond the canopy, and with soil depth. D. cinerea had the smallest seed bank associated with parent trees, particularly on the farm (8 vs. 1643 seeds/tree on the reserve), A. tortilis had the largest (6357, 31910), with A. nilotica intermediate (1789, 1906). The proportion of current (recently fallen) versus old (1 year old) seeds differed between species and sites. These species form at least short-term persistent seed banks with the old seeds largely representing the persistent seed bank. Seed densities in the open (inter-canopy) and those dispersed under either of the other two (non-parental) study species were much lower than those associated with parent trees. The latter were mostly found under the acacias (single-stemmed) rather than D. cinerea (multistemmed). Total seed store per parent plant increased with plant size (best fits were mostly power curves of canopy area). A large proportion of intact seeds were viable, namely 81–84% for A. tortilis, 68–77% for A. nilotica and 63–78% for D. cinerea, with no differences between sites. Viability tended to increase with depth of burial, except for A. nilotica seeds at the 3–5 cm depths on the farm. At the landscape scale there were 1.5 million and 140000 A. tortilis seeds/ha on the reserve and farm respectively, with corresponding values of 2000 and 31000 for D. cinerea, and 23000 and 86000 for A. nilotica.  相似文献

Inaccurate protein synthesis produces unstable -galactosidase, whose activity is rapidly lost at high temperature. Erythromycin, lincomycin, clindamycin, and celesticetin were shown to counteract the error-inducing effects of streptomycin on -galactosidase synthesized in the antibiotic-hypersensitive Escherichia coli strain DB-11 Met . Newly synthesized -galactosidase was more easily inactivated by high temperatures when synthesized by bacteria partially starved for arginine, threonine, or methionine. Simultaneous treatment with erythromycin or linocomycin yielded -galactosidase that was inactivated by high temperatures less easily than during starvation alone, an effect attributed to stimulation of ribosome editing. When synthesized in the presence of canavanine, -galactosidase was inactivated by high temperature more easily but this effect could not be reversed by erythromycin. The first arginine in -galactosidase occurs at residue 13, so the effect of erythromycin during arginine starvation is probably to stimulate dissociation of erroneous peptidyl-tRNAs of at least that length. Correction of errors induced by methionine starvation is probably due to stimulation of dissociation of erroneous peptidyl-tRNAs bearing peptides at least 92 residues in length. All the effects of erythromycin or the tested lincosamides on protein synthesis are probably the result of stimulating the dissociation from ribosomes of peptidyl-tRNAs that are erroneous or short.  相似文献

Summary There has been an ongoing controversy over how to decide whether the distribution of species is random — i.e., whether it is not greatly different from what it would be if species did not interact. We recently showed (Roberts and Stone (1990)) that in the case of the Vanuatu (formerly New Hebrides) avifauna, the number of islands shared by species pairs was incompatible with a random null hypothesis. However, it was difficult to determine the causes or direction of the community's exceptionality. In this paper, the latter problem is examined further. We use Diamond's (1975) notion of checkerboard distributions (originally developed as an indicator of competition) and construct a C-score statistic which quantifies checkerboardedness. This statistic is based on the way two species might colonise a pair of islands; whenever each species colonises a different island this adds 1 to the C-score. Following Connor and Simberloff (1979) we generate a control group of random colonisation patterns (matrices), and use the C-score to determine their checkerboard characteristics. As an alternative mode of enquiry, we make slight alterations to the observed data, repeating this process many times so as to obtain another control group. In both cases, when we compare the observed data for the Vanuatu avifauna and the Antillean bat communities with that given by their respective control group, we find that these communities have significantly large checkerboard distributions, making implausible the hypothesis that their species distributions are a product of random colonisation.  相似文献

High tree alpha-diversity in Amazonian Ecuador   总被引:14,自引:4,他引:10  
In a 1 ha square plot of terra firme forest at 260 m elevation in Amazonian Ecuador, all trees with diameter at breast height (dbh) 5 cm were studied. There were 1561 individuals, 473 species, 187 genera and 54 families. Of these, 693 individuals, 307 species, 138 genera and 46 families had a dbh 10 cm. This is the highest number of tree species ever recorded for a tropical rain forest sample of this size. In both dbh classes, the most species-rich families were: Fabaceae sensu lato (including Mimosaceae and Caesalpiniaceae), Lauraceae and Sapotaceae; the most species-rich genera, were Pouteria, Inga and Protium. The vertical space was partitioned among species: 166 species were found only in the 5–10 dbh cm class and were mostly sub-canopy treelets, and 307 species with dbh 10 cm were mostly large canopy trees.  相似文献

Little is known about the influence of forest management on the interaction between seed bank and aboveground vegetation. We surveyed seed banks and vegetation in 10 forest stands under similar abiotic conditions but submitted either to a coppice-with-standards treatment (n=5) or to a selective-cutting system (n=5). We analyzed species composition and diversity, community ecological profile, and distribution of taxa among different life forms, strategy, morphology and functional type categories. A total of 2085 seedlings (8296 seedsm–2) germinated-corresponding to 28 species, among which Juncus effusus was the most abundant. Fifty-seven percent of the species were also recorded in the aboveground vegetation, the dominant species being Rubus fruticosus agg., but only 28% of the aboveground species were present in the seed bank. Our results suggest that (1) vernal geophytes and shade-tolerant perennials, which group most true forest species, are not incorporated in the seed bank, (2) parent plants of most seeds were present either in the stand in an earlier dynamic stage or apart from the stand and long-distance dispersed, (3) as expected, early-successional species are well represented in the seed bank, (4) forestry vehicles seem to be a major means of dispersion for stress-tolerant species normally found in forest lanes and wheel tracks. We conclude that seed banks contain species that have a potentially negative impact on the true forest flora and, thus, forest management should minimize soil disturbance and retain remnants of old-coppice woods to conserve disturbance-sensitive true forest species.  相似文献

The mechanism of uptake of water-insoluble -sitosterol by a newly isolated strain of Arthrobacter simplex SS-7 was studied. The production of an extracellular sterol-pseudosolubilizing protein during growth of A. simplex on -sitosterol was demonstrated by isolating the factor from the cell-free supernatant and its subsequent purification by Sephadex G-150 column chromatography. The M r of the purified sterol-pseudosolubilizing protein determined by SDS–PAGE was 19kDa. The rate of sterol pseudosolubilization (5.2×10–3g l–1h–1) could not adequately account for the rate of sterol uptake (72×10–3g l–1h–1) and the specific growth rate (56×10–3 h–1). However in the unfavourable growth condition, when the cells were treated with sodium azide at the level of 30–60% of MIC, the sterol pseudosolubilization accounted for nearly 74% of the total growth containing 96% free cells. Cellular adherence to substrate particles was found to play an active role in the normal growth of the strain on -sitosterol. Unlike sodium acetate-grown cells, whose surface activity was negligible (60mNm–1), the sterol-grown cells had strong surface activity (40mNm–1). The high lipid content and long chain fatty acids in the cell-wall of -sitosterol-grown cells probably contribute to the high sterol adherence activity of the cells.  相似文献

Summary We investigated several ecological correlates of seed mass variation in the hemiparasitic, xylemtapping mistletoe, Phoradendron juniperinum. Mean seed mass varied two-fold among plants between the ages of 4 and 14 years old and was positively correlated with parental plant age. Both the standard deviation and the coefficient of variation in mean seed mass decreased with increasing plant age demonstrating that, on average, younger plants produced seed with more variable mass. Nitrogen concentrations (mg nitrogen per gram of seed) of both the seed and fruit (pericarp) were not correlated with mass or the age of the parent plant from which the seed was taken. However, the nitrogen content per seed (mg nitrogen per seed) was positively correlated with the mean seed dry mass and the age of the seed parent, suggesting that the carbon to nitrogen ratio of individual seeds remained relatively constant as seed mass increased and plants grew older. Seed germination ranged between 20% and 86% and was positively correlated with mass and parental plant age. Heavier seeds (seeds from older plants) also had the highest root radicle growth rates. Furthermore, the final root radicle length after 76 d of growth was positively correlated with seed dry mass. When grown on a medium containing an extract prepared from the host plant foliage, all seeds showed lower germination, grew more slowly and had shorter overall root radicles, but had significantly greater development of the haustorial disks (the holdfast which forms the host-parasite junction in Phoradendron) than seeds grown on a control medium. Our results suggest that, on average, seeds of greater mass produced by older plants have a greater total resource pool per propagule (fruit + seed). This resource pool may be important in conferring a greater potential for dispersal (fruit), survival, colonization, and establishment.  相似文献

Mature seeds of Glycine max (L.) Merr. contain two major storage proteins, a glycosylated 7S protein (conglycinin) and a non-glycosylated 11S protein (glycinin). Accumulation of these proteins and their mRNAs during seed development in cv. Provar was studied by SDS polyacrylamide gel electrophoresis and by Northern (DNA-RNA) hybridization. The 11S acidic and basic subunits and the 7S and subunits began to accumulate 18–20 d after pollination, shortly after the termination of cell division in developing cotyledons, whereas the 7S and 11S A-4 subunits were not detected until one to two weeks later, during the maturation phase of development. Messenger RNAs for 7S and 11S proteins were first detected 14–18 d after pollination, several days before the accumulation of storage proteins. Extracts from embryonic axes contained reduced levels of the 7S subunit, very little 11S protein, no detectable 7S or 11S A-4 subunits, and an additional 7S subunit not found in cotyledons. The effects of fertilization [control (C), 200kgNha–1+25kgP ha–1 (LNP) and 400kgNha–1+ 50kgP ha–1 (HNP)] on fine root dynamics were examined in a 40-year-old Larix leptolepis plantation in central Korea. The average fine root biomass during the growing season for C, LNP and HNP was 957, 934 and 814kgha–1, respectively, whereas the fine root production for C, LNP and HNP was 2103, 2131 and 2066kgha–1, respectively. Nitrogen and P inputs into the soil via fine root turnover for C, LNP and HNP were 23.0 and 1.2, 23.3 and 1.2 and 22.6 and 1.2kgha–1, respectively. There were no significant differences in fine root biomass, production and N and P inputs through fine root turnover between the fertilization treatments during the first growing season after fertilization.  相似文献

The effectiveness of Jeffrey pine (Pinus jeffreyi) seed dispersal performed by seed-caching yellow pine chipmunks (Tamias amoenus) and lodgepole chipmunks (Tamias speciosus) was compared to that of wind dispersal in the Sierra Nevada of western Nevada. Wind-dispersed seeds typically fall under or near the parent tree. Chipmunks removed 90 and 97% of 1064 radioactive seeds from each of two simulated wind-dispersed seed shadows in less than 24 h. Wind-dispersed seeds were deployed within 12 m of the two source trees, but chipmunk caches were found from 2–69 m from the trees. Chipmunks carried nearly all seeds away from source trees, greatly reducing the density of seeds under and near source trees. Caches contained from 1–35 seeds and most were buried 7–21 mm deep. Chipmunks cached in open bitterbrush shrubland with mineral soils much more than expected and cached in closed-canopy Jeffrey pine and lodgepole pine forests with thick needle litter much less than expected. Many Jeffrey pine seedlings and saplings grow in the bitterbrush habitat and few grow in the pine forests. Ten and 20% of the original caches survived until April, the time of seed germination, at the two sites. The movement of wind-dispersed seeds is random relative to environmental variables important in seedling survival, and the wind in coniferous forests cannot quickly bury seeds. The quality of seed dispersal rendered by chipmunks was superior to that provided by the wind because the chipmunks quickly harvested seeds on the ground, moved them away from source trees, and buried them in the ground in habitats and microhabitats where they were more likely to establish new seedlings. The increased quality of seed dispersal provided by animals relative to the wind may help explain why over twenty species of pines have evolved seeds and cones that are adapted for dispersal by seed-caching animals.  相似文献

Summary The pollination biology as related to divergent post-fire reestablishment strategies was examined during flowering sequences for two years in sympatric populations of Arctostaphylos pringlei and A. glandulosa var. mollis. Arctostaphylos pringlei reestablishes post-fire populations by means of fire-stimulated germination of previously dormant seeds, while A. glandulosa var. mollis resprouts from large subsurface burls. Arctostaphylos pringlei produced more flowers per unit plant size, exhibited faster nectar production and higher nectar concentration, attracted nearly twice the number of pollinator visitations, and set more seed when self-pollinated. This substantiated the hypothesis that in these two species the amount of reproductive energy allocated to flower, nectar, and seed production reflected the relative significance of seeding compared to a resprouting post-fire establishment strategy.  相似文献

Physiognomy, structure and floristic composition of one hectare of lowland tropical rain forest was studied in detail at Los Tuxtlas, Mexico. Physiognomically, the Los Tuxtlas forest should be classified as lowland tropical high evergreen rain forest. The forest showed a closed canopy at 30–35 m. Of all woody, non-climbing species with a DBH1.0 cm 89.4% (94.5% of all individuals) were evergreen, 25.4% (59.5% of the individuals) had compound leaves, and over 80% of species (and individuals) had leaves in the notophyll and mesophyll size classes. The forest structure was characterized by a low density (2976 individuals with a DBH1.0 cm, 346 individuals with a DBH10.0 cm, per ha, excluding vines) with an average basal area (38.1 m2, DBH1.0 cm, 34.9 m2, DBH10.0 cm, per ha, excluding vines). This was attributed to the relative maturity of the forest on the study plot. The study plot contained 234 species (11 208 individuals with a height 0.5 m), of which 55.1% (34.8% of individuals) were trees, 9.4% (6.8%) shrubs, 3.4% (44.3%) palms, 20.1% (5.2%) vines, 6.8% (8.7%) herbs and 5.1% (0.3%) of unknown lifeform. Furthermore, 58 species of epiphytes and hemi-epiphytes were found. Diversity of trees, shrubs and palms with a DBH1.0 cm was calculated as Shannon-Wiener index (4.65), Equitability index (0.65), and Simpson index (0.10). The dominance-diversity curve showed a lognormal form, characteristic for tropical rain forest. The community structure was characterized by a relative dominance of Astrocaryum mexicanum in the understorey, Pseudolmedia oxyphyllaria in the middle storeys, and Nectandra ambigens in the canopy. Species population structures of 31 species showed three characteristic patterns, differentiated by recruitment: continuously high, discontinuously high, and continuously low recruitment. Height/diameter and crown cover/diameter diagrams suggested a very gradual shift from height growth to crown growth during tree development. Forest turnover was calculated as 138 years. Compared to other tropical rain forests the Los Tuxtlas forest had 1. similar leaf physiognomical characteristics, 2. a lower diversity, 3. a lower density, 4. an average basal area, and 5. a slow canopy turnover.  相似文献

Connective tissue of the freshwater pulmonateLymnaea stagnalis was shown to contain fucosyltransferase activity capable of transferring fucose from GDP-Fuc in 1–2 linkage to terminal Gal of type 3 (Gal1–3GalNAc) acceptors, and in 1–3 linkage to GlcNAc of type 2 (Gal1–4GlcNAc) acceptors. The 1–2 fucosyltransferase was active with Gal1–3GalNAc1-OCH2CH=CH2 (K m=12 mM,V max=1.3 mU ml–1) and Gal1–3GalNAc (K m=20 mM,V max=2.1 mU ml–1), whereas the 1–3 fucosyltransferase was active with Gal1–4GlcNAc (K m=23 mM,V max=1.1 mU ml–1). The products formed from Gal1–3GalNAc1-OCH2CH=CH2 and Gal1–4GlcNAc were purified by high performance liquid chromatography, and identified by 500 MHz1H-NMR spectroscopy and methylation analysis to be Fuc1–2Gal1–3GalNAc1-OCH2CH=CH2 and Gal1–4(Fuc1–3)GlcNAc, respectively. The flow cytofluorometric susceptibility test (FCST) was incorporated into two in vitro synergy assays of amphotericin B-drug combinations. The FCST checkerboard assay and the FCST concentration-effect assay were developed to detect subtle modulation of amphotericin B fungicidal effects onCandida albicans andCryptococcus neoformans. Amphotericin B × cyclosporine A was a synergistic fungicidal combination against bothC. albicans andC. neoformans. Amphotericin B×gentamicin and amphotericin B×ketoconazole were synergistic combinations againstC. neoformans. In all cases tested, the synergy was effective when 0.50–0.62 g amphotericin B/ml was used with>-0.50 g of the other drug/ml. The fungicidal effect of 1.00 g amphotericin B/ml overwhelmed the synergistic effects. A number of other drug combinations were additive, autonomous, or antagonistic.  相似文献

Summary Rat liver nuclei were extracted with 0.14 M NaCl and the extracts submitted to sucrose gradient fractionation. Aliquots of the nuclear residue remaining after the 0.14 M NaCl extraction were also extracted either with 0.3 M NaCl or 1 M urea, and the extracts similarly submitted to sucrose gradient fractionation. Thereafter, both the presence and relative distribution of individual U-snRNA (U1–U6) species was followed. Results showed an extensive association of all U-snRNAs to RNP structures of 40 S. However, characteristic differences in the association of mostly U1 and U5 — which were the major identifiable species in the extracts — to these structures were observed. Only a small fraction of U1 appeared complexed to 40 S RNP structures, while most of it sedimenting in the > 20 S region of the gradient. In contrast, U5-snRNA had a tight and almost exclusive association to 40 S RNP structures. No pool of 10–12 S U5-snRNP complexes was detected.Combined immunoprecipitation and immunoblotting experiments on nuclear 0.14 M NaCl extracts using anti-Sm and/or anti-RNP antisera showed that all snRNA species, whether recovered as 10–12 S complexes or segregated with 40 S RNP components, existed as snRNP structures bearing at least their Sm-antigenic polypeptides.These and our previous results [Guialis, A., Arvanitopoulou, A, Patrinou-Georgoula, M. and Sekeris, C.E. (1983) FEBS Lett. 151, 127–133], support the existence of snRNP-enriched RNP structures of 40 S. In such structures the core polypeptides (Mr = 32000–45000) of 40 S monoparticles are not obligatory components.  相似文献

Summary In a pair of experiments conducted in old-field habitats in southwestern Michigan (USA), we examined rates of seed loss to post-dispersal predators (ants and rodents). Seeds from 4–6 species of biennial plants were tested over a range of seed densities and habitat types. We found that seed removal was significantly higher in vegetated habitats than in areas of disturbed soil (both simulated small-animal diggings and a plowed field). In the undisturbed vegetation, seed losses ranged from 1–20% of seeds removed/day.An exclosure experiment demonstrated that ants and rodents foraged selectively for seeds of the six plant species tested. Rodents (Peromyscus maniculatus) fed preferentially on species producing large seeds (predominantly Tragopogon dubius). Ants (Myrmica lobicornus) foraged on smaller seeds, although their foraging preferences were not based strictly on seed size.Seed density had only a minor effect on predation rate over the range of densities tested. Predators, instead appeared to treat each experimental group of seeds as a single prey patch. Consequently, predation intensity was quite variable over distances of <20 m within a relatively homogeneous section of habitat.These field experiments provide initial estimates of seed losses to post-dispersal predators in old-field habitats. Rates of seed loss were generally less than those reported from desert or semi-arid habitats. However, for some old-field species, seed losses averaged an appreciable 10–20% day. The selective nature of the seed predators, plusthe relative patchiness of predation intensity in space, suggest that postdispersal seed predation can play a role in determining the distribution and/or abundance of old-field herbs.  相似文献

Tumor promoters, proinflammatory cytokines, endotoxins, and protein synthesis inhibitors can modulate cell cycle kinetics of various cell types, stimulate production of reactive oxygen species, and induce keratinocytes to produce interleukin-8 (IL-8), a potent chemotactant for polymorphonuclear neutrophils and T lymphocytes. The aim of this study was to determine whether perturbations of cytogenetic responses correlated with the induction of IL-8 expression. Cultures of primary human keratinocytes were grown in serum-free medium with 5 mol/L bromodeoxyuridine to label DNA and exposed either to phorbol-13-myristate-12-acetate (PMA) (0.0001–100 ng/ml), cycloheximice (CHX) (0.01–50 g), lipopolysaccharide (0.1–100 g/ml), tumor necrosis factor- (TNF) (3.13–50 ng/ml), or interleukin-1 (IL-1) (1–182 pg/ml). Metaphase chromosome preparations were stained by a fluorescence-plus-Giemsa technique to differentiate sister chromatids. For IL-8 production, keratinocytes were grown to 70% confluency and then exposed to chemicals for 24 h. Immunoreactive IL-8 was quantitated from the supernatants by ELISA. With the exception of benzo(a)pyrene used as a positive control, none of the agents induced sister chromatid exchanges. However, PMA and TNF induced IL-8 production that coincided with significant cell cycle inhibition. IL-1 had no effect on cytogenetic endpoints, yet stimulated a 6.3-fold increase in IL-8. CHX inhibited cell cycle progression and mitotic activity at concentrations that were 200 times lower than required for IL-8 induction; however, puromycin (0.31–10 g/ml), another protein synthesis inhibitor, did not induce IL-8. At all concentrations tested, TNF reduced the mitotic index by 45%, slowed cell cycle progression by 3.5 h, and induced a flat, albeit large, IL-8 response at concentrations 12.5 ng/ml.
