A standardized protocol for the evaluation of hand disinfection by surgical scrub formulations was applied to volunteers in a multicenter trial. Povidone iodine (PVI), chlorhexidine (CHX), and a nonmedicated soap (NMS) were tested. The scrubbing procedure involved three daily hand washings for five consecutive days; surviving bacteria were counted daily after being collected in a suitable neutralizing solution. Immediate efficacy (IE), cumulative efficacy (CE), and remanent effect (RE) were calculated by reference to the control hand. Statistical analyses of IE, CE, and RE showed significant differences among the three scrub formulations. IEs of PVI and CHX were equivalent and different from IE of NMS; CE and RE of CHX were higher than those of PVI and NMS. On the basis of the statistical analysis, the population size required for further studies aimed at detecting significant differences between surgical scrub formulations could be estimated.  相似文献

The circular economy (CE) is a new model for the production and consumption of goods, which has attracted wide political attention as a strategy toward sustainability. However, the theoretical foundation of CE remains poorly structured and insufficiently explored. Recent studies have shown that the CE model draws on different schools of thought and that its origins are mainly rooted in fields such as industrial ecology (IE) and ecological economics (EE). In this article, we investigate the links between CE, IE, and EE and provide an overview of the similarities and differences between these fields. At the same time, we analyze to what extent the linkages between IE and EE can create a coherent body of knowledge for CE, and be used to identify further research opportunities. This paper shows that, until now, research on CE seems to be mainly rooted in the field of IE and based on concepts and tools that already exist in other fields, rather than inventing new ones. The reconciliation of IE and EE could provide a mechanism to extend beyond such a narrow focus, and increase knowledge of the theoretical and practical framework of CE to benefit sustainability.  相似文献

We determined myofibrillar and mitochondrial protein fractional synthesis rates (FSR), intramuscular signaling protein phosphorylation, and mRNA expression responses after isolated bouts of resistance exercise (RE), aerobic exercise (AE), or in combination [termed concurrent exercise (CE)] in sedentary middle-aged men. Eight subjects (age = 53.3 ± 1.8 yr; body mass index = 29.4 ± 1.4 kg·m(2)) randomly completed 8 × 8 leg extension repetitions at 70% of one repetition-maximum, 40 min of cycling at 55% peak aerobic power output (AE), or (consecutively) 50% of the RE and AE trials (CE). Biopsies were obtained (during a primed, constant infusion of l-[ring-(13)C(6)]phenylalanine) while fasted, and at 1 and 4 h following postexercise ingestion of 20 g of protein. All trials increased mitochondrial FSR above fasted rates (RE = 1.3-fold; AE = 1.5; CE = 1.4; P < 0.05), although only CE (2.2) and RE (1.8) increased myofibrillar FSR (P < 0.05). At 1 h postexercise, phosphorylation of Akt on Ser(473) (CE = 7.7; RE = 4.6) and Thr(308) (CE = 4.4; RE = 2.9), and PRAS40 on Thr(246) (CE = 3.8; AE = 2.5) increased (P < 0.05), with CE greater than AE for Akt Ser(473)-Thr(308) and greater than RE for PRAS40 (P < 0.05). Despite increased phosphorylation of Akt-PRAS40, phosphorylation of mammalian target of rapamycin (Ser(2448)) remained unchanged (P > 0.05), while rpS6 (Ser(235/236)) increased only in RE (10.4) (P < 0.05). CE and AE both resulted in increased peroxisome proliferator receptor-γ coactivator 1-α (PGC1α) expression at 1 h (CE = 2.9; AE = 2.8; P < 0.05) and 4 h (CE = 2.6; AE = 2.4) and PGC1β expression at 4 h (CE = 2.1; AE = 2.6; P < 0.05). These data suggest that CE-induced acute stimulation of myofibrillar and mitochondrial FSR, protein signaling, and mRNA expression are equivalent to either isolate mode (RE or AE). These results occurred without an interference effect on muscle protein subfractional synthesis rates, protein signaling, or mRNA expression.  相似文献

Cystic echinococcosis (CE) caused by the parasite Echinococcus granulosus is an important public health problem worldwide. Flubendazole has shown poor in vivo efficacy against CE in humans and mice. However, flubendazole causes marked in vitro damage on E. granulosus protoscoleces. The goals of the current work were: a) to compare the plasma pharmacokinetic behaviour of flubendazole formulated as a hydroxipropyl-β-cyclodextrin aqueous solution or as a carboxymethyl celullose suspension, both given by the oral route to mice, b) to compare flubendazole clinical efficacy in secondary CE in mice after its administration as both formulations, c) to evaluate the flubendazole-induced morphological changes in hydatid cysts recovered from infected mice treated with both drug formulations. Flubendazole administration as a solution resulted in significantly higher plasma maximum concentration (Cmax) and area under the concentration–time curve (AUC) values compared to those obtained after the flubendazole-suspension treatment. This enhanced drug availability correlated with an increased efficacy against secondary CE in mice observed for the flubendazole-solution formulation, while the suspension formulation did not reach differences with the untreated control group. Similar ultrastructural changes were observed in cysts recovered from flubendazole (both formulations) treated mice after 3, 6 and 9 months of infection, although the damage extension was greater after treatment with the flubendazole-solution formulation.  相似文献

A comparison of the immediate effects of resistance, aerobic, and concurrent exercise on postexercise hypotension. The influence of resistance exercise (RE), aerobic exercise (AE), and concurrent exercise (CE) on postexercise hypotension (PEH) is not known. We investigated the immediate blood pressure (BP) lowering effects of exercise after RE, AE, and CE sessions among healthy subjects. Twenty-one men (20.7 ± 0.7 years) performed 4 experimental sessions each in a within-subject design: control (CTL-seated rest for 60 minutes), RE (3 sets at 80% 1RM for 8 exercises, including upper and lower limbs), AE (7-minutes warm-up followed by 50 minutes of cycle ergometer exercise at 65% VO?peak and 3-minute cooldown), and CE (2 sets at 80% 1RM for 6 exercises among those which composed the RE session, plus 20 minutes of cycle ergometer exercise at 65% VO?peak, 7-minute warm-up and 3-minute cooldown, exactly in this order). The total duration of each exercise session was approximately 60 minutes. Systolic blood pressure (SBP) and diastolic blood pressure (DBP) were assessed by ambulatory monitoring at rest (20 minutes) and every 10 minutes after the exercise during 120 minutes while in the laboratory. The duration of the decrease in SBP was longer after AE and CE (120 minutes) compared to RE (80 minutes); and for DBP after AE (50 minutes) compared to CE (40 minutes) and RE (20 minutes) (p < 0.05). The magnitude of the decrease in SBP and DBP was similar after all exercise sessions and significantly different from CTL (p < 0.05) (SBP: RE = 4.1 ± 2.0 mm Hg, AE = 6.3 ± 1.3 mm Hg, CE = 5.1 ± 2.2 mm Hg; DBP: RE = 1.8 ± 1.1 mm Hg, AE = 1.8 ± 1.0 mm Hg, CE = 1.6 ± 0.6 mm Hg). It was concluded that exercise sessions combining aerobic and resistance activities are as effective as AE sessions and more effective than RE sessions to promote PEH.  相似文献

The addition of aerobic exercise (AE) to a resistance exercise (RE) program (concurrent exercise, CE) can interfere with maximum muscle fiber growth achieved with RE. Further, CE appears to markedly affect the growth of myosin heavy chain (MHC) I, but not MHC IIa fibers. The mechanism responsible for this "interference" is unclear. Satellite cell (SC) responsiveness to exercise appears to influence muscle adaptation but has not yet been examined following acute concurrent exercise. Thus, we assessed the fiber-type-specific SC response to RE, AE, and CE exercise. Eight college-aged males completed the following two exercise trials: the RE trial, which consisted of unilateral leg extensions and presses (4 sets ≥ 10 repetitions: 75% 1 repetition maximum, RM); and the AE/CE trial, which included an identical RE protocol with the opposite leg, immediately followed by subjects cycling for 90 min (60% W(max)). Muscle biopsies were obtained from the vastus lateralis before and 4 days after each session. Samples were cross-sectioned, stained with antibodies against NCAM, Ki-67, and MHC I, counterstained with DAPI, and analyzed for SC density (SC per fiber), SC activation, and fiber type. SC density increased to a greater extent following RE (38 ± 10%), compared with CE (-6 ± 8%). Similarly, MHC I muscle fiber SC density displayed a greater increase following RE (46 ± 14%), compared with AE (-7 ± 17%) and CE (-8 ± 8%). Our data indicate that the SC response to RE is blunted when immediately followed by AE, at least in MHC I muscle fibers, and possibly MHC II fibers. This suggests that the physiological environment evoked by AE might attenuate the eventual addition of myonuclei important for maximum muscle fiber growth and consequent force-producing capacity.  相似文献

In a field trial of three disinfectant-detergent preparations for the surgical scrub Hibiscrub (a 4% chlorhexidine detergent solution) maintained a significantly greater residual antibacterial action on the skin of the gloved hands of an operating team than was obtained with Disadine (povidone-iodine detergent preparation) (P<0·001). In comparison with Phisohex (3% hexachlorophane in detergent cream), the established surgical scrub of the operating team for many years, Hibiscrub maintained marginally more significant residual disinfection (P 0·05). Of the three preparations the members of the team found Hibiscrub to be more acceptable for use than either Disadine or Phisohex.  相似文献

Renewable energy (RE) technologies are looked upon favorably to provide for future energy demands and reduce greenhouse gas (GHG) emissions. However, the installation of these technologies requires large quantities of finite material resources. We apply life cycle assessment to 100 years of electricity generation from three stand‐alone RE technologies—solar photovoltaics, run‐of‐river hydro, and wind—to evaluate environmental burden profiles against baseline electricity generation from fossil fuels. We then devised scenarios to incorporate circular economy (CE) improvements targeting hotspots in systems’ life cycle, specifically (1) improved recycling rates for raw materials and (ii) the application of eco‐design. Hydro presented the lowest environmental burdens per kilowatt‐hour of electricity generation compared with other RE technologies, owing to its higher efficiency and longer life spans for main components. Distinct results were observed in the environmental performance of each system based on the consideration of improved recycling rates and eco‐design. CE measures produced similar modest savings in already low GHG emissions burdens for each technology, while eco‐design specifically had the potential to provide significant savings in abiotic resource depletion. Further research to explore the full potential of CE measures for RE technologies will curtail the resource intensity of RE technologies required to mitigate climate change.  相似文献

The aim of the present study was to evaluate the effectiveness of chlorhexidine digluconate (CHX) and commonly used mouthrinses to single- and poly-species biofilms by S. mutans, S. aureus and P. aeruginosa, on titanium discs of grade IV. The formation of single- and poly-species biofilms at 16.5, 40.5 and 64.5-h incubation on titanium surface was evaluated by plate count (CFU ml−1) before and after exposure to CHX and four mouthrinses (Curasept, Listerine, Meridol and Buccagel) and expressed as percentage of Inhibitory Activity (IA%). The application of the different anti-plaque formulations on biofilm can reduce the adhesion of bacteria to titanium surface with different degrees. The higher efficacy was observed for Listerine that shows IA% = 100 on the biofilm formed by S. mutans at 16.5 h. Log count of CFU was dependent to culture time and four mouthrinses for S. mutans and S. aureus, whilst was not dependent to culture time but to mouthrinses for P. aeruginosa. In general, the efficacy was particularly lesser to poly-species biofilms; no statistical differences were evidenced between all the mouthrinses and CHX as control group. The tested mouthrinses, compared to reference CHX 0.2%, have demonstrated a significant lower antibacterial activity than Listerine towards the experimental biofilms. This “in vitro” biofilm model should prove extremely useful for pre-clinical testing of anti-plaque agents, which inhibit biofilm formation, can prevent subsequent implant failure.  相似文献

Summary The red mangrove (Rhizophora mangle L.) occurs frequently in both scrub and fringe mangrove forests. Our previous study demonstrated that individuals of this mangrove species growing in scrub and fringe forests differ significantly in both morphological and physiological characteristics. To further characterize physiological differences between scrub and fringe mangroves, we compared their differences in water uptake and photosynthetic gas exchange during different seasons. In the wet season (June–October, 1990), scrub mangroves showed lower D and 18O values of stem water than fringe mangroves, indicating more usage of rain-derived freshwater. In the dry season (Jan–April, 1991), however, scrub mangroves utilized the same water source as fringe mangroves, reflected by their similar D and 18O values of stem water. Consistently, there were significant differences in predawn water potentials between scrub and fringe mangroves in the wet season (October 1990) with higher values for scrub mangroves, but no significant differences in the dry season (January 1991). Higher elevation in the scrub forest seems to be the major factor responsible for the shift of water sources in scrub mangroves. On Apr. 27 and Aug. 8, 1990, scrub mangroves showed lower CO2 assimilation rate, stomatal conductance, and intercellular CO2 concentration than fringe mangroves. There were no differences in these gas exchange characteristics on the other two measuring dates: Oct. 17, 1990 and Jan. 11, 1991. Instantaneous water use efficiency was significantly higher for scrub mangroves than for fringe mangroves on three of the four sampling dates. Similarly, leaf carbon isotope discrimination of scrub mangroves was always significantly lower than that of fringe mangroves, indicating higher long-term water use efficiency. Higher water use efficiency in scrub mangroves is a result of stomatal limitation on photosynthesis, which may entail considerable carbon cost to the plants.  相似文献

Oral bacterial microcosms, established using saliva inocula from three individuals, were maintained under a feast-famine regime within constant-depth film fermenters. Steady-state communities were exposed four times daily, postfeeding, to a chlorhexidine (CHX) gluconate-containing mouthwash (CHXM) diluted to 0.06% (wt/vol) antimicrobial content. The microcosms were characterized by heterotrophic plate counts and PCR-denaturing gradient gel electrophoresis (DGGE). CHXM caused significant decreases in both total anaerobe and total aerobe/facultative anaerobe counts (P < 0.05), together with lesser decreases in gram-negative anaerobes. The degree of streptococcal and actinomycete inhibition varied considerably among individuals. DGGE showed that CHXM exposure caused considerable decreases in microbial diversity, including marked reductions in Prevotella sp. and Selenomonas infelix. Pure-culture studies of 10 oral bacteria (eight genera) showed that Actinomyces naeslundii, Veillonella dispar, Prevotella nigrescens, and the streptococci were highly susceptible to CHX, while Lactobacillus rhamnosus, Fusobacterium nucleatum, and Neisseria subflava were the least susceptible. Determination of the MICs of triclosan, CHX, erythromycin, penicillin V, vancomycin, and metronidazole for microcosm isolates, before and after 5 days of CHXM exposure, showed that CHXM exposure altered the distribution of isolates toward those that were less susceptible to CHX (P < 0.05). Changes in susceptibility distributions for the other test agents were not statistically significant. In conclusion, population changes in plaque microcosms following repeated exposure to CHXM represented an inhibition of the most susceptible flora with a clonal expansion of less susceptible species.  相似文献

Objective: This study aimed to examine and compare the effects of continuous or intermittent exercises on adiposity and fatty liver in rats fed with high‐fat diet. Methods and Procedures: Wistar rats were divided according to diet composition—chow diet (C) or high‐fat diet (H)—and kinds of exercise—sedentary (S), continuous (CE), or intermittent (IE) exercises. The CE group swam 90 min/day, and the IE group swam 3 × 30 min/day (at 4‐h intervals between sessions); both groups exercised 5 days/week during 8 weeks. Body weight and food intake were recorded daily. Lipogenesis rate in vivo was determined by the incorporation of 3H2O into saponified lipids in retroperitoneal (RET), epididymal (EPI), and visceral (VIS) white adipose tissues, brown adipose tissue (BAT), liver (L), and gastrocnemius muscle (GAST) using the gravimetric method. Total cholesterol, high‐density lipoprotein (HDL)‐cholesterol, and triacylglycerol (TG) were analyzed. Results: The major finding of this study is that IE was more efficient than CE in reducing the adverse effects of high‐fat diet and sedentarism. There was an improvement in the lipid profile and a reduction in food intake, body weight gain, visceral and central adiposity, and fatty liver, contributing to the control of obesity and other comorbidities, including nonalcoholic fat liver diseases. Discussion: Earlier studies have discussed the effects of diet consumption on adiposity and their relation to chronic diseases and obesity. This study discusses the effects of high‐fat diet consumption and the different kinds of exercise on weight gain, adiposity, fatty liver, and lipid profile in rats. The results may depend on the exercise, time of each session, age, gender, and experimental period.  相似文献

Laboratory bioassays were carried out to evaluate the effect of insect density (10, 30, 60, and 100 adults), wheat quantity (10, 30, 60, and 100 g), and cracked kernel containment (5, 15, 30, and 50%) on the efficacy of diatomaceous earth (DE). Three beetle species, Sitophilus oryzae (L.), Rhyzopertha dominica (F.), and Tribolium confusum Jacquelin du Val, as well as two DE formulations, Insecto and SilicoSec, and one DE enhanced with pyrethrum, PyriSec (all commercially available) were tested. In the first two series of bioassays, the three DE formulations were applied at three dose rates, 500, 1000 and 1,500 ppm. In the third series, the dose rates used were 500 and 1,000 ppm. Dead adults were counted 14 d later. For insect density, wheat quantity, and cracked kernel containment, significant differences were noted in mortality levels of the tested species among the three DE formulations and among doses. No significant differences were noted in the mortality levels among the four adult densities of any of the insects tested. The increase of wheat quantity used in the bioassays increased significantly adult mortality of T. confusum. The increase of cracked wheat containment decreased significantly adult mortality of S. oryzae.  相似文献

Two wild groups of Callithrix penicillata, the Black Pincelled Marmoset, were observed from January to September 1998, in two areas, one an area of dense scrub savanna vegetation (cerrado) and the other, a semidecidual woodland (cerrad?o), both within the boundaries of the Ecological Reserve of IBGE (Brazilian Institute of Geography and Statistics), in an environmentally protected area, the APA (Portuguese abbreviation for "environmental protected area") Gama/Cabe?a-de-Veado, Brasília, DF. The behavioral data collected during the rainy (January 15 to April 15) and dry season (June 1 to September 15) were compared. Because of the proximity to the Reserve facilities, the group from the dense scrub savanna vegetation (CD) was submitted to antropic impacts different from the group in the semidecidual woodland (CE), which was using as territory an area that had been suffering from man-made fires every two years as part of a long-term experimental project on fire impacts. The behavioral data was quantified by instantaneous cross-section ("scan sampling") every ten minutes with records of locomotion, rest, foraging for insects, use of exudate, and feeding. During the whole year, the greatest percentage of time spent by CE and CD was in foraging for insects, with 44% and 39%, respectively. It was evident when comparing the data for the two seasons that, for both groups, foraging for insects was more intense during the dry season, possibly to complement the shortage of food, and locomotion increased during the rainy season. The greater the availability and distribution of fruit in the areas, the greater the locomotion of the groups to obtain these resources. None of the other behavioral patterns, including the use of exudates, presented significant differences between the two seasons. Both groups foraged more frequently during the dry season and locomoted more during the rainy one.  相似文献

Paralytic Shellfish Poisoning (PSP) toxins are annually recurrent along the Massachusetts coastline (USA), which includes many small embayments and salt ponds. Among these is the Nauset Marsh System (NMS), which has a long history of PSP toxicity. Little is known, however, about the bloom dynamics of the causative organism Alexandrium fundyense within that economically and socially important system. The overall goal of this work was to characterize the distribution and dynamics of A. fundyense blooms within the NMS and adjacent coastal waters by documenting the distribution and abundance of resting cysts and vegetative cells. Cysts were found predominantly in three drowned kettle holes or salt ponds at the distal ends of the NMS – Salt Pond, Mill Pond, and Town Cove. The central region of the NMS had a much lower concentration of cysts. Two types of A. fundyense blooms were observed. One originated entirely within the estuary, seeded by cysts in the three seedbeds. These blooms developed independently of each other and of the A. fundyense population observed in adjacent coastal waters outside the NMS. The temporal development of the blooms was different in the three salt ponds, with initiation differing by as much as 30 days. These differences do not appear to reflect the initial cyst abundances in these locations, and may simply result from higher cell retention and higher nutrient concentrations in Mill Pond, the first site to bloom. Germination of cysts accounted for a small percentage of the peak cell densities in the ponds, so population size was influenced more by the factors affecting growth than by cyst abundance. Subsurface cell aggregation (surface avoidance) limited advection of the vegetative A. fundyense cells out of the salt ponds through the shallow inlet channels. Thus, the upper reaches of the NMS are at the greatest risk for PSP since the highest cyst abundances and cell concentrations were found there. After these localized blooms in the salt ponds peaked and declined, a second, late season bloom occurred within the central portions of the NMS. The timing of this second bloom relative to those within the salt ponds and the coastal circulation patterns at that time strongly suggest that those cells originated from a regional A. fundyense bloom in the Gulf of Maine, delivered to the central marsh from coastal waters outside the NMS through Nauset Inlet. These results will guide policy decisions about water quality as well as shellfish monitoring and utilization within the NMS and highlight the potential for “surgical” closures of shellfish during PSP events, leaving some areas open for harvesting while others are closed.  相似文献

The antidiabetic activities of the aqueous (AqEx) and ethanolic (AlEx) extracts of Cleome droserifolia (Forssk.) Del., were tested in cultured C2C12 skeletal muscle cells and 3T3-L1 adipocytes. An 18-h treatment with the AqEx increased basal glucose uptake by 33% [insulin equivalent (IE)=1.3±0.04] in muscle cells comparable to a 25.5% increase caused by 100 nM insulin (IE=1±0.03). Fractionation of the tested AqEx yielded hexane (HxFr), chloroform (ClFr) and ethyl acetate (EtFr) fractions which exerted 38, 52 and 35% increase in the glucose uptake corresponding to an IE of 1.5±0.06, 2.0±0.04 and 1.4±0.04, respectively. Only the ClFr and EtFr accelerated the triglyceride accumulation [rosiglitazone equivalent (RE) was 0.9±0.13 and 0.63±0.12, respectively] in pre-adipocytes undergoing differentiation comparably with 10 μM rosiglitazone. Six terpenoids (C1-C6) and three flavonol glycosides (F1-F3) were isolated from the active ClFr and EtFr, respectively, and identified. C5, C2 and C4 had an IE of 0.86±0.05, 1.01±0.04 and 0.9±0.08, while F1, F2 and F3 gave an IE of 1.3±0.05, 2.3±0.05 and 2.0±0.04, respectively. We could conclude that the reported antihyperglycemic activity of Cleome droserifolia is attributed to significant insulin-like effects in peripheral tissues, and that compounds F2 and F3, being highly active, could be used as bioactive markers to standardize the C. droserifolia herbal extract.  相似文献

The entomopathogenic fungus, Beauveria bassiana, is highly susceptible to the damaging effects of solar radiation. This study demonstrates protection from simulated solar radiation by coating B. bassiana (GHA) spores with lignin and effects on pathogenicity to Lygus lineolaris (Palisot de Beauvois) adults in direct spray and contact uptake bioassays. Spores were coated with either lignin or cross-linked lignin by spray drying to produce spore coatings of high and low water solubility, respectively. Non-coated spores and the two spore coating treatments were suspended in either water (0.04% Silwet L77) or oil (Orchex 692) to produce six formulations. Rates of loss in spore viability under simulated solar radiation were approximately ten times lower for the three formulations in which spores remained coated in suspension (cross-linked lignin in water, cross-linked lignin in oil, and lignin in oil). However, these three formulations were the least pathogenic. Estimates of LC50 and LC75 values for the most pathogenic formulation (non-coated in oil) were 5 and 12 times lower, respectively, than the least pathogenic formulation (cross-linked lignin in oil), but these differences were not significant. The three lignin-coated formulations that provided the greatest UV protection were also the least pathogenic based on LT values, which were significant. Overall, L. lineolaris mortality was approximately 80 times less when exposed to treated broccoli rather than sprayed directly, based on LC50 values. If the contribution of spore uptake from plant surfaces to L. lineolaris mortality is similarly low under field conditions, then improving persistence may be less important for improving efficacy. However, under control conditions where solar radiation significantly impacts mycoinsecticide efficacy, the improved persistence of lignin coating formulations may outweigh negative effects on pathogenicity.  相似文献

Lipid droplets (LDs) in non-adipocytes contain triglycerides (TG) and cholesterol esters (CE) in variable ratios. TG-rich LDs are generated when unsaturated fatty acids are administered, but the conditions that induce CE-rich LD formation are less well characterized. In the present study, we found that protein translation inhibitors such as cycloheximide (CHX) induced generation of CE-rich LDs and that TIP47 (perilipin 3) was recruited to the LDs, although the expression of this protein was reduced drastically. Electron microscopy revealed that LDs formed in CHX-treated cells possess a distinct electron-dense rim that is not found in TG-rich LDs, whose formation is induced by oleic acid. CHX treatment caused upregulation of mTORC1, but the CHX-induced increase in CE-rich LDs occurred even when rapamycin or Torin1 was given along with CHX. Moreover, the increase in CE was seen in both wild-type and autophagy-deficient Atg5-null mouse embryonic fibroblasts, indicating that mTORC1 activation and suppression of autophagy are not necessary to induce the observed phenomenon. The results showed that translation inhibitors cause a significant change in the lipid ester composition of LDs by a mechanism independent of mTORC1 signaling and autophagy.  相似文献

We recently identified neuromedin S (NMS) as an endogenous ligand for the FM-4/TGR-1 receptor. Here, we examined the possible involvement of central NMS in regulation of urinary output and vasopressin (AVP) release in rats. Intracerebroventricular (icv) injection of NMS induced a dose-dependent increase in the plasma level of AVP, followed by a decrease of nocturnal urinary output. Expression of cFos after icv injection of NMS was observed in the supachiasmatic nucleus (SCN), arcuate nucleus, paraventricular nucleus (PVN), and supraoptic nucleus (SON). The cFos expressing cells in PVN and SON, but not SCN, were then double-stained using antibodies against the vasopressin. On the other hand, icv injection of neuromedin U, which also binds to the FM-4/TGR-1 receptor, required a concentration ten times higher than that of NMS in order to exert the same antidiuretic potency. These results suggest that central NMS may exert a physiological antidiuretic action via vasopressin release.  相似文献
