Actin filaments and photoreceptor membrane turnover   总被引:1,自引:0,他引:1  
The shape and turnover of photoreceptor membranes appears to depend on associated actin filaments. In dipterans, the photoreceptor membrane is microvillar. It is turned over by the addition of new membrane at the bases of the microvilli and by subsequent shedding, mostly from the distal ends. Each microvillus contains actin filaments as a component of its cytoskeletal core. Two myosin I-like proteins co-localize with the actin filaments. It is suggested that one of the myosin I-like proteins might be linked to the microvillar membrane. By interacting with the actin filaments, this motor should move the membrane of a microvillus in a distal direction, thus providing a possible mechanism for the turnover of the membrane. A vertebrate photoreceptor cell contains a small cluster of actin filaments in its connecting cilium at the site where new transductive disk membranes are formed. Disruption of the actin filaments perturbs disk morphogenesis. The most likely explanation for this perturbation is that the process of initiating a new disk is inhibited. Conventional myosin (myosin II) is found in the connecting cilium with the same distribution as actin. A simple model is proposed to illustrate how the actin-myosin system of the connecting cilium might function to initiate the morphogenesis of a disk membrane.  相似文献

Is there a prima facie obligation to produce additional individuals whose lives would be worth living? In his paper ‘Is it Good to Make Happy People?’, Stuart Rachels argues not only that there is, but, also, that precisely as much weight should be assigned to the quality of life that would be enjoyed by such potential persons, if they were to be actualized, as to the quality of life enjoyed by actually existing persons. In response, I shall argue, first, that Rachels’ view is exposed to very serious objections, and secondly, that his arguments in support of his position involve a crucial assumption, which cannot be sustained, concerning the relation between, on the one hand, propositions about good-making and bad-making properties, and, on the other, propositions about right-making and wrong-making ones. I shall then argue that there is a very plausible position concerning the conditions under which an action can be morally wrong which entails the following asymmetry: there is a prima facie obligation not to bring into existence individuals whose lives are not worth living, but there is no corresponding obligation to create additional individuals whose lives would be worth living.  相似文献

The thermal stability and kinetic properties of purified NADP+-malate dehydrogenase (NADP+-MDH; EC 1.1.1.82) isomorphs were analyzed from plants of two populations of Barnyard grass from contrasting thermal environments. Plants from Québec (QUE) and Mississippi (MISS) were acclimated under controlled conditions at 26/20°C and 14/8°C (day/night). While the enzyme from QUE showed one isomorph, 3 isomorphs were detected in all plants from MISS, suggesting the presence of gene duplication and fixed heterozygosity for the expression of this dimeric enzyme. This findig raises the possibility that the enhanced acclimatory potential of NADP+-MDH from MISS plants, as found from previous studies with the partially purified and unfractioned enzyme, may result from differential kinetic properties of isomorphs which would allow for the proper modulation of catalysis over a wide temperature range. The thermal stability of the QUE isomorph was significantly higher than that of any of the MISS isomorphs. The apparent activation energy of the QUE isomorph was within the range of values found for the 3 MISS isomorphs which were similar to each other. The Michaelis-Menten constants (Km) for oxalacetic acid were not significantly different among isomorphs or between thermoperiods, but Km (NADP+) values for the QUE isomorph were significantly higher than those of two of the MISS isomorphs over the 15–25°C assay range Vmax/Km ratios for OAA and NADP+ were not significantly different among isomorphs or between thermoperiods. Our data indicate that, under highly purified conditions, the single NADP+-MDH isomorph of QUE plants possesses good acclimatory potential for maintaining catalytic efficiency under a wide range of temperature conditions. In vitro thermal and kinetic data do not support the hypothesis that the the multiple NADP+-MDH isomorphs found in MISS plants may have been selected to optimize the thermal and catalytic efficiency of NADP+-MDH under warm temperature conditions.  相似文献

Most people believe it would be wrong to bring a child into the world if in all likelihood its life would be miserable. But if pain and suffering count against bringing someone into existence, why do pleasure and happiness not count in favour of bringing them into existence? Recently in this journal Michael Tooley has re‐affirmed his rights‐based explanation for this asymmetry. In a nutshell: to create an individual whose life is not worth living would be to wrong that individual – to create an obligation that cannot be fulfilled – but it is not possible to wrong an individual who is not brought into existence. In the same issue of this journal, in an article covering a range of arguments for and against the claim that it would be good for additional people to exist, Stuart Rachels objects to Tooley’s account on the ground that it has counterintuitive implications. His most interesting argument involves a Parfit‐style counterexample: a woman is about to take a fertility pill that will result in twins, one of whom will be healthy and the other of whom will not. Does it make a difference, morally speaking, if the woman knows which of the twins will be healthy and which will not? In this paper I argue that both Rachels’ criticism of Tooley’s rights‐based account, and Tooley’s own defence of it, are unsuccessful due to their failure to come to grips with the semantics of names for possible individuals. Both of them implicitly assume that it is possible to have a potential person in mind, in a way that misleads them about the fairness of actions that involve possible people. The significance of this extends to other areas such as abortion, population policy, and embryo experimentation, where examples involving possible people are common.  相似文献

One can formulate the statement contained in the title in two different ways, the one having a physical meaning only, the other having a biological meaning too. By using ideas of information theory the second sentence reads: In each interaction between an organism and its environment a flow of energy and a flow of information may be distinguished as different aspects of the same process. In connection with the proposed formulation of a new definition for “stimulus” it is to be recognized the use of the above sentence is advantageous. In many biological and other scientific journals, a reader's understanding of a paper to the argument of which statistical methods and analyses are important is often impeded by confusions of terminology and ambiguities of symbols. This is not solely because statistics is a difficult subject for biologists! If an editor were to formulate and make known a code of statistical symbols, abbreviations, and technical terms that in his journal will be regarded as part of the normal language of science, an author could use these without need for explanation each time. Every author would remain free to depart from the code, provided that he defined clearly his own usages. Such a policy, supported by the journal's referees, would do much to remove the frequent necessity for a reader to guess an author's meaning. Similarly considerations apply to the use of statistical software packages, where there is an evident need for an author to declare what software (if any) he has used, in much the same way as, by established custom, he will carefully specify his experimental materials and methods where these in any respect differ from the obvious. The present paper is written to stimulate constructive debate, and in no way to dogmatize on the merits or faults of particular statistical methods. Its underlying spirit is that the author of a scientific communication has a duty to describe the making of his observations, the conduct of his computations, and the performance of his computations with a clarity that would permit their repetition by another scientist who has access to the appropriate facilities and resources.  相似文献

An analysis of the sequences of scyllatoxin and charybdotoxin suggested that it would be possible to design a core peptide sequence which would still fold to give the β-hairpin and helix seen in the toxins, but which would eliminate one disulfide and connecting residues. The core sequence was modeled, then synthesized and purified. The cysteines oxidize in air to give the same disulfide pairings as seen in the parent toxins as the major product. The three-dimensional structure of the core sequence peptide, termed Max, was determined using proton NMR spectroscopy and found to be identical in secondary structure to the toxins. However differences were found in the relative orientation of the β-hairpin and helix. The use of this structural motif, found in many insect toxins, as a disulfide framework for exploring sequence/structure/activity relationships is discussed. © 1994 John Wiley & Sons, Inc.  相似文献

In this paper, we address the question, can biologically feasible neural nets compute more than can be computed by deterministic polynomial time algorithms? Since we want to maintain a claim of plausibility and reasonableness we restrict ourselves to algorithmically easy to construct nets and we rule out infinite precision in parameters and in any analog parts of the computation. Our approach is to consider the recent advances in randomized algorithms and see if such randomized computations can be described by neural nets. We start with a pair of neurons and show that by connecting them with reciprocal inhibition and some tonic input, then the steady-state will be one neuron ON and one neuron OFF, but which neuron will be ON and which neuron will be OFF will be chosen at random (perhaps, it would be better to say that microscopic noise in the analog computation will be turned into a megascale random bit). We then show that we can build a small network that uses this random bit process to generate repeatedly random bits. This random bit generator can then be connected with a neural net representing the deterministic part of randomized algorithm. We, therefore, demonstrate that these neural nets can carry out probabilistic computation and thus be less limited than classical neural nets.  相似文献

Conclusion My conclusion is that Mendel deliberately, though without any real falsification, tried to suggest to his audience and readers an unlikely and substantially wrong reconstruction of the first and most important phase of his research. In my book I offer many reasons for this strange and surprising behavior,53 but the main argument rests on the fact of linkage. Mendelian genetics cannot account for linkage because it was based on the idea of applying probability theory to the problem of species evolution. Central to the theory is the law of probability according to which the chance occurrence of a combination of independent events is the product of their separate probabilities. This is the common basis of Mendel's first and second laws, but this is why Mendel's second law on independent assortment is enunciated in too general a way. From Morgan's work we now know that characters may not always be independent if their genes are located very close one to the other on the same chromosome. And this was also the basis of Mendel's personal drama: he surely observed the effects of linkage, but he had no theoretical tools with which to explain it. So he presented his results in a logical structure consistent with the central idea of his theory. Had he described the real course of his experiments he would have had to admit that his law worked for only a few of the hundreds of Pisum characters — and it would thus have been considered more of an exception than a rule. This is why he insisted on the necessity of testing the law on other plants, and this is why in his second letter to Carl Nägeli he admits that the publication of his data was untimely and dangerous.54.We can argue that already in 1866 Mendel was less confident that his so-called second law had the same general validity as the first — and that later he lost his confidence altogether. Contemporary testimony indicates that in the end he became as skeptical as all his contemporaries as to the scientific relevance of his theory.55 But he was wrong. His research is in no way the fruit of methodological mistakes or forgery, and it remains a landmark in the history of science. He was only the victim of a strange destiny in which the use of probability theory was responsible, at the same time, for the strength and for the weakness of his theory. A rapid change in an interfacial electric potential of isolated bovine rod outer segment disk membranes occurs upon illumination. This potential change, which has been detected by the use of spin-labeled hydrophobic ions, apparently occurs within a low dielectric boundary region of the membrane near the external (cytoplasmic) surface and is positive with respect to the aqueous exterior of the disk. The magnitude of the potential change is pH-and temperature-dependent and appears with a first-order half-time of approximately 7 ms at 21 degrees C. A simple model in which one positive charge per bleached rhodopsin is translocated from the cytoplasmic aqueous space into the membrane low dielectric boundary region readily accounts for all experimental observations. The great similarity of the boundary potential change to the R2 phase of the early receptor potential suggests that the two have the same molecular origin.  相似文献

This paper reviews the major reports of the spontaneous or carcinogen-induced transformation of human fibroblasts to the malignant state, to infinite lifespan, or to anchorage independence. In some cases, the transformed cells and the parent cell with which the work began were made available to us to be tested to determine whether the cells shared common isozymes, HLA antigens, restriction-fragment length polymorphisms, marker chromosomes, etc., as one would expect. When we examined the normal fibroblastic cell line KD for these markers, and the transformed HuT cell lines developed from it by Kakunaga (Proc. Natl. Acad. Sci. (U.S.A.), 75, 1334, 1978) for these markers, we found marked differences, indicating that KD cells and HuT cells are derived from different individuals. When we applied these techniques to the 3 human fibroblast cell lines transformed by Namba to acquire infinite lifespan in culture (Gann, 27, 221, 1981), it became clear that KSMT-6 was derived from the parent cell, KMS-6, but that both cell lines CT-1 and SUSM-1 were derived from the same parental cell line, AD387. Similar studies with other sets of cell lines are also reported. In the light of these studies, it appears that there is no example of the malignant transformation of human fibroblasts by carcinogen treatment. However, neoplastic transformation and transformation to infinite lifespan by carcinogen treatment have been achieved by a number of workers. We speculate as to how malignant transformation might be obtained by carcinogen treatment.  相似文献

Surfaces of rod photoreceptor disk membranes: integral membrane components   总被引:8,自引:4,他引:4  
The membrane surfaces within the rod outer segment of the toad, Bufo marinus, were exposed by rapid-freezing followed by freeze-fracture and deep-etching. Platinum-carbon replicas of disk membranes prepared in this way demonstrate a distinct sidedness. The membrane surface that faces the lumen of the disk shows a fine granularity; particles of approximately 6 nm are packed at a density of approximately 30,000/micron 2. These dimensions suggest that the particles represent protrusions of the integral membrane protein, rhodopsin, into the intradisk space. In addition, when rhodopsin packing is intentionally perturbed by exhaustive digestion with phospholipase C, a concomitant change is observed in the appearance of the luminal surface granularity. The cytoplasmic surface of the disk rarely displays this rough texture; instead it exhibits a collection of much larger particles (8-12 nm) present at approximately 10% of the concentration of rhodopsin. This is about the size and concentration expected for certain light-regulated enzymes, cGMP phosphodiesterase and GTP-binding protein, which are currently thought to localize on or near the cytoplasmic surface of the disk. The molecular identity of the 8-12-nm particles will be identified in the following companion paper. A further differentiation of the cytoplasmic surface can be seen around the very edge, or rim, of each disk. This rim has relatively few 8-12- nm particles and instead displays short filamentlike structures connecting it to other membranes. These filaments extend between adjacent disks, across disk incisures, and from disk rims to the nearby plasma membrane.  相似文献

Information from oblique thin sections and from three-dimensional reconstructions of tilted, transverse thin sections (Cheng, N., and J. F. Deatherage. 1989. J. Cell Biol. 108:1761-1774) has been combined to determine the three-dimensional structure of the honeybee flight muscle Z disk at 70-A resolution. The overall symmetry and structure of the Z disk and its relationship to the rest of the myofibril have been determined by tracing filaments and connecting elements on electron images of oblique sections which have been enhanced by a local crystallographic averaging technique. In the three-dimensional structure, the connecting density between actin filaments can be described as five compact, crystallographically nonequivalent domains. Features C1 and C2 are located on the transverse twofold rotation axes in the central plane of the Z disk. They are associated with the sides of actin filaments of opposite polarity. Features C3, C4, and C5 are present in two symmetry-related sets which are located on opposite sides of the central plane. C3 and C5 are each associated with two filaments of opposite polarity, interacting with the side of one filament and the end of the other filament. C3 and C5 may be involved in stabilizing actin filament ends inside the Z disk. The location of the threefold symmetric connection C4, relative to the thick filament of the adjacent sarcomere, is determined and its possible relationship to the C filament is considered.  相似文献

Language transfers information on at least three levels; (1) what is said, (2) how it is said (what language is used), and, (3) that it is said (that speaker and listener both possess the ability to use language). The use of language is a form of honest cooperation on two of these levels; not necessarily on what is said, which can be deceitful, but always on how it is said and that it is said. This means that the language encoding and decoding systems had to evolve simultaneously, through mutual fitness benefits. Theoretical problems surrounding the evolution of cooperation disappear if a recognition system is present enabling cooperating individuals to identify each other – if they are equipped with “green beards”. Here, I outline how both the biological and cultural aspects of language are bestowed with such recognition systems. The biological capacities required for language signal their presence through speech and understanding. This signaling cannot be invaded by “false green beards” because the traits and the signal of their presence are one and the same. However, the real usefulness of language comes from its potential to convey an infinite number of meanings through the dynamic handling of symbols – through language itself. But any specific language also signals its presence to others through usage and understanding. Thus, languages themselves cannot be invaded by “false green beards” because, again, the trait and the signal of its presence are one and the same. These twin green beards, in both the biological and cultural realms, are unique to language.  相似文献

Some ethical dilemmas in health care, such as over the use of age as a criterion of patient selection, appeal to the notion of life expectancy. However, some features of this concept have not been discussed. Here I look in turn at two aspects: one positive — our expectation of further life — and the other negative — the loss of potential life brought about by death. The most common method of determining this loss, by counting only the period of time between death and some particular age, implies that those who die at ages not far from that one are regarded as losing very little potential life, while those who die at greater ages are regarded as losing none at all. This approach has methodological advantages but ethical disadvantages, in that it fails to correspond to our strong belief that anyone who dies is losing some period of life that he or she would otherwise have had. The normative role of life expectancy expressed in the 'fair innings' attitude arises from a particular historical situation: not the increase of life expectancy in modern societies, but a related narrowing in the distribution of projected life spans. Since life expectancy is really a representation of existing patterns of mortality, which in turn are determined by many influences, including the present allocation of health resources, it should not be taken as a prediction, and still less as a statement of entitlement.  相似文献

Concluding statement Wallace's contributions to biological thought tend to be overlooked or overly praised, neither of which produces a satisfactory assessment. Examples of the latter tendency are the recent expositions by Brackman and Brooks; although both books contain much worthwhile material, both are flawed. At critical points their theories fail to measure up, Brackman's because of his misinterpretations of events in the month of June 1858, and Brooks's Darwin's September 5 letter to Gray could, and probably did, represent an ordering of his ideas in response to a felt challenge.A fruitful way to characterize the relationship between Darwin and Wallace may be found in terms of game theory. Most scholars look upon the relationship as a zero-sum game, with a winner and a loser, the matter of priority being considered as a single event. Another approach would be to look upon it as a non-zero-sum game with each man influencing the other. In this case, the productivity of one is stimulated by the contributions of the other, resulting in a net gain in knowledge overall, and both men become winners, or codiscoverers. This approach is possible if Wallace's contributions to the theory of evolution by means of natural selection are recognized.  相似文献

Schr?dinger stated in his landmark book, What is Life?, that life feeds on negative entropy. In this contribution, the validity of this statement is discussed through a careful thermodynamic analysis of microbial growth processes. In principle, both feeding on negative entropy, i.e. yielding products of higher entropy than the substrates, and generating heat can be used by microorganisms to rid themselves of internal entropy production resulting from maintenance and growth processes. Literature data are reviewed in order to compare these two mechanisms. It is shown that entropy-neutral, entropy-driven, and entropy-retarded growth exist. The analysis of some particularly interesting microorganisms shows that enthalpy-retarded microbial growth may also exist, which would signify a net uptake of heat during growth. However, the existence of endothermic life has never been demonstrated in a calorimeter. The internal entropy production in live cells also reflects itself in the Gibbs energy dissipation accompanying growth, which is related quantitatively to the biomass yield. An empirical correlation of the Gibbs energy dissipation in terms of the physico-chemical nature of the growth substrate has been proposed in the literature and can be used to predict the biomass yield approximately. The ratio of enthalpy change and Gibbs energy change can also be predicted since it is shown to be approximately equal to the same ratio of the relevant catabolic process alone.  相似文献

We have measured the ionic strength dependence of the rate constants for the electron-transfer reactions of flavin mononucleotide (FMN) and flavodoxin semiquinones with 10 high redox potential ferredoxins (HiPIP's). The rate constants were extrapolated to infinite ionic strength by using a theoretical model of electrostatic interactions developed in our laboratory. In all cases, the sign of the electrostatic interaction was the same as the protein net charge, but the magnitudes were much smaller. The results are consistent with a model in which the electrical charges are approximately uniformly distributed over the HiPIP surface and in which there are both short- and long-range electrostatic interactions. An electrostatic field calculation for Chromatium vinosum HiPIP is consistent with this. The presumed site of electron transfer includes that region of the protein surface to which the iron-sulfur cluster is nearest and appears to be relatively hydrophobic. The principal short-range electrostatic interaction would involve the negative charge on the iron-sulfur cluster. For some net negatively charged proteins, this effect is magnified, and for net positively charged HiPIP's, it is counterbalanced. The rate constants extrapolated to infinite ionic strength can be correlated with redox potential differences between the reactants, as has previously been shown for cytochrome-flavin semiquinone reactions. Both electrostatic and redox potential effects are magnified for the flavodoxin semiquinone as compared to the FMN semiquinone-HiPIP reactions. This note reviews Léon Croizat's ideas about the evolution of human behavior, based on his evaluation of the tool use capabilities of a capuchin monkey. Croizat theorized mainly on his orthogenetic view to explain such behavior. Interesting enough is the timeframe (1962) and context of his statement and the monkey's model used for such explanation. In this direction, some comments are provided considering the current knowledge of capuchin tool using and its potential implications for modeling hominid evolution.  相似文献

We have considered the simplest model of the Brody effect: a circular inhomogeneity into a conducting medium of infinite extent energized by a source sink pair near the disk. To correlate with the reciprocal field equations, the Brody effect has been calculated in its integrated form by the Gabor-Nelson method (1954). In these conditions, our results do not depend upon any singular position on the closed contour; they are also invariable with respect to grounding the inhomogeneity and/or shifting the inhomogeneity centre from that of the compartment around it. Contrariwise, characterizing the Brody effect by an enhancement factor of local potential values is sensitive to the above factors and so, may become confusing because inducing the belief that the effect due to an inhomogeneity intrinsically depends upon removing the point from which it is viewed.  相似文献
