AIMS: The propionibacteria are commercially important due to their use in the cheese industry, and there is a growing interest for their probiotic effects. Stimulatory effects of lactic acid bacteria (LAB) on propionic acid bacteria have been observed. This study was designed to examine the possibility of using spent media previously used to grow LAB for the production of biomass and metabolites of Propionibacterium freudenreichii subsp. shermanii. METHODS AND RESULTS: Seventeen MRS and vegetable juice media were prefermented by various LAB and evaluated for their ability to subsequently support the growth of Propionibacterium, using automated spectrophotometry (AS). Growth of Propionibacterium in spent media was strongly affected by the LAB strain used to produce the spent medium. The native MRS medium (not prefermented) yielded the highest optical density values followed by prefermented media by Lactobacillus acidophilus, Bifidobacterium longum and Lactococcus lactis. Prefermented cabbage juice enabled good growth of Propionibacterium. For the production of organic acids and vitamin B12, cells of Propionibacterium were concentrated and immobilized in alginate beads in the aim of accelerating the bioconversions. More propionic acid was obtained in spent media than in native MRS. The concentration of vitamin B12 was higher in media fermented with free cells than those with immobilized cultures; with the free cells, its concentration varied from 900 to 1800 ng ml(-1) of media. CONCLUSIONS: It was demonstrated that spent media could be recycled for the production of Propionibacterium and metabolites, depending on the LAB strain that was previously grown. Media remediation is needed to improve the production of vitamin B12, especially with immobilized cells. SIGNIFICANCE AND IMPACT OF THE STUDY: This study presents an option for recycling of spent media generated by producers of LAB or producers of fermented vegetables. The propionic fermentation may result in three commercial products: biomass, vitamin B12 or organic acids, which may be used as starters, supplements or food preservatives. It is an attractive process from economical and environmental standpoints.  相似文献

The purpose of this article is to reveal the role of the lactic acid bacteria (LAB) in the beebread transformation/preservation, biochemical properties of 25 honeybee endogenous LAB strains, particularly: antifungal, proteolytic, and amylolytic activities putatively expressed in the beebread environment have been studied. Seventeen fungal strains isolated from beebread samples were identified and checked for their ability to grow on simulated beebread substrate (SBS) and then used to study mycotic propagation in the presence of LAB. Fungal strains identified as Aspergillus niger (Po1), Candida sp. (BB01), and Z. rouxii (BB02) were able to grow on SBS. Their growth was partly inhibited when co-cultured with the endogenous honeybee LAB strains studied. No proteolytic or amylolytic activities of the studied LAB were detected using pollen, casein starch based media as substrates. These findings suggest that some honeybee LAB symbionts are involved in maintaining a safe microbiological state in the host honeybee colonies by inhibiting beebread mycotic contaminations, starch, and protein predigestion in beebread by LAB is less probable. Honeybee endogenous LAB use pollen as a growth substrate and in the same time restricts fungal propagation, thus showing host beneficial action preserving larval food. This study also can have an impact on development of novel methods of pollen preservation and its processing as a food ingredient.  相似文献

Lactic acid bacteria (LAB) are known for their special nutritional requirements, being usually cultured in complex media to achieve optimal growth. In this paper, a protocol based on trichloroacetic acid precipitation of peptides and proteins is presented. The method has been tested on four probiotic LAB strains grown in De Man Rogosa Sharpe (MRS) broth, a complex medium that is often used for the culture of such bacteria. This protocol allowed the detection of 19 proteins after sodium dodecyl sulfate-polyacrylamide gel electrophoresis, 10 of them being successfully identified by tandem MS. Thereafter, the 10 were found to be secreted or surface associated by bioinformatic means. In conclusion, this work supplies a method for the identification of proteins secreted by LAB, allowing discrimination between the proteins present in the MRS and those produced by probiotic LAB.  相似文献

An evaluation of the ability of various solid and liquid media to support both growth and antigen expression, particularly lipopolysaccharide (LPS) expression, by Helicobacter pylori culture collection strains and clinical isolates was performed. Liquid-based basal media (brain heart infusion, Brucella broth, Mueller–Hinton broth and tryptone soya broth) supported the growth of strains, whereas solid basal media of the same formulation did not support growth. Optimal growth of all strains was obtained on solid and in liquid media containing blood. Supplemented solid media containing supplements other than blood supported growth but only to a small extent. In liquid media excluding blood, serum supplements enhanced growth and horse serum was found to be superior to fetal calf serum. In general, β-cyclodextrin did not increase growth. Mueller–Hinton broth or tryptone soya broth containing horse serum and a nitrogen source such as yeast extract or proteose peptone no. 3 were found to give optimal growth of H. pylori in a blood-free environment. Strains after cultivation in liquid media, irrespective of composition, maintained production of high-molecular weight (mol. wt) LPS with an O side chain independent of medium composition, whereas subculturing on solid media resulted in production of low-mol. wt LPS. Expression of proteins differed in liquid and on solid media, particularly proteins of 57 and 60 kDa, but qualitatively no differences were observed upon supplementation of basal media.  相似文献

AIMS: To investigate the influence of the culture medium on antibiotic susceptibility testing of food-associated lactic acid bacteria (LAB) with the agar overlay disc diffusion (DD) method. METHOD: The antibiotic resistance profile of 39 food-associated lactobacilli and enterococci was determined with the agar overlay DD method using a defined medium (i.e. Iso-sensitest agar; ISA) or an undefined medium (i.e. de Man, Rogosa, Sharpe or MRS agar). RESULTS: The study revealed that ampicillin discs and, although to a lesser extent, also tetracycline discs consistently produced larger zones on MRS medium compared to ISA medium. For the antibiotics gentamicin, bacitracin and erythromycin, the radius of the inhibition zones produced on MRS medium was significantly smaller in relation to ISA. For categorizing LAB isolates into resistant, intermediate and susceptible groups, it was demonstrated that major errors can occur in determining bacitracin and gentamicin resistance if MRS medium instead of ISA medium is used. On the other hand, the performance of both media was found to be equivalent for testing tetracycline resistance. CONCLUSIONS, SIGNIFICANCE AND IMPACT OF THE STUDY: Despite the fact that MRS medium generally supports the growth of lactic acid bacteria much better than the nutrient-poor ISA medium, the present study clearly demonstrates that both media are not compatible in susceptibility testing against various classes of antibiotics. These results may stimulate future discussions on a generally recommended DD method for susceptibility testing of food LAB strains.  相似文献

Vermicompost preparations are increasingly used in agricultural practice. There is a possibility, that crop plants are sensitive to negative effect of vermicompost at early stages of development. The aim of the present study was to test the effects of vermicompost on seed germination and seedling growth of different vegetable crop species. Vermicompost substitution inhibited seed germination and seedling growth with almost linear decrease of growth with increasing concentration of vermicopost in the substrate. However, both leaf chlorophyll content and photochemical activity of photosynthesis increased in all crop species with the exception of pea seedlings. Vermicompost extract as a watering solution showed positive effect on growth of bean and pea seedlings. Germination response of vermicompost extract-imbibed seeds was clearly crop species-dependent. Hypocotyl growth was stimulated by low and moderate vermicompost extract concentrations. Radicle growth was more sensitive to negative effect of vermicompost extract. It is reported that both solid vermicompost and vermicompost extract contain number of active substances of both phenolic and humic nature, each with own dose- and genotype-dependent effect of seed germination and early stages of seedling development. Findings of this study suggests that vermicompost must be used cautiously for practical purposes of plant propagation.  相似文献

Five yeast extracts (YE) were fractionated by ultrafiltration (UF) with 1, 3, and 10 kDa molecular weight cutoff membranes, concentrated by freeze-drying, and the resulting powders of yeast extract filtrates (YEF) were evaluated for their growth-promoting properties on nine cultures of lactic acid bacteria (LAB). There was an increase in alpha-amino nitrogen content of the YEF powders as the pore size of the UF membranes used to filter the YE solutions decreased. The source of YE had a much greater effect than UF on the growth of LAB. This was also the case for the YEF contents in total and alpha-amino nitrogen. Growth curves of the LAB showed that maximum growth rate (mumax) data were on average 30% higher with bakers' YE than with brewers' YE, while maximum optical density (ODmax) values were on average 16% higher with bakers' YE. This could be related to the higher nitrogen content of the bakers' YE used in this study. Modification by UF of the YE had no significant influence on the growth of 4 of the 9 LAB strains. The three strains of Lactobacillus casei were negatively influenced by UF, as they did not grow as well in the media containing the YEF obtained after filtering with 1 and 3 kDa membranes. On a total solids basis, the 2.5 x retentates from the 10 kDa membrane gave, on average, 4% lower mumax and 5% lower ODmax values as compared to cultures where the corresponding YEF was used as medium supplement. This could also be partially related to the different nitrogen contents of the filtrates and retentates.  相似文献

Eighteen lactic acid bacteria (LAB) strains, isolated from coffee pulp silages were characterized according to both growth and gallic acid (GA) consumption. Prussian blue method was adapted to 96-well microplates to quantify GA in LAB microcultures. Normalized data of growth and GA consumption were used to characterize strains into four phenotypes. A number of 5 LAB strains showed more than 60% of tolerance to GA at 2 g/l; whereas at 10 g/l GA growth inhibition was detected to a different extent depending on each strain, although GA consumption was observed in seven studied strains (>60%). Lactobacillus plantarum L-08 was selected for further studies based on its capacity to degrade GA at 10 g/l (97%). MRS broth and GA concentrations were varied to study the effect on growth of LAB. Cell density and growth rate were optimized by response surface methodology and kinetic analysis. Maximum growth was attained after 7.5 h of cultivation, with a dilution factor of 1–1/2 and a GA concentration between 0.625 and 2.5 g/l. Results indicated that the main factor affecting LAB growth was GA concentration. The main contribution of this study was to propose a novel adaptation of a methodology to characterize and select LAB strains with detoxifying potential of simple phenolics based on GA consumption and tolerance. In addition, the methodology presented in this study integrated the well-known RSM with an experimental design based on successive dilutions.  相似文献

The exchange of ammonia between the atmosphere and the canopy of barley, wheat, oilseed rape and pea crops was studied over two growing seasons by use of a modified aerodynamic gradient technique in which passive horizontal flux samplers were applied with a wind profile in gradient configuration. The crop foliage was a net source of NH3 to the atmosphere, with NH3 emissions on a seasonal basis between 1 and 5 kg NH3–N ha–1. The amount of NH3 lost constituted between 1 and 4% of the applied nitrogen and between 1 and 4% of the actual amount of nitrogen present in the mature shoots. The volatile NH3 losses depended on seasonal variations in climatic conditions affecting the growth and nitrogen economy of the crops and increased under conditions with excessive N absorption by roots and a high N concentration in the foliage. The accumulated NH3 loss was positively correlated with the above-ground crop N content at anthesis, but not with that at final maturity. There were no indications that NH3 emissions were larger under conditions unfavourable for nitrogen remobilization from vegetative plant parts (low N harvest index). Nevertheless, a distinct peak in NH3 emission occurred during senescence. It is concluded that crops in many areas will represent a significant input of ammonia to the atmosphere and that NH3 losses may become large enough to significantly affect crop N budgets.  相似文献

We used culture- and molecular-biology-based methods to investigate microbial diversity in the traditional Mongolian fermented milks “Airag” (fermented mare’s milk) and “Tarag” (fermented milk of cows, yaks, goats, or camels). By rRNA or functional gene sequencing, we identified 367 lactic acid bacteria (LAB) strains and 152 yeast strains isolated from 22 Airag and 31 Tarag samples. The total concentration of LAB in Airag (107.78 ± 0.50 c.f.u. ml–1; mean ± SD) was significantly lower (P < 0.01) than in Tarag (108.35 ± 0.62 c.f.u. ml−1), whereas the total concentration of yeasts in Airag (107.41 ± 0.61 c.f.u. ml-1) was significantly higher (P < 0.01) than in Tarag (105.86 ± 1.29 c.f.u. ml-1). Lactobacillus helveticus and Lactobacillus kefiranofaciens were isolated from Airag as the predominant LAB strains at levels of about 107 c.f.u. ml−1, whereas Lactobacillus delbrueckii subsp. bulgaricus, L. helveticus, and Streptococcus thermophilus were the predominant isolates from Tarag at about 107 c.f.u. ml−1. The lactose-fermenting Kluyveromyces marxianus was isolated predominantly from Airag as its major alcoholic fermentation component. Non-lactose-fermenting yeasts such as Saccharomyces cerevisiae, Issatchenkia orientalis, and Kazachstania unispora were the predominant isolates from Tarag, at about 105 c.f.u. ml−1. The apparent geographic differences in the L. kefiranofaciens and S. thermophilus contents of Tarag strongly suggested that differences among the animal species from which the milk was sourced, rather than geographic distances, were the most important factors influencing the diversity of the microbial composition of traditional fermented milks in Mongolia.  相似文献

The aim of the study was to characterize lactic acid bacteria (LAB) which are components of drugs administered orally in cases of intestinal disturbances, or antibiotic--related diarrhea. Biochemical properties, growth behavior, bile tolerance, and survival at low pH of six LAB strains (four strains L. rhamnosus and two L. acidophilus) were studied. The survival at low pH was determined in MRS broth (Difco) acidified to pH 1; 2; 3; and 4. Bile tolerance was tested on MRS broth with 0.3% oxgall (Difco). Between tested strains differences in ability to grow at low pH and survival in bile were observed. Only 0.01% inoculum of all examined strains survived at pH 1. Differences between strains in survival at low pH (pH 2 and pH 3) and tolerance of bile were observed. However, after 2 h incubation at pH 4, 100% of strains stayed alive. Examined strains demonstrated good 3% bile tolerance. All strains met the criteria for probiotic strains: ability to survive at pH 3 and in the presence of bile.  相似文献

The Acinetobacter sp. E11, isolated from Port Dickson Beach, Malaysia, was able to grow in media containing crude oil as the sole carbon and energy source. Substrate specificity studies showed that the bacterium exhibited substrate preference as growth was observed only in media containing aliphatic hydrocarbons, while aromatic and cyclic hydrocarbons inhibited growth. With the aliphatic hydrocarbons, growth was seen only in the long-chain alkanes tested (pentadecane, dodecane and hexadecane). No growth was recorded in the short-chain alkanes (pentane, hexane and heptane) tested. With complex hydrocarbons, only crude oil and 4T SHELL engine oil supported growth. No growth was observed in kerosene and PETRONAS gasoline. The isolate could grow in up to 10% and 20% [v/v] of the crude oil and alkanes tested, respectively. Among the long-chain alkanes tested, hexadecane was the most preferred, followed by pentadecane and dodecane. Nitrogen and phosphorous supplements were essential for growth and the best growth was achieved with 3% nitrogen/phosphorous additions. Microscopic observation revealed that the bacterium adhered to the hexadecane and crude oil droplets. GC analysis showed that the bacterium was able to degrade more than 60% of the hydrocarbons in the crude oil in 15 days at 37°C compared to the uninoculated media.  相似文献

Field experiments were carried out during three successive years to study through a dynamic approach the competition for soil N and its interaction with N2 fixation, leaf expansion and crop growth in pea–barley intercrops. The intensity of competition for soil N varied between experiments according to soil N supply and plant densities. This study demonstrates the key role of competition for soil N which occurs early in the crop cycle and greatly influences the subsequent growth and final performance of both species. Relative yield values for grain yield and N accumulation increased with the intensity of competition for soil N. Barley competed strongly for soil N in the intercrop. Its competitive ability increased steadily during the vegetative phase and remained constant after the beginning of pea flowering. The period of strong competition for soil N (500–800 degree-days after sowing) also corresponded to the period of rapid growth in leaf area for both species and therefore an increasing N demand. For each species, the leaf area per plant at the beginning of pea flowering was well correlated with crop nitrogen status. Barley may meet its N needs more easily in intercrops (IC) and has greater leaf area per plant than in sole crops (SC). Barley having a greater soil N supply results in an even higher crop N status and greater competitive ability relative to pea in intercrop. Competition by barley for soil N increased the proportion of pea N derived from fixation. The nitrogen nutrition index (NNI) values of pea were close to 1 whatever the soil N availability in contrast to barley. However N2 fixation started later than soil N uptake of pea and barley and was low when barley was very competitive for soil N. Due to the time necessary for the progressive development and activity of nodules, N2 fixation could not completely satisfy N demand at the beginning of the crop cycle. The amount of N2 fixed per plant in intercrops was not only a response to soil N availability but was largely determined by pea growth and was greatly affected when barley was too competitive.  相似文献

Aims: The present work tests the feasibility of the isothermal microcalorimetry method to study the performance of individual lactic acid bacteria during solid‐state fermentation in rye sourdough. Another aim was to elucidate the key factors leading to the formation of different microbial consortia in laboratory and industrial sourdough during continuous backslopping propagation. Methods and Results: Strains of the individual LAB isolated from industrial and laboratory sourdough cycle were grown in 10 kGy irradiated rye dough in vials of an isothermal calorimeter and the power–time curves were obtained. Sugars, organic acids and free amino acids in the sourdough were measured. The OD–time curves of the LAB strains during growth in flour extract or MRS (De Man, Rogosa and Sharpe) broth were also determined. The maximum specific growth rates of Lactobacillus sakei, Lactobacillus brevis, Lactobacillus curvatus and Leuconostoc citreum strains that dominated in backslopped laboratory sourdough were higher than those of Lactobacillus helveticus, Lactobacillus panis, Lactobacillus vaginalis, Lactobacillus casei and Lactobacillus pontis strains originating from industrial sourdough. Industrial strains had higher specific growth rates below pH 4·8. It was supposed that during long‐run industrial backslopping processes, the oxygen sensitive species start to dominate because of the O2 protective effect of rye sourdough. Conclusions: Measurements of the power–time curves revealed that the LAB strains dominating in the industrial sourdough cycle had better acid tolerance but lower maximum growth rate and oxygen tolerance than species isolated from a laboratory sourdough cycle. Significance and Impact of the Study: Isothermal microcalorimetry combined with chemical analysis is a powerful method for characterization of sourdough fermentation process and determination of growth characteristics of individual bacteria in sourdough.  相似文献

Culture-based technique was used to study the population dynamics of the bacteria and determine the dominant lactic acid bacteria (LAB) during cassava fermentation. LAB was consistently isolated from the fermented mash with an initial viable count of 6.00 log c.f.u. g−1 observed at 12 h. The aerobic viable count of amylolytic lactic acid bacteria (ALAB) was higher than other group of LAB throughout the fermentation up to 96 h with the highest viable count of 8.08 log c.f.u. g−1. Combination of phenotypic parameters and 16S rDNA gene sequencing identified the dominant group of LAB as Lactobacillus plantarum, L. fermentum and Leuconostoc mesenteroides while the pulse field gel electrophoresis determined that the strains were genotypically heterogeneous. The sugar fermentation profile of the isolates showed that indigestible sugars such as raffinose and stachyose can be fermented by the strains. Information was also generated about the functional properties of the strains. Only strain L. plantarum 9st0 isolate at 0 h of the fermentation produced bacteriocin with antagonism against closely related indicator strains. Quantitatively, the highest amylase activity was produced by strain L. plantarum 7st12, while appreciable amylase was also produced by L. fermentum 1st96. The result of this work showed that selection of mixed starter cultures of bacteriocin- and amylase-producing L. plantarum and L. fermentum will be highly relevant as starter cultures during the intermediate and large scale gari production.  相似文献

Rhizobacteria belonging to Bacillus sp. were isolated from the rhizosphere of green gram (Vigna radiata). Seed inoculation with the rhizobacteria showed stunting effect on root growth whereas four Bacillus strains caused stimulation of shoot growth at both 4 and 7 d of observations. Coinoculation of some Bacillus strains with effective Bradyrhizobium strain S24 resulted in enhanced nodulation and plant growth of green gram. The shoot dry mass (ratio to uninoculated control) varied from 1.32 to 6.33 at day 30 and from 1.28 to 3.55 at day 40 of plant growth. Nodule promoting effect after 40 d of plant growth was observed with majority of Bacillus strains except for MRS9 and MRS26. Maximum gains in nodulation, nitrogenase activity and plant growth were observed with Bacillus strains MRS12, MRS18, MRS22 and MRS27 after 40 d of plant growth, suggesting the usefulness of introduced rhizobacteria in improving crop productivity.  相似文献

Samples of camel's milk collected from different zones of Morocco were analysed to evaluate their microbiological quality and to identify predominating lactic acid bacteria (LAB). The following average colony-forming units (c.f.u.s) of aerobic total count, enterococci, faecal and total coliforms, LAB, yeasts,Staphylococcus aureus and spores of sulphite-reducing clostridia were recorded: 6.2 × 107, 2.9 × 104, 1.6 × 104, 7.0 × 106, 1.0 × 107, 3.8 × 104, 1.3 × 105 and 6.0 c.f.u./ml, respectively. The enumeration results were markedly variable and coliforms were not detected in 1 ml of some samples. Bacteriological identification revealed a definite dominance of enterococci with Enterococcus faecalis as the main representative species. Besides Enterococcus, other genera including Pediococcus (28.2%), Streptococcus (4%), Lactococcus (8%) and Leuconostoc(1%) were isolated on de Man, Rogosa and Sharp (MRS) agar.  相似文献

Nutrient agar plates stored in light and air for 48 h became inhibitory for Campylobacter jejuni, C. coli and nalidixic acid-resistant, therrnophilic campylobacter (NARTC) strains. All five campylobacter test strains showed > 5 log reduction in counts on media which had been stored in light and air. Media stored in the dark and/or in a reduced atmosphere did not become inhibitory and supported the growth of campylobacters. Ferrous sulphate, sodium pyruvate, blood, charcoal or sodium metabisulphite, compounds frequently used as supplements in campylobacter media, were added to nutrient agar prior to storage of media in light and air. All additives except sodium metabisulphite prevented the accumulation of photochemically generated toxic oxygen derivatives and allowed growth of test strains. In qualitative tests to determine the ability of supplements to neutralize hydrogen peroxide, blood was the most active, charcoal and sodium pyruvate slightly less active and ferrous sulphate and sodium metabisulphite the least active. The results of this study confirm that supplements in campylobacter media act as quenching or detoxifying agents and not as enrichment factors.  相似文献

Growth of a highly virulent strain of the phytopathogen Corynebacterium fascians on rich media at 37 degrees C resulted in a loss of virulence in a majority of the population within 10 generations. Strains retained virulence during cultivation at 30 degrees C on a minimal medium with ammonia as a nitrogen source. Populations of avirulent strains on the surfaces of pea seedlings decreased, whereas the number of cells of the virulent strain increased 1,000-fold during a 3-week period. All avirulent mutants isolated by growth on rich media at 37 degrees C were unable to grow on media containing agmatine or proline as sole sources of nitrogen. The ability of the mutants to grow on pea seedlings and cause fasciation disease appeared to be related to their ability to utilize nitrogen sources available on plant surfaces.  相似文献
