Summary Attempts at the reciprocal cross between Petunia parodii and P. inflata using standard emasculation and pollination techniques failed. Limited pollen tube growth down the style in reciprocal crosses led to reproductive isolation between the self-compatible P. parodii and self-incompatible P. inflata. The interspecific hybrid was successfully produced by bud-pollination of P. parodii with P. inflata as the male parent in 22 percent of attempts, but not in the opposite direction. In vitro pollination of P. parodii ovaries with P. inflata pollen also produced hybrids. The small size of the ovary made it technically impossible to use P. inflata as the female parent for in vitro pollination. The interspecific hybrids were intermediate, as compared to the two parents, for six of the seven plant and flower characters measured. Furthermore, the hybrids had high pollen fertility, set abundant seed upon self-pollination, and readily inter-crossed with the parental species. The results are consistent with a high degree of chromosomal homology in the parental species and with minor genetic divergency leading to reproductive isolation that is pre-zygotic in nature. Overcoming the barriers to cross-incompatibility by practical techniques resulted in fertile interspecific hybrids that segregated for parental characters. Larval growth and development of hybrid flounder were observed and compared with those of their parent species. The reciprocal hybrids of female starry flounder Platichthys stellatus and male stone flounder Kareius bicoloratus (hybrid Sb) and of female K. bicoloratus and male P. stellatus (hybrid Bs) both survived and grew to juveniles. Development was divided into nine stages (A–I). Many of the hybrids' traits were identical and intermediate to those of their parents. The position of the eye, however, was primarily sinistral in both hybrids (80% in Sb and 76% in Bs), a trait possessed by P. stellatus (80%) in the western Pacific Ocean. The daily growth rates of the larvae were similar. In the parent species, development was more rapid in P. stellatus than in K. bicoloratus whereas rate of development was intermediate in both Sb and Bs hybrids. The size at settlement [standard length (LS) at stage H (mean ± s.d. )] was 9·82 ± 1·47 mm for the hybrid Sb and 9·99 ± 0·90 mm for the hybrid Bs, while the minimum age at metamorphosis (initial age at stage H) was 29 days after hatching (DAH) in both hybrids. In comparison, LS at settlement in parent species was 6·43 ± 0·25 mm for P. stellatus and 12·87 ± 1·29 mm for K. bicoloratus. Minimum age at metamorphosis for the parents was 23 DAH at stage G in P. stellatus and 34 DAH at stage H in K. bicoloratus. Thus, the timing of settlement of hybrids was different from that of their parent species. These traits may occur with high frequency in a natural habitat.  相似文献

Synopsis The development of photobehavioural responses in brook (Salvelinus fontinalis) and lake (S. namaycush) charr was studied by monitoring the intrasubstrate movements and concurrent photoresponse behaviour of incubated embryos and alevins. Photoresponse behaviour of both F-1 hybrids of the parent species was also recorded. All embryos initially moved downward in the substrate, however brook charr descended farther and faster into the substrate than did lake charr. Photoresponse tests demonstrated a similar pattern of photoresponse transformation from a photonegative to a photopositive state in both species. However, photoresponse reversal was faster, more extensive and occurred later in brook charr than in lake charr. Patterns of photoresponse change in F-1 hybrids were intermediate between those of the parent species. Photoresponse shifts preceded the onset of alevin emergence in both species. occurring when differential development of various morphological characters existed. Developmental states of characters were synchronously maximal towards the end of alevin emergence. Intermediate measures of morphological development were observed for F-1 hybrids. Possible functions and mechanisms of photoresponse transitions are discussed in relation to ecological differences between the species.  相似文献

Summary Somatic hybrid plants regenerated following the fusion of leaf mesophyll protoplasts of Petunia parodii with those isolated from a cell suspension of albino P. inflata. These two species exhibit a unilateral cross-incompatability with a pre-zygotic mode of reproductive isolation preventing hybridizations with P. inflata as the maternal parent. Selection of somatic hybrids relied on the fact that unfused or homokaryon protoplasts of P. parodii did not develop beyond the cell colony stage while those of the putative somatic hybrids and albino P. inflata parent produced callus. Green somatic hybrid calluses were readily identified against the white background of P. inflata following complementation to chlorophyll synthesis proficiency and continued growth in hybrid cells. Shoots, and ultimately flowering plants, were identified as somatic hybrids based on their floral morphology and colour, chromosome number and the fact that they segregated for parental characters. The frequency of somatic hybrid production was comparable to that previously established for two sexually compatible Petunia species.  相似文献

A few individuals with intermediate morphology always appeared in the sympatric distributions of Gentiana straminea and G. siphonantha. These intermediate individuals were hypothesized to be the hybrids of two species after a careful evaluation of their morphological characteristics. To test this hypothesis, sequence comparison of the internal transcribed spacer (ITS) regions of the nuclear ribosomal and trnS (GCU)-trnG (UCC) intergenic spacer region of the chloroplast DNA from Gentiana straminea, G. siphonantha and the putative hybrids was performed. The results suggest that most intermediate individuals were the natural hybrids between G. straminea and G. siphonantha. In addition, we examined the sequence variation among the individuals of both parent species and analyzed the possibility leading to the incongruent identification in some individuals based on morphologic and molecular evidences, respectively. Emasculated and unemasculated crosses of tetraploid A. spicatum f. inerme X A. desertorum yielded four hybrids. The hybrids were morphologically intermediate between the parent species but resembled A. desertorum more closely than A. spicatum. Both parents behaved cytologically as autoploids. Mean chromosome associations of 0.04 I, 8.60 II, 0.01 III, and 2.67 IV were observed at diakinesis in the 28-chromosome A. spicatum. The A. desertorum parent contained 30 chromosomes, 2 of which were likely supernumeraries, and averaged 0.03 I, 9.85 II, and 2.57 IV at diakinesis. Three hybrids contained 30 chromosomes, and one had 29. The most common chromosome association in the 30-chromosome hybrids was 2 I and 14 II; and the average was 3.00 I, 13.40 II, 0.06 III, and 0.01 IV. A. spicatum and A. desertorum chromosomes were usually distinguishable from each other in the hybrid cell on the basis of size. All pairing in the hybrids was attributed to autosyndesis within parental genomes. A. spicatum, A. desertorum, and their hybrids were represented by genome formulas of SSSS, CCCC, and SSCC, respectively. The hybrids produced 5 to 439 seeds under open pollination. Three controlled crosses between the hybrids yielded 2, 5, and 23 seeds, respectively, on 10 maternal spikes in each cross. The prospects of developing a fertile, cytologically stable allotetraploid species from the hybrids appear favorable.  相似文献

Researchers recognized early that chromosome behavior, as other morphological characters, is under genetic control and gave some cytogenetical examples such as the homoeologous chromosome pairing in wheat. In the intergeneric sexual hybrids between cultivated Brassica species and another crucifer Orychophragmus violaceus, the phenomenon of parental genome separation was found under genetic control during mitosis and meiosis. The cytogenetics of these hybrids was species-specific for Brassica parents. The different chromosome behavior of hybrids with three Brassica diploids (B. rapa, B. nigra and B. oleracea) might contribute to the different cytology of hybrids with three tetraploids (B. napus, B. juncea and B. carinata). The finding that genome-specific retention or loss of chromosomes in hybrids of O. violaceus with B. carinata and synthetic Brassica hexaploids (2n=54, AABBCC) is likely related to nucleolar dominance gives new insight into the molecular mechanisms regarding the cytology in these hybrids. It is proposed that the preferential expressions of genes for centromeric proteins from one parent (such as the well presented centromeric histone H3) are related with chromosome stability in wide hybrids and nucleolar dominance is beneficial to the production of centromere-specific proteins of the rRNAs-donor parent and to the stability of its chromosomes.  相似文献

Summary The alkaloid profiles and morphological traits of the capsules of Papaver bracteatum, P. pseudo-orientale, and their hybrids were studied. Dominance of the hexaploid parent P. pseudo-orientale was observed for various characters. A genetic model assuming allelic additive effects and polysomic inheritance was elaborated for the control of isothebaine content in the capsules. The distribution of thebaine content in the segregating generations, F2 and BCF1 was evidence of the transfer of genes from the diploid parent P. bracteatum in the gametes of the interspecific hybrid and their expression in its progenies. Interspecific hybrids have been proposed to have reduced developmental stability in comparison to their parental species because the parental genomes have not undergone selection for the maintenance of developmental stability when they occur together. We present data from four interspecific hybrids of salmonid fishes that support this view. Natural hybrids of bull trout (Salvelinus confluentus) with brook trout (Salvelinus fontinalis) and laboratory hybrids of rainbow trout (Salmo gairdneri) with Yellowstone (Salmo clarki bouvieri), westslope (S. c. lewisi), and coastal (S. c. clarki) cutthroat trout all have higher levels of fluctuating asymmetry than either of their parental species raised in the same environment. Thus, the hybrids have reduced developmental stability. The hybrids do not have meristic counts intermediate to the counts of the parental species. The hybrids usually have counts as high as the species with the higher count for those characters that differ between the parental species and often have higher counts for those characters that do not differ between the parental species. We suggest that the tendency for interspecific hybrids to have high meristic counts may be related to differences between the species in the length and timing of the developmental periods during which the counts of the characters are determined.  相似文献

Summary Recurrent or introgressive backcrossing of Phaseolus vulgaris — P. acutifolius hybrids with either P. vulgaris or P. acutifolius increased fertility, as measured by pollen stainability and seed per pod from non-manipulated flowers, while invariably resulting in loss of traits from the non-recurrent parent. When hybrids were backcrossed with each of the parent species in alternate generations (congruity backcrossing), fertility decreased in early generations but gradually increased in later generations. By the fourth or fifth generation, congruity-backcross hybrids produced numbers of seeds per pod from non-manipulated flowers comparable to those of parent species, although the percent of stainable pollen tended to be lower. Congruity-backcross hybrids were intermediate to parent species when pedigrees contained equal representation of parents but favored the majority parent when pedigrees were unbalanced. Individuals exhibiting symptoms of developmental incongruity, such as abnormal leaflet number, foliar variagation, or unusual growth patterns, occurred in each generation. However, completely new characteristics appeared after the second backcross generation, presumably due to recombination. Hybridization and introgression are thought to be important for speciation and adaptation in many plants. However, little is known about the hybridization and introgression among Cypripedium species. To investigate the evidence for hybridization and the pattern of introgression between Cypripedium yunnanense and C. tibeticum in Shangrila County, Yunnan Province, China, morphological characters and amplified fragment length polymorphism (AFLP) data for both the species and their putative hybrids were studied. Hand pollination was also performed to verify the crossability of the putative parents. Principal coordinate analysis based on morphological characters and the AFLP data suggested that the putative hybrids were true hybrids of these two Cypripedium species. Analysis with the NewHybrids software indicated that the putative hybrids were F1 generation individuals and backcrosses to C. yunnanese, but no F2 generation was found. Analysis with the Structure software demonstrated asymmetric introgression from C. tibeticum to C. yunnanense. We conclude that natural hybridization and introgression can occur between these two species and that in situ conservation of the parental species is required before fully assessing the evolutionary potential of hybrids.  相似文献

In a numerical study of taxonomic relationships in Cucurbita, the inclusion of F1 interspecific hybrids did not materially alter the species grouping in the phenogram. The F1 interspecific hybrids tend to cluster with one of the parent species or species groups unless the parents are widely divergent. Also, the F1 interspecific hybrids between wild and domesticated species cluster with the wild parent Both parent species or species groups of an F1 interspecific hybrid can usually be determined from Q-correlation values. It is essential that compatibility relations be determined between species; i.e., species groups must be established before Q-correlation values can be successfully used in determining the parentage of hybrids. High amylose content (AC) in rice endosperm is correlated with poor grain quality, particularly in indica hybrid rice. We have generated several homozygous transgenic parent lines of indica hybrid rice carrying an antisense Waxy (Wx) gene and demonstrated that the AC in seeds of these lines decreased dramatically. Two transgenic maintainer lines (L25B and L18B), derived from one of the key maintainer parents of an indica hybrid rice in China, Long-te-fu B (LTF-B), were selected and the antisense Wx gene was subsequently introgressed into the male-sterile counterpart, LTF-A, with the aim to generate improved indica hybrids. The indica hybrids derived from the selected transgenic male-sterile lines and restorer lines were tested for quality and agronomic performance under normal field conditions. Our results demonstrated that the reduction of AC in the homozygous transgenic maintainer lines stably passed down in five successive generations and the improved quality was also found in their relevant transgenic hybrids produced. The other two key characters of rice cooking and eating quality, the gel consistence (GC) and gelatinization temperature (GT), were also improved in the grains of both the transgenic maintainer lines and their relevant hybrids. In addition, no change was observed for most of the agronomic characters of the transgenic maintainer lines and the relevant transgenic hybrids. Although the grain weight of the transgenic line was reduced, the grain yield of the homozygous transgenic parent lines and the transgenic hybrids was similar when compared with that of the wild-type controls. These results suggest that transgenic approaches are an effective way to obtain rice lines with both improved qualities and high yield, especially for indica hybrid rice.  相似文献

The objective was to identify spontaneous hybrids between P. mugo and P. sylvestris using organelle DNA markers in sympatric zones at the sea‐side spit of Kursiu Nerija in western Lithuania. A field inventory was carried out over the entire Lithuanian part of the spit and 203 individuals morphologically intermediate between P. sylvestris and P. mugo were tested for their male parent with chloroplast DNA PCR‐RFLP markers and for their female parent with mitochondrial DNA PCR markers. Unfortunately, the mitochondrial DNA Nad7 marker failed to identify the female parent species in our study. However, the chloroplast DNA PCR‐RFLP marker revealed that out of 203 tested individuals only 23 had a paternity different than that indicated by morphology. Of these, 13 individuals were morphologically identified as P. sylvestris but possessed cpDNA of P. mugo (putative hybrids with P. sylvestris (female) ×P. mugo (male parent), and 10 individuals morphologically identified as P. mugo possessed cpDNA of P. sylvestris and may be hybrids with P. mugo (female) ×P. sylvestris (male parent). The remainder of the 177 individuals identified in the field inventory were were considered as pure species. In conclusion, our study indicates ongoing spontaneous hybridization between P. mugo and P. syvestris in Kursiu Nerija. Human impact via seed transfer on altered hybridization rates as well as evolutionary consequences are discussed.  相似文献

Summary Crossability between the diploid species S. circaeifolium subsp. circaeifolium (crc) and other diploid species, primarily diploid S. tuberosum subsp. tuberosum (tbr-2x), was studied. Forty-seven hybrids were obtained from crosses between crc as female parent and tbr-2x and some other species from series Tuberosa as male parents. Of these hybrids 17% were diploids; the other 83% were triploids, probably carrying two genomes of crc. Female fertility was sufficient to obtain offspring from backcrosses with the cultivated parent. Pollen stainability of the f1 varied, and micro-pollen as well as unreduced pollen occurred. During meiosis of the diploids and triploids a rather high proportion of univalents was found, and in the triploids on average two or three trivalents per cell were found. All hybrids were resistant to Globodera pallida pathotypes 2 and 3, and 75% of the tested genotypes were highly resistant to Phytophthora infestans. Solanidine, tomatidine, tomatidenol, and demissidine glycosides were found in tubers of the hybrids. Comparisons with somatic hybrids between crc and tbr-2x are made. It is concluded that crc is a valuable Solanum species that can and should be included in potato breeding programs.  相似文献

Summary Somatic hybridization of rice (Oryza sativa L.) and barnyard grass (Echinochloa oryzicola), a close relative of barnyard millet, was attempted using electrofusion and a new culture method developed for rice protoplasts (Kyozuka et al. 1987) to incorporate some of the agronomically important characters of the latter species into rice. Selection of hybrids was based on inactivation of rice protoplasts by iodoacetamide and the inability of barnyard grass protoplasts to divide. A total of 166 calli were identified as hybrids by isozyme and chromosome analyses. Hybrid calli were highly morphogenic, and 44 shoots were obtained. Most of them, however, were abnormal, and nine grew to plantlets whose morphology was distinct from that of either parent. Our study clearly demonstrates the totipotency of protoplasts in graminaceous monocots.  相似文献

Summary F1 interspecific hybrids involving nine tetraploid Triticum species were studied. Some developed leaf tumours at the seedling stage. Tumorous hybrids were restricted to crosses involving either T. timopheevi or T. araraticum as one parent. The hybrids from the rest of the crosses, including those of T. timopheevi × T. araraticum, were non-tumorous. Genetically divergent and non-integrated parental species appeared to be inducing spontaneous tumour formation in their hybrids.  相似文献

Interspecific somatic hybrids between a dihaploid potato clone H-8105 susceptible to Phytophthora infestans (Mont.) de Bary and a resistant diploid tuberizing species Solanum bulbocastanum were generated and analysed. Only ten regenerants displaying the intermediate morphology with dominating characteristics of the wild parent (simple leaves, anthocyanin pigmentation) were produced in 15 weeks after a single PEG-mediated fusion event. The RAPD patterns confirmed the hybridity of all of them. The hybrids rooted poorly and grew slowly in vitro. The cytological analysis revealed a high degree of aneuploidy in the hybrids with morphological and growth anomalies in vitro, while the morphologically normal hybrids were tetraploids. All the S. bulbocastanum (+) H-8105 hybrids were unstable in culture and three of them were consequently lost during three years of propagation in vitro. The possible reasons for instability of somatic hybrids between the distantly related species are discussed.  相似文献

Summary Phenotypic and flowering characteristics of hybrid plants generated by protoplast fusion between a tetraploid S. tuberosum line and diploid S. brevidens were assessed under field conditions. Hybrids were compared to both clonal parental material and protoplast-derived plants of each parent. Almost all of the hybrids were hexaploid. A wide range of variation in morphological characters was observed for hybrids and protoclones. Flowering was markedly reduced in protoclones. The majority of hybrids flowered, had viable pollen and set tubers. Tuber and pollen characteristics of hybrids produced from individual fusion calli also varied. The potential usefulness of fusion hybrids in potato improvement is discussed.  相似文献

 Interspecific asymmetric somatic hybrids have been produced between the sexually incompatible annual Medicago species Medicago truncatula and M. scutellata using only one regenerable parent and improved protoplast techniques. Regenerable M. truncatula leaf protoplasts were inactivated by lethal doses of iodoacetamide. Cotyledon protoplasts of non-regenerable M. scutellata were produced from 9 to 13 day old γ-irradiated cotyledons. Polyethylene glycol was used for protoplast fusion, and its successful use depended on a strategy to eliminate toxic effects. More than 50 mature regenerated plants were obtained from the fusion products and amplified fragment length polymorphism and chromosome analysis of a sample of 20 plants allowed the identification of two asymmetric hybrid plants. This technique is suitable for transferring characters from incompatible annual Medicago species to Medicago truncatula.
