The DNAs from two virilis group species of Drosophila, D. lummei and D. kanekoi, have been analyzed. D. lummei DNA has a major satellite which, on the basis of CsCl equilibrium centrifugation, thermal denaturation, renaturation and in situ hybridization is identical to D. virilis satellite I. D. kanekoi DNA has a major satellite at the same buoyant density in neutral CsCl gradients as satellite III of D. virilis. However, on the basis of alkaline CsCl gradients, the satellite contains a major and a minor component, neither one of which is identical to D. virilis satellite III. By in situ hybridization experiments, sequences complementary to the major component of the D. kanekoi satellite are detected in only some species and in a way not consistent with the phylogeny of the group. However, by filter hybridization experiments using nick-translated D. kanekoi satellite as well as D. lummei satellite I and D. virilis satellite III DNAs as probes, homologous sequences are detected in the DNAs of all virilis group species. Surprisingly, sequences homologous to these satellite DNAs are detected in DNAs from non-virilis group Drosophila species as well as from yeast, sea urchin, Xenopus and mouse.  相似文献

PTL-1, a microtubule-associated protein of the structural MAP2/tau family, is the sole member of this gene family in Caenorhabditis elegans. Sequence analysis of available invertebrate genomes revealed a number of single, putative tau-like genes with high similarity to ptl-1. The ptl-1 gene is expressed in a number of cells, most notably mechanosensory neurons. We examined the role of ptl-1 in C. elegans in adult neurons as well as during development. A ptl-1 knockout strain of worms exhibited an egg-hatching defect, as well as a reduced sensitivity to touch stimuli. In addition, the knockout allele ptl-1(ok621) acts as a dominant enhancer of several temperature-sensitive alleles of mec-7 and mec-12, which code the isoforms of β-tubulin and α-tubulin that together form the unusual 15 protofilament microtubules involved in touch sensation. These results demonstrate for the first time a functional role for this microtubule-associated protein in nematodes and suggest that PTL-1 is involved in mechanosensation as well as some aspect of embryogenesis.  相似文献

Yonagunia Kawaguchi et Masuda, gen. nov. (Halymeniaceae, Rhodophyta) is proposed to accommodate a new species, Yonagunia tenuifolia Kawaguchi et Masuda and the species currently known as Prionitis formosana (Okamura) Kawaguchi et Nguyen. Based on auxiliary cell ampullar features, Yonagunia is included in the group of genera with the simplest type of ampulla (the Grateloupia type) that comprises Dermocorynus, Grateloupia, Kintokiocolax, Phyllymenia, and Zymurgia. However, Yonagunia differs from these genera in the behavior of cells in the ampullar filaments immediately after diploidization, most cells of the primary and secondary filaments simultaneously dividing to form grape‐like clusters of small globular cells that subsequently elongate and produce involucral filaments to laxly surround the maturing carposporophyte. Yonagunia is resolved by our rbcL gene sequence analyses as one of five monophyletic clades within the Halymeniaceae (an Aeodes/Pachymenia, a Polyopes, a Carpopertis/Cryptonemia/Halymenia, a Yonagunia, and a Grateloupia clade) that is positioned as sister to the Grateloupia clade. Carpogonial branch apparatuses are identified as a potential taxonomic significance on the same level as auxiliary cell ampullae.  相似文献

Acanthamoeba castellanii, known as the “Trojan horse of the microbial world,” is known to host a variety of microorganisms including viruses, yeasts, protists, and bacteria. Acanthamoeba can act as a vector and may aid in the transmission of various bacterial pathogens to potential hosts and are found in a variety of places, thus impacting the health of humans, animals, and the environment. These are interconnected in a system known as “one health.” With the global threat of antibiotic resistance, bacteria may avoid harsh conditions, antibiotics, and disinfectants by sheltering within Acanthamoeba. In this study, Acanthamoeba castellanii interaction with Morganella morganii, a Gram-negative bacterium was studied. Escherichia coli K1 interaction with Acanthamoeba was carried out as a control. Association, invasion, and survival assays were accomplished. Morganella morganii was found to associate, invade, and survive within Acanthamoeba castellanii. Additionally, Escherichia coli K1 was also found to associate, invade, and survive within the Acanthamoeba at a higher number in comparison to Morganella morganii. For the first time, we have shown that Morganella morganii interact, invade, and survive within Acanthamoeba castellanii, suggesting that Acanthamoeba may be a potential vector in the transmission of Morganella morganii to susceptible hosts. Taking a one health approach to tackle and develop disinfectants to target Acanthamoeba is warranted, as the amoebae may be hosting various microbes such as multiple drug-resistant bacteria and even viruses such as the novel coronavirus.

The Scorpaena cardinalis complex, including S. cardinalis, S. jacksoniensis and S. orgila, is defined. The genus Ruboralga (type species: S. jacksoniensis) is regarded as a junior synonym of Scorpaena. Scorpaena jacksoniensis Steindachner 1866, previously treated as a junior synonym of Scorpaena cardinalis Solander and Richardson 1842, is regarded here as a valid species. Scorpaena cookii Günther 1874, previously treated as a valid species, is regarded here as a junior synonym of S. cardinalis. Thus, recent recognition of the two Australasian scorpionfishes, i.e., S. cardinalis and S. cookii, are re-identified in this study as S. jacksoniensis and S. cardinalis, respectively. Scorpaena plebeia Solander 1842 is regarded as a junior synonym of S. cardinalis. Scorpaena jacksoniensis is distributed along the east coast of Australia from southern Queensland to eastern Victoria, whereas S. cardinalis occurs around northern New Zealand, the Kermadec Islands and offshore islands of the Tasman Sea. A neotype is designated for S. cardinalis. Morphological changes with growth in the two species are described in detail.  相似文献

Twenty-three genera of Amaranthaceae occur in the New World. Two endemic genera occur in the Hawaiian Islands. Among the genera of the subfamily Amaranthoideae, Celosia, Cyathula and Achyranthes have their main distributions in the Old World; the two last-named genera are represented in the Americas only by widespread weeds. All the New World genera of the subfamily Gomphrenoideae are mainly or entirely restricted to this region. Characters of androecium and gynoecium are fundamental in the recognition of genera within the family. Androecia of different genera may be structurally and phylogenetically more similar than would appear from a cursory examination. It is suggested that the type of staminal tubes found in Pseudogomphrena and Froelichia can be derived from that in Alternanthera and Froelichiella by reduction of filament length and a fusion of pseudostaminodia with the filaments. The staminal tube in Gomphrena could result from a further decrease in distance between pseudostaminodia of the Pseudogomphrena type, and a deeper forking of the pseudostaminodia; each so-called apical filament lobe in Gomphrena would then be homologous with half a pseudostaminodium in Pseudogomphrena. Much of the variation in the androecia of these and other genera, as well as within genera such as Pfaffia, can be explained as the combined results of coalescence and splitting-up tendencies. Splitting up of staminal tubes may not necessarily take place along the borders of phylogenetically original filaments and pseudostaminodia. The Amaranthus-type of pollen is found in the majority of genera of the subfamily Amaranthoideae, but also in the Chenopodiaceae. A group of genera within the subfamily Gomphrenoideae also has pollen very similar to, or identical with, this type. Most genera of the subfamily Gomphrenoideae have pollen of the Gomphrena-type. Pseudoplantago has unilocular (at anthesis) anthers, a characteristic of the subfamily Gomphrenoideae, but floral structure as well as pollen morphology connect the genus to a group of genera within the Amaranthoideae, subtribe Achyranthinae. The combination of subcuboidal shape and opercula with radially arranged hooked protuberances, makes the pollen of Pseudoplantago unique among the angiosperms studied so far. Floral morphology and palynological characteristics indicate a close relationship between Pfaffia and Alternanthera. Both genera, as currently accepted, are relatively homogeneous from pollen morphological points of view. There are no correlations between pollen morphology and the variation in the androecium in Pfaffia, nor would pollen structure support recognition of Hebanthe as a distinct genus. Woehleria and Irenella may be derived from, or be of the same origin as, Dicraurus and Iresine. All four genera are placed in the subfamily Gomphrenoideae because of the bisporangiate anthers, but their pollen structure is very close to, or identical with, that of the Amaranthus-type. Pseudogomphrena combines characteristics of Gomphrena and Pfaffia.  相似文献

An intracellular parasite occurring inCryptomonas rostratiformis and less numerously also inC. erosa andC. phaseolus. The parasite is described. It grows in the dorsal side of the host near the nucleus from which it is optically indistinguishable in young stages. When mature the parasite fills 1/3 to 1/2 the volume of theCryptomonas cell. It is seen as a colourless blister, which pushes back the plastid of the host. Reproduction occurs by separation of the protoplast into a hundred or moreBodo-like swarmers which perhaps represent the infectious phase. Under certain conditions, however, such as during decline of theCryptomonas population, the parasite transforms into thick-walled spindle-shaped cysts. Like the swarmers these cysts are released by rupture of the cryptomonad cells. The fate of the cysts is not known. TheCryptomonas population is destroyed by the infection in the course of a few days. Literature studies have shown that the parasite has been known for a long time, but considered incorrectly by different authors as part of theCryptomonas, or as a result of phagotrophic uptake of theCryptomonas. The parasite is compared with a somewhat similar parasite inMallomonas, and with certain parasitic dinoflagellates. The similarity with the latter is superficial only as it posesses a eucaryotic nucleus. The parasite shows some similarity with the genusParadinium (Mycetozoa) as well as with certainSporozoa.

Pythium spiculum, a recently described new taxon, has been frequently isolated from declining Quercus rotundifolia and Q. suber roots and rhizosphere since 2003 in southern Iberia. In soils of declining Quercus forests this species was found as frequently as Phytophthora cinnamomi which, until now, was the only oomycete described as a Quercus root rot pathogen in the region. Inoculation tests conducted on young Q. rotundifolia plants showed that Py. spiculum is an aggressive root pathogen, although producing severities of symptoms significantly lower than those of P. cinnamomi. This new pathogen could play a role as decline factor in southern Iberia. Another new species, Py. sterilum, was also found to be pathogenic to Quercus roots but there are presently only few records of this organism isolated from rhizosphere of declining oaks in central Spain. More than an active decline factor, this species should be considered as a potential risk for Quercus forests.  相似文献

The initial attempts at hyper-expressing buffalo/goat growth hormone (GH)-ORFs inEscherichia coli directly under various strong promoters were not successful despite the presence of a functional gene. High level expression of GH was achieved as a fusion protein with glutathione-S-transferase (GST). To produce native GH in an unfused state, we adapted an established strategy of two-cistronic approach in our system. In this strategy, utilizing one of the highly efficient reported sequences as the first cistron led to a nearly 1000-fold enhancement in the level of expression under anE. coli promoter (trc). In search of a newer first-cistron sequence as well as to see the generality of the two-cistronic approach, we explored the ability of different lengths of a highly expressing natural gene to act as an efficient first cistron. Surprisingly,GST, which is naturally highly expressible inE. coli, could not be fitted into a successful two-cistronic construct. In addition, placement of the entire two-cistronic expression cassette (which had earlier given high-level GH expression undertrc promoter) under theT7 promoter inE. coli failed to hyper-express GH. These results suggest that the successful exploitation of the two-cistron arrangement for hyper-expression of eukaryotic ORFs in bacteria is not as straightforward as was previously thought. It appears probable that factors such as the sequence context, together with the length and codons used in the first cistron are important as well.  相似文献

We have isolated and analysed a 2 kb region of the mitochondrial genome of Arabidopsis thaliana (Columbia) showing a high level of nucleotide identity with the mitochondrial (mt) rps14 small-subunit ribosomal protein gene from Oenothera berteriana and Vicia faba, as well as with an open reading frame (ORF) located upstream of the nad3 locus in O. berteriana. The rps14 locus is present as a single copy in the A. thaliana mt genome and has a translational stop codon located near the initiation codon, as well as a deletion of one nucleotide that disturbs the coding sequence. The cloning and sequencing of nine amplified mt rps14 cDNAs clearly demonstrated that this gene is transcribed and that the mRNA precursors are edited at three positions, all involving C-to-U conversions. No editing events changing the stop codon and restoring the correct coding sequence were witnessed within the 9 individual cDNA clones. Therefore, we conclude that the single rps14 sequence of the mitochondrial genome from A. thealiana is in fact a pseudogene that is transcribed and edited but not translated.  相似文献

In central Italy, Prunus cerasus var. austera is cultivated as small stands or scattered trees in marginal areas for the production of jam and wine. Thanks to the healthy attributes of its products and its ability to grow in different environmental conditions, this variety has gained new interest in the development of marginal areas. We assessed the level of the genetic variability of P. cerasus var. austera germplasm from central Italy and identified a ‘core collection’ representative of the present genetic diversity. A total of 161 trees, morphologically identified as var. austera, and one tree, identified as var. caproniana were collected and genotyped by 14 SSRs. Two individuals provided by a commercial plant nursery, one of P. cerasus var. caproniana and one of P. cerasus var. austera, were used as control. Thirteen SSRs presented private alleles in austera. Seven individuals morphologically identified as austera revealed private alleles specific to caproniana. The PCoA and Bayesian clustering analysis showed a main genetic group including var. austera, while a second group included all the caproniana-like genotypes. A core collection of 31 trees (46% of austera genotypes) was selected. This study can be considered as a starting point for future investigations on this variety.  相似文献

Rhizobium tropici CIAT899 is a broad-host-range strain that, in addition to Phaseolus, nodulates other plant legumes such as Leucaena and Macroptilium. The narrow-host-range of Rhizobium leguminosarum biovars phaseoli (strain CE3) and trifolii (strain RS1051) can be extended to Leucaena esculents and Phaseolus vulgaris plants, respectively, by the introduction of a DNA fragment 521 bp long, which carries 128 amino acids of the amino-terminal region of a nodD gene from R. tropici, as well as a putative nod-box-like sequence, divergently oriented. The 521 bp fragment, in the presence of L. esculenta or P. vulgaris root exudates, induced a R. leguminosarum bv. viciae nodA-lacZ fusion in either a CE3 or RS1051 background, respectively.  相似文献

Epbl41l4a (erythrocyte protein band 4.1-like 4a, also named Nbl4) is a member of the band 4.1/Nbl4 (novel band 4.1-like protein 4) group of the FERM (4.1, ezrin, radixin, moesin) protein superfamily. Proteins encoded by this gene family are involved in many cellular processes such as organization of epithelial cells and signal transduction. On a molecular level, band 4.1/Nbl4 proteins have been shown to link membrane-associated proteins and lipids to the actin cytoskeleton. Epbl41l4a has also recently been identified as a target gene of the Wnt/β-catenin pathway. Here, we describe for the first time the spatio-temporal expression of epbl41l4a using Xenopus laevis as a model system. We observed a strong and specific expression of epb41l4a in the developing somites, in particular during segmentation as well as in the nasal and cranial placodes, pronephros, and neural tube. Thus, epbl41l4a is expressed in tissues undergoing morphogenetic movements, suggesting a functional role of epbl41l4a during these processes.  相似文献

Brown algae of the order Laminariales, commonly referred to as kelps, are the largest and most productive primary producers in the coastal inshore environment. The genus Ecklonia (Lessoniaceae, Phaeophyceae) consists of seven species with four species in the Northern Hemisphere and three in the Southern Hemisphere. It was recently transferred to the family Lessoniaceae based on phylogenetic analyses of nuclear and chloroplastic markers, though the type of the genus was not included and its relationship with allied genera Eckloniopsis and Eisenia remained unresolved. The present study is the first to produce a phylogeny focussed on the genus Ecklonia. It included sequences from nuclear, mitochondrial, and chloroplastic DNA, for most of the distribution range of the three current Southern Hemisphere species (Ecklonia radiata, Ecklonia maxima, and a sample of a putative Ecklonia brevipes specimen), sequences for East Asiatic species (Ecklonia cava, Ecklonia kurome, and Ecklonia stolonifera), as well as the closely related genera Eckloniopsis and Eisenia. Results confirmed E. radiata and E. maxima as two distinct species in South Africa, E. radiata as a single species throughout the Southern Hemisphere (in South Africa, Australia, and New Zealand) and East Asiatic species as a distinct lineage from the Southern Hemisphere clade. Results further pointed out a close sister relationship between Eckloniopsis radicosa and two Eisenia species (including the type species: Eisenia arborea) to the genus Ecklonia suggesting that the genera Eckloniopsis and Eisenia are superfluous.  相似文献

The plasmid pNR333 is a kanamycin-resistant, deletion derivative of pNR113 with an extremely high copy number in Escherichia coli and in Proteus mirabilis. In order to determine the usefulness of pNR333 as a replication gene of vector, the genes encoding chloramphenicol acetyltransferase (CAT) and β-galactosidase (β-gal) were cloned individually into both pNR333 and other low-copy-number plasmids. The expression of the cloned genes was compared by measuring the specific activity of each enzyme and the amounts of the proteins produced. A hybrid plasmid pNR333-cat expressed 53 times as much activity of CAT as the low-copy plasmid S-a which had a copy number of four. The lacZ gene cloned in pNR333 produced 17 times as much β-gal as in the low-copy-number plasmid pNR1150. These results suggest that pNR333 is a useful vector plasmid for producing a large amount of polypeptides in E. coli hosts.  相似文献

Summary Insect herbivores that require young foliage for successful larval development are often restricted to a single generation during a year by the scarcity of suitable food over most of the growing season. The major specialist herbivore attacking shrubs in the genus Asimina in Florida, Eurytides marcellus, requires young foliage for successful larval development. Field manipulations were used to investigate the role of the young foliage produced by Asimina in response to defoliation by the late-season feeder Omphalocera munroei, a second specialist herbivore of Asimina in Florida, in maintaining Eurytides populations during the summer months when young foliage is otherwise scarce. Defoliation by Omphalocera proved to be the major inducer of young growth during the summer because Omphalocera defoliated Asimina shrubs so frequently and severely. When compared to young leaves produced in the absence of damage, the teaves produced by Asimina in response to defoliation were equally as suitable as food for Eurytides larvae and as acceptable as oviposition sites by Eurytides females. The availability of young foliage in an Asimina population was correlated with the size of the associated Eurytides population. The combination of regular, severe defoliation by Omphalocera and lack of a defensive response to damage by Asimina lead to a positive affect of Omphalocera on Eurytides population size, and may be central to other facilitative interactions between herbivores as well.  相似文献
