Long-term application of sewage sludge resulted in soil cadmium (Cd) and zinc (Zn) contamination in a pot experiment conducted to phytoextract Cd/Zn repeatedly using Sedum plumbizincicola and Apium graceolens in monoculture or intercropping mode eight times. Shoot yields and soil physicochemical properties changed markedly with increasing number of remediation crops when the two plant species were intercropped compared with the unplanted control soil and the two monoculture treatments. Changes in soil microbial indices such as average well colour development, soil enzyme activity and soil microbial counts were also significantly affected by the growth of the remediation plants, especially intercropping with S. plumbizincicola and A. graveolens. The higher yields and amounts of Cd taken up indicated that intercropping of the hyperaccumulator and the vegetable species may be suitable for simultaneous agricultural production and soil remediation, with larger crop yields and higher phytoremediation efficiencies than under monoculture conditions.  相似文献

Knowledge of soil heavy metal concentration is very important for assessing the purity and quality of the soil in an environment. The concentrations of nine heavy metals (NHM), Zn, Pb, Cr, Cu, Co, Ni, Cd, Hg, and As, from the near-surface soils (~ 0–15 cm) from an industrial cluster in Kumasi, Ghana, were qualitatively and quantitatively measured and analyzed using X-ray fluorescence (XRF) spectroscopy analysis. The sources of these NHM were mainly anthropogenic as a result of the indiscriminate industrial waste disposal. In all, a total of about 100 soil samples were taken from six sampling sites, four of which were industrial and the remaining two residential. Forty soil samples out of the total number were carefully selected for elemental analyses and the mean heavy metal concentrations were calculated using statistical methods. The results from locations of high industrial impact showed that the mean concentrations of the NHM present in the soil were in the order of Zn (189.2?908.6 mgkg?1), Pb (133.7?571.3 mgkg?1), Cr (91.3?545.8 mgkg?1), Cu (62.9?334.6 mgkg?1), Co (38.6?81.9 mgkg?1), Ni (12.4?30.9 mgkg?1), Cd (6.9?13.2 mgkg?1), Hg (5.5?10.4 mg kg?1), and As (2.3?18.6 mgkg?1). Apart from Ni and As, all the heavy metals recorded concentrations that ranged from 10?900% higher than their respective threshold limit values (TLVs). Heavy metal concentrations from the residential sites were comparatively far lower with only Cr, Cd, and Hg registering concentrations between 65?250% above their TLVs. The cluster with its residential communities is at a serious risk of soil heavy metal toxicity and awareness to this needs to be created as such.  相似文献

In this study we explored electrophoretic profiles, enzymatic activities and immunoreactivity of neonate and adult venoms from two snakes (Naja atra and Deinagkistrodon acutus) coexisting in southeastern China. Age-related variation in electrophoretic profiles was found in both species and proteolytic and fibrinogenolytic activity was higher in neonate than adult venoms. Neonate D. acutus venom had higher 5' nucleotidase, PLA2, hyaluronidase and gelatinolytie activity, but lower esterolytic activity, than adult venom. Neonate and adult D. acutus venoms showed identical phosphomonoesterase, LAO and fibrinolytic activities. Neonate N. atra venom had higher phosphomonoesterase and LAO activity, but lower 5' nucleotidase, PLA2, hyaluronidase and Ache activities than adult venom. Neonate and adult N. atra venoms showed similar gelatinolytic activity. Further, age-dependent immunoreactivity was found in both species, and cross-reactions between homologous venoms and antiserums were closely related to venom composition. We speculate that age-related variation in venom characteristics is possibly driven by evolutionary forces associated with ontogenetic shifts in dietary habits, competition and predation pressure.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are of global concern due to their ubiquitous presence, toxicity, and carcinogenicity. No data on PAHs in soils from South Africa have been published, even though it has the largest economy and industrial base in Africa. During this initial assessment, the levels of PAHs were determined in soils and sediments collected from central South Africa, specifically targeting industrial, residential, and agricultural areas. Analysis was performed by gas chromatography/mass spectrometry (GC/MS). The total concentration of PAHs (Σt-PAH) ranged between 44 and 39,000 ng/g, dw and the concentration of carcinogenic PAHs (Σc-PAH) ranged between 19 and 19,000 ng/g, dw. Pyrogenic processes were the most likely sources, with minimal petrogenic contributions. PAH levels were in the same range as levels reported from other countries, and the majority of the sites did not exceed Canadian environmental quality guidelines. Based on assumptions for dermal contact and ingestion of PAH-contaminated soil, we provisionally calculated only a small increase in cancer risk, but additional PAH inhalation could add considerably to this risk. Our data indicates a need for more analysis in industrial and residential areas, and should include air.  相似文献

The study of a decapod community in a Cymodocea nodosa meadow from Southeastern Spain (Western Mediterranean Sea) showed a stable structure, in which the families Hippolytidae, Processidae, Majidae and Portunidae were the most abundant and the species Hippolyte niezabitowskii dominated. The animal community was more numerous and diverse during the night, showing the existence of nychthemeral movements, which are essentially related to the trophic behaviour and shelter. In this way, many species increased their abundance as a result of an increasing activity and, also, of an influx of other species and specimens from adjacent sandy bottoms, such as Processa spp. (mainly P. modica) Sicyonia carinata, Liocarcinus spp. (mainly juveniles) and several species of hermit crabs, which were rare or absent during the day. All these changes produced modifications in the dominance curves and in the values of all ecological indices (richness, diversity and evenness). Monthly samples were grouped and ordered (MDS) by the factor “day–night”, which showed slight qualitative and quantitative differences (SIMPER, dissimilarity average of the factor day–night = 61.67). On the other hand, no global seasonal differences have been found (one way ANOSIM), but there was a significant level of similarity between winter and spring, while the summer samples were the most different. The differentiation of the summer 1999 can be attributed to a decrease in species abundance and richness, probably due to the dynamics of the decapod populations and the balance with predators (fishes), while that of the summer 2000, to an anomalous event: the massive proliferation of filamentous algae, mainly Ectocarpus s.l., which modified the environmental conditions.  相似文献

Four dipeptide complexes of the type [PtX(2)(dipeptide)] x H(2)O (X=Cl, I, dipeptide=l-methionylglycine, l-methionyl-l-leucine) were prepared. The complexes were characterized by (1)H, (13)C, (195)Pt NMR and infrared spectroscopy, DTG and elemental analysis. From the infrared, (1)H and (13)C NMR spectroscopy it was concluded that dipeptides coordinate bidentately via sulfur and amine nitrogen donor atoms. Confirmed with (13)C and (195)Pt NMR spectroscopy, each of the complexes exists in two diastereoisomeric forms, which are related by inversion of configuration at the sulfur atom. The (1)H NMR spectrum for the platinum(II) complex with l-methionylglycine and chloro ligands exhibited reversible, intramolecular inversion of configuration at the S atom; DeltaG( not equal)=72 kJ mol(-1) at coalescence temperature 349 K was calculated. In vitro cytotoxicity studies using the human tumor cell lines liposarcoma, lung carcinoma A549 and melanoma 518A2 revealed considerable activity of the platinum(II) complex with l-methionylglycine and chloro ligands. Further in vitro cytotoxic evaluation using human testicular germ cell tumor cell lines 1411HP and H12.1 and colon carcinoma cell line DLD-1 showed moderate cytotoxic activity for all platinum(II) complexes only in the cisplatin-sensitive cell line H12.1. Platinum uptake studies using atomic absorption spectroscopy indicated no relationship between uptake and activity. Potential antitumoral activity of this class of platinum(II) complexes is dependent on the kind of ligands as well as on tumor cell type.  相似文献

Abstract: Addition of endothelins (ETs) to neuroblastomaglioma hybrid cells (NG108-15) induced increases in cytosolic free Ca2+ ([Ca2+]i) levels of labeled inositol monophosphates and inositol 1,4,5-trisphosphate [Ins(1,4,5)P3]. The increases in [Ca2+]i elicited by the three ETs (ET-1, ET-2, and ET-3) were transient and did not show a sustained phase. Chelating extracellular Ca2+ in the medium by adding excess EGTA decreased the ET-mediated Ca2+ response by 40-50%. This result indicates that a substantial portion of the increase in [Ca2+]i was due to influx from an extracellular source. However, the increase in [Ca2+]i was not affected by verapamil or nifedipine (10?5M). A rank order potency of ET-1 ET-2 ET-3 is shown for the stimulated increase in [Ca2+]i, as well as labeled inositol phosphates, in these cells. ATP (10?4M) and bradykinin (10?7M) also induced the increases in [Ca2+]i and Ins(1,4,5)P3 in NG108-15 cells, albeit to a different extent. When compared at 10?7M, bradykinin elicited a five- to sixfold higher increase in the level of Ins(1,4,5)P3, but less than a twofold higher increase in [Ca2+]i than those induced by ET-1. Additive increases in both Ins(1,4,5)P3 and [Ca2+]i were observed when ET-1, ATP, and bradykinin were added to the cells in different combinations, suggesting that each receptor agonist is responsible for the hydrolysis of a pool of polyphosphoinositide within the membrane. ET-1 exhibited homologous desensitization of the Ca2+ response, but partial heterologous desensitization to the Ca2+ response elicited by ATP. On the contrary, ET-1 did not desensitize the response elicited by bradykinin, although bradykinin exhibited complete heterologous desensitization to the response elicited by ET-1. Taken together, these results illustrate that, in NG108-15 cells, a considerable amount of receptor cross talk occurs between ET and other receptors that transmit signals through the polyphosphoinositide pathway.  相似文献

Greenhouse gas emissions from small and medium enterprises (SMEs) account for 70% of the industrial pollution in the European Union. Owing to limited economic and human resources, only a few SMEs start procedures to evaluate the environmental impact of processes and products through life cycle assessment (LCA). In this work, a cluster life cycle assessment (Cluster‐LCA) is proposed as an instrument for the diffusion and realization of LCA analysis in clustered SMEs. This methodology is illustrated with a case study in the tanning cluster in Tuscany. The different characteristics of the methodology are analyzed by identifying the intrinsic strengths, weaknesses, opportunities, and threats. The application of this methodology in a particular cluster is then discussed in order to gather some helpful insight for the application of this methodology in different clusters.  相似文献

The S100 protein family is the largest subgroup within the superfamily of proteins carrying the Ca2+-binding EF-hand motif. Despite their small molecular size and their conserved functional domain of two distinct EF-hands, S100 proteins developed a plethora of tissue-specific intra- and extracellular functions. Accordingly, various diseases such as cardiomyopathies, neurodegenerative and inflammatory disorders, and cancer are associated with altered S100 protein levels. Here, we review the different S100 protein functions and related diseases from an evolutionary point of view. We analyzed the structural variations, which are the basis of functional diversification, as well as the genomic organization of the S100 family in human and compared it with the S100 repertoires in mouse and rat. S100 genes and proteins are highly conserved between the different mammalian species. Moreover, we identified evolutionary related subgroups of S100 proteins within the three species, which share functional similarity and form subclusters on the genomic level. The available S100-specific mouse models are summarized and the consequences of our results are discussed with regard to the use of genetically engineered mice as human disease models. An update of the S100 nomenclature is included, because some of the recently identified S100 genes and pseudogenes had to be renamed.  相似文献

Disseminated neoplasia (DN) was one of the most important pathological conditions found in cultured flat oysters (Ostrea edulis) from different geographical origins grow in Galicia (NW Spain), during a two years selective breeding programme to produce oysters less susceptible to bonamiosis. Histological characteristics observed in oysters affected by DN included intense infiltration of connective tissue of various organs (gills, stomach, digestive gland and gonad) by large undifferentiated cells, with a large nucleus and a high nucleus-to-cytoplasm ratio. The main ultrastructural features were predominance of euchromatin over heterochromatin that was arrayed in small clumps in the nucleus, prominent granular nucleolus, swollen mitochondria with few cristae and high number of free ribosomes in the cytoplasm. A seasonal pattern of DN prevalence was detected, with higher values in spring-summer, but there were no significant differences between geographic origins or families within these origins. However, the intensity of the disease was significantly different between origins; oysters originating outside of Galicia (particularly those originating from Ireland) were more susceptible to develop advanced DN. DN (8%) and bonamiosis (4.9%) were found concurrently in oysters. The nature and significance of this association warrants more investigation to determine its importance, if any.  相似文献

Diverse plant remains recovered from an archaeological site of Chalcolithic‐Early Historic age in the Bhairabdanga area of Pakhanna (latitude 23°25′N, longitude 87°23′E), situated on the west bank of the Damodar river, Bankura district, West Bengal, India, include food grains, wood charcoals, and palynomorphs. Radiocarbon dating of the recovered biological remains reveal the age of the site as (3 320 ± 400) to (2 080 ± 80) yr BP. The food grains were identified as Oryza sativa L. and Vigna mungo L, and seeds of Brassica cf. campestris L. were also found; these indicate the agricultural practice and food habits of the ancient people living at Pakhanna from the Chalcolithic to the Early Historic period. Sediments including plant remains have been broadly divided into two zones, considering archaeological findings and radiocarbon dating. Analysis of the plant remains (i.e. wood charcoals and palynomorphs) in addition to cultivated food grains has revealed that a rich vegetation cover existed in this area, with a prevailing tropical and humid climate, comprising the timber‐yielding plants Shorea sp., Terminalia sp., and Tamarindus sp., with undergrowths of diverse shrubs and herbs during the Chalcolithic period (zone I) dated (3 320 ± 400) yr BP. Comparatively poorer representation and frequency of plant remains indicate a drier climate during the Early Historic period (zone II) dated as (2 110 ± 340) to (2 080 ± 80) yr BP. Comparisons of the archaeobotanical data recovered from the Chalcolithic and Early Historic period and also a principle components analysis indicate a change in the climate of the area from tropical and humid at (3 320 ± 400) yr BP to tropical and drier conditions at (2 110 ± 340) to (2 080 ± 80) yr BP. The present‐day tropical, dry deciduous vegetation of the area suggests that climate change has occurred in the area since the contemporaneous past. The plant remains database has been utilized to reconstruct the settlement pattern of the community living in the site between (3 320 ± 400) and (2 080 ± 80) yr BP. The hypothesis was tested that isolated populations of Argia vivida Hagen living in habitats with different thermal regimes would show similar larval temperature preferenda, similar distributions in a temperature gradient, similar larval upper temperature acitivity thresholds, and similar adult minimum temperature flight thresholds. The hypothesis was supported in all cases, except for distribution within the gradient, where there were significantly fewer observations below 22°C in a population from a habitat with a fluctuating diel and annual temperature regime than in a population from a more thermally stable habitat. Larval modal temperature preferendum was 28°C; escape temperature (EST) was 35.4–36.4°C, critical thermal maximum (CTM) was 39.1–41.0°C, and upper lethal temperature (ULT) was 44.4–46.0°C. While technical difficulties affected the estimates of flight thresholds, there was no difference between the field estimates from different sites. Minimum body temperature for flight appears to be about 25°C, apparently higher than for several other zygopterans, while larval activity thresholds and 96 hr LD50 of 36.8°C are similar to those recorded for other odonates.  相似文献

The new macrocyclic complex 2-amino-2-deoxy-N-[2-(1,3,5,8,11-pentaazacyclotridecan-3-yl)ethyl]-beta-D-glucopyranosylamine copper(II) dichloride (1a) was prepared and thoroughly characterized by various techniques. Molar-conductance measurements showed that 1a (and its Ni analogue 1b) are ionic in nature. On the basis of spectroscopic data, both complexes were assigned a square-planar geometry, and found to be highly stabile and hydrolytically robust in H2O over a wide range of pH, as confirmed by cyclic voltammetry (CV). The Cu(II) complex 1a was found to bind to CT-DNA, with a binding constant Kb of 2.4x10(3) M(-1), as derived by UV/VIS titration, and confirmed by CV, circular dichroism (CD), and viscosity measurements. DNA binding seems to occur mostly via H-bonding. In an in vitro antitumor MTT assay, 1a exhibited significant anticancer activity against the SY5Y and PC-12 cell lines, with an estimated IC50 value in the micromolar range for SYSY, similar to the standard drug 5-fluorouracil.  相似文献

The effect of the four triadimenol stereoisomers on the purified yeast lanosterol 14 alpha-demethylase (cytochrome P-45014DM), the primary target of azole antifungal agents, was studied. (1S,2R)-Triadimenol was the most potent demethylase inhibitor and bound quantitatively to the enzyme below 0.05 microM. This isomer also interfered with the chemical reduction of cytochrome P-45014DM and the binding of CO to the cytochrome. The other isomers showed a lower inhibitory effect on the enzyme, and the order of activity was (1R,2R) greater than (1R,2S) greater than or equal to (1S,2S). Based on these findings and the reported preferred conformations for the triadimenol stereoisomers (Anderson, N.H. et al., Pestic. Sci. 15:310-316, 1984), it is predicted that orientation of the hydrophobic tert-butyl and p-chlorophenyl groups relative to the azole nitrogen is important to fit the antifungal agent in the active site of the demethylase.  相似文献

Structures, chemical properties, and in vitro insulinomimetic activities of new vanadyl [oxovanadium(IV), VO(2+)] complexes with five tripodal ligands containing an imidazole functionality were examined. The ligands, N-(carboxymethyl)- N-(4-imidazolylmethyl)amino acids, contain glycine, ( S)- and ( R)-alanine, and ( S)- and ( R)-leucine residues. The molecular structures of the latter four alanine- and leucine-containing complexes were determined by X-ray analysis. The coordination geometry around each vanadium center was octahedral, where an imino nitrogen occupied the apical site and two carboxylate oxygens, an imidazole nitrogen, and a water molecule coordinated in the equatorial plane. The spectroscopic properties of the complexes were characterized by means of IR, electronic absorption, and CD spectra. Acid dissociation constants (p K(a)) and protonation sites of the ligands were determined by a combination of potentiometric titrations and (1)H NMR spectra. The potentiometric study demonstrated that stability constants (log beta) were not so different among the present complexes (14.0-14.9) and a species of molecular complex with a 1:1 metal:ligand ratio existed predominantly at physiological pH 7.4. EPR parameters indicated that the species at pH 7.4 had an octahedral structure similar to the complex in the solid state. On the other hand, an EPR study in phosphate buffer (pH 7.4) suggested that inorganic phosphate coordinated to the vanadium center instead of the imidazole group in the presence of excess phosphate ion. Cyclic voltammograms in the phosphate buffer showed chemically reversible oxidation waves, whereas irreversible oxidation waves were observed in non-coordinating HEPES buffer. Moreover, the oxidation potential of each complex in phosphate buffer was more positive than that in HEPES buffer. Partition coefficients of the present complexes in a n-octanol/saline system were very low, probably due to hydrophilicity of the imidazole group. The in vitro insulinomimetic activities were estimated on the basis of the ability of the complexes to inhibit epinephrine-stimulated free fatty acid release from isolated rat adipocytes. The achiral glycine-derivative complex exhibited the highest insulinomimetic activity, which was higher than that of VOSO(4) as a positive control. Putting our previous observations together, it was found that the vanadyl complexes with tetradentate amino acid derivatives having no alkyl side chain tend to have high in vitro insulinomimetic activity.  相似文献

A new family of tridentate ligands PhimpH (2-((2-phenyl-2-(pyridin-2-yl)hydazono)methyl)phenol), N-PhimpH (2-((2-phenyl-2-(pyridin-2-yl)hydrazono)methyl)napthalen-1-ol), Me-PhimpH (2-(1-(2-phenyl-2-(pyridine-2-yl)hydrazono)ethyl)phenol) have been synthesized and characterized. The ligands PhimpH and N-PhimpH after deprotonation react with manganese(II) and manganese(III) starting materials affording [Mn(Phimp)2] (1), [Mn(Phimp)2](ClO4) (2), [Mn(N-Phimp)2] (3), [Mn(N-Phimp)2](ClO4) (4). Complexes [Mn(Phimp)2] (1) and [Mn(N-Phimp)2] (3) convert to [Mn(Phimp)2]+ (cation of 2) and [Mn(N-Phimp)2]+ (cation of 4) respectively upon oxidation. Ligand Me-PhimpH stabilized only manganese(III) centre resulting [Mn(Me-Phimp)2](ClO4) (5). The molecular structures of [Mn(Phimp)2], 1 and [Mn(Phimp)2](ClO4), 2 were determined by single crystal X-ray diffraction. X-ray crystal structures of 1 and 2 have revealed the presence of distorted octahedral MnN4O2 coordination sphere having meridionally spanning ligands. Electrochemical studies for the complexes showed Mn(II)/Mn(III), (E1/2 = 0.14-0.40 V) and Mn(III)/Mn(IV), (E1/2 = 0.80-1.06 V) couples vs. Ag/AgCl. The redox properties were exploited to examine superoxide dismutase (SOD) activity using Mn(II)/Mn(III) couple. The complexes 1, 2, 4 and 5 have been revealed to catalyze effectively the dismutation of superoxide () in xanthine-xanthine oxidase-nitro blue tetrazolium assay and IC50 values were found to be 0.29, 0.39, 1.12 and 0.76 μM respectively. DNA interaction studies with complex 2 showed binding of DNA in a non-intercalative pathway. Complexes 1, 2 and 4 exhibited nuclease activity in presence of H2O2 and inhibition of activity was noted in presence of KI.  相似文献

Abstract  The braconid parasitoid Microctonus aethiopoides Loan has been released in Australia and New Zealand for biological control of the lucerne pest Sitona discoideus Gyllenhal. In New Zealand, the parasitoid attacks a number of endemic weevil species. A survey of Curculionoidea found in and near lucerne in south-eastern Australia was carried out to investigate whether similar non-target parasitism was occurring, and to relate this to levels of parasitism found in the target host, S. discoideus . Some of the original M. aethiopoides release sites were particularly targeted in the survey of 25 sites in Victoria, New South Wales and South Australia. Almost 2500 weevils were collected, of which over 90% were S. discoideus , with the remaining 197 other weevils comprising 29 species found at 15 of the 25 sites. Parasitism of S. discoideus by M. aethiopoides occurred at 12 lucerne sites, with levels ranging from 0 to 25%. A single incidence of parasitism of a species of an Australian native weevil Prosayleus sp. by M. aethiopoides was recorded. No parasitism of any other weevil species was observed. The taxonomic affinities between Sitona and native Australian and New Zealand weevils are discussed, concluding that non-target host range in M. aethiopoides may be determined more by ecological factors than by taxonomic affinities among its hosts.  相似文献
