Membrane proteins present a hydrophobic surface to the surrounding lipid, whereas portions protruding into the aqueous milieu expose a polar surface. But how have proteins evolved to deal with the complex environment at the membrane-water interface? Some insights have been provided by high-resolution structures of membrane proteins, and recent studies of the role of individual amino acids in mediating protein-lipid contacts have shed further light on this issue. It now appears clear that the polar-aromatic residues Trp and Tyr have a specific affinity for a region near the lipid carbonyls, whereas positively charged residues extend into the lipid phosphate region.  相似文献

Recent attention has focused on the remarkable ability of adult stem cells to produce differentiated cells from embryologically unrelated tissues. This phenomenon is an example of metaplasia and shows that embryological commitments can be reversed or erased under certain circumstances. In some cases, even fully differentiated cells can change their phenotype (transdifferentiation). This review examines recently discovered cases of metaplasia, and speculates on the potential molecular and cellular mechanisms that underlie the switches, and their significance to developmental biology and medicine.  相似文献

Enzymes that are adapted to widely different temperature niches are being used to investigate the molecular basis of protein stability and enzyme function. However, natural evolution is complex: random noise, historical accidents and ignorance of the selection pressures at work during adaptation all cloud comparative studies. Here, we review how adaptation in the laboratory by directed evolution can complement studies of natural enzymes in the effort to understand stability and function. Laboratory evolution experiments can attempt to mimic natural evolution and identify different adaptive mechanisms. However, laboratory evolution might make its biggest contribution in explorations of nonnatural functions, by allowing us to distinguish the properties nutured by evolution from those dictated by the laws of physical chemistry.  相似文献

应对气候变化和保护生物多样性是2大全球性热点环境问题。气候变化导致物种多样性丧失、生态系统服务降低和区域生态安全屏障功能受损,威胁到中国国土生态安全格局和生态脆弱区域的可持续发展,给生物多样性保护带来新的挑战。做好生物多样性保护适应气候变化的风险管理工作,既是生物多样性应对气候变化风险的必要措施,也是减缓气候变化的重要途径。结合爱知目标10的实现情况,分析了欧盟、澳大利亚、美国等发达国家发布的生物多样性适应气候变化技术政策制定情况、中国生物多样性应对气候变化进展情况,剖析了中国生物多样性保护适应气候变化存在的问题,包括生物多样性应对气候变化的科学认知亟待提高、生物多样性保护适应气候变化的能力建设不足、自然保护地之间缺乏适应气候变化的生态廊道网络、生物多样性保护适应气候变化的技术标准缺乏。研究提出了中国生物多样性应对气候变化的适应性管理策略,包括制定《中国生物多样性保护协同应对气候变化的国家方案》、加强生物多样性保护适应气候变化的能力建设、开展自然保护区适应气候变化的风险管理试点、强化生物多样性应对气候变化的科技支撑,以期为推进纳入气候变化风险管理的生物多样性保护工作提供决策依据。  相似文献

Oxygen plays an important role in diverse biological processes. However, since quantitation of the partial pressure of cellular oxygen in vivo is challenging, the extent of oxygen perturbation in situ and its cellular response remains underexplored. Using two‐photon phosphorescence lifetime imaging microscopy, we determine the physiological range of oxygen tension in osteoclasts of live mice. We find that oxygen tension ranges from 17.4 to 36.4 mmHg, under hypoxic and normoxic conditions, respectively. Physiological normoxia thus corresponds to 5% and hypoxia to 2% oxygen in osteoclasts. Hypoxia in this range severely limits osteoclastogenesis, independent of energy metabolism and hypoxia‐inducible factor activity. We observe that hypoxia decreases ten‐eleven translocation (TET) activity. Tet2/3 cooperatively induces Prdm1 expression via oxygen‐dependent DNA demethylation, which in turn activates NFATc1 required for osteoclastogenesis. Taken together, our results reveal that TET enzymes, acting as functional oxygen sensors, regulate osteoclastogenesis within the physiological range of oxygen tension, thus opening new avenues for research on in vivo response to oxygen perturbation.  相似文献

The building stock consumes large amounts of resources for maintenance and expansion which is only exacerbated by disaster events where large‐scale reconstruction must occur quickly. Recent research has shown the potential for application of material stock (MS) accounts for informing disaster risk planning. In this research, we present a methodological approach to analyze the vulnerability of the material stock in buildings to extreme weather events and sea‐level rise (SLR) due to climate change. The main island of Grenada, a Small Island Developing State (SIDS) in the Caribbean region, was used as a case study. A bottom‐up approach based on a geographic information system (GIS) is used to calculate the total MS of aggregate, timber, concrete, and steel in buildings. The total MS in buildings in 2014 was calculated to be 11.9 million tonnes (Mt), which is equivalent to 112 tonnes per capita. Material gross addition to stock (GAS) between 1993 to 2009 was 6.8 Mt and the average value over the time period was 4.0 tonnes per capita per year. In the year following Hurricane Ivan (2004), the per capita GAS for timber increased by 172%, while for other metals, GAS spiked by 103% (compared to average growth rates of 11% and 8%, respectively, between 1993 and 2009). We also ran a future “Ivan‐II” scenario and estimated a hypothetical loss of between 135 and 216 kilotonnes (kt) of timber from the building stock. The potential impact of SLR is also assessed, with an estimated 1.6 Mt of building material stock exposed under a 2‐m scenario. We argue that spatial material stock accounts have an important application in planning for resilience and provide indication of the link between natural disaster recovery and resource use patterns.  相似文献

One of the most unusual features of RNA viruses is their enormous genetic variability. Among the different processes contributing to the continuous generation of new viral variants RNA recombination is of special importance. This process has been observed for human, animal, plant and bacterial viruses. The collected data reveal a great susceptibility of RNA viruses to recombination. They also indicate that genetic RNA recombination (especially the nonhomologous one) is a major factor responsible for the emergence of new viral strains or species. Although the formation and accumulation of viral recombinants was observed in numerous RNA viruses, the molecular basis of this phenomenon was studied in only a few viral species. Among them, brome mosaic virus (BMV), a model (+)RNA virus offers the best opportunities to investigate various aspects of genetic RNA recombination in vivo. Unlike any other, the BMV-based system enables homologous and nonhomologous recombination studies at both the protein and RNA levels. As a consequence, BMV is the virus for which the structural requirements for genetic RNA recombination have been most precisely established. Nevertheless, the previously proposed model of genetic recombination in BMV still had one weakness: it could not really explain the role of RNA structure in nonhomologous recombination. Recent discoveries concerning the latter problem give us a chance to fill this gap. That is why in this review we present and thoroughly discuss all results concerning nonhomologous recombination in BMV that have been obtained until now.  相似文献
