Larvae of the sawfly Arge (Hymenoptera, Argidae) are exposed to predators such as ants. Their defence mechanisms, which have been almost unstudied, were investigated by behavioural observations coupled to a morphological approach and by testing the bioactivity of several body parts. Arge larvae raised their abdomen when contacted by Myrmica rubra workers. The ants rarely bit a larva and generally retreated immediately, sometimes without contacting it. Most of those few ants that bit a larva then showed an uncoordinated walk. Crude hemolymph from a common species, A. pagana, was a feeding deterrent towards ants. Hemolymph extracts remained active up to a concentration of 0.8 microg DW extract per microlitre solution, and were more active than integument and gut extracts. We also observed ants paralysed by extracts, especially from the gut. It is likely that this toxicity is due to a polypeptide, lophyrotomin, which is known to occur in A. pullata. Six or seven non-eversible ventro-abdominal glands occurred in all species studied (A. fuscipes, A. nigripes, A. ochropus, A. pagana, A. pullata, A. ustulata). These glands contain volatiles. We consider both types of chemicals to be important in defence, and we propose that the paralysing effect is a common feature among Arge species.  相似文献

Insect midgut proteases are excellent targets for insecticidal agents such as protease inhibitors. These inhibitors are used for producing transgenic plants, resistant to pests. For achieving this goal, it is necessary to find the nature of specific proteases and their properties for adopting possible pest management procedure. Therefore, characterisation of the enzymes in the gut of the rose sawfly, Arge rosae (Hymenoptera: Argidae), responsible for proteolysis, was performed using a range of synthetic substrates and specific inhibitors. The optimum conditions for general proteases and trypsin were achieved at pH 10. The highest activity for general proteases was obtained at a temperature of 45°C. The use of specific inhibitors and SDS-PAGE (sodium dodecyl sulfate polyacrylamide gel electrophoresis) provided evidence to suggest that most of the proteases belonged to the serine group because of high inhibitory effect of phenyl methane sulfonyl fluoride on total proteolytic activity. Also, inhibition assays and zymogram analysis showed that metalloproteases are present in A. rosae digestive system. These results indicated that A. rosae larvae mainly used serine proteases for protein digestion, with chymotrypsin as the dominant form. The kinetic parameters of trypsin-like proteases using N-benzoyl-dl-arg-p-nitroanilide as substrate indicated that the K m and V max values of trypsin in the gut of the fifth instar larvae were 730 ± 17.3 μM and 456 ± 13.85 nmol min?1 mg?1 protein, respectively.  相似文献

The rose sawfly, Arge ochropus (Gmelin), is one of the most destructive pests attacking rose bushes in northern Iran. In the present study, the haemocyte reactions of A. ochropus larvae were assessed against two entomopathogenic nematodes, Steinernema carpocapsae (Weiser) and Heterorhabditis bacteriophora Poinar. With injection of infective juveniles of H. bactriophora into A. ochropus larvae, the total haemocyte count showed alternating fluctuations at all time intervals. Encapsulation occurred 22 hours post injection (hpi) of H. bacteriophora infective juveniles into the rose sawfly larvae, while melanization was observed after 24 h. In the case of S. carpocapsae, initial attachment of the haemocytes was detected at 18 hpi and complete encapsulation at 24 hpi; no melanization was observed. In general, findings showed strong immune responses of the rose sawfly larvae against H. bacteriophora, while these reactions were weakened for S. carpocapsae. Therefore, it is concluded that S. carpocapsae has more ability to overcome the host defence system, and this research provides a new window on its potential as a biocontrol agent.  相似文献

The sawfly, Arge rosae Linnaeus (Hymenoptera: Argidae) is a serious pest of rose plants in a vast area of the world including Iran. Pesticides are applied in order to control in areas where infestation is high. So, there is a need for more environmentally benign alternatives to control this insect pest in urban areas. Therefore, the aim of the current study was to study the effect of cereal seed proteinaceous extracts including wheat and triticale on the insect α-amylase. Wheat and triticale seed proteins as well as fourth instars larvae α-amylase were extracted. The results showed that there are two different α-amylase isoenzymes in the insect gut, one major band and the other minor band, which had optimal activity at alkaline pH (pH 8.0). The effect of five concentrations including 0.42, 0.21, 0.105, 0.05 and 0.025?mg?ml?1 of each seed extract was tested on α-amylase activity of the larvae gut. At the highest concentration (0.42?mg?ml?1), wheat seed extract caused 74% inhibition of the enzyme activity while triticale seed extract inhibited 62% enzyme activity. Experiments proved that seed proteinaceous extract affected the enzyme activity in a pH-dependant manner.  相似文献

In the seminal vesicle of the 'symphyta'Arge pagana the spermatozoa are stored in motile spermatodesm bundles, maintained by an anterior cap of extracellular material. This cap consists of a denser cortex and of an internal matrix, where part of the sperm heads are embedded. The number of spermatozoa per bundle is variable. The spermatozoa are short, only 30microm long, with a head region of about 23microm, and a very short flagellum of about 7microm. The head includes the acrosome, with a perforatorium, and the nucleus. The flagellum consists of an axoneme, with a 9+9+2 microtubule pattern, a centriolar adjunct, two mitochondrial derivatives and two accessory bodies. The mitochondrial derivatives are very slender and of different lengths. The longer begins at the base of the nucleus, while the shorter one starts just below the base of the centriolar adjunct. This latter is asymmetric and appears at the nuclear base, extending parallel to the axoneme up to the anterior end of the smaller mitochondrial derivative. The short spermatodesmata and the small mitochondrial derivatives characterize the A. pagana sperm. In addition, the centriolar adjunct asymmetry and the occurrence of spermatodesm bundles might be considered plesiomorphic states present in the basal Tenthredinoidea.  相似文献

The wheat stem sawfly,Cephus cinctusNorton (Hymenoptera: Cephidae) was first found in wild grasses and soon became an economically important pest of wheat after cultivation began in the northern Great Plains. Of the species of parasitoids that attackC. cinctusin wild grasses, onlyBracon cephi(Gahan) andB. lissogasterMuesebeck (Hymenoptera: Braconidae) have been found in wheat. Levels of parasitism vary between wheat-producing regions in Montana. Parasitism levels were increased by releasing individuals from a region with high levels of parasitism into a region where parasitism levels were low. Summary Relative abundance of natural food resource, leaves of wild rose, ofArge migrinodosa was evaluated by comparing the food quantity which the larvae composing a colony utilized throughout the larval stage with that which is available in the habitat. In this connection, behavioral pattern of larval feeding was also observed under natural conditions, and its ecological significance was discussed. It was quantitatively clarified that the larvae faced the severe shortage of food and had to disperse to other shoots with the expense of probably high risk. Under pressures of such insufficient supply of food, the larvae follow the most efficient pattern of feeding behavior that they move to the tip of shoot at first and feed regularly downward thereafter. Though individual larvae behaved completely independently of each other, the aggregation appeared to be formed and maintained because all the larvae from the same egg mass follow this fixed feeding pattern.  相似文献

Stemmata are peculiar visual organs of most larvae in holometabolous insects. In Hymenoptera, Symphyta larvae exclusively possess a pair of stemmata, whose cellular organizations have not been thoroughly elucidated to date. In this paper, the morphology and fine structure of stemmata were investigated in the large rose sawfly Arge pagana (Panzer, 1798) using light and electron microscopy. The larvae possess a pair of stemmata, which belong to the “unicorneal composite eye” or single-chamber stemmata. Each stemma is composed of a biconvex cornea lens, a layer of corneagenous cells, numerous pigment cells, and hundreds of retinula cells. According to the number of retinula cells forming a rhabdom, the stemma can be divided into two regions, the larger Region I and the smaller Region II. The former occupies the largest area of the stemma and contains the majority of rhabdoms, each of which is formed by the rhabdomeres of eight retinula cells. The latter occupies a narrow posterior margin, where each rhabdom consists of nine retinula cells. Based on the different cellular organizations of rhabdoms, the stemma of Argidae is likely developed by the fusion of two types of ommatidial units.  相似文献

Following the establishment of isofemale lines and subsequent inbreeding, the ichneumonid parasitoid wasp Diadegma chrysostictos (Gmelin) was shown by segregation of polymorphic alloenzyme loci to have single-locus complementary sex determination (sl-CSD). This and the biparental nature of diploid males was confirmed using two independent Mendelian recessive phenotypic markers. The existence of diploid males, sl-CSD, and the abrogation of diploid males following outbreeding was further confirmed by flow cytometry, a potentially general method that is independent of the maternal sex allocation or the need for genetic markers. Estimates of the number of sex alleles in several British populations demonstrated 17-19 alleles in Britain, with a decline toward the northerly limit of the parasitoid's range, varying from 16 in the south of England to 4-5 in central Scotland, in broad agreement with the rate of attainment of a male-biased sex ratio when used to establish en masse laboratory cultures. These data represent the second confirmation of the existence of sl-CSD in the Ichneumonidae (and the first in the Campopleginae subfamily), lending further support to the notion that sl-CSD was the ancestral condition in the Aculeata/Ichneumonoidea clade (Cook 1993a; Periquet et al. 1993).  相似文献

The egg deposition behavior of the turnip sawfly, Athalia rosae (Hymenoptera: Symphyta), is described. Both unmated and mated females lay eggs individually inside of fresh young leaves of cruciferous plants. During an oviposition event, females exhibit a distinct pause in abdominal contractions just before the actual egg deposition act. Unmated females show a longer pause (11.31 s on average) than mated females (4.38 s on overall average). By employing an eye color mutation, the sex of the eggs laid by females was ascertained. Females mated once lay mostly fertilized (diploid female) eggs initially but begin to lay a considerable number of unfertilized (haploid male) eggs later in life. The laying of an unfertilized egg is associated with a longer pause (6.98 s on average) than the laying of a fertilized egg (3.76 s on average). These results are in contrast to previous reports on apocritan Hymenoptera, where the presence of a pause or a longer pause during oviposition was associated with the deposition of fertilized eggs rather than unfertilized eggs. The possibility that mated Athalia rosae females control fertilization and its implications for sex allocation strategies are discussed.  相似文献
