A series of thirty N-(phenoxy)alkyl or N-{2-[2-(phenoxy)ethoxy]ethyl}aminoalkanols has been designed, synthesized and evaluated for anticonvulsant activity in MES, 6 Hz test, and pilocarpine-induced status epilepticus. Among the title compounds, the most promising seems R-(−)-2N-{2-[2-(2,6-dimethylphenoxy)ethoxy]ethyl}aminopropan-1-ol hydrochloride (22a) with proved absolute configuration with X-ray analysis and enantiomeric purity. The compound is effective in MES test with ED50 = 12.92 mg/kg b.w. and its rotarod TD50 = 33.26 mg/kg b.w. The activity dose is also effective in a neurogenic pain model—the formalin test. Within high throughput profile assay, among eighty one targets, the strongest affinity of the compound is observed towards σ receptors and 5-HT transporter and the compound does not bind to hERG. It also does not exhibit mutagenic properties in the Vibrio harveyi test. Moreover, murine liver microsomal assay and pharmacokinetics profile (mice, iv, p.o., ip) indicate that the liver is the primary site of biotransformation of the compound, suggesting that both 22a and its metabolite(s) are active, compensating probably low bioavailability of the parent molecule.  相似文献

Reaction of the N-tosyl-ethylenediamine and salicylaldehyde forms a new sulfonamide Schiff base N-[2-(2-hydroxybenzylideneamino)ethyl]-4-methyl-benzene-sulfonamide (H2L). Three novel complexes constructed from H2L, namely, [M(HL)2] · xH2O (M = Cu, x = 0 for 1, M = Ni, x = 0 for 2 and M = Zn, x = 1 for 3) have been prepared and characterized via X-ray single-crystal diffraction, elemental analysis, X-ray powder diffraction (XRPD), FT-IR, UV-Vis, TGA and photoluminescence measurements. Complex hydrogen bonds, C-H···π and π-π stacking interactions lead 1-3 to present 1-D, 2-D and 3-D supramolecular architectures, respectively.  相似文献

New cis-fused chromeno pyrano[4,3-c]isoxazole derivatives have been synthesized by intramolecular [1,3]-cycloaddition of the nitrones generated in situ from hydroxylamine derivatives and 7-O-prenyl derivatives of 8-formyl-2,3-disubstituted chromenones using PEG-400 as a reaction medium under catalyst-free conditions good to excellent yields. The structures were established by spectroscopic data and further confirmed by X-ray diffraction analysis. The results showed that compounds 4b, 4c, 4d, 4e and 4k exhibit very potent antiproliferative activity against MDA-MB-231 breast cancer cells. Compounds 4a, 4c, 4e, 4i and 4k displayed potent inhibitory activity against human MCF-7 breast cancer cell lines. Compounds 4h and 4i exhibited significant anti-proliferative activity against human cervical cancer cell line, HeLa. While 4b, 4d and 4j were active against human lung cancer cell line, A549. In addition, Compound 4j was found to be the most promising against A549 (Lung cancer) with IC50 value of 0.194 μM.  相似文献

A series of novel N-1,3-benzo[d]thiazol-2-yl-2-(2-oxo-2H-chromen-4-yl)acetamide derivatives has been synthesized. All the newly synthesized compounds were evaluated for their anti-HIV activity using MTT method. Most of these compounds showed moderate to potent activity against wild-type HIV-1 with an EC50 ranging from >7 EC50 [μg/ml] to <100 EC50 [μg/ml]. Among them, N-1,3-benzo[d]thiazol-2-yl-2-(2-oxo-2H-chromen-4-yl)acetamide 6v was identified as the most promising compound (EC50 = <7 μg/ml). Among all the compounds, three compounds 6m, 6v and 6u have been exhibits potent anti-HIV activity against MT-4 cells.  相似文献

In this study, some N-[4-(Benzothiazole-2-yl) phenyl]-2-aryloxyacetamide derivatives were prepared by reacting N-[4-(benzothiazole-2yl)phenyl]-2-chloroacetamide and different substituent phenol or thiophenol derivatives. The anticancer activities of the compounds obtained were investigated. It was observed that some of the compounds, namely 25 and 38, showed notable anticancer activity.  相似文献

The acidity constants of the two-fold protonated acyclic 9-[2-(phosphonomethoxy)ethyl]-8-azaadenine, H2(9,8aPMEA)(+)(-), and its 8-isomer, 8-[2-(phosphonomethoxy)ethyl]-8-azaadenine, H2(8,8aPMEA)(+)(-), both abbreviated as H2(PA)(+)(-), as well as the stability constants of their M(H;PA)+ and M(PA) complexes with the metal ions M2+=Mg2+, Ca2+, Sr2+, Ba2+, Mn2+, Co2+, Ni2+, Cu2+, Zn2+ or Cd2+, have been determined by potentiometric pH titrations in aqueous solution at I=0.1 M (NaNO3) and 25 degrees C. Application of previously determined straight-line plots of log K(M)M(R-PO3) versus pK(H)H(R-PO3)for simple phosph(on)ate ligands, R-PO3(2-), where R represents a residue without an affinity for metal ions, proves that for all M(PA) complexes a larger stability is observed than is expected for a sole phosphonate coordination of the metal ion. This increased stability is attributed to the formation of five-membered chelates involving the ether oxygen present in the aliphatic residue (-CH2-O-CH2-PO3(2-)) of the ligands. The formation degrees of these chelates were calculated; they vary between about 13% for Ca(8,8aPMEA) and 71% for Cu(8,8aPMEA). The adenine residue has no influence on complex stability except in the Cu(9,8aPMEA) and Zn(9,8aPMEA) systems, where an additional stability increase attributable to the adenine residue is observed and equilibria between four different isomers exist. This means (1) an open isomer with a sole phosphonate coordination, M(PA)op, where PA(2-)=9,8aPMEA2-, (2) an isomer with a five-membered chelate involving the ether oxygen, M(PA)cl/O, (3) an isomer which contains five- and seven-membered chelates formed by coordination of the phosphonate group, the ether oxygen and the N3 site of the adenine residue, M(PA)cl/O/N3, and finally (4) a macrochelated isomer involving N7, M(PA)cl/N7. For Cu(9,8aPMEA) the formation degrees are 15, 30, 48 and 7% for Cu(PA)op, Cu(PA)cl/O, Cu(PA)cl/O/N3 and Cu(PA)cl/N7, respectively; this proves that the macrochelate involving N7 is a minority species. The situation for the Cu(PMEA) system, where PMEA2- represents the parent compound, i.e. the dianion of 9-[2-(phosphonomethoxy)ethyl]adenine, is quite similar. The relationship between the antiviral activity of acyclic nucleoside phosphonates and the structures of the various complexes is discussed and an explanation is offered why 9,8aPMEA is biologically active but 8,8aPMEA is not.  相似文献

A set of two Cu(II) complexes, [Cu(cdXsalen)] and [Cu(cdXsalMeen)] derived from Schiff base ligands (H2cdXsalen: methyl-2-{[2-(2-X-phenyl)methylidynenitrilo]ethyl}amino-1-cyclopentenedithiocarboxylate and H2cdXsalMeen: methyl-2-{[1-methyl-2-(2-X-phenyl)methylidynenitrilo]ethyl}amino-1-cyclopenteneithiocarb-oxylate where X = hydroxyl, methoxy, nitro, sodiumsulfite, chloro, bromo and H2cdMesalen: methyl-2-{[2-(2-hydroxyphenyl)ethylidynenitrilo]ethyl}amino-1-cyclopentenedithiocarboxylate; H2cdPhsalen: methyl-2-{[2-(2-hydroxyphenyl)phenylidynenitrilo]ethyl}amino-1-cyclopentenedithiocarboxylate; H2cdMesalMeen: methyl-2-{[1-methyl-2-(2-hydroxyphenyl)ethylidynenitrilo]ethyl}amino-1-cyclopentenedithiocarboxylate; H2cdPhsalMeen: methyl-2-{[1-methyl-2-(2-hydroxyphenyl)phenylidynenitrilo]ethyl}amino-1-cyclopentenedi-thiocarboxylate) with an unsymmetric NNOS coordination sphere have been synthesized and characterized by elemental analysis, IR, UV-Vis and mass spectrometry. The thermodynamic formation constants of the complexes were measured spectrophotometrically, at constant ionic strength 0.1 M (NaClO4), at 25 °C in DMF solvent. The trend of the complex formation for copper is as follow:

Novel N-(1-(4-(dibenzo[b,f][1,4]thiazepin-11-yl)piperazin-1-yl)-1-oxo-3-phenylpropan-2-yl derivatives were designed, synthesized and their chemical structures were confirmed by 1H NMR, 13C NMR and Mass spectra. The anticancer activities of the newly synthesized compounds were evaluated in vitro against three human cancer cell lines including K562, Colo-205 and MDA-MB 231 by MTT assay. The screening results showed that five compounds (16b, 16d, 16i, 16p and 16q) exhibited potent cytotoxic activities with IC50 values between 20 and 40 μM. Further in vitro studies revealed that inhibition of sirtuins could be the possible mechanism of action of these molecules.  相似文献

Copper complexes: [Cu(phen)(L-Ser)(H2O)Cl] (1), [Cu(phen)(Gly)(H2O)]Cl·3H2O (2), [Cu(phen)(L-Ala)(H2O)]Cl·H2O (3), [Cu(phen)(L-Phe)(H2O)]Cl·2.5H2O (4), Cu(phen)2Cl2·6H2O (5) (phen = 1,10-phenanthroline) were synthesized and characterized. The structure of 1 was characterized by X-ray crystallography and showed in a triclinic system with space group P1, a = 6.8953(15) Å, b = 10.737(2) Å, c = 11.894(3) Å, α = 110.395(3)°, β = 94.183(4)° and γ = 100.540(3)°. The antibacterial activities on Escherichia coli (E. coli) of these five copper complexes and CuCl2 (6) were investigated by microcalorimetry. By analyzing the obtained metabolic thermogenic data and curves, crucial parameters such as rate constant of bacterial growth (k), half inhibitory concentration (IC50), and generation time (tG) were determined. All these copper complexes could stimulate the growth of the E. coli at their lower concentration. At their higher concentration they all showed antibacterial action. The inhibition on E. coli was 5 > 1 ≈ 2 ≈ 3 ≈ 4 > 6. And the antibacterial mechanism was discussed preliminarily.  相似文献

Complexes of the type [M(apash)Cl] and [M(Hapash)(H2O)SO4], where M = Mn(II), Co(II), Ni(II), Cu(II) and Zn(II); Hapash = acetone p-amino acetophenone salicyloyl hydrazone have been synthesized and characterized by elemental analyses, molar conductance, magnetic moments, electronic, ESR and IR spectra, thermal studies (TGA & DTA) and X-ray diffraction studies. The ligand coordinates through two >C=N and a deprotonated enolate group in all the chloro complexes, whereas through two >C=N- and a >C=O group in all the sulfato complexes. The electronic spectra suggest a square planar geometry for Co(II), Ni(II) and Cu(II) chloride complexes and an octahedral geometry for the sulfate complexes. ESR data show an isotropic symmetry for [Cu(apash)Cl] and [Cu(Hapash)(H2O)SO4] in solid state. However, ESR spectra of both Cu(II) complexes indicate the presence of unpaired electron in d x2-y2. The X-ray diffraction parameters for [Co(apash)Cl] and [Cu(Hapash)(H2O)SO4] complexes correspond to a tetragonal and an orthorhombic crystal lattices, respectively. Thermal studies of [Co(apash)Cl] complex shows a multi-step decomposition pattern. Most of the complexes show better antifungal activity than the standard miconazole against a number of pathogenic fungi. The antibacterial activity of these complexes has been evaluated against E. coli and Clostridium sp. which shows a moderate activity.  相似文献

The first tellurated derivative of morpholine, N-{2-(4-methoxyphenyltelluro)ethyl}morpholine (L1) has been synthesized by reacting in situ generated ArTe with 4-(2-chloroethyl)morpholine hydrochloride under N2 atmosphere. The compound L1 gives molecular ion peak at m/z 351 and is characterized structurally. The donor atoms N and Te in compound L1 are rightly oriented for its ligation in bidentate mode. The TeC(alkyl) is 0.02 Å longer than TeC(aryl). The complexes of ligand L1 having composition [PtCl2(L1)2] (1) and [RuCl2(p-cymene)L1] (2) have been synthesized. The compound 1 has been characterized structurally. The Pt has a square planar geometry in complex 1 and two molecules of ligand L1 bonded through Te alone are trans to each other (PtTe=2.583(2) Å). The 13C{1H} NMR spectrum of complex 1 is as expected. The 1H NMR spectrum of single crystals of complex 1 shows multiplication of signals, which is supported by HETCOR experiments. The complex 2 also has ligand L1 in a monodentate coordination mode, bonded through Te alone. This is supported by deshielded CH2Te and ArCTe signals in 1H and 13C{1H} NMR spectra of complex 2 with respect to those of free ligand L1. The HETCOR spectrum of complex 2 has been used to authenticate the assignments of CH2Te group, as its two protons appear to be magnetically non-equivalent.  相似文献

We have synthesized a new series of quinoline tripartite hybrids from chloroquine, ethambutol, and isoxyl drugs, using a short synthetic route. Compounds 18 were tested in vitro against five protozoa (Giardia intestinalis, Trichomonas vaginalis, Entamoeba histolytica, Leishmania mexicana and Trypanosoma cruzi) and Mycobacterium tuberculosis. N-(4-Butoxyphenyl)-N′-{2-[(7-chloroquinolin-4-yl)amino]ethyl}urea (6) was the most active compound against all parasites tested. Compound 6 was 670 times more active than metronidazole, against G. intestinalis. It was as active as pentamidine against L. mexicana, and it was twofold more potent than ethambutol and isoxyl versus M. tuberculosis. This compound could be considered as a new broad spectrum antimicrobial agent.  相似文献

Some new complexes of meclofenamic acid (N-(2,6-dichloro-m-tolyl)anthranilic acid), Hmeclo (1), with potentially interesting biological activities are described. Complexes [Mn(meclo)2] (2), [Cu(meclo)2(H2O)2] (3), [Zn(meclo)2(H2O)2] (4) and [Cd(meclo)2(H2O)2] (5) were prepared and structurally characterized by means of vibrational, electronic and 1H and 13C NMR spectroscopies. The crystal structure of complexes [Cu4(meclo)6(OH)2(DMSO)2]2DMSO (3a) and [Cd(meclo)2(DMSO)3] (5a) have been determined by X-ray crystallography. Complex (3a) is a centrosymmetric tetramer built up around the planar cyclic Cu2(OH)2 unit. Complex 5a is mononuclear seven-coordinated complex with the meclofenamato ligand behaving as a bidentate deprotonated chelating ligand. Intra and intermolecular hydrogen bonds stabilize these two structures, while the crystal packing is determined by π-π and C−H−-π interactions. Meclofenamic acid and its metal complexes have been evaluated for antiproliferative activity in vitro against the cells of three human cancer cell lines, MCF-7 (breast cancer cell line), T24 (bladder cancer cell line), and A-549 (non-small cell lung carcinoma), and a mouse fibroblast L-929 cell line. Complex 5 exhibits the highest selectivity against MCF-7 and 4 shows the highest selectivity against T-24. Complexes 2-5 were found to be more potent cytotoxic agents against T-24 and complex 5 against MCF-7 cancer cell lines than the prevalent benchmark metallodrug, cis-platin. The superoxide dismutase activity was measured by the Fridovich test which showed that complex [Cu(meclo)2(H2O)2] is a good superoxide scavenger.  相似文献

The complexes [Me2(Meclo)SnOSn(Meclo)Me2]2 (2) and [Ph3Sn(Meclo)] (3) where HMeclo is meclofenamic acid, N-(2,6-dichloro-m-tolylanthranilic acid)], have been prepared and structurally characterized by means of vibrational, 1H and 13C NMR spectroscopies. The crystal structure of complexes (2) and (3) have been determined by X-ray crystallography. Three distannoxane rings are present to the dimeric tetraorganodistannoxane of planar ladder arrangement of (2). The structure is centro symmetric and features a central rhombus Sn2O2 unit two additional tin atoms linked at the oxygen atoms. Five- and six-coordinated tin centers are present in the dimer distannoxane. X-ray analysis of (3) revealed a penta-coordinated structure containing Ph3Sn coordinated to the chelated carboxylato group. The polar imino hydrogen atom participates in intra-molecular hydrogen bonds. Complexes (2) and (3) are self-assembled via π → π, C-H-π, stacking interactions and intra-molecular hydrogen bonds. Meclofenamic acid and [Ph3Sn(Meclo)] have been evaluated for antiproliferative activity in vitro against three human cancer cell lines: MCF-7 (human breast cancer cell line), T24 (bladder cancer cell line), A-549 (non-small cell lung carcinoma) and a mouse L-929 (a fibroblast-like cell line cloned from strain L). The [Ph3Sn(Meclo)] complex exhibited high cytotoxic activity against all the cancer cell lines. Meclofenamic and [Ph3Sn(Meclo)] were tested for anti-mycobacterial activity against Mycobacterium tuberculosis H37Rv. The [Ph3Sn(Meclo)] complex was found to be a promising anti-mycobacterial lead compound, displaying high activity against M. tuberculosis H37Rv.  相似文献

A series of new 1,3,4-oxadiazole derivatives (6a6x) containing pyridine and acylhydrazone moieties were synthesized and developed as potential telomerase inhibitors. The bioassay tests demonstrated that compounds 6n, 6o, 6q, 6s and 6t exhibited significant broad-spectrum anticancer activity with IC50 range from 0.76 to 9.59 μM against the four cancer cell lines (HEPG2, MCF7, SW1116 and BGC823). Moreover, all the title compounds were assayed for telomerase inhibition using the TRAP-PCR-ELISA assay. Compound 6s showed the highest anticancer activity with IC50 of 0.76–1.54 μM against the tested cancer cell lines and exhibited the most potent telomerase inhibitory activity with IC50 of 1.18 ± 0.14 μM. The docking simulation was carried out to investigate a possible binding mode of compound 6s into the active site of telomerase (pdb. 3DU6) while the QSAR model was built to check the previous work as well as to introduce new directions.  相似文献

Using new nitronyl nitroxide radical ligand 2-[4-(1-imidazole)phenyl]-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (NITPhIm), three new complexes [M(hfac)2(NITPhIm)]2 (M = Cu(II) 1, Mn(II) 2, Co(II) 3; hfac = hexafluoroacetylacetonate) have been prepared. Three complexes possess cyclic dimer structure in which each NITPhIm radical links two different metal ions through the oxygen of nitroxide group and the nitrogen of imidazole. The magnetic studies show the copper(II) ion interacts ferromagnetically with the directly bonding nitronyl nitroxide while manganese(II) and cobalt(II) ions strong antiferromagnetically interact with the directly coordinated nitroxide groups. There is a weak antiferromagnetic coupling between the metal ion and the nitroxide through phenyl and imidazole rings of the radical ligand, which is agreement with spin polarization mechanism. The results show that the minor changes in the structure of radical ligand can change the magnetic behavior of radical-metal complex.  相似文献

In a previous study, it was demonstrated that N-(phenethyl)succinamic acid (PESA) derivatives form a new category of root-promoting substances which do not exhibit auxin-like activities, such as stem elongation and leaf epinasty (Soejima et al., 2000 [Plant Cell Physiol. 41s: 197]). In this study, N-[2-(3-indolyl)ethyl]succinamic acid (IESA) and N-[2-(1-naphthyl)ethyl]succinamic acid (NESA) were synthesized, and their biological activities were evaluated. In an adzuki root-promoting assay, IESA and NESA exhibited root-promoting activity equivalent to PESA. In adzuki stem elongation assays, elongation activity was not observed in the stem segments soaked in either an IESA or NESA aqueous solution, whereas the stem segments immersed in Indole-3-acetic acid (IAA) or 1-naphthylacetic acid (NAA) aqueous solution were clearly elongated. In an epinastic bending study, IAA and NAA exhibited leaf epinasty, whereas IESA and NESA did not, suggesting that the IESA and NESA derivatives belong to the same category of root-promoting substances as PESA derivatives and are different from auxin-like substances. In addition, eleven kinds of IESA derivatives and nineteen kinds of NESA derivatives were synthesized, and their root-promoting activities were measured. The activities of methyl ester derivatives were approximately three times higher than that of the acid compounds, with exceptions for some compounds. The partition coefficient (P) between 1-octanol and water for each IESA, NESA, and PESA derivative was measured in order to evaluate the hydrophobicity of their molecules and to determine their structure–activity relationship. The results indicate that the root-promoting activity of the acid compounds was significantly correlated with their hydrophobicity, whereas that of ester derivatives was not correlated.  相似文献

Reaction of the five-coordinate trigonal-bipyramidal platinum(II) complex, [Pt(pt)(pp3)](BF4) (pt = 1-propanethiolate, pp3 = tris[2-(diphenylphosphino)ethyl]phosphine), with I in chloroform gave the five-coordinate square-pyramidal complex with a dissociated terminal phosphino group and an apically coordinated iodide ion in equilibrium. The thermodynamic parameters for the equilibrium between the trigonal-bipyramidal and square-pyramidal geometries, [Pt(pt)(pp3)]+ + I ? [PtI(pt) (pp3)], and the kinetic parameters for the chemical exchange were obtained as follows: , ΔH0 = − 10 ± 2.4 kJ mol−1, ΔS0 = − 36 ± 10 J K−1 mol−1, , ΔH = 34 ± 4.7 kJ mol−1, ΔS = − 50 ± 21 J K−1 mol−1. The square-planar trinuclear platinum(II) complex was formed by bridging reaction of one of the terminal phosphino groups of trigonal-bipyramidal [PtCl(pp3)]Cl with trans-[PtCl2(NCC6H5)2] in chloroform. From these facts, ligand substitution reactions of [PtX(pp3)]+ (X = monodentate anion) are expected to proceed via an intermediate with a dissociated phosphino group. The rate constants for the chloro-ligand substitution reactions of [PtCl(pp3)]+ with Br and I in chloroform approached the respective limiting values as concentrations of the entering halide ions are increased. These kinetic results confirmed the preassociation mechanism in which the square pyramidal intermediate with a dissociated phosphino group and an apically coordinated halide ion is present in the rapid pre-equilibrium.  相似文献

The synthesis, characterization, crystal structure, thermal stability, optical spectroscopic, electronic structure, and second-order nonlinear optical (NLO) properties of a novel class of materials, based on planar [N4]Ni(II) complexes, are reported. The single-crystal X-ray structure of the dichloro derivative indicates a planar structure of the molecule. The Ni(II) ion is tetracoordinated in a square planar environment, with the four nitrogen atoms as donors. The unsubstituted derivative exhibits efficiency 1.2 times that of urea in second-harmonic generation at 1.9 μm. INDO/SCI-SOS quantum-chemical calculations predict a sizeable molecular response and a significant octupolar contribution to optical nonlinearity.  相似文献

To investigate the effect of organic anions on the coordination frameworks, we synthesized three new complexes, namely, Zn(DPA)(bpimb)0.5(H2O) (1), Zn(BDC)(bpimb)0.5 (2) and Zn2(SDBA)2(bpimb)·H2O (3) (H2DPA = diphenic acid; H2BDC = isophthalic acid; H2SDBA = 4,4′-dicarboxybiphenylsulfone), which were obtained by the reactions of 1,4-bis((2-(pyridin-2-yl)-1H-imidazol-1-yl)methyl)benzene (bpimb) as main ligand, and several aromatic polycarboxylate as organic anions with Zn(NO3)2·6H2O. Single-crystal structure analysis shows that complex 1 is a one-dimensional chain structure, which is further interlinked into a higher-dimensional supramolecular framework by hydrogen-bonding interactions. In 2, BDC bridge Zn(II) atoms to give dimeric units, which are further linked by bpimb ligands to form sql nets. In 3, SDBA ligands and bpimb ligands connect Zn(II) ions into catenane-like two-dimensional layers. These catenane-like two-dimensional layers stack in an ABAB fashion to form a 3D supramolecular network. The distinct structures indicate three kinds of carboxylic ligands with different lengths and angles play fundamental roles in the formation of the final products.
