澳大利亚亚热带不同森林土壤微生物群落对碳源的利用   总被引:5,自引:0,他引:5  
为阐明土壤微生物群落对碳源利用类型和强度,采用MicroRespTM方法研究3种森林类型不同土壤含水量微生物群落对不同类型碳源的利用情况,结果表明:湿地松(Pinus elliottii Engelm. var. elliotttii)林地土壤微生物碳源利用率依次为60%WHC>20%WHC>40%WHC,南洋杉(Araucaria cunninghamii)和贝壳杉(Agathis australis)林地土壤微生物对碳源利用的格局相似,为20%WHC>60%WHC>40%WHC。南洋杉和贝壳杉林地土壤对碳源利用率趋势相同,主要是对L-苹果酸、草酸和L-赖氨酸利用比较高。在40%WHC处理中,3种树种对碳源的利用均很低,差异不明显。除精氨酸和L-赖氨酸外,60%WHC处理土壤微生物利用单一碳源能力的大小顺序为:南洋杉>湿地松>贝壳杉。3种树种土壤Shannon多样性指数(H’)、Shannon均匀度(E)和Simpson指数(D)均无显著差异。土壤pH值影响微生物对L-丙氨酸、精氨酸、D-( )-葡萄糖、N-乙酰基-氨基葡萄糖的利用率较大,这些类群的微生物主要分布在贝壳杉林地;分布在南洋杉林地的微生物对柠檬酸、L-苹果酸和γ-酪氨酸利用率较大,且主要是受TP的影响;D-( )-果糖、柠檬酸和L-半胱氨酸-盐酸等受水分、TN和TC等影响较大,这类微生物类群主要分布在湿地松林地。  相似文献

As shown before [C. Ottander et al. (1995) Planta 197:176-183], there is a severe inhibition of the photosystem (PS) II photochemical efficiency of Scots pine (Pinus sylvestris L.) during the winter. In contrast, the in vivo PSI photochemistry is less inhibited during winter as shown by in vivo measurements of deltaA820/A820 (P700+). There was also an enhanced cyclic electron transfer around PSI in winter-stressed needles as indicated by 4-fold faster reduction kinetics of P700+. The differential functional stability of PSII and PSI was accompanied by a 3.7-fold higher intersystem electron pool size, and a 5-fold increase in the stromal electron pool available for P700+ reduction. There was also a strong reduction of the QB band in the thermoluminescence glow curve and markedly slower Q-A re-oxidation in needles of winter pine, indicating an inhibition of electron transfer between QA and QB. The data presented indicate that the plastoquinone pool is largely reduced in winter pine, and that this reduced state is likely to be of metabolic rather than photochemical origin. The retention of PSI photochemistry, and the suggested metabolic reduction of the plastoquinone pool in winter stressed needles of Scots pine are discussed in terms of the need for enhanced photoprotection of the needles during the winter and the role of metabolically supplied energy for the recovery of photosynthesis from winter stress in evergreens.  相似文献

为探讨混交林中松树对害虫的生态调控作用机制,研究了不同林分类型下马尾松林和湿地松林对马尾松毛虫生长发育的影响。结果表明,取食马尾松针叶的马尾松毛虫与取食湿地松针叶的相比,其发育历期缩短,死亡率降低,幼虫及蛹(雌、雄)重增加,化蛹率、羽化率和产卵量均提高;且其幼虫的代谢量(取食量与排泄量)更大,对食物的利用与消耗的各项指标均高。取食不同林分类型下的同一种松树(马尾松或湿地松)针叶的马尾松毛虫,以取食纯林松针的幼虫生长发育更快,死亡率更低,化蛹率、羽化率和产卵量更高,对食物的利用、转化和消耗率更强。混交林中的松针不利于马尾松毛虫的生长发育和种群增长。  相似文献

The impact of the Mondi Syktyvkar Timber Industry Complex on the chemical composition of pine needles was investigated. A comparative analysis of the mineral composition of pine needles showed that the age dynamics of the content of the vast majority of investigated elements in the case of both the studied background region and the zone affected by anthropogenic emissions demonstrated similar trends. The total content of chemical elements in bilberry pine forests showed no difference depending on the needle age or the distance from the source of pollution. An examination carried out in 2007 revealed that the total element concentration in one-year-old pine needles was higher than that in older ones. Over the period of studies, a decrease in pine needle mineralization was observed under the influence of pollution in comparison with the background territory. Additionally, aerotechnogenic emissions of nitrogen and sulfur oxides as well as carbon containing compounds resulted in no accumulation of the above-mentioned substances in pine needles except for the impacted stand of 1.3 km.  相似文献

【目的】为探讨包埋法固定化过程对硫氧化菌群硫化物去除能力及菌群微生物群落结构的影响,【方法】以聚乙烯醇-海藻酸钠-活性炭为载体,对硫氧化菌群进行了固定化,并采用富含硫化物的无机盐培养基,对比固定化与非固定化硫氧化菌群对硫化物的氧化去除能力。同时,利用PCR-DGGE技术,探讨硫氧化菌群在固定化前后以及在硫化物氧化去除过程中微生物群落结构变化。【结果】在对硫氧化菌群进行固定化之后,12 h之内对硫化物的最大去除能力从1000 mg/L下降为600 mg/L。硫氧化菌群的微生物群落结构发生了明显变化,但菌群中的硫氧化菌Catenococcus thiocycli未受影响,硫氧化菌Thioclava pacifica在菌群中的地位反而得到了强化。【结论】受制于底物在载体材料中的扩散迁移效率,硫氧化菌群对硫化物的氧化去除能力在固定化之后有所下降。由于不同微生物对固定化形成的微环境的适应能力以及对载体附着能力的不同,固定化对硫氧化菌群的微生物群落结构产生较大影响。  相似文献

Methods were developed to determine proteinase activity in germinating seeds of Scots pine. The assays were based on the liberation of TCA-soluble peptides from haemoglobin at pH 3.7 and from casein at pH 5.4 and pH 7.0; the reaction products were determined by the Lowry method. — Endosperms separated from seeds at the time of rapid storage protein mobilization (seedling length between 20 and 50 mm) showed high proteinase activities in all three assays. Experiments with different inhibitors suggested that at least four enzymes were involved. One of the enzymes resembled mammalian and microbial pepsin-like acid proteinases: the pH optimum was 3.7 and the enzyme was inhibited by pepstatin.—The proteinase activities in the endosperms were high enough to account for the mobilization of the reserve proteins during germination. Moreover the activities at pH 3.7.5.4. and 7.0 in the endosperms were 10-, 25-, and 50-fold the corresponding activities in the growing seedlings (a “reference” tissue). Consequently, it seems that both the acid and neutral proteinases take part in the mobilization of storage proteins in the germinating seed.  相似文献

The renewable pine needles was used as an adsorbent to remove phosphorus from aqueous solutions. Using batch experiments, pine needles pretreated with alkali-isopropanol (AI) failed to effectively remove phosphorus, while pine needles modified with lanthanum hydroxide (LH) showed relatively high removal efficiency. LH pine needles were effective at a wide pH ranges, with the highest removal efficiency reaching approximately 85% at a pH of 3. The removal efficiency was kept above 65% using 10 mg/L phosphorus solutions at desired pH values. There was no apparent significant competitive behavior between co-existing anions of sulfate, nitrate, and chloride (SO4 2-, NO3 - and Cl-); however, CO3 2- exhibited increased interfering behavior as concentrations increased. An intraparticle diffusion model showed that the adsorption process occurred in three phases, suggesting that a boundary layer adsorption phenomena slightly affected the adsorption process, and that intraparticle diffusion was dominant. The adsorption process was thermodynamically unfavorable and non-spontaneous; temperature increases improved phosphorus removal. Total organic carbon (TOC) assays indicated that chemical modification reduced the release of soluble organic compounds from 135.6 mg/L to 7.76 mg/L. This new information about adsorption performances provides valuable information, and can inform future technological applications designed to remove phosphorus from aqueous solutions.  相似文献

用马尾松老叶、新叶,湿地松老叶、新叶,当年生马尾松新叶、湿地松新叶和当年生以20%和40%β-蒎烯处理过的马尾松新叶作食料饲养马尾松毛虫。结果表明:马尾松毛虫幼虫在第1代发生期取食马尾松老叶、马尾松新叶、湿地松老叶和湿地松新叶等4种不同质量的食料后,因其食料中10种必需氨基酸含量和挥发性物质β-蒎烯含量的不同,导致幼虫的存活率差异显著。其中,取食马尾松老叶的幼虫成活率最高(44%),依次为马尾松新叶(32.7%)、湿地松老叶(4%),而取食湿地松新叶的幼虫全部死亡。通过马尾松针叶增加外援β-蒎烯喂养马尾松毛虫实验,表明增加了外源β-蒎烯的松针饲养的马尾松毛虫幼虫与用同样松针未增加外源β-蒎烯饲养的幼虫相比,死亡率明显提高;松树中β-蒎烯含量的增加与幼虫成活率呈显著负相关。由此,我们阐明了马尾松与湿地松对马尾松毛虫的抗虫机制。  相似文献

Nitrogen metabolism of the needles of 40-year-old Douglas fir and Scots pine trees, growing in two forest stands on cation-poor and acidic sandy soil with a relatively high atmospheric nitrogen deposition was studied. The composition of the free amino acid (FAA) pool, the concentrations of total nitrogen and soluble protein and the activities of glutamine synthetase (GS) and glutamate dehydrogenase (GDH) were determined in the needles. An excessive nitrogen supply by a high atmospheric nitrogen deposition in both forest stands was indicated by the high concentrations of total nitrogen and the amino acids arginine, glutamic acid, glutamine and aspartic acid in control trees. In addition the effect of optimal nutrition and water supply (fertigation) on the needle nitrogen metabolism was evaluated. The total concentration of the FAA pool in needles of both tree species was lower in the fertigated than in the non-fertigated (control) trees, except for 1-year-old needles of Scots pine, in which the concentration after fertigation did not differ from the control. The lower total FAA concentration in the fertigated trees could be attributed to arginine, the concentration of which was on average 60% lower than in the control. Neither the concentration of soluble protein nor the activity of GS were influenced by fertigation. The activity of GDH in fertigated trees only differed significantly from the control in October. Scots pine needles had higher concentrations of protein (50%) and higher activities of GS (44%) and GDH (25%) than Douglas fir needles. Possible explanations for the lower vitality of Douglas fir compared to Scots pine are given.  相似文献

通过接种马尾松毛虫Dendrolimus punctatus和人工剪叶,分析比较了不同受害方式对马尾松针叶内化学物质的影响。结果表明,害虫取食松针诱导的次生代谢物质(单宁、酚类物质)比人工剪叶处理略有增加,而营养物质-可溶性糖变化不大。对马尾松进行害虫取食为害、人工剪叶受害、未受害三种处理后,连续3年跟踪测定了松针叶内化学物质含量的变化。结果发现,无论是害虫取食为害,还是人工剪叶受害,针叶内次生代谢物质含量都减少,可溶性糖含量降低,直至一年后才恢复到原有的水平;蛋白质变化的趋势则始终是受害处理的含量比未受害处理的含量高,表明马尾松受害后诱导的化学物质变化具有滞后特性。  相似文献

在微生物层面研究枯梢病与植物针叶内生微生物的互作关系,分析内生真菌的多样性差异,为松枯梢病的防控提供基础数据。利用高通量测序技术测定赤松(Pinus densiflora)不同染病程度的针叶内生真菌的多样性。结果显示,随着病害的加重,P. densiflora针叶内生真菌丰富度呈现出上升的趋势,多样性指数表现为先下降后上升的趋势。无病斑针叶内生真菌中,子囊菌门(Ascomycota)与担子菌门(Basidiomycota)相对丰度最高,优势属为Lapidomyces和Selenophoma,病害导致优势菌相对丰度的降低。通过对不同染病情况的P. densiflora针叶内生真菌的测定,明确了枯梢病不同发病程度的P. densiflora针叶内生真菌的多样性及群落结构组成。  相似文献

Foliage of Scots pine (Pinus sylvestris L.) and pedunculate oak (Quercus robur L.) was collected in a mixed pine/oak forest at canopy positions differing in radiation environment. In both species, chlorophyll (Chl) a/b ratios were higher in foliage of canopy positions exposed to higher irradiance as compared to more shaded crown layers. Throughout the growing season, pine needles exhibited significantly lower Chl a/b ratios than oak leaves acclimated to a similar photon availability. Hence, pine needles showed shade-type pigment characteristics relative to foliage of oak. At a given radiation environment, pine needles tended to contain more neoxanthin and lutein per unit of Chl than oak leaves. The differences in pigment composition between foliage of pine and oak can be explained by a higher ratio of outer antennae Chl to core complex Chl in needles of P. sylvestris which enhances the efficiency of photon capture under limiting irradiance. The shade-type pigment composition of pine relative to oak foliage could have been due to a reduced mesophyll internal photon exposure of chloroplasts in needles of Scots pine, resulting from their xeromorphic anatomy. Hence, the higher drought tolerance of pine needles could be achieved at the expense of shade tolerance.  相似文献

Results of a long-term (1997-2002) study of the Scots pine populations growing in the vicinity of the radioactive waste storage facility ('Radon' LWPE) are presented. Cytogenetic disturbances in reproductive (seeds) and vegetative (needles) tissues sampled from Scots pine populations were studied to examine whether Scots pine trees have experienced environmental stress in areas with relatively low levels of pollution. The data clearly indicate the presence of mutagenic contaminants in the environment of the pine trees. An increased number of mitotic abnormalities, especially multipolar mitoses was found in the pine tree populations submitted to man-made exposure, which suggests that the cytogenetic damage is mainly caused by chemical contamination. A higher radioresistance of the Scots pine seeds from the impacted populations was shown by use of acute gamma-irradiation. During the observation period 1997-2002, pine trees exposed to anthropogenic pollution showed a steady increase of cytogenetic alterations in the root meristem cells.  相似文献

Antioxidant metabolites in eastern white pine (Pinus strobus L.) needles increased two- to fourfold from the summer to the winter season. Antioxidant enzymes in needle tissue increased between 2- and 122-fold during this same period. These seasonal changes were determined by monitoring ascorbate and glutathione concentrations and the activity of ascorbate peroxidase, glutathione reductase (GR), and superoxide dismutase. Levels of antioxidant metabolites and enzymes were observed always to be lowest during the summer, or active growing season, and highest during the winter, or dormant season. These data correlated well with the thermal kinetic window for purified GR obtained from summer needles. The minimum, apparent Km,NADPH for two isoforms of GR (GRA and GRB) occurred at 5 and 10°C, respectively. The upper limit of the thermal kinetic window (200% of the minimum Km) for GRA and GRB was 20 and 25°C, respectively, indicating that needle temperatures exceeding 25°C may result in impairment of antioxidant metabolism. The needle content and kinetic properties of GR, the increased activities of other enzymes, and the high substrate concentrations observed during the winter are consistent with the protective function this pathway may provide against photooxidative, winter injury.  相似文献

Formation of pinosylvin (PS) and pinosylvin 3-O-monomethyl ether (PSM), as well as the activities of stilbene synthase (STS) and S-adenosyl-l-methionine (SAM):pinosylvin O-methyltransferase (PMT), were induced strongly in needles of Scots pine seedlings upon ozone treatment, as well as in cell suspension cultures of Scots pine upon fungal elicitation. A SAM-dependent PMT protein was purified and partially characterised. A cDNA encoding PMT was isolated from an ozone-induced Scots pine cDNA library. Southern blot analysis of the genomic DNA suggested the presence of a gene family. The deduced protein sequence showed the typical highly conserved regions of O-methyltransferases (OMTs), and average identities of 20–56% to known OMTs. PMT expressed in Escherichia coli corresponded to that of purified PMT (40 kDa) from pine cell cultures. The recombinant enzyme catalysed the methylation of PS, caffeic acid, caffeoyl-CoA and quercetin. Several other substances, such as astringenin, resveratrol, 5-OH-ferulic acid, catechol and luteolin, were also methylated. Recombinant PMT thus had a relatively broad substrate specificity. Treatment of 7-year old Scots pine trees with ozone markedly increased the PMT mRNA level. Our results show that PMT represents a new SAM-dependent OMT for the methylation of stress-induced pinosylvin in Scots pine needles.  相似文献

We studied the effects of phosphorus fertilisation on foliar terpene concentrations and foliar volatile terpene emission rates in six half-sib families of Pinus pinaster Ait. seedlings. Half of the seedlings were resistant to attack of the pine weevil Hylobius abietis L., a generalist phloem feeder, and the remaining seedlings were susceptible to this insect. We hypothesised that P stress could modify the terpene concentration in the needles and thus lead to altered terpene emission patterns relevant to plant-insect signalling. The total concentration and emission rate ranged between 5732 and 13,995 μg·g(-1) DW and between 2 and 22 μg·g(-1) DW·h(-1), respectively. Storage and emission were dominated by the isomers α- and β-pinene (77.2% and 84.2% of the total terpene amount amassed and released, respectively). In both resistant and susceptible families, P stress caused an increase of 31% in foliar terpene concentration with an associated 5-fold decrease in terpene emission rates. A higher terpene content in the leaves implies that the 'excess carbon', available under limiting growth conditions (P scarcity), is allocated to terpene production. Sensitive families showed a greater increase in terpene emission rates with increasing P concentrations, which could explain their susceptibility to H. abietis.  相似文献

Laboratory investigations were conducted to study potential mechanisms for prolonged survival of Pseudomonas aeruginosa in poloxamer-iodine (PxI). P. aeruginosa organisms isolated from PxI and adapted for growth in distilled water or found as part of a mixed microbial population from water in a manufacturing plant did not survive more than 15 s after challenge in stock PxI solution. Batches of PxI were compounded in the laboratory to determine the survival and growth of P. aeruginosa during the various stages of preparation. No P. aeruginosa organisms were recovered from the finished product at 1 min after the addition of iodine-iodide. However, we found P. aeruginosa in PxI 48 h after adding sterile PxI to the inside of a naturally contaminated polyvinyl chloride water distribution pipe. These organisms (10(4) CFU/ml) survived for as long as 98 days in contaminated stock PxI after it was removed from the polyvinyl chloride pipe. Both decreasing the free iodine level through addition of potassium iodide and increasing the free iodine level through dilution of the product resulted in an increased length of survival of P. aeruginosa in contaminated PxI solution. Comparative survival studies with pipes of different composition revealed that other materials may exert an effect similar to polyvinyl chloride. We concluded that polyvinyl chloride and perhaps other materials may play an important role in the survival of P. aeruginosa in iodophors and may be one source of resistant microbial populations when used in manufacturing plants which produce these antimicrobial solutions.  相似文献

The autochthonous microbiota from a granitic aquifer in Spain were studied. Several bacterial strains were isolated and identified. The major components of the microbiota were Pseudomonas-like strains, Flavobacterium, Acinetobacter, and Alcaligenes. The variation in the number of microorganisms after the bottling process was studied. The initial bacterial population increased over the first 5 days after bottling. This increase was higher in samples from polyvinyl chloride bottles. Sonication usually increased the total cell counts.
