A hyposmotic challenge elicited contraction of isolated canine basilar arteries. The contractile response was nearly abolished by the removal of extracellular Ca2+ and by the voltage-dependent Ca2+ channel (VDCC) blocker nicardipine, but it was unaffected by thapsigargin, which depletes intracellular Ca2+ stores. The contraction was also inhibited by Gd3+ and ruthenium red, cation channel blockers, and Cl channel blockers DIDS and niflumic acid. The reduction of extracellular Cl concentrations enhanced the hypotonically induced contraction. Patch-clamp analysis showed that a hyposmotic challenge activated outwardly rectifying whole cell currents in isolated canine basilar artery myocytes. The reversal potential of the current was shifted toward negative potentials by reductions in intracellular Cl concentration, indicating that the currents were carried by Cl. Moreover, the currents were abolished by 10 mM BAPTA in the pipette solution and by the removal of extracellular Ca2+. Taken together, these results suggest that a hyposmotic challenge activates cation channels, which presumably cause Ca2+ influx, thereby activating Ca2+-activated Cl channels. The role of nitric oxide (NO) in the occurrence of intracellular Ca2+ concentration ([Ca2+]i) oscillations in pituitary GH3 cells was evaluated by studying the effect of increasing or decreasing endogenous NO synthesis with L-arginine and nitro-L-arginine methyl ester (L-NAME), respectively. When NO synthesis was blocked with L-NAME (1 mM) [Ca2+]i, oscillations disappeared in 68% of spontaneously active cells, whereas 41% of the quiescent cells showed [Ca2+]i oscillations in response to the NO synthase (NOS) substrate L-arginine (10 mM). This effect was reproduced by the NO donors NOC-18 and S-nitroso-N-acetylpenicillamine (SNAP). NOC-18 was ineffective in the presence of the L-type voltage-dependent Ca2+ channels (VDCC) blocker nimodipine (1 µM) or in Ca2+-free medium. Conversely, its effect was preserved when Ca2+ release from intracellular Ca2+ stores was inhibited either with the ryanodine-receptor blocker ryanodine (500 µM) or with the inositol 1,4,5-trisphosphate receptor blocker xestospongin C (3 µM). These results suggest that NO induces the appearance of [Ca2+]i oscillations by determining Ca2+ influx. Patch-clamp experiments excluded that NO acted directly on VDCC but suggested that NO determined membrane depolarization because of the inhibition of voltage-gated K+ channels. NOC-18 and SNAP caused a decrease in the amplitude of slow-inactivating (IDR) and ether-à-go-go-related gene (ERG) hyperpolarization-evoked, deactivating K+ currents. Similar results were obtained when GH3 cells were treated with L-arginine. Decoding of fast cytosolic Ca2+ concentration ([Ca2+]i) transients by mitochondria was studied in permeabilized cat ventricular myocytes. Mitochondrial [Ca2+] ([Ca2+]m) was measured with fluo-3 trapped inside mitochondria after removal of cytosolic indicator by plasma membrane permeabilization with digitonin. Elevation of extramitochondrial [Ca2+] ([Ca2+]em) to >0.5 µM resulted in a [Ca2+]em-dependent increase in the rate of mitochondrial Ca2+ accumulation ([Ca2+]em resulting in half-maximal rate of Ca2+ accumulation = 4.4 µM) via Ca2+ uniporter. Ca2+ uptake was sensitive to the Ca2+ uniporter blocker ruthenium red and the protonophore carbonyl cyanide p-trifluoromethoxyphenylhydrazone and depended on inorganic phosphate concentration. The rates of [Ca2+]m increase and recovery were dependent on the extramitochondrial [Na+] ([Na+]em) due to Ca2+ extrusion via mitochondrial Na+/Ca2+ exchanger. The maximal rate of Ca2+ extrusion was observed with [Na+]em in the range of 20–40 mM. Rapid switching (0.25–1 Hz) of [Ca2+]em between 0 and 100 µM simulated rapid beat-to-beat changes in [Ca2+]i (with [Ca2+]i transient duration of 100–500 ms). No [Ca2+]m oscillations were observed, either under conditions of maximal rate of Ca2+ uptake (100 µM [Ca2+]em, 0 [Na+]em) or with maximal rate of Ca2+ removal (0 [Ca2+]em, 40 mM [Na+]em). The slow frequency-dependent increase of [Ca2+]m argues against a rapid transmission of Ca2+ signals between cytosol and mitochondria on a beat-to-beat basis in the heart. [Ca2+]m changes elicited by continuous or pulsatile exposure to elevated [Ca2+]em showed no difference in mitochondrial Ca2+ uptake. Gustatory transduction mechanisms for sucrose and amino acidsweeteners in gerbil taste cells were studied with Ca2+ imagingand whole cell recording techniques. A 100 mM sucrose stimuluswith Ca2+ increased the intracellular Ca2+ concentration ([Ca2+]i)in sweet-sensitive taste cells of the taste bud, but the sucrosestimulus without Ca2+ did not change the [Ca2+]i. A 10 mM D-phenylalaninesweet stimulus with or without Ca2+ similarly increased the[Ca2+]i in the taste bud. The addition of 5 µM ionomysin,a Ca2+-ionophore, without Ca2+ greatly increased the [Ca2+]iin the taste bud. The application of 10 mM D-phenylalanine stimuluswithout Ca2+ enhanced the outward K+ current in isolated tastecells. These results suggest that a sugar sweetener such assucrose induces a depolarization in gerbil taste cells whichactivates voltage-dependent Ca2+ channels and that a non-sugarsweetener such as D-phenylalanine releases Ca2+ from the internalstores without a depolarization. Cl is essential for the vasoconstrictive response to angiotensin II (ANG II). In vascular smooth muscle cells (VSMC), we determined whether ANG II-induced transient increase in intracellular Ca2+ concentration ([Ca2+]i) is Cl dependent. After incubating the cells at different extracellular Cl concentration ([Cl]e) for 40 min, the ANG II-induced Ca2+ transients at 120 meq/l Cl were more than twice those at either 80 or 20 meq/l Cl. Replacing Cl with bicarbonate or gluconate yielded similar results. In addition, after removal of extracellular Ca2+, ANG II-induced as well as platelet-derived growth factor-induced Ca2+ release exhibited Cl dependency. The difference of Ca2+ release with high vs. low [Cl]e was not affected by acutely altering [Cl]e 1 min before administration of ANG II when [Cl]i was yet to be equilibrated with [Cl]e. Pretreatment of a Cl channel inhibitor, 5-nitro-2-(3-phenylpropylamino)benzoic acid, increased ANG II-induced Ca2+ release and entry at 20 meq/l Cl but did not alter those at 120 meq/l Cl. However, after equilibration, a reduced [Cl]e did not affect thapsigargin-induced Ca2+ release, suggesting that Cl may not affect the size of intracellular Ca2+ stores. Nevertheless, at high [Cl], the peak increase of inositol 1,4,5-trisphosphate [Ins(1,4,5)P3] induced by ANG II was approximately sixfold that at low [Cl]. In response to hypotonic treatment internodal cells of the brackishwater Characeae Lamprothamnium regulate turgor pressure by releasingK+ and Cl, accompanying membrane depolarization and atransient increase in membrane electrical conductance (Okazakiet al. 1984b). The hypothesis that a transient increase in cytoplasmicfree Ca2+ concentration ([Ca2+]c) caused by hypotonic treatmenttriggers release of K+ and Cl from the cell (Okazakiand Tazawa 1986a, b, c) was tested using tonoplast-removed cells.These cells did not regulate turgor pressure. The plasmalemmaconductance remained almost constant for a change in the intracellularfree Ca2+ concentration ([Ca2+],) from 10–6 to 10–2mol?m–3. The results suggest that some cytoplasmic Ca2+-sensitizingsoluble components, which work as mediators to activate K+ and/orCl channels in the plasmalemma and/or the tonoplast,were lost after desintegration of the tonoplast. The plasmalemmapotential was depolarized under high [Ca2+]i. However, no membranedepolarization was observed upon hypotonic treatment. Volume changes and whole cell ionic currents activated by gradual osmolarity reductions (GOR) of 1.8 mosM/min were characterized in C6 glioma cells. Cells swell less in GOR than after sudden osmolarity reductions (SOR), the extent of swelling being partly Ca2+ dependent. In nominally Ca2+-free conditions, GOR activated predominantly whole cell outward currents. Cells depolarized from the initial –79 mV to a steady state of –54 mV reached at 18% osmolarity reduction [hyposmolarity of –18% (H-18%)]. Recordings of Cl and K+ currents showed activation at H-3% of an outwardly rectifying Cl current, with conductance of 1.6 nS, sensitive to niflumic acid and 5-nitro-2-(3-phenylpropylamino)benzoic acid, followed at H-18% by an outwardly rectifying K+ current with conductance of 4.1 nS, inhibited by clofilium but insensitive to the typical K+ channel blockers. With 200 nM Ca2+ in the patch pipette, whole cell currents activated at H-3% and at H-13% cells depolarized from –77 to –63 mV. A K+ current activated at H-1%, showing a rapid increase in conductance, suppressed by charybdotoxin and insensitive to clofilium. These results show the operation of two different K+ channels in response to GOR in the same cell type, activated by Ca2+ and osmolarity and with different osmolarity activation thresholds. Taurine and glutamate efflux, monitored by labeled tracers, showed delayed osmolarity thresholds of H-39 and H-33%, respectively. This observation clearly separates the Cl and amino acid osmosensitive pathways. Effects of cytoplasmic Ca2+ on the electrical properties ofthe plasma membrane were investigated in tonoplast-free cellsof Chara australis that had been internally perfused with media,containing either 1 mM ATP to fuel the electrogenic pump orhexokinase and glucose to deplete the ATP and stop the pump. In the presence of ATP, cytoplasmic Ca2+ up to 2.5?10–5M did not affect the membrane potential (about -190 mV), butmembrane resistance decreased uniformly with increasing [Ca2+]i.In the absence of ATP, the membrane potential, which was onlyabout -110 mV, was depolarized further by raising [Ca2+]i from1.4?10–6 to 2.5?10–5 M. Membrane resistance, whichwas nearly the twofold that of ATP-provided cells, decreasedmarkedly with an increase in [Ca2+]i from zero to 1.38?10–6M, but showed no change for further increases. Internodal cellsof Nitellopsis obtusa were more sensitive to intracellular Ca2+with respect to membrane potential than were those of Charaaustralis, reconfirming the results obtained by Mimura and Tazawa(1983). The effect of cytoplasmic Ca2+ on the ATP-dependent H+ effluxwas measured. No marked difference in H+ effluxes was detectedbetween zero and 2.5?10–5 M [Ca2+]i; but, at 10–4M the ATP-dependent H+ efflux was almost zero. Ca2+ efflux experimentswere done to investigate dependencies on [Ca2+]i and [ATP]i.The efflux was about 1 pmol cm–2 s–1 at all [Ca2+]iconcentrations tested (1.38?10–6, 2.5?10–5, 10–4M).This value is much higher than the influx reported by Hayamaet al. (1979), and this efflux was independent of [ATP]i. Localized Ca2+ transients resulting from inositoltrisphosphate (IP3)-dependent Ca2+ releasecouple to spontaneous transient outward currents (STOCs) in murinecolonic myocytes. Confocal microscopy and whole cell patch-clamptechniques were used to investigate coupling between localizedCa2+ transients and STOCs. Colonic myocytes were loadedwith fluo 3. Reduction in external Ca2+([Ca2+]o) reduced localized Ca2+transients but increased STOC amplitude and frequency. Simultaneous recordings of Ca2+ transients and STOCs showed increasedcoupling strength between Ca2+ transients and STOCs when[Ca2+]o was reduced. Gd3+ (10 µM) did not affect Ca2+ transients but increased STOCamplitude and frequency. Similarly, an inhibitor of Ca2+influx,1-2-(4-methoxyphenyl)-2-[3-(4-methoxyphenyl)propoxy]ethyl-1H-imidazole (SKF-96365), increased STOC amplitude and frequency. A protein kinase C(PKC) inhibitor, GF-109203X, also increased the amplitude and frequencyof STOCs but had no effect on Ca2+ transients. Phorbol12-myristate 13-acetate (1 µM) reduced STOC amplitude and frequencybut did not affect Ca2+ transients. 4-Phorbol (1 µM)had no effect on STOCs or Ca2+ transients. Single channelstudies indicated that large-conductance Ca2+-activatedK+ (BK) channels were inhibited by aCa2+-dependent PKC. In summary 1)Ca2+ release from IP3 receptor-operated storesactivates Ca2+-activated K+ channels;2) Ca2+ influx through nonselective cationchannels facilitates activation of PKC; and 3) PKC reducesthe Ca2+ sensitivity of BK channels, reducing the couplingstrength between localized Ca2+ transients and BK channels.

Ricinus communis L. var. Gibsonii was grown in Long Ashton nutrientmedium with either 12mol m–3 NO3 or 8.0 mol m–3NH+4 as N source. Two plants from each N treatment were harvestedtwice a week and analysed for C, N, P, S, NO3, SO2–4ClK+Na+, Ca2+ Mg2+ and ash alkalinity. Statistical analysis of thedata showed that the effect of age and N source was differentfor the chemical variables analysed. Thus [Na+] was unaffectedby age or N source, and for both N sources [Mg2+] started atthe same level and decreased at the same rate as the plantsmatured. With NH+4 as N source, [SO2–4] was higher thanwith NO3, but did not alter with age. The concentrations,in mmol g–1 dry wt, of C, organic N, K+ and Ca2+ weredifferent for the two N sources, but the levels of these variablesaltered with age in the same way for both N sources; i.e. therewas no age x N interaction. In the case of P, NO3, Cl and COO, however,age-related variations were different for the two N sources.It is concluded, inter alia, that [Na+] is determined by external[Na+] alone, and that K+, Ca2+ and Cl are the inorganicions actively involved in charge balance during ion uptake bythe roots. Using the whole cell patch clamp technique, we measured changesin outward K+ currents of gerbil taste cells in response todifferent kinds of sweeteners. Outward K+ currents of the tastecell induced by depolarizing pulses were suppressed by sweetstimuli such as 10 mM Na-saccharin. The membrane-permeable analogof cAMP, cpt-cAMP, also decreased outward K+ currents. On theother hand, the K+ currents were enhanced by amino acid sweetenerssuch as 10 mM D-tryptophan. The outward K+ current was enhancedby external application of Ca2+-transporting ionophore, 5 µMionomycin, and intracellular application of 5 µM inositol-1,4,5-trisphosphate(IP3). The outward K+ currents were no longer suppressed by10 mM Na-saccharin containing 20 µM gurmarin, but werestill enhanced by 10 mM D-tryptophan containing 20 µMgurmarin. These results suggest that sweet taste transductionfor one group of sweeteners such as Na-saccharin in gerbilsis concerned with an increase of the intracellular cAMP level,and that the transduction for the other group of sweetenerssuch as D-tryptophan is concerned with an increase of the intracellularIP3 level which releases Ca2+ from the internal stores. We investigatedthe role of intracellular calcium concentration([Ca2+]i) in endothelin-1 (ET-1) production,the effects of potential vasospastic agents on[Ca2+]i, and the presence of L-typevoltage-dependent Ca2+ channels in cerebral microvascularendothelial cells. Primary cultures of endothelial cells isolated frompiglet cerebral microvessels were used. Confluent cells were exposed toeither the thromboxane receptor agonist U-46619 (1 µM),5-hydroxytryptamine (5-HT; 0.1 mM), or lysophosphatidic acid (LPA; 1 µM) alone or after pretreatment with the Ca2+-chelatingagent EDTA (100 mM), the L-type Ca2+ channel blockerverapamil (10 µM), or the antagonist of receptor-operated Ca2+ channel SKF-96365 HCl (10 µM) for 15 min. ET-1production increased from 1.2 (control) to 8.2 (U-46619), 4.9 (5-HT),or 3.9 (LPA) fmol/µg protein, respectively. Such elevated ET-1biosynthesis was attenuated by verapamil, EDTA, or SKF-96365 HCl. Toinvestigate the presence of L-type voltage-dependent Ca2+channels in endothelial cells, the [Ca2+]isignal was determined fluorometrically by using fura 2-AM. Superfusionof confluent endothelial cells with U-46619, 5-HT, or LPA significantlyincreased [Ca2+]i. Pretreatment ofendothelial cells with high K+ (60 mM) or nifedipine (4 µM) diminished increases in [Ca2+]i inducedby the vasoactive agents. These results indicate that 1)elevated [Ca2+]i signals are involved in ET-1biosynthesis induced by specific spasmogenic agents, 2) theincreases in [Ca2+]i induced by thevasoactive agents tested involve receptor as well as L-typevoltage-dependent Ca2+ channels, and 3) primarycultures of cerebral microvascular endothelial cells express L-typevoltage-dependent Ca2+ channels.

The role of mitochondria inCa2+ homeostasis is controversial.We employed the Ca2+-sensitive dyerhod 2 with novel, high temporal and spatial resolution imaging toevaluate changes in the matrix freeCa2+ concentration of individualmitochondria([Ca2+]m)in agonist-stimulated, primary cultured aortic myocytes. Stimulation with 10 µM serotonin (5-HT) evoked modest cytosolicCa2+ transients[cytosolic freeCa2+ concentration([Ca2+]cyt)<500 nM; measured with fura 2] and triggered contractions inshort-term cultured myocytes. However, 5-HT triggered a large mitochondrial rhod 2 signal (indicating pronounced elevation of [Ca2+]m)in only 4% of cells. This revealed heterogeneity in the responses ofindividual mitochondria, all of which stained with MitoTracker GreenFM. In contrast, stimulation with 100 µM ATP evoked large cytosolicCa2+ transients (>1,000 nM) andinduced pronounced, reversible elevation of[Ca2+]m(measured as rhod 2 fluorescence) in 60% of cells. This mitochondrial Ca2+ uptake usually lagged behindthe cytosolic Ca2+ transient peakby 3-5 s, and[Ca2+]mdeclined more slowly than did bulk[Ca2+]cyt.The uptake delay may prevent mitochondria from interfering with rapidsignaling events while enhancing the mitochondrial response to large,long-duration elevations of[Ca2+]cyt.The responses of arterial myocytes to modest physiological stimulationdo not, however, depend on such marked changes in [Ca2+]m.  相似文献

We previously showed that plasma membrane Ca2+-ATPase (PMCA) activity accounted for 25–30% of relaxation in bladder smooth muscle (8). Among the four PMCA isoforms only PMCA1 and PMCA4 are expressed in smooth muscle. To address the role of these isoforms, we measured cytosolic Ca2+ ([Ca2+]i) using fura-PE3 and simultaneously measured contractility in bladder smooth muscle from wild-type (WT), Pmca1+/–, Pmca4+/–, Pmca4–/–, and Pmca1+/–Pmca4–/– mice. There were no differences in basal [Ca2+]i values between bladder preparations. KCl (80 mM) elicited both larger forces (150–190%) and increases in [Ca2+]i (130–180%) in smooth muscle from Pmca1+/– and Pmca1+/–Pmca4–/– bladders than those in WT or Pmca4–/–. The responses to carbachol (CCh: 10 µM) were also greater in Pmca1+/– (120–150%) than in WT bladders. In contrast, the responses in Pmca4–/– and Pmca1+/–Pmca4–/– bladders to CCh were significantly smaller (40–50%) than WT. The rise in half-times of force and [Ca2+]i increases in response to KCl and CCh, and the concomitant half-times of their decrease upon washout of agonist were prolonged in Pmca4–/– (130–190%) and Pmca1+/–Pmca4–/– (120–250%) bladders, but not in Pmca1+/– bladders with respect to WT. Our evidence indicates distinct isoform functions with the PMCA1 isoform involved in overall Ca2+ clearance, while PMCA4 is essential for the [Ca2+]i increase and contractile response to the CCh receptor-mediated signal transduction pathway. In isolated rat pancreatic -cells, hypotonic stimulation elicited an increase in cytosolic Ca2+ concentration ([Ca2+]c) at 2.8 mM glucose. The hypotonically induced [Ca2+]c elevation was significantly suppressed by nicardipine, a voltage-dependent Ca2+ channel blocker, and by Gd3+, amiloride, 2-aminoethoxydiphenylborate, and ruthenium red, all cation channel blockers. In contrast, the [Ca2+]c elevation was not inhibited by suramin, a P2 purinoceptor antagonist. Whole cell patch-clamp analyses showed that hypotonic stimulation induced membrane depolarization of -cells and produced outwardly rectifying cation currents; Gd3+ inhibited both responses. Hypotonic stimulation also increased insulin secretion from isolated rat islets, and Gd3+ significantly suppressed this secretion. Palytoxin-induced cell death cascade in bovine aortic endothelial cells   总被引:1,自引:0,他引:1  
The plasmalemmal Na+-K+-ATPase (NKA) pump is the receptor for the potent marine toxin palytoxin (PTX). PTX binds to the NKA and converts the pump into a monovalent cation channel that exhibits a slight permeability to Ca2+. However, the ability of PTX to directly increase cytosolic free Ca2+ concentration ([Ca2+]i) via Na+ pump channels and to initiate Ca2+ overload-induced oncotic cell death has not been examined. Thus the purpose of this study was to determine the effect of PTX on [Ca2+]i and the downstream events associated with cell death in bovine aortic endothelial cells. PTX (3–100 nM) produced a graded increase in [Ca2+]i that was dependent on extracellular Ca2+. The increase in [Ca2+]i initiated by 100 nM PTX was blocked by pretreatment with ouabain with an IC50 < 1 µM. The elevation in [Ca2+]i could be reversed by addition of ouabain at various times after PTX, but this required much higher concentrations of ouabain (0.5 mM). These results suggest that the PTX-induced rise in [Ca2+]i occurs via the Na+ pump. Subsequent to the rise in [Ca2+]i, PTX also caused a concentration-dependent increase in uptake of the vital dye ethidium bromide (EB) but not YO-PRO-1. EB uptake was also blocked by ouabain added either before or after PTX. Time-lapse video microscopy showed that PTX ultimately caused cell lysis as indicated by release of transiently expressed green fluorescent protein (molecular mass 27 kDa) and rapid uptake of propidium iodide. Cell lysis was 1) greatly delayed by removing extracellular Ca2+ or by adding ouabain after PTX, 2) blocked by the cytoprotective amino acid glycine, and 3) accompanied by dramatic membrane blebbing. Allosteric regulation by cytosolic Ca2+ of Na+/Ca2+ exchange activity in the Ca2+ efflux mode has received little attention because it has been technically difficult to distinguish between the roles of Ca2+ as allosteric activator and transport substrate. In this study, we used transfected Chinese hamster ovary cells to compare the Ca2+ efflux activities in nontransfected cells and in cells expressing either the wild-type exchanger or a mutant, (241–680), that operates constitutively; i.e., its activity does not require allosteric Ca2+ activation. Expression of the wild-type exchanger did not significantly lower the cytosolic Ca2+ concentration ([Ca2+]i) compared with nontransfected cells. During Ca2+ entry through store-operated Ca2+ channels, Ca2+ efflux by the wild-type exchanger became evident only after [Ca2+]i approached 100–200 nM. A subsequent decline in [Ca2+]i was observed, suggesting that the activation process was time dependent. In contrast, Ca2+ efflux activity was evident under all experimental conditions in cells expressing the constitutive exchanger mutant. After transient exposure to elevated [Ca2+]i, the wild-type exchanger behaved similarly to the constitutive mutant for tens of seconds after [Ca2+]i had returned to resting levels. Pulmonary vasoconstriction and vascularmedial hypertrophy greatly contribute to the elevated pulmonaryvascular resistance in patients with pulmonary hypertension. A rise incytosolic free Ca2+ ([Ca2+]cyt)in pulmonary artery smooth muscle cells (PASMC) triggers vasoconstriction and stimulates cell growth. Membrane potential (Em) regulates[Ca2+]cyt by governing Ca2+influx through voltage-dependent Ca2+ channels. Thusintracellular Ca2+ may serve as a shared signaltransduction element that leads to pulmonary vasoconstriction andvascular remodeling. In PASMC, activity of voltage-gated K+(Kv) channels regulates resting Em. In thisstudy, we investigated whether changes of Kv currents[IK(V)], Em, and[Ca2+]cyt affect cell growth by comparingthese parameters in proliferating and growth-arrested PASMC. Serumdeprivation induced growth arrest of PASMC, whereas chelation ofextracellular Ca2+ abolished PASMC growth. Resting[Ca2+]cyt was significantly higher, andresting Em was more depolarized, inproliferating PASMC than in growth-arrested cells. Consistently, wholecell IK(V) was significantly attenuated in PASMCduring proliferation. Furthermore, Emdepolarization significantly increased resting[Ca2+]cyt and augmented agonist-mediatedrises in [Ca2+]cyt in the absence ofextracellular Ca2+. These results demonstrate that reducedIK(V), depolarized Em, and elevated [Ca2+]cyt may play a criticalrole in stimulating PASMC proliferation. Pulmonary vascular medialhypertrophy in patients with pulmonary hypertension may be partlycaused by a membrane depolarization-mediated increase in[Ca2+]cyt in PASMC.

Addition of salicylic acid (SA) to tobacco (Nicotiana tabacum)suspension culture immediately induced a rapid and transientgeneration of superoxide anion (O2), followed by a transientincrease in cytosolic free calcium ion concentration ([Ca2+]c).The level of SA-induced O2 was lowered by treatment withseveral scavengers of active oxygen species and a peroxidaseinhibitor, but not with an NADPH oxidase inhibitor. The SA-induced[Ca2+]c elevation was also lowered by inhibitors which effectivelylowered the O2 level. Inhibition of [Ca2+]c elevationby Ca2+ channel blockers and a Ca2+ chelator indicated thatextracellular Ca2+ was responsible for the increased [Ca2+]c.Among the several SA analogs, only compounds that actively inducedthe O2 generation also elevated [Ca2+]cIn addition, theinhibitory effects of SA analogs on catalase activity correlatedwell with their effects on the O2 generation and the[Ca2+]c elevation. SA-dependent O2 generation was shownto occur extracellularly, requiring both H2O2 and at least oneproteinaceous factor excreted from the cells. Ethanol strongly augments secretin-stimulated, but not acetylcholine (ACh)-stimulated, fluid secretion from pancreatic duct cells. To understand its mechanism of action, we examined the effect of short-chain n-alcohols on fluid secretion and intracellular Ca2+ concentration ([Ca2+]i) in guinea pig pancreatic ducts. Fluid secretion was measured by monitoring the luminal volume of isolated interlobular ducts. [Ca2+]i was estimated using fura-2 microfluorometry. Methanol and ethanol at 0.3–10 mM concentrations significantly augmented fluid secretion and induced a transient elevation of [Ca2+]i in secretin- or dibutyryl adenosine 3',5'-cyclic monophosphate (DBcAMP)-stimulated ducts. However, they failed to affect fluid secretion and [Ca2+]i in unstimulated and ACh-stimulated ducts. In contrast, propanol and butanol at 0.3–10 mM concentrations significantly reduced fluid secretion and decreased [Ca2+]i in unstimulated ducts and in ducts stimulated with secretin, DBcAMP, or ACh. Both stimulatory and inhibitory effects of n-alcohols completely disappeared after their removal from the perfusate. Propanol and butanol inhibited the plateau phase, but not the initial peak, of [Ca2+]i response to ACh as well as the [Ca2+]i elevation induced by thapsigargin, suggesting that they inhibit Ca2+ influx. Removal of extracellular Ca2+ reduced [Ca2+]i in duct cells and completely abolished secretin-stimulated fluid secretion. In conclusion, there is a distinct cutoff point between ethanol (C2) and propanol (C3) in their effects on fluid secretion and [Ca2+]i in duct cells.
