A study has been made of the content and composition of RNA in cytoplasm, nucleoplasm, and nucleoli from growing oocytes of the starfish Asterias rubens. The determinations were carried out, using ultramicrochemical methods, on units isolated by microdissection from fixed sections. Macrochemical and interferometric control experiments show that RNA can be quantitatively evaluated in this way. The results show that the growing oocyte represents a system in which the relations between the quantities of nucleolar, nucleoplasmic, and cytoplasmic RNA undergo great changes. These changes are continuous for nucleolar and cytoplasmic RNA so that their amounts may be predicted from the size of the cell. Nucleoplasmic RNA, on the other hand, shows great variations among different cells, independent of cell size. Purine-pyrimidine analyses show that each cell component contains an RNA which differs significantly from that of the other two. Cytoplasmic and nucleolar RNA are closely related, the only difference being a slightly higher guanine/uracil quotient for the nucleolar RNA. They are both of the usual tissue RNA type, i.e., they show a preponderance of guanine and cytosine over adenine and uracil. Nucleoplasmic RNA deviates grossly from the RNA of the other two components. Here the concentrations of adenine and uracil are higher than those of guanine and cytosine, respectively. This RNA consequently shows some resemblance to the general type of animal DNA although the purine/pyrimidine ratio is far from unity. Our data favor a nucleolar origin for the stable part of the ribosomal RNA and a nucleoplasmic one for the unstable part (the messenger RNA).  相似文献

The base composition of RNA's extracted from chromosomes, nucleoli, nuclear sap, and cytoplasm of Triturus oocytes has been determined by microelectrophoresis. The chromosomal RNA has a content of guanine+cytosine equal to that of DNA, but there is no complementarity in the composition as for DNA. Nuclear sap contains a highly variable RNA with a tendency towards high uracil values. Nucleolar and cytoplasmic RNA's are similar in composition and both are of the guanine-cytosine rich type. The chromosomes and nucleoli contain roughly equivalent amounts of RNA, somewhat less than is present in the nuclear sap. The RNA/DNA ratio of the whole chromosomes is about 10. However, the ratio in the synthetically active regions, the loops, is much higher, since the loops contain all the chromosomal RNA but only a small fraction of the DNA.  相似文献

Abstract— The fluorescence of chlorotetracycline (CTC) in the presence of synaptosomes isolated from sheep brain is selectively increased by Ca2+ under conditions in which Mg2+, Na+, K+, Li+ or choline have only a small effect. The monovalent cations release bound Ca2+ from synaptosomes, and this effect is reflected by a decrease in the CTC fluorescence. Under optimal conditions there is a near parallelism between Ca2+ and CTC binding to the synaptosomes membranes, and Li+ is the monovalent cation tested which interferes the most with the binding of both substances. These results obtained in a predominantly sucrose medium become less distinct when media simulating physiological composition are utilized, which limits the usefulness of the method. Brain mitochondria and myelin also bind Ca2+ and CTC. The ratio of the fluorescence signal (or CTC bound) to Ca2+ bound is highest of all for mitochondrial membranes, and the apparent fluorescence quantum yield of CTC is also the highest in these membranes, which suggests that the Ca2+ in these membranes is localized in a more apolar region than is the case for synaptosomes and myelin.  相似文献

Abstract— Segments of spinal cords from goldfish or from carp were incubated in vitro in the presence of RNA precursors for varying periods of time. Mauthner nerve fibres were isolated from the fresh unfixed tissue, or, for the separate analyses of axon and myelin sheath, from the fixed spinal cord.
The myelinated Mauthner fibre isolated from the incubated spinal cord showed RNA synthesis. A considerable part of the material sedimented at 4 S , but part of the nucleic acid was recovered at higher sedimentation values, up to 30 S. Newly synthesized RNA was extracted from the isolated myelin sheath as well as from the axon. Isolated myelinated Mauthner fibres were also incubated in vitro with RNA precursors. In this case incorporation occurred exclusively in material sedimenting at 4 S or lower. The turnover rate for RNA from the fibre was of a higher order than that of the bulk of RNA from the spinal cord. The findings of RNA synthesis in these tissue components lacking nuclei could possibly be explained as owing to mitochondria.
Studies by electron microscopy demonstrated the extent of purity of the isolated components and it was found that contamination was so small as to make it unlikely that the RNA investigated originated in contaminating tissue.  相似文献

The structure of the vitelline nuclei of Lycosidae and Thomisidae was described as follows: Vitelline nuclei are constituted of two parts: (a) a peripheral layer (vitelline body cortex), and (b) a central core. The vitelline body cortex is demonstrated to be formed by many cisternae of the endoplasmic reticulum among which mitochondria and Golgi elements are intermingled. The central core is made up mainly of a special type of body described under the name of "capsulated body." Capsulated bodies comprise a capsular layer, limited by a membrane, and two central masses called "geminated masses," each one limited by a double membrane. Irregular masses of closely packed vesicles are found in some cases among the capsulated bodies and free vesicles are present in large numbers. The optical properties of the vitelline body cortex compared with the electron microscope findings lead us to the concept that this layer is a "composite body" according to Weiner's theory.  相似文献

Nuclei from isolated nerve cells were sampled by microdissection. The content and composition of the nuclear RNA was studied and compared with that of the cytoplasmic RNA of Deiters' nerve cells of rabbits. Analyses were made of control nerve cells and of cells in which an enhanced RNA and protein production had been induced by chemical means, tricyano-amino-propene, for 60 minutes. The nuclear RNA content of the control nerve cells was 56 µµg, i.e. 3 per cent of the total RNA content of the nerve cell. The base ratios were: adenine 21.3, guanine 26.6, cytosine 30.8, uracil 21.3. Purine-pyrimidine analyses showed that the nuclear RNA differed significantly from the cytoplasmic RNA in having higher adenine and uracil values. The guanine and cytosine values were high, however, and the ratio G/C was 0.86 as compared with 1.16 for the cytoplasmic RNA. The composition of the nuclear RNA was interpreted as reflecting the extraordinarily strong development of the nucleolus in these neurons. During the 60 minutes of enhanced neuronal RNA production (+25 per cent) the guanine value increased and the uracil value decreased significantly in the nuclear RNA. In the cytoplasmic RNA the guanine value also increased although not so much as the nuclear guanine. The cytoplasmic cytosine value decreased. The result indicated that the production of the characteristic cytoplasmic RNA had been influenced by the change in the nuclear RNA  相似文献

The incorporation of uridine into RNA in brain slices was studied. Optimal conditions for uridine incorporation were determined. The characteristics of the product suggest that de novo DNA-directcd synthesis of fairly high molecular weight material takes place. Incorporation into RNA of several areas of brain was studied. The incorporation was also studied as a function of the age of the animal. Finally, an apparent correlation was observed between the decrease in uridine incorporation with age and the increase of the enzyme uridine nucleosidase which hydrolyses uridine to uracil, a material which cannot be incorporated into RNA.  相似文献

嗜热微生物的嗜热特性与其蛋白质的高度热稳定性紧密相关。为了探索嗜热蛋白质的热稳定机制,比较嗜热和嗜常温微生物的蛋白质在氨基酸组成上的差别,收集110对分别来自嗜热和嗜常温微生物的同源蛋白质序列,比较两组蛋白质各种氨基酸含量以及疏水性氨基酸组成、疏水性指数和荷电氨基酸组成的差别,结果两者在多种氨基酸含量上存在微小但统计学上显著的差别,嗜热蛋白质比嗜常温蛋白质具有较高的平均疏水性和荷电氨基酸组成。对两组蛋白质的“脂肪族氨基酸指数”进行分析,证明嗜热蛋白质之所以具有较高的脂肪族氨基酸指数是由于其亮氨酸含量较高,与影响该指数的其它几种氨基酸无关;从而认为该指数的意义值得怀疑。通过对大量同源嗜热蛋白质和嗜常温蛋白质氨基酸组成的比较,能够揭示一些有关蛋白质热稳定性的普遍规律。  相似文献

The labile precursors of ribosomal RNA in mouse kidney are preserved when nuclei rapidly isolated after sieving through multiple screens are swollen and cleansed in the presence of an RNase inhibitor before digestion with DNase and phenol extraction. The kinetics of nucleolar labeling analyzed on polyacrylamide gels show that 36S RNA is the major intermediate product in the catabolism of the original 45S RNA precursor to 32S RNA, from which 28S RNA is derived. Each kidney nucleus contains about 200–600 molecules of 45S RNA; the turnover time of the 45S pool is about 3 ± 2 min. Compared with HeLa cells, kidney nuclei have a different major intermediate product and a much smaller and more rapidly turning-over pool of ribosomal precursor RNA.  相似文献

栝楼属(Trichosanthes)属葫芦科,系多年生草质藤本植物。全世界有60余种,我国分布55种。中药栝楼为葫芦科植物栝楼(Trichosanthes ki-rilowii Maxim.)的果实,本植物的根(天花粉),果皮(栝楼皮),种子(栝楼子)均供药用,具抗菌抗癌作用[1],为一种重要的中药。该属植物的种子富含油脂,我们所研究的样品,多数含油率都在20%以上。本文研究了该属七种植物的种子油,经气相色谱分析,紫外和红外光谱鉴定,同时采用安石榴(Punica granatum L.)种子油中的石榴酸作标准样品进行相应的平行测定作参照,证实这七种种子油均含有石榴酸,而且含量丰富。石榴酸(顺-9,反-11,顺-13-十八碳三烯酸)是一种特殊结构的脂肪酸,在植物界中相当少见。它于1935年首次被发现于安石榴(Punica granatum L.)的种子油中[5],因而得名。至今半个世纪过去,该酸仅仅在葫芦科等的二、三个属中少数的几个种的种子油中被发现有它的分布。近几年来,先后已有瓜蒌子(Trichosanthes quinquangulata A.Gray),双边栝楼(Trchosanthes uniflora Hao),大苞栝楼(Trichosanthes bracteata(Lam.) Voigt)和毛果栝楼(Trichosanthes mushaensis Hayata)四种栝楼属植物的种子油脂肪酸化学组成的研究见于国内文献的报导[2,3],在这些报导当中均没有石榴酸组成成分,其中双边栝楼,大苞栝楼和毛果栝楼被包括在我们的研究里面,我们得出不同的结果。本文报导中其余的几种,多数未见于国内外文献的报导。  相似文献

A procedure for the purine-pyrimidine analysis of RNA in the 100- to 1000 µµg. range is presented. It includes hydrolysis and electrophoretic analysis of RNA, which is extracted from single isolated tissue units, like single cells. The quantitative determination of the separated compounds is carried out by a photographic-photometric procedure in ultraviolet light. The determined values show a coefficient of variation of about ±7 per cent on test substance. Microelectrophoretic analyses of RNA from different sources have been performed and are compared to macrochemical analyses. The agreement is good in those cases in which it is possible to get any information at all through macrochemical analyses.  相似文献

The cellular RNA content of mouse fibroblasts incubated with actinomycin decreases at a rate of about 1 to 1.5 per cent per hour, while DNA and protein content remain unchanged. This degradation affects nuclear and cytoplasmic RNA, ribosomal and soluble RNA. The breakdown products appear quantitatively in the acid-soluble fraction of the cells and the medium. Polynucleotides synthesized a short period (120 minutes) prior to exposure to actinomycin are degraded before those synthesized 8 to 12 hours previously.  相似文献

用高效液相色谱法和酶解的方法检测了银杏叶片磷脂酰甘油(PG)脂肪酸的分子种组成和位置分布,确定银杏叶片PG主要分子种的脂肪酸组成(sn1/sn2)是18∶3/16∶1(3t),18∶3/16∶0,18∶2/16∶1(3t),18∶2/16∶0,18∶1/16∶1(3t),16∶0/16∶1(3t),18∶1/18∶1,18∶1/16∶0和16∶0/16∶0。银杏叶片PG脂肪酸组成和位置分布的分析结果表明,C18脂肪酸主要位于sn1位,16∶1(3t)只分布于sn2位,16∶0在sn1位和sn2位上均有发现。sn1位上的不饱和度∑u大于sn2位上的∑u。  相似文献
