A large number of different mutations in the tumor suppressor gene p53 gene have been identified in all types of cancer. As of September 1995, this database contains over 4200 mutations. This substantial increase since our previous report can enable epidemiological analyses which were not previously possible. In order to capture all these new data, the software permitting analysis has been improved. This report describes the various improvements since first release of the database.  相似文献

Calorimetric methods have been used to determine equilibrium constants since 1937, but no comprehensive review of the various calorimeters and methods has been done previously. This article reports methods for quantitative comparison of the capabilities of calorimeters for simultaneous determination of equilibrium constants and enthalpy changes, for determining optimal experimental conditions, and for assessing the effects of systematic and random errors on the accuracy and precision of equilibrium constants and enthalpy changes determined by this method.  相似文献

Targeted therapy has replaced the conventional methods of disease management with the advances in recombinant technology and increased understanding of molecular mechanisms of diseases. The immunotoxin strategy for diseases like cancer and a variety of autoimmune disorders has been used successfully in the past since its discovery. Since bacterial, fungal and plant toxins have various limitations like toxicity and immunogenicity, studies on fully humanized immunotoxins have gained attraction recently, which reduced toxicity significantly. Improved methods of antibody engineering have led to the emergence of various new formats of immunotoxins. This review summarizes the target moieties used in immunotoxin constructs in different diseases and describes the recent advances in immunotoxin targeting.  相似文献

Autophagy has long been thought of as a bulk degradation system in which cytoplasmic components are sequestered by double-membrane structures called autophagosomes, and the contents are then degraded after autophagosomes fuse with lysosomes. Genetic experiments in yeast identified a set of Autophagy-related (ATG) genes that are essential for autophagy. We have since elucidated many of the molecular underpinnings of autophagy and the physiologic roles of these processes in various systems. This review summarizes the physiologic roles of autophagy with a particular focus on liver autophagy based on analyses of knockout mice lacking Atg genes.  相似文献

The quinazolinone-containing 2,3-disubstituted piperidines febrifugine and isofebrifugine have been the subject of significant research efforts since their occurrence in Dichroa febrifuga and their anti-malarial actions were first described in the late 1940s. Subsequently they have also been shown to be present in other plants belonging to the hydrangea family and various analogues of febrifugine have been prepared in attempts to tune biological properties. The most notable analogue is termed halofuginone and a substantial body of work now demonstrates that this compound possesses potent human disease relevant activities. This review focuses on the literature associated with efforts dedicated towards uncovering the structures of febrifugine and isofebrifugine, the development of practical methods for their synthesis and the syntheses of structural analogues.  相似文献

Nation-wide phenological observations have been made in Switzerland since 1951. In addition to these observation programmes, there are two very long phenological series in Switzerland: leaf bud burst of horse-chestnut trees has been observed in Geneva since 1808 and full flowering of cherry trees in Liestal since 1894. In addition to the presentation of these two long phenological series, trends for 896 phenological time series have been calculated with national data from 1951 to 1998. The earlier bud burst of horse-chestnut trees in Geneva can be attributed mainly to the city effect (warmth island). This phenomenon was not observed with the cherry tree flowering in Liestal. A clear trend towards earlier appearance dates in spring and a weak tendency towards later appearance dates in autumn could be shown with data from the national observation network. Amylose is a linear polymer of α-1,4-linked glucose and is expected to be used in various industries as a functional biomaterial. However, pure amylose is currently not available for industrial purposes, since the separation of natural amylose from amylopectin is difficult. It is known that amylose has been synthesized using various enzymes. Glucan phosphorylase, together with its substrate, glucose-1-phosphate, is the most suitable system for the production of amylose since the molecular size of amylose can be controlled precisely. However, the problem with this system is that glucose-1-phosphate is too expensive for industrial purposes. This review summarizes our work on the enzymatic synthesis of essentially linear amylose, together with recent progress in the production of synthetic amylose using sucrose or cellobiose through the combined actions of phosphorylases.  相似文献

The assertion that a significant portion of the mammalian genome has not been translated and that non-coding RNA accounts for over half of polyadenylate RNA have received much attention. In recent years, increasing evidence proposes non-coding RNAs (ncRNAs) as new regulators of various cellular processes, including cancer progression and nerve damage. Apoptosis is a type of programmed cell death critical for homeostasis and tissue development. Cancer cells often have inhibited apoptotic pathways. It has recently been demonstrated that up/down-regulation of various lncRNAs in certain types of tumors shapes cancer cells' response to apoptotic stimuli. This review discusses the most recent studies on lncRNAs and apoptosis in healthy and cancer cells. In addition, the role of lncRNAs as novel targets for cancer therapy is reviewed here. Finally, since it has been shown that lncRNA expression is associated with specific types of cancer, the potential for using lncRNAs as biomarkers is also discussed.  相似文献

It has been almost a decade since the last review appeared comparing and contrasting the influences that the different families of High Mobility Group proteins (HMGA, HMGB and HMGN) have on the various DNA repair pathways in mammalian cells. During that time considerable progress has been made in our understanding of how these non-histone proteins modulate the efficiency of DNA repair by all of the major cellular pathways: nucleotide excision repair, base excision repair, double-stand break repair and mismatch repair. Although there are often similar and over-lapping biological activities shared by all HMG proteins, members of each of the different families appear to have a somewhat ‘individualistic’ impact on various DNA repair pathways. This review will focus on what is currently known about the roles that different HMG proteins play in DNA repair processes and discuss possible future research areas in this rapidly evolving field.  相似文献

The world literature since 1962 gives further substantiation of the usefulness of serologic measurement of glutamic-oxalacetic transaminase (GOT) activity and lactic dehydrogenase (LDH) activity in the diagnosis of myocardial infarction. LDH has not been found superior to GOT. The isoenzymes of LDH are now creating more and more interest because of their specificity for damage in various organs. As of October, 1997, this database (http:// perso.curie.fr/tsoussi ) contained >7500 mutations. Such a substantial increase since our previous reports should enable epidemiological analyses which were not previously possible. In order to analyse these new data, the UMD software has been improved. A new Web version of the UMD software enables online analysis of the database. Censuses of buffalo and wildebeest in the Serengeti-Mara region have been carried out by various workers at intervals since 1958. The methods of these censuses, which normally employ a total count, are described and the errors inherent in such techniques are analysed in detail. The perception of the color of a surface can be influenced by many factors including its material properties and the composition of the illuminant. McCollough demonstrated that sensory conditioning could also influence the perception of surface color by inducing a long-lasting pattern specific color aftereffect. This effect has been extensively studied since its original report and a number of increasingly complex explanations have been proposed. In this article I examine the temporal properties of a simple learning model of the McCollough effect (ME). This model has previously been used to account for quantitative data sets obtained from a series of monocular and binocular variants of the ME. The model replicates the acquisition and decay of the ME, pre- and post-induction interference effects, and can also simulate the effects of various cholinergic and anticholinergic drugs that have been shown to influence ME induction and decay.  相似文献

The White Stork (Ciconia ciconia) has an extensive European breeding distribution. There have been significant demographic changes since the 1930s, with country-specific extinctions throughout the western distribution since the mid-1940s. Following various reintroduction programs significant concerns have been raised about the geographic origins of introduced storks, and the effect this may have on individual biology, particularly breeding success. Here we report on seven new microsatellite markers developed for C. ciconia that have been successfully combined in a series of multiplex PCR’s with six markers previously described for the Wood Stork.  相似文献

Continuing periosteal apposition (CPA) of small amounts of new lamellar bone, leading to absolutely larger size, has been identified in a number of adult cranial and postcranial bones. This paper reviews 42 studies published since 1964 that have found both significant and nonsignificant age-related change in various skeletal size dimensions, e.g., length, diameter, width, and area. Also considered are four hypotheses that have, or may be, postulated for the occurrence of CPA. To date, however, these hypotheses (cohort effect, mechanical compensation, bone repair and/or mechanical response potential, and heterochrony) have not been rigorously tested, hence remain speculative. An important interpretive problem that befalls the investigation of CPA is its small effect size (i.e., the magnitude of change between observations), since most studies have restricted sample sizes. This problem is illustrated by power analysis of three reviewed studies that reported nonsignificant age-related change. The analysis indicates that these studies had very little likelihood of finding a statistically significant result, i.e., a low probability of rejecting the null hypothesis stipulating no size change with age. This finding has implications for interpreting CPA and for distinguishing between the statistical and biological significance of this phenomenon.  相似文献
