Ion homeostasis is considered to be one of the most important mechanisms underlying salt stress tolerance. We used the Steptoe × Morex barley doubled haploid population to screen for genetic variation in response to salinity stress at an early development stage in a hydroponics system, focusing on ion homeostasis. Salinity induced a strong adverse effect on growth of the parents and their derived population, with Steptoe as the more tolerant parent. Steptoe maintained higher concentrations of K+, Na+ and Cl? in the roots and a similar shoot/root ion ratio (<1) under stress conditions compared to control conditions. In contrast, Morex had higher concentrations of these ions in the shoots under stress and a doubled shoot/root ion ratio relative to control conditions, indicating that salt exclusion might contribute to the higher tolerance of Steptoe. Correlation and path analysis demonstrated that shoot Cl? contents most strongly affected salt tolerance and suggest that both Na+ and Cl? contents are important for salinity stress tolerance in barley. We identified 11 chromosomal regions involved in the control of the variation observed for salt tolerance and various salt stress response traits, including Na+, Cl? and K+ contents in shoots. Two specific regions on chromosomes 2H and 3H were found controlling ion contents and salt tolerance, pointing to genes involved in ion homeostasis that contribute to salt tolerance.  相似文献

Salinity is one of the major abiotic constraints to agriculture. The physiological and molecular mechanisms of salt tolerance have been studied in plants for many years. The regulation of osmosis and ion homeostasis is crucial. A lot of important components involved in plant responses to salt stress have been identified. Among them, ion transporters and channels take an essential role in ion homeostasis, mainly for Na+, Cl-, and K+. Until now, many cation antiporters important for salt tolerance in plants have been characterized. Among them, the monovalent cation/proton antiporters (CPA) family is one of the most important families, including sodium proton exchangers (NHXs), K+-efflux antiporters (KEAs), and cation/H+ exchangers (CHXs). Here, the current knowledge of the plant CPA family in responses to salt stress is reviewed. The regulation mechanisms were also included and discussed.  相似文献

Potassium (K+) is an essential macronutrient in plants and a lack of K+ significantly reduces the potential for plant growth and development. By contrast, sodium (Na+), while beneficial to some extent, at high concentrations it disturbs and inhibits various physiological processes and plant growth. Due to their chemical similarities, some functions of K+ can be undertaken by Na+ but K+ homeostasis is severely affected by salt stress, on the other hand. Recent advances have highlighted the fascinating regulatory mechanisms of K+ and Na+ transport and signaling in plants. This review summarizes three major topics: (i) the transport mechanisms of K+ and Na+ from the soil to the shoot and to the cellular - compartments; (ii) the mechanisms through which plants sense and respond to K+ and Na+ availability; and (iii) the components involved in maintenance of K+/Na+ homeostasis in plants under salt stress.  相似文献

The SOS signal-transduction pathway is known to be important for ion homeostasis and salt tolerance in plants. However, there is a lack of in planta electrophysiological data about how the changes in signalling and ion transport activity are integrated at the cellular and tissue level. In this study, using the non-invasive ion flux MIFE technique, we compared net K+, H+ and Na+ fluxes from elongation and mature root zones of Arabidopsis wild type Columbia and sos mutants. Our results can be summarised as follows: (1) SOS mutations affect the function of the entire root, not just the root apex; (2) SOS signalling pathway is highly branched; (3) Na+ effects on SOS1 may by-pass the SOS2/SOS3 complex in the root apex; (4) SOS mutation affects H+ transport even in the absence of salt stress; (5) SOS1 mutation affects intracellular K+ homeostasis with a plasma membrane depolarisation-activated outward-rectifying K+ channel being a likely target; (6) H+ pump also may be a target of SOS signalling. We provide an improved model of SOS signalling and discuss physiological mechanisms underlying salt stress perception and signalling in plants. Our work shows that in planta studies are essential for understanding the functional genomics of plant salt tolerance.  相似文献

不同浓度NaCl盐处理下,AM真菌对羊草(Leymus chinensis)的侵染能力和对植物生长的影响,从植物形态和离子含量角度探讨了AM真菌提高羊草耐盐性的作用机理。结果表明,在高盐胁迫下,AM真菌显著降低了盐胁迫效应,提高了羊草生物量,菌根效应明显。菌根化羊草的根茎比显著增加,并且N、P浓度较高,Na~+和Cl~-离子浓度较低,表明AM真菌即促进羊草对营养元素的吸收,又减少了离子毒害。菌根化羊草的Ca~(2+)和K~+离子浓度,以及P/Na~+和K~+/Na~+比高于非菌根化羊草,表明AM真菌可通过调节渗透势以避免或减缓盐胁迫造成的生理缺水。随着盐胁迫的增加,菌根化羊草对磷的依赖性逐渐转换为对钾的依赖性。研究结果有助于揭示AM真菌提高植物耐盐能力的作用机理,并对应用菌根技术修复盐化草地具有理论指导意义。  相似文献

Salt is an important factor affecting the growth and development of soybean in saline soil. In this study, a novel soybean gene encoding a transporter (GmHKT1) was identified and its function analyzed using transgenic plants. GmHKT1 encoded a protein of 419 amino acids, with a potential molecular mass of 47.06 kDa and a predicted pI value of 8.59. Comparison of the genomic and cDNA sequences of GmHKT1 identified no intron. The deduced amino acid sequence of GmHKT1 showed 38–49% identity with other plant HKT‐like sequences. RT‐PCR analysis showed that the expression of GmHKT1 was upregulated by salt stress (150 mM NaCl) in roots and leaves but not in stems. Overexpression of GmHKT1 significantly enhanced the tolerance of transgenic tobacco plants to salt stress, compared with non‐transgenic plants. To investigate the role of GmHKT1 in K+ and Na+ transport, we compared K+ and Na+ accumulation in roots and shoots of wild‐type and transgenic tobacco plants. The results suggested that GmHKT1 is a transporter that affected K+ and Na+ transport in roots and shoots, and regulated Na+/K+ homeostasis in these organs. Our findings suggest that GmHKT1 plays an important role in response to salt stress and would be useful in engineering crop plants for enhanced tolerance to salt stress.  相似文献

In this research, the effect of different SA concentrations (0, 0.5, 1.0, 1.5, and 2.0 mM) on biological and grain yield as well as Na+, K+, Cl?, Ca2+, and Mg2+ distribution and accumulation in barley plants was examined under nonsaline (2 dS m?1) and saline (12 dS m?1) conditions in a three-year field study (2012–2015 growing seasons). Storage factor (SF) was defined as the concentration of an ion in the root, as a proportion of total uptake of that ion, to quantify ion partitioning between root and shoot. Salt stress decreased SF for K+, Ca2+, and Mg2+ and enhanced it for Na+ and Cl?, which led to reduce grain and biological yield. Nonetheless, foliar-applied SA in varying concentrations could lower some of these adverse effects on ion transport and accumulation. At the 2nd and 3rd years, unfavorable climatic conditions such as less precipitation and higher temperature intensified salt stress and decreased the alleviating impact of SA. Foliar application of SA at higher levels increased SF for Na+ and Cl? ions and decreased that for K+ indicating that SA helped barley plants keep more Na+ and Cl? and less K+ ions in the root system, which suggested the probable role of SA in altering ion transport within the plant in favor of salt stress tolerance. SF was found to be more correlated with grain yield under both nonsaline and saline conditions. Overall, SF might be considered as a potential criterion for salt tolerance in barley plants.  相似文献

为研究盐胁迫下小麦幼苗生长及Na+、K+的吸收和积累规律,以中国春、洲元9369和长武134等3种耐盐性不同小麦品种为材料,采用非损伤微测技术检测盐胁迫2 d后的根系K+离子流变化,并对植株体内的Na+、K+含量进行测定。结果表明:短期(2d)盐胁迫对小麦生长有抑制作用,且对根系的抑制大于地上部,耐盐品种下降幅度小于盐敏感品种。盐胁迫下,小麦根际的 K+大量外流,盐敏感品种中国春K+流速显著高于耐盐品种长武134,最高可达15倍。小麦幼苗地上部分和根系均表现为Na+积累增加,K+积累减少,Na+/K+比随盐浓度增加而上升。中国春限Na+能力显著低于长武134,Na+/K+则显著高于长武134。综上所述,盐胁迫下造成小麦组织器官中Na+/K+比上升的主要原因是根系K+大量外流和Na+的过量积累,耐盐性不同的小麦品种间差异显著,并认为根系对K+的保有能力可能是作物耐盐性评价的一个重要指标。  相似文献

Atriplex nummularia plants are able to grow well in the absence of significant amounts of Na+. Medium levels of salinity (100 mM NaCl or KCl) did not cause substantial inhibition of growth but increasing concentrations of salt induced a progressive decline in length and weight of the plants. This inhibition was significantly higher in KCl grown plants than in NaCl grown plants. In addition, although it has been proposed that both K+ and Na+ are involved in the osmotic adjustment of plants in response to high soil salinity, we show that Na+ ions contribute more efficiently than K+ ions to perform this function. Our results also indicate that most of the osmotic adjustment of the plant was due to the accumulation of inorganic ions. The strong inhibition of Rb+ transport caused by internal sodium suggests that this cation could be efficiently used by the plant and, as a consequence, the transport of other monovalent cations is down-regulated.  相似文献

Rice is a salt-sensitive crop whose productivity is strongly reduced by salinity around the world. Plants growing in saline soils are subjected to the toxicity of specific ions such as sodium, which damage cell organelles and disrupt metabolism. Plants have evolved biochemical and molecular mechanisms to cope with the negative effects of salinity. These include the regulation of genes with a role in the uptake, transport or compartmentation of Na+ and/or K+. Studies have shown that the arbuscular mycorrhizal (AM) symbiosis alleviates salt stress in several host plant species. However, despite the abundant literature showing mitigation of ionic imbalance by the AM symbiosis, the molecular mechanisms involved are barely explored. The objective of this study was to elucidate the effects of the AM symbiosis on the expression of several well-known rice transporters involved in Na+/K+ homeostasis and measure Na+ and K+ contents and their ratios in different plant tissues. Results showed that OsNHX3, OsSOS1, OsHKT2;1 and OsHKT1;5 genes were considerably upregulated in AM plants under saline conditions as compared to non-AM plants. Results suggest that the AM symbiosis favours Na+ extrusion from the cytoplasm, its sequestration into the vacuole, the unloading of Na+ from the xylem and its recirculation from photosynthetic organs to roots. As a result, there is a decrease of Na+ root-to-shoot distribution and an increase of Na+ accumulation in rice roots which seems to enhance the plant tolerance to salinity and allows AM rice plants to maintain their growing processes under salt conditions.  相似文献

Adaptation to high salinity is achieved by cellular ion homeostasis which involves regulation of toxic sodium ion (Na+) and Chloride ion (Cl) uptake, preventing the transport of these ions to the aerial parts of the plants and vacuolar sequestration of these toxic ions. Ion transporters have long been known to play roles in maintaining ion homeostasis. Na+ enters the cell through various voltage dependent selective and non-selective ion channels. High Na+ concentration in the plasma membrane is balanced either by uptake of potassium ion (K+) by various potassium importing channels, by salt exclusion mechanism or by sequestration of Na+ in the vacuoles. Therefore, the role of high-affinity potassium transporter, the salt overly sensitive pathway, the most well-defined Na+ exclusion pathway that exports Na+ from cell into xylem and tonoplast localized cation transporters that compartmentalizes Na+ in vacuoles need to be studied in detail and applied to make the plant adaptable to saline soil. Knowledge on the regulation of expression of these transporters by the hormones, microRNAs and other non-coding RNAs can be utilized to manipulate the ion transport. Here, we reviewed paradigm of the ion transporters in salt stress signalling pathways from the recent and past studies aiding transformation of basic knowledge into biotechnological applications to generate engineered salt stress tolerant crops.

High salinity is one of the most serious environmental stresses that limit crop growth. Expansins are cell wall proteins that regulate plant development and abiotic stress tolerance by mediating cell wall expansion. We studied the function of a wheat expansin gene, TaEXPA2, in salt stress tolerance by overexpressing it in tobacco. Overexpression of TaEXPA2 enhanced the salt stress tolerance of transgenic tobacco plants as indicated by the presence of higher germination rates, longer root length, more lateral roots, higher survival rates and more green leaves under salt stress than in the wild type (WT). Further, when leaf disks of WT plants were incubated in cell wall protein extracts from the transgenic tobacco plants, their chlorophyll content was higher under salt stress, and this improvement from TaEXPA2 overexpression in transgenic tobacco was inhibited by TaEXPA2 protein antibody. The water status of transgenic tobacco plants was improved, perhaps by the accumulation of osmolytes such as proline and soluble sugar. TaEXPA2‐overexpressing tobacco lines exhibited lower Na+ but higher K+ accumulation than WT plants. Antioxidant competence increased in the transgenic plants because of the increased activity of antioxidant enzymes. TaEXPA2 protein abundance in wheat was induced by NaCl, and ABA signaling was involved. Gene expression regulation was involved in the enhanced salt stress tolerance of the TaEXPA2 transgenic plants. Our results suggest that TaEXPA2 overexpression confers salt stress tolerance on the transgenic plants, and this is associated with improved water status, Na+/K+ homeostasis, and antioxidant competence. ABA signaling participates in TaEXPA2‐regulated salt stress tolerance.  相似文献

Salt stress is one of the most serious factors limiting the productivity of agricultural crops. Increasing evidence has demonstrated that vacuolar Na+/H+ antiporters play a crucial role in plant salt tolerance. In the present study, we expressed the Suaeda salsa vacuolar Na+/H+ antiporter SsNHX1 in transgenic rice to investigate whether this can increase the salt tolerance of rice, and to study how overexpression of this gene affected other salt-tolerant mechanisms. It was found that transgenic rice plants showed markedly enhanced tolerance to salt stress and to water deprivation compared with non-transgenic controls upon salt stress imposition under outdoor conditions. Measurements of ion levels indicated that K+, Ca2+ and Mg2+ contents were all higher in transgenic plants than in non-transformed controls. Furthermore, shoot V-ATPase hydrolytic activity was dramatically increased in transgenics compared to that of non-transformed controls under salt stress conditions. Physiological analysis also showed that the photosynthetic activity of the transformed plants was higher whereas the same plants had reduced reactive oxygen species generation. In addition, the soluble sugar content increased in the transgenics compared with that in non-transgenics. These results imply that up-regulation of a vacuolar Na+/H+ antiporter gene in transgenic rice might cause pleiotropic up-regulation of other salt-resistance-related mechanisms to improve salt tolerance.Fengyun Zhao and Zenglan Wang contributed equally to this work.  相似文献

The mechanisms of potato (Solanum tuberosum L.) plants’ tolerance to chloride salinity were investigated in cv. Lugovskoi regionalized in Russia. Regenerated plants were produced in vitro from apical meristem and grown on half-strength Murashige and Skoog medium (0.5 MS) using a hydroponic unit in controlled-climate conditions. At the age of six weeks, the plants were exposed to salt stress (50–150 mM NaCl, 7 days). Plant response to salt stress was estimated by growth parameters (fresh and dry biomass of the aboveground and underground parts of plants, linear dimensions of shoot and root, area of leaf surface, and number of stolons) and physiological characteristics (level of photosynthetic pigments, accumulation of sodium, potassium, and calcium ions in the aboveground and underground parts of plants, content of proline, activity of antioxidant enzymes, plant tissue hydration, osmotic potential, and POL). It was found that, in response to salinity, the plants of potato, cv. Lugovskoi, showed a considerable inhibition of growth processes, reduction in chlorophyll a content, and suppression of stolon formation, which points to a rather low salinity tolerance of the cultivar. At the same time, under weak or moderate salt stress, the plants preserved water homeostasis owing to effective osmoregulation, actively accumulated proline that acted as a stress protector, and showed hardly any signs of oxidative stress. It was assumed that low salt tolerance of this cultivar depends on the inability of its root system to retain sodium ions and ensure selective ion transport to the aboveground part of the plant and on inefficiency of the system of sodium ions’ removal from the cytoplasm of leaf cells and their compartmentalization in the central vacuole with the purpose of reducing their toxic effect. The obtained results may be useful for working out a technique of improving salt tolerance of this cultivar by the methods of molecular genetics.  相似文献
