Chick embryo heart cells in tissue culture actively oxidize [1-14C]palmitate to 14CO2. Fatty acid oxidation by cell monolayers was linear with time and increasing protein concentration. The addition of carnitine to the assay medium resulted in a 30–70% increase in the rate of fatty acid oxidation. The specific activity of palmitic acid oxidation did not change significantly with time in culture and was also the same in rapidly proliferating and density-inhibited cell cultures. Addition of unlabeled glucose to the assay medium resulted in a 50% decrease in 14CO2 production from [1-14C]palmitate. Conversely, palmitate had a similar sparing effect on [14C]glucose oxidation to 14CO2. Lactate production accounted for most of the glucose depleted from the medium and was not inhibited by the presence of palmitate in the assay. Thus, the sparing action of the fatty acids on glucose oxidation appears to be at the mitochondrial level. The results indicate that although chick heart cells in culture are primarily anaerobic, they can oxidize fatty acid actively.  相似文献

The effect of methionine supplementation on glycine and serine metabolism was studied in vitamin B-12-deficient rats which received only 0.2% methionine in the diet. In the perfused liver, incorporation of the C-2 of glycine to the C-3 of serine was increased by addition of methionine to the perfusate. The oxidation of [1-14C]glycine to 14CO2 was however depressed. Unlike methionine, glycine did not have any significant effect on the liver folate coenzyme distribution. Oxidation of [3-14C]serine to 14CO2 both in vivo and in perfused liver was increased by methionine. A major portion of the C-3 radioactivity however was recovered in glucose. Data presented indicate that the rate of oxidation of [2-14C]histidine to 14CO2 is more sensitive indicator of folate deficiency than the rate of oxidation of [3-14C] serine to 14CO2 although both are presumably tetrahydrofolate dependent.  相似文献

Abstract Radioactive acetyl groups and lipids are produced from dl -[5-14C]glutamate. Degradation studies indicate that approximately 90 per cent of the radioactivity is localized in the original carboxyl groups of the two carbon unit. Since these results are shown not to be due to a 14CO2 fixation, it is concluded that the oxoglutarate shunt as an acetyl group transport system is functional in brain. The highest ratio of fatty’acid activity/CO2 activity in this pathway is found in the newborn rat brain and steadily decreases with development. This pattern is observed with incubations of brain slices with labelled glutamate or citrate and is similar to the changes observed in the activity of the citrate cleavage enzyme with brain maturation. In contrast to the previous studies with liver preparations, the conversion of [2-14C]- and [5-14C]glutamate to fatty acids is relatively small. This is particularly true during the period of maximal lipid synthesis.  相似文献

A sensitive assay for arginase activity was developed using [guanidino-14C]arginine as substrate and measuring the production of 14CO2 from [14C]urea in the presence of urease. Arginase activity was measured in bovine lymphocytes after activation by Concanavalin A. The specific enzymatic activity of arginase doubled in 6 hours and increased nearly 4-fold by 24 hours after stimulation. It is suggested that the role of arginase in these cells is to provide ornithine as substrate for the synthesis of putrescine, precursor of the polyamines spermidine and spermine.  相似文献

A new assay for phospholipase C activity that uses alkaline phosphatase to convert phosphorylcholine to inorganic phosphate is described. The determination of inorganic phosphate is performed in the presence of phosphatidylcholine and protein after the addition of sodium dodecyl sulfate. Phospholipase C activity determined by this coupled enzyme assay agrees well with data obtained by extracting and measuring phosphoryl[14C]choline produced from phosphatidyl[methyl-14C]choline. The assay is sensitive to 1 nmol of phosphate, requires no removal of protein or phospholipid, and will work with a variety of phospholipid substrates. The assay is faster and more sensitive than previously published procedures. Stimulation of phospholipase C from Clostridium perfringens by ammonium sulfate is also reported.  相似文献

—The regulation of [14C]ACh synthesis was studied in rat striatal synaptosomes incubated in presence of various concentrations of Triton X-100, using [2-14C]pyruvate or [6-14C]glucose as precursors. The progressive rupture of the cytoplasmic and mitochondrial compartments induced by the non-ionic detergent was followed by studying the release, into the incubating medium, of lactate dehydrogenase and choline acetyltransferase (ChAc) and of fumarate hydratase, respectively. [3H]Choline uptake (1 μm ) was measured to determine the activity of the high affinity choline permease. 14CO2 formation from [2-14C]pyruvate was used as an index of the Krebs cycle activity. The rate of [14C]ACh synthesis from [2-14C] pyruvate was dependent on the Triton X-100 concentration; the ester formation decreased between 0·001% (v/v) and 0·010%, but increased again beyond this concentration of detergent. This last phenomenon was interpreted as the result of an extracellular synthesis of ACh involving pyruvate dehydrogenase and ChAc. At 0·002% Triton X-100 the 14CO2 formation was not affected, indicating a normal mitochondrial activity. The decrease of [14C]ACh synthesis observed up to this detergent concentration could be correlated to the decline of the highaffinity choline permease activity. In these experimental conditions, the ester synthesis could not be restored by the addition of large amounts of choline in the incubating medium suggesting that the molecules of choline must cross the high-affinity choline permease system in order to be acetylated. This could indicate a close association between the permease and choline acetyltransferase.  相似文献

A simple, rapid, and sensitive assay for phospholipase A2 in post-heparin plasma that uses commercially available l-α-dipalmitoyl-(2-[1-14C]palmitoyl) phosphatidylcholine is described. The incubation mixture, containing the enzyme substrate and products, is extracted with a two-phase heptane-isopropyl alcohol-aqueous sulfuric acid system, and the labeled fatty acid in the heptane phase is separated by the absorption of unreacted substrate on silicic acid. The heptane phase, containing the labeled fatty acid, is counted after the addition of commercial liquid scintillation fluid. Phospholipase A2 activity determined by this method agrees well with the data obtained by an earlier published method. The enzyme assay is faster and more sensitive than previously published procedures and is sensitive to levels as low as 1 nmol palmitate/h/200 μl of plasma. The enzyme activity could not be found in plasma obtained prior to the injection of heparin. Plasma phospholipase A2 is thermolabile, and the enzyme activity is enhanced by 2 mm sodium deoxycholate and calcium chloride, and inhibited by EDTA.  相似文献

In the absence of any other oxidizable substrate, the perfused rat heart oxidizes [1-14C]leucine to 14CO2 at a rapid rate and releases only small amounts of α-[1-14C]ketoisocaproate into the perfusion medium. The branched-chain α-keto acid dehydrogenase complex, assayed in extracts of mitochondria prepared from such perfused hearts, is very active. Under such perfusion conditions, dichloroacetate has almost no effect on [1-14C]leucine oxidation, α-[1-14C]ketoisocaproate release, or branched-chain α-keto acid dehydrogenase activity. Perfusion of the heart with some other oxidizable substrate, e.g., glucose, pyruvate, ketone bodies, or palmitate, results in an inhibition of [1-14C]leucine oxidation to 14CO2 and the release of large amounts of α-[1-14C]ketoisocaproate into the perfusion medium. The branched-chain α-keto acid dehydrogenase complex, assayed in extracts of mitochondria prepared from such hearts, is almost completely inactivated. The enzyme can be reactivated, however, by incubating the mitochondria at 30 °C without an oxidizable substrate. With hearts perfused with glucose or ketone bodies, dichloroacetate greatly increases [1-14C]leucine oxidation, decreases α-[1-14C]ketoisocaproate release into the perfusion medium, and activates the branched-chain α-keto acid dehydrogenase complex. Pyruvate may block dichloroacetate uptake because dichloroacetate neither stimulates [1-14C]leucine oxidation nor activates the branched-chain α-keto acid dehydrogenase complex of pyruvate-perfused hearts. It is suggested that leucine oxidation by heart is regulated by the activity of the branched-chain α-keto acid dehydrogenase complex which is subject to interconversion between active and inactive forms. Oxidizable substrates establish conditions which inactivate the enzyme. Dichloroacetate, known to activate the pyruvate dehydrogenase complex by inhibition of pyruvate dehydrogenase kinase, causes activation of the branched-chain α-keto acid dehydrogenase complex, suggesting the existence of a kinase for this complex.  相似文献

The production of 14CO2 from l-[1-14C]fucose and d-[1-14C]arabinose has been studied in five mammalian species.Cats, guinea pigs, mice, and rabbits respired about 22% of the label of l[1-14C]fucose or of d-[1-14C]arabinose within 6 h after intraperitoneal injection of the sugar. Rats respired only 1.5% of the l-fucose label and 5% of the d-arabinose label in the same time period.Liver homogenates from cat, guinea pig, and rabbit produced significantly more 14CO2 from l-[1-14C]fucose or d-[1-14C]arabinose than mouse or rat liver homogenates. Unlike those of the other species, guinea pig liver homogenates had very low l-fucose dehydrogenase activity.The results suggest that substantial catabolism of l-fucose and d-arabinose occurs in the tissues of some animal species. Investigators wishing to employ l-fucose as a tracer of glycoprotein metabolism must, therefore, ensure that the species that they employ does not metabolize l-fucose to products interfering with their studies.  相似文献

Tolbutamide partially inhibited the growth but increased the glycogen content of Tetrahymena pyriformis in logarithmically growing cultures. Tolbutamide slightly increased 14CO2 production from [1-14C] and [6-14HC] glucose and [2-14C] pyruvate, but had little effect on the oxidation of [1-14C] acetate when any of these substrates were added to the proteose-peptone medium in which the cells had been grown. Measurement of 14CO2 production from [1-14C] and [2-I4C]-glyoxylate showed that this substrate was primarily oxidized via the glyoxylate cycle, with little if any oxidation occurring via the peroxisomal glyoxylate oxidase. Addition of tolbutamide inhibited the glyoxylate cycle as indicated by a marked reduction in label appearing in CO2 and in glycogen from labeled acetate. In control cells, addition of acetate strongly inhibited the oxidation of [2-14C]-pyruvate whereas addition of pyruvate had little effect on the oxidation of [1-14C]-acetate. Acetate was more effective than pyruvate in preventing the growth inhibitory and glycogen-increasing effects of tolbutamide. The data suggest that one effect of tolbutamide may be to interfere with the transfer of isocitrate and acetyl CoA across mitochondrial membranes.  相似文献

This study focuses on the activity of the pentose-phosphate pathway and its relationship to de novo synthesis of fatty acids and cholesterol in oligodendrocyte-enriched glial cell cultures derived from 1-week old rat brain. The proportion of glucose that was metabolized along the pentose-phosphate pathway was estimated by measuring 14CO2 production from [1-14C]-, [2-14C]- and [6-14C]glucose, the utilization of glucose and the production of lactate. Incorporation of 14C from [14C]glucose and from [3-14C]acetoacetate into lipids was analysed. The pentose- phosphate pathway produced much more CO2 from glucose than the Krebs cycle, although it accounted for only a small part of the consumption of glucose (< 3%). The higher 14CO2 production from [2-14C]glucose than from [6-14C]glucose indicated that recycling of the products of the pentose-phosphate pathway takes place in these cells.Gradual inhibition of the pathway with increasing concentrations of 6-aminonicotinamide resulted in a parallel inhibition of the conversion of acetoacetate and of glucose into fatty acids and into cholesterol. Glycolysis was also strongly inhibited in the presence of 6-aminonicotinamide whereas the activity of the Krebs cycle was not affected.These results suggest that de novo synthesis of fatty acids and cholesterol by oligodendrocytes of neonatal rats is closely geared to the activity of the pentose-phosphate pathway in these cells.  相似文献

Extracts of Pseudomonas C grown on methanol as sole carbon and energy source contain a methanol dehydrogenase activity which can be coupled to phenazine methosulfate. This enzyme catalyzes two reactions namely the conversion of methanol to formaldehyde (phenazine methosulfate coupled) and the oxidation of formaldehyde to formate (2,6-dichloroindophenol-coupled). Activities of glutathione-dependent formaldehyde dehydrogenase (NAD+) and formate dehydrogenase (NAD+) were also detected in the extracts.The addition of d-ribulose 5-phosphate to the reaction mixtures caused a marked increase in the formaldehyde-dependent reduction of NAD+ or NADP+. In addition, the oxidation of [14C]formaldehyde to CO2, by extracts of Pseudomonas C, increased when d-ribulose 5-phosphate was present in the assay mixtures.The amount of radioactivity found in CO2, was 6.8-times higher when extracts of methanol-grown Pseudomona C were incubated for a short period of time with [1-14C]glucose 6-phosphate than with [U-14C]glucose 6-phosphate.These data, and the presence of high specific activities of hexulose phosphate synthase, phosphoglucoisomerase, glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase indicate that in methanol-grown Pseudomonas C, formaldehyde carbon is oxidized to CO2 both via a cyclic pathway which includes the enzymes mentioned and via formate as an oxidation intermediate, with the former predominant.  相似文献

A sensitive and rapid radiochemical micromethod is described for measuring the activity of acetyl-CoA hydrolase (EC 3.1.2.1). [1-14C]Acetyl-CoA is incubated with tissue homogenates; unhydrolyzed [1-14C]acetyl-CoA is separated from the radiolabeled product, [1-14C]acetate, by adsorption to charcoal. The soluble [1-14C]acetate is measured by liquid scintillation techniques. This procedure makes it possible to measure as little as 0.2 to 0.4 nmol acetate generated per assay.  相似文献

A new, simple, and accurate method for the sequential determination of the specific radioactivity of [1-14C]glutamic acid and [1-14C]glutamine is described. Using this method, radioactivity in H14CO3?, in [14C]glutamic acid, and in [14C]-glutamine can be readily determined on a single sample of blood plasma. Radioactivity is released as 14CO2 in a stepwise fashion, trapped in the center wells, and counted in a liquid scintillation counter. The applicability of the method is discussed.  相似文献

Summary We did this work to see if there is a correlation between lignin synthesis and the activity of the pentose phosphate pathway. Excision of the third internode of the stem of Coleus blumei Benth. followed by incubation on sucrose and indoleacetic acid led to extensive formation of tracheids. During this lignification we determined the activities of glucose-6-phosphate dehydrogenase and fructose-1,6-diphosphate aldolase, and the extent to which [1-14C]-,[3,4-14C]-, and [6-14C]glucose labelled CO2 and the major cellular components. The results indicate that the pentose phosphate pathway was active during lignification, and that the activity of this pathway relative to glycolysis increased at the onset of lignification. Explants of storage tissue of Helianthus tuberosus L. were cultured under conditions which caused extensive lignification. 14CO2 production from [1-14C]-, [3,4-14C]-, and [6-14C]glucose indicated activity of the pentose phosphate pathway during tracheid formation. In islets from adult rats injected with streptozotocin during the neonatal period, both a nonmetabolized analog of L-leucine and 3-phenylpyruvate augmented 14CO2 output from islets either prelabeled with L-[U-14C]glutamine or exposed to D-[2-14C]glucose and D-[6-14C]glucose in a manner qualitatively comparable to that found in islets from control rats. The islets of diabetic rats differed, however, from those of control rats by their unresponsiveness to both the L-leucine analog and a high concentration of D-glucose in terms of increasing 3HOH generation from [2-3H]glycerol, an impaired sparing action of the hexose upon 14CO2 output from islets prelabeled with [U-14C]palmitate, and, most importantly, by a decreased rate of D-[2-14C]glucose and D-[6-14C]glucose oxidation when either incubated at a high concentration of the hexose (16.7 mM) or stimulated by nonglucidic nutrient secretagogues at a low concentration of D-glucose (2.8 mM). In islet homogenates, the activity of glyceraldehyde phosphate dehydrogenase, glutamate decarboxylase, and NADP-malate dehydrogenase was lower in diabetic than control islets. Such was not the case for glutamatealanine transaminase, glutamate-aspartate transaminase, or glutamate dehydrogenase. The neonatal injection of streptozotocin thus affected, in the adult rats, the activity of several islet enzymes. Nevertheless, the metabolic data suggest that an impaired circulation in the glycerol phosphate shuttle, as observed in response to stimulation of the islets by either a high concentration of D-glucose or nonglucidic nutrient secretagogues, represents an essential determinant of the preferential impairment of glucose-induced insulin release in this model of non-insulin-dependent diabetes.  相似文献

Glycine decarboxylase has been successfully solubilized from pea (Pisum sativum) leaf mitochondria as an acetone powder. The enzyme was dependent on added dithiothreitol and pyridoxal phosphate for maximal activity. The enzyme preparation could catalyze the exchange of CO2 into the carboxyl carbon of glycine, the reverse of the glycine decarboxylase reaction by converting serine, NH4+, and CO2 into glycine, and 14CO2 release from [1-14C]glycine. The half-maximal concentrations for the glycine-bicarbonate exchange reaction were 1.7 millimolar glycine, 16 millimolar NaH14CO2, and 0.006 millimolar pyridoxal phosphate. The enzyme (glycine-bicarbonate exchange reaction) was active in the assay conditions for 1 hour and could be stored for over 1 month. The enzymic mechanism appeared similar to that reported for the enzyme from animals and bacteria but some quantitative differences were noted. These included the tenacity of binding to the mitochondrial membrane, the concentration of pyridoxal phosphate needed for maximum activity, the requirement for dithiothreitol for maximum activity, and the total amount of activity present. Now that this enzyme has been solubilized, a more detailed understanding of this important step in photorespiration should be possible.  相似文献

We demonstrate that allantoate is catabolized in soybean seedcoat extracts by an enzyme complex that has allantoate amidohydrolase and ureidoglycolate amidohydrolase activities. Soybean seedcoat extracts released 14CO2 from [ureido-14C]ureidoglycolate under conditions in which urease is not detectable. CO2 and glyoxylate are enzymically released in a one to one ratio indicating that ureidoglycolate amidohydrolase is the responsible activity. Ureidoglycolate amidohydrolase has a Km of 85 micromolar for ureidoglycolate. Glyoxylate and CO2 are enzymically released from allantoate at linear rates in a one to 2.3 ratio from 5 to 30 min. This ratio is consistent with the degradation of allantoate to two CO2 and one glyoxylate with approximately 23% of the allantoate degraded reacting with 2-mercaptoethanol to yield 2-hydroxyethylthio, 2′-ureido, acetate (RG Winkler, JC Polacco, DG Blevins, DD Randall 1985 Plant Physiol 79: 787-793). That [14C]urea production from [2,7-14C]allantoate is not detectable indicates that allantoate-dependent glyoxylate production is enzymic and not a result of nonenzymic hydrolysis of a ureido intermediate (nonenzymic hydrolysis releases urea). These results and those from intact tissue studies (RG Winkler DG Blevins, JC Polacco, DD Randall 1987 Plant Physiol 83: 585-591) suggest that soybeans have a second amidohydrolase reaction (ureidoglycolate amidohydrolase) that follows allantoate amidohydrolase in allantoate catabolism. The rate of 14CO2 release from [2,7-14C]allantoate is not reduced when the volume of the reaction mixture is increased, suggesting that the release of 14CO2 is not dependent on the accumulation of free intermediates. That [2,7-14C]allantoate dependent 14CO2 release is not proportionally diluted by unlabeled ureidoglycolate indicates that the reaction is carried out by an enzyme complex. This is the first report of ureidoglycolate amidohydrolase activity in any organism and the first in vitro demonstration in plants that the ureido-carbons of allantoate can be completely degraded to CO2 without a urea intermediate.  相似文献

Measurements have been made of the activities of enzymes of the pentose phosphate pathway, the glucuronate-xylulose pathway, hexokinase and phosphofructokinase in kidney of diabetic and normal rats. The activities of these enzymes keep pace with kidney growth, remaining constant per gram tissue but showing a marked increase on the basis of total activity per 100 g body wt. The formation of 14CO2 from [1-14C]glucose and [6-14C]glucose by kidney slices from diabetic rats was decreased to approximately half the control value; evidence was obtained for an equivalent dilution of the glucose 6-phosphate pool. Correction of the 14CO2 yields for the change in specific activity of glucose 6-phosphate yielded values consistent with the enzyme profile. Calculations from specific yields of 14CO2 provided evidence for an increased flux of glucose via the pentose phosphate pathway in the kidney in diabetes. The results are discussed in relation to kidney hypertrophy in diabetes and the requirement for ribose 5-phosphate and NADPH for biosynthetic reactions and in relation to the thickening of the basement membrane in diabetes. These results are in accord with the concept of glucose overutilization by non-insulin-requiring tissues.  相似文献

13C-nuciear magnetic resonance (NMR) spectroscopy was used to investigate the products of glycerol and acetate metabolism released by Leishmania braziliensis panamensis promastigotes and also to examine the interaction of each of these substrates with glucose or alanine. The NMR data were supplemented by measurements of the rates of oxygen consumption and substrate utilization, and of 14CO2 production from 14C-labeIed substrate. Cells incubated with [2-13C]glycerol released acetate, succinate and D-lactate in addition to CO2. Cells incubated with acetate released only CO2. More succinate C-2/C-3 than C-l/C-4 was released from both [2-13C]glycerol and [2-13C]glucose, indicating that succinate was formed predominantly by CO2 fixation followed by reverse flux through part of the Krebs cycle. Some redistribution of the position of labeling was also seen in alanine and pyruvate, suggesting cycling through pyruvate/oxaloacetate/phosphoenolpyruvate. Cells incubated with combinations of 2 substrates consumed oxygen at the same rate as cells incubated with 1 or no substrate, even though the total substrate utilization had increased. When promastigotes were incubated with both glycerol and glucose, the rate of glucose consumption was unchanged but glycerol consumption decreased about 50%, and the rate of 14CO2 production from [l,(3)-14C]glycerol decreased about 60%. Alanine did not affect the rates of consumption of glucose or glycerol, but decreased 14CO2 production from these substrates by increasing flow of label into alanine. Although glucose decreased alanine consumption by 70%, it increased the rate of 14CO2 production from [U-14C]- and [l-14C]alanine by about 20%. This is consistent with rapid equilibration of alanine with pyruvate derived from glucose and yet little decrease in the specific activity of the large alanine pool.  相似文献
