The involvement of present-day diploid bluegrass species in the formation of polyploid genomes was investigated using comparison of sequences of internal transcribed spacers ITS1 and ITS2, and the 5.8S rDNA sequence. It was demonstrated that highly polyploid New Zealand bluegrasses, P. cita (2n = 84; ca. 96 to 100), P. chathamica (2n = 112), and P. litorosa (2n = 263 to 266) formed separate highly supported clade together with tetraploids (2n = 28) P. intrusa, P. anceps, and P. trioides (Austrofestuca littoralis). Among the diploid species (2n = 14), the closest relatives of these species, as well as of the polyploid species of section Poa, are the genomes of Eurasian species P. remota, P. chaixcii (sect. Homalopoa), P densa (Bolbophorum), and P. sibirica (sect. Macropoa). Nuclear genomes of polyploid Stenopoa, Tichopoa, Oreinos, and Secundae are definitely related to the genome of Arctic species P. pseudabbreviata (sect. Abbreviatae). On the contrary, judging by the genes for nuclear 45S rRNA, genomes of diploid P. trivialis (sect. Pandemos), P. annua, and P. supina (sect. Ochlopoa both) are only remotely related to the genomes of highly polyploid species (distances p between them and other bluegrass species from different sections of subgenus Poa constitute 6-10% and 11-15%, respectively). The conclusion on the relationships between highly polyploid and diploid bluegrass species was tested using analysis of synapomorphic mutations in the 5.8S rRNA gene. It was demonstrated that genomes of Poa eminens (2n = 42) and P. schischkinii (2n = 70) (sect. Arctopoa both) were noticeably different in ITS regions from the genomes of the members of the type subgenus Poa. A comparison of the Arctopoa ITS regions showed that the differences between them constituted only 0.2%. At the same time, p distances between the Arctopoa ITS and those from the species belonging to other sections of the genus Poa varied from 5 to 14%. South American species P chonotica (sect. Andinae) (=Ncoraepoa chonotica) (2n = 42) was found to be related to Arctagrostis, Festucella, and Hookerochloa, being at the same time quite distant from the other species of the genus Poa. Polymorphic in chromosome number highly polyploid species of Northern Hemisphere, P. arctica (2n = 42 to 106), P. turneri (2n = 42, 63 to 64), and P. smirnovii (2n = 42, 70) (sect. Malacanthae) are relative to a large group of tetraploid (2n = 28) endemic bluegrass species from New Zealand and sub-Antarctic islands (P. novae-zelandiae and allied species).  相似文献

Fifty-eight quantitative and qualitative exomorphological characters were recorded on 48 herbarium specimens belonging to rhizomatous species belonging to the genus Poa. These were treated as OTUs and analysed, together with the data from caespitose species treated in a previous paper, using cluster analysis, principal component analyses and stepwise discriminant analyses. The following hypotheses are proposed and discussed. (1) There is no evidence supporting the hypothesis that growth habit characterizes supraspecific entities. (2) P. trivialis, P. secunda, P. subenervis and probably P. compressa seem to be well defined species; those remaining are not discriminated through phenetic analysis. (3) There are no phenetic gaps between the rhizomatous species included in the P. oligeria—P. yaganica-P. chrysantha complex and the caespitose P. hachadoensis it is not clear that this single character supports its segregation. (4) The obscurity of the pattern of the species related to P. pratensis-P. nemoralis is probably explained by the capability of P. pratensis to introgress with taxonomically distant species and by the possibility of seasonal intraindividual variation of some characters. (5) The geographical origin of some species postulated as introduced seems to be doubtful.  相似文献

A segregating second locus, PgiC2, for the enzyme phosphoglucose isomerase (PGIC) is found in the grass sheep's fescue, Festuca ovina. We have earlier reported that a phylogenetic analysis indicates that PgiC2 has been horizontally transferred from the reproductively separated grass genus Poa. Here we extend our analysis to include intron and exon information on 27 PgiC sequences from 18 species representing five genera, and confirm our earlier finding. The origin of PgiC2 can be traced to a group of closely interrelated, polyploid and partially asexual Poa species. The sequence most similar to PgiC2 is found in Poa palustris with a divergence, based on synonymous substitutions, of only 0.67%. This value suggests that the transfer took place less than 600,000 years ago (late Pleistocene), at a time when most extant Poa and Festuca species already existed.  相似文献

BACKGROUND AND AIMS: To improve the management of grass communities, early plant development was compared in three species with contrasting growth forms, a caespitose (Lolium perenne), a rhizomatous (Poa pratensis) and a caespitose-stoloniferous species (Agrostis stolonifera). METHODS: Isolated seedlings were grown in a glasshouse without trophic constraints for 37 d (761 degrees Cd). The appearance of leaves and their location on tillers were recorded. Leaf appearance rate (LAR) on the tillers and site-filling were calculated. Tillering was modelled based on the assumption that tiller number increases with the number of leaves produced on the seedling main stem. Above- and below-ground parts were harvested to compare biomass. KEY RESULTS: Lolium perenne and A. stolonifera expressed similar bunch-type developments. However, root biomass was approx. 30 % lower in A. stolonifera than in L. perenne. Poa pratensis was rhizomatous. Nevertheless, the ratio of above-ground : below-ground biomass of P. pratensis was similar to that of L. perenne. LAR was approximately equal to 0.30 leaf d(-1) in L. perenne, and on the main stem and first primary tillers of A. stolonifera. LAR on the other tillers of A. stolonifera was 30 % higher than on L. perenne. For P. pratensis, LAR was 30 % lower than on L. perenne, but the interval between the appearance of two successive shoots from rhizomes was 30 % higher than the interval between two successive leaf stages on the main stem. Above-ground parts of P. pratensis first grew slower than in the other species to the benefit of the rhizomes, whose development enhanced tiller production. CONCLUSIONS: Lolium perenne had the fastest tiller production at the earliest stages of seedling development. Agrostis stolonifera and P. pratensis compensated almost completely for the delay due to higher LAR on tillers or ramets compared with L. perenne. We investigated whether leaf dark respiration (nonphotorespiratory mitochondrial CO2 release) is inhibited by light in several Poa species, and whether differences in light inhibition between the species are related to differences in the rate of leaf net photosynthesis. Four lowland (Poa annua L., Poa compressa L., Poa pratensis L., and Poa trivialis L.), one subalpine (Poa alpina L.), and two alpine (Poa costiniana Vick. and Poa fawcettiae Vick.) Poa species differing in whole plant relative growth rates were grown under identical controlled conditions. Nonphotorespiratory mitochondrial CO2 release in the light (Rd) was estimated according to the Laisk method. Photosynthesis was measured at ambient CO2 partial pressure (35 Pa) and 500 [mu]mol photons m-2 s-1. The rate of photosynthesis per unit leaf mass was positively correlated with the relative growth rate, with the slow-growing alpine Poa species exhibiting the lowest photosynthetic rates. Rates of both Rd and respiration in darkness were also substantially lower in the alpine species. Nonphotorespiratory CO2 release in darkness was higher than Rd in all species. However, despite some variation between the species in the level of light inhibition of respiration, no relationship was observed between the level of inhibition and the rate of photosynthesis. Similarly, the level of inhibition was not correlated with the relative growth rate. Our results support the suggestion that rates of leaf respiration in the light are closely associated with rates in darkness.  相似文献

In this study, sequence data from the inert nuclear region ITS1‐5.8S rDNA‐ITS2 and the chloroplast region trnL–F, as well as a few morphological characters, are analysed to the relationships among known annual Poa (bluegrasses). It is shown that all taxa from the Poa annua aggregate distinguished by lemma characters and growth form have identical ITS and trnL–trnF sequences, all ITS sequences of this aggregate are the same as thethose of P. supina, and all trnL–trnF sequences are homologous with those of P. infirma. Furthermore, no differences were found between unusual morphotypes of P. supina with short spinules on their panicle branches and typical plants, but Siberian samples were found to have a slightly differentiated trnL–trnF region. These results suggest a hybrid origin of the Asian annual bluegrasses. Their maternal genome is close to that of P. sect. Homalopoa, but their ITS sequences are different. Some ITS sequences from annual Asian bluegrasses are resolved among representatives of P. sect. Stenopoa while for other (morphologically closely similar) species they fall in a clade with representatives of P. sect. Malacanthae. The latter group is distant from P. sect. Ochlopoa and is better treated as a separate section, viz P. sect. Acroleucae. The American annual bluegrasses are heterogeneous and also rather distant from P. sect. Ochlopoa. Poa chapmaniana, a species with cleistogamic flowers, is nested among the basal Subantarctic sections, far away from the taxa with which it has previously been considered related. It is indeed closer to P. sect. Ochlopoa than to other annual American bluegrasses. Thus, the studied annual species in fact belong to four independent evolutionary lines (or six including the separate genus Eremopoa and the Turkish Poa jubata), one of which, Acroleucae, has gone through three reticulation events. As in previous studies, our analysis did not support the generic status of P. sect. Ochlopoa.  相似文献

We describe a new Epichlo? species found in symbiosis with Poa pratensis ssp. pratensis in Liyang, China. Stromata characteristic of Epichlo? spp. were present on some of the reproductive tillers of individual host grasses. Only three of the 98 stromata observed on field plants became orange and produced perithecia. Phylogenetic analyses based on sequences of tubB and tefA indicated that this Epichlo? sp. was an interspecific hybrid related to both E. yangzii and members in the E. typhina complex clade (ETC). Allele-1 of tefA and tubB grouped in the E. bromicola/E. yangzii clade; allele-2 of these two genes clustered in a distinct subclade in the ETC. This is the first report of an Epichlo? species that has interspecific hybrid origins. We propose the name Epichlo? liyangensis Z. Wang, Y. Kang et H. Miao, sp. nov. for this species.  相似文献

Poa pratensis L. is a forage and turf grass species well adapted to a wide range of mesic to moist habitats. Due to its genome complexity little is known regarding evolution, genome composition and intraspecific phylogenetic relationships of this species. In the present study we investigated the morphological and genetic diversity of 33 P. pratensis accessions from 23 different countries using both nuclear and chloroplast molecular markers as well as flow cytometry of somatic tissues. This with the aim of shedding light on the genetic diversity and phylogenetic relationships of the collection that includes both cultivated and wild materials. Morphological characterization showed that the most relevant traits able to distinguish cultivated from wild forms were spring growth habit and leaf colour. The genome size analysis revealed high variability both within and between accessions in both wild and cultivated materials. The sequence analysis of the trnL-F chloroplast region revealed a low polymorphism level that could be the result of the complex mode of reproduction of this species. In addition, a strong reduction of chloroplast SSR variability was detected in cultivated materials, where only two alleles were conserved out of the four present in wild accessions. Contrarily, at nuclear level, high variability exist in the collection where the analysis of 11 SSR loci allowed the detection of a total of 91 different alleles. A Bayesian analysis performed on nuclear SSR data revealed that studied materials belong to two main clusters. While wild materials are equally represented in both clusters, the domesticated forms are mostly belonging to cluster P2 which is characterized by lower genetic diversity compared to the cluster P1. In the Neighbour Joining tree no clear distinction was found between accessions with the exception of those from China and Mongolia that were clearly separated from all the others.  相似文献

It has been suggested that some perennial grasses secrete phytosiderophores in response to iron (Fe) deficiency, but the compounds have not been identified. Here, we identified and characterized the phytosiderophores secreted by two perennial grasses, Lolium perenne cv. Tove and Poa pratensis cv. Baron. Root exudates were collected from the roots of Fe-deficient grasses and then purified with various chromatographies. The structure of the purified compounds was determined using both nuclear magnetic resonance and fast atom bombardment mass spectrometry. Both species secreted phytosiderophores in response to Fe deficiency, and the amount of phytosiderophores secreted increased with the development of Fe deficiency. The type of phytosiderophores secreted differed with plant species; L. perenne cv. Tove secreted 3-epihydroxy-2'-deoxymugineic acid (epiHDMA), 2'-deoxymugineic acid (DMA) and an unknown compound, whereas P. pratensis cv. Baron secreted DMA, avenic acid A (AVA) and an unknown compound. Purification and subsequent analysis with nuclear magnetic resonance and mass led to identification of the two novel phytosiderophores; 3-hydroxy-2'-deoxymugineic acid (HDMA) from L. perenne, and 2'-hydroxyavenic acid A (HAVA) from P. pratensis. Both novel phytosiderophores have similar chelating activity to known phytosiderophores.  相似文献

In sheep's fescue, Festuca ovina, genes coding for the cytosolic enzyme phosphoglucose isomerase, PGIC, are not only found at the standard locus, PgiC1, but also at a segregating second locus, PgiC2. We have used PCR-based sequencing to characterize the molecular structure and evolution of five PgiC1 and three PgiC2 alleles in F. ovina. The three PgiC2 alleles were complex in that they carried two gene copies: either two active genes or one active and one pseudogene. All the PgiC2 sequences were very similar to each other but highly diverged from the five PgiC1 sequences. We also sequenced PgiC genes from several other grass species. Phylogenetic analysis of these sequences indicates that PgiC2 has introgressed into F. ovina from the distant genus Poa. Such an introgression may, for example, follow from a non-standard fertilization with more than one pollen grain, or a direct horizontal gene transfer mediated by a plant virus.  相似文献

Black cutworm, Agrotis ipsilon (Hufnagel) (Lepidoptera: Noctuidae), feeding bioassays were conducted on young and mature turfgrass species to determine their potential resistance. Measures of resistance included larval weight, survival rate, instar development, pupal weight, duration of pupation, and days to pupation and adult emergence. Black cutworm reared on Poa pratensis 'Midnight', Poa arachnigera 'Tejas', and Poa pratensis X Poa arachnigera 'Reveille' exhibited slower development, lower weight, and higher overall mortality than those fed upon other turfgrasses tested. Larvae reared on Reveille did not survive to pupation and all died within 14 d. Black cutworm larvae reared on Midnight died within 17 d in trial 1 but attained pupation in trial 2. However, development of black cutworm larvae was slower on Poa pratensis Midnight compared with other susceptible turfgrass species such as Agrostis stolonifera 'Penncross', Poa annua ('DW194', 'Q98-4-6', and 'Q98-6-18'), Lolium perenne, and Poa supina 'Supranova'. Generally, larval performance on young plant tissues was better than on mature plant tissues. Larvae reared on P. pratensis 'Midnight' exhibited the most distinctive difference on young versus mature plant tissue. These results suggest that plant age may play an important role in turfgrass susceptibility and resistance.  相似文献

The phylogeny of diploid Hordeum species has been studied using both chloroplast and nuclear gene sequences. However, the studies of different nuclear datasets of Hordeum species often arrived at similar conclusions, whereas the studies of different chloroplast DNA data generally resulted in inconsistent conclusions. Although the monophyly of the genus is well supported by both morphological and molecular data, the intrageneric phylogeny is still a matter of controversy. To better understand the evolutionary history of Hordeum species, two chloroplast gene loci (trnD-trnT intergenic spacer and rps16 gene) and one nuclear marker (thioreoxin-like gene (HTL)) were used to explore the phylogeny of Hordeum species. Two obviously different types of trnD-trnT sequences were observed, with an approximately 210 base pair difference between these two types: one for American species, another for Eurasian species. The trnD-trnT data generally separated the diploid Hordeum species into Eurasian and American clades, with the exception of Hordeum marinum subsp. gussoneanum. The rps16 data also grouped most American species together and suggested that Hordeum flexuosum has a different plastid type from the remaining American species. The nuclear gene HTL data clearly divided Hordeum species into two clades: the Xu+H genome clade and the Xa+I genome clade. Within clades, H genome species were well separated from the Xu species, and the I genome species were well separated from the Xa genome species. The incongruence between chloroplast and nuclear datasets was found and discussed.  相似文献

Detached 3-week-old leaves of Agrostis palustris, Lolium perenne, Poa annua, and Poa pratensis were inoculated with conidial suspensions of two isolates of Colletotrichum graminicola obtained from A. palustris. Inoculated leaves were incubated at 23 degrees C under high relative humidity (>95%). The infection process was investigated by light microscopy from 2 to 168 h after inoculation (AI). Spore germination was observed within 2 h AI, appressoria within 6 h AI, and penetration pores within 8 h AI on all four hosts. Infection hyphae were observed inside epidermal cells within 24 h AI on all four hosts, but significantly greater infection was observed in A. palustris and P. annua than in L. perenne or P. pratensis at both 96 and 120 h AI. Acervuli appeared on leaves of A. palustris at 72 h AI and on L. perenne at 96 h AI but were not found on either P. annua or P. pratensis during the first 168 h AI. The infection process was similar to that reported for C. graminicola from other hosts; however, disease development of the two isolates of C. graminicola from A. palustris was faster or fungal growth more extensive on detached leaf tissue of A. palustris than on other turfgrass species tested.  相似文献

Morphometric and isozyme data were used to test the hypothesis that the pseudoviviparous Poa jemtlandica has originated as a hybrid between the sexual P flexuosa and the facultative apomict P alpina. Plants were sampled from three localities in southern Norway. In addition, some Icelandic plants were included. Both morphometric and isozyme data clearly support the hybrid hypothesis. Poa jemtlandica was morphologically intermediate between its supposed parents in several individual characters as well as in a discriminant analysis. Seven enzyme systems were analysed. Eight areas of activity were interpreted as "loci", each of which certainly represented several duplicated genes in these plants which all are polyploid. A total of 27 different multilocus phenotypes was observed: ten phenotypes in P alpina var. vivipara, eight phenotypes in I. alpina var. alpina, seven phenotypes in P jemtlandica, and three phenotypes in P flexuosa. The phenotype of Icelandic P jemtlandica was identical to that of P. jemtlandica from one of the Norwegian localities. Additive electrophoretic patterns were observed for P jemtlandica at four individual "loci". At the remaining "loci" some phenotypes of P. jemtlandica were most similar to P. flexuosa, others to P alpina. The data also suggest that P jemtlandica has originated several times, but there is no indication of frequent or recent hybridization where the species coccur today.  相似文献

The structure of the abaxial epidermis of the ligules of mid- and upper-culm leaves of the grasses Poa annua, P. nemoralis, P. pratensis and P. trivialis as seen by light microscopy is described. Amongst the structural elements found were long cells, short cells and unicellular prickle hairs over the surface, with unicellular hairs and papillate cells on the free edges of the ligules. Ligules of upper-culm leaves of some plants of P. trivialis L. contained vascular tissue with associated green (chlorenchyma) regions and stomata-like structures. The value of ligule characters in grass identification and classification is briefly discussed.  相似文献

Flowering in Poa pratensis L. cv. Holt and Bromus inermis Leyss. cv. Manchar requires exposure to short days (SD) for primary induction to occur, followed by long days (LD) to allow the inflorescence to develop. Weekly sprays with gibberellic acid (GA3) during primary induction inhibited flower initiation in both P. pratensis and B. inermis . With 10−4 M GA3 flowering of P. pratensis was suppressed even after an induction period of 10 weeks. Since both GA3 and non-inductive LD conditions greatly stimulate leaf elongation, the degree of primary induction was closely negatively correlated with plant height (leaf sheath and blade length) at the end of the induction period. GA3 application or the interpolation of LD during SD induction were most inhibitory during the later middle part of the SD period, whereas they were stimulatory near the beginning or immediately before the SD period. We suggest that changes in the portfolio or levels of endogenous gibberellins mediate photoperiodic control of growth and floral initiation in these plants. However, GA3 sprays could not substitute for LD in causing heading and culm elongation in SD induced plants of the two species. The results are discussed in the light of results with other plants with dual floral induction requirements.  相似文献
