We describe the structure of the spleen of the African lungfish Protopterus annectens in freshwater conditions, and after 6?months of aestivation. The spleen is formed by cortical tissue that surrounds the splenic parenchyma. The cortex is a reticulum that contains two types of granulocytes, developing and mature plasma cells, and melanomacrophage centres (MMCs). The parenchyma is divided into lobules that show a subcapsular sinus and areas of red pulp and white pulp. Red pulp contains vascular sinuses and atypical cords formed by delicate trabeculae. White pulp also contains vascular sinuses and cords. Structural data indicate that red pulp is involved in erythropoiesis, destruction of effete erythrocytes, and plasma cell differentiation. White pulp appears to be involved in the production of immune responses. Macrophages and sinus endothelial cells constitute the reticulo-endothelial system of the spleen. After aestivation, the number of MMCs increases, and spleen tissue is infiltrated by lymphocytes, granulocytes, and monocytes. Also, white pulp is reduced, and sinus endothelial cells undergo vacuolar degeneration. Lungfish spleen shares structural characteristics with secondary lymphoid organs of both ectothermic and endothermic vertebrates, but appears to have evolved in unique ways.  相似文献

Experimental infection of hamsters with Leishmania donovani caused visceral leishmaniasis in which hematological changes occurred. The infected hamsters were anemic and reticulocyte counts were high. No significant change in the serum erythropoietin level was noted. Red cell membrane Na(+)-K(+)-ATPase and acetylcholinesterase activities increased. Osmotic fragility of the erythrocytes from infected animals increased. The level of 2,3-diphosphoglycerate of the red cells increased with the degree of anemia.  相似文献

Complete and differential blood counts were conducted on 23 male squirrel monkeys (Saimiri sciureus) of Bolivian origin. The sample included 9 laboratory-bred and 14 wild-caught adult monkeys weighing between 600 and 1,500 g. The means of total white blood cell (WBC) counts, total hemoglobin and hematocrits of laboratory-bred animals were found to be significantly different from those of wild-caught monkeys (p less than 0.01). Other hematologic parameters showed no significant differences between the two experimental groups. 3 of 14 (21.0%) wild-caught animals each had 1 reactive lymphocyte and 7% of the same group had 10 nucleated erythrocytes per 100 WBC. No reactive lymphocytes or nucleated erythrocytes were observed in laboratory-bred animals. The importance of these baseline data and the basis for the differences observed between laboratory and wild-caught monkeys are discussed within the context of experimental studies involving primates in which hematologic parameters are valuable.  相似文献

Using 3H-thymidine autordiography, we studied the cellular proliferation of the spleen of rats after cryolesions in liver, kidney, spleen, and stomach.In the germinal centers at first, a dissociation develops, followed by a hyperplasia with high labeling indices of the germinal center cells with a maximum between the second and third postoperative day. In the surrounding lymphatic mantle zone of the white pulp, as well as in the marginal zone, an increased labeling index of the cells can be observed between the first and second day. The highest percentages of labeled cells in the red pulp are seen on the fifth postoperative day.These cell kinetic results correspond very well with those after antigenic stimulation, for instance, after intravenous injection of sheep erythrocytes. Therefore, these findings suggest that an immunologic reaction occurs in the spleen after cryolesions on parenchymal organs.  相似文献

Proliferative and migratory changes of lymphoid cells in the spleen were observed in turtles stimulated with KLH and maintained at 30°C. Small foci of pyroninophilic lymphoblasts were first seen in the white pulp at about day 5. Progressive enlargement of these centers continued and peaked by days 8-12. By days 15-20 the white pulp returned to a normal (unimmunized) state, while the number of pyroninophilic cells, primarily plasma cells, increased markedly in the red pulp. At days 22-25, the number of plasma cells returned to normal levels and the spleen appeared normal for the remainder of the 60 day observation period. These events suggest that at 30°C,-the turtle is capable of a strong and prompt proliferative response in the white pulp sheaths, followed by migration and differentiation of lymphoblasts into plasma cells, n the red pulp., Observations of pyroninophilic cells in sinuses, venules and veins of the spleen and a concomitant depletion of cells in red pulp, further suggest a migration from the spleen to other sites. Following a second antigenic challenge, at day 60, no significant histological changes were observed at 30°C. Nor were any changes observed following primary or secondary antigenic challenge, in animals maintained at 10°C. These findings are discussed with regard to immunological memory and low temperature immunosuppression in ectothermic vertebrates.  相似文献

As little comprehensive baseline data are available on age-related haematological changes in genetically-defined rat strains, the haematology of female F344 rats is described in animals sampled at 2, 4, 8, 20, 66 and 121 weeks of age. Values for Hb, RBC and PCV increased from 2 weeks of age to reach adult levels at 8 weeks, whereas MCV, MCH and reticulocyte counts were high initially but decreased to reach the adult range at 8 weeks. Between 66 and 121 weeks, reticulocyte counts were significantly increased and values for MCHC significantly decreased. Lymphocytes were the predominant white cell type in each age group. The absolute numbers of neutrophils and lymphocytes showed slight variations between 2 and 66 weeks and both cell types increased significantly between 66 and 121 weeks. Platelet counts showed no overall age-related trends. Fibrinogen values increased from 2 weeks of age to reach the adult level at 8 weeks. One animal of the 14 sampled at 121 weeks showed changes in the blood, liver and spleen consistent with a diagnosis of lymphoid leukaemia.  相似文献

The spleen of sea bass (Dicentrarchus labrax) is composed mainly of red pulp, whereas the white pulp is poorly developed. The red pulp consists of clear reticular cells intermingled with blood cells, sinusoids, and melanomacrophage centers (MMCs). The MMCs are enclosed by an interrupted connective tissue capsule and show some areas in continuity with the adjacent pulp. The MMCs are formed by the association of free macrophages that have phagocytosed some blood cells. Sparse white pulp is diffuse, forming a cuff around the pulp arteries and MMCs, or occurring in small groups between the splenic cords. A longitudinal artery and vein, lying side by side, extend the length of the spleen. Frequently the capillaries are surrounded by a sheath of macrophages or ellipsoids. These macrophages may contain erythrocytes in varying degrees of degradation. Lymphopoiesis and plasmapoiesis occur in the sparse lymphold areas. Abundant plasma cell groups may indicate the presence of antibody production.  相似文献

  • 1 In studies of birds and their pathogens, spleen size has frequently been used to make inferences about immune system strength. However, the use of spleen size in mammals is more complicated because, in addition to having an immune function, the mammalian spleen is also a reservoir for red blood cells.
  • 2 To assess the reliability of mammalian spleen mass as an indicator of immune activity, we quantified the white and red pulp mass by histology of spleens from shot red deer Cervus elaphus. We then analysed the relationships among spleen mass, the amounts of white and red pulp, and the deer's body condition relative to faecal counts of the nematode parasite Elaphostrongylus cervi.
  • 3 White and red pulp mass were positively correlated so that an increase in spleen mass was a positive function of both components of the spleen. In male deer, which had significantly lower body condition and higher parasite loads than females, parasite counts were negatively correlated with spleen mass, white pulp mass, and red pulp mass.
  • 4 Our findings suggest that (i) spleen mass in shot red deer is a reliable measure of white and red pulp content; and (ii) when looking at the red deer life history, which is greatly influenced by sex of the deer, splenic mass and white pulp mass could be used as reflections of immune system strength.
  • 5 Future studies of mammalian spleens can contribute to the understanding of evolved strategies of immune response investment in mammals. However, determination of the white and red pulp spleen components using various sampling methods must be made prior to their application.

ABSTRACT. A survey for Babesia microti in rodents was conducted at six sites within Grand Teton National Park, Wyoming. Blood and spleen smears, hematocrits, and reticulocyte counts were made on all of the animals to evaluate parameters for the diagnosis of babesiosis. Ticks were removed for identification. Of 257 Microtus montanus , 103 were infected with B. microti. In addition, five of 12 Microtus pennsylvanicus and one of three Arvicola richardsoni were parasitized by B. microti. Peromyscus maniculatus (n = 40) were not infected. Concurrent infections by Hepatozoon sp., Trypanosoma sp., and the bacterium, Grahamella sp., were noted in blood smears from a number of M. montanus. Splenomegaly and reticulocytosis were significant parameters associated with babesiosis while decreased hematocrit was not. Ticks removed from the voles were identified as Ixodes eastoni and were the probable vectors of the B. microti.  相似文献

Hematologic reference values have been established for captive adult cotton-top tamarins (Saguinus oedipus oedipus) by carrying out full blood counts and fibrinogen estimation on 43 clinically normal animals. Females were shown to have significantly lower hemoglobin levels, red cell counts and packed cell volumes, and higher reticulocyte counts than males. The reference values were used to identify abnormal changes in the blood of 13 clinical cases. Marked neutrophilia was found in animals with localized bacterial infections, and a degenerative left shift was recorded in an individual with streptococcal septicemia. Three cases of unexplained progressive muscle wasting showed Heinz body anemia and abnormal white cell changes. These hematologic responses have been compared where possible with those recorded in other species of Callithricidae.  相似文献

The influence of retinyl acetate, retinoic acid and its esterified cis- and trans-derivatives, retinoids C15 and C20 on erythrocytes, liver macrophages and white pulp of the spleen was studied in the experiments on mice of different strains. It was found that administration of these compounds leads to the damage of the blood cells, increase in the number of liver macrophages and the share of white pulp in the spleen and stimulation of the local response to tuberculin. The interdependence of changes in the blood and adjuvant is suggested.  相似文献

The hematology of the common marmoset (Callithrix jacchus) was investigated to provide reference values for the normal animal. Red blood cell counts, white blood cell counts, hemoglobins, mean cell volumes and differential white blood cell mean values were determined for Texas A&M colony-born animals and those obtained from the wild. The analyses were completed on 31 animals, three times each, for a total of 93 analyses, which included 16 colony-born and 15 wild-born marmosets. The hematological mean values found for marmosets were similar to those mean values reported for humans. The ranges for the hematology values were much narrower in the colony-born marmosets, and the average white blood cell count was significantly lower in these animals. This indicated that it is possible to produce and maintain a more uniform animal in the colony environment and, consequently, these animals are more suitable than wild-born animals for use as models for human research.  相似文献

SYNOPSIS. The presence of anemia that often seems excessive for the amount of parasitemia in Plasmodium berghei infections had led to the suggestion that autoimmunity might be in part responsible for the anemia. In another erythrocytic infection, Anaplasma marginale of cattle, the association of erythrophagocytosis, autohemagglutination and anemia with infection has led to the suggestion that autoimmunization may occur in anaplasmosis. The possibility that similar findings might be present in P. berghei infections of rats has been investigated. Groups of rats infected with P. berghei were examined at 2–3 day intervals during the course of infection. Red blood cell counts, hematocrit values and percentages of parasitized erythrocytes were determined. The rats were bled at intervals and the sera tested for agglutinins for trypsinized rat erythrocytes. Other infected rats were killed, and their spleens and bone marrow were examined for evidence of erythrophagocytosis. Parasitemia reached a peak on the 9th day of infection and became subpatent by the 14th. The greatest depression in erythrocyte numbers occurred on the 11th day, and the counts remained below normal until the 23rd day. Phagocytized erythrocytes, predominantly uninfected, were found in the phagocytes of the spleen and bone marrow from the 5th through the 21st day. Agglutinins for trypsinized normal rat erythrocytes were present in the sera of the rats in titers as high as 1:64 from the 5th through 14th day of infection. Lower agglutinin titers (1:8) were found from time to time in sera of rats made anemic by repeated bleedings. It is not clear whether these agglutinins are responsible for erythrophagocytosis; however, the fact that predominantly uninfected erythrocytes were phagocytized suggests that the erythrocytes might have been opsonized by an autoantibody associated with the P. berghei infections.  相似文献

The presence and ultrastructural features of reticulum cells and macrophages were studied in the spleen of the dogfish Scyliorhinus canicula. Three morphologically distinguishable regions of the spleen were identified: the white pulp, the red pulp and the ellipsoids. In all three, the splenic parenchyma was a meshwork supported by reticulum cells and fibres. Reticulum cells in both the white and the red pulp are irregular elements, the processes of which are joined by cell junctions and embrace developing reticular fibres. The ellipsoids of the dogfish spleen are terminal branches of the splenic arteries of the white pulp, with a sheath consisting of reticulum cells, reticular fibres, ground substance, macrophages and occasional lymphocytes. Isolated melanomacrophages also occur in the ellipsoid walls as well as in the red pulp. In both the white and the red pulp phagocytic reticulum cells, and macrophages appear frequently forming cell associations with surrounding blood cells, mainly lymphocytes. The functional significance of the ellipsoids and the cell-cell clusters of the white and the red pulp is discussed in relation to the immune capacities demonstrated in elasmobranchs.  相似文献

The spleen of Psammophis sibilans is composed mainly of red pulp, the white pulp being poorly developed. The white pulp lymphoid clusters are scattered throughout the organ and contain lymphocytes, reticular cells, and some plasma cells. The red pulp consists of reticular cells intermingled with blood cells, sinusoids, and melanomacrophage centers (MMCs). Filtering of particulate matter from the blood occurs in the red pulp by phagocytes of the pulp cord. MMCs are formed by the association of free macrophages that have phagocytosed some blood cells. This paper describes the distribution of thymus-derived lymphocytes in the spleen of the lizard, Calotes versicolor. Our studies reveal the existence of red and white pulp in the spleen. The red pulp consists of collagenous fibrous septae containing lymphocytes, erythrocytes and sinuses. The white pulp is an area enclosed by fibrous septae, containing lymphocytes, reticular cells and arterioles. The region around the arteriole in the white pulp was depleted of lymphocytes either one month after adult thymectomy or after anti-thymocyte serum treatment. The repopulation of this region was observed in sham-thymectomized controls, but not in thymectomized lizards. Therefore, this peri-arteriolar region may be designated as a thymus-dependent area, as described previously in higher vertebrates. The significance of these findings is discussed in relation to the phylogeny of the immune system.  相似文献

In several ectotherms, including all members of the Osteichthyes studied so far, the spleen is capable of storing and releasing erythrocytes according to the animal's respiratory needs. The tropical cave cyprinid Phreatichthys andruzzii uses its liver rather than the spleen as the site of accumulation in the respiratory compensation process, like the amphibian Rana esculenta. The reversible process of erythrocyte accumulation in the liver is very evident in animals anaesthetized with chlorobutanol; MS-222, an anaesthetic widely used in lower vertebrates alters all the haematological parameters and is not suitable for studies on blood and respiration. The hepatic respiratory compensation mechanism is as efficient as the splenic one: in animals kept at 18 °C for 24 h the mean liver weight percentage was 70% higher than in specimens kept at 28 °C (2.43% of the body weight compared to 1.39%, in groups of six specimens) while mean red blood cell counts fell from 2.49 to 1.60 · 1012 per l, in agreement with the haematocrit value and haemoglobin concentration; mean corpuscular volume remained constant (at about 177 fl). Summary In the present study comparative aspects of the ultrastructure of the spleen were analyzed in non-immunized and T-dependent antigen-challenged natterjacks, Bufo calamita. Special attention is focused on the role of the non-lymphoid components in the splenic immunoreactivity. Ten days after primary immunization with sheep erythrocytes, splenic lymphoid follicles increase considerably in number and size. By that time, lymphoblasts, medium and large lymphocytes abound in the periphery of the white pulp near the marginal zone. Meanwhile, in the red pulp numerous monocytes migrating across the sinusoidal walls apparently transform into giant, dendritic-like cells. Twenty days after immunization the splenic lymphoid follicles decrease in number, although certain reactivity persists and numerous plasma cells occur in the cell cords and sinusoids of the red pulp. These results are discussed comparatively with those reported in other lower vertebrates.  相似文献

Heat stress results in a multitude of biological and physiological responses which can become lethal if not properly managed. It has been shown that heat stress causes significant adverse effects in both human and animals. Different approaches have been proposed to mitigate the adverse effects caused by heat stress, among which are special diet and probiotics. We characterized the effect of the yeast fermentate EpiCor (EH) on the prevention of heat stress-related complications in rats. We found that increasing the body temperature of animals from 37.1±0.2 to 40.6±0.2 °C by exposure to heat (45 °C for 25 min) resulted in significant morphological changes in the intestine. Villi height and total mucosal thickness decreased in heat-stressed rats pre-treated with PBS in comparison with control animals not exposed to the heat. Oral treatment of rats with EH before heat stress prevented the traumatic effects of heat on the intestine. Changes in intestinal morphology of heat-stressed rats, pre-treated with PBS resulted in significant elevation of lipopolysaccharides (LPS) level in the serum of these animals. Pre-treatment with EH was effective in the prevention of LPS release into the bloodstream of heat-stressed rats. Our study revealed that elevation of body temperature also resulted in a significant increase of the concentration of vesicles released by erythrocytes in rats, pre-treated with PBS. This is an indication of a pathological impact of heat on the erythrocyte structure. Treatment of rats with EH completely protected their erythrocytes from this heat-induced pathology. Finally, exposure to heat stress conditions resulted in a significant increase of white blood cells in rats. In the group of animals pre-treated with EH before heat stress, the white blood cell count remained the same as in non-heated controls. These results showed the protective effect of the EH product in the prevention of complications, caused by heat stress.  相似文献
