Heterozygotes from matings of the mouse strains YBR/Cv and C3H/As have about 3 times more YBR-amylase than C3H-amylase in the saliva. The determinant for this quantitative effect is located on linkage group XVI close to or within the structural gene for salivary amylase. The quantitative effect is the result of an increase in the rate of synthesis of YBR-amylase, and the determinant is cis acting. Studies of other mouse strains suggest that regulatory genetic elements may modulate salivary amylase production.This work was supported by the Danish Natural Science Research Council and a grant from the United States Public Health Service (Grant GM-19521). 相似文献

to the cytoskeleton. The effects of ritanserin and inmecarb are prevented or attenuated by lipophilic serotonin analogs (serotoninamides
of polyenoic fatty acids), as well as by polypeptides isolated from neurons Pd5 and Pd6 of the pedal ganglia of the adultTritonia. In late embryos (stage of veligers), serotonin and to a lesser extent its lipophilic analogs strongly increase embryonic
motility. This effect of serotonin is potentiated by some neuropeptides and inhibited by others. These results provide evidence

In this study, we have further characterized the morphology and physiology of the neuroglandular synapse between the identified
buccal neuron, B4, and the salivary gland ofHelisoma. We demonstrate that the coupling coefficient between salivary cells within an individual acinus is approximately 1.0. We
also demonstrate that synapses within the salivary gland are located near a superficial muscle layer. We examine the effects
of glutamate on the salivary gland and on the B4-salivary gland EPSP.l-glutamate produces a transient, rapid onset depolarization of salivary gland cells. The response is mimicked by high concentrations
ofl-homocysteic acid, but not by NMDA,l-aspartate,d-glutamate or kainate. The response is blocked by the presence ofl- ord-glutamate in the bath, but not by CNQX, DNQX, DGG,d-AP5, orl-AP3. The depolarization is primarily dependent on the presence of calcium in the bathing solution. When eitherl- ord-glutamate is present in the bathing solution, the amplitude of the B4-salivary gland EPSP is reversibly reduced. The similar

Summary The heart of the nudibranch mollusc Archidoris montereyensis is regulated by a small number of powerful effector neurons located in the right pleural and visceral ganglia. Two identifiable neurons in the pleural ganglion, a heart excitor (plHE) and a heart inhibitor (PlHI), are especially important regulators of cardiac function in that low levels of spontaneous activity in either cell significantly alters the amplitude and rate of heart contractions. These neurons have extensive dendritic arbors within the right pleural ganglion and branching axonal processes within the visceral ganglion. The visceral ganglion also contains a heart excitor neuron (VHE) and at least two heart inhibitor neurons (VHI cells), but their influence on cardiac activity is weaker than that of the pleural ganglion cells. All of these heart effector cells appear to be motor neurons with axons that terminate predominately in the atrio-ventricular valve region of the heart via the pericardial nerve. The simplicity and strength of these neuronal connections to the heart of Archidoris make this a favorable preparation for studies of cardiac regulation.Abbreviations

Human proline-rich proteins (PRPs) are polymorphic, homologous in sequence, and linked in a cluster called the human salivary protein complex (SPC). Recently this complex was localized to human chromosome band 12p13.2 (Mamulaet al., Cytogenet. Cell Genet. 39:279, 1985). We have isolated a PRP cDNA, EO27, from a human parotid gland library, identified it by DNA sequencing, and used it to study the molecular and cellular biology of PRP production. Cell-free translation and mRNA characterization with EO27 indicate that the numerous PRPs seen in saliva are produced from relatively few, large precursors, probably by posttranslational cleavage. This supports an hypothesis originally proposed by Friedman and Karn in 1977 (Am. J. Hum. Genet. 29:44A;Biochem. Genet. 15:549) and later supported by biochemical studies (Karnet al., Biochem Genet. 17:1061, 1979) and molecular studies (Mamulaet al., Fed. Proc. 43:1522, 1984; Maedaet al., J. Biol. Chem. 260:1123, 1985). EO27 was also used in this study to localize PRP mRNA production to the acinar cells of the parotid gland byin situ hybridization. 相似文献

Summary In order to study the relationship between the inward Ca current activated by hyperpolarization and the frequency increase in ciliary beating, Paramecium cells were voltage clamped under conditions where K current was suppressed by use of CsCl electrodes and by extracellular tetraethyl ammonium. A 2-s pulse of hyperpolarization from the resting potential activated an inward current consisting of two components, an initial transient current peaking at 0.1–0.2 s (which had been identified as a Ca current) and a subsequent sustained current. The initial component was not associated with the frequency increase because the frequency increase was normally induced even when the peak current was almost completely inhibited by external addition of Ba2+. The second sustained current was closely correlated with the frequency increase. The frequency rose steeply with the sustained current and saturated at –0.6 nA. External addition of La3+ or replacement of Ca2+ by Mg2+ suppressed this current, and at the same time the frequency increase was inhibited. As the amplitude of the sustained current was not changed by deciliation, this current must pass through the somatic membrane. These results suggest that the frequency increase upon hyperpolarization is triggered by the voltage-activated inward current passing through the somatic membrane of the interciliary compartment.Abbreviations cAMP

Homologues of a neuron that contributes to a species-specific behavior were identified and characterized in species lacking
that behavior. The nudibranch Tritonia diomedea swims by flexing its body dorsally and ventrally. The dorsal swim interneurons (DSIs) are components of the central pattern
generator (CPG) underlying this rhythmic motor pattern and also activate crawling. Homologues of the DSIs were identified
in six nudibranchs that do not exhibit dorsal–ventral swimming: Tochuina tetraquetra, Melibe leonina, Dendronotus iris, D. frondosus, Armina californica, and Triopha catalinae. Homology was based upon shared features that distinguish the DSIs from all other neurons: (1) serotonin immunoreactivity,
(2) location in the Cerebral serotonergic posterior (CeSP) cluster, and (3) axon projection to the contralateral pedal ganglion. The DSI homologues, named CeSP-A neurons, share additional features with the DSIs: irregular basal firing, synchronous inputs, electrical coupling, and
reciprocal inhibition. Unlike the DSIs, the CeSP-A neurons were not rhythmically active in response to nerve stimulation. The CeSP-A neurons in Tochuina and Triopha also excited homologues of the Tritonia Pd5 neuron, a crawling efferent. Thus, the CeSP-A neurons and the DSIs may be part of a conserved network related to crawling that may have been co-opted into a rhythmic
swim CPG in Tritonia.

Summary We have identified 13 pairs of neurons in the pedal ganglia of the marine nudibranch slug Tritonia diomedea that responded tonically and/or phasically to water-flow directed at the rhinophore sheaths and oral veil tips. Most of the neurons responded equally to inputs from either side of the body, but 6 pairs responded with greater intensity to ipsilateral water-flow stimuli. When stimulated intracellularly in a semi-intact, whole-animal preparation, 4 of these 6 pairs of neurons caused ipsilateral movements that may turn the animal towards that side. These observations suggest a role for these current-sensitive neurons in the previously described orientation to water-currents in Tritonia diomedea. 相似文献

The nostrils of the seabird Calonectris diomedea are endowed with a salt-excreting gland that could produce a suitable environment for the colonization of extreme halophilic
prokaryotes. We have studied in this organ the presence of extreme halophiles by means of culturing techniques. We could easily
cultivate members of haloarchaea, and all cultures studied were identified as members of one of the two species Halococcus morrhuae and Hcc. dombrowskii. In order to reveal the diversity of these colonizers, we undertook a taxonomic study. Altogether, the results indicated
that members of the genus Halococcus may constitute a part of the natural epizootic microbiota of C. diomedea, and that they exhibit such an important degree of taxonomic variability that appeals for a pragmatic species definition.
This seabird nests in the west Mediterranean coasts, but its migratory habits, reaching locations as distant from the Mediterranean
as the South Atlantic, may help in the dispersal mechanisms of haloarchaea through the Earth’s surface. 相似文献

Summary We stayed on the Portugese island of Selvagem Grande, 30° N 16° W, 300 km south of Madeira and 200 km north of Tenerife, from 9 May to 3 June 1991 in order to study the prelaying exodus in Cory's Shearwaters (Calonectris diomedea borealis) and its connection with the colony attendance cycles described earlier. Laying took place on 1.5 June ±3.4 days. During the preceding weeks males and females of Cory's Shearwater show very different attendance patterns. Females are absent for 19±4.8 days at minimum. Absences of males are much shorter and also much more variable. They last 7.9±6.0 days (coefficient of variation 76 %, as against 12 % in females). The first incubation stint of the males is badly synchronised with the laying: the delay between laying and the start of the males' first stint may amount up to one week, without compromising the breeding success. Two well marked attendance peaks have been observed, separated by an interval of 18 days, twice the length of that observed in June 1988. We suggest that this anomaly in the daily attendance graph is correlated with the prelaying exodus. The function of the prelaying exodus and its relation with the energy cost of producing the egg and the necessity of storing lipid reserves are discussed.
Vorbrutzeitliches Verlassen der Kolonie beim Gelbschnabelsturmtaucher (Calonectris diomedea borealis) auf Selvagem Grande
Zusammenfassung Zwischen 9. Mai und 3. Juni 1991 untersuchten wir auf der portugisischen Insel Selvagem Grande, 30° N 16° W, 300 km südlich von Madeira und 200 km nördlich von Teneriffa gelegen, das Verlassen der Kolonie vor der Eiablage des Gelbschnabelsturmtauchers (Calonectris diomedea borealis) und die Beziehung zur Anwesenheit in der Kolonie. Gelegt wurde am 1,5 Juni ±3,4 Tage. In den Monaten vor der Eiablage zeigten Männchen und Weibchen sehr unterschiedliche Muster der Anwesenheit in der Kolonie. Während die Weibchen im Minimum durchschnittlich 19±4,8 Tage abwesend waren, war diese Abwesenheit bei den Männchen mit 7,9±6,0 Tagen wesentlich kürzer und variabler (Variationskoeffizient 76 % gegenüber 12 % bei den Weibchen). Die erste Brutbeteiligung des Männchens war kaum mit der Eiablage synchronisiert, doch war dadurch der Bruterfolg nicht beeinträchtigt. Die Anwesenheit in der Kolonie zeigte zwei Gipfel, die 18 Tage auseinander lagen. In 1988 betrug dieser zeitliche Abstand nur etwa die Hälfte. Es wird vermutet, dass dieser Unterschied mit dem Ausmaß des vorbrutzeitlichen Verlassens der Kolonie korreliert. Die Funktion dieses vorbrutzeitlichen Exodus und seine Beziehung zu den energetischen Kosten der Eiproduktion und der Bildung von Lipidreserven werden diskutiert.相似文献

Effect of nitrogen supply rate on disease resistance in tomato depends on the pathogen 总被引:1,自引:0,他引:1
The aim of this study was to investigate the effect of tissue nitrogen concentration, as a consequence of nitrogen supply
rate, on the susceptibility of tomato plants to three pathogens. We varied tissue N concentration by supplying N at different
rates by adding nitrate in different, exponentially increasing amounts to the nutrient solution on which the tomato plants
were grown. Separate experiments were carried out to test susceptibility of tomato plants to the bacterial speck-causing Pseudomonas syringae pv tomato, to the wilt agent Fusarium oxysporum f.sp. lycopersici and to tomato powdery mildew caused by Oidium lycopersicum. The effect of tissue N concentration appeared to be highly pathogen-dependent: there was no effect on susceptibility to
F. oxysporum, but susceptibility to P. syringae and O. lycopersicum increased significantly with increasing N concentration. We have previously demonstrated the opposite for susceptibility
to Botrytis cinerea: decreasing susceptibility with increasing N concentration. The apparent contradictory effects are discussed in relation
to the effect of N supply on both the nutritional value of the plant tissue to the pathogen and on the concentration of resistance-related
compounds. We conclude that the effect of changing both characteristics on disease susceptibility is highly pathogen-specific
and is probably dependent on differences in resource requirements of the pathogen or the sensitivity of the pathogen to plant
resistance reactions or on both these factors.

An immunogold-labelling electron-microscopic study of the frontal ganglion of two noctuids, Lacanobia oleracea and Helicoverpa armigera, has been carried out with antisera directed against three neuropeptides; allatostatins of the Y/FXFGL-NH2 type, Manduca sexta allatostatin (Mas-AS) and M. sexta allatotropin. The ganglion of both noctuids has two pairs of large peptidergic neurones with many clusters of electron-dense granules, one pair being situated anteriorly and the other posteriorly. By means of a double-labelling (flip-flop) technique, with different sizes of gold particles, all possible paired combinations of the three different types of peptide have been visualised within granules of the anterior neurones, leading to the conclusion that the three peptides are co-packaged and co-stored in these cells. Within the posterior neurones of L. oleracea, gold labelling of granules is only linked to the Y/FXFGL-NH2 allatostatin antisera and, in contrast to the anterior cells of this species in which double gold labelling results in a sparse accumulation of gold particles for any one peptide type, single labelling gives a more intense, uniform pattern of gold particles. In contrast to L. oleracea, the gold-labelling pattern seen in the posterior neurones of H. armigera reflects the co-localisation of allatostatins of the Y/FXFGL-NH2 type with Mas-AS in this species. The Salton Sea is one of the few saline, inland lakes in the world with a population of barnacles, Balanus amphitrite. It is also one of California’s most impaired water bodies due to excessive nutrient loading which leads to phytoplankton blooms and low dissolved oxygen. Currently, B. amphitrite growth is limited due to lack of hard substrate in and around the Sea. We have hypothesized that artificial substrate could support the growth of B. amphitrite and their filter-feeding would lead to improved water quality. Periodic harvesting of the barnacles would result in the permanent removal of nitrogen and phosphorus from the Sea. A 44-day in-situ experiment was carried out in the Salton Sea to assess the rate of barnacle growth and phosphorus and nitrogen sequestration on burlap sheets suspended vertically from a floating line. Burlap panels were collected weekly and the barnacles analyzed for Ca, total-P, inorganic-P, total-N, total-C, CaCO3, and organic matter content. After 44 days of growth, the barnacle mats weighed 7.4 kg m−2 on a dry weight basis, with 80% of the mass as shell material. The nutrient sequestration was 9.4 g P m−2 and 100 g N m−2. Approximately half of the P was inorganic and appears to be coprecipitated with the calcium carbonate shell material. Results indicate that harvesting barnacles grown on artificial substrate in the Salton Sea would not be an effective method for removing N or P from the lake because of the relative proportions of shell material and organic material. Peptides of the RFamide family occur in neurosecretory cells of all nervous systems of Cnidaria so far studied. Photoreceptive
organs – if evolved in a cnidarian species – are always associated with neural cells showing RFamide immunoreactivity. Experimental
evidence for the function of RFamides and other neuropeptides in nervous systems and photoreceptive organs is, however, scarce
or lacking. RFamide and LWamide immunoreactivity were surveyed in photoreceptive organs of the hydromedusa Cladonema radiatum, in rhopalia of the scyphozoan Aurelia aurita, and in rhopalia of the cubomedusa Tripedalia cystophora. A possible function of neuropeptides in transmission of photic stimuli was assayed by analysing photic behavior in Tripedalia, which has highly developed eyes, and in the simply constructed planula of the hydroid Hydractinia echinata, in which the mode of light perception is unknown. In both species, light orientation was effectively prevented by RFamides
administered to the animals in micromolar concentration. In contrast, among four other neuropeptides occurring in the larva
of Hydractinia, only one interfered with phototaxis and then only at 10× higher concentrations. Planulae depleted of bioactive peptideamides
also lost phototaxis while still locomotorily active. The results support the hypothesis that one possible function of RFamides

Autophagy, a form of programmed cell death (PCD) that is morphologically distinguished from apoptosis, is thought to be as prevalent as apoptosis, at least during development. In insect metamorphosis, the steroid hormone 20-hydroxyecdysone (ecdysone) activates autophagic PCD to eliminate larval structures that are no longer needed. However, in comparison with apoptosis, there are not many studies on the regulation mechanisms of autophagy. To provide a useful model for studying autophagic PCD, I established an in vitro culture system that enables real-time observation of the autophagic cell destruction of Drosophila salivary glands. The new system revealed that de novo gene expression was still required for the destruction of salivary glands dissected from phanerocephalic pupae. This indicates the usefulness of the system for exploring genes that participate in the last processes of autophagic PCD.Edited by N. Satoh 相似文献

The protein and peptide composition of medicinal leech salivary gland secretion (SGS) was analyzed in preparations obtained in July from three species--Hirudo verbana, H. medicinalis, and H. orientalis. Two-dimensional electrophoresis (molecular mass 10-150 kD and pI 3-10) revealed no distinctions in the distribution of over 100 silver-stained proteins. Differences were noted only in intensity of 10 protein spots at 30-90 kD and pI 4.7-7.5. Mass spectrometric profiling of SGS of the three leech species using the Zip-Tip/golden chip scheme and cation-exchanging chips CM-10 revealed over 50 components in SGS of each of the three leech species. It was noted that 30-40% of the individual masses of the SGS of each leech species fall within the masses present in SGS of at least one of the two other species. This rather small part of the total mass may be indicative of a high polymorphism of amino acid sequences or a high frequency of posttranslational modifications of the SGS proteins and peptides. Calculation of Jacquard's coefficient showed that H. medicinalis and H. orientalis are closest to each other in SGS composition, which is consistent with data in the literature on the phylogenetic relationship between these two species of medicinal leech. Comparison of detected molecular masses with those of six known biologically active compounds produced by medicinal leeches revealed their uneven distribution in SGS of each of the three medicinal leech species. This opens prospects for using certain species of medicinal leech for targeted therapy of various pathologies. 相似文献

Using light-grown plants of Sinapis alba an analysis has been made of the effect on extension growth of adding far red light to a background photosynthetic source. It has been possible to distinguish between the increase in fluence rate and the reduction of the amount of phytochrome present as Pfr, which are both consequences of the addition of supplementary far red light, and to determine that the response of increased extension growth is due only to the latter. It is shown that the degree of fluence rate dependency varies with photoequilibrium and the significance of this interaction is discussed in terms of the mode of action of phytochrome and of its role in the natural light environment.Abbreviations PAR
