Structural development and lipid absorption in anterior intestine of gilt-head sea bream Sparus aurata , were studied by light and electron microscopy during three stages of post-embryonic development: (1) the endotrophic period from hatching day (day 0) to mouth opening day (day 3); (2) the endo-exotrophic period from days 3 to 15; (3) the exotrophic period after day 15. During the 2 days following hatching, there was no trace of lipids in intestinal epithelia. Before mouth opening day, the first lipoproteic particles of endogenous origin appeared in enterocyte endoplasmic reticulum and Golgi apparatus. During the endo-exotrophic period, lipoproteinogenesis increased weakly until day 9, and more greatly between days 9 and 15. It intensified at the beginning of the exotrophic period to remain at a high level afterwards. Until day 15, few transfers of lipoproteins to interenterocyte spaces occurred, whereas no lipoproteins were detectable in the blood flow from days 7 to 9. Their concentration increased slightly between days 9 and 15 to become intense afterwards. Lipid droplets appeared from day 7, and subsisted until the end of endo-exotrophic period. Possible relationships between very low density lipoproteins and chylomicron type lipoproteins and lipid droplets related to lipid excess in food are discussed. 相似文献

The basal metabolic rate ( BMR) of mice losing weight about two weeks after infection by Nematospiroides dubius was lower than that of uninfected mice gaining weight when fed ad libitum or losing weight on quantitatively reduced rations. There was no difference in BMR between the latter two groups.Following the injection of 14C-glucose, the high specific activity of expired CO 2 from infected and reduced ration mice was considered to be due to the utilization of energy reserves. The levels of lipid and liver glycogen were low in these two groups of mice and their specific activities, particularly in the severely affected animals, were high.It was concluded that the depressed BMR of infected mice is unrelated to anorexia, which did, however, explain the low levels of lipid and liver glycogen. 相似文献

The levels of glycogen and glucose, as well as of the activities of several key enzymes of glycogenolysis, glycolysis, gluconeogenesis and the pentose phosphate shunt were assessed in livers of small (97 g) and large (166 g) rainbow trout Oncorhynchus mykiss during gradual transfer to sea water (salinities of 0, 9, 18 and 28%). Plasma glucose and protein levels were also evaluated. Liver carbohydrate metabolism changed during gradual adaptation of rainbow trout to sea water. Glucose increased with salinity in livers of both sizes of animals, as did glycogenolysis and gluconeogenesis, but this latter only in large animals, and glycolytic rates in small animals. The different responses in metabolic parameters between sizes of animals may reflect a higher stress of the small animals and/or a better adaptation of large animals to increased salinities, in a way similar to that previously suggested by other authors after evaluation of non-metabolic parameters. The changes observed at 28% can also be the result of reduced food consumption. 相似文献

Livers of juvenile cabrilla sea basses ( Serranus cabrilla ) were subjected to light and electron transmission microscopy following different periods of maintenance in an aquarium. Since this fish is easy to feed in captivity and the hepatic structure was found to be comparable at the four periods tested (0, 5, 10 and 20 days), both at the histological and ultrastructural level, the liver of S. cabrilla could be an available model for marine contamination experimental studies. As in other fish species, it is not possible to distinguish the portal lobules and the triads. The Melano-Macrophage Centres contain tipofuscins, ceroids and haemosiderin, but they do not contain any melanin. The hepatocytes are arranged in cords (two cells thick), and, at the ultrastructural level, they show numerous microvilli in the perisinusoidal and canalicular areas. The hyaloplasm includes a considerable amount of glycogen and some lipid droplets are occasionally observed. Mitochondria, rough endoplasmic reticulum and the Golgi apparatus are relatively scarce. 相似文献

Cobalt decreases blood glucose in diabetic rats but the mechanisms involved are unclear. To determine the contribution of glycogen metabolism to glycemia-lowering effect, glycogen contents of liver and muscle in the streptozotocin-induced diabetic rats were determined. The liver glycogen was depleted in diabetic rats. But when cobalt was administered to the rats, the glycogen returned to the level of healthy rats, concomitantly with the decrease in blood glucose. The cobalt treatment had no effect on the muscle glycogen in the diabetic rats. The tissue-specific responses of glycogen metabolism suggest the involvement of suppressed glucagon signaling due to cobalt treatment. 相似文献

The plasma membrane of spermatogenic cells of the teleost Xiphophorus helleri was examined ultrastructurally and cytochemically in order to characterize the temporal development of the membrane specializations characteristic of the mature spermatozoon. Mature sperm display a mosaic distribution of Concanavalin A and Ricinus comrnunis I binding sites; the anterior region of the head displays an intense binding that is not seen in other surface regions. This asymmetric binding is evident in early spermatids and the area of lectin binding appears associated with the plasma membrane overlying the nucleus. Transmission electron microscopy reveals that the plasma membrane over the anterior region of the head is characterized by an ordered glycocalyx and a tight adherence to the underlying nucleus. Additional membrane differentiations were revealed both in the midpiece region where a “submitochondrial net” is attached to the plasma membrane and at the base of the axoneme where the plasma membrane possesses a “collar-like” arrangement of circumferential rings. The possible functions of these differentiations, as well as their correlation to differentiations seen in sperm of other animal groups, are discussed. 相似文献

Summary Glycogen synthase kinase-3 (GSK-3) was purified from rabbit liver to homogeneity by ultracentrifugation, ion-exchange chromatography on DEAE-cellulose, Cellulose phosphate, CM-Sephadex and Fast Protein Liquid Chromatography (FPLC) on Mono-S column. The enzyme was purified approximately 20,000 fold with an approximate 2% recovery. The purified enzyme showed a single band on SDS-polyacrylamide gel electrophoresis. GSK-3 is a monomeric enzyme with a molecular weight of 50,000–52,000 as derived from SDS-polyacrylamide gel electrophoresis and gel filtration. The purified enzyme was indeed a GSK-3 since it phosphorylated three sites, i.e., 3a, 3b, and 3c on liver glycogen synthase. GSK-3 incorporated up to 2.6 mol Pi/mol glycogen synthase subunit with a concomitant inactivation of glycogen synthase activity. 相似文献

Hypertension is known to be associated with an oxidative stress resulting from an imbalance of antioxidant defense mechanisms
in various tissues. The purpose of this study was to investigate the relationship between the increase of arterial blood pressure,
measured during the gradual development of experimental hypertension in deoxycorticosterone (DOCA)-salt-treated rats, and
an early imbalance of liver antioxidant status. The levels of liver oxidant/antioxidant markers and iron were studied during
the induction of hypertension in 3-, 6-, and 8-wk DOCA-salt-treated Sprague-Dawley rats. Hepatic antioxidant defenses were
decreased as early as 3 wk of hypertensive treatment: the decrease of peroxidase-reductase-transferase and catalase activities
was associated with a significant increase of thiobarbituric acid reactive substances (TBARS) levels. Liver oxidative stress
increased until 6 wk, and remained stable at 8 wk of DOCA-salt treatment. Concurrently, liver iron levels were increased at
6 wk and returned to normal values after 8 wk of hypertensive treatment. Iron seems to be an inductor of liver oxidative stress
and responsible for the persistent oxidative stress, most likely through secondary free-radical release. Thus, our data (1)
confirm that hypertension in DOCA-salt-treated rats might be a free-radical-dependent disease where hepatic oxidant/antioxidant
imbalance is obviously involved from the beginning of blood pressure elevation and (2) suggest that the use of suitable iron
chelators might reverse liver oxidative stress associated with the increase of blood pressure. 相似文献

Effects of somatostatin (SOM) on tissue contents of proteins, total lipids and phospholipids were investigated in regenerating and intact liver tissue of Y-59 rats. Whereas SOM inhibited protein accumulation in regenerating liver, the hormone evoked and increase in total lipids, and specially in phosphatidylcholine, phosphatidylethnolamine, phosphatidylserine (PS) and phosphatidylinositol (PI). Since the same effects were not seen when intact liver was analyzed, it is assumed that SOM acts primarily on tissue stimulated to rapid growth. The increase of PS+PI fractions indicates a specific effect of SOM on the metabolism of phosphatidylinositides. Such an effect might result from the interference of the hormone with the action of growth factors that accelerate phosphatidylinositol breakdown. 相似文献

Effect of realimentation was studied on the structure and function of liver tissue of carp, Cyprinus carpio. Yearling carp, after a 3-month starvation period, were renourished at a feeding rate of 1% body weight per day. Samples were taken at refeeding days 0, 1, 2, 5, 22 and 78. Analyses were made of blood metabolites, liver RNA, DNA, lipids, glycogen and protein and of liver enzyme activities. Additionally, liver cytology was examined by means of qualitative and quantitative electron microscopy. The early refeeding period (up to day 5) was characterized by a fast recovery of plasma metabolite concentrations (protein, total lipids, free fatty acids, glucose), a drastic augmentation of hepatic glycogen reserves, and a pronounced increase of total liver weight and liver-somatic index. Constant values of total hepatic DNA showed that liver weight augmentation was not due to cell proliferation, but to a pronounced enlargement of the existing hepatocytes. Major hunger-related structural modifications of carp hepatocytes such as enlarged mitochondria or prominence of the lysosomal compartment were reversed. A significant volume increase of cell nuclei, together with a particularly strong elevation of hepatic RNA concentrations during initial realimentation suggest an immediate stimulation of protein synthesis. Since the cisternae of the endoplasmic reticulum were not reconstituted during that early phase, protein synthesis may have been executed mainly by free ribosomes. With prolonged realimentation, the volume of the endoplasmic reticulum as well as total and relative contents of liver soluble protein continuously increased, whereas RNA concentrations decreased again. An enforcement of liver oxidative capacity was indicated by the augmentation of cellular number and volume of mitochondria. The activities of the enzymes glucose-6-phosphate dehydrogenase and malic enzyme, which convert excess energy into NADPH, increased steadily. Concomitantly, hepatic lipid accumulation was enhanced. In conclusion, liver metabolism during the early recovery phase seems to be dominated both by repair processes and by intensive protein and glycogen synthesis. The liver slows down these processes during prolonged refeeding and directs an increasing percentage of energy and metabolites toward the generation of reducing equivalents and lipid reserves.Abbreviations BW

Four overlapping cDNA fragments encoding a partial sequence for uncoupling protein 2 (UCP2) were amplified by PCR using degenerate primers from the liver of a marine teleost fish, red sea bream (Pagrus major). The partial sequence was 674 bp long, encoding 224 amino acids. The deduced amino acid sequence from the cDNA partial sequence contained the signature motifs for mitochondrial transporter protein and revealed positional identity higher than 72.8% with UCP2 from mammals. The fish UCP2 gene was highly expressed in the liver but almost undetectable in the visceral mesenteric adipose tissue. Using beta-actin as control, the UCP2 mRNA level was determined to be at least 20-fold higher in the liver than in the visceral mesenteric adipose tissues. Neither 48 h starvation nor high lipid diet had any significant effect on liver UCP2 gene expression, indicating that the abundant UCP2 gene expression was stable and might have some basic function in a fish liver that always contains high lipid content. The striking contrast of UCP2 gene expression in the two fish fat-depot organs is consistent with their large differences in oxidative capacity. We suggest that the fish liver may adapt to a constantly high fat deposit by maintaining high UCP2 expression to constrain reactive oxygen species (ROS) production and protect hepatocytes from apoptosis. 相似文献

1. 1.|The metabolic role of the thyroid gland was studied in intact snakes, Naja naja and Ptyas korros treated with tri-iodo-thyronine (T3) and thyroxine (T4) and in thyroidectomized (Tx) N. naja kept at 21°C by analyzing tissue composition and glycogen phosphorylase a activity. 2. 2.|Liver weight was unaffected by thyroid hormone injection in both species but decreases in liver glycogen followed T3 or T4 injection, and there was an increase in liver glycogen in N. naja. These changes in liver glycogen were accompanied by a decrease in glycogen phosphorylase a activity with T3 injection. T3 decreased muscle glycogen in Ptyas and Tx increased it in N. naja. 3. 3.|T3 increased % liver lipid in Ptyas but not in Naja. 4. 4.|Between species, there were differences in liver weight, blood glucose level, cholesterol level and % muscle lipid. 5. 5.|The results showed that thyroid hormones affected carbohydrate and lipid metabolism at a low temperature of 21°C, although the significance is not known.

The histology of the gonad of the red sea bream, Pagrus major, was examined in order to study the early gonadal development, sexual maturation and sex ratio in a natural population. A
total of 1,117 fish between the ages of 4 months and 8 years were examined. Gonads of 4-month-old fish were either sexually
undifferentiated with a central cavity, or ovarian in form. Gonads of 12- and 18-month-old fish were ovaries or bisexual gonads,
while those of 2-year-old fish were ovaries, bisexual gonads or testes. Fish aged between 3 and 8 years had ovaries or testes,
except for a few bisexual gonads found in 3- and 4-year-old fish. The chronological appearance of females, hermaphrodites
and males in that order, and histological evidence, suggested that the testis originates from the ovary via a bisexual gonad
in the juvenile stage. The sex ratio of females to males at the age of 2 years and over was about 1:1, suggesting that hermaphroditic
red sea bream appear in about 50% of the juvenile population. The sexual pattern in this species, therefore, is concluded
to be gonochorism with a bisexual juvenile stage. 相似文献

Scanning and transmission electron microscopy were used to investigate the fine structure of the sperm of the sparid fish Pagellus bogaraveo.The spermatozoon of P. bogaraveo belongs, like that of the other sparid fish, to the teleostean “type I” spermatozoon with the flagellar axis insert perpendicular to the nuclear fossa. It has an ovoidal head, a short, cylindrically shaped midpiece and a long tail region. The nucleus reveals a deep invagination (nuclear fossa), in which the centriolar complex is located, and a satellite nuclear notch shaped like a golf club. The two centrioles are perpendicular to each other and show a conventional “9+0” pattern. The distal centriole is attached to the nuclear envelope by means of basal feet and radial fibers made of electron-dense material. Below the basal plate, plasma membrane pinches in, and the necklace, a specialized connection joining axonemal doublets to the plasma membrane, is visible. The short midpiece houses one mitochondrion. The flagellum is perpendicularly and eccentrically with respect to the nucleus and contains the conventional “9+2” axoneme. 相似文献

Abstract Liver glycogen, liver lipid, liver triglycerides, plasma glucose, plasma total lipid, plasma cholesterol, plasma corticoids, hypothalamic serotonin and pituitary pro‐lactin levels were assayed at five times over a 24‐h period in Carassius auratus maintained under a specific photoperiod regime at various times throughout the year. Diurnal variations were observed in all parameters monitored. Daily variations of liver glycogen, plasma glucose, plasma lipid, plasma corticoids and hypothalamic serotonin were affected by time of feeding. Liver glycogen, plasma lipid and plasma corticoid levels were also affected by time of feeding. Diurnal variations of liver glycogen, plasma glucose and plasma lipid were influenced by light‐dark cycles. These data illustrate that feeding time, photoperiod and time of sacrifice are important considerations in the study of metabolic and hormonal parameters in fishes. 相似文献

Mitochondria are cellular organelles where the generation of reactive oxygen species may be high. They are, however, effectively protected by their high capacities of antioxidative systems, as enzymes and either water or lipid soluble low molecular weight antioxidants.These antioxidative defence systems can be effectively regenerated after or during an oxidative stress as long as the mitochondria are in an energized state. Energization of mitochondria mainly depends on the availability of suitable respiratory substrates which can provide hydrogen for the reduction of either the glutathione- or -tocopherol-system, since GSH is regenerated by glutathione reductase with the substrate NADPH and the -tocopheroxyl-radical likely by reduced coenzyme Q. It was shown that mitochondria do not undergo damages as long as they can keep a high energy state. The delicate balance between prooxidative/antioxidative activities can be shifted towards oxidation, if experimentally prooxidants were added. After exhaustion of the antioxidative defence systems damages of rnitochondrial functions become expressed followed by membrane injuries along with the oxidation and degradation of mitochondrial lipids and proteins leading finally to the total degradation of the mitoc hondria.Extramitochondrial antioxidants may assist the mitochondrial antioxidative defence systems in a complex way, whereby particularly ascorbic acid can act both as prooxidant and as antioxidant.
