Bioremediation of petroleum spills requires aerobic soil conditions and readily available N, which may be susceptible to leaching. Our objectives were to determine the influence of soil water potential on nitrification in the presence of crude oil, the toxicity of oil to NHj‐oxidizing bacteria, and the preferences of microorganisms for NH+ 4 or NO? 3. A Weswood clay loam was amended with crude oil to contain 0, 5, and 10% by soil dry weight, and N was added to achieve C:N ratios of 90:1 and 120:1. Soil water potentials were maintained at ‐0.02, ‐0.1, and ‐1.0 kJ/kg or allowed to fluctuate between ‐0.02 and ‐3 kJ/kg. Concentrations of NH+ 4 and NO3 ?were measured during an incubation period of 40 d. Nitrification in soil not amended with oil was rapid at water potentials of ‐0.02 and ‐0.1 kJ/kg but inactive at a water potential of ‐1.0 kJ/kg. Oil reduced nitrification rates and populations of NH+ 4‐oxidizing bacteria. Little NO? 3 accumulated when the C:N ratio was 120:1, but when the C:N ratio was 90:1, up to 150 μg of NO3‐N/g of soil accumulated at a soil water potential of ‐0.02 kJ/kg. Soil water potential in the range used did not greatly influence the extent of oil bioremediation but significantly influenced nitrification. Ammonium was preferentially used over NO? 3 by microorganisms during oil bioremediation. Nitrate accumulation from urea applied to stimulate oil bioremediation was low when N application matched requirements for oil bioremediation, and nitrification was restricted by controlling soil water content.

In the present study, we investigated whether growth and main nutrient ion concentrations of cabbage (Brassica campestris L.) could be increased when plants were subjected to different NH4^+/NO3- ratios. Cabbage seedlings were grown in a greenhouse in nutrient solutions with five NH4^+/NO3- ratios (1:0; 0.75:0.25; 0.5:0.5; 0.25:0.75; and 0:1). The results showed that cabbage growth was reduced by 87% when the proportion of NH4^+-N in the nutrient solution was more than 75% compared with a ratio NH4^+/NO3- of 0.5:0.5 35 d after transplanting, suggesting a possible toxicity due to the accumulation of a large amount of free ammonia in the leaves. When the NH4+/NO3- ratio was 0.5:0.5, fresh seedling weight, root length, and H2PO4- (P), K^+, Ca^2+, and Mg^2+ concentrations were all higher than those in plants grown under other NH4^+/NO3- ratios. The nitrate concentration in the leaves was the lowest in plants grown at 0.5: 0.5 NH4^+/NO3-. The present results indicate that an appropriate NH4^+/NO3- ratio improves the absorption of other nutrients and maintains a suitable proportion of N assimilation and storage that should benefit plant growth and the quality of cabbage as a vegetable.

The intensification of land use constitutes one of the main drivers of global change and alters nutrient fluxes on all spatial scales, causing landscape‐level eutrophication and contamination of natural resources. Changes in soil nutrient concentrations are thus indicative for crucial environmental issues associated with intensive land use. We measured concentrations of NO3–N, NH4–N, P, K, Mg, and Ca using 1,326 ion‐exchange resin bags buried in 20 cm depth beneath the main root zone in 150 temperate grasslands. Nutrient concentrations were related to land use intensity, that is, fertilization, mowing, grazing intensities, and plant diversity by structural equation modeling. Furthermore, we assessed the response of soil nutrients to mechanical sward disturbance and subsequent reseeding, a common practice for grassland renewal. Land use intensity, especially fertilization, significantly increased the concentrations of NO3–N, NH4–N, K, P, and also Mg. Besides fertilization (and tightly correlated mowing) intensity, grazing strongly increased NO3–N and K concentrations. Plant species richness decreased P and NO3–N concentrations in soil when grassland productivity of the actual year was statistically taken into account, but not when long‐term averages of productivity were used. Thus, we assume that, in the actual study year, a distinct drought period might have caused the observed decoupling of productivity from fertilization and soil nutrients. Breaking up the grassland sward drastically increased NO3–N concentrations (+146%) but reduced NH4–N, P, and K concentrations, unbalancing soil nutrient stoichiometry and boosting the risk of N leaching. Reseeding the sward after disturbance did not have a short‐term effect on nutrient concentrations. We conclude that renewal of permanent grassland should be avoided as far as possible and future grassland management has to strongly rise the effectiveness of fertilization. Additionally, grassland management might have to increasingly taking care of periods of drought, in which nutrient additions might not increase plant growth but potentially only facilitate leaching.

Plant growth in saline soils may be increased by fertilisation, but little is known about the effect of different forms of N on wheat growth in soils with different salinity levels. The aim of this study was to investigate the response of wheat (Triticum aestivum L., cv Krichauff) to (NH4)2SO4 or KNO3 or NH4NO3 at 0 (N0), 50 (N50), 100 (N100) and 200 (N200) mg N?kg?1 soil in a saline sandy loam. Salinity was induced using Na+ and Ca2+ salts to achieve three ECe levels, 2.8, 6.6 and 11.8 dS m?1 denoted S1, S2 and S3, respectively, while maintaining a low SAR (>1). Dry weights of shoot and root were reduced by salinity in all N treatments. Addition of N significantly increased shoot and root dry weights with significant differences between N forms. Under non-saline conditions (S1), addition of NO3???N at rates higher than N50 had a negative effect, while N100 as NH4???N or NH4NO3???N increased shoot and root dry weights. At N100, shoot concentrations of N and K were higher and P, Ca, Fe, Mn, Cu and Zn were lower with NO3???N than with NH4???N nutrition. The concentration of all nutrients however fell in ranges did not appear to be directly associated with poor plant growth with NO3???N. At all N additions, calculations indicated that soil salinity was highest with N addition as NO3???N and decreased in the following order: NO3?N > NH4?N > NH4NO3?N. Addition of greater than N50 as NO3???N, compared to NH4???N or NH4???NO3, increased soil salinity and reduced micronutrient uptake both of which likely limited plant growth. It can be concluded that in saline soils addition of 100 mg N?kg?1 as NH4???N or NH4NO3???N is beneficial for wheat growth, whereas NO3???N can cause growth depression.

1. Benthivorous fish may play an important role in internal nutrient loading. Ruffe are highly specialised, feeding exclusively on bottom animals; thus all nutrients released via their feeding are derived from the bottom and are new to the water column. The fish can also release nutrients from the sediment through resuspension while searching for food. 2. The aim of this study was to estimate experimentally in the laboratory the effect on water quality of resuspension and nutrient release by ruffe and bottom animals (chironomids). 3. Ruffe released nutrients during 8 h experiments as follows: total P 1.4, dissolved PO4 0.6, total N 24.0 and NH4‐N 15.9 μg g?1 WW h?1. A decreasing trend in mass‐specific release was observed over time, probably because of starvation. The mass‐specific release of total N and NH4‐N decreased as the mean weight of fish increased. The mean ratio of excreted N : P was 32. 4. In 26 h experiments with sediment and both ruffe and chironomids, ruffe increased nutrient concentrations and turbidity values significantly but chironomids had an effect only on turbidity. Neither ruffe nor chironomids affected the ratio of inorganic N : P concentrations. An interaction between ruffe and chironomids was found for turbidity. 5. According to these results, benthivorous fish may increase nutrient concentrations in the water column and need to be taken into account when estimating internal loading.

In short-term water culture experiments with different 15N labeled ammonium or nitrate concentrations, citrus seedlings absorbed NH4 + at a higher rate than NO3 . Maximum NO3 uptake by the whole plant occurred at 120 mg L–1 NO3 -N, whereas NH4 + absorption was saturated at 240 mg L–1 NH4 +-N. 15NH4 + accumulated in roots and to a lesser degree in both leaves and stems. However, 15NO3 was mostly partitioned between leaves and roots.Adding increasing amounts of unlabeled NH4 + (15–60 mg L–1 N) to nutrient solutions containing 120 mg L–1 N as 15N labeled nitrate reduced 15NO3 uptake. Maximum inhibition of 15NO3 uptake was about 55% at 2.14 mM NH4 + (30 mg L–1 NH4 +-N) and it did not increase any further at higher NH4 + proportions. In a long-term experiment, the effects of concentration and source of added N (NO3 or NH4 +) on nutrient concentrations in leaves from plants grown in sand were evaluated. Leaf concentration of N, P, Mg, Fe and Cu were increased by NH4 + versus NO3 nutrition, whereas the reverse was true for Ca, K, Zn and Mn.The effects of different NO3 -N:NH4 +-N ratios (100:0, 75:25, 50:50, 25:75 and 0:100) at 120 mg L–1 total N on leaf nutrient concentrations, fruit yield and fruit characteristics were investigated in another long-term experiment with plants grown in sand cultures. Nitrogen concentrations in leaves were highest when plants were provided with either NO3 or NH4 + as a sole source of N. Lowest N concentration in leaves was found with a 75:25 NO3 -N/NH4 +-N ratio. With increasing proportions of NH4 + in the N supply, leaf nutrients such as P, Mg, Fe and Cu increased, whereas Ca, K, Mn and Zn decreased. Yield in number of fruits per tree was increased significantly by supplying all N as NH4 +, although fruit weight was reduced. The number of fruits per tree was lowest with the 75:25 NO3 -N:NH4 +-N ratio, but in this treatment fruits reached their highest weight. Rind thickness, juice acidity, and colour index of fruits decreased with increasing NH4 + in the N supply, whereas the % pulp and maturity index increased. Percent of juice in fruits and total soluble solids were only slightly affected by NO3 :NH4 + ratio.

Three-year-old Scots pine (Pinus sylvestris) trees were grown on a sandy forest soil in pots, with the objective to determine their NH4/NO3 uptake ratio and proton efflux. N was supplied in three NH4-N/NO3-N ratios, 3:1, 1:1 and 1:3, either as 15NH4+14NO3 or as 14NH4+15NO3. Total N and 15N acquisition of different plant parts were measured. Averaged over the whole tree, the NH4/NO3 uptake ratios throughout the growing season were found to be 4.2, 2.5, and 1.5 for the three application ratios, respectively. The excess cation-over-anion uptake value (Ca-Aa) appeared to be linearly related to the natural logarithm of the NH4/NO3 uptake ratio. Further, this uptake ratio was related to the NH4/NO3 ratio of the soil solution. From these relationship it was estimated that Scots pine exhibits an acidifying uptake pattern as long as the contribution of nitrate to the N nutrition is lower than 70%. Under field circumstances root uptake may cause soil acidification in the topsoil, containing the largest part of the root system, and soil alkalization in deeper soil layers.

1. Nutrient diffusing substrata (NDS) were used to determine the relative importance of nutrients and light as potential limiting factors of periphyton biomass and nitrogen (N) uptake in Mediterranean streams subjected to different human impacts. The nutrients examined were phosphorus (P) and N, and we also further differentiated between the response of periphyton communities to N species (i.e. NO3‐N and NH4‐N). To examine the effect of light and nutrients on periphyton biomass, chlorophyll a accrual rates on NDS located at open and closed canopy sites were compared. The effect of nutrient availability on periphyton uptake was measured by 15N changes on the NDS after NO315N short‐term nutrient additions. 2. Results show that light was the main factor affecting algal biomass in the study streams. Algal biomass was in general higher at open than at closed canopy sites. Nutrient availability, as simulated with the NDS experiments, did not enhance algal biomass accrual in either of the 2 light conditions. 3. In the control treatments (i.e. ambient concentrations), periphyton NO3‐N uptake rates increased and C : N molar ratios decreased consistently with increases in N availability across streams. NO3‐N uptake rates were altered when ambient N concentrations were increased artificially in the N amended NDS. Periphyton assemblages growing on N enriched substrata seemed to preferentially take up N diffusing from the substratum rather than N from the water column. This response differed among streams, and depended on ambient N availability. 4. Periphyton biomass was not significantly different between substrata exposed to the two forms of available N sources. Nonetheless, we found differences in the effects of both N sources on the uptake of N from the water column. NH4‐N seemed to be the preferred source of N for periphyton growing on NDS. 5. Results suggest that the effect of riparian zones on light availability, although seldom considered by water managers, may be more important than nutrients in controlling eutrophication effects derived from human activities. Finally, our results confirm that not only increases in concentration, but also stoichiometric imbalances should be considered when examining N retention in human altered streams.

An investigation was untertaken to evaluate the nutrient status of the River Rhine (two stations) and eight of its tributaries (total of ten samplings). Determinations of the following inorganic substances were made: PO4 ?3-P; NO3 ?-N; NO2 ?-N; NH4 + -N and Cl?. In addition, pH and carbonate alkalinity were measured. Bioassays to obtain the algal growth potential (AGP) were carried out using periphyton from the River Rhine. A linear relationship could be established between NO3 ?-N and the AGP, while the AGP showed a non-linear dependence on the PO4 3?-P concentration. The critical N/P ratio for N or P limitation of the algal growth in bioassays was evaluated graphically and by calculation. The results of the two methods are in good agreement: N is the limiting factor at NO3 ?N/PO4 3?-P ratios less than 10, while P is limiting at ratios greater than 20. At values between 10 and 20 neither N nor P can be supposed with certainty to be limiting.

The growth of Salvinia molesta D.S. Mitchell was studied in a greenhouse using controlled-temperature water-baths at 16, 19 and 22°C and 4 different nitrogen compounds (NO3?, NH4+, NH4NO3 and urea) at levels up to 60 mg N l?1. Little growth occurred at 16°C even if 20 mg N l?1 was supplied together with other nutrients including phosphorus (2 mg H2PO4-P l?1). The highest relative growth rate and total dry matter production occurred at 22°C when plants were supplied with 20 mg NH4-N l?1. At this temperature, the NH4+ ion was superior to the NO3? ion or urea as a nitrogen source (almost doubling the biomass), but was not significantly better than NH4NO3. Over a period of 19 days for plants receiving 0.02 mg NH4-N l?, biomass increased 4-fold at 16°C, 9-fold at 19°C and 10-fold at 22°C. In contrast, for plants receiving 20 mg NH4-N l?1, biomass increased 4-fold at 16°C, 18-fold at 19°C and 38-fold at 22°C.

We have developed a strategy for the optimization of plant cell suspension culture media using a combination of fractional factorial designs (FFDs) and response surface methodology (RSM). This sequential approach was applied to transformed tobacco BY‐2 cells secreting a human antibody (M12) into the culture medium, in an effort to maximize yields. We found that the nutrients KNO3, NH4NO3 and CaCl2 and the hormones 2,4‐dichlorophenoxyacetic acid (2,4–D) and 6‐benzylaminopurine (BAP) had the most significant impact on antibody accumulation. The factorial screening revealed strong interactions within the nutrients group (KNO3, NH4NO3 and CaCl2) and also individually between 2,4‐D and three other components (KNO3, NH4NO3 and BAP). The RSM design resulted in a fivefold increase in the antibody concentration after 5 days and a twofold reduction in the packed cell volume (PCV). Longer cultivation in the optimized medium led to the further accumulation of antibody M12 in the culture medium (up to 107 μg/mL, day 10). Because the packed cell volume was reduced in the optimized medium, this enhanced the overall yield by 20‐fold (day 7) and 31‐fold (day 10) compared to the conventional MS medium.

Bioremediation of diesel-oil-contaminated alpine soils at low temperatures
Bioremediation of two diesel-oil-contaminated alpine subsoils, differing in soil type and bedrock, was investigated in laboratory experiments at 10 °C after supplementation with an inorganic fertilizer. Initial diesel oil contamination of 4000 mg kg−1 soil dry matter (dm) was reduced to 380–400 mg kg−1 dm after 155 days of incubation. In both soils, about 30 % of the diesel oil contamination (1200 mg kg−1 dm) was eliminated by abiotic processes. The residual decontamination (60 %–65 %) could be attributed to microbial degradation activities. In both soils, the addition of a cold-adapted diesel-oil-degrading inoculum enhanced biodegradation rates only slightly and temporarily. From C/N and N/P ratios (determined by measuring the contents of total hydrocarbons, NH4 + N, NO3 N and PO4 3− P) of soils␣it could be deduced that there was no nutrient deficiency during the whole incubation period. Soil biological activities (basal respiration and dehydrogenase activity) corresponded to the course of biodegradation activities in the soils. When NH4 + or NO3 ? was supplied to NO3 ? ‐stressed cells of the microalga Dunaliella tertiolecta Butcher, immediate transient changes in chl a fluorescence were observed over several minutes that were not seen in N‐replete cells. These changes were predominantly due to nonphotochemical fluorescence quenching. Fluorescence changes were accompanied by changes in photosynthetic oxygen evolution, indicating interactions between photosynthesis and N assimilation. The magnitude of the fluorescence change showed a Michaelis‐Menten relationship with half‐saturation concentration of 0.5 μM for NO3 ? and 10 μM for NH4 + . Changes in fluorescence responses were characterized in D. tertiolecta both over 5 days of N starvation and in cells cultured at a range of NO3 ? ‐limited growth rates. Variation in responses was more marked in starved than in limited cells. During N starvation, the timing and onset of the fluorescence responses were different for NO3 ? versus NH4 + and were correlated with changes in maximum N uptake rate during N starvation. In severely N‐starved cells, the major fluorescence response to NO3 ? disappeared, whereas the response to NH4 + persisted. N‐starved cells previously grown with NH4 + alone showed fluorescence responses with NH4 + but not NO3 ? additions. The distinct responses to NO3 ? and NH4 + may be due to the differences between regulation of the uptake mechanisms for the two N sources during N starvation. This method offers potential for assessing the importance of NO3 ? or NH4 + as an N source to phytoplankton populations and as a diagnostic tool for N limitation.

Across northern Alberta, Canada, bogs experience periodic wildfire and, in the Fort McMurray region, are exposed to increasing atmospheric N deposition related to oil sands development. As the fire return interval shortens and/or growing season temperatures increase, the regional peatland CO2–C sink across northern Alberta will likely decrease, but the magnitude of the decrease could be diminished if increasing atmospheric N deposition alters N cycling in a way that stimulates post-fire successional development in bogs. We quantified net ammonification, nitrification, and dissolved organic N (DON) production in surface peat along a post-fire chronosequence of five bogs where we also experimentally manipulated N deposition (no water controls plus 0, 10, and 20 kg N ha?1 yr?1 simulated deposition, as NH4NO3). Initial KCl-extractable NH4+–N, NO3?–N and DON averaged 176?±?6, 54?±?0.2, and 3580?±?40 ng N cm?3, respectively, with no consistent changes as a function of time since fire and no consistent effects of experimental N addition. Net ammonification, nitrification, and DON production averaged 3.8?±?0.3, 1.6?±?0.2, and 14.3?±?2.0 ng N cm?3 d?1, also with no consistent changes as a function of time since fire and no consistent effects of experimental N addition. Our hypothesis that N mineralization would be stimulated after fire because root death would create a pulse of labile soil organic C was not supported, most likely because ericaceous plant roots typically are not killed in boreal bog wildfires. The absence of any N mineralization response to experimental N addition is most likely a result of rapid immobilization of added NH4+–N and NO3?–N in peat with a wide C:N ratio. In these boreal bogs, belowground N cycling is likely characterized by large DON pools that turn over relatively slowly and small DIN pools that turn over relatively rapidly. For Alberta bogs that have persisted at historically low N deposition values and begin to receive higher N deposition related to anthropogenic activities, peat N mineralization processes may be largely unaffected until the peat C:N ratio reaches a point that no longer favors immobilization of NH4+–N and NO3?–N.

The effect of two N-forms (NH4 + and NO3 ) and NaCl on pattern of accumulation of some essential inorganic nutrients was examined in sunflower (Helianthus annuus L.) cv. Hisun 33. Eight-day-old plants of were subjected for 21 d to Hoagland's nutrient solution containing 8 mM N as NH4 + or NO3 ·, and salinized with and addition of NaCl to the growth medium had no significant effect on total leaf N. However, root N of NH4-supplied plants decreased significantly with increase in NaCl concentration, whereas that of NO3-supplied plants remained unaffected. There was no significant effect of NaCl on leaf or root P, but the NO3-supplied plants had significa concentration of leaf P than that of NH4-supplied plants at varying salt treatments. Salinity of the rooting med did not show any significant effect on Na+ concentrations of leaves or roots of plants subjected to two differen N. NH4-treated plants generally had greater concentrations of Cl in leaves and roots and lower K+ content in leaves than NO3-supplied plants. Ca2+ concentrations of leaves and roots and Mg2+ concentrations of leaves decreased in NH4-supplied plants due to NaCl, but they remained unaffected in NO3-treated plants.

1. While phosphorus (P) is often considered the most important growth limiting factor for plants in lakes, recent studies of shallow lakes indicate that nitrogen (N) may be of greater importance than realized hitherto and that submerged macrophytes may be lost when the N concentration exceeds a certain threshold, as long as the concentration of P is sufficiently high. 2. We studied the effects of different loadings of NH4‐N and NO3‐N on chlorophyll a and on a macrophyte tolerant of eutrophication, Vallisneria spinulosa (Hydrocharitaceae). In outdoor mesocosms we used water from a pond as control and created four levels of NH4‐N and NO3‐N (approximately 2.5, 5, 7.5 and 10 mg L−1) by dosing with NH4Cl and NaNO3, respectively. After the experiment, the plants were transferred back to a holding pond to study their recovery. In contrast to previous research, we used a low background concentration of phosphorus (TP 0.024 ± 0.003 mg L−1) so we could judge whether any effects of N were apparent when P is in short supply. 3. Chlorophyll a increased significantly with N dosing for both forms of N, but the increase was highest in the NH4‐N dosed mesocosms (maximum 58 μg L−1 versus 42 μg L−1 in the NO3‐N mesocosms), probably due to a higher total inorganic N concentration (part of the added ammonia was converted to nitrate during the experiment). 4. Although the number of ramets of V. spinulosa was not affected by the N treatment, the biomass increased up to concentrations of 7.5 mg L−1, while biomass at 10 mg L−1 remained at the control level for both N ions treatments. A similar pattern was apparent for the content of N and soluble sugar of the plant, while there were no differences in the plant P content among treatments. Five months after transplantation back to the pond no difference was found in the number of ramets or in biomass, except that the biomass of plants grown at 10 mg N L−1 during the experiment was greater than that in the control, while the N and P contents of plants were similar to those of the controls. 5. Nitrogen concentration had little influence on the growth of the eutrophication tolerant submerged macrophyte at moderately low concentrations of phosphorus. Moreover, the two N ions showed no toxic effects, suggesting that loss of macrophytes observed in other studies, run at higher phosphorus concentrations, was probably related to enhanced shading by periphyton and/or phytoplankton rather than to any toxic effects of N.

Plant debris, naturaiiy infested with the take-all fungus (Ophiobolus graminis), was washed from soil and added to a leached sandy loam, deficient in nitrate nitrogen (NO3-N) and magnesium. Nutrient solutions containing potassium and phosphorus, with and without magnesium, were added to the amended soil unsupplemented, or with either NO3-N, ammonium nitrogen (NH4+-N), or both. Nitrification of NH4+-N was inhibited by 2–chloro-6–(trichloromethy1)-pyridine (N-Serve). After 38 days at 19°C, fewer plants had take-all with N (75 or 100 mg/kg soil) than without and root systems were most discoloured and had most diseased axes when nutrients were not added. Plants given NH4+-N developed less take-all when magnesium was present. A comparison of forms of N in the presence of added magnesium showed that take-all was least with a mixture of both forms of N, intermediate with NO3-N alone and worst with NH4+-N alone. The most extensive lesions on individual root axes occurred on plants given NH4+-N. It is suggested that take-all will be least when the amounts and ratio of NH4+-N and NO3-N are optimum for the growth of the host.

The assimilation of N‐NO3? requires more energy than that of N‐NH4+. This becomes relevant when energy is limiting and may impinge differently on cell energy budget depending on depth, time of the day and season. We hypothesize that N‐limited and energy‐limited cells of the oceanic cyanobacterium Synechococcus sp. differ in their response to the N source with respect to growth, elemental stoichiometry and carbon allocation. Under N limitation, cells retained almost absolute homeostasis of elemental and organic composition, and the use of NH4+ did not stimulate growth. When energy was limiting, however, Synechococcus grew faster in NH4+ than in NO3? and had higher C (20%), N (38%) and S (30%) cell quotas. Furthermore, more C was allocated to protein, whereas the carbohydrate and lipid pool size did not change appreciably. Energy limitation also led to a higher photosynthetic rate relative to N limitation. We interpret these results as an indication that, under energy limitation, the use of the least expensive N source allowed a spillover of the energy saved from N assimilation to the assimilation of other nutrients. The change in elemental stoichiometry influenced C allocation, inducing an increase in cell protein, which resulted in a stimulation of photosynthesis and growth.
