A cell suspension culture of Devil’s claw (Harpagophytum procumbens), a South African plant with high medicinal value, cultivated under submerged conditions showed stable growth and accumulated high amounts of biomass (18.2 g l−1). Flow cytometry analyses of the suspension’s cell cycle kinetics showed that proportions of cells in G0/G1 and S phases varied insignificantly (between 69–76% and 9–13%, respectively) during the cultivation, while the proportion of G2/M-phase cells increased until day 8 of cultivation, when the exponential phase of cell growth ended. Metabolite production in the culture was studied through simultaneous determination of three bioactive phenylethanoid glycosides (verbascoside, β-OH-verbascoside and leucosceptoside A) by high performance liquid chromatography. It was found that suspended Devil’s claw cells accumulated mainly verbascoside (517.3 mg l−1), followed by leucosceptoside A (107.1 mg l−1) and β-OH-verbascoside (80.3 mg l−1). In addition, several fatty acids and β-sitosterol were identified in the cell suspension by gas chromatographic-mass spectrometry analysis. Comparison of the results with previously acquired data for Harpagophytum procumbens transformed roots indicate that cell suspensions cultures are more promising as potential commercial sources of metabolites such as phenylethanoid glycosides.  相似文献

Devil's Claw (Harpagophytum procumbens) a plant native to Southern Africa, has historically been used in traditional medicine to treat a wide range of diseases and currently is widely employed as anti-inflammatory and pain-relieving natural remedy in Europe and other parts of the world.Aim of the studyLittle is known about possible herb-drug interactions arising from effects of Devil's Claw on the major drug metabolizing enzymes or transporters. This study evaluated in vitro the effects of Devil's Claw on the multidrug transporter ABCB1/P-glycoprotein.Materials and methodsThe effects of three commercially available Devil's Claw preparations and that of pure harpagoside were studied in the human kidney (HK-2) proximal tubule cell line, constitutively expressing ABCB1/P-glycoprotein (P-gp). Pgp activity and expression were tested by the calcein-AM test and by Western blotting, respectively.ResultsCommercial preparations inhibited P-gp activity, even if to a different extent, while pure harpagoside was almost ineffective. In cells cultured for three days in the presence of Devil's Claw preparations or pure harpagoside, a dose-dependent P-gp upregulation was found.ConclusionsOur results demonstrate for the first time that Devil's Claw may interact with the multidrug transporter ABCB1/P-gp, the effect not appearing strictly related to the harpagoside relative content. Modulation of both P-gp activity and P-gp expression by Devil's Claw raise the possibility of herb-drug interactions, to be further explored in depth.  相似文献

Ghrelin is a stomach-derived peptide that has been identified as the only circulating hunger hormone that exerts a potent orexigenic effect via activation of its receptor, the growth hormone secretagogue receptor (GHS-R1a). Hence, the ghrelinergic system represents a promising target to treat obesity and obesity-related diseases. In this study we analysed the GHS-R1a receptor activating potential of Harpagophytum procumbens, popularly known as Devil''s Claw, and its effect on food intake in vivo. H. procumbens is an important traditional medicinal plant from Southern Africa with potent anti-inflammatory and analgesic effects. This plant has been also used as an appetite modulator but most evidences are anecdotal and to our knowledge, no clear scientific studies relating to appetite modulation have been done to this date. The ghrelin receptor activation potential of an extract derived from the dried tuberous roots of H. procumbens was analysed by calcium mobilization and receptor internalization assays in human embryonic kidney cells (Hek) stably expressing the GHS-R1a receptor. Food intake was investigated in male C57BL/6 mice following intraperitoneal administration of H. procumbens root extract in ad libitum and food restricted conditions. Exposure to H. procumbens extract demonstrated a significant increased cellular calcium influx but did not induce subsequent GHS-R1a receptor internalization, which is a characteristic for full receptor activation. A significant anorexigenic effect was observed in male C57BL/6 mice following peripheral administration of H. procumbens extract. We conclude that H. procumbens root extract is a potential novel source for potent anti-obesity bioactives. These results reinforce the promising potential of natural bioactives to be developed into functional foods with weight-loss and weight maintenance benefits.  相似文献

Harpagoside is an iridoid glycoside that was first isolated from Harpagophytum procumbens (devil’s claw, Pedaliaceae), a medicinal plant in which it is the major constituent of the iridoid pool. Both the pure compound and devil’s claw extracts have potent anti-rheumatic, anti-inflammatory and analgesic effects. According to the European Pharmacopoeia commercial devil’s claw products should contain at least 1.2% harpagoside. However, the compound has also been isolated from several other plant species and in vitro plant culture systems. Recent advances in knowledge of harpagoside distribution, biosynthesis/accumulation and pharmacology are summarized in this review. We also discuss the possible synergism and/or antagonism between major constituents in harpagoside-containing phytopharmaceutical products. Finally, future perspectives for its potential application are highlighted.  相似文献

In a previous report, we showed that cell cultures of Harpagophytum procumbens, a South African plant with high medicinal value, accumulate high amounts of anti-inflammatory phenylethanoid glycosides during cultivation in shake-flasks. The aim of the present study was to transfer the phenylethanoid biosynthetic process to a 3-L stirred tank reactor and a 1-L glass-column bioreactor (operated with pulsed aeration). We found that, with stepwise increases in aeration, the stirred tank reactor yielded similar productivities of verbascoside (the major phenylethanoid glycoside in the cells) to those reported for shake-flask cultures (55.68 vs. 54.78 mg verbascoside/L/day, respectively). Transfer in the pulse-aerated column reactor resulted in 165.42 mg verbascoside/L/day, one of the highest yields reported to date. Further, to evaluate the physiological status of the suspended cells in the bioreactors cultures, we examined their hormone levels and compared them to those of cells in shake-flask cultures. While indole-3-acetic acid levels did not differ significantly between the bioreactor and shake-flask cultures, there were considerable differences in their levels of abscisic, jasmonic, and salicylic acids. These results are discussed with respect to relative stress levels in the different cultivation systems.  相似文献

Devil''s Claw (Harpagophytum procumbens), an herbal product being marketed in Canada as a home remedy for the relief of arthritic disease, was screened for efficacy with standard preclinical screening methods. At doses 100 times or greater than the recommended daily dose for humans, Devil''s Claw was completely ineffective in reducing edema of the rat hind foot induced by either lambda-carrageenan or Mycobacterium butyricum. At concentrations of up to 1 x 10(5) microgram/ml, Devil''s Claw was also ineffective as an in-vitro inhibitor of prostaglandin synthetase. These results indicate that Devil''s Claw lacks the anti-inflammatory properties possessed by all antiarthritic drugs of the nonsteroidal, anti-inflammatory analgesic type.  相似文献

Harpagophytum procumbens, popularly known as devil’s claw, is a plant commonly used in the treatment of diseases of inflammatory origin. The anti-inflammatory effects of H. procumbens have been studied; however, the mechanism of action is not elucidated. It is known that excess of reactive oxygen and nitrogen species may contribute to increasing tissue damage due to inflammation. In the present study, we examined the effects of H. procumbens infusion, crude extract and fractions on lipid peroxidation (brain homogenates) induced by different pro-oxidants (Fe2+ or sodium nitroprusside) and the effects of ethyl acetate fraction (rich in phenolic compounds) on antioxidant defenses (catalase activity and thiol levels) and cell damage (brain cortical slices) induced by different pro-oxidants. All tested extracts of H. procumbens inhibited lipid peroxidation in a concentration-dependent manner. Furthermore, the ethyl acetate fraction had the highest antioxidant effects either by decreasing lipid peroxidation and cellular damage or restoring thiols levels and catalase activity. Taken together, our results showed that H. procumbens acts either by preventing oxidative stress or loss of cell viability. Thus, the previously reported anti-inflammatory effect of H. procumbens could also be attributed to its antioxidant activity.  相似文献

Harpagophytum procumbens (Burch.) DC. ex Meisn. subsp. procumbens (Pedaliaceae) is an important African medicinal plant growing in the Kalahari region of southern Africa. This species, together with its close taxonomic ally Harpagophytum zeyheri are collectively referred to as Devil's Claw and are used interchangeably for the treatment of inflammation-related disorders. Although the two taxa are botanically and chemically similar, H. zeyheri contains lower levels of harpagoside and these two species have not been proven to exhibit equipotent pharmacological activity. Due to these taxonomic similarities, effective quality control methods are required to distinguish between the two species. Differentiation between the two species was achieved using single point mid-infrared spectroscopy in combination with chemometric data analysis. The orthogonal projections to latent structures discriminant analysis (OPLS-DA) model had good predictive ability, as illustrated by the model statistics: R2X (cum predictive + orthogonal) = 0.86 and Q2 (cum) = 0.63. Short wave infrared (SWIR) hyperspectral imaging could distinguish between the two species with acceptable model statistics: R2X and R2Y of 0.99 and 0.78, respectively. This study demonstrated that both MIR single point spectroscopy and SWIR hyperspectral imaging coupled with chemometric modelling are reliable and rapid methods to determine the authenticity of Harpagophytum spp.  相似文献

Two‐phase bioreactor cultivation system was developed and applied for in sito recovery of extracellular galanthamine during the cultivation of Leucojum aestivum L. shoot culture in a modified column bioreactor system. The inclusion of an external circulation column with adsorbent resin Amberlite XAD‐4 as a second phase, on the 21st day of the beginning of cultivation resulted in 1.25 folds increase in biomass accumulation and maximal amounts of accumulated galanthamine of 6 mg/L (3.1 mg/L intracellular and 2.9 mg/L extracellular). It was demonstrated that the inclusion of a second phase at the cultivation of the L. aestivum shoot culture in a bubble column bioreactor with internal sections redirected the alkaloid metabolism to galanthamine synthesis and inhibits the synthesis of hemanthamine and lycorine type alkaloids. Our research demonstrated that the application of the two‐phase cultivation systems could be an important tool to increase the yields of valuable secondary metabolites in plant tissue culture‐based bioprocess.  相似文献

Gynura procumbens (Lour.) Merris one of medicinal plant which was carried out used as antioxidant, anticancer, anti-inflammatory, hepatoprotective, and antimicrobial. Many strategies were used to increase the production of biomass and valuable compounds. This study was to investigate the variation effect of growth regulators and immersion frequency on production of biomass and flavonoid contained of G. procumbens shoots culture in temporary immersion bioreactor. Stem nodes were used as an explants and induction of shoots were done in solid MS medium supplemented with many kinds of growth regulator. The best treatments were used to produce biomass and flavonoid compounds in temporary immersion bioreactor; there are combination of IAA 2?mg/L and BA 4, 6, 8?mg/L and immersion frequency (5 min each 3?h; 15?min each 12?h). Results showed that the growths of G. procumbens shoots in solid MS medium were influenced by supplementation of growth regulators. MS medium supplemented with single cytokinine (6 mg/L kinetin) and combination of auxin (IAA) and cytokinine (BA) caused increasing of shoots growth. Production of biomass of G. procumbens in temporary immersion bioreactor was achieved in long immersion interval (12?h) and highest flavonoid production was obtained in combination treatment of immersion frequency 15?min each 12?h and MS medium supplemented with IAA 2?mg/L, BA 8?mg/L.  相似文献

This study investigated factors affecting transplantation success of native forest understory vegetation on metal‐contaminated soils. One year after transplantation along a gradient of historical Cu–Ni smelter pollution in Sudbury, Canada, community‐level characteristics of the transplanted plots and short‐term indicators expressing the performance of selected species were assessed. Thirty 16‐m2 plots were studied, each established with 45 transplanted 0.56 × 0.64?m turfs 10 cm thick, extracted from mixed‐hardwood forests slated for development. Species richness, diversity, and evenness were not affected by environmental conditions but short‐term responses of root growth and sexual reproduction of selected species to environmental variables indicate a need for long‐term monitoring. Root growth of Clintonia borealis (Aiton) Raf. and Gaultheria procumbens L. was positively related to soil temperature. Root growth of G. procumbens correlated negatively with soil Ni availability, but in all plots, root growth was comparable with the literature values from unaffected forests. Flowering frequency of G. procumbens correlated negatively with soil pH and positively with tree cover, corresponding to ecological requirements of this species. Soil had a negative effect on sexual reproduction of C. borealis. Unexpectedly, fruiting of C. borealis responded positively to Ni, and fruiting of Maianthemum canadense Desf. positively to As, possibly due to interdependencies among soil variables. The results are encouraging with respect to transplant success, as effects of smelter‐related variables were relatively minor, but species‐specific responses of the selected species to environmental factors indicate that species performance is dependent on site‐specific conditions, potentially influencing long‐term success of the transplants.  相似文献

The time courses of growth and rosmarinic acid production by Lavandula vera MM cell suspension were investigated. The uptake of the main nutrients (sucrose, nitrogen, phosphorus, K, Ca, Mg) was followed during cultivation and the data on the physiology of the L. vera MM cell culture are presented. It was established that the cell culture synthesizes rosmarinic acid during the linear phase of growth for a relatively short period (between the 4th and 8th days of cultivation). The influence of sucrose concentration in the nutrient medium on cell growth and accumulation of rosmarinic acid by L. vera MM cell culture was investigated. The results showed that 7% sucrose in the nutrient medium ensured a steady growth of the cell suspension and increased the yield of rosmarinic acid (29.2 g/l dry biomass and 507.5 mg/l rosmarinic acid compared to 13.0 g/l dry biomass and 68.6 mg/l rosmarinic acid for the control cultivation with 3% sucrose). New biomass sources for alternative fuels has become a subject of increasing importance as the nation strives to resolve the economic and strategic impacts of limited fossil fuel resources on our national security, environment, and global climate. Algae are among the most promising non‐food‐crop‐based biomass feedstocks. However, there are currently no commercially viable microalgae‐based production systems for biofuel production that have been developed, as limitations include less‐than optimal oil content, growth rates, and cultivation techniques. While batch studies are critical for determining basic growth phases and characteristics of the algal species, steady‐state studies are necessary to better understand and measure the specific growth parameters. This study evaluated the effects of dilution rate on microalgal biomass productivity, lipid content, and fatty acid profile under steady‐state conditions with continuous illumination and carbon dioxide supplemention for two types of algae. Continuous cultures were conducted for more that 3 months. Our results show that the productivity of Chlorella minutissima varied from 39 to 137 mg/L/day (dry mass) when the dilution rate varied from 0.08 to 0.64 day?1. The biomass productivity of C. minutissima reached a maximum value (137 mg/L/day) at a dilution rate of 0.33 day?1, while the productivity of Dunaliella tertiolecta varied from 46 to 91 mg/L/day at a dilution rate of 0.17 to 0.74 day?1. The biomass productivity of D. tertiolecta reached a maximum value of 91 mg/L/day at a dilution rate of 0.42 day?1. Moreover, the lipid content had no significant change with various dilution rates. 为探明爵床(Justicia procumbens)甲醇提取物对小菜蛾的生物活性,采用室内生测法测定了爵床甲醇提取物对小菜蛾的触杀、拒食、胃毒、生长发育抑制和产卵忌避作用。结果表明,爵床甲醇提取物对小菜蛾幼虫具有较强的触杀、拒食、胃毒和生长发育抑制活性,对小菜蛾成虫具有较强的产卵忌避活性。在触杀试验中,药后1、2 d和3 d爵床甲醇提取物对小菜蛾3龄幼虫的致死中浓度(LC50)分别为5.17、4.05和3.06 mg/m L;在拒食试验中,药后1 d和2 d提取物对3龄幼虫的选择性拒食中浓度(AFC50)分别为2.64和3.13 mg/m L,药后1 d和2 d提取物对3龄幼虫的非选择性拒食中浓度(AFC50)分别为3.70、4.54 mg/m L;在胃毒试验中,药后4、5、6 d和7 d提取物对3龄幼虫的致死中浓度(LC50)分别为8.13、3.65、2.88、2.23 mg/m L;在生长发育抑制试验中,药后1 d和2 d提取物对3龄幼虫的抑制中浓度(IC50)分别为2.02、1.40 mg/m L;在产卵忌避试验中,药后1、2 d和3 d提取物对小菜蛾成虫的选择性产卵忌避中浓度(AOC50)分别为2.61、3.66、4.58 mg/m L,药后1、2和3 d提取物对小菜蛾成虫的非选择性产卵忌避中浓度(AOC50)分别为3.19、4.52、5.65 mg/m L。由此证实,爵床提取物对小菜蛾具有显著的毒杀活性,具有开发为新型高效、低毒植物源农药的潜在价值。  相似文献

Determining the relative contributions of different ecological factors for herbivore fitness is one key to understanding the ecology and evolution of host plant choice by herbivores. Natural enemies are increasingly being recognized as an important factor: host plants of inferior quality for development may still be used by herbivores if they provide enemy‐free space (EFS). Here we used the tobacco hornworm, Manduca sexta, to experimentally disentangle the effects of natural enemies from the potentially confounding factors of host plant quality, competition and microhabitat. We explored the consequences for both individual components of fitness and total fitness of M. sexta feeding on a typical high quality host plant, tobacco Nicotiana tabacum and a novel, low quality host plant, devil's claw Proboscidea louisianica in an experimental field environment in the presence of a parasitoid natural enemy, Cotesia congregata. Although early larval survival, development and growth rates, final body size and fecundity were all reduced for M. sexta feeding on devil's claw, a high rate of parasitism on tobacco and an absence of parasitism on devil's claw contributed to similar total fitness (net reproductive rate, R0) across the two host plant species. Our results suggest M. sexta has adopted a novel host plant (devil's claw) outside its typical host range because this host plant provides enemy free space. In addition, oviposition behavior of adult female M. sexta appears to be well suited to exploiting the enemy‐free space on devil's claw; oviposition by M. sexta on devil's claw appears to correspond with seasonal variation in parasitoid abundance.  相似文献

Harpagophytum procumbens is a medicinal plant containing several compounds with pharmaceutical activity. Previously, we established shoot culture and in vitro regenerated plants of H. procumbens. In this study, HPLC and LC-ESI-MS were used to identify harpagoside, harpagide, verbascoside and isoverbascoside in various tissues (stems, leaves and callus) of shoots multiplied on Schenk and Hildebrandt (SH) solid medium supplemented with 0.57 μM indole-3-acetic acid (IAA) and 8 μM 6-benzylaminopurine (BAP), as well as in stems, leaves and root tubers of in vitro propagated plants grown in the greenhouse for 3, 6 and 12 months. The content of the compounds was also determined by HPLC. For comparison, control H. procumbens plants initiated from seeds were analyzed. H. procumbens shoots grown under in vitro conditions accumulated lower amounts of iridoids and phenylethanoids than the plants derived from them. The levels of analyzed compounds were higher in the organs of 3- or 6-month-old plants than in those of 12-month-old plants. Differences in the distribution of secondary metabolites were also observed between organs. The aerial parts (stems, leaves) of 3-month-old in vitro regenerated plants were characterized by the highest amounts of phenylethanoids, which significantly exceeded those detected in control plants. Total iridoid content, calculated as the sum of harpagoside and harpagide, was highest in the root tubers of 6-month-old plants. In these organs the level of harpagoside was comparable to that in root tubers of 6-month-old seed-propagated plants, but the level of harpagide was much lower.  相似文献

Summary The growth of Lavandula vera MM cell suspension and biosynthesis of rosmarinic acid were followed during its cultivation in Linsmayer - Skoog (LS) medium (containing 170 mg/L KH2PO4, control cultivation) and in modified LS media, containing different concentration of KH2PO4. Doubled concentration of KH2PO4 (340 mg/L) caused an increase in the amount of biosynthesised cell biomass (17 g/L vs 13 g/L for control cultivation) and rosmarinic acid (140 mg/L vs 68 mg/L for control cultivation).  相似文献

Effects of experimental warming on shoot developmental growth and biomass production were preliminarily investigated in two evergreen dwarf shrubs Empetrum nigrum and Loiseleuria procumbens, using the International Tundra Experiments open-top chamber (OTC) method, in the Tateyama Range, central Japan. An OTC was installed over shrub (E. nigrum and L. procumbens) -dominated vegetation and over shrub-forb (such as Anemone narcissiflora var. nipponica and Solidago virga-aurea ssp. leiocarpa) mixed vegetation, and stem samples of the evergreen shrubs were obtained at 26 months after installing the OTC. The OTC increased the daily mean temperature by 0.1°C to 1.8°C, on average, during the growing season. Shoot developmental growth and biomass production were considerably different between species of different vegetation types. The boreal species E. nigrum generally showed better growth inside the OTC than the arctic and subarctic species L. procumbens. Both species showed significantly larger shoot elongation and biomass production inside the OTC over shrub-dominated vegetation, whereas smaller or reduced growth was detected inside the OTC over shrub-forb mixed vegetation. The variations of growth responses to warming between species of different vegetation types are discussed, especially in relation to interspecific competition under a simulated environmental change.  相似文献

A previous study showed that the contents of caffeoylquinic acids and iridoids, the major bioactive components in the postharvest Lonicera japonica Thunb., were induced by enhanced ultraviolet (UV)‐A or UV‐B irradiation. To clarify the UV‐responsive key enzymes in the bioactive metabolites biosynthetic pathway and the related plant defense mechanism in L. japonica, 2DE in combination with MALDI‐TOF/TOF MS was employed. Seventy‐five out of 196 differential proteins were positively identified. Based on the functions, these proteins were grouped into nine categories, covering a wide range of molecular processes including the secondary metabolites (caffeoylquinic acids and iridoids) biosynthetic‐related proteins, photosynthesis, carbohydrate and energy metabolism, stress, DNA, transport‐related proteins, lipid metabolism, amino acid metabolism, cell wall. Of note is the increasing expression of 1‐deoxy‐d ‐xylulose 5‐phosphate reductoisomerase and 5‐enol‐pyruvylshikimate‐phosphate synthase, which was crucial to supply more precursor for the secondary metabolites including caffeoylquinic acids and iridoids. Thus, this study provides both the clues at the protein level for the increase of the two bioactive components upon UV irradiation and the profile of UV‐responsive proteins in L. japonica.  相似文献

Grapes are rich in phytochemicals with many proven health benefits. Phenolic profiles, antioxidant and antiproliferative activities of twenty-four selected Vitis vinifera grape cultivars were investigated in this study. Large ranges of variation were found in these cultivars for the contents of total phenolics (95.3 to 686.5 mg/100 g) and flavonoids (94.7 to 1055 mg/100 g) and antioxidant activities (oxygen radical absorbance capacity 378.7 to 3386.0 mg of Trolox equivalents/100 g and peroxylradical scavenging capacity14.2 to 557 mg of vitamin C equivalents/100 g), cellular antioxidant activities (3.9 to 139.9 µmol of quercetin equivalents/100 g without PBS wash and 1.4 to 95.8 µmol of quercetin equivalents /100 g with PBS wash) and antiproliferative activities (25 to 82% at the concentrations of 100 mg/mL extracts).The total antioxidant activities were significantly correlated with the total phenolics and flavonoids. However, no significant correlations were found between antiproliferative activities and total phenolics or total flavonoids content. Wine grapes and color grapes showed much higher levels of phytochemicals and antioxidant activities than table grapes and green/yellow grapes. Several germplasm accessions with much high contents of phenolics and flavonoids, and total antioxidant activity were identified. These germplasm can be valuable sources of genes for breeding grape cultivars with better nutritional qualities of wine and table grapes in the future.  相似文献
