Many organisms use proton pump to earn energy for living. Some proton pumps start to work by light and one of the famous proteins are called proteorhodopsin (PR). From recent study it used not only protons but also mono-valent cations, divalent cations, or mono-valent anions during pumping activity. The goal of this study is to find new types of proton pumping proteins in the surface of the ocean. Metagenome samples were collected from the beach in Taean-gun and Incheon (Kkotji beach (36°30′0′′N, 126°19′56′′E), Kkotji mud (36°30′8′′N, 126°19′60′′E), Duegi beach (36°31′6′′N, 126°19′39′′E), Sorae salt pond (37°24′25′′N, 126°44′41′′E), swamp (37°24′59′′N, 126°44′54′′E) and reservoir (37°24′39′′N, 126°45′5′′E) in West Sea of Korea. Genomic DNA of each sample was isolated and used for PCR with specific primers for PR and sodium pumping rhodopsin. As a result, we obtained an unidentified PR in Duegi beach sample. The unidentified PR was expressed with chimeric expression system. It has 528 nm absorption maximum at pH 7. By the light differential spectrum measurement, putative M and O photo-intermediates were detected at around 400 and 600 nm, respectively. Length–weight relationship (LWR) was estimated for 12 elasmobranch species; five shark species, four species of rays and three species of guitar fishes from north‐eastern Arabian Sea, India. Five major landing centres of Maharashtra were selected; Satpati (Lat. 19°43′15″N, Long. 72°42′00″E), Naigaon (Lat. 19°19′32″N, Long. 72°48′54″E), Versova (Lat. 19°08′33″N, Long. 72°48′11″E), New ferry Wharf (Lat. 18°57′29″N Long. 72°51′01″E) and Sassoon dock (Lat. 18°54′42″N, Long. 72°49′33″E). Samples were collected fortnightly during August 2016 to October 2017 from various gears; drift gill nets (Hung length 114–143 m and #100–270 mm) off Satpati coast at 35–50 m depth, dol nets (length 50–65 m and cod end # 30–69 mm) in Naigaon at 38–50 m depth and trawl (length 33–72 m and cod end # 17–32 mm) in Versova, New ferry Wharf and Sassoon dock operated at 20–50 m depth. Multiday fishing was carried out with 2–3 fishing trips in a month, each trip with duration of 7–13 days. Soaking time of gill net and dol net varied from 4 to 8 hr while each trawl haul lasted for 3–4 hr. Length–weight/Disc‐width‐weight relationship showed good fit with r2 values varying from 0.818 to 0.999. In addition to information on LWR, new maximum size for three species of elasmobranchs is reported in this paper.  相似文献

Chlormadinone acetate (CAP) was given orally for 42 days to four groups of 15 regularly cycling Hereford heifers in Central Mexico (19°N, altitude 2200 m). The CAP doses (0.625; 1.25 and 2.5 mg/day per animal) represent 1/16th, 1/8th and 1/4th, respectively, of the cycle synchronizing dose of 10 mg. Heat was observed for 1 hour at 06.00, 12.00 and 18.00 h and animals in heat were rectally palpated at 6 h intervals until ovulation had occurred. Observations continued for the duration of the treatment period and 26 days thereafter. CAP treatment had significant effects. With 2.5 mg, occurrence of the next heat was delayed in 8 and inhibited in 6 animals; nine animals had synchronized heat after CAP withdrawal; five animals did not ovulate but developed luteinized follicles; animals not cycling had prolonged periods of vulvar swelling. With 1.25 mg CAP, most animals continued cycling, but estrous intervals were prolonged and 3 animals developed luteinized follicles. With 0.625 mg CAP, only a slight prolongation in estrous intervals and of the heat periods was observed.Individual differences in response to CAP were marked in all treatment groups. CAP at the higher doses interfered with mechanisms of ovulation and was partially effective in preventing heat and follicular development.In cycling animals the length of the cycle, the duration of heat and the daily variations of the onset of heat were within reported limits. The interval between disappearance of heat symptoms and ovulations (21.23 h) was about twice that reported in the literature. Ovulations followed a daily rhythm with a major peak between 24.00 and 0.600 h and a minor elevation between 12.00 and 18.00 h.  相似文献

Seasonal adaptations of populations of the southwestern corn borer, Diatraea grandiosella, obtained from south-central Mexico (19°N latitude) and southeast Missouri (37°N latitude) were compared. Day length and temperature were found to serve as environmental cues to programme the larval diapause of both populations, but different critical values were observed. The critical day length for diapause induction was about 13 hr light/day for Mexican larvae and about 15 hr light/day for Missouri larvae, and was relatively stable at 20 to 30°C. Mexican larvae displayed a less-intense diapause than did Missouri larvae. Some diapausing Mexican larvae maintained at 25 or 30°C pupated in about 15 days, regardless of the day length to which they were exposed. The rate of diapause development of Mexican larvae was high at day lengths between 14 hr and 16 hr, whereas that of Missouri larvae was accelerated at day lengths of 16 hr at 25 and 30°C. Diapause development of Mexican larvae was virtually unaffected by chilling at 10°C, whereas that of Missouri larvae continued at a low rate at 10°C. Selection of Mexican larvae for diapause showed that only four generations were needed to significantly increase the incidence of diapause.  相似文献

Abstract Due to the risk of extinction and ornamental value of the swallowtail butterfly, Sericinus montelus Gray (Lepidoptera: Papilionidae) in China, knowledge about local adaptations is important for the conservation and economical utilization of the species. In the present study, photoperiodic diapause induction and diapause intensity of S. montelus populations from Jiamusi (46°37′N), Beijing (40°15′N), Zibo (36°48′N), Fangxian (32°36′N), Wuhan (30°33′N) and Huaihua (27°33′N) were characterized at 25°C. Logistic regression analysis revealed a significant population × hours of light interaction, confirming that photoperiodic responses varied among populations. The critical photoperiod was positively correlated with latitude and increased toward the north at a rate of about 1 h for each 6.67 degrees of latitude. Survival analyses indicated that survival time of diapausing pupae before adult eclosion differed significantly among populations at 25°C and 16 : 8 L : D h. The mean duration of pupal diapause was also positively correlated with latitude. Our study reveals geographic variation in the critical photoperiod for diapause induction and in diapause intensity of S. montelus. These results provide useful information for our general understanding about seasonal adaptation in insects and may also be used to predict how geographic populations respond to climate warming.  相似文献

L-phenylalanine mustard (L-PAM) was incubated at 37° C in bile of bovine, canine and human origin. Recovery rate constants of L-PAM from bile were 0.1/hr for canine bile (0–3 hours); 0.18/hr for bovine bile; 0.45/hr for human bile. No significant hydrolysis of L-PAM in canine bile was noted for the period of 3 to 6 hours at 37° C. The incubation of L-PAM in sodium taurocholate solution (1000 molar excess) gave a recovery rate constant 0.15/hr at 37° C. However, the incubation of L-PAM in bilirubin solution (2.5 mg/ml H2O) gave a recovery rate constant of 0.52/hr at 37° C. The high concentration of the parent compound L-PAM seen in vivo in canine bile after i.v. administration may be related to its low in vitro degradation rate in canine bile.  相似文献

To develop an oral formulation for PG301029, a novel potent agent for the treatment of Hepatitis C virus infection, that not only has very low aqueous solubility but also degrades rapidly in water. The solubility of PG301029 was determined in water, various aqueous media, and several neat organic solvents. The stability of PG301029 was monitored at room temperature in buffess for 4 days, and in several neat organic solvents for up to 8 mo. Drug concentrations were measured by high-performance liquid chromatography (HPLC). Based on solubility and stability data, Gelucire 44/14 and DMA (N,N-dimethylacetamide) at a weight ratio of 2 to 1 were chosen as the formulation vehicle. After the vehicle was prepared, it was maintained in liquid form at ∼40°C until the PG301029 was dissolved. The final formulation product was a semisolid at room temperature. The bioavailability of the formulation was tested on 4 female BALB/c mice. PG301029 is insoluble in all tested aqueous media, while its solubility is promising in DMA. This compound is unstable in aqueous media and some organic solvents; however, it is stable in DMA. This proposed formulation is able to hold up to 10 mg/mL of drug and is stable at 4°C. The shelf life for this formulation stored at 4°C is extrapolated to be greater than 4 years. This formulation dramatically increases the bioavailability of PG301029. This nonaqueous formulation solves the stability, solubility, and bioavailability problems for PG301029. This semisolid formulation can easily be incorporated into soft elastic capsules.  相似文献

Cycloheximide(Cyclo), an inhibitor of protein synthesis by a direct action on protein synthesis at the ribosomal level, was used to reversibly inhibit estrogen-induced sexual receptivity. Cyclo (100 μg per rat) was infused into the preoptic area(POA) of ovariectomized rats at varying times before, simultaneously with, and after 3 μg of subcutaneous estradiol benzoate (EB). All animals received 0.5 mg progesterone (P) 36 hr after EB, and were tested for sexual receptivity 4–6 hr after P. The females were placed with stud males and a lordosis quotient was computed for each female (lordosis quotient = number of lordosis responses/20 mounts by the male × 100). Females receiving Cyclo 6 hr before, simultaneously with, or 12 hr after EB showed significantly lower levels of sexual receptivity when compared to females receiving Cyclo 36 hr before and 18 and 24 hr after EB. When those animals that showed low levels of sexual behavior after Cyclo infusion were reprimed with EB and P 7 days later and presented with a male they showed high levels of sexual receptivity. Thus, the effect of Cyclo was reversible. Only Cyclo infusions into the POA (bilateral) and third ventricle were effective in suppressing sexual behavior. Caudate nucleus, lateral ventricle, and unilateral POA infusions were without effect.The data presented are in agreement with earlier work that utilized actinomycin D to inhibit steroid-induced sexual behavior. Cyclo was found to be less toxic than actinomycin D. All of the available evidence is consistent with the hypothesis that estrogen stimulates RNA and/or protein synthesis in its facilitation of sexual behavior in the female rat.  相似文献

The effects of estrogen and progesterone on the social and sexual behavior of brown lemmings, Lemmus trimucronatus, were investigated. The behavior of hormone-treated and untreated ovariectomized females and sexually vigorous males was observed in six consecutive daily 5-min dyadic encounters. Sexual receptivity, as measured by lordosis, and other social behaviors including nasonasal contact, boxing postures, allogrooming, perineal investigation, and male mounting increased following 48 hr of exposure to daily injections of 0.5 μg estradiol benzoate (EB). Lordosis in EB-primed females was not facilitated or inhibited by short-term (4 hr) exposure to 0.5 mg progesterone (P). Long-term (greater than 24 hr) exposure to P apparently inhibited lordosis and other social behaviors in EB-treated females, although males continued to attempt to mount these females. In EB-treated females a dramatic increase in threat-leaps, directed by the female toward the male, was observed within 4 hr of P injection. Threat-leaps declined when P was withdrawn. Threat-leaps were also observed in ovariectomized females after prolonged exposure to P only (0.5 mg/day). Vaginal perforation and cornification were first apparent 48 hr after EB injection. P-alone treated ovariectomized females also showed vaginal perforation but cornified cells were infrequent and these animals did not show lordosis.  相似文献

This is the first documentation on the feeding habits of the black drum, Pogonias cromis, in waters of South America. The aim of the investigation was to determine the trophic ecology of P. cromis in the Mar Chiquita coastal lagoon (37°32′–37°45′S; 57°19′–57°26′W) from stomach content analyses of 109 P. cromis specimens. Samples in various sizes were collected monthly using various types of fishing gear. The results showed that this species is a benthic predator focusing on bivalves and crabs. permanova results indicated significant seasonal differences in the diet. This pattern reflected seasonal changes in prey abundance and availability, which produced changes in both foraging strategy and trophic niche breadth. This indicates that P. cromis is a versatile predator, adapting its feeding strategy in response to a seasonal shift of prey abundance, and indicating an opportunist behavior.  相似文献

The respiratory metabolism in larvae of the Antarctic fly, Belgica antarctica Jacobs (Diptera: Chironomidae) was investigated at Palmer Station, Anvers Island (64°46′S, 64°03′W). Oxygen consumption was linearly related to temperature from 0 to 20°C, respectively, 49 and 338 nl/mg live wt/hr. Maintenance at 0 and 10°C for 8 days had no differential effect on the metabolic rate, suggesting that larvae lack the ability for compensatory acclimation. A comparison of standard metabolism for polar and temperate chironomids revealed no elevation of metabolic rate in polar forms. However, polar species exhibited lower activation energies than temperate forms indicating that the respiratory metabolism of polar chironomids is relatively temperature independent.  相似文献

Immediate membrane sealing after spinal cord injury (SCI) can prevent further degradation and result in ultimate functional recovery. It has been reported that polyethylene glycol (PEG) can repair membrane damage caused by mechanical insults to the spinal cord. Furthermore, membrane fluidity and its sealing process vary at different temperatures. Here, we have assessed the possible synergistic effects of PEG and temperature on the repair of neural membranes in an SCI model. The effects of PEGs (400, 1,000 and 2,000 Da) were studied at different temperatures (25, 37 and 40 °C) by means of compound action potential (CAP) recovery and a lactate dehydrogenase (LDH) assay. Isolated spinal cords were mounted in a double sucrose gap chamber, where the amplitude and area of CAPs were recorded after implementing injury, in the presence and absence of PEG. Moreover, the LDH assay was used to assess the effects of PEG on membrane resealing. Data showed that the least CAP recovery occurred at 25 °C, followed by 37 and 40 °C, in all treated groups. Moreover, maximum CAP amplitude recovery, 65.46 ± 5.04 %, was monitored in the presence of PEG400 at 40 °C, followed by 41.49 ± 2.41 % in PEG1000 and 37.36 ± 1.62 % in PEG2000. Furthermore, raising the temperature from 37 to 40 °C significantly increased CAP recovery in the PEG2000 group. Similar recovery patterns were obtained by CAP area measurements and LDH assay. The results suggest that application of low-molecular weight PEG (PEG400) in mild hyperthermia conditions (40 °C) provides the optimum condition for membrane sealing in SCI model.  相似文献

Aim This paper examines the migration of the tropical nectarivorous bat Leptonycteris curasoae considered as a latitudinal migrant that breeds in south-west United States and northern Mexico in spring and migrates southward during fall. We tested the hypothesis that the latitudinal migration occurs only locally given by the local availability of bat resources, leading to migratory movements in zones with seasonal scarcity of resources and to resident bat populations where resources are available throughout the year. Localization We analysed the presence of L. curasoae along its distribution range in North America (between 14°N and 33°N). Study cases were also conducted in three Mexican localities: the Tehuacán Valley (17°48′–18°58′N and 96°48′–97°43′W), Sonoran Desert (28°41′N and 110°15′W), and the coast of Jalisco (19°32′N and 105°07′W). Methods Geographic evidence for latitudinal migration of L. curasoae was analysed using 94-year capture records housed in twenty-two collections of North America. Records were analysed using a Geographical Information System (GIS), in which floral resources and capture records were integrated. Monthly captures in the Tehuacán Valley were conducted during three years and bats abundance and reproductive status were correlated with the phenology of bat resources. Bat captures were also conducted during two consecutive years in an extratropical desert during winter and spring, and during one spring in the coast of Jalisco. Results The latitudinal migration of L. curasoae in North America only occurs at latitudes near 30°N, whereas bats may be residents at latitudes lower than 21°N. Captures were associated always to the availability of floral resources in both geographical and local scales. Main conclusions The existence of resident populations in the tropics with two reproductive events support the hypothesis that migration only occurs in the northern distribution limit of this nectar-feeding bat.  相似文献

Questions

A robust ecosystem requires a functionally heterogeneous community of organisms with ecological traits that permit broad resource partitioning. Understanding community diversity patterns can help investigate drivers of community assembly and assess restoration success. Do biodiversity patterns differ among grassland communities sown with different ecotypes of dominant species during restoration along a rainfall gradient in the tallgrass prairie of the central US Great Plains?

Location

Four field sites across a rainfall gradient within the North American Great Plains: Colby, Kansas (39°23′17.8″N, 101°04′57.4″W), Hays, Kansas (38°51′13.2″N, 99°19′08.6″W), Manhattan, Kansas (39°08′22.3″N, 96°38′23.3″W), and Carbondale, Illinois (IL, 37°41′47.0″N, 89°14′19.2″W).

Methods

We applied linear mixed models to assess the effect of dominant species ecotype, year, and location on grassland taxonomic, phylogenetic, and functional diversity.

Results

The non-local grass ecotype (compared to the local ecotype) promoted species richness. In contrast, the effect of the dominant species ecotype on phylogenetic or functional diversity was site-specific over the 10-year restoration. Richness decreased across the rainfall gradient from dry to moist sites, and the wettest site had the highest phylogenetic and functional diversity.

Conclusions

Our results suggest that abiotic filtering by rainfall is a key assembly mechanism that could predict grassland changes in biodiversity in the early restoration phases. Given the community response across the tallgrass prairie, restoration practitioners should consider the impact of regional sources of dominant species used in restoration when biodiversity is a restoration goal. It is recommended for future grassland restoration to detect gaps and limitations in evolutionary and trait structure that will reveal which diversity components to evaluate.  相似文献

Stages of Metaseiulus occidentalis sensitive to photoperiod induction of diapause were determined by transferring various stadia into diapause-inducing conditions, and rearing them until adult females could be scored for reproductive condition. When eggs were transferred to 10 hr light at 19°C from 24 hr light at 25°C and the mites reared to adults, 92 per cent entered diapause. When larvae and all subsequent stages were kept under the inductive conditions, 62 per cent of adult females diapaused. Mites transferred as protonymphs into inductive conditions yielded only 10 per cent in diapause, and mites transferred as deutonymphs or newly emerged females did not enter diapause.However, adult females reared from eggs at 19°C under 12 hr light (which is near the critical photophase of 11·2 hr at 19°C) showed an unexpected sensitivity to photoperiod. Some newly emerged females oviposited upon transfer to an 8 hr photophase at 19°C. Some then stopped ovipositing and apparently entered diapause; these females resumed ovipositing after intervals ranging from 34 to 100 days. This was termed ‘switching’ into diapause. Some females reared under a 16 hr photophase at 19°C ‘switched’ also upon transfer as adults to shorter photophases—either 8 or 12 hr at 19°C. Thus, ‘switching’ may be due to transfer to shorter photophases. Promptness of mating vs delayed mating allowed ‘switching’ to be more easily detected.  相似文献
