Five newly identified species of Fomitiporia (F. alpina, F. gaoligongensis, F. hainaniana, F. subrobusta and F. subtropica) and their morphological and molecular characterisation are described in this paper. Fomitiporia alpina sp. nov. is distinguished by its pileate basidiomata, parallel tramal hyphae and large basidiospores (6.5–8 × 6–8 μm), and by its gymnosperm wood-living habitat. Fomitiporia gaoligongensis sp. nov. is distinct from other species due to its semicircular pileus and subglobose to globose basidiospores (6.5–7.6 × 6–7.4 μm). Fomitiporia hainaniana sp. nov. is marked by its resupinate basidiomata, the presence of setae and small globose basidiospores (4–5 × 3.8–4.4 μm). Fomitiporia subrobusta sp. nov. is characterised by its triquetrous basidiomata, small pores (6–9 per mm) with entire and thick dissepiments, and subglobose to obovoid basidiospores (6.2–6.8 × 5.2–6 μm). Fomitiporia subtropica sp. nov. can be differentiated by its resupinate basidiomata, smaller pores (6–10 per mm) and smaller basidiospores (5.2–6 × 4.4–5 μm). Phylogenetic analysis, based on multi-gene comparison of the internal transcribed spacer regions (ITS), nuclear large subunit ribosomal RNA gene regions (nLSU), the translation elongation factor 1-α gene (tef1α) and the second subunit of RNA polymerase II (rpb2), confirmed affinity with the Fomitiporia species and showed association with similar fungi in the genus.  相似文献

Two new species of Coltricia, C. austrosinensis and C. minima, are described from southern China on the basis of morphological characters and molecular evidence. Phylogenetic analysis based on the internal transcribed spacer (ITS) regions and nuclear large subunit (nLSU) ribosomal RNA gene regions indicated that the two new species were nested within the Coltricia clade in Hymenochaetales. Coltricia austrosinensis is characterized by centrally stipitate basidiocarps, lobed pileal margin, distinctly swollen stipe tip, cinnamon pore surface, large pores (1–3 per mm), and broadly ellipsoid basidiospores measuring 8–10?×?5.5–6.5 μm, with a distribution to date in subtropical China. Coltricia minima is characterized by tiny, centrally stipitate basidiocarps, entire pileal margin, uniform stipe, pileus bearing distinct concentric zones and pore surface dark greyish blue when fresh, small pores (3–4 per mm), narrow tramal hyphae (3.5–4 μm), and broadly ellipsoid to subglobose basidiospores measuring 6–7?×?4–5 μm, and occur in mixed tropical forests.  相似文献

Phylloporia is a monophyletic genus within the Hymenochaetaceae as recovered by nuclear large subunit ribosomal DNA (nLSU) sequences. According to the summarization of 38 species accepted in this genus, Phylloporia is characterized by an absence of setae and the presence of abundant thick-walled, colored and tiny basidiospores, although its other morphological characters are highly diverse. Nine herbarium specimens from China, fitting the morphological concept of Phylloporia, were morphologically and phylogenetically studied in detail. The phylogeny inferred from nLSU sequences shows that the nine specimens formed three terminal lineages within the Phylloporia clade. Two lineages being composed of four specimens from Hainan and three from Guizhou were newly described as Phylloporia minutipora and P. radiata, respectively. In Phylloporia, P. minutipora is distinct by a combination of annual, sessile and imbricate basidiocarps, distinctly concentrically sulcate pileal surface with obtuse margin, angular pores of 12–15 per mm, duplex context separated by a black zone, a dimitic hyphal system, and broadly ellipsoid basidiospores of 2.5–3?×?2–2.5 μm, while P. radiata is distinct by a combination of annual, sessile and imbricate basidiocarps, faintly sulcate and radially striate pileal surface, sharp pileal margin, angular pores of 8–10 per mm, duplex context separated by a black zone, a monomitic hyphal system, and broadly ellipsoid basidiospores of 2.5–3.5?×?2–2.5 μm. The third lineage, comprising two specimens from Hainan, was morphologically determined as Phylloporia pulla. This species was recently combined to Phylloporia based on only morphological characters, and the current study for the first time generated its molecular sequences for phylogenetic reference. Spadicoides lushanensis sp. nov., S. multiseptata sp. nov. and S. rostrata sp. nov. are described and illustrated from specimens collected on dead branches of unidentified plants in Lushan Mountain, China. Spadicoides lushanensis is characterized by obclavate, rostrate, pale brown, 40–65?×?4.5–5.5 μm, smooth, 6–8-euseptate conidia. Spadicoides multiseptata is easily distinguished by obclavate, smooth, 11–15-euseptate, 65–115?×?11.5–13 μm, brown to pale brown conidia with a subacute apex. Spadicoides rostrata differs from other described Spadicoides species by obclavate, rostrate, pale brown, 40–65?×?10.5–12.5 μm, smooth, predominantly 5-euseptate conidia. A new poroid wood-inhabiting fungal genus, Leifiporia, is proposed, based on morphological and molecular evidence, which is typified by L. rhizomorpha sp. nov. The genus is characterized by an annual growth habit, resupinate basidiocarps with white to cream pore surface, a dimitic hyphal system with generative hyphae bearing clamp connections and branching mostly at right angles, skeletal hyphae present in the subiculum only and distinctly thinner than generative hyphae, IKI–, CB–, and ellipsoid, hyaline, thin-walled, smooth, IKI–, CB– basidiospores. Sequences of ITS and LSU nrRNA gene regions of the studied samples were generated, and phylogenetic analyses were performed with maximum likelihood, maximum parsimony and Bayesian inference methods. The phylogenetic analysis based on molecular data of ITS + nLSU sequences showed that Leifiporia belonged to the core polyporoid clade and was closely related to Diplomitoporus overholtsii and Lopharia cinerascens, and then grouped with Pycnoporus and Trametes. Further investigation was obtained for more representative taxa in the Polyporaceae based on nLSU sequences, in which the results demonstrated that the genus Leifiporia formed a monophyletic lineage with a strong support (100 % BS, 100 % BP, 1.00 BPP). Both morphological and molecular evidence confirmed the placement of the new genus in the core polyporoid clade. In addition, a new combination, Leifiporia eucalypti, is proposed based on examination of its type material and phylogeny.  相似文献

Taxonomic and phylogenetic studies of the Funalia gallica species complex and its related species were carried out. The phylogenetic analyses were based on datasets of either combined ITS, nrLSU and RPB2 sequences or single ITS sequences. The results reveal that at least four strongly hispid to strigose Funalia species can be recognised, i.e. the cosmopolitan F. trogii, Eurasian F. gallica, North American “F. gallica” and a new species F. subgallica discovered from tropical China. Morphologically, F. subgallica is characterised by its strongly hispid to strigose pileal surface, large pores (1–3 per mm) and basidiospores (11–15?×?4–5 μm).  相似文献

Rhizoglomus venetianum, a new arbuscular mycorrhizal fungal species, has been isolated and propagated from a heavy metal-contaminated site in Sacca San Biagio island, downtown Venice, Italy. Interestingly, under the high levels of heavy metals occurring in the site, the new fungus was able to grow only intraradically. In greenhouse trap and single species cultures under low heavy metal levels, the fungus produced innumerous spores, clusters, and sporocarps extraradically, which were formed terminally on subtending hyphae either singly, in small spore clusters, or, preferably, in loose to compact non-organized sporocarps up to 2500?×?2000?×?2000 μm. Spores are golden-yellow to bright yellow brown, globose to subglobose to rarely oblong, 75–145?×?72–140 μm in diameter, and have four spore wall layers. Morphologically, the new fungus is similar to R. intraradices, and phylogenetically, it forms a monophyletic clade next to R. irregulare, which generally forms irregular spores and lacks, like R. intraradices, the flexible innermost wall layer beneath the structural/persistent third wall layer. A key for the species identification is presented comprising all 18 Rhizoglomus species, so far described or newly combined.  相似文献

The objective of this study comprises of developing novel co-spray dried rifampicin phospholipid lipospheres (SDRPL) to investigate its influence on rifampicin solubility and oral bioavailability. Solid-state techniques were employed to characterize the liposphere formulation. SDRPL solubility was determined in distilled water. BACTEC 460TB System was employed to evaluate SDRPL antimycobacterial activity. The oral bioavailability of the lipospheres was evaluated in Sprague Dawley rats. Lipospheres exhibited amorphous, smooth spherical morphology with a significant increase (p?<?0.001) in solubility of SDRPL (2:1), 350.9?±?23 versus 105.1?±?12 μg/ml and SDRPL (1:1) 306.4?±?20 versus 105.1?±?12 μg/ml in comparison to rifampicin (RMP). SDRPL exhibited enhanced activity against Mycobacterium tuberculosis, H37Rv strain, with over twofolds less minimum inhibitory concentration (MIC) than the free drug. Lipospheres exhibited higher peak plasma concentration (109.92?±?25 versus 54.31?±?18 μg/ml), faster T max (two versus four hours), and enhanced area under the curve (AUC0–∞) (406.92?±?18 versus 147.72?±?15 μg h/L) in comparison to pure RMP. Thus, SDRPL represents a promising carrier system exhibiting enhanced antimycobacterial activity and oral bioavailability of rifampicin.  相似文献

A new poroid wood-inhabiting fungal species, Ceriporiopsis kunmingensis sp. nov., is proposed based on morphological and molecular characters. The species is characterized by resupinate basidiocarps with pale cinnamon-buff to ochreous color when dry; generative hyphae unbranched and subparallel along the tubes; presence of hyphal ends; allantoid basidiospores, 4.5–5 × 1.5–2 μm. The internal transcribed spacer (ITS) and large subunit (LSU) regions of nuclear ribosomal RNA gene sequences of the studied samples were generated, and phylogenetic analyses were performed with maximum likelihood, maximum parsimony, and Bayesian inference methods. The phylogenetic analysis based on molecular data of ITS?+?nLSU sequences showed that C. kunmingensis belonged to the phlebioid clade, formed a monophyletic lineage with a strong support (100% BS, 100% BP, 1.00 BPP), and was closely related to Phlebia aurea, and then grouped with P. livida and P. subserialis. Two new species of the feather mite genus Proctophyllodes Robin, 1868 (Analgoidea: Proctophyllodidae) are described from two passerine birds (Passeriformes) in Europe: Proctophyllodes markovetsi n. sp. from the tawny pipit Anthus campestris (L.) (Motacillidae) and P. loxiae n. sp. from the red crossbill Loxia curvirostra (L.) (Fringillidae). Males of P. markovetsi are most clearly distinguished from the closely related P. tchagrae Atyeo & Braasch, 1966 by having greater terminal lamellae (30–40 × 20–25 µm), the tips of genital arch curved medially, and the corolla of the anal sucker with 14–15 denticles; females of this species are characterised by the terminal appendages distinctly longer than the lobar region width. Males of P. loxiae differ from the closest species, P. fuchsi Mironov, 1997, by having smaller terminal lamellae (45–50 × 22–28 μm), the genital organ extending beyond the posterior margin of lamellae by half their length; females can be distinguished by having the terminal cleft noticeably wider than long (28–30 × 35–40 μm).  相似文献

Lumostatic operation was applied for efficient astaxanthin production in autotrophic Haematococcus lacustris cultures using 0.4-L bubble column photobioreactors. The lumostatic operation in this study was performed with three different specific light uptake rates (q e) based on cell concentration, cell projection area, and fresh weight as one-, two- and three-dimensional characteristics values, respectively. The q e value from the cell concentration (q e1D) obtained was 13.5 × 10?8 μE cell?1 s?1, and the maximum astaxanthin concentration was increased to 150 % compared to that of a control with constant light intensity. The other optimum q e values by cell projection area (q e2D) and fresh weight (q e3D) were determined to be 195 μE m?2 s?1 and 10.5 μE g?1 s?1 for astaxanthin production, respectively. The maximum astaxanthin production from the lumostatic cultures using the parameters controlled by cell projection area (2D) and fresh weight (3D) also increased by 36 and 22 % over that of the controls, respectively. When comparing the optimal q e values among the three different types, the lumostatic cultures using q e based on fresh weight showed the highest astaxanthin productivity (22.8 mg L?1 day?1), which was a higher level than previously reported. The lumostatic operations reported here demonstrated that more efficient and effective astaxanthin production was obtained by H. lacustris than providing a constant light intensity, regardless of which parameter is used to calculate the specific light uptake rate.  相似文献

Russula lotus, R. nivalis, and R. purpureoverrucosa are proposed here as new taxa based on morphological and molecular evidences. The new species are described with illustration photographs and line drawings, and compared against related species. Morphologically, R. lotus (subgenus Heterophyllidia, section Heterophyllae, subsection Cyanoxanthinae) is characterized by a medium-sized basidioma with a pale pink to purplish pink pileus whose center area is yellowish white, white to cream white lamellae occasionally forked with lamellulae, a cream white smooth stipe, broadly ellipsoid to ellipsoid basidiospores ornamented with disconnected amyloid warts and inamyloid suprahilar plage, dispersed sulphovanillin (SV)-negative pileocystidia, and the pileipellis with suprapellis cells claw-assembled. Russula nivalis (subgenus Russula, section Russula, subsection Russula) is characterized by a very small pure white basidioma with a pileus 7–12 mm in diameter, abundant clavate pileocystidia and caulocystidia changing purplish red in SV, and broadly ellipsoidal to ellipsoid basidiospores ornamented with strongly amyloid warts and ridges interconnected by fine lines in an uncompleted or completed reticulum, mostly with inamyloid suprahilar plage. Russula purpureoverrucosa (subgenus Incrustatula, section Lilaceinae, subsection Lilaceinae) is characterized by a medium-sized basidioma with a red to grayish magenta pileus slightly areolate in small irregular warts from center to margin, a stipe with the same color and warts as the pileus, white to cream white occasionally forked lamellae with lamellulae, broadly ellipsoidal to ellipsoid basidiospores ornamented with amyloid warts and ridges rarely connected, abundant clavate pleurocystidia covered with thick yellowish incrustations, and the pileipellis with suprapellis cells a typical trichoderm, some of which covered with yellowish incrustations, pileocystidia absent. Phylogenetic analysis of nucleotide sequences of the internal transcribed spacer (ITS) region provided further evidence that the described species belong to the subsections above respectively, and represent new taxa.  相似文献

Horse heart carboxymethylated cytc (CM-cytc) displays myoglobin-like properties. Here, the effect of cardiolipin (CL) liposomes on the nitrite reductase activity of ferrous CM-cytc [CM-cytc-Fe(II)], in the presence of sodium dithionite, is reported between pH 5.5 and 7.6, at 20.0 °C. Cytc-Fe(II) displays a very low value of the apparent second-order rate constant for the NO2 ?-mediated conversion of cytc-Fe(II) to cytc-Fe(II)-NO [k on = (7.3 ± 0.7) × 10?2 M?1 s?1; at pH 7.4], whereas the value of k on for NO2 ? reduction by CM-cytc-Fe(II) is 1.1 ± 0.2 M?1 s?1 (at pH 7.4). CL facilitates the NO2 ?-mediated nitrosylation of CM-cytc-Fe(II) in a dose-dependent manner, the value of k on for the NO2 ?-mediated conversion of CL–CM-cytc-Fe(II) to CL–CM-cytc-Fe(II)-NO (5.6 ± 0.6 M?1 s?1; at pH 7.4) being slightly higher than that for the NO2 ?-mediated conversion of CL–cytc-Fe(II) to CL–cytc-Fe(II)-NO (2.6 ± 0.3 M?1 s?1; at pH 7.4). The apparent affinity of CL for CM-cytc-Fe(II) is essentially pH independent, the average value of B being (1.3 ± 0.3) × 10?6 M. In the absence and presence of CL liposomes, the nitrite reductase activity of CM-cytc-Fe(II) increases linearly on lowering pH and the values of the slope of the linear fittings of Log k on versus pH are ?1.05 ± 0.07 and ?1.03 ± 0.03, respectively, reflecting the involvement of one proton for the formation of the transient ferric form, NO, and OH?. These results indicate that Met80 carboxymethylation and CL binding cooperate in the stabilization of the highly reactive heme-Fe atom of CL–CM-cytc.  相似文献

The role of 4.1 or 8.2 μM meta-topolin (mT) on shoot multiplication, rooting and ex vitro acclimatization of micropropagated Corylus colurna L., a promising non-suckering rootstock for hazelnut (Corylus avellana L.), was examined in comparison to N6-benzyladenine (BA), the most used cytokinin in tissue culture of Corylus spp. The influence of 8.2 μM mT and BA on photosynthetic pigments content and antioxidant enzymes activity, catalase (CAT) and guaiacol peroxidase (POD), in regenerated shoots, and on the preparation of the rootstock for micrografting was also evaluated. The highest shoot multiplication was recorded on medium containing 8.2 μM mT and an overall positive effect of mT on growth and quality of micropropagated shoots was found. The highest chlorophyll a content (1.236 mg g?1 fresh weight, FW) and chlorophyll a/b ratio (2.48), and the lowest total carotenoids content (0.292 mg g?1 FW) and CAT activity (25.8 μmol min?1 mg?1 protein) were detected after 8.2 μM mT application, while no significant differences were found in chlorophyll b content and POD activity between the two cytokinins. The best rhizogenesis response (98% for 4.1 μM and 100% for 8.2 μM mT) and ex vitro acclimatization competence (higher than 78%) were exhibited from shoots multiplied on mT. Furthermore, the multiplication of rootstock on mT allowed obtaining the highest (70%) response of successful micrografting. The present findings provide the first evidence of the successful applicability of mT in C. colurna tissue culture and development of micrografted plantlets.  相似文献

Single cells of five different Microcystis species (M. ichthyoblabe, M. viridis, M. flos-aquae, M. wesenbergii, and M. aeruginosa) were batch-cultured at different temperatures and light intensities: (a) 25 °C and 50 μmol photons m?2 s?1 (control culture); (b) 25 °C and 10 μmol photons m?2 s?1; and (c) 15 °C and 50 μmol photons m?2 s?1. The extracellular polysaccharide content was significantly higher in treatments b and c than in the control treatment. All Microcystis species existed as single cells under the control treatment but formed colonies in treatments b and c. All of the colonies were irregular with indistinct margins. M. ichthyoblabe, M. viridis, M. flos-aquae, and M. wesenbergii formed colonies with similar morphologies and their cells were loosely aggregated. In contrast, M. aeruginosa formed denser colonies with no distinct holes. The colony morphologies differed from the classic morphology of M. ichthyoblabe field-grown colonies but resembled that of small colonies found in Lake Taihu (Yangtze Delta Plain, China) during early spring. This indicates that field- and laboratory-grown colonies are governed by similar formation processes. We suggest that in laboratory and field environments, M. ichthyoblabe (or M. flos-aquae) colonies are representative of small colonies formed from single Microcystis cells, whereas the morphology of older colonies evolves to resemble M. wesenbergii and M. aeruginosa colonies.  相似文献

The cotton bollworm, Helicoverpa armigera (Hubner) (Lepidoptera: Noctuidae) is a polyphagous pest that exist all over the world. It is also very destructive in Turkey on various agricultural products. In this study, we have detected an alphabaculovirus from Helicoverpa armigera larvae collected from cotton field. The virus isolate was named as Helicoverpa armigera nucleopolyhedrovirus (HearNPV-O1) based on morphological and molecular properties. This is the first record of baculovirus from H. armigera in Eurasia region. Scanning electron microscopy examinations of polyhedral inclusion bodies (PIBs) showed their irregular morphology with average diameter of 0.85 to 1.25 μm. Transmission electron microscopy studies revealed that the nucleocapsids were multiple enveloped and bacilliform shaped. The size of nucleocapsid was found as 279 nm with a width of 56 nm. Digestion of viral genome by PstI generated 8 fragments with total size of 130.9 kbp. According to the phylogenetic analysis of the partial polyhedrin (polh), late expression factor 8 (lef8) and late expression factor 9 (lef9) genes, HearNPV-O1 isolate is very close to H. armigera MNPV Indian isolate-443. Five different concentrations of HearNPV-O1 between 103 and 107 were used in dose-response test on neonate, 3rd and 5th instars larvae of H. armigera. The highest dose (107) showed 92%, 88% and 57% mortality, respectively within 14 days. LC50 values of HearNPV-O1 isolate were calculated as 6?×?104, 7?×?104 and 8?×?106 PIBs/mL?1 against neonate, 3rd and 5th instars larval stages, respectively. These results demonstrate that HearNPV-O1 isolate may be a potential biocontrol agent to be utilized against H. armigera.  相似文献

Objectives

To investigate the outcomes of capsule lost on cell transformation efficiency and chemicals (1,3-propanediol, 2,3-butanediol, and 2-ketogluconic acid) production by Klebsiella pneumoniae.

Results

The cps gene cluster showed low sequence homology with pathogenic strains. The wza is a highly conserved gene in the cps cluster that encodes an outer membrane protein. A non-capsulated mutant was constructed by deletion of wza. Phenotype studies demonstrated that non-capsulated cells were less buoyant and easy to sediment. The transformation efficiency of the non-capsulated mutant reached 6.4 × 105 CFU μg?1 DNA, which is 10 times higher than that of the wild strain. 52.2 g 1,3-propanediol L?1, 30.7 g 2,3-butanediol L?1, and 175.9 g 2-ketogluconic acid L?1 were produced by non-capsulated mutants, which were 10–40% lower compared to wild strain. Furthermore, viscosities of the three fermentation broths decreased to approximately 1.3 cP from the range of 1.8–2.2 cP.

Conclusions

Non-capsulated K. pneumoniae mutants should allay concerns regarding biological safety, improve transformation efficiency, lower viscosity, and subsequently ameliorate the financial burden of the downstream process of chemicals production.

Hypoaspis larvicolus (Acari: Laelapidae) (first report from Turkey) occurred together with Sancassania polyphyllae (Acari: Acaridae) on the larvae of the scarab beetle, Polyphylla fullo (Coleoptera: Scarabaeidae), that were feeding on the roots of strawberry in Aydin, Turkey. Laboratory studies were conducted to (1) observe whether H. larvicolus feeds and completes its life cycle on the various stages of S. polyphyllae or other astigmatid mites, such as Acarus siro, Carpoglyphus lactis and Tyrophagus putrescentiae (Acaridae), and to determine its population growth when feeding on these prey, and (2) to determine development periods, longevity and fecundity of H. larvicolus feeding on C. lactis. Hypoaspis larvicolus females did not feed on S. polyphyllae, but fed, developed and reproduced when A. siro, C. lactis or T. putrescentiae were provided as prey. Hypoaspis larvicolus is larviparous as well as oviparous. The female lays eggs or gives birth to larvae. If a female gives birth to a larva, it is attached under the female’s venter for 1–2 days, a phenomenon recorded for the first time in Hypoaspis; in fact, for the first time in mites. The results of the population growth experiments revealed that H. larvicolus feeding on C. lactis produced the highest number of eggs, juveniles and adults. The developmental periods of H. larvicolus feeding on C. lactis at life-cycle path I (larva to adult) and II (egg to adult) were 12.2?±?0.3 and 15.6?±?0.6 days (females) and 19.5?±?0.2 and 20.9?±?0.4 days (males), respectively. Longevity of females versus males of H. larvicolus was 120.6?±?7.2 versus 91.6?±?13.1 days (life cycle I) and 110.0?±?27.7 versus 118.3?±?10.9 days (life cycle II), respectively.  相似文献
