In the Kol basin, one of the typical salmon rivers of Western Kamchatka, special traits of reproduction ecology of masu salmon Oncorhynchus masou are studied (characteristics and behavior of spawners during spawning, localization of spawning grounds, topography and hydrology of redds, and interaction with other species of salmonids). The masu salmon spawns in the Kol basin in tributaries of the upper reaches of the river and only in downwelling. Spawning grounds of masu samlmon are confined to stretches with overhanging banks, log jams creating shelters for spawners and favorable hydrological conditions for spawning and egg development. All over its range, masu salmon requires similar conditions for reproduction. In the north of its range, in Kamchatka, masu salmon retains the properties of the most warm-water species in the genus Oncorhynchus and selects for spawning the grounds characterized by the highest temperature in the period of spawning and development.  相似文献

There has been very little effort to understand genetic divergence between wild and hatchery populations of masu salmon (Oncorhynchus masou). In this study, we used mitochondrial (mt) NADH dehydrogenase subunit 5 gene (ND5) and six polymorphic nuclear microsatellite DNA loci to compare the genetic variability in three hatchery broodstocks of masu salmon with the variability in eight putative wild masu populations sampled in five rivers including one known source river for the hatchery broodstocks. Both ND5 and microsatellites showed no significant genetic divergence (based on FST estimates) between four annual collections from the source river population, suggesting no change in genetic diversity over this time period. The FST estimates, an analysis of molecular variance (AMOVA), and a neighbor-joining tree using both DNA markers suggested significant differentiation between the three hatchery and all eight putative wild populations. We conclude that genetic diversity of hatchery populations are low relative to putative wild populations of masu salmon, and we discuss the implications for conservation and fisheries management in Hokkaido.  相似文献

A superficially located periventricular proliferative area with PCNA-immunopositive (PCNA+) cells, which corresponds to the pallial periventricular zone (PVZ) of other fish species, including its dorsal, lateral, and medial compartments, is discovered in the telencephalon of the juvenile masu salmon Oncorhynchus masou. The PCNA+ cells are also identified in the parenchyma of the masu salmon intact brain, and their maximum concentration is observed in the medial zone. After a mechanical injury, the zones of induced neurogenesis—neurogenic niches and sites of secondary neurogenesis surrounded by radial glial fibers—appear in the masu salmon telencephalon. The PVZ of the juvenile masu salmon pallium contains clusters of undifferentiated HuCD-immunopositive (HuCD+) neurons. A change in the HuCD+ cell topography is observed in the mechanically injured masu salmon telencephalon, namely, neurogenic niches in the lateral zone and an increase in the cell distribution density and cell migration patterns in the medial zone. A high level of persistent neurogenesis is characteristic of the juvenile masu salmon brain.  相似文献

The presented data on the distribution of the masu salmon Oncorhynchus masou in the Amur River Basin are collected in 1976–2015. In the analyzed region the species’ spawning range includes the lower part of the Amur River basin from the rivers flowing into the Amur estuary to the basins of the Anyui and Amgun’ Rivers in their middle courses. The information on the catch of a masu salmon specimen in the Khor River Basin (approximately 1300 km from the Amur River mouth) in 2015 is presented. It is assumed that the total Amur population does not exceed several thousand of adult anadromous fish.  相似文献

The amago salmon, Oncorhynchus masou ishikawae, is an endemic subspecies of O. masou in Japan. Owing to the extensive stocking of hatchery fish throughout Japan, indigenous populations of O. m. ishikawae are now on the verge of extinction. We examined the genetic effects of stocking hatchery fish on wild populations in the River Koza, Japan, using microsatellite and mitochondrial DNA (mtDNA) markers. For mtDNA, haplotype mt1, which is common in wild populations, was present exclusively in isolated wild populations assumed to be unaffected by previous stocking, while it was never observed in hatchery fish. Genetic diversity was much higher in wild populations in the stocked area, which shared many mtDNA haplotypes with hatchery fish, than in isolated wild populations with haplotype mt1. Pairwise F ST estimates based on microsatellites showed significant differentiation among the isolated populations with many microsatellite loci monomorphic. Significant deviation from Hardy–Weinberg equilibrium was observed in wild populations in the area subject to stocking, where a Bayesian-based assignment test showed a high level of introgression with hatchery fish. These results suggest that wild populations with haplotype mt1, which became isolated through anthropogenic environmental change in the 1950–1960s, represent indigenous populations of O. m. ishikawae in the River Koza. They have low genetic diversity, most likely caused by genetic bottlenecks following damming and environmental deterioration, while stocking of hatchery fish over the past 30 years apparently had a large impact on the genetic structure of wild populations in the main channel of the River Koza.  相似文献

Distribution of nitroxidergic and H2S-producing neurons in the brain of the masu salmon Oncorhynchus masou was studied by methods of histochemical labeling of NADPH-diaphorase and by immunohistochemical labeling of the neuronal nitric oxide synthase and cystathionine β-synthase (CBS). The established distribution of CBS and nNOS/NADPH-d of neurons and fibers in the masu salmon telencephalon, optic tectum, and cerebellum allows suggesting that the NO- and H2S-producing systems represent individual, non-overlapping neuronal complexes performing specialized functions in the activity of local neuronal networks. In the medullar part, the nNOS-ir and NADPH-d-positive neurons were detected in the composition of viscerosensory (V, VII, and IX–X) and visceromotor (III, IV, and VI) nuclei of craniocerebral nerves, octavolateral afferent complex, reticulospinal neurons, and medial reticular formation. CBS in the masu salmon medulla was revealed in neurons of the nerve X nucleus, reticulospinal neurons, and ventrolateral reticular formation. Distribution of NO-ergic and H2S-producing neurons in the masu salmon medullar nuclei indicates that NO in masu salmon is the predominant neuromodulator of the medullar viscerosensory systems, while H2S seems to modulate only the descending motor systems. The results of the performed study allow suggesting that NO in the masu salmon medulla periventricular area can act as a regulator of postnatal ontogenesis.  相似文献

The genetic variability distribution of two mtDNA segments of chum salmon (Oncorhynchus keta) (Walbaum, 1792) and Sakhalin taimen (Parahucho perryi) (Brevoort, 1856) was examined in populations of the Sea of Japan and the Sea of Okhotsk. The values of haplotype and nucleotide variability in these species are, in general, of the same level. The dating of the divergence time of species haplotypes revealed four evolutionary periods in Sakhalin taimen and three in chum salmon. In the taimen, the first divergence time occurred approximately 430 thousand years (kyr) ago, the second 220 kyr ago, and the third 70 kyr ago. In the chum salmon, the first divergence time corresponds to 220 kyr; the second is approximately 100 kyr ago. In both species, the main portion of presently revealed haplotypes evolved over the past 50–10 kyr. Certain glacioeustatic sea level fluctuations influenced each stage of evolution history of species, contributing to their geographic isolation. Demographic population history research found that the initial stage of population growth in the taimen occurred at the time period of approximately 12 kyr ago and was apparently associated with the end of the Last Glacial Maximum. In the chum salmon, this period began somewhat earlier, 30–35 kyr ago; it has accelerated in the past 10–15 kyr. The last glaciation to a lesser extent impacted the demographics of chum salmon, probably due to the greater eurythermity and to the larger range of this species.  相似文献

Results from the 1993–2004 genetic monitoring of pink salmon populations reproducing in the rivers of Tauy Bay on the Sea of Okhotsk are analyzed. A statistically significant heterogeneity of samples as determined by gene frequencies is found only in the pink salmon generations of even years. The genetic differentiation of samples from even years (GST = 1.39 ± 0.41) is higher than that of odd years (GST = 0.740.09). The pattern for the indicator of genetic variability (heterozygosity) is exactly the opposite (0.076 ± 0.02564 vs. 0.8760 ± 0.01950). Consequently, the lower-heterozygosity samples of lines from even years are on average more genetically distinct than the analogous indicator for odd years. In addition, the interpopulation ratio in the general value of genetic diversity is almost always smaller than both the intraannual and interannual ratios, leading to a low level of interpopulation genetic differences. Cluster analysis reveals that most 2001–2004 samples are grouped separately from samples collected prior to 2000. In our opinion, the reason for this could be the turnover of a numerically dominant generation of northern Sea of Okhtosk pink salmon and the change in gene frequencies accompanying it.  相似文献

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes.  相似文献

According to materials from original investigations in Kamchatka as well as published data, the population structure and geographical variation of the masu salmon Oncorhynchus masou are considered in the species area. The intraspecies structure, dates of their run to rivers, dates of spawning, traits of spawning, size-age and sex composition, fecundity, biological traits of parr, downstream migration of smolts are investigated. The relationship is shown between the manifestations of geographical variation of masu salmon and the factor of temperature. It is assumed that the global warming of climate would contribute to the advancement of this species to northern areas and to the increase in its abundance.  相似文献

Studying Pneumocystis has proven to be a challenge from the perspective of propagating a significant amount of the pathogen in a facile manner. The study of several fungal pathogens has been aided by the use of invertebrate model hosts. Our efforts to infect the invertebrate larvae Galleria mellonella with Pneumocystis proved futile since P. murina neither caused disease nor was able to proliferate within G. mellonella. It did, however, show that the pathogen could be rapidly cleared from the host.  相似文献

Characteristics of early and late migrants of pink salmon Oncorhynchus gorbuscha that spawned in the rivers of the coast of Aniva Bay (southern Sakhalin) and Iturup Island (southern Kuril Islands) are described. Upstream migration of the late form is characterized by the presence of large exemplars, especially males, and increasing proportion of these fishes in the catches. Absolute fecundity of the females of the early and late forms can be similar (Iturup Island) or different (Aniva Bay), but relative fecundity is lower in the females of the late form in both regions. Production of a larger number of eggs by the females of the early form is associated with a larger level of mortality: the abundance of the early form is comparatively low, and substantial annual variation of the abundance is registered. Relative abundance of pink salmon from the temporal groups depends mainly on reproductive conditions during the freshwater part of their life cycle. Analysis of size composition of the fish conducted over many years shows a trend in the ratio between the body lengths of males and females: unfavorable foraging conditions (and slower growth rate) lead to the appearance of females that are larger than males. To take into account a positive correlation between the body length and fecundity of females, this feature is regarded as a compensatory reaction of the population directed to more intensive reproduction at poor foraging conditions.  相似文献

A genetic transformation system has been developed for callus cells of Crataegus aronia using Agrobacterium tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with 5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Putative transgenic calli were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this is the first time to report an Agrobacterium-mediated transformation system in Crataegus aronia.  相似文献

Heparan sulfate proteoglycans play a vital role in signaling of various growth factors in both Drosophila and vertebrates. In Drosophila, mutations in the tout velu (ttv) gene, a homolog of the mammalian EXT1 tumor suppressor gene, leads to abrogation of glycosaminoglycan (GAG) biosynthesis. This impairs distribution and signaling activities of various morphogens such as Hedgehog (Hh), Wingless (Wg), and Decapentaplegic (Dpp). Mutations in members of the exostosin (EXT) gene family lead to hereditary multiple exostosis in humans leading to bone outgrowths and tumors. In this study, we provide genetic and biochemical evidence that the human EXT1 (hEXT1) gene is conserved through species and can functionally complement the ttv mutation in Drosophila. The hEXT1 gene was able to rescue a ttv null mutant to adulthood and restore GAG biosynthesis.  相似文献

New combinations are proposed in anticipation of the Polygonaceae treatment in the forthcoming volume of Intermountain Flora: Polygonum kelloggii var. esotericum, P. kelloggii var. watsonii , Rumex densiflorus var. pycnanthus , R. salicifolius var. utahensis, and R. occidentalis var. tomentellus. Typifications are proposed to facilitate ongoing studies in Polygonaceae and to maintain current usage.  相似文献

We surveyed microsatellite variation from 22 spawning populations of coho salmon (Oncorhynchus kisutch) from the Oregon Coast to help identify populations for conservation planning. All of our samples were temporally replicated, with most samples obtained in 2000 and 2001. We had three goals: (1) to confirm the status of populations identified on the basis of spawning location and life history; (2) to estimate effective population sizes and migration rates in order to determine demographic independence at different spatial scales; and (3) to determine if releases of Washington hatchery coho salmon in the 1980's into Oregon Coast streams resulted in measurable introgression into nearby wild Oregon Coast coho populations. For the last question, our study included a hatchery broodstock sample from 1985, after the Puget Sound introduction, and a 1975 sample taken from the same area prior to the introduction. Our results generally supported previously hypothesized population structure. Most importantly, we found unique lake-rearing groups identified on the basis of a common life-history type were genetically related. Estimates of immigrant fraction using several different methods also generally supported previously identified populations. Estimates of effective population size were highly correlated with estimates of spawning abundance. The 1985 hatchery sample was genetically similar to contemporary Washington samples, and the contemporary Oregon Coast samples were similar to the 1975 Oregon Coast sample, suggesting that introductions of Washington coho salmon did not result in large scale introgression into Oregon populations.  相似文献

The maT clade of transposons is a group of transposable elements intermediate in sequence and predicted protein structure to mariner and Tc transposons, with a distribution thus far limited to a few invertebrate species. We present evidence, based on searches of publicly available databases, that the nematode Caenorhabditis briggsae has several maT-like transposons, which we have designated as CbmaT elements, dispersed throughout its genome. We also describe two additional transposon sequences that probably share their evolutionary history with the CbmaT transposons. One resembles a fold back variant of a CbmaT element, with long (380-bp) inverted terminal repeats (ITRs) that show a high degree (71%) of identity to CbmaT1. The other, which shares only the 26-bp ITR sequences with one of the CbmaT variants, is present in eight nearly identical copies, but does not have a transposase gene and may therefore be cross mobilised by a CbmaT transposase. Using PCR-based mobility assays, we show that CbmaT1 transposons are capable of excising from the C. briggsae genome. CbmaT1 excised approximately 500 times less frequently than Tcb1 in the reference strain AF16, but both CbmaT1 and Tcb1 excised at extremely high frequencies in the HK105 strain. The HK105 strain also exhibited a high frequency of spontaneous induction of unc-22 mutants, suggesting that it may be a mutator strain of C. briggsae.  相似文献

Traditionally viewed as an Andean grain crop,Chenopodium quinoa Willd. includes domesticated populations that are not Andean, and Andean populations that are not domesticated. Comparative analysis of leaf morphology and allozyme frequencies have demonstrated that Andean populations, both domesticated(quinua) and free-living(ajara), represent an exceptionally homogeneous unit that is well differentiated from allied domesticates of coastal Chile(quingua) and freeliving populations of the Argentine lowlands(C. hircinum). This pattern of relationships indicates that Andean populations represent a monophyletic crop/weed system that has possibly developed through cyclic differentiation (natural vs. human selection) and introgressive hybridization. Relative levels of variation suggest that this complex originated in the southern Andes, possibly from wild types allied withC. hircinum, with subsequent dispersal north to Colombia and south to the Chilean coast. Coastal populations were apparently isolated from post-dispersal differentiation and homogenization that occurred in the Andes. Other data point toward a center of origin in the northern Andes with secondary centers of genetic diversity subsequently developing in the southern Andes and the plains of Argentina. Comparative linkage of South American taxa, all tetraploid, with North American tetraploids of the subsection will eventually clarify this problem. While the possibility of a direct phyletic connection betweenC. quinoa and the Mexican domesticate(C. berlandieri subsp. nuttalliae,) cannot be excluded, available evidence indicates that the latter represents an autonomous lineage that is associated with the basal tetraploid, C. b. subsp.berlandieri, through var.sinuatum, whereas South American taxa show possible affinities to either var. zschackei or var.berlandieri. An extinct domesticate of eastern North America,C. b. subsp.jonesianum, represents either another instance of independent domestication, possibly from subsp. b. var.zschackei, or a northeastern outlier of subsp.nuttalliae.  相似文献
