A species of the flea beetle (Epitrix sp.) transmitted Andean potato latent virus (APLV) from Datura stramonium to D. stramonium, Nicandra physalodes and potato, and from N. physalodes to N. physalodes and D. stramonium. Although the acquisition and inoculation feeding times used were varied, this transmission always occurred at low efficiency. By contrast, APLV was very readily transmitted in tests in which infected plants of D. stramonium, N. physalodes, Nicotiana bigelovii and potato were brushed against healthy plants of one or more of these species. A low level of seed transmission of APLV occurred in potato.  相似文献

Potato cybrids result from the fusion between cytoplasm and nuclear gene donors. Such genetic materials are an alternative means to broaden the breeding pool by non‐sexual gene transfer. Tetrad pollen sterility provides also another source of male sterility with some potential for true potato seed breeding. The objective of this research was to investigate cybrid‐derived offspring for both agronomic and reproductive characteristics in two contrasting Peruvian locations, and to examine new exotic germplasm for tetrad sterility, with the aim of broadening the breeding pool available at the Centro Internacional de la Papa (CIP). The cybrids were derived from fusions between Y‐245.7, a clone with tetrad sterility, and Atzimba. These cybrids were crossed with selected male parents from the CIP breeding population, and their hybrid offspring were tested in La Molina (coastal desert) and Huancayo (cool highlands). In addition, other clones with tetrad sterility were also crossed with selected testers to determine their breeding value. There were significant differences for tuber yield, style length, and berry number among the hybrid offspring, and the genotype by environment interaction was significant for tuber yield and berry number. The top 25% highest yielding cybrid‐derived offspring across both locations showed the same tuber yield although they were significantly different for some of the reproductive characteristics. With the exception of one cybrid, the others did not exhibit segregation for tetrad sterility in their hybrid offspring, which were male fertile. However, the offspring derived from crosses between other sources of tetrad sterility and the same testers all showed tetrad sterility, and some of them had outstanding tuber yield at La Molina. The lack of segregation for tetrad sterility in these new crosses suggests that the non‐cybrid, male sterile, female parents are triplex or quadriplex for the Tr nuclear locus, which interacts with a sensitive cytoplasm (e.g. Trs from S. verrucosum or S. stoloniferum) to produce tetrad sterility in potato.  相似文献

根据马铃薯病毒PVX、PVY、PVA、PLRV的CP基因序列设计4对特异性引物,通过对试剂浓度和反应条件进行优化,建立了能够同步检测PVX、PVY、PVA、PLRV的一步法多重RT-PCR检测方法。该方法对PVX、PVY、PVA、PLRV扩增出的靶带大小分别为732、422、132和336 bp,凝胶电泳易辨别区分。病毒RNA最低检测限度为7.8 pg/μL,对PVM、PVS、AMV、TMV及PSTVd的扩增为阴性。研究结果表明,该方法特异、灵敏,比两步法多重RT-PCR检测更加快速、简便,提高了检测效率,降低检测成本,为马铃薯病毒的高效检测提供了有效手段。  相似文献

A true potato seed (TPS) synthetic cultivar from open pollination may broaden the genetic base in potato. A synthetic cultivar can also reduce the effect of inbreeding and maintain the productivity from generation to generation. Selected tetraploid parental genotypes, based on combining-ability tests, were chosen for isolated polycrosses to obtain tetraploid offspring. A series of experiments were undertaken to investigate the development of these TPS synthetic populations in two Peruvian locations: San Ramon, a rainfed, humid, mid-altitude environment, and La Molina, an arid, coastal environment under irrigation. Natural open-pollinated synthetics or control-mixture synthetics, involving two to six parents, had a similar tuber set but plant survival and tuber weight were higher in control synthetics including two or six parents. The results suggest that two-parent open-pollinated TPS synthetics may be a feasible option by selecting the right parents for the base population. A subsequent experiment showed that four-parent control synthetics (i.e. double crosses) were the best for tuber weight and size, followed by the two-parent control synthetics. Synthetic breeding populations could be shared with other breeders, who in cooperation with their local farmers may select promising genotypes for further cultivar release. The original variation in the source population as well as the selection method may influence the genetic variation in further cycles of genetic improvement. Therefore, the objectives of this research were to determine genetic parameters (variance components and heritability) in source and intermediate stages of a true potato seed (TPS) breeding population and to calculate the genetic and phenotypic correlations in this breeding material developed by the Centro Internacional de la Papa (CIP). The intermediate stage was derived from a source population adapted to the warm lowland tropics plus introduction of exotic germplasm from North America and Europe. Non-additive genetic variation was almost nil for plant survival, tuber yield and tuber shape uniformity in both stages of the breeding population and no quantitative genetic variation for uniformity of tuber color was observed in both source and intermediate breeding materials. Heritability was higher in the intermediate stage than in the source population for plant survival (0.86 vs 0.66), tuber yield (0.30 vs 0.14) and tuber shape (0.77 vs 0.51), but it was the reverse for tuber uniformity (0.11 vs 0.72). These results suggest that potato breeders at CIP were able to keep enough genetic variation for most important characteristics for potato production from true seed in their intermediate breeding materials by adding new sources of variation to the original breeding population. Additive genetic and phenotypic correlations were significant and positive between plant vigor after transplanting and tuber yield, and tuber shape and tuber uniformity, which suggest that high yielding offspring result from early vigorous growth, and that tuber uniformity could depend on tuber shape uniformity in this breeding material.  相似文献

A small proportion (1–4%) of the seeds of Stellaria media extracted from fallow soil from three widely separated areas contained cucumber mosaic virus (CMV). S. media seeds buried for 21 months produced 5 % infected seedlings. S. media plants from Britain, N. America and Australia were least severely affected by the CMV strain obtained from their country of origin and showed more severe reactions when infected with two alien strains. Several weed species were experimentally infected with lettuce mosaic, turnip mosaic and cauliflower mosaic viruses but, although virus was detected in the seeds of some species, it was not transmitted to any of their seedlings.  相似文献

Twenty-five true potato seed progenies and five clonally maintained cultivars were grown in a glasshouse. Tubers of each clone of each progeny were grown in a field experiment. At harvest, one seed tuber was taken from each plant and field grown for a second year. After each experiment, the tubers from every plant were weighed and counted and their colour, shape, form (regularity) and skin finish assessed. Progeny means and variances for each character were calculated. There were significant differences between means for each character in all experiments, except mean yield in the glasshouse, variances for each character in both field experiments and tuber number variances and colour variances in the glasshouse. In general, Pearson's product-moment correlations between glasshouse and field assessments were moderate or poor (r ≤ 0.6) and inconsistent between experiments. Spearman's rank correlations between glasshouse and field assessments for progeny means were similar to the linear correlations, while those for variances were somewhat better. Rank correlation gave more consistent results between experiments. Ranking glasshouse means will allow some worthwhile progeny selection for tuber colour, shape and skin finish. It can be a useful screen for progenies with the highest levels of uniformity for all characters.  相似文献

Tuber yield in open-pollinated (OP) offspring of potato cultivars appears to be correlated with the proportion of hybrids within each population. OP true potato seed (TPS) from ten selected clones was tested in an experiment at two Peruvian locations, San Ramón (mid-altitude warm tropics) and Huancayo (highland cool tropics), to investigate the early selection effect on OP populations. Another experiment in La Molina (dry coast) was included to confirm the results from these other two locations. The results clearly showed that plant survival, tuber yield and berry number can be increased by a strong early selection. Likewise, the results suggested that the most-promising OP offspring for TPS production could be derived from tetraploid clones with a high rate of outcrossing. This experiment demonstrates that high tuber yield may be obtained in OP TPS cultivars with little effort, using early selection for seedling vigor in the nursery. Genetic interpretation of this response to early selection indicates that both the intensity of selection and non-additive genetic variation for tuber yield account for these observed gains. Transmission of potato vims Y to sweet pepper by the green peach aphid, Myzus persicae (Sulzer), was inhibited by foliar applications of 1.0% or 2.0% neem seed oil to infected source plants or to uninfected recipient plants. Neem seed oil interfered with virus acquisition and inoculation in a manner comparable to that of a commercial horticultural oil, while an oil-free neem seed extract did not reduce rates of transmission compared with controls. The finding that neem seed oil inhibits virus transmission, while oil-free neem seed extract does not, suggests that the presence of the oil rather than biologically active limonoids such as azadirachtin interfere with virus transmission. None of the treatments affected rates of infection when potato virus Y was transmitted mechanically, or the resulting virus titre and symptom expression. In addition to direct control of insect pests, formulated neem oils may help reduce or delay the spread of non-persistent plant viruses.  相似文献

The aim of this research was to determine the genetic variation available in some important characteristics for true potato seed breeding in intermediate and advanced stages of a breeding population developed by the Centro Internacional de la Papa. A factorial mating design was used to calculate variance components and heritability at both selection stages. Four males were crossed with four females within each set (4 for intermediate stage and 5 for advanced stage) and their resulting offspring tested across two contrasting locations. Tuber yield and set were the only common characteristics recorded in both selection stages. In the intermediate stage early development characteristics (seed germination plus root and internode length) were also measured. In the advanced breeding material, vine earliness and other reproductive traits (days to flowering, flowering intensity, style length and pollen production) were scored. The heritability for tuber yield (0.35) and tuber set (0.32) in the advanced selection stage was higher than in the intermediate stage (0.26 and 0.13 respectively), which suggest that recombination through more cycles of recurrent selection brought untapped variation for both characteristics in this breeding material. Significant additive genetic variation, and thereby high heritability, was observed for internode length (0.52) in the intermediate selection stage, and for days to flowering (0.53) in the advance stage. Progress through selection may be expected for both characteristics. The heritability for pollen production was intermediate (0.23), while it was low (i.e.< 0.10) for the other characteristics recorded in both populations.  相似文献

The world's climate is changing. This can affect agricultural insects and the damage they cause by altering their ecology, behavior and habitats. Aphids are emerging as an indicator of climate change, and they are a good model for understanding these environmental changes, especially based on their migratory behavior. In this study, the occurrence of aphids in a seed potato field in Pyeongchang, Korea, was monitored daily using a yellow water pan trap from 1977 to 2017. During these 40 years, the annual migration fluctuations of the aphids were analyzed, with particular focus on four species known to vector potato viruses, Aphis gossypii, Aulacorthum solani, Macrosiphum euphorbiae, and Myzus persicae. It was found that, as time goes by, the start point of spring migration in these aphid species was getting earlier while the peak in autumn migration gradually moved to the winter season. Moreover, the annual total number of collected aphids increased by 3.74 times during the 40 years. Together with a mean annual temperature increase of 1.03°C in Pyeongchang over the past four decades, there is a potential threat of expansion in the distribution of aphid species. Therefore, we carefully hypothesized that this temperature change could influence the aphid's population changes in the Korean peninsula. This information can be used in an integrated aphid management program in seed potato cultivation.  相似文献

On reaching the stem each virus usually first moved downward, but in some plants both upward and downward movement occurred simultaneously and in a few
upward movement occurred first.
All three viruses travelled through the stem at approximately the same rate. Each was capable of travelling more than 80 cm. during the first 12 hr. after entering the stem, giving a minimal average rate of about 8 cm. per hr.
Uninfected pieces of stem invariably occurred between infected pieces. Maximum lengths of stem through which virus particles had apparently passed without causing infection, were 44.5, 49 and 39 cm. for the three viruses.  相似文献

The infection of viruses to a neighboring cell is considered to be beneficial in terms of evasion from host anti-virus defense systems. There are two pathways for viral infection to "right next door": one is the virus transmission through cell-cell fusion by forming syncytium without production of progeny virions, and the other is mediated by virions without virus diffusion, generally designated cell-to-cell transmission. Influenza viruses are believed to be transmitted as cell-free virus from infected cells to uninfected cells. Here, we demonstrated that influenza virus can utilize cell-to-cell transmission pathway through apical membranes, by handover of virions on the surface of an infected cell to adjacent host cells. Live cell imaging techniques showed that a recombinant influenza virus, in which the neuraminidase gene was replaced with the green fluorescence protein gene, spreads from an infected cell to adjacent cells forming infected cell clusters. This type of virus spreading requires HA activation by protease treatment. The cell-to-cell transmission was also blocked by amantadine, which inhibits the acidification of endosomes required for uncoating of influenza virus particles in endosomes, indicating that functional hemagglutinin and endosome acidification by M2 ion channel were essential for the cell-to-cell influenza virus transmission. Furthermore, in the cell-to-cell transmission of influenza virus, progeny virions could remain associated with the surface of infected cell even after budding, for the progeny virions to be passed on to adjacent uninfected cells. The evidence that cell-to-cell transmission occurs in influenza virus lead to the caution that local infection proceeds even when treated with neuraminidase inhibitors.  相似文献

The production of attractive, uniform true potato seed (TPS) progenies was investigated. Four breeding schemes were compared: intercrossing tetraploid cultivars (cv x cv); doubled dihaploids x cultivars (ddh x cv); cultivars x diploid unreduced-gamete producers (cv x FDR) and doubled dihaploids x diploid unreduced-gamete producers (ddh x FDR). Fifty three progenies and five clones were grown in a glasshouse in a randomised complete block design with three replicates of 25 plants per progeny and clone. Each plant's tubers were counted and the colour, shape, quality of skin finish, flesh colour, and commencai attractiveness (which includes yield) recorded. The most uniform progenies were also selected by visual comparison with the clones. For mean attractiveness, differences (P < 0.001) between breeding schemes and between progenies within breeding schemes were detected. The cv x cv and cv x FDR progenies were more attractive than clonal controls. There were significant additive and non-additive effects for attractiveness in all breeding schemes except cv x FDR. There were between-progeny differences (P < 0.001) for uniformity for all characters. Progenies uniform for one character could be variable for other traits. Breeding schemes gave different levels of uniformity (P < 0.001) for all characters except shape and flesh colour, but none gave low levels of variation for all traits. Doubled-dihaploid parents increased the variation in progenies. There were uniformity differences (P < 0.001) between progenies within breeding schemes for all characters. Evidence of additive and nonadditive genetic variation for uniformity in all traits was detected. In each breeding scheme, parents with good general combining ability (GCA) for uniformity in several characters were identified. Visually selected uniform progenies had parents with good GCAs for uniformity in a range of traits and high specific combining abilities (SCAs) for several traits. A desynaptic first-division restitution (FDR) clone and a male-sterile doubled-dihaploid clone had the best GCAs for tuber uniformity in TPS progenies. Achieving multitrait uniformity in TPS is problematic but may be aided by the selection of parents with GCAs for uniformity coupled with progeny testing to allow for non-additive effects.  相似文献

Western Australia has a unique seed potato production scheme which has remained virtually unchanged for more than 60 years, consisting of summer plantings of predominantly one cultivar in wind-exposed coastal swamplands. No rotation is used and the scheme relies on natural winter flooding and 'grazing' by sheep to eliminate unharvested tubers. Stocks are recycled every year with only limited inputs of pathogen-tested seed tubers in recent times. Virus spread in the crop is controlled by selecting large tubers for planting, roguing, aphicide application and growing season inspections. Potato viruses X and S were commonly detected in old seed stocks produced by this scheme attaining 100% infection in some. Both viruses were less frequently found in newly introduced seed stocks. By contrast, potato virus Y was never detected and potato leaf roll virus rarely found.  相似文献

Grape seed extract (GSE) is reported to have many pharmacological benefits, including antioxidant, anti-inflammatory, anticarcinogenic, and antimicrobial properties. However, the effect of this inexpensive rich source of natural phenolic compounds on human enteric viruses has not been well documented. In the present study, the effect of commercial GSE, Gravinol-S, on the infectivity of human enteric virus surrogates (feline calicivirus, FCV-F9; murine norovirus, MNV-1; and bacteriophage MS2) and hepatitis A virus (HAV; strain HM175) was evaluated. GSE at concentrations of 0.5, 1, and 2 mg/ml was individually mixed with equal volumes of each virus at titers of ~7 log(10) PFU/ml or ~5 log(10) PFU/ml and incubated for 2 h at room temperature or 37°C. The infectivity of the recovered viruses after triplicate treatments was evaluated by standardized plaque assays. At high titers (~7 log(10) PFU/ml), FCV-F9 was significantly reduced by 3.64, 4.10, and 4.61 log(10) PFU/ml; MNV-1 by 0.82, 1.35, and 1.73 log(10) PFU/ml; MS2 by 1.13, 1.43, and 1.60 log(10) PFU/ml; and HAV by 1.81, 2.66, and 3.20 log(10) PFU/ml after treatment at 37°C with 0.25, 0.50, and 1 mg/ml GSE, respectively (P < 0.05) in a dose-dependent manner. GSE treatment of low titers (~5 log(10) PFU/ml) at 37°C also showed viral reductions. Room-temperature treatments with GSE caused significant reduction of the four viruses, with higher reduction for low-titer FCV-F9, MNV-1, and HAV compared to high titers. Our results indicate that GSE shows promise for application in the food industry as an inexpensive novel natural alternative to reduce viral contamination and enhance food safety.  相似文献

 Inbreeding depression may affect the performance of consecutive generations of potatoes propagated by true potato seed (TPS). The effect of inbreeding was established using selfed and sib-mated generations of five TPS families. Correlation coefficients were calculated between the level of inbreeding and different traits. Inbreeding depression was expressed mainly by pollen viability, as measured by its stainability (r=−0.912, P<0.01), and tuber yield (r=−0.837, 0.01<P<0.05). The results also indicated that without unavoidable selection inbreeding depression is expected to be more evident. Furthermore, the TPS families responded quite differently to inbreeding depression. They did not show the same amount of depression for yield as they did for the characters concerning fertility. Attachment of virus particles to antiserum-coated electron microscope grids (immunosorbent electron microscopy) provided a test that was at least a thousand times more sensitive than conventional electron microscopy for detecting potato leafroll (PLRV) and potato mop-top (PMTV) viruses. The identity of the attached virus particles was confirmed by exposing them to additional virus antibody, which coated the particles.
PLRV particles (up to 50/μm2 of grid area) were detected in extracts of infected potato leaves and tubers, infected Physalis floridana leaves, and single virus-carrying aphids. On average, Myzus persicae yielded 10–30 times more PLRV particles than did Macrosiphum euphorbiae .
PMTV particles (up to 10/μm2 of grid area) were detected in extracts of inoculated tobacco leaves, and of infected Arran Pilot potato tubers with symptoms of primary infection. Particles from tobacco leaves were of two predominant lengths, about 125 nm or about 290 nm, and fewer particles of other lengths were found than in previous work, in which partially purified or purified preparations of virus particles were examined, using grids not coated with antiserum.  相似文献
