An increase in the K+ concentration in the medium to 60 mM which causes depolarization of cell membranes in the rat cerebral cortex is studied for its effect on the cGMP level, guanylate cyclase and cGMP phosphodiesterase activities in norm and one hour after X-ray irradiation. The cGMP content and guanylate cyclase activity in normal rats and 1 min after depolarization are shown to increase with the external K+ concentration. One hour after irradiation the activity of enzymes under examination is three times as high. The character of changes in the cGMP content caused by a rise of the external KCP concentration is mainly determined by variations in the guanylate cyclase activity under these conditions.  相似文献

It has been shown that the malonic dialdehyde (MDA) content in spleen lymphocytes of rats increases after whole-body X irradiation with a dose of 0.5 Gy to reach the maximum level in 24 h. Simultaneously, the concentration of cGMP and free cytosol Ca2+ increases. With a dose of 1 Gy MDA content of cells increases 6 h following irradiation. The maximum drop of the release of viable lymphocytes from the spleen and thymus, observed 24 h and 3 days after irradiation respectively, coincides with the appearance of the second peak of the MDA content. The level of cGMP remains decreased throughout the period of about 6 days. The onset of lymphocyte repopulation in the spleen on day 6 coincides with the decrease in the MDA level and increase in the cytosol Ca2+ concentration.  相似文献

The effect of different chemical compounds on the cAMP/cGMP ratio in the bone marrow of mice and radioresistance of animals has been studied. It has been shown that all compounds possessing radioprotective properties give rise to the cAMP/cGMP ratio in the bone marrow of mice. No changes in cAMP and cGMP level were noted after the administration of nonradioprotective substances. The maximal radioprotective effect coincide in time with the largest increase of the cAMP/cGMP ratio. The injection of radioprotectors at different doses demonstrate clearly that only at radioprotective doses the increase in the cAMP/cGMP ratio takes place. The administration of some substances 2, 15 and 60 min after the irradiation of mice shows that the radioprotective effect, though modest, was evident only in one case of elevated cAMP/cGMP ratio (the injection of 2-Mercaptoethylamine 2 min after the irradiation). Evident radioprotective effect occurs at the cAMP/cGMP ratio of about 170-200%; the ratio of about 130-140% corresponds to small radioprotection.  相似文献

Ceruloplasmin administered 60 min before irradiation diminished cAMP and cGMP levels, which were increased by irradiation at LD50 and LD100, and normalized cAMP/cGMP ratio in the rat liver during the first 24 h following irradiation with a dose of 6.24 Gy. The content of phospholipids increased and that of cholesterol decreased under the effect of ceruloplasmin leading to normalization of the molar cholesterol/phospholipid ratio in the rat liver on the 7th day of radiation sickness (LD50, 6.24 Gy).  相似文献

Ovariectomized, female Sprague-Dawley rats were trained to run on a treadmill for up to several minutes. On the day of the experiment the treadmill speed was adjusted to either 680, 1360 or 2720 cm/minute and separate groups of 5 rats were required to run at each of these speeds for either 15, 30, 45 or 60 seconds. Immediately after running on the treadmill, each rat was killed by microwave irradiation, and the cerebellum was collected for subsequent determination of cyclic guanosine monophosphate (cGMP). The time to the onset of an elevation of cGMP and the maximal elevation obtained were functions of the speed of running. The first significant increase in cerebellar cGMP over that observed in inactive control rats occurred within 15 seconds after the onset of running. Regardless of the speed of running, the cGMP content reached a plateau after 45 seconds.  相似文献

Changes have been revealed in the function of cyclic GMP system of thymus and liver of irradiated (8 Gy) mice. In the thymus the cGMP level increased during the first 60 min following irradiation. In the liver the concentration of cGMP exhibited two peaks: 30 min and 24 hr after irradiation. The changes observed in the cGMP level are connected with the increased guanylate cyclase activity of thymocytes and liver of irradiated mice and, less likely, with changes in the activity of cGMP phosphodiesterase of these tissues.  相似文献

In experiments with dogs and monkeys a study was made of the dynamics of the content of prostaglandins (PG) and cyclic nucleotides after gamma irradiation. In dogs irradiation with lethal doses (3.1 and 50 Gy) caused a short-term, evidently stress growth of cAMP, PGE and PGF2 alpha levels. At the height of radiation sickness PGF2 alpha and cGMP content decreased considerably. Irradiation of monkeys with a nonlethal dose of 3.2 Gy changed PGE and PGF2 alpha levels to a lesser extent, while concentrations of cyclic nucleotides varied but their ratio remained stable.  相似文献

Strips of bovine mesenteric arteries brought to sustained contraction by the addition of 3.0 microM phenylephrine relaxed when exposed to ultraviolet radiation (366 nm). The relaxation was reversible and associated with a rapid increase in the cGMP content. After termination of the radiation the cGMP level rapidly decreased below the basal level. The crude soluble guanylate cyclase from the artery was stimulated about 8-fold by ultraviolet radiation (366 nm). Neither the cGMP-phosphodiesterase activity nor the cAMP level were found to be changed during irradiation. The ultraviolet light-induced relaxation was not dependent on an intact intimal surface. Furthermore, the relaxing effect was found to be enhanced and accompanied by a larger increase of the cGMP level in nitroglycerin-tolerant arteries. The present results show that the ultraviolet light-induced relaxation in bovine mesenteric arteries is associated with a rapid increase in the cGMP content and that ultraviolet light and nitrocompounds may exert their relaxing actions through a common substance.  相似文献

Phosphodiesterase of cyclic nucleotides from membranes of rat brain synaptosomes hydrolyzes cAMP and cGMP; the maximal rate of cAMP hydrolysis is 2.5 times higher than the values of the analogous index for GMP. The enzyme is found to be activated by calmodulin. A different direction of changes in the rate of cAMP and cGMP hydrolysis is observed 1 h after total X-ray irradiation. The process of cAMP hydrolysis by phosphodiesterase is characterized by positive cooperativity which is also shown after irradiation and for the process of cGMP hydrolysis. It is established that enzyme inhibition by the reaction product takes place both in control and after irradiation.  相似文献

The secretory response implicated in the intestinal response to luminal attack is altered by radiation. The cAMP, cGMP and Ca(2+)(i) pathways leading to secretion as well as the interactions between the cAMP pathway and the cGMP or Ca(2+)(i) pathway were studied in the rat distal colon 4 days after a 9-Gy abdominal X irradiation, when modifications mainly occurred. The secretory response in Ussing chambers and cAMP and cGMP accumulation in single isolated crypts were measured. The muscarinic receptor characteristics were determined in mucosal membrane preparations. The secretory response by the cAMP pathway (stimulated by vasoactive intestinal peptide or forskolin) and the cAMP accumulation in crypts were decreased (P < 0.05) after irradiation. The weak secretory response induced by the cGMP pathway (stimulated by nitric oxide or guanylin) was unaltered by radiation, and the small amount of cGMP determined in isolated crypts from the control group became undetectable in the irradiated group. Inducible NOS was not involved in the hyporesponsiveness to VIP after irradiation (there was no effect of an iNOS inhibitor). The secretory response by the Ca(2+)(i) pathway (stimulated by carbachol) was unaffected despite a decreased number and increased affinity of muscarinic receptors. The non-additivity of VIP and carbachol co-stimulated responses was unmodified. In contrast, VIP and SNP co-stimulation showed that NO enhanced the radiation-induced hyporesponsiveness to VIP through a reduced accumulation of cAMP in crypts. This study provides further understanding of the effect of ionizing radiation on the intracellular signaling pathways.  相似文献

An increase (123%) of cyclic GMP (cGMP) was observed in the hippocampus of the rat killed by microwave irradiation 45 min after administration of 500 mg/kg gamma-hydroxybutyrate (GHB) IP. This increase is time and dose dependent. No modification in cyclic nucleotide content was observed in striatum and in cerebellum. As the role of GHB has been implicated in neurotransmission, the fact that this compound increases cyclic GMP accumulation in hippocampus in vivo may represent a mechanism by which the actions of GHB are mediated at the cellular level. Valproate (400 mg/kg) or naloxone (10 mg/kg) pretreatment completely abolish the cGMP increase due to GHB. A GABAergic and/or opiate phenomenon may be involved in the mechanism of GHB induced increase of cGMP.  相似文献

Changes in the content of cyclic nucleotides (cAMP and cGMP) and related enzyme activities were observed in the rat thyroid, pituitary and plasma during the prolonged increase of endogenous TSH produced by treatment with methylthiouracil (MTU). Experiments were performed after 4 weeks treatment with MTU. The wet weight and cAMP content per wet weight of the thyroid increased 3 and 1.4 times respectively, but cGMP showed a slight decrease. Pituitary weight increased 1.3 times, but cAMP and cGMP content did not change. The cAMP level in plasma also increased about 1.3 times, but cGMP did not increase. The cAMP-phosphodiesterase activity in the thyroid, pituitary and plasma was increased 1.9, 1.4 and 1.3 times respectively after MTU treatment, while cGMP-phosphodiesterase showed no significant change. ATPase activity in the thyroid and pituitary was also increased more than 1.5 times after MTU treatment, while 5'-nucleotidase activitity decreased remarkably. These data indicate that the metabolism of the cyclic nucleotide system in the thyroid is stimulated by TSH.  相似文献

cGMP content of axoplasm from the giant axon of Loligo forbesi was investigated after subjecting the axon to various treatments. Repetitive electrical stimulation or depolarisation by high K+ caused no change in cGMP content. Glutamate and serotonin were also without effect. The nicotinic agonist carbachol (100 microM) increased cGMP levels by 90% (n = 5). A large transient elevation of cGMP content was evoked by external nitroprusside (10 nM-20 microM in intact axons. Nitroprusside injected into both extruded axoplasm and intact axons also increased cGMP content, the stimulation being considerably higher in intact axons where the axolemma was also present. Nitroprusside was also active in axons where the soluble cytoplasmic components were washed out by internal perfusion.  相似文献

Male and female rats were given oestradiol benzoate (1 mg s.c. twice a week for 3 weeks) and/or sodium nitroprusside (SN), a donor of nitric oxide (NO), which was administered in their food in amounts of 0.2 or 0.6 mg/rat/day. Neither oestradiol-induced hypertrophy of the hypophysis, nor the serum prolactin (PRL) level, was affected by the simultaneous administration of SN. The PRL content of the hypophysis rose after oestradiol in the males, but the increase was again uninfluenced by the simultaneous administration of SN and the cAMP content of the hypophysis--raised after oestradiol--was likewise unaffected. The amount of cGMP in the hypophysis after oestradiol rose only in males. Both the serum and the hypophyseal prolactin level were found to be correlated to the cAMP and the cGMP content of the hypophysis. It was found that the simultaneous administration of SN together with oestradiol slightly reduced the increase in the cGMP content of the hypophysis elicited with oestradiol treatment only.  相似文献

The effect of estradiol-17beta, estriol, diethylstilbestrol and nafoxidine on rat uterine cGMP content was studied in relation with their respective estrogenic potency. Confirming previous results from Kuehl et al. (5), we observed a rise in uterine cGMP and a simultaneous decrease in cAMP content in treated animals. The reverse effect was obtained in the vagina after stimulation with estradiol or estriol. In the uterus, all compounds tested induced two main waves of cGMP increase corresponding to the two main phases of the estrogenic response i.e. the early fluid imbibition and the later period of true growth. No direct relationship could be established between the late rise in cGMP and true growth responses. A causal link between the first accumulation of uterine cGMP and the wet weight response in the early phase of estrogenic action is suggested by the comparison of time-course effects of the different compounds used on those two parameters.  相似文献

The effect of intraperitoneal administration of cGMP (0.5 mg per animal) on carbohydrate metabolism of wound area muscle tissue was studied in experiments on rats with linear skin wounds. The content of glycogen, gluconeogenesis, activity of glycogen phosphorylase, lactate dehydrogenase and malate dehydrogenase were studied. Cyclic GMP induced a substantial activation of glycogen metabolism (elevation of gluconeogenesis, increase in the activity of glycogen phosphorylase) even the third day after the operation. The animals not given cGMP demonstrated such an activation only the fifth day following the operation. Under the effect of cGMP the activity of lactate dehydrogenase rose the third day after the operation. Thus cGMP administration to the animals with wounds leads to an earlier mobilization of energy resources thereby promoting the acceleration of wound healing.  相似文献

Nitroglycerin (NG) caused a dose dependent-relaxation of the bovine mesenteric artery with an ED50-value of 2.7 × 10?8M. The relaxant effect of NG was significantly correlated to an increase in the cGMP content of the artery. There was a significant non-linear component in the data. At moderate cGMP levels relaxation and cGMP changes were correlated. At high levels of cGMP, however, the mechanism responsible for the nitroglycerin-mediated relaxation seemed to be completely activated and a further increase in cGMP did not induce additional relaxation. The cGMP content of the preparation was not significantly changed by nitroglycerin. The cGMP increase induced by nitroglycerin preceded the relaxation. A maximal increase of cGMP was observed after 2 min and the levels subsequently declined. This decline was not accompanied by an increase in the tissue tension. It is suggested from these experiments that cGMP might cause a relaxation of the vascular smooth muscle. Furthermore, if this suggestion is true, there seems to exist a “receptor reserve” for NG with respect to its relaxing action, since an over-capacity for cGMP production is present.  相似文献

The concentration of cGMP in the tissues of oat (Avena sativaL.) seedlings was shown to depend on seedling age and the light regime of their growth. The level of cGMP in the etiolated seedlings was lower than in the green ones and declined with seedling age. Red and blue light recognized by phytochrome and cryptochrome, respectively, affected the cGMP content. The effectors of cGMP metabolism, guanylin, protoporphyrin IX, and zaprinast, elevated the cGMP content in tissue extracts from oat seedlings.  相似文献
