This study determined the potential for short-term adaptation to fescue toxicosis and heat stress in rats. Male CD outbred rats (n=24) were implanted with temperature transmitters (Respironics, Bend, OR) to measure core temperature (Tc) and general activity. All rats were initially fed diets with ground, uninfected tall fescue seed (E−) and exposed to 21 °C (thermoneutral, TN) to establish baseline values. In Period 1, all groups were maintained at TN for 7 days, with one group fed a diet containing ground, endophyte-infected tall fescue seed (E+, approximately 165 μg ergovaline/kg BW/d) and two groups fed E− diet. Ergovaline is thought to be the primary toxin responsible for many symptoms associated with fescue toxicosis. Period 1 was followed by 7 days at 31 °C (heat stress, HS, Period 2) on the same diets. All animals were fed E− diet during the second 7 day of HS (Period 3). In the final 7 day (Period 4), E+ diet was returned to the original group and fed to one of the previously E− groups, with the third group remaining on E− diet. A 40% decrease in FI occurred with E+ treatment at TN (P<0.05), with a comparable BW reduction (P<0.05) after 4 day. Both responses worsened during HS. Treatment with E+ in Period 4 indicated that FI and BW had not adapted to fescue toxicosis. A reduction in daily Tc occurred with E+ treatment at TN (P<0.05) followed by hyperthermia during the initial stage of HS (P<0.05). Although feed intake and growth rate showed no change over time, there was a reduction in fescue toxicosis-induced hyperthermia in the heat with repeat treatment. Conditioning animals to fescue toxicosis and heat stress prior to exposure may be beneficial in reducing impacts on thermal status of the animal.  相似文献

Intake of endophyte-infected tall fescue by cattle results in fescue toxicosis, which is characterized by increased hyperthermia during heat stress and concomitant reductions in feed intake and growth. Rats were monitored at 21 or 31 °C for short- or long-term periods to determine temporal changes associated with the intake of endophyte-infected (E+) or uninfected (E−) fescue seed diets. Core temperature only changed in rats fed E+ diet at 31 °C. Intake of E+ diet reduced feed intake, daily gain, and serum prolactin. There were temporal and thermal differences in the response to endophytic toxins, with short-term changes diminishing over time at 21 °C, but increasing for certain parameters at 31 °C.  相似文献

Fescue toxicosis is a common syndrome of poor growth and reproductive performance of beef cattle grazing endophyte‐infected tall fescue infected with Lolium arundinaceum Schreb. Together with decreased feed intake, decreased growth rates and tissue necrosis due to vasoconstriction, depressed circulating serum prolactin concentrations are typically observed in cattle afflicted with fescue toxicosis. Polymorphisms within the XK, Kell blood group complex subunit‐related family, member 4 (XKR4) gene located on BTA14 have been previously reported to be associated with rump fat thickness, residual feed intake, average daily feed intake and average daily gain in cattle. Associations also have been reported between XKR4 genotype and effectiveness of the dopamine antagonist iloperidone as a treatment of schizophrenia in humans. Domperidone, a related dopamine antagonist, mediates effects of fescue toxicosis on livestock, including restoring depressed concentrations of prolactin. A mixed‐breed population of 592 beef cattle grazing endophyte‐infected tall fescue was used to examine the association between XKR4 genotype and circulating prolactin concentrations. The SNP rs42646708 was significantly (= 0.0002) associated with serum prolactin concentrations and explained 2.45% of the phenotypic variation. Effect of genotype at the SNP was tested across five breeds, with significant associations within both Angus (= 0.0275) and Simmental (= 0.0224) breeds. These results suggest XKR4 may play a role in mediating the negative effects of fescue toxicosis, and polymorphisms within this gene may be useful markers for selection for genetic resistance to the debilitating effects of endophyte‐infected tall fescue.  相似文献

The incidence ofAspergillus terreus recovered fromAcremonium coenophialum-infected and non-infected tall fescue grass and from the rumens of heifers grazing on the grasses was determined. The recovery ofA. terreus fromA. coenophialum-infected grass was similar to that from non-infected grass. The same was true of the recovery from the rumens of heifers on infected and non-infected grass. All heifers grazing onA. coenophialum-infected grass showed symptoms of the summer syndrome manifestation of fescue toxicosis while those grazing on non-infected grass did not;A. terreus is not a factor in fescue toxicosis in cattle. Acremonium coenophialum, a fungal endosymbiont in tall fescue, is responsible for the production of alkaloid toxins. Animals grazing endophyte-infected tall fescue often show toxicosis. In marginal environments, the endophyte is important for long-term survival of tall fescue. Few differences in endophyte isolates from individual tall fescue plants have been reported. To aid development of a toxicosis-free tall fescue, it is important to identify differences in endophyte isolates. This report describes variation in nitrogen utilization in a defined culture medium by endophyte isolates from Kentucky-31 tall fescue. Overall, the best nitrogen sources for dry weight production of mycelium were proline and potassium nitrate. Thirty-four isolates grown on agar-solidified defined media with single nitrogen sources showed variation in nitrogen utilization. Fifty percent of the isolates were unable to utilize two or more amino acids. Manipulation of endophyte variation could lead to development of a toxicosis-free tall fescue cultivar.  相似文献

Our objective was to determine whether consumption of endophyte-infected fescue seed affected male reproduction differently in a mouse line previously selected for susceptibility (S) to fescue toxicosis than in a line previously selected for fescue toxicosis resistance (R). For 8 weeks following weaning, 48 males per line were provided diets containing 50% of either endophyte-infected (E+) or endophyte-free (E-) fescue seed. Each male was then paired with a female for 1 week, with litter size and weight recorded from subsequent births. Males were then killed, testes and seminal vesicles were weighed, cauda epididymal sperm were collected and testis cross-sections were fixed. The E+ diet reduced litter size by 0.5 in mates of S males but increased it by 1.0 in mates of R males (line by diet interaction P=0.05). Testis traits were not affected by diet or the line by diet interaction. Sperm integrity was adversely affected by the E+ diet (P<0.01) but did not differ significantly between lines, nor were line by diet interactions important. In earlier work, the E+ diet reduced long-term reproduction by a larger amount in S- than in R-line mated pairs. Because the E+ diet had similar effects on reproductive traits in R and S males in the current experiment, we infer that the differential impact previously reported acted primarily through traits expressed in females.  相似文献

Tall fescue is one of the most widely grown forage grasses for horses in the United States. However, it is frequently infected with the endophyte Neotyphodium coenophialum which produces ergot alkaloids that cause severe adverse effects in the pregnant mare. The objectives of this study were to determine the effects of fescue toxicosis and fluphenazine on circulating relaxin in pregnant pony mares and evaluate the usefulness of relaxin as a monitor of treatment efficacy. Twelve mares were maintained on endophyte-infected tall fescue pasture. Group TRT (n = 6), received 25 mg of fluphenazine decanoate (i.m.) on Day 320 of gestation while Group UTRT served as untreated controls. Daily blood samples were collected from Day 300 of gestation until Day 3 post partum and analyzed for plasma relaxin concentrations using a homologous equine radioimmunoassay. Mean gestation lengths were 330 +/- 0.7 and 336.5 +/- 3.2 days for TRT and UTRT mares, respectively (P = 0.07). Mean plasma relaxin concentrations in both groups of mares during the week before treatment (Day 313 to 319) were not different (UTRT, 53.4 +/- 11.3 ng/mL; TRT, 61.4 +/- 9.3 ng/mL). In the week after treatment (Day 320 to 326), mean plasma relaxin tended to be higher (P = 0.1) in TRT mares (66.7 +/- 6.2 ng/mL) when compared with UTRT mares (49.6 +/- 6.6 ng/mL), representing a 17.1 ng/mL difference in circulating relaxin between the two groups. Systemic relaxin during the last week before delivery (days relative to parturition) for UTRT and TRT mares was 45.7 +/- 6.7 and 64.7 +/- 6.4 ng/mL (P = 0.06), respectively. At Day -8 and Day -5 relative to parturition, systemic relaxin in TRT mares was significantly higher (P < 0.05) than in UTRT mares. Three of the six UTRT mares and one TRT mare showed clinical symptoms of fescue toxicosis. In the week before delivery, circulating relaxin in mares with problematic pregnancies (39.9 +/- 7.8 ng/mL) was significantly lower than concentrations measured in mares with normal pregnancies (63.4 +/- 5.4 ng/mL; P = 0.03). Clinical observations suggest that a one-time injection with fluphenazine improved pregnancy outcome by reducing the adverse effects of fescue toxicosis concomitant with a stabilization of plasma relaxin concentrations. These data support the hypothesis that systemic relaxin may be a useful biochemical means of monitoring placental function and treatment efficacy in the mare.  相似文献

The objective was to evaluate the efficacy of domperidone in the prevention of reproductive complications of fescue toxicosis in periparturient mares. Pregnant mares at ≤310 days of gestation were fed ≥200 μg ergovaline per kg diet daily in endophyte-infected fescue hay and seed, starting ≥30 days before their expected foaling date (EFD: 340 days after breeding). Thirty-five mares were randomized to a treatment group to receive either domperidone gel (n = 20, 1.1 mg/kg, PO, once daily) or placebo (n = 15). Treatment was initiated 10 to 15 days before the EFD and continued for 5 days after foaling. “Treatment success” was defined as foaling within 14 days of the EFD, adequate mammary development on the day of foaling, and adequate lactation for 5 days postpartum. Twenty-seven mares were included in the effectiveness analysis. More mares in the domperidone group (12/13, P < 0.0001) were treatment successes than in the control group (1/14). Gestation length was shorter (P = 0.0011), and lactation at foaling (P = 0.0011) was better for the domperidone-group mares. Foals from two control mares were born dead and four others died or were euthanized within a few days after birth, compared with one foal death (an autolyzed twin) from a domperidone-treated mare. Plasma IgG concentrations were evaluated in 24 foals. Failure of passive transfer of immunoglobulins (IgG <800 mg/dL) occurred in 13/16 (81%) foals of domperidone-group mares and 7/8 (88%) foals of control mares. In conclusion, the reproductive complications of fescue toxicosis in periparturient mares induced by a fescue seed/hay model were prevented by treatment with domperidone.  相似文献

Tall fescue (Festuca arundinacea Schreb.), a highly competitive European grass that invades US grasslands, is reportedly allelopathic to many agronomic plants, but its ability to inhibit the germination or growth of native grassland plants is unknown. In three factorial glasshouse experiments, we tested the potential allelopathic effects of endophyte-infected (E+) and uninfected (E−) tall fescue on native grasses and forbs from Midwestern tallgrass prairies. Relative to a water control, at least one extract made from ground seed, or ground whole plant tissue of E+ or E− tall fescue reduced the germination of 10 of 11 species in petri dishes. In addition, the emergence of two native grasses in potting soil was lower when sown with E+ and E− tall fescue seedlings than when sown with seeds of conspecifics or tall fescue. However, when seeds of 13 prairie species were sown in sterilized, field-collected soil and given water or one of the four tall fescue extracts daily, seedling emergence was lower in one extract relative to water for only one species, and subsequent height growth did not differ among treatments for any species. We conclude that if tall fescue is allelopathic, its inhibitory effects on the germination and seedling growth of native prairie plants are limited, irrespective of endophyte infection. On the other hand, the apparent inability of these plants to detect tall fescue in field soil could hinder prairie restoration efforts if germination near this strong competitor confers fitness consequences. We propose that lack of chemical recognition may be common among resident and recently introduced non-indigenous plants because of temporally limited ecological interactions, and offer a view that challenges the existing allelopathy paradigm. Lastly, we suggest that tall fescue removal will have immediate benefits to the establishment of native grassland plants.  相似文献

The presence of an endophytic fungus, Acremonium coenophialum, in tall fescue (Festuca arundinacea) deterred aphid feeding by Rhopalosiphum padi and Schizaphis graminum. Both species of aphid were unable to survive when confined to endophyte-infected tall fescue plants. Feeding deterrents and toxic factors to R. padi and Oncopeltus fasciatus, large milkweed bug, were primarily associated with a methanol extract obtained when endophyte-infected tall fescue seed was serially extracted with hexane, ethyl acetate, and methanol. The concentrations of pyrrolizidine alkaloids were determined to be 30 to 100 times greater in the methanol extract than in the hexane and ethyl acetate extracts.  相似文献

The presented work was conducted on seedlings of spring barley and meadow fescue which differ in the degree of sensitivity to leaf spot pathogen Bipolaris sorokiniana (Sacc.) Shoem. The seedling reaction to inoculation with mycelium and conidia was examined in glasshouse conditions on the basis of respiration intensity and heat production. The leaf respiration was measured using Clark-type electrode, while heat emission was evaluated by means of isotermic microcalorimeter. The measurements were performed after 1, 3, 6, 10, 24, 48, 72, 168 and 240 hours since the inoculation moment. Leaves of meadow fescue were characterized by the most intense respiration at the 6th hour, while barley leaves at the 24th and 72nd hour after inoculation. In the case of meadow fescue the greatest heat production was noted in the period between 24 and 168 hours after inoculation. Simultaneously, at the 48th hour the smallest rate of respiration was observed. Barley leaves emitted the greatest amount of heat only in the first 3 hours of the pathogenesis. In these hours the smallest respiration rate was noted. The observed, opposing reaction of respiration intensity and heat emission in the infected seedlings of both species may illustrate a disorder in metabolic processes in plants during pathogenesis. The plants studied differed in the time of their reaction to pathogen attack: barley responded earlier in heat production, while fescue extended respiration rate in the first hours after inoculation. This is clearly observable, when coefficients of metabolic inefficiency (heat rates per mole O2) are compared. In the case of barley the highest rates were noticed just after inoculation, whereas in fescue at the 48th hour. In both species attack of pathogen caused high metabolic efficiency.  相似文献

Four experiments were conducted to assess the effect of foliar applications of various nutrient solutions on the phylloplane yeast community of tall fescue (Festuca arundinacea Schreb.). In the first three experiments, increasing concentrations of sucrose (2–16%), yeast extract (0.5–2.5%), and sucrose plus yeast extract (2.5–18.5% total) were applied and the yeast colony forming units (cfu) enumerated 14 h later by dilution plating. Significant positive linear relationships were observed between the number of yeast cfu and applications of both yeast extract and sucrose plus yeast extract. Foliar applications of sucrose alone had no significant effect on yeast community abundance, indicating that phylloplane yeasts of turfgrass are not limited by the amount or availability of carbohydrates. In the fourth experiment, five different solutions were applied to tall fescue to investigate the response of the yeast community to organic and inorganic nitrogen sources. Tryptone or yeast extract, both with considerable amino acid composition, significantly increased the yeast population, while yeast nitrogen base (with or without amino acids) and ammonium sulfate had no affect on yeast abundance. These results suggest that organic nitrogen stimulate yeast community growth and development on the phylloplane of tall fescue, while carbohydrates, inorganic nitrogen, and non-nitrogenous nutrients have little positive effect.  相似文献

Summary Ergovaline's role in the direct causation of fescue toxicosis first requires establishment of its dietary absorption. Therefore, ergovaline movement across human intestinal cells was assessed using Caco-2 cells derived from human colon carcinoma. A pre-equilibrated mixture of ergovaline/ergovalinine (60∶40 ratio of isomers) was added to the apical compartment, and isomer movements were assessed by high-performance liquid chromatography (HPLC) of extracted media (initial pre-isomerized ergovaline concentrations of 6 and 22 μM, two doses). Mathematical models for ergot alkaloid movement were developed. Rates of movement were not different for the isomers. In the absence of cells, basal accumulation of isomers was essentially linear for 3 h regardless of loading concentration, after which basal accumulation of ergovaline/ergovalinine plateaued. Rates of ergovaline/ergovalinine movement in the presence of cells slowed to about 25% the rate of movement in the absence of cells (22 μM, kt=0.0133 no cells, 0.0036 with cells, P<0.05). Mass transfer rate was 7.5 ng·cm−2·min−1 and was similar to that reported for ergovaline using a parabiotic chamber with sheep omasum. After 6 h in the presence of cells, ∼25 and 40% of the total ergovaline/ergovalinine administered had accumulated in the basal compartment for 6.6 and 22 μM treatments, respectively. Ergovaline and its naturally occurring isomer, ergovalinine, readily crossed intestinal cells intact and at similar rates. Either isomer, or a combination of both, could be involved in the pathogenesis of fescue toxicosis at sites distal to the intestine.  相似文献

The aim of our work is to investigate the changes in phenolic level, PAL activity and heat production rate induced during pre-hardening at 12°C and cold acclimation at 2°C of the forage grasses Festulolium, meadow fescue, tall fescue and Italian ryegrass in relation to their resistance to snow mould caused by Microdochium nivale. Meadow fescue and tall fescue were most resistant to M. nivale infection, while Italian ryegrass demonstrated the least resistance to this fungus inoculation. Festulolium, meadow fescue and tall fescue responded similarly to low temperature, while Italian ryegrass demonstrated considerable disturbance of energy balance and lower phenolic concentration, which could explain a higher susceptibility of the latter species to infection by M. nivale. The enhanced level of phenolic compounds, probably utilised for cell wall lignification as well as equilibrium of the metabolic activity observed in meadow fescue and tall fescue, is very important for both cold and pathogen-resistance mechanisms. The studied Festulolium cultivar ‘Felopa’, a hybrid of the Lolium multiflorum and Festuca pratensis genomes, was characterised by changes in biochemical parameters similar to the resistant meadow fescue and tall fescue.  相似文献

The objective was to examine follicular and luteal development and function in mature, lactating beef cows grazing endophyte free (E-) or endophyte infected (E+) tall fescue during the early postpartum period. Angus, Hereford, and Angus x Hereford cows were exposed to pasture for 37-39 days before synchronized estrus. Serum concentrations of prolactin were evaluated during the luteal phase before the synchronized estrus. Every Monday, Wednesday, and Friday for one estrous cycle ovaries were monitored by transrectal ultrasonography and blood was collected for determination of serum concentrations of progesterone and estradiol in cows that responded to synchronization. Signs of fescue toxicosis in E+ cows included decreased serum concentrations of prolactin (84.9+/-13.6 pg/ml versus 32.3+/-12.0 pg/ml; P < 0.009) measured during the luteal phase (day 37 of grazing) and decreased body condition of cows and weight of cows and calves (P < 0.001). Neither serum concentrations of progesterone or estradiol, nor diameter of the CL differed between treatments. Diameter of the largest follicle tended to be smaller for cows grazing E+ fescue, especially between days 8 and 12 of the estrous cycle (P < 0.08). Numbers of class 1 (3-5 mm) and class 3 (>10 mm) follicles were similar (P > 0.05) between treatments, but number of class 2 (6-9 mm) follicles was reduced in E+ cows for most of the cycle (days 10 through 20; P < 0.03). Length of synchronized estrous cycle, days open, calving interval, and pregnancy rate at 30, 45, 60, and 90 days post-breeding was similar (P > 0.05) among treatment groups. Even though follicular dynamics (diameter of the largest follicle and number of class 2 follicles) were altered in cows grazing E+ tall fescue, follicular function was apparently not affected by ergot alkaloids.  相似文献

Tall fescue toxicosis and ergot alkaloids cost U.S. livestock producers approximately one billion dollars in annual livestock production loss annually. Ergovaline (EV) is the tall fescue alkaloid primarily responsible for clinical disease in livestock. Since native ruminal microorganisms have not been attributed to the detoxification of EV, finding detoxifying microbes from other environments is desirable. One possible source for potential microorganisms that can degrade EV is the anaerobic gut of the earthworm, Eisenia fetida. This study describes a comparative microbial analysis of earthworm digestive tracts receiving 10,000 ppb EV (E+ treatment) when compared with a control treatment with no detectable amounts of EV (E− treatment). An HPLC assay determined a 25% loss of EV from the E+ treatment was microbial in nature. A community microbiomic approach of constructing 16S-rRNA gene clone libraries was used to compare the microbes affected by the two treatments. RDPII tools such as Classifier and Libcompare were used in the analysis of 16S sequences. DOTUR analysis was used to examine the richness and diversity of the two microbial populations in these experiments. The results indicate there are few significant differences in the microbial community structure between the two microbiomes.  相似文献

Small heat shock proteins are involved in stress tolerance. We previously isolated and characterized a rice cDNA clone, Oshsp26, encoding a chloroplast-localized small heat shock protein that is expressed following oxidative or heat stress. In this study, we transferred this gene to tall fescue plants by an Agrobacterium-mediated transformation system. The integration and expression of the transgene was confirmed by PCR, Southern, northern, and immunoblot analyzes. Compared to the control plants, the transgenic plants had significantly lower electrolyte leakage and accumulation of thiobarbituric acid-reactive substances when exposed to heat or methyl viologen. The photochemical efficiency of photosystem II (PSII) (Fv/Fm) in the transgenic tall fescue plants was higher than that in the control plants during heat stress (42°C). These results suggest that the OsHSP26 protein plays an important role in the protection of PSII during heat and oxidative stress in vivo.  相似文献
