Two hundred and eighty-eight arsenic-resistant bacteria were isolated by an enrichment culture method from a total of 69 arsenic-contaminated soil-samples collected from Dantchaeng district in Suphanburi province (47 samples), and from Ron Phiboon district in Nakhon Sri Thammarat province (22 samples), in Central and Southern Thailand, respectively. Twenty-four of the 288 isolated arsenic-resistant bacteria were found to be arsenite-oxidizing bacteria. On the basis of their morphological, cultural, physiological, biochemical and chemotaxonomic characteristics, and supported by phylogenetic analysis based upon their 16S rRNA gene sequences, they were divided into five groups, within the genera Acinetobacter, Flavobacterium, Pseudomonas, Sinorhizobium and Sphingomonas, respectively. Within genera, phylogenetic analysis using the 16S rRNA gene sequences suggested that they were comprised of at least ten species, five isolates being closely related to known bacteria (Acinetobacter calcoaceticus NCCB 22016T, Pseudomonas plecoglossicida FPC951T, Ps. knackmussii B13T, Sinorhizobium morelense Lc04T, and Sphingomonas subterranea IFO16086T). The other five proposed species are likely to be new species closely related to Flavobacterium johnsoniae, Sinorhizobium morelense, Acinetobacter calcoaceticus and Pseudomonas plecoglossicida, but this awaits further characterization for confirmation of the taxonomic status. No overlap in isolated species or strains was observed between the two sites. The strain distribution and characterization are described.  相似文献

微生物产生的胞外多糖(exopolysaccharides, EPS)可促进大粒径土壤团聚体形成,高产EPS的菌株在土壤改良、促进作物生长方面具有较好的应用前景。【目的】从土壤样品中筛选高产胞外多糖的细菌,研究其在土壤改良、环境适应性、广谱抗病等方面的功能,为制备土壤改良型功能菌剂提供候选菌株。【方法】采用蒽酮硫酸法测定菌株胞外多糖的产量,通过形态学观察、生理生化试验及16S rRNA基因序列测定确定其分类地位,结合土壤培养试验研究菌株对土壤团聚体形成的影响。【结果】获得3株胞外多糖产量大于500 mg/L的细菌,经鉴定A-5为地衣芽孢杆菌(Bacillus licheniformis),XJ-3为萎缩芽孢杆菌(Bacillus atrophaeus),KW3-10为耐盐芽孢杆菌(Bacillus halotolerans)。菌株A-5、XJ-3、KW3-10处理后,土壤大团聚体(>0.25 mm)含量较对照分别提高了4.07、2.14和3.16倍。3株菌株对疮痂链霉菌(Streptomyces scabies)、尖孢镰刀菌(Fusarium oxysporum)、茄链格孢菌(Alternaria solani)和立枯丝核菌(Rhizoctonia solani)等多种植物病原菌具有明显的抑制效果,可耐受pH为5-9和NaCl含量1%‒9%的盐碱环境,促进植物生长,其中KW3-10的代谢产物中IAA含量为25.58 mg/L。【结论】菌株A-5、XJ-3、KW3-10可显著促进土壤团粒结构形成,具有较好的广谱抗病性和促生长特性,可作为高效复合功能菌剂的候选菌株。  相似文献

Local adaptation to serpentine soils is studied using both transplant experiments and molecular genetic techniques. In long-lived species, such as pines, it is unclear how soon after germination local adaptation becomes detectable. Here I present results of a 36-year reciprocal transplant experiment using Pinus ponderosa, along with allozyme analyses from the same trees. Using a repeated measures analysis of variance, there is evidence for adaptation to serpentine soils; however, significant differences between source soil types do not become apparent until 20 years after the start of the experiment. Analysis of allozyme data showed no evidence for differentiation between the serpentine and non-serpentine populations. Comparing the performance of families over the course of the experiment found that there was little correlation between performance after 1 or 4 years of growth in the field and performance after 36 years. This suggests that short-term transplant experiments may not provide definitive evidence for adaptation to serpentine soils. A literature survey of all transplant studies using pine species growing on and off of serpentine soils found that studies that lasted fewer than 2 years showed no evidence for adaptation. However, in the two experiments (this one included) that lasted more than 2 years, both showed evidence for adaptation to serpentine soils. Serpentine soils, which contain relatively high concentrations of nickel and some other metals, are the preferred substrate for some plants, especially those that accumulate Ni in their tissues. In temperate regions more Ni-hyperaccumulator plants are found in Alyssum than in any other genus. In this study, serpentine soils of two areas (Marivan and Dizaj) in the west/northwest of Iran and also perennial Alyssum plants growing on these soils were analyzed for Ni and some other metals. The highest concentrations of total metals in the soils of these areas for Ni, Cr, Co and Mn were 1,350, 265, 94 and 1,150 μg g−1, respectively, while concentrations of Fe, Mg and Ca reached 3.55%, 16.8% and 0.585% respectively. The concentration of exchangeable Ni in these soils is up to 4.5 μg g−1. In this study two Alyssum species, A. inflatum and A. longistylum, have been collected from Marivan and Dizaj, respectively. Analysis of leaf dry matter shows that they can contain up to 3,700 and 8,100 μg Ni g−1, respectively. This is the first time that such high Ni concentrations have been found in these species. The concentrations of other metals determined in these species were in the normal range for serpentine plants, except for Ca, which was higher, up to 5.3% and 3.5%, respectively  相似文献

【目的】获得锰氧化细菌,对锰矿周边土壤中生物所参与的锰氧化过程进行初探。【方法】依据细菌是否能氧化Mn(Ⅱ),形成棕褐色锰氧化物进行筛选。利用染料LBB对生成的锰氧化物进行检测。通过考察分离菌株的形态、生理特征和16S r RNA基因、gyr B基因、gyr A基因序列的同源性对分离菌株进行鉴定。分析筛选菌与所在属已知锰氧化菌的亲缘关系。利用LBB显色法检测氧化锰的动态生成,通过扫描电镜-能谱分析和X射线衍射技术分析生物氧化锰的表征。【结果】获得1株锰氧化细菌菌株,命名为CP133,综合形态、生理及分子分析结果,鉴定为蜡样芽孢杆菌(Bacillus cereus),分离菌株与多株分离自海洋及土壤的芽孢类锰氧化菌在进化上具有一定的差异。与其他菌株比较菌株CP133具有较强的锰氧化能力,进入稳定期后可生成紧密结合在菌体周围的无定形态生物氧化锰。【结论】从锰矿周边土壤分离出1株具有较强锰氧化功能的蜡样芽孢杆菌,丰富了土壤芽孢类锰氧化菌的资源,同时也为锰矿周围土壤与锰氧化菌间的生物地球化学循环提供了线索及材料。  相似文献

Selection for metal-tolerant ecotypes of ectomycorrhizal (ECM) fungi has been reported in instances of metal contamination of soils as a result of human activities. However, no study has yet provided evidence that natural metalliferous soils, such as serpentine soils, can drive the evolution of metal tolerance in ECM fungi. We examined in vitro Ni tolerance in isolates of Cenococcum geophilum from serpentine and non-serpentine soils to assess whether isolates from serpentine soils exhibited patterns consistent with adaptation to elevated levels of Ni, a typical feature of serpentine. A second objective was to investigate the relationship between Ni tolerance and specific growth rates (μ) among isolates to increase our understanding of possible tolerance/growth trade-offs. Isolates from both soil types were screened for Ni tolerance by measuring biomass production in liquid media with increasing Ni concentrations, so that the effective concentration of Ni inhibiting fungal growth by 50% (EC50) could be determined. Isolates of C. geophilum from serpentine soils exhibited significantly higher tolerance to Ni than non-serpentine isolates. The mean Ni EC50 value for serpentine isolates (23.4 μg ml−1) was approximately seven times higher than the estimated value for non-serpentine isolates (3.38 μg ml−1). Although there was still a considerable variation in Ni sensitivity among the isolates, none of the serpentine isolates had EC50 values for Ni within the range found for non-serpentine isolates. We found a negative correlation between EC50 and μ values among isolates (r = −0.555). This trend, albeit only marginally significant (P = 0.06), indicates a potential trade-off between tolerance and growth, in agreement with selection against Ni tolerance in “normal” habitats. Overall, these results suggest that Ni tolerance arose among serpentine isolates of C. geophilum as an adaptive response to Ni exposure in serpentine soils. Two proteases, designated I and II, have been isolated from sporulating cells of Bacillus subtilis. They were partially purified by ammonium sulfate fractionation, Sephadex chromatography and affinity columns. Protease I was found to be similar to an already characterized B. subtilis protease. Protease II is trypsin-like in its substrate specificity and is distinct from protease I in its pH optimum, pH stability, molecular weight, substrate specificity, heat stability and sensitivity to various inhibitors. While both enzymes were produced primarily during sporulation, they attained maximum levels of activity at different times. Distinct functions for these proteases in post exponential B. subtilis are likely.  相似文献

In this study, we isolated bacteria from rhizosphere and endorhizophere of wheat crops of the central region of Argentina. The isolates were phenotypically characterized and the restriction patterns of 16S rDNA (ARDRA) using endonuclease AluI were analysed. Representative isolates were used to evaluate the effect of the inoculation on the growth of wheat under greenhouse conditions. The effects of plant growth-promoting bacteria on wheat plants were studied by evaluating shoot fresh and dry weights and root fresh and dry weights. One native strain increased the shoot and root dry biomass by 23% and 45% respectively. Other strains increased the shoot dry biomass. A 1.5 kb fragment of the 16S rRNA gene of one isolate was sequenced. This isolate showed high identity with different species of Pseudomonas.  相似文献

【背景】开发、筛选优良益生菌菌种是当下畜牧业的研究热点,益生菌的潜在功能也被广为挖掘。【目的】分离、筛选具有良好耐受性且高产胞外蛋白酶的菌株,研究其生物学特性和酶学性质,为后续微生物蛋白酶的制备和微生态制剂的开发提供菌种资源。【方法】采集健康水貂新鲜粪便,配制酪蛋白培养基初筛和优化Folin-酚法复筛,对筛选菌进行生物学特性研究,获得产蛋白酶能力较强、耐受性较优的菌株,并进行常规鉴定和分子生物学鉴定,最后对蛋白酶酶学性质进行研究。【结果】筛选得到一株高产蛋白酶、耐受性较优的芽孢杆菌,经鉴定为枯草芽孢杆菌(Bacillussubtilis),编号为3。在初始发酵培养基条件下,酶学性质研究结果表明,该蛋白酶的最适反应温度为70℃,最适反应pH值为9.0,最佳金属离子激活剂为K+,Cu2+和Fe2+对酶活力有明显的抑制作用,在20%浓度的有机溶剂作用时蛋白酶未变性失活。【结论】从水貂粪便中分离获得一株具有良好的生物学特性、酶学性质和碱性蛋白酶活性的枯草芽孢杆菌,为该菌株在实际生产应用中提供了基础保障。  相似文献

【背景】利用微生物促进植物健康生长是农业可持续发展的重要方向之一,而种子相关的促生菌可在植物生命周期早期与植物相互作用,对植物健康生长具有重要意义。【目的】发掘与利用种子相关促生菌的前提是筛选获得促生菌菌种资源,验证其益生能力,为其进一步应用与机理研究提供依据与支持。【方法】以花生种子为研究对象,从种子表面及种子内部分离纯化多株菌,测定菌株的固氮、解磷、解钾、吲哚乙酸合成和铁载体合成等促生能力,并验证菌株对常见植物病原菌的生长抑制特性;通过16S rRNA基因序列进行系统发育分析,确定分类地位;通过生物膜形成能力及根际定殖能力测定菌株在植物根际的生存能力;最后通过催芽及盆栽试验测定菌株对花生种子发芽及幼苗生长的影响。【结果】从花生种子表面、种子内和胚根内分离筛选到41株菌,均有吲哚乙酸合成能力,其中35株有固氮能力,2株有铁载体分泌能力,14株有植物病原菌生长抑制能力。各选一株为代表的菌株,即PS3、PE5和PR5,经16S rRNA基因序列比对分析鉴定为芽孢杆菌属(Bacillus)。PS3、PE5和PR5均可在MSgg液体培养基表面形成褶皱较强的生物膜,也可在花生根际形成有效定殖。催芽试验结果表明经过促生菌浸种后花生种子萌发率明显提高,在第2天时,PS5将发芽率由14.17%提高至38.33%,PE5发芽率提高至30.83%,PR5发芽率提高至39.17%。三株菌能够明显促进花生幼苗生长,PS5对花生幼苗苗高、根长、鲜重和干重分别提高21.82%、22.20%、37.11%和35.64%,PE5分别提高17.45%、18.93%、26.10%和21.18%,PR5分别提高23.11%、23.92%、38.66%和37.47%。【结论】筛选获得的花生种子相关促生菌,具有促进植物生长的潜力,明显促进种子萌发及幼苗生长,是良好的促生菌生物资源,具有较好的应用潜力。  相似文献

【背景】蓝细菌是水生和陆地生态系统中生物固氮的主要贡献者。【目的】增加对稻田土壤固氮蓝细菌的了解,获得用于进一步研究的可培养固氮蓝细菌菌株。【方法】选择3种具有不同固氮能力的水稻土,采用BG11-N培养基分离培养固氮蓝细菌菌株,对新分离菌株进行形态特征观察,通过基因组DNA的nifH基因扩增明确其固氮潜力,进一步采用乙炔还原法和~(15)N_2示踪法定量测定其固氮能力,通过基因组DNA的16SrRNA基因序列比对进行鉴定。【结果】在光照培养条件下,采用BG11-N培养基共分离纯化得到自养菌株7株,细胞呈圆形或椭圆形、单列、无分枝、丝状和念珠状,在固体培养基上形成团垫状菌落。新分离菌株在BG11-N培养基中生长状况良好,以基因组DNA为模板可扩增出nifH基因,乙炔还原法和~(15)N_2示踪法测定结果显示具有较高固氮能力,同时具有铁载体生成能力。结合16S rRNA基因序列比对和形态特征,7株菌被初步鉴定隶属于念珠藻科(Nostocaceae)。【结论】从水稻土中分离到在稻田生物固氮中发挥重要作用的蓝细菌(念珠藻科)菌株,可培养固氮蓝细菌菌株固氮能力较高,兼具铁载体生成能力,可作为进一步深入研究的微生物资源,具有潜在的研究应用价值。  相似文献

We isolated the feather-degrading Bacillus pseudofirmus FA30-01 from the soil sample of poultry farm. The isolate completely degraded feather pieces after liquid culture at 30°C (pH 10.5) for 3 days. Strain FA30-01 is a Gram-positive, spore-forming, rod-shaped bacterium and was identified with B. pseudofirmus based on 16S rDNA analysis. The keratinase enzyme produced by strain FA30-01 was refined using ammonium sulfate precipitation, negative-ion DEAE Toyopearl exchange chromatography, and hydroxyapatite chromatography. The refinement level was 14.5-fold. The molecular weight of this enzyme was 27.5 kDa and it had an isoelectric point of 5.9. The enzyme exhibited activity at pH 5.1–11.5 and 30–80°C with azokeratin as a substrate, although the optimum pH and temperature for keratinase activity were pH 8.8–10.3 and 60°C, respectively. This enzyme is one of the serine-type proteases. Subtilisin ALP I and this enzyme had 90% homology in the N-terminal amino acid sequence. Since this enzyme differed from ALP I in molecular weight, heat resistance and isoelectric point, they are suggested to be different enzymes.  相似文献

Enumeration of bacteria and recovery of the dominant species have been conducted for soils contaminated with fuel spills for more than 10 years at seven gas stations in Jordan. Bacterial counts of these polluted soils ranged between 0.68 × 108 and 32.8 × 108 c.f.u./g soil with two different bacterial colony types recovered on agar plates. Phenotypic examination of the recovered bacteria revealed that they belonged mainly to the genus Pseudomonas and was represented by the following species: P. acidovorans, P. putrefaciens, P. cepacia, P. vesicularis and P. fluorescens. The ability of these bacteria to grow on hexane or heptane was revealed by a colorimetric test. Action of five Pseudomonas spp.: Pseudomonas putrefaciens, P. cepacia, P. acidovorans, P. vesicularis and P. fluorescens and Rhodococcus erythropolis on 0.1% (v/v) hexane or heptane was followed at 1, 2, 6 and 12 h. Pseudomonas putrefaciens, P. cepacia and P. acidovorans were capable of degrading both hydrocarbons as indicated by the yellow colour formation (positive reaction); however, P. vesicularis and P. fluorescens showed no such capability.  相似文献

Amplified fragment length polymorphism (AFLP) analysis was used to investigate the genetic diversity in isolates of the ectomycorrhizal fungus Cenococcum geophilum from serpentine and non-serpentine soils in Portugal. A high degree of genetic diversity was found among C. geophilum isolates; AFLP fingerprints showed that all the isolates were genetically distinct. We also assessed the in vitro Ni sensitivity in three serpentine isolates and one non-serpentine isolate. Only the non-serpentine isolate was significantly affected by the addition of Ni to the growth medium. At 30 microg g(-1) Ni, radial growth rate and biomass accumulation decreased to 73.3 and 71.6% of control, respectively, a highly significant inhibitory effect. Nickel at this concentration had no significant inhibitory effect on serpentine isolates, and so the fitness of serpentine isolates, as evaluated by radial growth rate and biomass yield, is likely unaffected by Ni in the field. In all isolates, the Ni concentration in the mycelia increased with increasing Ni concentration in the growth medium, but two profiles of Ni accumulation were identified. One serpentine isolate showed a linear trend of Ni accumulation. At the highest Ni exposure, the concentration of Ni in the mycelium of this isolate was in the hyperaccumulation range for Ni as defined for higher plants. In the remaining isolates, Ni accumulation was less pronounced and seems to approach a plateau at 30 microg g(-1) Ni. Because two profiles of Ni accumulation emerged among our Ni-insensitive serpentine isolates, this result suggests that different Ni detoxification pathways may be operating. The non-serpentine isolate whose growth was significantly affected by Ni was separated from the other isolates in the genetic analysis, suggesting a genetic basis for the Ni-sensitivity trait. This hypothesis is further supported by the fact that all isolates were maintained on medium without added Ni to avoid carry-over effects. However, because AFLP analysis failed to distinguish between serpentine and non-serpentine isolates, we cannot conclude that Ni insensitivity among our serpentine isolates is due to evolutionary adaptation. Screening a larger number of isolates, from different geographical origins and environments, should clarify the relationships between genetic diversity, morphology, and physiology in this important species.  相似文献

Ecologists have long sought mechanistic explanations for the patterns of plant distribution and endemism associated with serpentine soils. We conducted the first empirical test of the serpentine pathogen refuge hypothesis, which posits that the low levels of calcium found in serpentine soils provide associated plants with a refuge from attack by pathogens. We measured the range of soil calcium concentrations experienced by 16 wild population of California dwarf flax (Hesperolinon californicum) and experimentally recreated part of this range in the greenhouse by soaking serpentine soils in calcium chloride solutions of varying molarity. When flax plants grown in these soils were inoculated with spores of the rust fungus Melampsora lini we found a significant negative relationship between infection rates and soil calcium concentrations. This result refutes the pathogen refuge hypothesis and suggests that serpentine plants, by virtue of their association with low calcium soils, may be highly vulnerable to attack by pathogens. This interaction between plant nutrition and disease may in part explain demographic patterns associated with serpentine plant populations and suggests scenarios for the evolution of life history traits and the distribution of genetic resistance to infection in serpentine plant communities.  相似文献

【背景】抗生素污染越来越引起人们的关注。利用微生物处理抗生素污染被认为是一种环境友好型的方法。【目的】筛选林可霉素高效降解菌并研究其降解机制。【方法】经形态学观察、生理生化鉴定和16S rRNA基因测序分析进行鉴定;通过PCR技术和质谱分析技术对该菌抗性基因和降解产物等进行分析。【结果】从林可霉素菌渣堆肥样本中获得一株高效降解林可霉素的假单胞菌(Pseudomonas RST-1),该菌在林可霉素浓度为3.0 g/L的牛肉膏蛋白胨培养基上培养40 h后,林可霉素降解率高达57.3%。该菌含有intI1、sul1、sul2等抗性基因,降解产物为去甲基林可霉素和2-丙基-N-甲基脯氨酸。【结论】菌株RST-1具有高效降解林可霉素的能力,推测可能的降解机制为去甲基化和酰胺键水解作用,该菌株降解特性及降解机制研究为林可霉素降解工程菌及其高效降解菌剂的研制奠定了基础。  相似文献

Peroxisomes were isolated by gradient centrifugation from two different diatoms: Nitzschia laevis (subgroup of Pennales) and Thalassiosira fluviatilis (subgroup of Centrales). In neither of these organelles could catalase or any H2O2-forming oxidase be demonstrated. The glycolate-oxidizing enzyme present in the peroxisomes is a dehydrogenase capable of oxidizing l-lactate as well. The peroxisomes also contain the glyoxysomal markers isocitrate lyase and malate synthase. However, enzymes of the fatty-acid -oxidation pathway are located exclusively in the mitochondria. The mitochondria additionally possess glutamate-glyoxylate aminotransferase and a glycolate dehydrogenase which differs from the peroxisomal glycolate dehydrogenase since it preferably utilizes d-lactate as an alternative substrate. Hydroxypyruvate reductase and glyoxylate carboligase were not found in the cells of either diatom. By culturing Nitzschia laevis it could be demonstrated that decreasing the CO2 concentration in the aeration mixture from 2% to 0.03% and increasing the irradiance from 40 to 250 mol quanta · m–2 · s–1 resulted in an increase of all peroxisomal enzyme activities. In addition, enzyme activities of the -oxidation pathway were increased. However, mitochondrial glycolate dehydrogenase and aminotransferase did not alter their activities under these conditions. Summarizing all results, it is postulated that there are two different pathways for the metabolism of glycolate in the diatoms.This work was supported by the Deutsche Forschungsgemeinschaft.  相似文献

We have undertaken a study to estimate the species diversity of zoosporogenous actinomycetes that can be isolated from an arid environment. The study site encompassed an area of approximately 22 000 square kilometers of the Mojave Desert along the California-Nevada border. A series of 29 soil samples was collected along two intersecting transects of approximately 190 and 240 km which traversed a number of distinct ecosystems. A0 horizon soils were collected from the rhizosphere of the predominant vegetation at each sampling site and screened for the target genera using selective isolation techniques: chemoattraction (xylose and -collidine) and baiting with hair. Following incubation of primary isolation plates for 28 days at 28°C, all colonies that exhibited filamentous growth, presence of sporangia and/or motile spores upon direct microscopic observation (450 and 1000×) were further characterized by fatty acid analysis (FAME). Most of the isolates fell into three broad clusters that roughly correlated with presumptive genus assignments. Individual isolates could be assigned to 226 FAME biotypes based on chromatographic similarity (85%). The dominant species (514/826 isolates) belong to a previously undescribed taxon that morphologically resemblesGeodermatophilus but possesses unique FAME profiles that include at least three novel lipids. Rhizosphere microorganisms harboring nickel hyperaccumulators, Rinorea bengalensis (Wall.) O. K. and Dichapetalum gelonioides ssp. andamanicum (King) Leenh. endemic to serpentine outcrops of Andaman Islands, India, were screened for their tolerance and accumulation of Ni. The rhizosphere soils from both the plants were rich in total and available Ni along with Co, Cr, Fe and Mg but poor in microbial density and were dominated by bacteria. Out of total 123 rhizosphere microorganisms (99 bacteria and 24 fungi), bacteria were more tolerant to Ni than fungi. Viable cells of selected Ni-tolerant bacterial isolates (MIC = 13.6–28.9 mM Ni) belonging to Pseudomonas, Bacillus and Cupriavidus were capable of accumulating nickel (209.5–224.0 μM Ni g−1 protein) from aqueous solution. Cupriavidus pauculus KPS 201 (MTCC 6280), showing highest degree of nickel tolerance (MIC 28.9 mM Ni) and uptake (224.0 μM Ni g−1 protein, 60 min) was used for detailed study. Kinetics of nickel uptake in C. pauculus KPS 201 followed a linearized Lineweaver-Burk plot. The K m and V max for nickel uptake by minimal medium grown-cells approximated 1.5 mM Ni and 636.9 μM Ni g−1 protein, respectively. The uptake process was inhibited by Co, Cu, Cd, Mg, Mn and Zn, however, complete inhibition was not achieved even in presence of 500 mM Mg. Metabolic inhibitors, sodium azide (1.0 mM) and carbonyl cyanide m-chlorophenylhydrazone (0.4 mM) strongly inhibited nickel uptake suggesting the process as an energy dependent one. The present study clearly shows that bacteria in the rhizosphere of Ni-hyperaccumulators are capable of tolerating high concentration of Ni and also possesses nickel uptake potential. The Ni-hyperaccumulators in combination with these Ni-resistant bacteria could be an ideal tool for nickel bioremediation.  相似文献
