利用线粒体C0II基因序列分析法,研究了我国尾蛱蝶属5种蝴蝶的系统分化.结果表明,在尾蛱蝶属5个种的12个样品中,405 bp长的COII片段有11.4%的位点为多态性位点,大部分的碱基改变是转换.各物种内不同个体间的差异明显小于不同物种间的差异,种内个体间的差异一般为0.5%~1.5%,各物种间的差异绝大多数在4%以上.利用最大似然性法构建的尾蛱蝶属聚类关系图显示,尾蛱蝶属蝴蝶分为两大分支,一支包括大二尾蛱蝶、二尾蛱蝶和忘忧尾蛱蝶,另外一个分支包括窄斑凤尾蛱蝶和黑凤尾蛱蝶聚在一起.在大二尾蛱蝶、二尾蛱蝶和忘忧尾蛱蝶这一分支中,大二尾蛱蝶和忘忧尾蛱蝶的亲缘关系较近,而二尾蛱蝶较远.这些分子系统学的结果均与形态学的结果相一致,是对形态分类的有力支持.  相似文献

在对中国尾蛱蝶属雄性外生殖器解剖研究的基础上,选取了23个雄性外生殖器解剖特征,利用Paup4.0b10系统发育分析软件,分析探讨了中国尾蛱蝶属系统发育关系。结果表明:中国尾蛱蝶属9个种分为2个支系,一支系由凤尾蛱蝶Polyura arja、窄斑凤尾蛱蝶P.athamas及黑凤尾蛱蝶P.schreiber3种组成,另一支系由二尾蛱蝶P.narcaea、沾襟尾蛱蝶P.posidonius、忘忧尾蛱蝶P.nepenthes、异纹尾蛱蝶P.mutata、大二尾蛱蝶P.eudamippus及针尾蛱蝶P.dolon6种组成。两大支系均以渐次方式演化;凤尾蛱蝶与窄斑凤尾蛱蝶、大二尾蛱蝶与针尾蛱蝶各自互为姊妹群关系。  相似文献

【目的】了解闪蛱蝶亚科属间及种间的分子系统进化关系。【方法】采用PCR步移法对武铠蛱蝶 Chitoria ulupi 线粒体基因组全序列进行了测定和分析。基于线粒体基因组13个蛋白质编码基因的核苷酸序列构建了38种鳞翅目昆虫的系统发育树。【结果】分析结果表明,武铠蛱蝶线粒体基因组全长15 279 bp,包括13个蛋白质编码基因、22个tRNA基因、2个rRNA基因和一段长度为391 bp的A+T富含区,基因排列顺序与其他已知近缘种昆虫相同。武铠蛱蝶线粒体基因组中存在很高的A+T含量(79.9%)。13个蛋白质编码基因中,COII以TTG作为起始密码子,COI以CGA作为起始密码子外,其余均为昆虫典型的起始密码子ATN。COII和ND4基因使用了不完全终止密码子T,其余基因均以典型的TAA为终止密码子。在所测得的22个tRNA基因中,除tRNASer(AGN)缺少DHU臂外,其余tRNA均能形成典型的三叶草结构。与其他多数鳞翅目昆虫一样,武铠蛱蝶的A+T富含区中有一段由ATAGAA引导的保守的多聚T结构,长度为21 bp,并散布着一些长短不一的串联重复单元。系统发育树结果显示,总科级别的系统发育关系为:卷蛾总科+(凤蝶总科+(螟蛾总科+(夜蛾总科+蚕蛾总科+尺蛾总科)));在蛱蝶科物种中,武铠蛱蝶与猫蛱蝶Timelaea maculate 亲缘关系最近。【结论】基于分子标记构建的鳞翅目昆虫系统发育关系与传统的形态学分类结果基本一致。  相似文献

采用PCR步移法对猫蛱蝶Timelaea maculata线粒体基因组全序列进行了测定和分析.分析结果表明:猫蛱蝶线粒体基因组全长15 178 bp,包括13个蛋白编码基因、22个tRNA基因、2个rRNA基因和一段长度为382 bp的A+T富含区,基因排列顺序与其它已知鳞翅目昆虫相同.猫蛱蝶线粒体基因组中存在很高的A+T含量(81.1%).13个蛋白编码基因中除CO Ⅰ以CGA作为起始密码外,其余蛋白质基因均以ATN作为起始密码子.COⅡ和ND4基因使用了不完全终止密码子T,其余基因均以典型的TAA、TAG为终止密码子.在所测得的22个tRNA基因中,除tRNAser(AGN)缺少DHU臂外,其余tRNA均能形成典型的三叶草结构.与其它多数鳞翅目昆虫一样,猫蛱蝶的A+T富含区中有一段由“ATAGAA”引导的保守的多聚T结构,长度为19 bp,并散在着一些长短不一的串联重复单元.  相似文献

该文对柳紫闪蛱蝶Apaturailia(鳞翅目:蛱蝶科)的线粒体基因组全序列进行了测定,同时结合其它已知蛱蝶类的相应序列进行了比较分析。结果显示:柳紫闪蛱蝶的线粒体基因组(GenBankaccessionno.:JF437925)是一个15242bp的环状DNA分子,包含13个蛋白质编码基因、2个rRNA基因和22个tRNA基因。13个蛋白编码基因中,除了COI基因的起始密码子是CGA外,其余12个蛋白编码基因都具有标准的ATN起始密码子;柳紫闪蛱蝶与其它已测的10种蛱蝶在基因定位和排列顺序方面几乎相同,只是在非编码序列上存在细微的差异,其核苷酸的构成及密码子使用频率都处于鳞翅目昆虫的范围之内。22个的tRNA基因中,除了tRNASer(AGN)缺少DHU臂,其余的tRNA基因都显示为典型的三叶草结构。基因组共存在9处基因间重叠区(总长度为33bp)以及12个基因间隔区(总长为155bp,最长间隔是49bp,最短的是1bp)。在ND6和Cytb间的间隔区中还发现有(TA)23似微卫星结构。与其他蛱蝶类相似,403bp的AT富集区包含有ATAGA,ATTTA二个保守模块(一个21bp的poly-T,一个10bp的poly-A),以及二个似微卫星的重复结构((TA)10和(TA)7)。  相似文献

该研究对斐豹蛱蝶(Argyreus hyperbius)(鳞翅目:蛱蝶科)线粒体基因组全序列进行了测定和初步分析。结果表明:斐豹蛱蝶线粒体基因全序列全长为15156bp,包含13个蛋白质编码基因、22个tRNA和2个rRNA基因以及1个非编码的A+T富集区,基因排列顺序与其它鳞翅目种类一致;线粒体全序列核苷酸组成和密码子使用显示出明显的A+T偏好(80.8%)和轻微的AT偏移(AT skew,?0.019)。基因组中共存在11个2~52bp不等的基因间隔区,总长96bp;以及14个1~8bp不等的基因重叠区,总长34bp。除COI以CGA作为起始密码子外,13个蛋白质编码基因中的其余12个基因是以ATN作为起始密码子。除COI和COII基因是以单独的一个T为终止密码子,其余11个蛋白质编码基因都是以TAA结尾的。除了缺少DHU臂的tRNASer(AGN),其余的tRNA基因都显示典型的三叶草结构。tRNA(AGN)和ND1之间的基因间隔区包含一个ATACTAA结构域,这个结构域在鳞翅目中是保守的。A+T富集区没有较大的多拷贝重复序列,但是包含一些微小重复结构:ATAGA结构域下游的20bp poly-T结构,ATTTA结构域后的(AT)9重复,以及位于tRNAMet上游的5bp poly-A结构等。这项研究所揭示的斐豹蛱蝶的线粒体基因组特征,不仅为认识蛱蝶科的遗传多样性贡献数据,而且对于该物种的保护生物学、群体遗传学、谱系地理及演化研究等具有重要意义。  相似文献

对残锷线蛱蝶(Parathymasulpitia)(鳞翅目:蛱蝶科)线粒体基因组全序列进行了测定。结果表明:残锷线蛱蝶线粒体基因组全序列全长为15268bp,除了在trnS1(AGN)和trnE基因之间有一段121bp长的基因间隔外,其基因的排列顺序及排列方向与大多数已测鳞翅目物种基本一致。在蛋白质编码基因中,除cox1以CGA作为其起始密码子之外,其余12个蛋白质编码基因都以标准的ATN作为起始密码子。此外,除nad4基因以单独的T为终止密码子,其余12个蛋白质编码基因都以TAA结尾。除trnS1(AGN)缺少DHU臂之外,22个tRNA基因都显示典型的三叶草形二级结构。除A+T富集区外的非编码序列中,线粒体基因组共含有11个基因间隔区。其中,最长的一个121bp的基因间隔区位于trnS1(AGN)和trnE之间,其A+T含量高达100%。另外,和其他鳞翅目物种一样,在其A+T富集区的3’端有一段长达18bp的poly-T结构。A+T富集区内部没有明显的小卫星样多拷贝重复序列,而含有一些微卫星样的重复结构。本研究基于13种蛋白编码基因序列的组合数据,用最大似然法和贝叶斯法对蛱蝶科几个主要亚科间共9个代表物种间的系统发生关系进行了分析。结果表明,本研究的结果与前人的分子系统学研究结论基本吻合(其中,线蛱蝶亚科和釉蛱蝶亚科互为姐妹群),而与形态学的研究结论不一致。  相似文献

Sequence divergence of mitochondrial COII was analyzed in 50 specimens belonging to five subspecies of Polyura eudamippus (Lepidoptera: Nymphalidae) collected from southern China. There were nine haplotypes in the 405 bp of partial COII sequence. Distribution of the five subspecies was primarily consistent with the spatial distribution of haplotypes. The K st statistic showed genetic differentiation among these subspecies, except between the pair of P. e. kuangtungensis and P. e. formosana, which were separated by the Taiwan Strait. This is consistent with the 10,000-year history of the Taiwan Strait, not long enough for detectable differentiation. The present distribution pattern of COII haplotypes of P. eudamippus should be shaped by the alteration of Pleistocene glaciations, and Yunnan might be the refugium of P. eudamippus in the ice age, judging from the abundant haplotypes remaining. There were two routes for P. eudamippus in the postglacial expansion, one northward to Sichuan, Chongqing, and Hubei and another eastward to the southeastern coast of mainland China and Taiwan Island. Because the haplotype of butterflies on Hainan Island (P. e. whiteheadi) was completely different from that of mainland China, it was estimated that butterflies on Hainan Island might be from the Indo-China Peninsula rather than from mainland China.  相似文献

对我国东北地区典型常见蛱蝶科15属20种蝴蝶翅鳞片的超微结构进行了扫描电镜观察。结果显示:蛱蝶翅鳞片形态上可分为窄叶形、阔叶形和圆叶形3种,鳞片长65~135 μm,宽35~85 μm,间距48~112 μm。蛱蝶翅鳞片的超微结构可分为拱桥形、棋盘形和筛孔形3 种。拱桥形结构和棋盘形结构比较接近,二者与筛孔形结构差异较明显。在已观察的种类中,线蛱蝶属红线蛱蝶翅鳞片上的纵肋突起最小(200 nm×300 nm),闪蛱蝶属柳紫闪蛱蝶翅鳞片上的纵肋突起最大(590 nm×560 nm)。鳞片具有相似的形状、结构和排列,尤其是同属蝴蝶翅鳞片超微结构的形状和尺寸差异较小,表明它们之间的亲缘关系接近。  相似文献

Phylogenetic inference of mitochondrial control region (501 bp) of 167 individuals from 12 regional populations revealed that Chinese swamp eels fell into five genetic lineages, Lineage A, B, C, D and E. Lineage A was speculated to share the same mitochondrial ancestors with the populations from Taiwan Island. Lineage C harboured most of the haplotypes (39/60) of populations and may have experienced population expansion. The distribution pattern of Lineage C from east to west regions may have resulted from the occurrence of the major glaciation and inter‐regional introduction. Lineage A, B and E inhabiting coastline regions were immune from the expansion of Lineage C due to isolation from inland areas blocking gene flow between inland and coastal populations. On the other hand, Chinese swamp eels were revealed to be maintaining substantially differentiated population structures, while three populations from the Sichuan basin (MY, LC and YA) were genetically closely related. This was attributed to the geographical isolation of Sichuan populations from other populations, facilitating gene flow among the three populations from the Sichuan basin.  相似文献

Four polymorphic microsatellite loci were identified in the butterfly Speyeria idalia. We constructed a phagemid library and screened approximately 120 000 inserts. Probing with GT15, we identified 36 positives and designed polymerase chain reaction (PCR) primers for 12 potential loci. Of those loci, only four consistently produced polymorphic, diallelic PCR products in the expected size range. These results are consistent with previous studies concerning the low frequency of microsatellite loci in the lepidoptera, although these four loci are highly polymorphic and therefore likely to provide information on the fine scale genetic structure among populations in this species.  相似文献

The complete mitochondrial genome sequence is determined for Paracymoriza prodigalis (Leech, 1889). The 15,326 bp circular molecule possesses a gene organization and order identical to other sequenced Pyraloidea mitochondrial genomes. All tRNAs have the typical clover-leaf structure except for tRNASer(AGN), which lacks the dihydrouridine (DHU) arm. The A+T-rich region of 343 bp includes the features common to the Lepidoptera, including the ‘ATAGA’ followed by an 19-bp poly-T stretch, but the tandem repeat sequences often appearing in available insects are not found. Phylogenetic relationships of eight subfamilies of 14 Pyraloidea species were constructed based on 13 PCGs of mitochondrial genomes using Bayesian inference (BI) and maximum likelihood (ML) methods. These phylogenies of the subfamilies within Pyraloidea accord well with morphological phylogenetic analysis except for the position of Schoenobiinae.  相似文献

Despite the fact that Bicyclus anynana has become an important model species for wing-pattern developmental biology and studies of phenotypic plasticity, little is known of the evolutionary history of the genus Bicyclus and the position of B. anynana. Understanding the evolution of development as well as the evolution of plasticity can be attempted in this species-rich genus that displays a large range of wing patterns with variable degrees of phenotypic responses to the environment. A context to guide extrapolations from population genetic studies within B. anynana to those between closely related species has been long overdue. A phylogeny of 54 of the 80 known Bicyclus species is presented based on the combined 3000-bp sequences of two mitochondrial genes, cytochrome oxidase I and II, and the nuclear gene, elongation factor 1alpha. A series of tree topologies, constructed either from the individual genes or from the combined data, using heuristic searches under a variety of weighting schemes were compared under the best maximum-likelihood models fitted for each gene separately. The most likely tree topology to have generated the three data sets was found to be a tree resulting from a combined MP analysis with equal weights. Most phylogenetic signal for the analysis comes from silent substitutions at the third position, and despite the faster rate of evolution and higher levels of homoplasy of the mitochondrial genes relative to the nuclear gene, the latter does not show substantially stronger support for basal clades. Finally, moving branches from the chosen tree topology to other positions on the tree so as to comply better with a previous morphological study did not significantly affect tree length.  相似文献
